In a 2-year study, fruiting plants of strawberry (Fragaria × ananassa Duch.) cv. ‘Korona’ and ‘Elsanta’ were exposed for 2 months to 78 ppb ozone on average or filtered air without ozone in controlled environment chambers. Plant growth, photosynthesis, carbohydrate accumulation, and macronutrient concentrations were investigated in order to demonstrate cultivar-specific differences in the ozone sensitivity of ‘Korona’ and ‘Elsanta’ on the whole plant level. Moreover, the hypothesis was tested whether properties of the root system in strawberry were involved in ozone tolerance, for example, the roots’ ability to store or make available carbohydrates and their capacity to secure plants’ supply with nitrogen during a stress situation. In strawberry, ozone reduced leaf area by reducing leaf number. Moreover, specific leaf area (SLA) and relative leaf water content were reduced. Net photosynthesis was only slightly impaired, but activity of Rubisco and chlorophyll content in older leaves of cv. ‘Elsanta’ were significantly reduced. The most important, indirect impairment of photosynthesis was the reduction of plants’ total leaf area, which resulted in a decrease in plant biomass. The reduction of root biomass, the root/shoot ratio, and also the distribution of carbohydrates indicated a partitioning priority of the shoot at expense of the root system. Cultivar ‘Elsanta’ was characterized by significantly lower carbohydrate levels in ozone-exposed leaves, whereas levels remained fairly stable in ‘Korona’ leaves. In addition, nitrogen concentrations in leaves and roots decreased significantly in ‘Elsanta’, not in ‘Korona’. The reduced nitrogen concentration in leaves may be related with the more distinct reduction in Rubisco activity and chlorophyll content in older leaves of ‘Elsanta’.  相似文献

Hydrogen peroxide generation rates of uninfected and infected leaves of two tomato (Lycopersicon esculentum) cultivars showing differential susceptibility to Botrytis cinerea were determined. The superoxide anion, hydroxyl radical, ascorbate contents and changes in NADH peroxidase, superoxide dismutase (SOD), ascorbate peroxidase (APX) and catalase (CAT) activities in the apoplast fraction were analysed. Infected leaves had an increased hydrogen peroxide level. It was greater and generally occurred earlier in plants of the less susceptible cv. Perkoz than in those of the more susceptible cv. Corindo. Induction of nitrotetrazolium blue reducing activity and SOD levels in apoplast were higher in cv. Perkoz 24 h after inoculation. In the controls, NADH peroxidase activity in apoplast was higher in the more susceptible cv. Corindo, but after infection it increased faster and to a higher level in the less susceptible cv. Perkoz. NADH oxidation was inhibited by only 15% by a specific inhibitor DPI (diphenylene‐iodonium) but was completely inhibited by KCN and NaN3. Similar increases in APX activity after 48 h and a small increase in catalase activities were observed in both cultivars soon after infection. These results indicate that resistance of tomato plants to infection by the necrotrophic fungus B. cinerea may result from early stimulation of hydrogen peroxide and superoxide radical generations by NADH peroxidase and SOD in apoplastic space, and they confirm the important role of their enhanced production in apoplastic spaces of plants.  相似文献

The aim of this work was to examine the correspondence between apoplastic/symplastic antioxidant status and previously reported plant age-related shifts in the ozone (O3) resistance of Plantago major L. Seed-grown plants were fumigated in duplicate controlled environment chambers with charcoal/Purafil®-filtered air (CFA) or CFA plus 70 nmol mol−1 O3 for 7 h d−1 over a 42 d period. Measurements of stomatal conductance and antioxidants were made after 14, 28 and 42 d fumigation, on leaves at an equivalent stage of development (youngest fully expanded leaf, measured c . 9 d after emergence). Ozone exposure resulted in a similar decline in stomatal conductance across plant ages, indicating that increases in O3 resistance with plant age were mediated through changes in the tolerance of leaf tissue rather than enhanced pollutant exclusion. Leaf apoplastic washing fluid was found to contain 'unspecific' peroxidase, ascorbate peroxidase, superoxide dismutase and ascorbate, but not glutathione and the enzymes required to facilitate the regeneration of ascorbate from its oxidized forms. A weak induction in the activity of certain symplastic antioxidants was found after 14 d O3 fumigation, despite a lack of visible symptoms of injury, but shifts in symplastic antioxidant enzyme activity were not consistent with previously observed increases in resistance to O3 with plant age. By contrast, changes in 'unspecific' peroxidase activity and in the small pool of ascorbate in the leaf apoplast were found to accompany age-related shifts in O3 resistance. It is concluded that constituents of the leaf apoplast may constitute a potentially important front line defence against O3.  相似文献

The present work describes, for the first time, the changes that take place in the leaf apoplastic antioxidant defenses in response to NaCl stress in two pea (Pisum sativum) cultivars (cv Lincoln and cv Puget) showing different degrees of sensitivity to high NaCl concentrations. The results showed that only superoxide dismutase, and probably dehydroascorbate reductase (DHAR), were present in the leaf apoplastic space, whereas ascorbate (ASC) peroxidase, monodehydroascorbate reductase (MDHAR), and glutathione (GSH) reductase (GR) seemed to be absent. Both ASC and GSH were detected in the leaf apoplastic space and although their absolute levels did not change in response to salt stress, the ASC/dehydroascorbate and GSH to GSH oxidized form ratios decreased progressively with the severity of the stress. Apoplastic superoxide dismutase activity was induced in NaCl-treated pea cv Puget but decreased in NaCl-treated pea cv Lincoln. An increase in DHAR and GR and a decrease in ASC peroxidase, MDHAR, ASC, and GSH levels was observed in the symplast from NaCl-treated pea cv Lincoln, whereas in pea cv Puget an increase in DHAR, GR, and MDHAR occurred. The results suggest a strong interaction between both cell compartments in the control of the apoplastic ASC content in pea leaves. However, this anti-oxidative response does not seem to be sufficient to remove the harmful effects of high salinity. This finding is more evident in pea cv Lincoln, which is characterized by a greater inhibition of the growth response and by a higher rise in the apoplastic hydrogen peroxide content, O(2)(.-) production and thiobarbituric acid-reactive substances, and CO protein levels. This NaCl-induced oxidative stress in the apoplasts might be related to the appearance of highly localized O(2)(.-)/H(2)O(2)-induced necrotic lesions in the minor veins in NaCl-treated pea plants. It is possible that both the different anti-oxidative capacity and the NaCl-induced response in the apoplast and in the symplast from pea cv Puget in comparison with pea cv Lincoln contributes to a better protection of pea cv Puget against salt stress.  相似文献

Two strawberry (Fragaria × ananassa Duch.) cvs Korona and Elsanta differing in their tolerance to NaCl salinity were exposed to 40 and 80 mmol NaCl L?1 for over 4 months in the growing seasons of 2002 and 2003, respectively. However, the osmotic potential, i.e. the NaCl concentration of the root medium, varied during the experiments, because Hoagland solution and demineralized water were added usually once a week in order to push NaCl uptake on the one hand, but to allow leaching the soil after application of demineralized water on the other. Leaching the soil should quickly improve the water relations of the plant, but not affect salt levels within the plant. This strategy was chosen to reduce the effects of water stress and to focus onto the salt-specific impacts of NaCl stress. The salt stress reduced fresh and dry matter of the whole plants and photosynthetically active leaf area, especially in cv. Elsanta. Typical leaf symptoms of Na and Cl stress were detected in both cvs and the combined effects of both toxic ions resulted in the leaf scorching symptoms. Na uptake of both cvs was similar, but distribution of Na within plants was different. Korona was able to protect leaves more efficiently from Na accumulation.Under NaCl stress Korona plants achieved a significant increase of K content in leaves and crowns, while Elsanta showed an increase of K in fruits and petioles. The accumulation of K under evaluated NaCl levels suggests an efficient K uptake system in strawberry plants. Concentrations of Ca were not significantly affected, with the exception of rising levels in roots of Elsanta plants. Concentrations of Mg, Mn and Fe significantly decreased in leaves, while those of Mg and Mn remarkably rose in crowns of both cvs. N content in leaves, petioles, and roots of both cvs increased. In addition it rose in fruits and crowns in cv. Elsanta. A significant limitation of N uptake by competition with Cl did not occur in these plants. Concentrations of P increased in roots and petioles of both cvs, and in fruits of cv. Elsanta. With respect to Zn and Cu, significant concentration changes related to NaCl stress could not be detected.  相似文献

Phaseolus vulgaris cv. Korona plants were inoculated with the bacteria Pseudomonas syringae pv. phaseolicola (Psp), necrotrophic fungus Botrytis cinerea (Bc) or with both pathogens sequentially. The aim of the experiment was to determine how plants cope with multiple infection with pathogens having different attack strategy. Possible suppression of the non-specific infection with the necrotrophic fungus Bc by earlier Psp inoculation was examined. Concentration of reactive oxygen species (ROS), such as superoxide anion (O2 ?) and H2O2 and activities of antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD) were determined 6, 12, 24 and 48 h after inoculation. The measurements were done for ROS cytosolic fraction and enzymatic cytosolic or apoplastic fraction. Infection with Psp caused significant increase in ROS levels since the beginning of experiment. Activity of the apoplastic enzymes also increased remarkably at the beginning of experiment in contrast to the cytosolic ones. Cytosolic SOD and guaiacol peroxidase (GPOD) activities achieved the maximum values 48 h after treatment. Additional forms of the examined enzymes after specific Psp infection were identified; however, they were not present after single Bc inoculation. Subsequent Bc infection resulted only in changes of H2O2 and SOD that occurred to be especially important during plant–pathogen interaction. Cultivar Korona of common bean is considered to be resistant to Psp and mobilises its system upon infection with these bacteria. We put forward a hypothesis that the extent of defence reaction was so great that subsequent infection did not trigger significant additional response.  相似文献

Summary The apoplast of barley and oat leaves contained superoxide dismutase (SOD), catalase, ascorbate peroxidase, dehydroascorbate reductase, monodehydroascorbate reductase, and glutathione reductase activities. The activities of these enzymes in the apoplastic extracts were greatly modified 24 h after inoculation with the biotrophic fungal pathogenBlumeria graminis. The quantum efficiency of photosystem II, which is related to photosynthetic electron transport flux, was comparable in inoculated and healthy leaves during this period. Apoplastic soluble acid invertase activity was also modified in inoculated leaves. Inoculation-dependent increases in apoplastic SOD activity were observed in all lines. Major bands of SOD activity, observed in apoplastic protein extracts by activity staining of gels following isoelectric focusing, were similar to those observed in whole leaves but two additional minor bands were found in the apoplastic fraction. The apoplastic extracts contained substantial amounts of dehydroascorbate (DHA) but little or no glutathione (GSH). Biotic stress decreased apoplastic ascorbate and DHA but increased apoplastic GSH in resistant lines. The antioxidant cycle enzymes may function to remove apoplastic H2O2 with ascorbate and GSH derived from the cytoplasm. The effects of nitric oxide (NO) on chilling tolerance (freezing injury, ice nucleation activity, contents of hydrogen peroxide and superoxide anion, and lipid peroxidation level) and the activities of apoplastic antioxidant enzymes (peroxidase and superoxide dismutase) were investigated in the leaves of maize (Zea mays) exposed to short-term chilling. NO treatment was carried out through spraying of sodium nitroprusside (SNP), which is a donor of NO, in concentrations of 0.0, 0.1 and 1 μM on the leaves of 10-day plants. The plants then were transferred into the chilling condition (10/7 °C) 2 days before the harvesting of leaves (14th and 21th days). Application of 0.1 μM NO had more effect on the alleviation by decreasing the freezing injury in maize at least for 11 days after the application. Both concentrations of NO generally increased ice nucleation activity of apoplastic proteins extracted from leaves. The SNP applications decreased the contents of reactive oxygen species such as hydrogen peroxide and superoxide anion and the level of lipid peroxidation, while further increasing the activities of the apoplastic antioxidant enzymes studied. The results show that exogenous NO treatment provides important contributions to increasing the chilling tolerance of maize by regulating the biochemical mechanisms of chilling response, including apoplastic antioxidant enzymes. It can be seen that the NO treatment can play positive roles in alleviating chilling-induced damage in maize. Therefore, it is suggested that NO treatments may contribute to research studies related to diminishing chilling-induced damage in agricultural applications.  相似文献

Atmospheric ozone causes formation of various highly reactive intermediates (e.g. peroxyl and superoxide radicals, H2O2, etc.) in plant tissues. A plant's productivity in environments with ozone may be related to its ability to scavenge the free radicals formed. The effects of ozone on photosynthesis and some free radical scavengers were measured in the fifth emergent leaf of poplars. Clonal poplars (Populus deltoides × Populus cv caudina) were fumigated with 180 parts per billion ozone for 3 hours. Photosynthesis was measured before, during, and after fumigation. During the first 90 minutes of ozone exposure, photosynthetic rates were unaffected but glutathione levels and superoxide dismutase activity increased. After 90 minutes of ozone exposure, photosynthetic rates began to decline while glutathione and superoxide dismutase continued to increase. Total glutathione (reduced plus oxidized) increased in fumigated leaves throughout the exposure period. The ratio of GSH/GSSG also decreased from 12.8 to 1.2 in ozone exposed trees. Superoxide dismutase levels increased twofold in fumigated plants. After 4 hours of ozone exposure, the photosynthetic rate was approximately half that of controls while glutathione levels and superoxide dismutase activity remained above that of the controls. The elevated antioxidant levels were maintained 21 hours after ozone exposure while photosynthetic rates recovered to about 75% of that of controls. Electron transport and NADPH levels remained unaffected by the treatment. Hence, elevated antioxidant metabolism may protect the photosynthetic apparatus during exposure to ozone.  相似文献

In zinc-deficient bush beans (Phaseolus vulgaris L. cv. Saxa)ozone sensitivity was enhanced compared to plants sufficientlysupplied with this nutrient. This was correlated with reducedlevels of Cu/ZnSOD activity, but was unrelated to effects ofzinc deficiency on transpiration, rates of ethylene formation,ascorbic acid content or levels of MnSOD and ascorbatedependentperoxidase activities. Thus, these results show that detoxificationof superoxide anions by Cu/ZnSOD is important in plant resistanceto ozone. Ozone stimulates apoplastic antioxidant systems in pumpkin leaves   总被引:5,自引:0,他引:5  
The phytotoxiticky of ozone is due to its high oxidant capacity and to its ability to generate toxic molecular species. It is well known that intracellular peroxidases play an important role in eliminating toxic forms of oxygen but little evidence has been reported on the role of peroxidases in the apoplastic compartment. The detoxification systems located in the foliar extracellular matrix and in the intracellular fluid of sensitive pumpkin plants ( Cucurbita pepo L. cv. Ambassador) exposed to ozone (150 ppb. 5 days. 5 h day-1) in a fumigation chamber, were analyzed. The analyses were carried out on both young and mature leaves. Ascorbate peroxidase (EC 1.11.1.11) was found in the extracellular matrix of the pumpkin tissues. Its activity increased in both young and mature leaves as a consequence of the treatment, while at intraeellular levels its effect was most prominent in mature leaves. Analysis of the ascorbie-dehydroascorbic acid system revealed an enhancement of the pool content in the extracellular matrix of both kinds of leaves as a consequence of fumigation, while at the intracelluiar level small variations were found. Very little variation was observed in the glutathione pool as a consequence of fumigation. The analysis of a lipid peroxidation marker, malondi-aldehyde. showed the significant effect of ozone on membrane lipids. Following fumigation, the free phenols in the extracellular matrix decreased in both young and mature leaves, while the free and glycoside-bound phenols of the intracellular fluid showed little increase. The results support the hypothesis that ozone stimulates the an-tioxidant systems mainly in the apoplast and that ascorbic peroxidase activity, ascorbic acid levels and cell wall stiffening are the most influenced parameters.  相似文献

Changes in the activities of peroxidase, ascorbate peroxidase, catalase and superoxide dismutase in rice in response to infection by Rhizoctonia solani were studied. A significant increase in peroxidase activity was observed in R. solani-inoculated rice leaf sheaths 1 day after inoculation and the maximum enzyme activity was recorded 3 days after inoculation at which period a 3-fold increase in peroxidase activity was observed compared to the untreated control. Three peroxidase isozymes viz., PO-4, PO-5 and PO-6 were induced in rice upon infection by R. solani. Ascorbate peroxidase and catalase activities significantly increased 1–2 days after inoculation and the maximum enzyme activities were recorded 5 days after inoculation. Superoxide dismutase activity increased significantly 2 days after inoculation and increased progressively, reaching four times the control value at 7 days after inoculation.  相似文献

The apoplastic fluid of pine ( Pinus pinaster Aiton) hypocotyls contains ascorbic acid (AA) and dehydroascorbic acid (DHA). The amounts of ascorbic and dehydroascorbic acids were in the nmol (g fresh weight)−1 range and decreased with the hypocotyl age as well as along the hypocotyl axis. The ratio AA/(AA+DHA) also decreased with the hypocotyl age and along the hypocotyl. Both ascorbic oxidase and peroxidase activity against ascorbic acid showed very low activity not only in the apoplastic fluid but also in the fractions ionically and covalently bound to the cell walls. However, the peroxidase activity in the three abovementioned fractions was strongly increased in the presence of ferulic acid. That stimulation effect increased with the hypocotyl age and from the apical towards the basal region of the hypocotyls of 10-day-old seedlings. Furthermore, the oxidation of ferulic acid by apoplastic and ionically- and covalently-bound peroxidases was inhibited by ascorbic acid as long as ascorbate was available. A regulatory role of apoplastic ascorbic acid levels in the formation of dehydrodiferulic bridges between wall polysaccharides catalysed by cell wall peroxidases and thus in the cell wall stiffening during plant growth is proposed.  相似文献

Ozone is an air pollutant that damages a variety of biomolecules. We investigated ozone-induced inactivation of three major antioxidant enzymes. Cu/Zn superoxide dismutase was inactivated by ozone in a concentration-dependent manner. The concentration of ozone for 50% inactivation was approximately 45 microM when 10 microM Cu/Zn superoxide dismutase was incubated for 30 min in the presence of ozone. SDS-polyacrylamide gel electrophoresis (PAGE) showed that the enzyme was randomly fragmented. Both ascorbate and glutathione were very effective in protecting Cu/Zn superoxide dismutase from ozone-induced inactivation. The other two enzymes, catalase and glutathione peroxidase, were much more resistant to ozone than Cu/Zn superoxide dismutase. The ozone concentrations for 50% inactivation of 10 microM catalase and glutathione peroxidase were 500 and 240 microM, respectively. SDS-PAGE demonstrated that ozone caused formation of high molecular weight aggregates in catalase and dimerization in glutathione peroxidase. Glutathione protected catalase and glutathione peroxidase from ozone but the effective concentrations were much higher than that for Cu/Zn superoxide dismutase. Ascorbate was almost ineffective. The result suggests that, among the three antioxidant enzymes, Cu/Zn superoxide dismutase is a major target for ozone-induced inactivation and both glutathione and ascorbate are very effective in protecting the enzyme from ozone.  相似文献

The current study confirmed earlier conclusions regarding differential ozone (O3) tolerances of two soybean cultivars, Essex and Forrest, and evaluated antioxidant enzyme activities of these two varieties based on their performance under environmentally relevant, elevated O3 conditions. The experiment was conducted in open-top chambers in the field during the 1994 and 1995 growing seasons. Exposure of plants to moderately high O3 levels (62.9 nl l−1 air, 2-year seasonal average) caused chlorophyll loss and increased membrane permeability when compared to control plants grown in charcoal filtered air (24.2 nl l−1 air). The other effects of O3 treatment were decrease in seed yield, loss of total sulfhydryl groups, reduction of soluble protein content, and increase in guaiacol peroxidase activity in leaves of both cultivars. The O3-induced increase in guaiacol peroxidase activity was much smaller in cv. Essex leaflets. Cv. Essex had less leaf oxidative damage and smaller reduction in seed yield than cv. Forrest under elevated O3 conditions. During ozonation, mature leaflets of the more O3 tolerant cv. Essex had higher levels of glutathione reductase (30%), ascorbate peroxidase (13%), and superoxide dismutase (45%) activity than did mature leaflets of cv. Forrest. Cu,Zn-superoxide dismutase, which represented 95% of total superoxide dismutase activity in the two cultivars, appeared to be increased by O3 exposure in the leaflets of O3 tolerant cv. Essex but not in those of cv. Forrest. Cytosolic ascorbate peroxidase activity was also higher in leaflets of cv. Essex than in cv. Forrest regardless of O3 level. Stromal ascorbate peroxidase and Mn-superoxide dismutase activity did not appear to be involved in the O3 tolerance of the two soybean cultivars. This study was carried out to better understand the role of salicylic acid (SA) applied before cold stress in the cold tolerance mechanism. Two barley (Hordeum vulgare) cultivars, cold-sensitive (Akhisar) and cold-tolerant (Tokak), were used and 0.1 mM SA was applied to 7-d-old barley seedlings growing under control conditions (20/18 °C). The seedlings were transferred to cold chamber (7/5 °C) at the age 14, 21, and 28 d. After three days, the leaves were harvested to determine the activities of apoplastic antioxidant enzymes, such as superoxide dismutase (SOD), catalase (CAT), and peroxidase (POX) and ice nucleation activity and electrophoretic patterns of apoplastic proteins. Cold treatment decreased the activities of all enzymes in cold-sensitive cultivar, however, it increased CAT and POX activities in cold-tolerant cultivar. Exogenous SA increased enzyme activities in both cultivars. Ice nucleation activity increased by cold treatment, especially in 17-d-old seedlings in both cultivars. In addition, SA treatment increased ice nucleation activity in all examined samplings in both cultivars. SA treatment caused accumulation or de novo synthesis of some apoplastic proteins. The results of the present study show that exogenous SA can improve cold tolerance by regulating the activities of apoplastic antioxidative enzymes, ice nucleation activity, and the patterns of apoplastic proteins.  相似文献

Lipid peroxidation and peroxide-scavenging in soybean seeds during aging   总被引:8,自引:1,他引:7  
The possible role of lipid peroxidation in seed deterioration was investigated during natural aging and accelerated aging of seeds of edible soybean ( Glycine max [L], Merr. cv. Kaohsiung Selection No. 1). Natural aging was achieved by sealing the seeds in aluminum foil bags coated with polyethylene and storing the seeds at room temperature for 3 to 12 months. Accelerated aging was obtained by incubating the seeds at 45°C and close to 100% relative humidity for 3 to 12 days, after which the seeds were air dried to their original moisture level (8%). The results indicate that both natural and accelerated aging enhanced lipid peroxidation, as germination was depressed. Aging also inhibited the activity of superoxide dismutase, catalase, ascorbate peroxidase and peroxidase. The changes in germination and physiological activities, expressed as a function of aging duration, were somewhat similar in the two aging treatments.  相似文献

Peaches (Prunus persica cv. Hermoza) were stored at 0C in regular air (RA) or in controlled atmosphere (CA 10% CO2, 3% O2) for 4 weeks and then ripened for 4 days at 20 degrees C. Woolliness developed in the regular air stored fruit while the controlled atmosphere stored fruit ripened normally. In the woolly fruit symptoms of the disorder were greater in the inner mesocarp than in the outer. Polygalacturonase (PG) and pectin esterase (PE) activities differed in the outer and inner mesocarp of the affected fruit. PG activity was low and PE activity was high in the inner mesocarp of the woolly fruit during ripening relative to the outer mesocarp, while in the healthy fruit, activities were similar in both areas. Cell wall fractions of water-soluble, CDTA-soluble and carbonate-soluble pectins were prepared from freshly harvested peaches and incubated with PE and PG from ripe peaches at different ratios. Only the CDTA-soluble fraction formed a gel with peach enzymes, and the rate of gelation increased with increasing amounts of PE relative to PG. Both water-soluble and CDTA-soluble pectin fractions formed gels with commercial PE (extracted from orange peel). The PE extracted from peaches was stable when stored at 0 degrees C for 9 days, while PG activity was stable only for 1 day. We suggest that PE, acting on pectins in the cell wall in vivo may cause gel formation and that the CDTA-soluble polymers have the capacity to bind apoplastic water and create the dry appearance observed in woolly fruit.  相似文献

Changes in apoplastic peroxidases during auxin-induced in vitro rooting of cultured grapevine (Vitis vinifera L. cv. Touriga) stems have been studied. The largest increase in peroxidase activity (EC 1.11.1.7) was associated with the early stages of root initiation and could be attributed to an increase in activity of an apoplastic 36 kDa cationic peroxidase (PxB2). Relative to other peroxidases, PxB2 demonstrated high indole-3-acetic acid (IAA) oxidase activity and apparently contributed the majority of potential IAA oxidase activity in rooting tissues. The distribution of this peroxidase in developing roots additionally associates it with early phases of growth restriction. PxB2 was purified from cell wall extracts prepared from the basal 1 cm of rooting stems. Microsequencing and subsequent cloning of its corresponding 3' truncated cDNA (encoding 255 amino acids of the mature protein) revealed it to have a typical class III peroxidase structure. The results suggest that this class III peroxidase with IAA oxidase activity is important for the control of IAA levels during root initiation and development.  相似文献
