The rice indica/japonica hybrid shows strong heterosis.However,such inter-subspecific hybrid can't be directly used in rice production due to its low spikelet fertility.The S5 locus was proved to be associated with fertility of indica/japonica hybrid and its S5n allele from wide-compatibility variety(WCV)is capable to overcome fertility barrier.In the present study,we reported the causal sites in the S5 locus responsible for compatibility of indica/japonica hybrid.Fine-mapping of the S5 locus using the 11 test-cross families pinpoints a candidate S5 locus encoding aspartic protease(Asp).Intragenic recombination within the Asp gene happened in a number of recombinants,resulting in chimeric S5j-S5n alleles.Just like S5n,the chimeric s5j-S5n allele displayed higher spikelet fertility when combined with the S5i allele.In the complementary test,however,the S5n allele from WCVs failed to enhance fertilities of the indica/japonica hybrids.Compared to both indica and japonica varieties.all nine WCVs from different resources are characterized with a 136 bp deletion in the Asp N-terminus.which probably renders the S5n allele non-functional.Furthermore,an A/C polymorphic site is detected 1,233 bp downstream of the Asp start codon.The heterozygous A/C site of the Asp gene in indica/japonica hybrid is believed to be the casual factorto causc partial sterility.The functional makers based on the two polymorphic sites will be broadly used in developing wide-compatibility rice varieties.  相似文献

The S5 locus regulates spikelet fertility of indica/japonica hybrid rice (Oryza sativa). There are three alleles at the S5 locus, including an indica allele (S5i), a japonica allele (S5j), and a wide-compatibility allele (S5n). This study analyzed the molecular basis for S5-dependent reproductive isolation and compatibility of indica/japonica rice hybrids. Three S5 alleles were expressed at extremely low levels, and only in the ovary. S5n was more similar to S5i in both RNA and protein expression profiles. The S5 locus was not essential for embryo sac development, although deleterious interactions between S5i and S5j resulted in reduced rates of spikelet fertility. The yeast two-hybrid system was used to test direct interactions between S5-encoded proteins. The results indicated that the S5i- and S5j-encoded eukaryotic aspartyl proteases formed both homodimers and heterodimers, whereas the S5n-encoded aspartyl protease was incapable of dimerization. Site-directed mutagenesis revealed that a single amino acid difference between S5i- and S5j-encoded aspartyl proteases (phenylalanine/leucine at residue 273) was primarily responsible for embryo sac abortion. The S5 locus may have promoted the subspeciation of indica and japonica, but it also enables gene flow between them.  相似文献

 The discovery of wide-compatibility varieties (WCVs) that are able to produce normal fertility hybrids when crossed both to indica and japonica rice has enabled the fertility barrier between indica and japonica subspecies to be broken and provided the possibility of developing inter-subspecific hybrids in rice breeding programs. However, a considerable variation in the fertility level of hybrids from the same WCV crossed to different varieties has often been observed. One hypothesis for this variable fertility is that additional genes are involved in hybrid fertility besides the wide-compatibility gene (WCG). To assess such a possibility, we performed a genome-wide analysis by assaying a large population from a three-way cross ‘02428’/‘Nanjing 11’//‘Balilla’ using a total of 171 RFLP probes detecting 191 polymorphic loci distributed throughout the entire rice linkage map. Our analysis recovered 3 loci conferring significant effects on hybrid fertility. The major locus on chromosome 6 coincided in chromosomal location with the previously identified S 5 locus, and the 2 minor loci that mapped to chromosomes 2 and 12, respectively, were apparently distinct from all previously reported hybrid sterility genes. Interaction between the indica and japonica alleles at each of the loci caused a reduction in hybrid fertility. The joint effect of the 2 minor loci could lead to partial sterility even in the presence of the WCG. The location of the S 5 locus on the molecular marker linkage map was determined to be approximately 1.0 cM from the RFLP locus R2349.  The partial sterility of hybrids between the indica and japonica rice subspecies of Asian cultivated rice is a serious constraint for utilizing inter-subspecific heterosis in hybrid rice breeding. In this study, we have investigated the relationship between molecular-marker polymorphism and indica-japonica hybrid fertility using a diallel set involving 20 rice accessions including 9 indica and 11 japonica varieties. Spikelet fertility of the resulting 190 F1s and their parents was examined in a replicated field trial. Intra-subspecific hybrids showed much higher spikelet fertility than inter-subspecific hybrids except in crosses involving wide-compatibility varieties. The parents were surveyed for DNA polymorphism using 96 RFLP and ten SSR markers, which revealed extensive genetic differentiation between indica and japonica varieties. A large number of markers detected highly significant effects on hybrid fertility. The chromosomal locations for many of the positive markers coincided well with previously identified loci for hybrid sterility. The correlation between hybrid fertility and parental distance was low in both intra- and inter-subspecific crosses. The results suggest that the genetic basis of indica-japonica hybrid sterility is complex. In intraspecific crosses between cultivated rice (Oryza sativa) subspecies indica and japonica, the hybrid male sterility gene S24 causes the selective abortion of male gametes carrying the japonica allele (S24-j) via an allelic interaction in the heterozygous hybrids. In this study, we first examined whether male sterility is due solely to the single locus S24. An analysis of near-isogenic lines (NIL-F(1)) showed different phenotypes for S24 in different genetic backgrounds. The S24 heterozygote with the japonica genetic background showed male semisterility, but no sterility was found in heterozygotes with the indica background. This result indicates that S24 is regulated epistatically. A QTL analysis of a BC(2)F(1) population revealed a novel sterility locus that interacts with S24 and is found on rice chromosome 2. The locus was named Epistatic Factor for S24 (EFS). Further genetic analyses revealed that S24 causes male sterility when in combination with the homozygous japonica EFS allele (efs-j). The results suggest that efs-j is a recessive sporophytic allele, while the indica allele (EFS-i) can dominantly counteract the pollen sterility caused by S24 heterozygosity. In summary, our results demonstrate that an additional epistatic locus is an essential element in the hybrid sterility caused by allelic interaction at a single locus in rice. This finding provides a significant contribution to our understanding of the complex molecular mechanisms underlying hybrid sterility and microsporogenesis.  相似文献

Hybrids between the indica and japonica subspecies of rice(Oryza sativa) are usually sterile, which hinders utilization of heterosis in the inter-subspecific hybrid breeding. The complex locus Sa comprises two adjacently located genes, SaF and SaM, which interact to cause abortion of pollen grains carrying the japonica allele in japonica-indica hybrids. Here we showed that silencing of SaF or SaM by RNA interference restored male fertility in indica-japonica hybrids with heterozygous Sa. We further used clustered regularly interspaced short palindromic repeats(CRISPR)/Cas9-based genome editing to knockout the SaF and SaM alleles, respectively, of an indica rice line to create hybrid-compatible lines. The resultant artificial neutral alleles did not affect pollen viability and other agricultural traits, but did break down the reproductive barrier in the hybrids. We found that some rice lines have natural neutral allele Sa-n, which was compatible with the typical japonica or indica Sa alleles in hybrids. Our results demonstrate that SaF and SaM are required for hybrid male sterility, but are not essential for pollen development. This study provides effective approaches for the generation of hybrid-compatible lines by knocking out the Sa locus or using the natural Sa-n allele to overcome hybrid male sterility in rice breeding.  相似文献

Hybrid sterility frequently occurs in crosses between indica and japonica subspecies of Asian cultivated rice. In this study, we investigated the cytological processes involved in formation and development of male and female gametes as well as their interactions in fertilization, using an indica/japonica hybrid in comparison with an indica/indica hybrid. It was found that more than 50% of the microspores generated in the indica/japonica hybrid could not develop into functional pollen. The abortion rate of microspores in the indica/japonica hybrid was much higher than that in the indica/indica hybrid. Abortive embryo sacs made up roughly 70% of the embryo sacs examined in the indica/japonica hybrid, which was also much higher than that detected in the indica/indica hybrid. Moreover, the amount of pollen adherence on stigmas of the indica variety upon hand-pollination with pollen from the japonica variety was much lower than the indica/indica pollination, and the number of pollen adhered on the stigmas by natural self-pollination was much greater in the indica/indica hybrid than in the indica/japonica hybrid. The indica/japonica hybrid also encountered difficulties in pollen tube growth after pollination, and the fertilization rate of the indica/japonica hybrid was much lower than that of the indica/indica hybrid. These results clearly illustrate the complexity of the mechanisms underlying inter-subspecific hybrid sterility in rice involving both pre- and post-zygotic reproductive isolation mechanisms.  相似文献

F(1) hybrid sterility and ``hybrid breakdown' of F(2) and later generations in rice (Oryza sativa L.) are common and genetically complicated. We used a restriction fragment length polymorphism linkage map and F(4) progeny testing to investigate hybrid sterility and hybrid breakdown in a cross between ``widely compatible' O. sativa ssp. japonica cultivar Lemont from the Southern U.S. and ssp. indica cultivar Teqing from China. Our results implicate different genetic mechanisms in hybrid sterility and hybrid breakdown, respectively. Hybrid sterility appeared to be due to recombination within a number of putative differentiated ``supergenes' in the rice genome, which may reflect cryptic structural rearrangements. The cytoplasmic genome had a large effect on fertility of both male and female gametes in the F(1) hybrids. There appeared to be a pair of complementary genes that behaved like ``wide compatibility' genes. This pair of genes and the ``gamete eliminator' (S(1)) or ``egg killer' (S-5) may influence the phenotypic effects of presumed supergenes in hybrids. Hybrid breakdown appeared to be largely due to incompatibilities between indica and japonica alleles at many unlinked epistatic loci in the genome. These proposed mechanisms may partly account for the complicated nature of postreproductive barriers in rice.  相似文献

Wide-compatibility varieties (WCVs), comprising a special class of rice germplasm, are able to produce fertile hybrids when crossed with both indica and japonica varieties. Dular, a landrace variety from India, has both a wide spectrum and high level of wide-compatibility when crossed with a range of indica and japonica varieties. In previous studies, an allele at the f5 locus from Dular (f5-Du) was identified as a neutral allele conferring wide-compatibility with a large effect on both pollen and spikelet fertility. Using a population of 1993 hybrid plants derived from a cross between ZS(f5-Du/f5-ZS) (F1 of near isogenic line of Zhenshan 97 containing f5-Du) and Balilla (a japonica tester), f5-Du was genetically mapped to an interval of about 1.6 cM, with 0.8 cM from both SSR markers WFPM3 and WFPR1. Combined with bioinformatic analysis, a contig map was constructed for the f5 region, consisting of five bacterial artificial chromosome (BAC) or P1 artificial chromosome (PAC) clones and spanning approximately 437 kb in length. By assaying the recombinant events in the region with markers developed using the sequence information, the f5 locus was further narrowed down to a 70 kb fragment containing nine predicted genes. The result will be very useful for cloning this gene and marker-assisted transferring of the neutral allele in rice breeding programs.  相似文献

采用田间试验研究了421个常规籼稻、63个常规粳稻和82个籼型杂交稻品种种子含氮量的基因型差异。结果表明,栽培稻种子含氮量变幅在0.85%~2.55%,平均为1.39%。种子平均含氮量最高的是籼型杂交稻,高达1.46%,显著高于常规粳稻(1.39%)和常规籼稻(1.37%)。不同类型的栽培稻种子含氮量呈正态分布,但频数分布最高的区间存在一定的差异。以50%品种的种子含氮量(累积百分数从25%~75%)为依据,常规籼稻、常规粳稻、籼型杂交稻分别集中在1.21%~1.52%、1.21%~1.51%、1.36%~1.53%。LSR测验表明,籼型杂交稻极差显著小于常规籼稻和常规粳稻,但常规粳稻与常规籼稻的差异未达显著水平,即籼型杂交稻种子含氮量比常规籼稻和常规粳稻的分布更加集中。不同类型栽培稻内的种子含氮量极差较大,可分为若干组,且各组间达极显著差异。  相似文献

水稻广亲和基因的利用是克服亚种间杂种不育性的重要途径。但在广亲和性的遗传上不同研究者的结论不尽一致。以3种有广亲和品种参加的三交组合为研究材料,研究了品种Ketan Nangka的广亲和性遗传。结果表明水稻亚种杂交F1同时存在着雄性不育和雌性不育,但雄性不育对小穗育性的作用大小因组合而异;无论是在雄性不育位点还是雄性不育位点上,Ketan Nangka均具有相对应的中性基因(广亲和基因);广亲和性的遗传特点与所用的籼粳测验品种间的杂种不育性密切相关;S-5位点的广亲和基因遗传符合单位点孢子体-配子体互作模型。  相似文献

利用一套籼粳交DH系与两份具有广亲和性的水稻光(温)敏核不育系测交,构建两个测交F1群体,同时用92个多型性的RFLP标记分析了杂交亲本和另外30个不同类型的籼、粳品种(对照组)的基因型。利用对照组籼或粳表型与分子标记的关联性,检测出41个与籼粳分化高度相关的RFLP标记。结果表明,87.8%的这种标记参与了对杂种优势显著或极显著的影响,表明在水稻的演化过程中,参与适应性进化的基因与控制经济产量的  相似文献

Hybrid sterility between indica and japonica subspecies in rice is basically caused by partial abortion of gametes and hybrid fertility could be recovered by a single wide compatibility (WC) allele. In this study, a typical indica germplasm source of rice, UPRI 95-162, with strong wide compatibility in cross with japonica rice was studied for location of its WC locus. Bulked segregant analysis was performed and SSRs (simple sequence repeats) were conducted on a F1 population derived from a three-way cross (UPRI 95-162/T8//Akihikari). The locus was located on chromosome 1 approximately 0.2 cM to SSR markers RM581 on one side and 1.5 cM to RM292 on another. This WC locus, tentatively designated as S-20 n (t), and its tight linkage markers, RM581 and RM292, would be very useful for efficient marker-assisted selection for breeding new WC varieties and for map-based cloning of the gene.  相似文献

Physical mapping of the 5S ribosomal RNA genes on rice chromosome 11   总被引:9,自引:0,他引:9  
One 5S ribosomal RNA gene (5S rDNA) locus was localized on chromosome 11 of japonica rice by in situ hybridization. The biotinylated DNA probe used was prepared by direct cloning and direct labeling methods, and the locus was localized to the proximal region of the short arm of chromosome 11 (llpl.l) by imaging methods. The distance between the signal site and the centromere is 4.0 arbitrary units, where the total length of the short arm is 43.3 units. The 5S rDNA locus physically identified and mapped in rice was designated as 5SRrn. The position of the 5S rDNA locus reported here differs from that in indica rice; possible reasons for this difference are discussed. DNA sequences of 5S rDNA are also reported.  相似文献

近年来,灰飞虱Laodelphax striatellus(Fall n)在江浙等地呈明显上升态势。采用不同温度及水稻类型处理灰飞虱若虫,获得灰飞虱种群在不同处理组合下的存活率、发育进度及抗寒力。结果表明,35℃的高温下,灰飞虱各龄若虫无论在杂交籼稻还是粳稻上,其发育进度均比25~30℃推迟2~7d。35℃高温下若虫死亡率较高,并且不能正常羽化。粳稻上饲养的灰飞虱若虫的存活率明显高于杂交籼稻,但水稻类型和温度对灰飞虱存活率的影响不存在明显的互作作用。在适宜温度下(27~30℃)用杂交籼稻饲养的灰飞虱,其过冷却点及结冰点均要比在高温(35℃)或低温(25℃)下饲养的低。在26~30.5℃变温条件下,杂交籼稻上饲养的灰飞虱的过冷却点和结冰点显著低于粳稻上饲养的,而且不同温度和水稻类型上的灰飞虱的过冷却点均低于零下10℃。可见江浙稻区冬季低温对灰飞虱的越冬已不存在制约作用。温度和水稻类型的变化是近年灰飞虱暴发成灾的重要原因。  相似文献

 Low-temperature-sensitive sterility (LTSS) has become one of the major obstacles in indica-japonica hybrid rice breeding. In this study, we determined, using RFLP markers, the genetic basis of LTSS in two populations derived from crosses between indica and japonica parents, the BC1F1 of 3037/02428//3037 and the F2 of 3037/02428. The fertility segregation in the two populations under low-temperature conditions was used as a measurement of the temperature sensitivity of the various genotypes in the populations. A RFLP survey of bulked extremes from the BC1F1 population identified three genomic regions, two on chromosome 1 and one on chromosome 12, that were likely to contain genes for LTSS (or Ste loci). One-way ANOVA and QTL analysis using a total of 19 markers from these three genomic regions resolved three Ste loci in the BC1F1 population and two Ste loci in the F2 population. On the basis of chromosomal location these loci were distinct from those governing wide-compatibility identified in previous studies. Two- and three-way ANOVA showed that these loci acted essentially independent of each other in conditioning LTSS. The main mode of gene action was an interaction between the indica and the japonica alleles within each locus. For each respective locus this resulted in a drastic fertility reduction in the heterozygote state relative to the homozygote state. Wide-compatibility varieties (WCVs) are a special class of rice germplasm that is able to produce fertile hybrids when crossed to both indica and japonica rice varieties. WCVs may differ greatly in their spectrum and level of compatibility. The objective of this study was to determine the genetic basis of wide-compatibility conferred by ‘Dular’, a landrace variety from India that has demonstrated a high level of wide-compatibility in previous studies with a broad range of indica and japonica varieties. A three-way cross (‘Balilla/Dular//Nanjing 11’) was made and the resulting F1 population evaluated in the field for spikelet fertility. A total of 235 plants from this population was assayed individually for restriction fragment length polymorphisms (RFLPs) at 159 marker loci covering the entire rice genome at regular intervals. Quantitative trait locus (QTL) analysis identified 5 loci, located on chromosomes 1, 3, 5, 6 and 8, as having significant effects on hybrid fertility, which jointly explained 55.5% of the fertility variation in this population. The QTL on chromosome 5 ( f5) showed the largest effect on hybrid fertility, followed by those on chromosomes 6 ( f6), 3 ( f3) and 1 ( f1), with the one on chromosome 8 ( f8) having the smallest effect. Genotypes each composed of an allele from ‘Dular’ and an allele from ‘Nanjing 11’ at four ( f3, f5, f6 and f8) of the five QTLs contributed to the increase of fertility in the population. In contrast, the genotype composed of alleles from ‘Balilla’ and ‘Nanjing 11’ at the fifth locus ( f1) was in the direction of increasing fertility. Analysis of variance using marker genotypes at the five QTLs as the groups detected two interactions involving four of the five loci, a 2-locus interaction between f5 and f8 and a 3-locus interaction among f3, f5 and f6. The level of hybrid fertility is the result of complex interactions among these loci. The implication of the present findings in the utilization of the wide-compatibility of ‘Dular’ in rice breeding programs is also discussed. Hybrid sterility hinders the exploitation of the heterosis displayed by japonica?×?indica rice hybrids. The variation in pollen semi-sterility observed among hybrids between the japonica recipient cultivar and each of two sets of chromosome segment substitution lines involving introgression from an indica cultivar was due to a factor on chromosome 5 known to harbor the gene S24. S24 was fine mapped to a 42?kb segment by analyzing a large F(2) population bred from the cross S24-NIL?×?Asominori, while the semi-sterility shown by the F(1) hybrid was ascribable to mitotic failure at the early bicellular pollen stage. Interestingly, two other pollen sterility genes (f5-Du and Sb) map to the same region (Li et al. in Chin Sci Bull 51:675-680, 2006; Wang et al. in Theor Appl Genet 112:382-387, 2006), allowing a search for candidate genes in the 6.4?kb overlap between the three genes. By sequencing the overlapped fragment in wild rice, indica cultivars and japonica cultivars, a protein ankyrin-3 encoded by the ORF2 was identified as the molecular base for S24. A cultivar Dular was found to have a hybrid-sterility-neutral allele, S24-n, in which an insertion of 30?bp was confirmed. Thus, it was possible to add one more case of molecular bases for the hybrid sterility. No gamete abortion is caused on heterozygous maternal genotype with an impaired sequence from the hybrid-sterility-neutral genotype. We have recently cloned a pathogen inducible blast resistance gene Pi-kh from the indica rice line Tetep using a positional cloning approach. In this study, we carried out structural organization analysis of the Pi-kh locus in both indica and japonica rice lines. A 100 kb region containing 50 kb upstream and 50 kb down- stream sequences flanking to the Pi-kh locus was selected for the investigation. A total of 16 genes in indica and 15 genes in japonica were predicted and anno- tated in this region. The average GC content of indica and japonica genes in this region was 53.15% and 49.3%, respectively. Both indica and japonica sequences were polymorphic for simple sequence repeats having mono-, di-, tri-, tetra-, and pentanucleotides. Sequence analysis of the specific blast resistant Pi-kh allele of Tetep and the susceptible Pi-kh allele of the japonica rice line Nipponbare showed differences in the number and distribution of motifs involved in phosphorylation, resulting in the resistance phenotype in Tetep.  相似文献
