By the age of 80 yr, the skeletal muscles of men and women decrease in mass and maximum force by approximately 30%. Severe contraction-induced injury may contribute to these age-related declines. One to two months after a 225 lengthening contraction protocol (LCP), muscles of young/adult male mice recovered completely, whereas those of old male mice sustained deficits of approximately 15% in mass and approximately 25% in maximum force. Although gender-related differences in the early events of contraction-induced injury have been reported, the recovery phase of muscles in old female animals has not been investigated. The hypothesis tested was that 2 mo after a severe LCP to the plantar flexor muscle group, the magnitude of recovery of mass and force for old female mice is less than that for adult female mice. The LCP was administered to muscles of adult and old, female C57BL/6 mice. At 3 days, 1 mo, and 2 mo following the LCP, maximum isometric force was measured, and muscles were removed and weighed. Two months following the LCP, the muscles of adult female mice recovered mass and force. In contrast, for old female mice, even after 2 mo, muscle masses were decreased by 11% and maximum forces by 38%. We conclude that, as reported previously for old male mice, a severe contraction-induced injury to muscles of old female mice results in prolonged deficits in mass and force.  相似文献

IV CS mice (ddN origin) aged 90 days (control) and 180, 210, 240, 270, 300, 330, 360 and 420 days were used for observation of the length of the estrous cycle, and the numbers of ovulated and fertilized ova. The estrous cycle between the ages of 90 and 330 days presented a regular 4-day pattern. Thereafter, regularity of the cycle declined steadily between the ages of 360 and 420 days. All mice over 420 days old exhibited cessation of cyclicity and ultimately persistent diestrus. The average number of ova ovulated in mice aged between 90 and 240 days was consistent (11.8 to 11.4 ova), whereas a steady decline was observed for mice between 270 days old (10.5 ova) and 360 days old (4.8 ova). Ova were not recovered from oviducts of mice aged about 420 days. The average number of fertilized ova (2-cell stage) in mice between 90 and 210 days old showed no significant change (11.5 to 10.3), whereas the number began to decrease in mice aged between 240 days (8.7 fertilized ova) and 330 days (2.5 fertilized ova). No fertilized ova were recovered from the oviducts of mice around 360 days old. These findings demonstrate that the decline of reproductive activity is initially observed in the number of fertilized ova, followed by a decline in the number of ovulations and finally by loss of the estrous cycle.  相似文献

Campylobacter fetus subsp. jejuni was commonly present in the feces of unweaned calves (2 to 3 weeks old) and from two of four groups of sheep. One new season lamb (12 to 16 weeks old) carried the organism, but the bacteria were not isolated from cattle. With unweaned calves, the fractions of animals infected and carcasses contaminated were similar. Contamination of carcasses usually involved low densities of C. fetus subsp. jejuni (ca. 1 to 10/cm2), which were isolated from flank but not rump areas. The organism was recovered less frequently from chilled carcasses and deboned veal. Small numbers of C. fetus subsp. jejuni could be recovered from equipment during the processing of unweaned calves but not after routine cleaning.  相似文献

Mares (n = 39) were classified according to age as young (less than 1.5 yr, n = 17) or old (more than 1.5 yr, n = 22) and sacrificed. Ovaries were measured and weighed, and the number of follicles and CL were counted. Follicle size and distribution were recorded (external: > 20 mm, < 20 mm; internal: > 5 mm, < 5 mm). External follicles were aspirated while internal follicles were sliced. The number and Type of oocytes recovered using each method were recorded. Oocyte recovery rates (oocytes/ovary) resulted in a mean of 0.92 oocytes by aspiration and 1.36 oocytes by additional slicing. The mean numbers of available follicles (8.11 and 5.02) oocytes recovered (4.94 and 3.02) and oocytes selected per ovary (3.29 and 2.32) were not significantly different in the young or old mares, respectively.  相似文献

A scanning electron microscopic study was performed to observe the tegumental ultrastructures of Paragonimus iloktsuenensis according to its developmental stages. The metacercariae were obtained from the liver of the brackish water crab, Sesarma dehaani. Juvenile and adult P. iloktsuenensis were recovered from the experimental rats on 2, 4 and 8 weeks after infection. The findings were summarized as follows: 1. The excysted metacercariae were characteristically gourd-shape, with their whole body surface beset with numerous spade-shape spines. The large, type II sensory papillae (non-ciliated round swellings) were arranged along the rim of the oral and ventral suckers, 11-12 and 6-8 in numbers respectively. 2. Two-week old juvenile worms, recovered chiefly from the liver of the experimental rats, were slender in body shape, with their ventral sucker near the anterior one-third level. The distribution of tegumental spines was less dense than in the excysted metacercariae. The spines were with 1-2 pointed tips and 3-4 longitudinal splits. Numerous ciliated knob-like, type I papillae were observed in both sides of the oral sucker, and 6 large, type II papillae were arranged along the rim of the ventral sucker. 3. Four-week old worms, recovered from the thoracic cavity and/or lung parenchyme of the experimental rats, were thicker than wide in body configuration, and their ventral sucker was located near the anterior one-fourth level. The tegumental spines at ventral surface were grouped, each group with 3-5 aggregated ones. The type I and type II papillae (small-sized) were distributed chiefly around the rim of two suckers. 4. Adult (eight-week old) worms, recovered from the capsules in the lung parenchyme, were very stout, and covered densely with bearfoot-like spines. At dorsal surface, cobblestone-like cytoplasmic processes were well-developed, with many tegumental spines embedded in them. It was observed in this study that the tegument of P. iloktsuenesis continued to change and differentiate as the worms grew to be adults.  相似文献

The European corn borer, Ostrinia nubilalis (Hübner) (Lepidoptera: Crambidae), is often controlled with genetically modified corn, Zea mays L., hybrids (Bacillus thuringiensis [Bt] corn) in the United States. If Bt-resistant insects are detected in the field, mitigation-remediation tactics must be implemented to sustain the efficacy of insecticidal, transgenic corn. Mass releasing laboratory-reared, susceptible adults near aggregation sites to mate with locally emerging resistant adults is a possible remediation tactic, but it is imperative that the former remain in or near the release site long enough to mate. Understanding adult dispersal behavior relative to the timing of mating is important, because it directly affects patterns of gene flow and the rate at which Bt resistance moves through a population. Previous work shows that newly eclosed adults do not remain in proximity to their natal field. However, moth age, reproductive development, or mating status may influence the propensity to disperse. The objectives of this study were to determine the effect of adult age (0-3, 4-6, and 7-10 d old) and mating status on dispersal of adults released in small-grain aggregation plots. Less than 1% of the marked adults released in the aggregation plots remained after one night. More males than females were recovered. Age influenced dispersal, with mostly 4-6-d old adults being recovered. Conversely, mating status did not affect the number of adults recovered. Given the paucity of marked adult moths recovered near their release sites, mass releases of adults may not be a viable tactic to combat the spread of resistance to Bt corn.  相似文献

A simple method is described whereby pure chloroauric acid (HAuCl4; "brown gold chloride") is recovered from old solutions used for toning silver-stained sections. The procedure can be carried out in any laboratory and 60 to 80% of the gold initially present in a solution is recovered. Immunogenicity of Plasmodium gallinaceum sporozoites for chicks and their in vitro reactivity with normal and specific immune sera were studied. Two sporozoite populations recovered from experimentally infected Aedes fluviatilis were used: sporozoites from salivary glands and sporozoites from midgut oocysts. Populations seven to nine days old of sporozoites recovered from salivary glands were infective for all chicks until the chicks were three weeks old; however, sporozoites recovered from midguts containing oocysts infected these chicks only if isolated on days 8-9, but not on day 7 after the mosquitoes' infective blood meal. Infectivity of the sporozoites was lost after exposure to ultraviolet (UV) light (30 min) or X-rays (13 krad). Inactivated sporozoites from both sources proved highly immunogenic to chicks that were immunized by several intravenous or intramuscular injections. These parasites elicited a strong humoral immune response in the chicks, as measured by the circumsporozoite precipitation (CSP) reaction. The levels of the CSP antibodies were similar with sporozoites from both sources, there being no detectable differences in the percentage of reactive sporozoites or the intensity of the CSP reaction with sera containing antibodies to either sporozoites from salivary glands or sporozoites from oocysts. These results provide the first evidence that avian malaria sporozoites express the circumsporozoite protein that has been extensively characterized in mammalian malaria (rodent, simian, human sporozoites). Furthermore, we observed that the yields of sporozoites obtained from mosquito midguts, on days 8 and 9 of the P. gallinaceum infection, were at least twice as great as those obtained by salivary gland dissection, even 20 days after a blood meal.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

In vitro fertilisation after stimulation of the ovulatory cycle has led to successful pregnancy. If more oocytes are recovered than are needed they may be left unfertilised, preserved, or donated to a recipient couple from whom oocytes cannot be obtained. A case of human pregnancy initiated by transfer of a donated embryo fertilised in vitro is reported. The donor was a 42 year old woman with primary infertility from whom six follicles were aspirated after stimulation of the ovulatory cycle. The recipient was a 38 year old infertile woman who had undergone several unsuccessful attempts for artificial insemination from a donor. Five oocytes were recovered from the donor''s six follicles, four of which were inseminated with spermatozoa of the donor''s husband and the fifth with a frozen sample of semen. Three of the four embryos fertilised by her husband were returned to the donor and the fifth was transferred to the recipient. No pregnancy was recorded in the donor, but pregnancy was confirmed in the recipient, though spontaneous abortion occurred after 10 weeks. This case will give useful information for further study of in vitro fertilisation, but also raises many ethical issues.  相似文献

Human skeletal remains recovered at a new archaeological site, dating back to the Italian Mousterian age are analyzed. The finds consist of a left parietal of a child about 2-3 years old. The metrical, morphometrical, and morphological traits of this parietal are compared to those of other Neanderthal children. The geological features of the site, its fauna, and stone artifacts recovered there during quarry operations are also analyzed.  相似文献

Three 58-day old Small-Ear-Miniature (SEM) pigs, six 36-day old Landrace-Small-Ear-Miniature (L-SEM) pigs, and two 5-day old Holstein calves were each fed 3000 or 30,000 Indonesia Taenia (Samosir strain) eggs and sacrificed 27-195 days after inoculation. A total of 4922 cysticerci were recovered only from the livers of the three SEM pigs (1977 cysticerci) and six L-SEM pigs (2945 cysticerci). The infection rate in pigs was 100%. Cysticerci recovery rates of SEM and L-SEM pigs were 22.0% and 1.6%, respectively. Calves were not susceptible to Indonesia Taenia. More cysticerci were found in the liver parenchyma (L-SEM, 66.4%; SEM, 76.2%) than on the liver surface (L-SEM, 33.6%; SEM, 23.8%) of the infected animals. Most (99.86%) of the cysticerci recovered from the livers of L-SEM pigs were degenerated or calcified but 77.9% of those in the livers of SEM pigs were mature and only 22.1% were degenerated or calcified. Measurements of length, width, diameters of protoscolex, rostellum, and sucker and hooklet pattern indicated that Indonesia Taenia is very similar to Taiwan Taenia and very different from Taenia saginata and Taenia solium. The present findings indicate that Indonesia Taenia and Taiwan Taenia may be the same new species.  相似文献

There was no significant difference in the number of E. caproni recovered 21 days post-infection from 1-, 2-, 4-, 5-, 6-, 7-, 12-, or 21-month-old ICR mice infected with 25 metacercarial cysts. A range of 10.8-17.3 worms was recovered from mice in the age groups. In a similar experiment with E. trivolvis, there was no significant difference in worms recovered in young vs old mice. A range of 0-1.7 worms was recovered from mice in each age group. Contrary to a previous study on E. caproni in NMRI mice, our results indicate that host age does not affect establishment of E. caproni or E. trivolvis in the ICR mouse.  相似文献

Young and old hybrid female mice were given 0.5 Gy or 2 Gy acute x-irradiation, followed by (i) in utero examination for dominant lethal mutations, or (ii) examination of metaphase I oocytes for chromosome aberrations 2-3 weeks after the irradiation. Some of the old females had been mated when young to males of a specific locus stock. Others were left unmated until after the irradiation when they, and the young females, were mated to the same specific locus stock and allowed to have 1 (if given 2 Gy) or 2 (if given 0.5 Gy) litters before the dominant lethal test. In both the 0.5-Gy and 2-Gy series, mean sizes of first litters in the old late-mated group were markedly lower than in the old early-mated or young groups, the differences being significant at the 2-Gy level. The intrauterine examinations showed that this difference was largely the result of a reduced ovulation rate in the old late-mated females. Preimplantation loss tended to be higher in all the old females than in the young ones, but differences between the groups in postimplantation lethality were less pronounced. In the chromosome studies, only about half as many oocytes were recovered from the ovaries of old females than from young ones. At both the 0.5-Gy and 2-Gy dose levels interchange frequencies were non-significantly higher in old than in young females (with no clear-cut effect of mating status), while the overall frequency of aberrations (interchanges + fragments) was significantly higher in oocytes of old than young females after 2 Gy X-rays (35.5% against 12.5%). No specific locus mutations were found in 5616 offspring of unirradiated females.  相似文献

An outbreak of a pneumoenteric disease occurred in neonates in a zoo nursery. Four of seven affected animals died. Rotaviruses were observed in the feces of an affected 4-day old impala (Aepyceros melampus), a Thomson's gazelle (Gazella thomsonii) and an addax (Addax nasomaculatus). Encapsulated Escherichia coli also were isolated from the feces. The recovered rotaviruses was antigenically related to bovine rotavirus. A bovine rotavirus vaccine was given orally and no adverse effects were noted.  相似文献

One year old fillies are able to conceive but, usually, not to give birth to a living foal. Although embryo transfer allows the production of foals from mature mares with repeated pregnancy losses, no reports are available on the use of one year old fillies as embryo donors. To evaluate this possibility, eleven 12-16 months old Haflinger mares were inseminated with fresh semen and subjected to embryo recovery. Some of the recovered embryos were non-surgically transferred into synchronized mature recipients. Pregnancies were terminated using PGF2alpha at day 25. Fillies' embryo recovery rate and their recipients' pregnancy rate at day 25 were compared with those achieved in two years old and mature mares of the same breed, subjected to the same management. Embryo recovery rate was 21/44 (47.7%), 12/16 (75%) and 22/26 (84.6%) (P>0.01) for one year old, two years old and mature mares, respectively. Five/7 (71.4%) one year old donors' embryos resulted in a pregnancy after transfer and 4/7 (57.1%) developed until day 25. Significant differences in pregnancy rates after transfer between donors' age groups were not observed; no short term side effects resulted from the use of fillies as embryo donors. This study showed that one year old mares employed as embryo donors produce embryos both morphologically normal and able to develop in recipient mares at least up to day 25 of pregnancy.  相似文献

Gas vacuolate populations of Aphanizomenon and Anabaena were cultured from sediment up to 18 and 64 years old respectively. 兰州生物制品研究所研制的流行性感冒病毒裂解疫苗于2003年9月~12月在广西进行Ⅰ~Ⅲ临床试验,对疫苗的安全性和免疫原性进行考核评价。试验中随机选取852人(6月龄~67岁)接种试验疫苗,227人接种对照疫苗。6~36月龄的婴幼儿接种2针,每次0.25m l,间隔28天;成人接种0.5 m l。所有接种对象均未见红肿和硬结等局部反应;发生低热反应(37.1℃~37.5℃)率为3.5%,均于48小时内恢复正常。疫苗接种后易感人群的H I抗体总阳转率为100%,非易感人群的H I抗体几何平均效价增长7.1~16.8倍,抗体4倍增长率为73.1%~91.7%。证实该疫苗具有良好的安全性和免疫原性。  相似文献

With aging, the skeletal muscles of humans sustain decreases of approximately 30% in mass and maximum force. Contraction-induced injury may contribute to these declines. When a 225 lengthening contraction protocol (LCP) was administered to small, non-weight-bearing muscles of mice, muscles of young/adult mice recovered completely, whereas those of old mice sustained permanent deficits of 20% in muscle mass and maximum force. Despite these observations, whether a large, frequently recruited, weight-bearing muscle sustains such permanent damage is not known. The hypothesis tested is that after a severe contraction-induced injury, large, weight-bearing muscles of old mice sustain permanent reductions in mass and force. The LCP was administered to plantar flexor muscles of adult and old, male C57BL/6 mice. At 3 days, 1 mo, and 2 mo after the LCP, maximum isometric forces were measured, anesthetized mice were euthanized, and muscles were removed and weighed. Two months after the LCP, the muscles of the adult mice regained control values of mass and force, whereas for muscles of old mice the mass decreased by 24% and the maximum force decreased by 32%. We conclude that a severe contraction-induced injury to large, weight-bearing muscles of old mice causes permanent deficits in mass and force.  相似文献

The peculiarities of dynamics of quantitative changes of some classes of lipid and phospholipid spectra of blood plasma of calves recovered after dyspepsia were studied. Obtained reliable changes of the blood plasma lipidogrammas testify to development of dyslipidemia. It is characterized by hypercholesterolemia and hypertriacylglycerolemia of recovered 30 days old calves 3 weeks after diseases symptoms past. These changes give evidence concerning deficiency of phosphatides choline fraction - main structural components of cell membranes. It was established that changes of lipid and phospholipid spectra of blood plasma caused by enteropathology can be corrected by the inclusion of reparative therapy preparations to dyspepsia treatment plan in particular--experimental phospholipid containing a drug, which is prepared on the basis of milk phospholipids--its natural source for newborn calves.  相似文献
