The effects of Cl and pretreatment on 36Clinflux and influx into Characorallina cells were examined. Both treatments reduced 36Clinflux into C. corallina cells in the acid pH range (4.5–7.0). pretreatment stimulated influx into C. corallina cells, but Cl pretreatment hadno effect. There was a direct inhibitory effect of CI on influx into C. corallina cells, but no apparenteffect on net NO uptake. The time course of 36Cl accumulation into cytoplasmic and vacuolarcompartments during incubation of the cells with showed that significant radioactivity appeared in the vacuolarsap after 30 min. There was a linear increase in the percentageof total 36CI which crossed the tonoplast (c-v). There was noeffect of Cl or pretreatment on accumulation of radioactivity in the vacuole. Thin layer chromatography ofthe vacuolar sap showed that after 2 h only one component waspresent which had an RF which was similar to 36CI. We describe theuse of an in vivo human bronchial xenograft model of cystic fibrosis(CF) and non-CF airways to investigate pathophysiological alterationsin airway surface fluid (ASF) volume (Vs) and Cl content.Vs was calculated based on thedilution of an impermeable marker,[3H]inulin, duringharvesting of ASF from xenografts with an isosmotic Cl-free solution.These calculations demonstrated thatVs in CF xenographs (28 ± 3.0 µl/cm2;n = 17) was significantly less thanthat of non-CF xenografts (35 ± 2.4 µl/cm2;n = 30). The Cl concentration of ASF([Cl]s) wasdetermined using a solid-state AgCl electrode and adjusted for dilutionduring harvesting using the impermeable[3H]inulin marker.Cumulative results demonstrate small but significant elevations(P < 0.045) in[Cl]s in CF (125 ± 4 mM; n = 27) compared with non-CF(114 ± 4 mM; n = 48) xenografts.To investigate potential mechanisms by which CF airways may facilitatea higher level of fluid absorption yet retain slightly elevated levelsof Cl, we sought to evaluate the capacity of CF and non-CF airways toabsorb both 22Na and36Cl. Two consistent findings wereevident from these studies. First, in both CF and non-CF xenografts,22Na and36Cl were always absorbed in anequal molar ratio. Second, CF xenografts hyperabsorbed (~1.5-foldhigher) both 22Na and36Cl compared with non-CFxenografts. These results substantiate previously documented findingsof elevated Na absorption in CF airways and also suggest that theslightly elevated[Cl]s found in thisstudy of CF xenograft epithelia does not occur through a mechanism ofdecreased apical permeability to Cl.  相似文献

Smith, J. R., Smith, F. A. and Walker, N. A. 1987. Potassiumtransport across the membrane of Chara. I. The relationshipbetween radioactive tracer influx and electrical conductance.—J.exp. Bot. 38:731–751. The 42K influx () and the electrical conductance (Gm) were measured simultaneously for the ‘membrane’of internodal cells of Chara australis as a function of theexternal [KCl] (K?. In bathing solutions of pH = 5?0, progressively increased from 20?5to 430?60 nmol m–2 s–1 and Gm increased from 0?36?0?02to 3?8?0?8 S m–2 when K? was increased from 0?1 to 10mol m–3. The resting membrane potential difference (p.d.)was approximately -135 mV for low K? and approached the expectedNernst equilibrium p.d. for K+ ions when K? > 1?0 mol m–3.Measurements of 36Cl influx suggested that the 42K influx waspredominantly electrogenic. The equivalent Goldman permeabilityto K+ ions (Pk) was approximately 20–30 nm s–1 anddid not vary significantly with increasing K?. The equivalentconductance attributable to the electrogenic transport of K+ ions was calculated from assuming passive, independent diffusionof K+ ions and the ratio was found to be typically close to one. It was also found that themagnitudes of and Gm measuredsimultaneously for each individual cell were also well correlatedfor K? 1?0 mol m–3, and that the slope of the line ofbest fit was close to one. For each K? it was found that theconductance not attributable to K+ translocation and presumablyassociated primarily with the transport of protons or theirequivalents was typically 0?2–0?5 Sm–2. For K? >1?0 mol m–3 the results indicated that the transport ofK+ ions was essentially independent, i.e. there was no evidencefor flux interactions. The results also indicated that the equivalentconductance derived from the measured 42K influx could usefullyindicate the fraction of the electrical conductance attributableto the translocation of K+ ions. Stable carbon isotopes (13C) were determined for phytoplanktonand dissolved inorganic carbon (DIC) from Lake Apopka, a shallow,polymictic and hypereutrophic lake in Florida, USA. Bulk planktondominated by pico- and nanqanobacteria were enriched in 13(–13.1± 1.1%) as a result of assimilation of extremely 13C-richDIC (13C = 9.6 ± 3.0%). Diatoms (Aulacoseira spp.) hada 13C of –14.3 ± 0.6% that was slightly more negativethan that of small cyanobacteria. Meroplanktonic diatoms hada 13C (–13.6 ± 1.8%), similar to their planktoniccounterparts. The 13C of a colonial cyanobacterium (Microcystisincerta) was exceptionally heavy (–3.0 ± 1.0%)and attributed to localized carbon limitation. Seasonal variationin 13C of bulk plankton was small (4%) relative to reports forother lacustrine systems No difference in the 13C of bulk planktonhorn surface water between stratified and non-stratified periodswas found. No measurable changes in 13C of bulk plankton wereindicated in light and dark incubation experiments Frequentwind mixing of the water column, high DIC concentration, andconsistently high lake productivity were used to explain thetemporal and spatial isotope consistency of phytoplankton inthis lake.  相似文献

Naturally occurring microbial communities are complex, withautotrophs and heterotrophs often similarly sized and impossibleto separate by conventional size fractionation approaches. However,if it was possible to identify specific compounds that are characteristicof particular groups of microbes and determine the stable isotopecomposition of these biomarkers, the requirement for size fractionationcould potentially be negated. This work considered the usefulnessof such an approach by analysis of a simple laboratory predator–preysystem comprising Nanochloropsis oculata, an autotrophic flagellateprey and Oxyrrhis marina, a heterotrophic flagellate predator.In growth-grazing experiments the fatty acids 20:5(n–3)and 22:6(n–3) were used as biomarkers for N. oculata andO. marina respectively. Interpretation of 13C values of thesepredator and prey fatty acid biomarkers was not straightforwardsince although isotopic signature of the O. marina biomarkerwas consistently enriched compared to that of its N. oculataprey, the magnitude of enrichment in 13C increased with ageof culture (1.0–5.4 %). Given the variability we observedin our experimental cultures, it will be difficult to applythis approach to complex field situations without a comprehensiveunderstanding of the factors determining the 13C values of specificbiomarker molecules.  相似文献

Hajibagheri, M. A., Gilmour, D. J., Collins, J. C. and Flowers,T. J. 1986. X-ray microanalysis and ultrastructural studiesof cell compartments of Dunaliella parva. -J. exp. Bot. 37:1725–1732. Ultrastructural studies of the unicellular green alga Dunaliellaparva showed the presence of cytoplasmic vacuoles. X-ray microanalysiswas performed on sections of cells which had been freeze substitutedin acetone. It was found that the concentrations of both Naand Cl were much higher in the vacuoles than in the cytoplasm.When cells were grown in 0·4 kmol m–3 NaCl theNa and Cl concentrations in the vacuoles were 349 and 283 molm –3 respectively, whilst cytoplasmic Na and Cl concentrationswere 37 and 26 mol m–3. Corresponding values for cellsgrown in 1·5 kmol m–3 NaCl were 392 mol m–3Na and 325 mol m–3 Cl in the vacuoles and 36 mol m–3Na and 30 mol m–3 Cl in the cytoplasm. Immediately afterexposure to an increase in external salinity Na and Q concentrationsincreased in both vacuoles and cytoplasm. To find out whether the decrease in muscle performance of isolated mammalian skeletal muscle associated with the increase in temperature toward physiological levels is related to the increase in muscle superoxide (O2) production, O2 released extracellularly by intact isolated rat and mouse extensor digitorum longus (EDL) muscles was measured at 22, 32, and 37°C in Krebs-Ringer solution, and tetanic force was measured in both preparations at 22 and 37°C under the same conditions. The rate of O2 production increased marginally when the temperature was increased from 22 to 32°C, but increased fivefold when the temperature was increased from 22 to 37°C in both rat and mouse preparations. This increase was accompanied by a marked decrease in tetanic force after 30 min incubation at 37°C in both rat and mouse EDL muscles. Tetanic force remained largely depressed after return to 22°C for up to 120 min. The specific maximum Ca2+-activated force measured in mechanically skinned fibers after the temperature treatment was markedly depressed in mouse fibers but was not significantly depressed in rat muscle fibers. The resting membrane and intracellular action potentials were, however, significantly affected by the temperature treatment in the rat fibers. The effects of the temperature treatment on tetanic force, maximum Ca2+-activated force, and membrane potential were largely prevented by 1 mM Tempol (4-hydroxy-2,2,6,6-tetramethylpiperidine-1-oxyl), a membrane-permeable superoxide dismutase mimetic, indicating that the increased O2 production at physiological temperatures is largely responsible for the observed depression in tetanic force at 37°C by affecting the contractile apparatus and plasma membrane. intact mammalian muscle; physiological temperature; superoxide; excitation-contraction coupling; maximum Ca2+-activated force; muscle excitability; cytochrome c assay  相似文献

When grown under conditions of low relative humidity, the C3–C4intermediate Panicum milioides, as well as the C3 grasses Triticumaestivum and Poa pratense, exhibited 13C values which were upto 2–7%o less negative than the 13C values of the correspondingplants grown at high relative humidity. The luciferin-luciferase method was used to determine ATP extractedfrom darkmaintained and light-exposed samples of the green algaChlorella pyrenoidosa and of the blue-green alga Anacystis nidulans.A few measurements on Synechococcus lividus (a bluegreen thermophile,clone 65?C) are also reported.
  1. The light-minus-dark ATP levels (ATP) from aerobic cells ofChlorella and Anacystis were negative; however, ATP from Synechococcuswas positive. Large positive ATP was obtained in regularly grown(RG: moderate light) Chlorella treated with oligomycin; darklevels were reduced, light levels remained essentially unaffected.In high-light exposed (HLE) Chlorella, oligomycin reduced bothlight and dark ATP levels, but positive ATP was still obtained.However, in Anacystis, which has a different organization ofthylakoid membrane, oligomycin severely reduced both the lightand the dark ATP levels and the ATP remained negative.

Theoligomycin (12 µM) treated Chlorella and the untreatedAnacystis and Synechococcus show the presence of cyclic photophosphorylationunder conditions in which the non-cyclic electron flow fromphotosystem II to photosystem I is blocked by 10 µM 3-(3,4-dichlorophenyl)-l,l-dimethylurea(DCMU), or not allowed to operate by the absence of CO2. Cyclicphotophosphorylation ranged from 10–30% of the maximumATP in RG, to 40–50% in HLE Chlorella. In RG Chlorella,cyclic and non-cyclic (in the absence of DCMU) photophosphorylation(ATP) saturate at about 103 ergs cm–2 sec–1 and104 ergs cm–2 sec–1 and 104 ergs cm–2 sec–1red (>640 nm) light, respectively; a lag was observed inthe light curve.
  3. In Chlorella, the addition of the photosystemI electron acceptormethyl viologen (MV; 1 mM) increased ATPby twofold. Furtheraddition of DCMU (25 µm) reduced thisto the level observedwith DCMU alone. If 1 mM reduced dichlorophenolindophenol orphenazine methosulphate (DCPIPH2 or PMSH2, respectively)wasadded along with DCMU, the ATP level was 30–40% ofthecontrol. Further addition of MV increased the JATP to be70–80%of that of the control. These and other resultsconfirm thepresence of both non-cyclic and cyclic photophosphorylationin vivo, the former predominating in Chlorella, and the latterin Anacystis and Synechococcus.

The neuronal K-Cl cotransporter isoform (KCC2) was functionallyexpressed in human embryonic kidney (HEK-293) cell lines. Two stablytransfected HEK-293 cell lines were prepared: one expressing anepitope-tagged KCC2 (KCC2-22T) and another expressing theunaltered KCC2 (KCC2-9). The KCC2-22T cells produced aglycoprotein of ~150 kDa that was absent from HEK-293 control cells.The 86Rb influx in both cell lineswas significantly greater than untransfected control HEK-293 cells. TheKCC2-9 cells displayed a constitutively active86Rb influx that could beincreased further by 1 mMN-ethylmaleimide (NEM) but not by cellswelling. Both furosemide [inhibition constant (Ki) ~25µM] and bumetanide (Ki~55 µM) inhibited the NEM-stimulated 86Rb influx in the KCC2-9cells. This diuretic-sensitive86Rb influx in theKCC2-9 cells, operationally defined as KCC2 mediated, required external Clbut not external Na+ and exhibiteda high apparent affinity for externalRb+(K+)[Michaelis constant(Km) = 5.2 ± 0.9 (SE) mM; n = 5] but alow apparent affinity for externalCl(Km >50 mM). Onthe basis of thermodynamic considerations as well as the unique kineticproperties of the KCC2 isoform, it is hypothesized that KCC2 may servea dual function in neurons: 1) themaintenance of low intracellularCl concentration so as toallow Cl influx vialigand-gated Cl channelsand 2) the buffering of externalK+ concentration([K+]o) in the brain.

Resting egg production and oviducal cycling were investigatedfor the calanoid copepods Arctodiaptomus alpinus and Acanthodiaptomusdenticornis both in the laboratory and in a small karstic alpinelake by making a census of the number of eggs produced and theproportion of females in each of four morphologically distinguishedreproductive conditions each day in the laboratory or duringa 2–3 week period in lake enclosures. In the laboratory,individuals were maintained on a mixed diet of natural phytoplanktonat constant temperatures of 4, 10, 15 and 20C, respectively.Both species differed considerably in their temperature requirementsfor reproduction. Lifetime fecundity was highest at 10C in A.alpinusand at 20C in A.denticornis, with up to 327 eggs –1 spawnedin the former and up to 582 eggs –1 in the latter species.Unfavorable temperatures were further reflected in an increasein egg mortality and the allocation of time spent in a post-reproductivephase, as well as in a decrease of longevity. Increasing temperaturesenhanced egg production rates due to decreasing clutch productionperiods, although clutch size was negatively correlated withtemperature. Maximum rates reached 5.88 and 7.98 eggs –1day–1 in the laboratory, and 0.73 and 0.55 eggs –1day–1 in enclosures in A.alpinus and A.denticornis, respectively.Egg production rates and clutch size were clearly governed bynutritional conditions in the lake, but were less affected byfood supply in the laboratory. Here, rates of egg productionwere adapted to improving food supply by increasing the frequencyof spawning events, rather than the number of eggs per clutch.No correlation was found between female body size and reproductiveparameters in the laboratory. A very low proportion of totalclutch production resulted in clutches composed of subitaneouseggs, i.e. 0.14% in A.denticornis and 1.20% in A.alpinus. Oviducalphase duration allocations indicate that there exists a temperatureoptimum for gamete maturation.  相似文献

The stable carbon isotope composition of particulate organicmatter expressed as 13C was measured in cultures of 13 speciesof marine microalgae in different phylogenetic groups. The effectsof salinity variations and changes in photoperiod were alsoassayed for three of them (i.e. Skeletonema costatum, Amphidiniumopercularum and Isochrysis galbana); the effect of nature ofnitrogen supply (nitrate. ammonium) was studied for one (S.costatum).These environmental parameters were chosen because of theirvariability in the ocean and their possible effects on 13C valuesof phytoplankton organic carbon. Batch culture conditions andsampling time after inoculum were strongly controlled in orderto provide cells in good physiological state which were comparablefrom one culture to the other. In the same way, sampling waslimited to the first 2 days of exponential growth, in orderto avoid a possible dissolved inorganic carbon (DIC) limitation.Carboxylase activities [of the enzyme ribulose 1,5-bisphosphatecarboxylase oxygenase (Rubisco), and the three ß carboxylases:phosphoenolpyruvate carboxylase (PEPC), phosphoenolpyruvatecarboxykinase (PEPCK) and pyruvate carboxylase (PC)] and totalchlorophyll a concentrations were assayed simultaneously. The13C values observed were between –30.2 and –12.7i.e. comparable to those observed in the world's oceans. Theisotopic composition of phytoplankton organic carbon was shownto be under the influence of the parameters tested but 13C variationsare specific to the species considered. The nature of ßcarboxylase found in each species, or systematic position, couldnot be linked to the isotopic composition of organic carbon.No linear or single correlation between 13C variations and environmentalmodifications were observed and there is no evidence for a simpleand universal relation between 13C of phytoplankters and theirenvironment. In monospecific cultures as in the field, 13C fractionationby Rubisco (and eventually by PEPCK) may be counterbalancedby other mechanisms.  相似文献

The natural abundance of 13C in different parts of beech (Fagussylvatica L.) leaves was analysed. Values for leaf ribs wereconsistently higher than those for intercostal tissue. Similardifferences occur between petiole and stem, with petiole beingless negative. The pattern of results is the same, independentof the position in the tree. However, the absolute values differby up to 6%. from the bottom to the top of the tree. Valuesof 13C are in the range of – 29 to – 32%. for thelower leaf strata; while values between – 24 and –26%. have been measured for the top of the tree. Absolute 13Cvalues of the whole tissue and cellulose differ by about 2%.,but relative 13C trends are almost identical. However, 13C trendsare not identical for different leaf parts. A comparison ofcellulose and whole tissue 13C data makes it unlikely that the13C variations are primarily due to different compositions ofchemical compounds. No fractionation seems to exist betweenleaf and wood cellulose. Tissue from different areas of a leafrevealed identical carbon isotope compositions. The vacuolar pH (pHv) and the cytoplasmic pH (pHc) of the marinegiant-celled green alga Chaetomorpha darwinii were measuredby pH microelectrode techniques on extracted vacuolar sap, andby the [I4C]DMO distribution method respectively. Equilibrationof DMO occurred with a half-time of about 2 h, with an apparentPDMO of 3.6 x 10–5 cm s–1, but the vacuolar concentrationof free, undissociated DMO was always less than the externalconcentration. The explanation offered for freshwater giant-celledalgae of net DMO leakage across the plasmalemma cannotapply to Chaetomorpha darwinii, since electrically-driven DMOexit from the cytoplasm should be similar across the plasmalemmaand the tonoplast in these cells with large, vacuole-positivepotential differences across the tonoplast. pHc was accordinglycomputed assuming either tonoplast or plasmalemma equilibrationof DMO, with correction for DMO metabolism within the cell.pHc was 8.0–8.3 in the light in artificial seawater (pHoabout 8.0), was some 0.5 units lower in the dark, and was slightlylower with an external pH of 7. Vacuolar pH was 6.5–6.9,without consistent effects of illumination or of external pHof 7 rather than 8. While µH+ at the tonoplast was similarto that in giant-celled freshwater algae (although with a greatercontribution from relative to pH), µH+ at the plasmalemmawas less than 8 kJ mol–1, i.e. less than one-third ofthe value in freshwater green algae. µNa+ was some 13kJ mol–1 at the plasmalemma. The possibility that theprimary active transport process at the plasmalemma of Chaetomorphadarwinii (and certain other marine algae) is Na+ efflux ratherthan H+ efflux is discussed.  相似文献

Using carefully standardized test conditions and tandem pairsof cells of Nitella flexilis, the influx of 14C (added as H14CO3and transnodal transport were studied under various pressuregradients applied across the node up to P=± 2·5bar. When mannitol was used as the osmoticum, influx was foundto increase only when the mannitol solution was around the cellproximal to the feed. 14C was transported across the node tothe distal cell, probably as 14C-photosynthate products, evenagainst a pressure gradient of 2 bars or more, as was 36Cland 32P. The % transported in general decreased with increasingP, whether assisted or opposed by added pressure. It was unchangedby the presence of mannitol at the node and was essentiallythe same whether the pressure gradient was produced by directpressure or by use of the osmoticum. Transnodal transport of14C products is almost certainly via plasmodesmata and appearsto be largely by an active mechanism. In absolute amounts itis the same whether the pressure gradient assists or opposesflow. Valving is evident at the node, increasing the resistanceto transport as the pressure gradient increases whether 14C(asHCO3), 42K+ (as KCl), 36Cl (as NaCl) or 32P (asNa3PO4) are used to detect it. NHE1, NHE2, andNHE3 are well-characterized cloned members of the mammalianNa+/H+exchanger (NHE) gene family. Given the specialized function and regulation of NHE1, NHE2, and NHE3, we compared basal turnover numbersof NHE1, NHE2, and NHE3 measured in the same cell system: PS120fibroblasts lacking endogenous NHEs. NHE1, NHE2, and NHE3 were epitopetagged with vesicular stomatitis virus glycoprotein (VSVG). Thefollowing characteristics were determined on the same passage of cellstransfected with NHE1V, NHE2V, or NHE3V:1) maximal reaction velocity(Vmax) by22Na+uptake and fluorometery, 2) totalamount of NHE protein by quantitative Western analysis with internalstandards of VSVG-tagged maltose-binding protein, and3) cell surface expression by cellsurface biotinylation. Cell surface expression (percentage of totalNHE) was 88.8 ± 3.5, 64.6 ± 3.3, 20.0 ± 2.6, and 14.0 ± 1.3 for NHE1V, 85- and 75-kDa NHE2V, and NHE3V, respectively. Despitethese divergent cell surface expression levels, turnover numbers forNHE1, NHE2, and NHE3 were similar (80.3 ± 9.6, 92.1 ± 8.6, and99.2 ± 9.1 s1, whenVmax wasdetermined using 22Na uptake at22°C and 742 ± 47, 459 ± 16, and 609 ± 39 s1 whenVmax wasdetermined using fluorometry at 37°C). These data indicate that, inthe same cell system, intrinsic properties that determine turnovernumber are conserved among NHE1, NHE2, and NHE3.

Measurements have been made of the electrical potential differencebetween the vacuoles of single potato tuber cells and externalCl- solutions over the range 1–40 mM. With K+ as the counter-ion,the relationship between this transmembrane electrical potentialand external Cl- concentration, for fresh cells at 20° C,was found to be one of decreasing negative polarity with increasingCl- concentration (E at 1 mM Cl- external = – 81 m V;change in E for a 10-fold change in external concentration,E10 = 46 m V). The linearity of this relationship, apparenton a semi-logarithmic plot, was virtually unaltered by low temperature(0.5–2.5° C) or by previous ageing of the cells forperiods up to four days (indicating that metabolic ion absorptionis not an electrogenic process). When the counter-ion was maintainedat a constant high concentration (40 mM K+), the change in potentialover the Cl- concentration range was only 4 m V, polarity becomingmore negative with increasing Cl- concentration. With Ca++ asthe counter-ion, the potential to external Cl- concentrationrelationship was similar to that found in KCl solutions, exceptthat E10 was only about 20 m V. Curves for the influx of C1- to be expected on the basis ofthese electrochemical data alone have been shown to run closelyparallel to Cl absorption isotherms previously determinedexperimentally. This confirms the opinion, already formed onthe basis of theoretically derived values for passive Cl- influx,that Cl- uptake by both fresh and one-day-aged potato tissue,from KCl solutions and Cl- solutions with a fixed high K+ concentration,is rate-determined at o° C by passive movement across theplasmalemma. Uptake of Cl- by fresh tissue at 20° C appearsto be similarly regulated. No such parallelism was found between observed and expectedpatterns of Cl- uptake from CaCl2 solutions, or from KCl bytwo-day-aged tissue, and here factors in addition to the electrochemicalones must determine low temperature Cl-uptake.  相似文献

Net accumulation of Cl by intact barley plants was virtuallyeliminated in roots and reduced by 40% in shoots when externalmedia (0.5 mol m–3 CaSO4 plus 0–5 mol m–3KCI) were supplemented with 0.25 mol m Ca(NO3)2. Plasmalemma36Cl influx (oc) was shown to be insensitive to externalNO3- in plants which had previously been grown in solutionslacking –3, but oc became sensitive to NO3-after a lagperiod of 3–6 h. Kinetic analyses revealed that the inhibitionof 36C1 influx by external NO3- was complex. At 0.25mol m–3 NO3- the Vmax for Cl influx was reducedby greater than 50%, with insignificant effects upon Km. At0.5 mol m–3 NO3- there was no further effect upon Vmaxbut Km for influx increased from 38±5 mmol m–3to 116±26 mmol m–3. By contrast, Cl effluxwas found to be insensitive to external NO3-. A model for theregulation of Cl influx is proposed which involves bothnegative feedback effects from vacuolar NO3- +Cl) concentrationand (external) NO3- inhibition of Cl influx at the plasmalemma.These combined effects serve to discriminate against Claccumulation, favouring NO3- accumulation, when the latter ionis available. Such observations are inconsistent with recentproposals for the existence of bona fide homeostats for chlorideaccumulation in higher plants. The vapour pressures of aqueous solutions of polyethylene glycol6000 have been measured (by equilibration with sucrose solutions)up to the saturation point at 25 °C (1.45 g g–1 water).The reduced-osmotic-pressure (/c), when plotted versus concentration(c), rapidly and linearly increased up to a concentration ofabout 0.8 g g–1 (crossing the similar plot for sucrose).Above this concentration, the reduced-osmotic-pressure rosemore slowly, but still more rapidly than sucrose. The maximumosmotic pressure achieved at saturation was nearly 18 MPa. Usingthe virial equation: /c= RT/M + RTA2c, the calculated secondvirial coefficient (A2) for the linear part is 4.5 x 10–3mol g–1, a value slightly greater than most literaturevalues at 25 °C. Using NaH 14CO3 (0.1 to 3.0 mol m –3) fed to 5.0 mm ofan internodal cell of Nitella flexilis in artificial pond waterat pH 6.8 and 19–25 °C, we have found that the carbohydrates(lactose, xylose, mannose, galactoseand sucrose) are formedby photo-assimilation from 14C-DIC (dissolved organic carbon)in 1 h and are carried in the cytoplasm with amino acids (glutamineand alanine in particular) to the node attached to a tandeminternodal cell. These small sacchandes and some amino acidspassed, apparently unchanged, across the node into the sinkcell. Influx of DIC was highly sensitive to inhibitors of photosystemsI and II (at concentrations around 1.0 mol m–3) and touncouplers of phosphorylation. Most influx inhibitors, exceptfor NaN3, also reduced % transnodal transport. NH4+ (1.0 to5.0 mol m–3) appeared to reduce % transport in light (butnot in dark) with much less effect on influx. Dinitrophenoland Na citrate (at pH 8.2) also strongly reduced apparent %transport without altering cytoplasmic streaming rates. Someof the apparent reduction of transport could be due to an alterationof metabolism or of sequestering in the feed cell, but withNH4+ the latter was not detectable. Our findings support thehypothesis that transnodal transport, including that via theplasmodesmata, is at least partly ‘active’ and requiresmetabolic energy to sustain it.
