Apomicts provide the opportunity to investigate the variation in achene size and performance that is due to factors other than genetic differences between achenes. Achene characteristics of a single capitulum from each of 15 biotypes of apomicticTaraxacumwere investigated. Within a capitulum, achene fresh weight ranged from 4.45-fold to 13.74-fold. Overall, 59.8% of achenes germinated. The probability of any one achene germinating was influenced both by its source and by its weight and ranged from 0.049 to 0.902 between capitula and from 0.028 to 0.873 between weight classes. An investigation of the relationship between achene parts in three biotypes identified a linear trend for embryos to acquire a greater proportion of total achene weight as achene weight increases. Although the heaviest achenes showed the best germination, it is predicted that, for a given investment of maternal resources, seedling number would be maximized by producing achenes of a single intermediate weight.  相似文献

The effects of the cytokinin benzyladenine (BA) and the auxin naphthalene acetic acid (NAA) on Camptotheca acuminata Decaisne growth and camptothecin (CPT) accumulation (leaf CPT concentration and total leaf CPT yield) were studied in a hydroponic culture system for three weeks. Increasing BA concentrations from 0 to 3 mg l–1 in growth medium decreased plant height, stem weight, and leaf weight but increased root weight. High BA levels (1 and 3 mg l–1) increased leaf CPT concentration (% of dry weight), whereas BA applications had no effect on total leaf CPT yield, the product of leaf CPT concentration and total leaf dry weight per seedling. There was a positive correlation between root weight and leaf CPT concentration under BA treatments. NAA supplementations (from 0.5 to 4 mg l–1) to growth medium reduced plant height, leaf number, leaf length, specific leaf weight, plant weight, stem weight, and leaf weight compared with the NAA control. Meanwhile, there were no differences in plant height, leaf length, and specific leaf weight among the NAA supplementations. NAA applications had no effect on leaf CPT concentration and NAA applications decreased total leaf CPT yield. There were negative correlations between leaf number and leaf CPT concentration, leaf length and leaf CPT concentration under NAA treatments. Our results suggest that BA applications from 0.3 to 3 mg l–1 are not helpful for achieving high total leaf CPT yield and NAA applications from 0.5 to 4 mg l–1 decrease total leaf CPT yield.  相似文献

Systems for establishing suspension cultures and for inducing plant regeneration from these cultures for the Liliaceous ornamental plant, Hosta sieboldiana (Lodd.) Engl. have been developed. Pale-yellow and nodular calluses were induced from more than 20% of scape segments on MS medium containing 1 mg l–1 picloram (PIC), 30 g l–1 sucrose, and 2 g l–1 gellan gum. Upon transfer of calluses to the same medium lacking gellan gum, stably-growing suspension cultures were established after 1 month. Suspension cell clusters regenerated a large number of adventitious shoots following transfer to MS media containing 0.1 mg l–1 NAA in combination with either BA or TDZ. Over 20 shoots per 0.3 g FW of cell clusters were obtained on media containing 0.1 mg l–1 NAA and either 1 or 5 mg l–1 TDZ. Shoots rooted easily on plant growth regulator (PGR)-free MS medium, and plantlets were successfully transferred to soil. Plants showed no visible morphological alterations and maintained the diploid level as indicated by flow cytometric analysis.  相似文献

Tissue culture propagation system was developed for zedoary (Curcuma zedoaria Roscoe), a valuable medicinal plant, using rhizome sprout cultures. Shoots were induced from rhizomes on basal MS medium containing 20 g l–1 sucrose and 5 g l–1 agar, supplemented with 20 (v/v) coconut water (CW) and benzylaminopurine (BA) concentrations from 0.5 to 5.0 m g l–1. The excised shoots were subcultured on Murashige-Skoog (MS) medium with 20 (v/v) CW and different concentrations of BA and kinetin (Kin), either alone or in combination with indolebutyric acid (IBA) or naphthaleneacetic acid (NAA). MS medium with 20 (v/v) CW, 3 mg l–1 BA, and 0.5 mg l–1 IBA resulted in a multiplication rate per shoot; 5.6 shoots per explant were obtained on average after 30 days of culture. Well-developed shoots (30–40 mm in length) were rooted on MS medium containing 20 g l–1 sucrose and 8 g l–1 agar, supplemented with 20 (v/v) CW and 2 mg l–1 NAA. Petioles from in vitro grown plants of interspecific grapevine hybrids cvs `Bianca', `Podarok Magaracha' and `Intervitis Magaracha' were cultured on solid NN medium supplemented with 2,4-D and BA at various concentrations. The callus developed was cultured in liquid NN medium supplemented with 0.5 mg l–1 BA to induce formation of somatic embryos. Somatic embryos of globular and heart-stage developed in suspensions of `Podarok Magaracha' and `Intervitis Magaracha'. In contrast, `Bianca' did not undergo embryogenesis beyond globular stage. This made it necessary to perform subculture of the suspensions to HTE liquid medium supplemented with 0.2 mg l–1 BA for the development of globular embryos into heart stage. Heart-stage embryos developed into torpedo-stage after subculturing suspensions of all three cultivars to liquid HTE medium supplemented with 0.1 mg l–1 IAA and 30 mg l–1 sodium hummate. Torpedo-stage embryo suspensions were subcultured in liquid HTE medium supplemented with 0.5 mg l–1 BA, 0.5 mg l–1 GA3 and 0.5 mg l–1 GA3 + 0.2 mg l–1 BA. After 12 days of incubation, plantlets were cultured on solid M2MS medium: without growth regulators and with 0.5 mg l–1 BA. Plantlets that developed in liquid HTE media with 0.5 mg l–1 GA3 or 0.5 mg l–1 GA3 + 0.2 mg l–1 BA produced 82–90% shoots on solid M2MS medium with 0.5 mg l–1 BA after 50 days of culture.  相似文献

The purpose of these experiments is to determine the effects of foliar applications of benzylaminopurine (BAP) and gibberellic acid (GA) on tuber number production of seed potatoes. In field experiments conducted during 1989/90 cv. Mailén was used and BAP, 50 mg·l–1 was foliarly applied at (1) tuber initiation, 36 days after emergence (DAE); (2) 54 DAE; and, (3) 64 DAE. Under glasshouse conditions, in 1991/92 cv. Spunta was used and BAP 50 mg·l–1+GA 50 mg·l–1 were applied 30 and 37 days after planting/transplanting. In 1992 cv. Huinkul, Kennebec and Spunta were used and BAP 50 mg·l–1+GA 50 mg·l–1 and Biozyme (Techic SA), a commercial product with auxin (IAA, 32.2 mg·l–1), gibberellic acid (GA3, 32.2 mg·l–1) and cytokinins (zeatin, 83.2 mg·l–1) at 5 ml·l–1 were applied. In cv. Mailén, a higher tuber number in the seed fraction (<80 g) was found when BAP was applied at each of the three crop stages, while applications 54 and 62 DAE also increased tuber number in the 80–400 g fraction. As a result of BAP applications, tuber yield was also significantly increased. In the glasshouse experiments, cv. Spunta showed a significant increase in minituber production in 3 out of 4 cases, either if the mother plant came from in vitro generated plantlets or minitubers, or if GA + BAP or Biozyme was applied. It can be concluded that the use of these PGRs under both field and glasshouse conditions in cvs. Mailén and Spunta can result in increased tuber number in the seed fraction.  相似文献

A protocol for the transformation of castor embryo axes using the pCAMBIA vector 1304 in disarmed Agrobacterium tumefaciens strain EHA105 is presented. Co-cultivated explants were initially subjected to expansion and proliferation on MS medium with 0.5 mg l–1 TDZ followed by three cycles of selection on medium with 0.5 mg l–1 BA and increasing concentrations of hygromycin (20–40–60 mg l–1). Selected shoot clusters were transferred to medium with 0.5 mg l–1 BA for proliferation and 0.2 mg l–1 BA for shoot elongation. Elongated shoots were rooted on half-strength MS medium with 2.0 mg l–1 NAA. The presence and stable integration of the hpt gene was confirmed through PCR, RT-PCR, PCR-Southern blot, sequence analysis, Southern blot analysis and PCR analysis of progeny. Southern blot analysis of the primary transformants showed single copy integration and progeny analysis revealed monogenic inheritance of the introduced gene. This paper reports the first successful attempt at producing transgenic castor.  相似文献

High percentages of micro-calli and micro-derived embryos were produced from isolated asparagus microspores at late uninucleate stage on MS liquid medium supplemented with 1.0 mg l–1 2,4-D and 0.5 mg l–1 BA. Two types of calli, namely compact callus (CC) and loose callus (LC), were found. Plantlets were regenerated via organogenesis, when these calli were transferred onto MS solid medium supplemented with 1.0 mg l–1 BA and 0.2 mg l–1 IBA 6 weeks. Embryos were produced from liquid cultured microspores, or from solid cultured micro-calli. The frequencies of haploid plant production from organogenesis and embryogenesis were compared. Effects of plant growth regulators on callus production, plantlet regeneration, and haploid plant production were tested. The effect of brassinolide (BL) on cultured calluses of Spartina patens (Ait.) Muhl. (Poaceae), a halophyte monocot was studied. BL at 0.03–0.04 mg l–1 at fixed concentrations of IAA (0.2 mg l–1) and BA (3.0 mg l–1) added in MS medium increased the ratio for fresh weight (CIRFW) to dry weight (CIRDW) by 96–111% and 235–326%. Similarly, in callus regeneration capacity, BL at 0.03 mg l–1 was most effective, increasing the shoot regeneration ratio (SRR) by 425%. BL at 0.04 mg l–1 had not such an increasing effect as BL at 0.03 mg l–1, which increased SRR by 79%. However, BL at 0.005 mg l–1 promoted regenerated shoot growth most significantly, increasing the shoot height increasing ratio (SHIR) by 395% after a 40-day culture. BL at 0.05 mg l–1 was least effective in the callus regeneration and regenerated shoot growth, decreasing SRR by 27% and SHIR by 52%. Present results suggest that BL at 0.03 mg l–1 is suitable for the callus growth and shoot regeneration, while BL at 0.005 mg l–1 effectively enhanced the regenerated shoot growth.  相似文献

An efficient plant regeneration system was developed from isolated protoplasts of Echinacea purpurea L. using an alginate block/liquid culture system. Viable protoplasts could be routinely isolated from young leaves of Echinacea seedlings in an isolation mixture containing 1.0% cellulase Onozuka R-10, 0.5% pectinase and 0.3 mol l–1 mannitol. Purified protoplasts were embedded in 0.6% Na-alginate block at a density of 1 × 105/ml and cultured in a modified MS medium containing 0.3 mol l–1 sucrose, 2.5 µmol l–1 BA and 5.0 µmol l–1 2,4-D. Cell colonies were observed after 4 weeks of culture, and the protoplast-derived colonies formed calluses when transferred onto 0.25% gellan gum-solidified MS medium supplemented with 1.0 µmol l–1 BA and 2.0 µmol l–1 IBA. Shoot organogenesis from protoplast-derived callus was induced on MS medium supplemented with 5.0 µmol l–1 BA and 2.0 µmol l–1 IBA. Complete plantlets were obtained from the regenerated shoots on MS basal medium. The protoplast to plant regeneration protocol developed in this study provides the prerequisite for creating novel genotypes of this valuable medicinal species through genetic manipulation.  相似文献

A rapid micropropagation protocol was established for Aloe vera L. var. chinensis (Haw.) Berger, Chinese Aloe. The effects of three factors, namely BA, NAA and sucrose, on bud initiation were evaluated by L9 (34) orthogonal design. The variance analysis of the experimental results showed that the actions of the three factors were all considerable. Among the three factors, sucrose was the most important for bud initiation followed by BA, and NAA had the weakest effect. The best medium for bud initiation was semi-solid MS supplemented with 2.0 mg l–1 BA, 0.3 mg l–1 NAA, 30 g l–1 sucrose and 0.6 g l–1 PVP (pH 5.8), on which Chinese aloe could multiply 15 times in 4 weeks. Some shoots rooted spontaneously on 1/2 strength MS medium, but the rooting percentage was improved in the presence of 0.2 mg l–1 NAA. Rooted plantlets were acclimatized to greenhouse conditions. The young plantlets from tissue culture were transplanted successfully. In vitro propagation can be a useful tool in the conservation of this endangered medicinal species.  相似文献

疏齿千里光(Senecio subdentatus)是分布在新疆北部古尔班通古特沙漠中的一种具异形瘦果的菊科一年生短命植物。将野外观测与室内实验相结合, 对该物种异形瘦果的形态、扩散和萌发特性, 以及异形瘦果产量与植株大小的关系进行了研究, 并对其生态学意义进行了探讨。结果表明: 疏齿千里光果序中的外围果和中央果均为柱形, 但前者为淡黄色, 后者为褐色, 且二者在大小、冠毛长度及果皮微形态等方面均存在明显差异。两种瘦果均以单个果实为扩散单元, 且在静止空气中的降落速度和在1 m·s-1与2 m·s-1风速下的扩散距离无显著差异, 说明虽然果实大小和冠毛长度对瘦果扩散具有不同的影响, 但对其整体扩散能力无明显影响。在各温变周期(5/2、15/2、20/10、25/15和30/15 ℃)处理中, 淡黄色外围果的萌发率均高于褐色中央果的, 且不同温度间两种瘦果的萌发率均存在显著差异, 但光照条件对其无显著影响。果序中的中央瘦果数明显多于外围果的, 且植株中外围果所占比例与植株大小间呈显著负相关关系, 而中央果所占比例与植株大小间呈显著正相关关系。这些特点说明, 该物种的小植株倾向于产生较多较易萌发的外围果, 大植株倾向于产生较多不易萌发的中央果。在古尔班通古特沙漠不可预测的极端环境中, 疏齿千里光可通过异形瘦果间的萌发差异及调节其不同大小植株中异形瘦果的比例, 来减少同胞后代之间的竞争, 增加其对不同微环境条件的生态适应性。  相似文献

Anthers of Morus indica L., with microspores at the uninucleate stage were cultured; and the influence of temperature and kinetin pretreatment on induction of androgenic calluses was examined. The effects of various pretreatments revealed that 24 h cold pretreatment increased the percentage of cultures inducing callus. First microspore division was observed after 16 to 20 days of culture. Th anthers split and developed embryogenic calluses on MB medium supplemented with NAA (0.5 mg l–1 and BA (1.0 mg l–1)) using 8% sucrose. Rhizogenesis was induced on medium supplemented with NAA and BA (each 0.5 mg l–1) with reduced myo-inositol (75 mg l–1). Cytological study of induced roots confirmed the haploid nature of calluses. Different type of embryos were initiated upon transfer of calluses to medium supplemented with NAA, BA (each 0.5 mg l–1), 2,4-d (1.0 mg l–1) and PVP (600 mg l–1). The control of in vitro direct main stem formation by culturing receptacles, and a protocol for the micropropagation of Lilium longiflorum using in vitro main stem nodes derived from receptacle culture were developed. Receptacles from flowers cultured on MS medium containing 1.0 mg l–1 gibberellic acid (GA3) and 0.5 mg l–1 6-benzyladenine (BA) resulted in direct main stem formation after 3 months culture. These stems were isolated and cut into nodal stem segments, which were then cultured on MS medium supplemented with 0.2 mg l–1 BA. Shoots formed on each node after one month culture. These shoots were subcultured on MS medium containing 0.5 mg l–1 BA for their mass propagation. An average of 30 vigorous and uniform shoots were formed per single shoot after each subculture. A cyclic and continuous system of propagation by multiplication of shoots was developed. Shoots were rooted on 1/2 MS medium containing 0.2 mg l–1-naphthaleneacetic acid (NAA). One hundred plantlets that were acclimatized in the greenhouse had a 100% survival. A comparison was made with the traditional culture of explants derived from bulb-scales and with that from main stems.  相似文献

Isolated embryos ofKarwinskia humboldtiana were cultured in vitro. The growth of embryos and development to plantlets on woody plant medium supplemented with indole-3-acetic acid 6.10-2 mol l–1, gibberellic acid (GA3) 3.10-2 mol l–1, and 6-benzylaminopurine (BA) 2 mol l–1 was obtained. Multiplication of shoots and rooting of excised shoots has been achieved. The effects of foliar applications of nitrogen and benzyladenine (BA) on grain yield and grain protein of wheat grown under field conditions were studied over 2 years with 5 cultivars at 2 locations. Nitrogen (N) at 20 kg.ha–1, and BA at 100 or 800 mg.l–1 were applied alone or combined at pre and post-anthesis; applications of BA at 8 mg.l–1 were also made on individual ears in order to study the effect on cell number. Weekly determinations of the chlorophyll content of the flag leaf were conducted after anthesis to study leaf senescence. At harvest, yield, yield components and grain protein percentage were determined. N and BA applications delayed chlorophyll loss in the flag leaf, but modified neither yield nor yield components. Foliarly applied BA increased grain protein in four of the five cultivars tested. The association between mating systems and dispersal in plants has been studied mostly in cleistogamous species where, generally, seeds produced by cleistogamous (selfed) flowers are less dispersed than seeds produced by chasmogamous (potentially outcrossed) flowers. In heterocarpic Asteraceae, non-dispersing fruits (achenes) are produced at the periphery of the capitulum (outer florets) whereas dispersing achenes are produced by inner florets in the same capitulum. Since all the florets are protandrous, the outer floret developing first are in female phasis when anthesis of inner florets takes place. Thus, outer florets can be potentially selfed by the inner florets of the same capitulum whereas the latter must be pollinated by flowers of other capitula. Therefore outer florets should be more inbred than inner florets. To test this hypothesis, we measured the natural outcrossing rate in outer and inner florets using allozymes in three populations of the heterocarpic Crepis sancta. The results showed that the outcrossing rate was highest for non-dispersed achenes. Moreover, among the outcrossed achenes within a capitulum it was observed that the number of paternal parents of non-dispersing achenes was higher than for dispersing achenes. The pattern observed was therefore the opposite to the pattern of cleistogamous plants and contradicts the putative pollination mechanism we proposed for Asteraceae. The results agree with the predictions of sib competition theory which considers that outcrossing may minimize competitive interactions among relatives (sibs) falling near the mother plant. Higher outcrossing rate in outer florets could also occur because pollinators are more attracted to these florets.  相似文献

Callus tissue was induced in young stem segments cultured on MS based media supplemented with 0.25–0.5 mg l-1 2, 4-D. Shoots were differentiated on media containing 0.5–1.0 mg l-1 BA and 0.5–2.0 mg l-1 IBA or 0.1–0.2 mg l-1 NAA. The same media were suitable for shoot multiplication. Shoot elongation and rooting were strongly inhibited by BA and stimulated by auxins IBA and NAA. Medium containing 0.5 mg l-1 IBA was optimal for rooting. Root elongation was stimulated by light and inhibited in darkness. Transfer of rooted plantlets to outdoor conditions was feasible and special hardening procedures were not required. Among more than 5000 plants produced by this procedure only 9 off-type plants with variegated leaves were found.  相似文献

Superoxide dismutase (SOD) plays an important role in cellular defense against oxidative stress in aerobic organisms. To generate cucumber (Cucumis sativus L.) fruits producing high yields of SOD for an anti-aging cosmetic material as a plant bioreactor, the CuZnSOD cDNA (mSOD1) from cassava was introduced into cucumber fruits by Agrobacterium-mediated transformation using the ascorbate oxidase promoter with high expression in fruits. The bialaphos-resistant shoots were selected on medium containing MS basal salts, 2 mg l–1 BA, 0.1 mg l–1 IAA, 300 mg l–1 claforan, and 2 mg l–1 bialaphos. After 6 weeks of culture on the selection medium, the shoots were transferred to MS medium containing 1 mg l–1 IAA, 300 mg l–1 claforan, 2 mg l–1 bialaphos to induce roots. Southern blot analysis confirmed that the mSOD1 gene was properly integrated into the nuclear genomes of three cucumber plants tested. The mSOD1 gene was highly expressed in the transgenic cucumber fruits, whereas it was expressed at a low level in the transgenic leaves. The SOD specific activity (units/mg protein) in transgenic fruits was approximately 3 times higher than in those of non-transgenic plants.  相似文献

Poor seed development in sunflower may result from insufficient assimilate supply (source limitation). To test this hypothesis, the effects of changed source–sink ratio on seed set (measured as percentage of empty achenes) and seed filling (measured as dry mass per filled achene) in individual plants were investigated. Source–sink ratio, defined as leaf area per floret (LAF), was experimentally altered using invasive (floret removal, defoliation) and non‐invasive (pulse of chilling, short days or shading during leaf or floret initiation) treatments. Shading at floret initiation proved the most effective non‐invasive method. Generally, an increase, or decrease, in LAF improved, or impaired, both seed set and filling. Increasing LAF by 2.0 cm2[95% confidence interval (1.5, 2.5)] decreased the percentage of empty achenes by 36.9%‐points (?41.9, ?30.9) and increased dry mass per filled achene by 20.1 mg (13.6, 26.7) in the capitulum centre. The effect of source–sink ratio on seed set was always strongest in the centre, whereas peripheral whorls were not affected. Achene mass was affected in all parts of the capitulum. It is concluded that source limitation is a major cause for empty achenes in sunflower plants grown under non‐stress conditions.  相似文献
