There is a strong association between metabolic syndrome (MS) and increased cardiovascular risk. Moreover, elevated nighttime blood pressure (BP) and non-dipping (subjects with <10% decline in the asleep relative to the awake BP mean) have been also linked to increased cardiovascular morbidity and mortality. We investigated the relation between MS, circadian time of hypertension treatment, and impaired nighttime BP decline in a cross-sectional study on 3352 (1576 men/1776 women) non-diabetic hypertensive subjects, 53.7?±?13.1 (mean?±?SD) yrs of age. Among them, 2056 were ingesting all their prescribed hypertension medication upon awakening, and 1296 were ingesting at least one of their BP medications at bedtime. BP was measured by ambulatory monitoring for 48 consecutive hours to substantiate reproducibility of the dipping pattern. Physical activity was simultaneously monitored every minute by wrist actigraphy to accurately calculate mean BP when awake and asleep for each subject. MS was present in 52.6% of the subjects. The prevalence of an altered non-dipper BP profile was significantly higher among subjects with MS (52.0% vs. 39.5% in subjects without MS, p < .001). Non-dipping was significantly more prevalent among subjects ingesting all BP-lowering medications upon awakening (56.8%) than among those ingesting at least one of their BP medications at bedtime (29.1%; p < .001). Subjects with MS had significantly higher values of uric acid (6.0 vs. 5.3?mg/dL, p < .001), plasma fibrinogen (331 vs. 315?mg/dL, p < .001), and erythrocyte sedimentation rate (14.8 vs. 12.4?mm, p < .001). Non-dipping was significantly associated with the presence of MS and treatment upon awakening in a multiple logistic regression model adjusted by significant confounding factors, including age, creatinine, erythrocyte sedimentation rate, and cigarette smoking. This cross-sectional study documents a significant increase of a blunted sleep-time BP decline in treated hypertensive subjects with MS. ABSTRACT

As the population ages the number of older adults living with hypertension (HTN) is rising dramatically. Uncontrolled HTN increases cardiovascular and renal mortality. Ambulatory (A) blood pressure (BP) is a better predictor of HTN-related morbidity and mortality than office BP. Lower BP is the most important therapeutic goal in treating HTN. Any reduction in BP has significant benefits for older adults. The current study was designed to evaluate the impact of the presence of pet dogs and cats on ABP during the daily lives of independently living, older pet owners with pre- to mild HTN. A repeated measures observational study of ABP of 32 pet owners (21 dogs, 8 cats, 3 cat and dog; 29 women) aged 50–83 years with BP 120–150/80–100 mmHg or < 150/100 with anti-hypertensive medication was conducted. Owner's ABPs were recorded every 20 minutes for one day during waking hours at study entry, one month, and three months. Activity monitors and diaries were used to obtain information about activity, mood, and whether the pet and/or another person was present in the room (indoors) or in close proximity (outdoors) with the owner at each assessment. Generalized estimating equation (GEE) analyses for hierarchical data (unstructured correlations) were performed for systolic and diastolic ABP. Mean ABPs were significantly (systolic BP/diastolic BP: dog p = 0.008/ p = 0.002; cat p < 0.009/ p < 0.001) different (systolic BP/diastolic BP mmHg: dog 3.1/1.5; cat –3.0/2.2) when pets were present after controlling for participant's mood (p > 0.05/ p < 0.001), activity intensity (p = 0.026/ p = 0.441), location (p = 0.013/ p = 0.004), and the presence of other people (p = 0.947/ p = 0.723). The presence of a dog was associated with lower systolic and diastolic BP and of a cat was associated with lower diastolic BP and higher systolic BP during their owners' normal daily lives. This finding suggests that pets, especially dogs, may be effective as an adjunctive intervention to slow the development or progression of HTN in older adults. Comparison of ABPs of pet owners with non-owners during their daily lives is warranted and underway.  相似文献

The Swedenborg whale Balaena swedenborgii Liljeborg, 1867, is a baleen whale species believed to have existed in the North Sea from the period when the inland ice melted around 13,000 before present (BP) until about 8000 years ago. The first bones attributed to this species were found in Sweden in 1705. When whale remains were discovered on the Swedish west coast during the extension work of a motorway extension, it was speculated that this could be a specimen of the extinct Swedenborg whale. The bones were found 72 m above the present-day sea level embedded in glacial mud. Shelly remains of marine organisms were present in the deposit surrounding the whale-fall, and sediments with the associated specimens were therefore collected for further analyses. We applied radiocarbon dating, thin sectioning, morphological analyses, ancient DNA typing and analyses of the associated shelly assemblage in an interdisciplinary effort to understand the circumstances of this fossil whale-fall. Our results show that the whale is not the putative species B. swedenborgii, but a bowhead whale Balaena mysticetus. The results also indicate that the whale must have been rapidly covered by glacial sediments, highlighting the speed of the deglacial process in the area.  相似文献

The late Quaternary vegetation communities of the south‐central highlands of Victoria are constructed from analyses of pollen and charcoal, and macroscopic plant remains preserved in Sphagnum bogs. The sites, located in eucalypt forest or woodland, form an altitudinal sequence with the component Eucalyptus species varying with altitude and with small pockets of Nothofagus cunninghamii (Hook.) Oerst. in close proximity to the higher sites. The record from the sites above 900 m covers the last 32 000 years, and the record from the lower sites extends from at least 12 000 BP . Around 32 000 BP the region was predominantly covered by a mosaic of alpine feldmark and herbfield, with small patches of Eucalyptus and Nothofagus woodland close to sea level when summer temperatures were probably 5°C lower than present. Lowest values, probably 7°–8°C below present, occurred between 19 800 and 16 900 BP , when alpine communities were most widespread and much of the Central Highlands was treeless. Around 12 000 BP alpine taxa disappeared or were greatly reduced, first at the lower sites. There was an associated rise in the treeline with the movement upslope of Nothofagus and eucalypt forest as a result of a general increase in temperature and probably effective precipitation. By 6000 BP wet eucalypt forest and Nothofagus reached their maximum postglacial extent at all sites, possibly related to a further increase in temperature, at least 2°C lower than present, and higher effective precipitation. A continuing increase in temperature, or an increase in continentality, and a decrease in effective precipitation led to increased fire hazard and retraction of rainforest and wet sclerophyll or tall open forest toward present‐day values. Nothofagus disappeared from the sites below 900 m. The activities of humans pose further threats to remaining forest communities. The record of vegetation and environmental change derived from the local and regional picture from eight sites reinforces and complements that from the individual sites. For example, combining the records overcomes to some extent taphonomic problems such as the effect of streams that flow close to all sites, and other limitations including problems of dating, poor preservation and variable sedimentation rates.  相似文献

Abstract The late Quaternary vegetation communities of the south-central highlands of Victoria are reconstructed from analyses of pollen and charcoal, and associated environmental conditions derived from the record of Nothofagus cunninghamii and alpine and sclerophyll taxa preserved in four subalpine Sphagnum bogs. The highest site occurs amid Eucalyptus paudflora woodland, the two intermediate sites are surrounded by Eucalyptus delegatensis forest and the lowest by a mixed forest of E. delegatensis/Eucalyptus regnans. Small pockets of N. cunninghamii occur within the eucalypt forests, and in close proximity to all four sites. Around 32 000 BP the vegetation consisted of a mosaic of alpine feldmark and herbfield, with small scattered groves of Nothofagus and Eucalyptus well below 1100m. Summer temperatures were probably 5°C lower than present with lowest values, probably 7° to 8°C below present, possibly between 17 000 and 13 500 BP, at which time alpine communities reached their greatest extent and much of the Central Highlands was treeless. After ca 13 500 BP herbaceous alpine taxa disappeared and there was an associated movement upslope of Nothofagus and tall open forest taxa to their maximum post-glacial extent, as temperatures and effective precipitation increased, ca 6000 BP. The retraction of cool temperate rainforest and wet sclerophyll or tall open forest towards present day values indicates lower effective precipitation, generally rising temperatures and increased fire hazard. More recently, European activities have increased the stress on the remaining forests. The study of four sites has demonstrated die importance of analysing a number of sites within a given area in order to overcome the limitations imposed by sites which were sub-optimal due to one or more factors including poor preservation, problems of dating, variable sedimentation rates, and the influence of streams which flow close to all sites. While die local environment varies between sites, and some vegetation changes are successional, this study shows that the local records complement one another, to some extent reinforcing die regional picture of vegetation and environmental change.  相似文献

Evidence from rodents and association analyses in humans suggest the presence on chromosome Y of one or more genes affecting blood pressure (BP). The HindIII centromeric alphoid polymorphism has been reported to be associated with BP in three independent human populations, although other studies reported null associations with this trait. Our objective was to test for association between BP and genetic variation of the Y chromosome. To this end, 2,743 unrelated Caucasian men recruited from nine UK practices were analysed for five SNPs (including the HindIII site) and two microsatellites spanning the non-recombining region of the Y chromosome. Systolic and diastolic BP were analysed both as quantitative traits and as categorical variables. Differences between locations were tested. Haplotypic and linkage disequilibrium (LD) analyses were also performed. Overall, no significant association was found between any of the loci analysed and BP, although post hoc analyses suggest a possible relation of specific Y haplogroups to BP. The HindIII polymorphism marks major structural differences in the Y centromere which could infuence mitotic loss during ageing, or other somatic events. Wetlands provide the ecosystem services of enhancing water quality, attenuating floods, sequestrating carbon and supporting biodiversity. In southern Africa, the pattern and intensity of land use is influenced by whether land tenure is public (state), private (individual ownership), or communal (shared agricultural and grazing resources). The influence of land tenure and its associated use on service provision was compared for communal tenure (grazing, maize production), wildlife conservation, and commercial agriculture (grazing, planted pastures) in the southern Drakensberg. Ordination analyses revealed that oxbow marshes, hill slope seepages and hygrophilous grasslands, the main hydro-geomorphic units, supported distinct plant communities that differed in their response to land use because of wetness or slope. Oxbows, uncultivated because of wetness, were inherently species poor with few exotics. Composition of hill slope seepages, uncultivated because of saturated slopes, varied among tenure types most likely in relation to grazing pressure. Seepages were threatened by the exotic invasive Rubus cuneifolius. Eighty-five percent of hygrophilous grassland had been cultivated by 1953, most of which was subsequently abandoned to secondary grassland. Primary hygrophilous grassland and hill slope seepages were the main repository for indigenous plant diversity, while communal maize fields supported a diverse mixture of mainly exotic species. Soil carbon concentrations decreased from oxbows to pastures, seepages, primary hygrophilous grassland, secondary grassland, and maize on former grassland (7.0, 4.1, 4.0, 3.5, 2.4, and 1.7%, respectively). The pattern for total soil nitrogen and sulphur were the same. Cultivation of hygrophilous grassland was estimated to have reduced soil carbon stocks to 69% of pre-settlement levels by 1953 (∼150 years BP). Stocks then increased by 8% to 2001 following crop abandonment. Cultivation has impaired water quality enhancement and flood attenuation because of greater amounts of bare ground and shorter vegetation. Further improvement of ecosystem services will depend on the influence of socio-economic factors on communal cropping.  相似文献

Appropriate management of contemporary environments requires knowledge of their long-term history. We use palaeoecological data to explore how contemporary forest-steppe environments have been shaped by climate change and human impacts through the Holocene using the western Mid-Russian Upland as a case-study. Our paper presents new reconstructions of Mid- and Late Holocene climate, vegetation dynamics and local environmental change based on pollen, plant macrofossil and testate amoeba records from a site at Selikhovo (Mid-Russian Upland, Russia). Eutrophic fen vegetation dominated by Phragmites australis developed around 6800 cal year BP and has been resilient to episodes of local burning and variable input of mineral material through the Holocene. New and previously-published data show that the boundary between broadleaf forest and steppe occupied a similar position to present during the period 7000–4800 cal year BP, despite a warmer and drier climate, but shifted to the south following climate cooling and an increase in precipitation from 4800–2500 cal year BP. A subsequent decline in woodland cover was caused by both climate change and human impacts, with human activity becoming increasingly significant over the last two millennia. Prior to major human disturbance (about 1700 cal year BP) the landscape was dominated by mixed broadleaf-pine forests with some spruce covering about 60 % of the study area. Our results emphasize the variability of steppe-forest habitats over long time periods and the need to consider human impacts and climate change when setting targets for habitat conservation.  相似文献

Questions

As the dominant tree in many European forests, Fagus sylvatica functions as an ecosystem engineer, yet its istory remains little understood. Here we ask: (a) are there indications for its presence in southeast France during the last Glacial period; (b) what was the timing of the expansion and decline of F. sylvatica dominated forests; (c) which factors influenced their dynamics and in particular to what extent did past precipitation changes impact upon them; and (d) at which altitudes did these beech forests occur within the region?

Location

Languedoc, the French Mediterranean area.

Method

This article presents a well dated and high‐resolution pollen sequence covering the last 7,800 years from the Palavas Lagoon in the Languedoc together with a review of Fagus charcoal occurrences in the Languedoc and the lower Rhône Valley, and a review of pollen data from a compilation of 69 sites in southeast France.

Results

The Palavas pollen sequence provides a regional summary of F. sylvatica abundance changes near the Mediterranean coast. Around 6,000 years cal BP , an abrupt transition from small beech populations to well‐developed forests is recorded. The maximum development of beech forests occurred between 4,000 and 3,000 years cal BP , while F. sylvatica started to regress after 3,000 years cal BP .

Conclusion

Scattered F. sylvatica populations probably survived throughout southern France during the last Glacial period. F. sylvatica started to spread around 8,000 years cal BP while beech forests never expanded before 6,000 years cal BP . The complex patterns of F. sylvatica expansion in southern France after 6,000 years cal BP suggests that a combination of global (climate change) and local (human impact) factors were responsible for this major change. Recurrent abrupt climate changes, the aridity trend and human deforestation caused beech forests to decline after 3,000 years cal BP .

Abstract— A new CNS myelin autoantigen(s) (referred to as M2), different from the encephalitogenic basic protein (BP), can be detected with guinea-pig demyelinating and complement fixing (CF) sera raised against guinea pig CNS tissue or myelin (Lebar et al., 1976). M2 and BP were present in mouse, rat, rabbit, bovine and human CNS tissues when tested with guinea-pig homologous specific antisera; they were not present in non-CNS tissues. Both autoantigens were also detected in newborn guinea-pig myelin and myelin-like fractions. The CF activity of myelin with demyelinating (anti-M2) sera was not altered by trypsin; however, absorption experiments showed that M2 was partly trypsin sensitive. Both antibodies against the trypsin sensitive and the trypsin resistant determinants of M2 were demyelinating. Both determinants of M2 were preselit in mouse, rat, rabbit, bovine‘and human CNS tissues and in guinea-pig newborn myelin. CF BP activity of myelin was partially or even totally abolished by trypsin, but the persistent encephali-togenicity of trypsin-treated myelin could be attributed to non-CF encephalitogenic peptides from BP. In accordance with recent work our results tend to support an inner localization of BP in myelin; M2, on the other hand, would be a surface antigen(s).  相似文献

Data on hundreds or thousands of single nucleotide polymorphisms (SNPs) provide detailed information about the relationships between individuals, but currently few tools can turn this information into a multigenerational pedigree. I present the r package sequoia , which assigns parents, clusters half‐siblings sharing an unsampled parent and assigns grandparents to half‐sibships. Assignments are made after consideration of the likelihoods of all possible first‐, second‐ and third‐degree relationships between the focal individuals, as well as the traditional alternative of being unrelated. This careful exploration of the local likelihood surface is implemented in a fast, heuristic hill‐climbing algorithm. Distinction between the various categories of second‐degree relatives is possible when likelihoods are calculated conditional on at least one parent of each focal individual. Performance was tested on simulated data sets with realistic genotyping error rate and missingness, based on three different large pedigrees (= 1000–2000). This included a complex pedigree with overlapping generations, occasional close inbreeding and some unknown birth years. Parentage assignment was highly accurate down to about 100 independent SNPs (error rate <0.1%) and fast (<1 min) as most pairs can be excluded from being parent–offspring based on opposite homozygosity. For full pedigree reconstruction, 40% of parents were assumed nongenotyped. Reconstruction resulted in low error rates (<0.3%), high assignment rates (>99%) in limited computation time (typically <1 h) when at least 200 independent SNPs were used. In three empirical data sets, relatedness estimated from the inferred pedigree was strongly correlated to genomic relatedness.  相似文献

Blood pressure (BP) is largely determined by quantitative trait loci (QTLs) in Dahl salt-sensitive (DSS) rats. Little is known about QTLs controlling kidney (K), cardiac (C), and aortic (A) mass (i.e. Km, Cm, and Am, respectively) of DSS rats independent of BP. Their identification can facilitate our understanding of end organ damage. In this work, 36 congenic strains were employed to define QTLs for Km, Cm, and Am either independent of or associated with BP. Five new QTLs, i.e., KmQTLs, that influence Km independent of Cm, Am, and BP were defined. Four new CakmQTLs were defined for Cm, Am, and Km independent of BP. Among them, the CakmC10QTL1 interval contained 13 genes and undefined loci, and none was known to influence the phenotypes in question, paving the way for a novel gene discovery. Among 17 individual QTLs for BP, 14 also affected Cm, Km, and Am, i.e., they are BpcakmQTLs. In contrast, one BpQTL had no effect on Cm, Am, and Kam. Therefore, BP and Cm, Am, and Km have distinct and shared genetic determinants. The discovery of individual Km and Cakm QTLs will likely facilitate the identification of mechanisms underlying renal, cardiac, and/or aortic hypertrophy independent of hypertension. The Late Pleistocene and Holocene history of five lakes in the central part of Karelian Isthmus, south of the present Vuoksi River, are described on the basis of sediment stratigraphical investigations. Two of the Lakes, Michurinskoe (94 m a.s.l.) and Uzornoe (55 m a.s.l.) are situated in an upland area that remained dry land after the deglaciation even during the early high water stages of the Baltic Sea (Baltic Ice Lake until c. 10 000 yr BP and Ancylus Lake 9500–8800 BP). The low-lying central parts of the Isthmus were flooded by the outflow of Lake Ladoga that took place across this area until the formation of Neva River, c. 3 100 yr BP, and further by the waters of River Vuoksi that started flowing into the area from the NW c. 5000 yr BP as a new outlet of Lake Saimaa. The basins of the lakes Krasnoe and Vishnevskoe (both 16 m a.s.l.) became isolated when River Neva was formed and Lake Ladoga sank to its present level. Lake Rakovoe (12 m a.s.l.) was on the level of River Vuoksi up until the mid-19th century, when water level in the central stretch of the river was artificially lowered. Each of the lakes has been variously affected by hydrological and climatic changes and consequences of human activities, e.g. eutrophication due to intensified land use. As a consequence to artificial lowering, done in order to gain field and meadowland, the large, shallow Lake Rakovoe has been largely overgrown by macrophytic vegetation.  相似文献

Colonies of the ascomycete fungus Penicillium brevicompactum Dierckx produce bright yellow-green fluorescent exudate bubbles on its surface when grown on standard plant cell culture medium. According to SDS-PAGE analysis, the exudate is enriched in one protein, named bubble protein (BP). Detailed characteristics of BP are described, and also its corresponding genomic promoter and terminator sequences that flank sequences encoding signal peptide and a precursor sequence upstream of that of the mature protein. Following on previous work, the protein is now biochemically characterized. BP, the structure of which mainly consists of beta sheets, has four very stable disulfide bridges that resist standard procedures for reduction. With such traits, BP can now be categorized as a new member of the ever growing class of defensins. Indeed, the protein revealed anti-fungal effects as it inhibits growth of the yeast Saccharomyces cerevisiae in a dose-dependent manner. Structural classification places BP into the group of proteins with a knottin fold, founding the BP superfamily. Based on genomic alignments that revealed very high homology to four proteins of related fungi, a 3D structure prediction of the corresponding proteins was made. In addition, it was discovered that the closely related fungus Penicillium chrysogenum encodes a BP homolog – in addition to its PAF protein, which also is similar to BP – further suggesting that fungi may possess more than one defensin.  相似文献

Aim To reconstruct the history of vegetation and environments using pollen, charcoal and sediment analysis, and to identify the timing and nature of climate change and human impact on the vegetation of a remote Pacific island. Location Cerro de Los Inocentes, 1000 m above sea level, Alexander Selkirk Island (33°45′S, 80°45′W), Chile. The westernmost island of the Juan Fernandez Archipelago, south‐east Pacific Ocean. Methods A 150‐cm long sediment core comprising 87 cm dark brown peat overlying 63 cm of yellow grey clay was extracted from a shallow depression on the southern slopes of Cerro de Los Inocentes. Pollen, charcoal, sediment and accelerator mass spectrometry radiocarbon analyses were used to construct a record of vegetation change through time. Numerical analysis of multispecies data allowed the classification of fossil assemblages into distinct pollen zones. Results Pollen and spores are preserved throughout the sediment with high concentrations coinciding with the beginning of organic sediment accumulation at around 8000 14C yr BP. Prior to 8000 14C yr BP, the deposition of clays, presumably from upslope erosion, occurred in a landscape sparsely vegetated by grasses, ferns and Pernettya rigida heath, including several plants that are only found 100–200 m above the site today (Zone CI‐1). After 8000 14C yr BP, a P. rigida heath was the dominant vegetation (Zone CI‐2). A shift to a wet heath–shrubland (Zone CI‐3) occurred at 6000 14C yr BP and was followed by a transition to a treefern–shrubland mosaic accompanied by periodic burning (Zone CI‐4) after 4500 14C yr BP. The impact of human occupation is evident in Zone CI‐5 at 450 14C yr BP with the loss of forest species, increased burning and invasion of the exotic plant Rumex. Main conclusions The pollen and charcoal record provides the first evidence of vegetation changes spanning at least the last 8000 14C yr BP from the high altitude environment of Alexander Selkirk Island. Prior to 8000 14C yr BP, the altitudinal ranges of different plant species may have been suppressed by a cooler and drier climate. Increasing precipitation and temperatures at the end of the last glacial period may have mobilized exposed sediments in a sparsely vegetated upland environment, altering local drainage patterns, eventually leading to slope stabilization and deposition of organic detritus under an increasing density of heath and shrub vegetation. The subalpine heath–shrubland persisted until 4500 14C yr BP when first evidence for sustained burning is found in association with the establishment of a more open treefern–shrubland vegetation pattern. In the absence of human occupation at this time, the influence of increased climatic variability associated with more frequent El Niño‐Southern Oscillation events during the mid to late Holocene is considered one of the main driving forces behind increased vegetation disturbance during this period. The record provides evidence that island vegetation patterns have been highly dynamic over millennial to decadal time‐scales and that the flora has persisted through periods of rapid and major climate change. This changed with the discovery of the island by European explorers in the late sixteenth century and the subsequent introduction of goats and exploitation and burning of forests, which resulted in the progressive destruction of native vegetation and the invasion of introduced plants. There is evidence that reduced burning and control of the goat population within the last 50 years has resulted in marginal recovery of some high altitude native plant species.  相似文献

Moreover, elevated nighttime blood pressure (BP) and non-dipping (subjects with <10% decline in the asleep relative to the awake BP mean) have been also linked to increased cardiovascular morbidity and mortality. We investigated the relation between MS, circadian time of hypertension treatment, and impaired nighttime BP decline in a cross-sectional study on 3352 (1576 men/1776 women) non-diabetic hypertensive subjects, 53.7 ± 13.1 (mean ± SD) yrs of age. Among them, 2056 were ingesting all their prescribed hypertension medication upon awakening, and 1296 were ingesting at least one of their BP medications at bedtime. BP was measured by ambulatory monitoring for 48 consecutive hours to substantiate reproducibility of the dipping pattern. Physical activity was simultaneously monitored every minute by wrist actigraphy to accurately calculate mean BP when awake and asleep for each subject. MS was present in 52.6% of the subjects. The prevalence of an altered non-dipper BP profile was significantly higher among subjects with MS (52.0% vs. 39.5% in subjects without MS, p < .001). Non-dipping was significantly more prevalent among subjects ingesting all BP-lowering medications upon awakening (56.8%) than among those ingesting at least one of their BP medications at bedtime (29.1%; p < .001). Subjects with MS had significantly higher values of uric acid (6.0 vs. 5.3 g/dL, p < .001), plasma fibrinogen (331 vs. 315 mg/dL, p < .001), and erythrocyte sedimentation rate (14.8 vs. 12.4 mm, p < .001). Non-dipping was significantly associated with the presence of MS and treatment upon awakening in a multiple logistic regression model adjusted by significant confounding factors, including age, creatinine, erythrocyte sedimentation rate, and cigarette smoking. This cross-sectional study documents a significant increase of a blunted sleep-time BP decline in treated hypertensive subjects with MS. Even in the presence of MS, treatment at bedtime is significantly associated with lower prevalence of a high-risk non-dipper BP profile.  相似文献

Objective:

Circulating cortisol and psychosocial stress may contribute to the pathogenesis of obesity and metabolic syndrome (MS). To evaluate these relationships, a cross‐sectional study of 369 overweight and obese subjects and 60 healthy volunteers was performed and reviewed the previous literature.

Design and Methods:

Overweight and obese subjects had at least two other features of Cushing's syndrome. They underwent measurements representing cortisol dynamics (24 h urine cortisol excretion (UFC), bedtime salivary cortisol, 1 mg dexamethasone suppression test) and metabolic parameters (BMI, blood pressure (BP); fasting serum triglycerides, HDL, insulin, and glucose). Subjects also completed the Perceived Stress Scale (PSS). UFC, salivary cortisol, and weight from 60 healthy volunteers were analyzed.

Results:

No subject had Cushing's syndrome. UFC and dexamethasone responses were not associated with BMI or weight. However, salivary cortisol showed a trend to increase as BMI increased (P < 0.0001), and correlated with waist circumference (WC) in men (rs = 0.28, P = 0.02) and systolic BP in women (rs = 0.24, P = 0.0008). Post‐dexamethasone cortisol levels were weak to moderately correlated with fasting insulin (rs = ?0.31, P = 0.01) and HOMA‐IR (rs = ?0.31, P = 0.01) in men and systolic (rs = 0.18, P = 0.02) and diastolic BP (rs = 0.20, P = 0.009) in women. PSS results were higher in obese subjects than controls, but were not associated with cortisol or metabolic parameters. As expected, WC correlated with fasting insulin, HOMA‐IR, and systolic BP (adjusted for BMI and gender; P < 0.01). Literature showed inconsistent relationships between cortisol and metabolic parameters.

Conclusion:

Taken together, these data do not support a strong relationship between systemic cortisol or stress and obesity or MS.
