The presence in rhizomorphs of Armillaria mellea and cords ofPhallus impudicus of wide diameter hyphae (5–20 {diaeresis}m),which run for considerable distances along these linear organs,has been demonstrated. The longitudinal hydraulic conductivityof these organs has been determined experimentally and similarvalues were obtained when the hydraulic conductivity was calculatedtheoretically on the basis that the vessel hyphae were the solechannel for water movement along these organs. The experimentaldata have been discussed in relation to other data for long-distancetranslocation in basidiomycete linear organs. Fungal Genetics and Biology 20, 289-298. Polarized cell growth is exhibited by a diverse range of eukaryotic and prokaryotic cells. The events which are responsible for this growth are poorly understood. However, the existence of ion gradients may play an important role in establishing and driving cell polarity. Using a pH-sensitive, ratiometric fluorescent dye to monitor intracellular pH in growing fungal hyphae, we report a gradient at the extending hyphal tip that is up to 1.4 pH units more alkaline than more distal regions. Both the magnitude and the length of the pH gradient were strongly correlated with the rate of hyphal extension and eradication of the gradient-arrested growth. These results suggest that alkaline pH gradients may be integral to hyphal extension in fungi.  相似文献

Previous studies have shown the leafy liverwort Cephaloziella varians to associate consistently with fungi, typically the ericoid mycorrhizal symbiont Rhizoscyphus ericae, across a wide latitudinal gradient in the maritime and sub-Antarctic. Hitherto, however, there are no quantitative data on the intensity of colonisation of C. varians by fungal structures in the natural environment and how colonisation might vary with changing environmental conditions. A study is hence reported showing that the frequency of colonisation by fungal structures of C. varians alters along a latitudinal transect from South Georgia (54° S, 38° W) to Moutonnée Valley on Alexander Island (71° S, 68° W). The percentage of stem length colonised by dark septate (DS) hyphae increased significantly along the transect, from 30% at South Georgia to 97% at Moutonnée Valley. In contrast, the percentage of stem length colonised by hyaline hyphae decreased significantly, from 85% at South Georgia to 13% at Moutonnée Valley, and that colonised by hyphal coils similarly decreased from 71% at the former location to 15% at the latter. The frequencies of DS hyphae were negatively associated with mean annual and seasonal air temperatures, whereas those of hyaline septate hyphae and hyphal coils were positively associated with air temperatures. Coils at northerly locations were more convoluted than those at southerly locations. The data indicate that hyphal coils, usually associated with nutrient exchange between partners in ericoid mycorrhizas, do form in C. varians tissues in the maritime and sub-Antarctic, but that the frequency of these structures diminishes in colder habitats.  相似文献

The fruiting ofFavolus arcularius in culture is described. When the cultures, which have been pre-incubated in darkness to allow the inoculum mycelia to become thick and white wooly in texture, are exposed to light, fruit-body primordia, 1 mm in height, are formed about 4 days after the start of illumination. The primordium develops into a cylindrical stipe, the growth of which mainly occurs in the final 1 mm of the terminal region. Hyphal elongation in the region within 3 mm of the apex is predominant in the growth of the pileate stipe. With maturation of the stipe, changes in hyphal orientation occur on the periphery of the subapical region, and then the pileus-primordium is formed. The differentiation into the inner layer and the outer layer (pre-hymenial layer) in the pileus tissue is completed at this stage. The early growth of the pileus may be due to rapid elongation of the hyphae on the margin in addition to gradual expansion of the hyphae in the preformed pseudo-tissue. When the pileus has grown to about 3 mm in diameter, the subsequent three to four fold increase in size may be due to parallel expansion of the hyphae constituting the young pileus tissue.  相似文献

Seeking new yeast strains having the ability to protect apple fruits against blue mould for a long time under different storage conditions was the main goal of this work. Based on the in vitro test, yeast strains KKUY0017 and KKUY0051 were selected as the most effective antagonists against Penicillium expansum. Sequencing of 26S rDNA of both yeasts confirmed that the identity of KKUY0017 and KKUY0051 was Cryptococcus albidus and Wickerhamomyces anomalus, respectively. The two strains protected the apple fruits from the blue mould disease under a wide range of temperature (5–30°C); however, W. anomalus KKUY0051 was more effective. At 25°C, W. anomalus KKUY0051 involved in the reduction of disease severity and disease incidence of blue mould by 56.49% and 57.78%, respectively. When either of the two yeasts was applied in concentration of 108 or 109 cells/mL, the maximum reduction in disease severity and disease incidence was achieved. Under cold storage (5°C), both yeast strains succeeded to protect the apple fruits free from the infection up to 24 days. Electron micrograph showed a fit attachment between the cells of C. albidus KKUY0017 and the fungal hyphae leading to the degrading of the hyphae; however, W. anomalus killed the fungal hyphae without direct attachment to them. Gas chromatography–mass spectrometry analysis of the cell-free extract of W. anomalus KKUY0051 revealed the presence of toxic compounds such as the nitrophenol derivatives. The results support the assumption that the main mode of action of this yeast is by killer toxins. We conclude that application of these yeasts under cold storage condition could keep the apple fruits free from blue mould infection for a long time.  相似文献

The colony of a filamentous ascomycete fungus typically grows as a multinucleate syncytium. While this syncytial organization has developmental advantages, it bears the risk of extensive damage caused by local injury of hyphae. Loss of cytoplasm in injured hyphae is restricted by the fast and efficient sealing of the central pores of hyphal crosswalls, or septa, by a peroxisome-derived organelle called the Woronin body. The formation of septal plugs is also associated with development and leads to separation of certain parts of the colony. Septal plugs associated with developmental processes or aging hyphae typically occur by the accumulation of sealing material. Here we report that in Neurospora crassa, a protein necessary for hyphal fusion and proper colony development called SO (SOFT) localizes to septal plugs. In response to injury, SO accumulates at the septal plug in a Woronin body-independent manner. However, the presence of the Woronin body affects the speed of accumulation of SO at the septal pore. We determined that SO contributes to, but is not essential for, septal plugging. SO accumulation was also observed at septal plugs formed during hyphal aging and during programmed cell death mediated by genetic differences at heterokaryon incompatibility (het) loci.  相似文献

The characteristics of the Polyporus Msjndus , when grown on artificial media, both solid and liquid, are described and compared with those given by Baxter.
Growth on wood of ash under laboratory conditions produces a rot which is indistinguishable from that occurring naturally. The distribution of the hyphae in the wood is described.
The mode of penetration of the cell-wall by the hyphae is figured. It is apparently by pits in the early stages of decay, but by bore-holes, formed entirely by enzyme activity, in-the more advanced rot.
The rate of growth of the fungus under controlled conditions has been measured, and shown to be 0–5 cm. per month.
Successful inoculation experiments have been carried out with young trees, confirming the results of Baxter, who states that the fungus can attack young, living sap wood.
An investigation of the enzymes produced by the mycelium has been carried out, and a method evolved for demonstrating the presence of a ligninase. This is a modification of Czapek's "hadromal" reaction.
The following enzymes are shown to be present in the mycelium: emulsin, diastase, invertase, ligninase, hemicellulase, oxidase, and catalase. The list is not intended to be exhaustive.

在植物病害研究中,观察寄主植物被病原菌入侵的过程非常重要。Solophenyl Flavine 7GFE染料可附着于菌丝,在波长为330~380 nm的激发光下被激发出蓝色荧光。为了更好的观察寄主植物中病原真菌的侵染情况,本实验以Solophenyl Flavine 7GFE为染剂,对寄主植物中病原真菌的侵染情况进行了观察研究。结果显示,用0.002%(M/V)Solophenyl Flavine 7GFE溶于0.1 mol/L Tris-HCl(p H 8.5)配制的染色液染色5 min的效果最佳;使用95%乙醇溶液替代0.15%三氯乙酸(W/V)酒精溶液∶氯仿(V/V)(4∶1)对寄主植物叶片脱色的方法操作简便、毒害较低;染色时省略了番红预染步骤。将改良的染色方法用于晚疫病菌入侵的马铃薯叶片观察取得了良好的效果。该技术是一种改良的、快速有效、安全无毒的观察真菌菌丝入侵植物的荧光染色方法,也适用于观察其他真菌入侵寄主植物组织的过程。  相似文献

【目的】确定新疆辐射污染土样中分离筛选获得的一株产多糖的耐辐射菌株W36-1的分类学地位和抗氧化特性。【方法】对该菌株进行的60Co辐照,结合菌落形态和菌丝特征显微观察、Biolog鉴定以及16S rRNA基因序列分析。采用苯酚硫酸法测定其多糖含量,采用Fenton法测定多糖的粗提物对超氧阴离子清除率;用DPPH法测定抗氧化性;采用邻苯三酚自氧化法测定对超氧阴离子的清除率。【结果】W36-1可耐5 kGy辐照,为一株欧文氏菌属(Erwinia)新种;其粗多糖含量为53.17%,该多糖对超氧阴离子清除率为75.63%,对DPPH清除率为62.43%,羟基自由基去除率为54.89%。【结论】菌株W36-1产的多糖具有抗氧化性。  相似文献

天麻(Gastrodia elata Bl.)种子可与紫萁小菇(Mycena osmundicola Lange),兰小菇(M.orchidicola Fan et Guo)等一类小菇属真菌共生萌发形成原球茎。侵入种皮的菌丝集结在柄状细胞外周的胚柄残迹中,首先侵入胚的柄状细胞,然后自柄状细胞侵入其他原胚细胞。原胚细胞发生功能分化,形成菌丝结细胞和消化细胞,初被菌丝定殖的原胚细胞具有消化菌丝的功能,随后,部分原胚细胞逐渐被菌丝充满,充育成菌丝结细胞。菌丝由菌丝结细胞进一步侵入消化细胞后最终被消化。由原球茎分化形成的营养繁殖茎(以下简称营繁茎)进一步被蜜环菌(Armilariella mellea(Vahl.Fr.)Karst.)定植,蜜环菌与紫箕小菇的菌丝同时存在于营繁茎中,但两菌相遇时都停止蔓延,互不交错侵染。  相似文献

The stipecell, subepidermal parenchyma cells and inner cortical parenchyma cells were differentiated from Gastrodia elata Bl. seed and protocorm cells when they were colonized by the fungal hyphae of Mycena osmundicola Lange and M. orchidicola Fan et Guo. The hyphae aggregated in the suspensor remnant surrounding stipecell, primarily penetrated the stipecell, and then colonized the embryo of seed. Stipecell is the unique invading site of the hyphae. Subepidermal parenchyma cells containing pelotons of hyphae is also a kind of passage cells of hyphae, but, when primarily colonized by hyphae, they can degenerate a little of hyphae. The hyphae colonizing inner cortical parenchyma cells were totally degenerated, and the function of inner ocrtical parenchyma cells is digestive. The vegetative propagation corms, which differentiated from protocorms, were recolonized by Armilariella mellea (Vahl:Fr.) Karst., and the hyphae of A. mellea and M. osmundicola were found in the same cell, but there is a layer of cells uncolonized by mycorrhizal fungal hyphae. This means the two fungal species can not crisscross colonize the cell of G. elata.  相似文献

利用光学显微镜、电子显微镜及细胞化学方法,对墨兰菌根的结构和酸性磷酸酶定位进行了初步研究。结果表明墨兰具有典型的兰科植物根结构,发现该兰花的根的外皮层不具薄壁通道细胞,菌根真菌通过破坏部分根被和外皮层细胞而侵入根的皮层细胞并在细胞内形成菌丝结,侵入的菌丝被染菌皮层细胞质膜和电子透明物质包围,进一步被消化并聚集成衰败菌丝团块。酸性磷酸酶在染菌皮层细胞及包围菌丝的皮层细胞质膜和衰败菌丝细胞壁上有强烈的酶反应,衰败菌丝周围分布有许多单层膜的含酶小泡,它们可相互愈合形成大的含酶泡或与包围菌丝的质膜融合,类似于兰科植物共生原球茎中观察到的现象。说明皮层细胞可主动释放水解酶参与对菌丝的消化  相似文献

The distribution of filamentous actin (F-actin) in invasive and noninvasive hyphae of the ascomycete Neurospora crassa was investigated. Eighty six percent of noninvasive hyphae had F-actin in the tip region compared to only 9% of invasive hyphae. The remaining 91% of the invasive hyphae had no obvious tip high concentration of F-actin staining; instead they had an F-actin-depleted zone in this region, although some F-actin, possibly associated with the Spitzenk?rper, remained at the tip. The size of the F-actin-depleted zone in invasive hyphae increased with an increase in agar concentration. The membrane stain FM 4-64 reveals a slightly larger accumulation of vesicles at the tips of invasive hyphae relative to noninvasive hyphae, although this difference is unlikely to be sufficient to account for the exclusion of F-actin from the depleted zone. Antibodies raised against the actin filament-severing protein cofilin from both yeast and human cells localize to the tips of invasive hyphae. The human cofilin antibody shows a more random distribution in noninvasive hyphae locating primarily at the hyphal periphery but with some diffuse cytoplasmic staining. This antibody also identifies a single band at 21 kDa in immunoblots of whole hyphal fractions. These data suggest that a protein with epitopic similarity to cofilin may function in F-actin dynamics that underlie invasive growth. The F-actin-depleted zone may play a role in the regulation of tip yielding to turgor pressure, thus increasing the protrusive force necessary for invasive growth.  相似文献

Using image analysis the growth kinetics of the single hyphae of the filamentous fungus Aspergillus oryzae has been determined on-line in a flow-through cell at different glucose concentrations in the range from 26 mg L-1 to 20 g L-1. The tip extension rate of the individual hyphae can be described with saturation type kinetics with respect to the length of the hyphae. The maximum tip extension rate is constant for all hyphae measured at the same glucose concentration, whereas the saturation constant for the hyphae varies significantly between the hyphae even within the same hyphal element. When apical branching occurs, it is observed that the tip extension rate decreases temporarily. The number of branches formed on a hypha is proportional to the length of the hypha that exceeds a certain minimum length required to support the growth of a new branch. The observed kinetics has been used to simulate the outgrowth of a hyphal element from a single spore using a Monte Carlo simulation technique. The simulations shows that the observed kinetics for the individual hyphae result in an experimentally verified growth pattern with exponential growth in both total hyphal length and number of tips.  相似文献

Summary After hyphal fusions between the secondary mutant and wild-type strains ofSchizophyllum commune some of the fused hyphae show several nuclei per cell and dissolved septa. These hyphae are designated migration hyphae because they are evidently the main routes of nuclear exchange between the two strains. On the wild-type side of the mating the nuclei spread gradually from the main part of the migration hyphae into the side branches, which develop into an extensive network with many anastomoses. The first cells with pseudoclamps or clamp connections are observed in this network.On the mutant side of the mating the branching is less developed and the number of anastomoses is smaller. The cross walls of the septa are poorly dissolved, and nuclear aggregations occur in the hyphae. No development of clamp connections or pseudoclamps is observed. It is suggested that the unilateral mating response of the secondary mutant strain might possibly be caused by the failure of the septa in the mutant hyphae to dissolve, which inhibits the distribution of the nuclei into the side branches of migration hyphae.  相似文献

The internal mycelium of Pseudoperonospora cubensis has been observed in transmission and scanning electron microscopic preparations. The internal mycelium may be inter- or intracellular. Haustoria of short swollen bundles of hyphae have been observed. Actively growing hyphae contain numerous mitochondria, nuclei, active dictyosomes, low amounts of storage materials (lipid) and microbody-like structures with a laminate inclusion. Thick walled hyphae with a diameter which is smaller than the actively growing hyphae have been observed. These thick wallcd hyphae contain large amounts of reserve material (lipid) and it is suggested that they may function as resting propagules.  相似文献
