【目的】开发一种高效地从造礁石珊瑚中分离、培养共生虫黄藻的技术方法,为珊瑚共生虫黄藻藻种资源储备和生理功能研究积累基础。【方法】首先采用微孔滤网过滤法和密度梯度离心法从造礁石珊瑚组织中直接分离或富集共生虫黄藻细胞,然后用改良的L1培养基在96孔板上对所得细胞进行离体培养,最后进行单细胞分离、培养和(或)平板划线培养获得单克隆虫黄藻细胞系。对所得虫黄藻单克隆藻株进行聚合酶链式反应-限制性内切酶片段长度多态性(polymerase chainreaction-restrictionfragmentlengthpolymorphism,PCR-RFLP)分析,结合内转录间隔区2(internal transcribed spacer2,ITS2)和大亚基(large subunit,LSU)测序进行物种鉴定及系统发育分析。【结果】采用上述方法从涠洲岛的霜鹿角珊瑚(Acropora pruinose)和西沙群岛的丛生盔形珊瑚(Galaxea fascicularis)及柔枝鹿角珊瑚(Acropora tenuis)中分离、培养得到3个虫黄藻株系,编号分别为AP21C1、GF21D1和AT21A...  相似文献

Many groups of tropical cnidarians including scleractinian corals, octocorals, corallimorphs, and anemones contain the tertiary sulfonium compound dimethylsulfoniopropionate (DMSP). It is not known if the compound is synthesized by the animals, their microalgal symbionts, or derived through their diet. We determined the source of the DMSP in several species of tropical and temperate anemones using three approaches: (1) conducting comparative measurements of DMSP in aposymbiotic and zooxanthellate anemones of three species that harbor zooxanthellae, and similar measurements in one species that can harbor both zooxanthellae and zoochlorellae, (2) manipulating the presence or absence of zooxanthellae by inoculating juvenile aposymbiotic anemones (Aiptasia pallida) with their symbiont, Symbiodinium bermudense, and (3) manipulating the numbers of S. bermudense by growing aposymbiotic and zooxanthellate A. pallida in the light and the dark. DMSP was present in zooxanthellate anemones in concentrations of 3.4–15 μmol g−1 fresh mass (FM). In aposymbiotic Aiptasia spp. and Anthopleura elegantissima that lacked large numbers of zooxanthellae, concentrations ranged from being undetectable to 0.43 μmol g−1 FM. When aposymbiotic A. pallida were inoculated with zooxanthellae, concentrations of DMSP were an average of 4.24 μmol g−1 FM after 5 weeks; DMSP was undetectable in uninoculated control animals. Aposymbiotic anemones maintained in the light or the dark for 6 weeks contained no DMSP or zooxanthellae. Zooxanthellate anemones in the light contained five times as many zooxanthellae and approximately 7.5 times as much DMSP as zooxanthellate anemones maintained in the dark. Taken together, these data show that the zooxanthellae are the sole source of DMSP in A. pallida. The trends in DMSP concentrations in other species of zooxanthellate anemones suggest that this phenomenon is not limited to A. pallida but may be more generally true for other anemones or even other cnidarians hosting species of Symbiodinium. Relationship among symbiotic dinoflagellates of the genusSymbiodinium derived from seven different host species has been studied by means of DNA/DNA hybridization. DNA homologies range from about 70 to 30%D. Highest homology is regarded as subspecific level. Based on the characteristics of DNA and phenotypes,Symbiodinium microadriaticum subsp.microadriaticum is separated fromSymbiodinium microadriaticum subsp.condylactis. Lowest homology occurs at the methodical background, and is similar to that obtained with DNA of algae belonging to different classes. The data are in excellent agreement with DNA base composition, karyotypes, and morpohological as well as biochemical markers, emphasizing speciation among these gymnodinioid zooxanthellae.Dedicated to DrRobert K. The comparative floral ontogeny of five species belonging to the primuloid clade of the Ericales are investigated, viz. Maesa japonica, M. perlarius, Aegiceras corniculatum, Embelia laeta and E. ribes. All five species basically show 2/5-spiral phyllotaxis of the sepal primordia, although with some minor modification (particularly in Embelia, where the flowers are predominantly tetramerous). The phyllotaxis of the common petal-stamen primordia is also 2/5-spiral in the Maesa and Aegiceras species investigated, but appears to be unidirectional in Embelia. All five species develop common petal-stamen primordia in which the resultant petal primordia are larger than the stamen primordia, and in which the stamens develop proximally on the adaxial flank of the common primordia. Growth of the placenta in Maesa and Aegiceras partially embeds the ovules, but in Embelia the ovules are almost fully immersed in placental tissue at maturity. A comprehensive review of all previously published studies of floral ontogeny of primuloid genera is presented, and the phylogenetic significance of the variation between genera is evaluated with reference to recently published cladograms.  相似文献

Frequent occurrences of coral bleaching and the ensuing damage to coral reefs have generated interest in documenting stress responses that precede bleaching. The objective of this study was to assess and compare physiological changes in healthy, semi-bleached and totally bleached colonies of two coral species, Porites lutea and Acropora formosa, during a natural bleaching event in the Lakshadweep Archipelago in the Arabian Sea to determine the traits that will be useful in the diagnosis of coral health. In April 2002, three “health conditions” were observed as “appearing healthy,” “semi-bleached” and “bleached” specimens for two dominant and co-occurring coral species in these islands. Changes in the pigment composition, zooxanthellae density (ZD), mitotic index (MI) of zooxanthellae, RNA/DNA ratios and protein profile in the two coral species showing different levels of bleaching in the field were compared to address the hypothesis of no difference in health condition between species and bleaching status. The loss in chlorophyll (chl) a, chl c and ZD in the transitional stage of semi-bleaching in the branched coral A. formosa was 80, 75 and 80%, respectively. The losses were much less in the massive coral P. lutea, being 20, 50 and 25%, respectively. The decrease in zooxanthellar density and chl a was accompanied by an increased MI of zooxanthellae and RNA/DNA ratios in both the species. There was an increase in accumulation of lipofuscin granules in partially bleached P. lutea tissue, which is an indication of cellular senescence. Multivariate statistical analyses showed that colonies of P. lutea ranked in different health conditions differed significantly in chl a, chl c, ZD, RNA/DNA ratios, and protein concentrations, whereas in A. formosa chl a, chl c, chl a/c, phaeopigments and MI contributed to the variance between health conditions.  相似文献

Amplified fragment length polymorphism (AFLP) markers were used to assess the genetic diversity of 57 Achillea accessions belonging to five species, A. millefolium, A. filipendulina, A. tenuifolia, A. santolina and A. biebersteinii. Nine AFLP primer combinations were used, which produced 301 polymorphic bands. In most species, a high level of genetic variation was detected among the genotypes. The Jaccard's similarity indices (J), based on AFLP profiles, were subjected to UPGMA cluster analysis. Application of Mantel's test for cophenetic correlation to the cluster analysis indicated the high fitness of the accessions to a group (r = 0.918). The dendrogram generated revealed five major groups corresponding to five species. The principle coordinate analysis (PCoA) data confirmed the results of the clustering. Among the species, A. teunifolia and A. santolina showed the greatest and the least genetic diversity, respectively. A. filipendulina accessions were acquired primarily from the same ecological regions of western Iran. Accessions belonging to A. biebersteinii originated from the Isfahan province and were separated from other species at the root of the dendrogram. The results of the clustering method, based on AFLP markers, corresponded closely with the geographical origins of the genotypes. The results of the present study could contribute to a better understanding and management of conservation and exploitation of the Achillea germplasm.  相似文献

The anatomy of seven species of aeolid opisthobranch molluscs is described. They are all considered to be specialized feeders on alcyonarian coelenterates and to belong to one genus Phyllodesmium (Glaucidae; Favorininae). Two species are considered to be new. Favorinus horridus Macnae, Aeolidia poindimiei Risbec, Cratena macphersonae Burn and another species not studied in this paper, Hervia serrata Baba, are transferred to Phyllodesmium from the genera in which they are currently placed. Myrrhine Bergh, Babaiella Risso-Dominguez and Phyllodesmiopsis Risso-Dominguez are considered synonyms of Phyllodesmium. The two new species feed on Xenia and P. poindimiei feeds on Telesto. It is shown that the species of Phyllodesmium exhibit an evolutionary series in the development of symbiosis with zooxanthellae. The zooxanthellae are obtained from the alcyonarians upon which the aeolids feed. Morphological peculiarities present in some members of the genus are considered to be associated with the symbiosis.  相似文献

This study focused on the association between corals of the genus Pocillopora, a major constituent of Pacific reefs, and their zooxanthellae. Samples of P. meandrina, P. verrucosa, P. damicornis, P. eydouxi, P. ligulata and P. molokensis were collected from French Polynesia, Tonga, Okinawa and Hawaii. Symbiodinium diversity was explored by looking at the 28S and ITS1 regions of the ribosomal DNA. Most zooxanthellae were found to belong to clade C, sub-clade C1, with little differentiation between populations. Interestingly, individuals of P. damicornis harbored sub-clade C1, clade D and clade A, depending on location. The symbiotic association of P. damicornis with its zooxanthellae may be somewhat more flexible than those of other Pocillopora species.  相似文献

Previous efforts at understanding the evolution of the genus Phyllodesmium, based on morphological analyses, have been plagued by poorly supported phylogenies (Ortiz and Gosliner, 2008; Moore and Gosliner, 2009, in press). It has been suggested (Moore and Gosliner, 2009) that a molecular phylogeny might provide more insight into this history than can be easily discovered using morphological data. In this study, 658bp of the cytochrome c oxidase subunit I gene (CO1), 441bp of the mitochondrial large ribosomal subunit (16S) gene, and 328bp of a protein-coding nuclear gene (histone 3) were sequenced for 18 species of Phyllodesmium and six outgroup species. A total of 464 parsimony informative sites were used for parsimony, maximum likelihood, and Bayesian inference of phylogeny analyses. All three analyses produced similar topologies, with the exception of a single difference within the parsimony analysis. Bootstrap values and posterior probabilities provided strong support at many shallow nodes, and the monophyly of Phyllodesmium was supported in every case. Three distinct clades of Phyllodesmium are evident in this analysis. One of these represents the majority of asymbiotic taxa. Phyllodesmium poindimiei, an asymbiotic species, is clearly a member of a symbiotic clade and appears to have secondarily lost its symbiotic relationship with zooxanthellae. There was moderate support confirming similar topological trends seen in earlier morphological phylogenies, including the hypothesis that symbiotic species associating with zooxanthellae have evolved more recently than non-symbiotic species. Despite the inclusion of a presumably conservative nuclear locus, some deep nodes are still unresolved or are not well supported. Future inclusion of additional taxa and more slowly evolving loci will likely improve resolution of these deeper nodes. The subsequent phylogeny supports previous hypotheses by Rudman (1991), Kempf (1991) and Burghardt et al. (2008b) that evolution of more complex digestive gland structures is related to increased complexity of symbiosis with zooxanthellae and greater efficiency of photosynthetic activity. Our phylogeny also demonstrates that this symbiosis has evolved only once in Phyllodesmium and that azooxanthellate species are sister, rather than basal, to zooxanthellate species.  相似文献

The colonization and diversity of arbuscular mycorrhizal (AM) fungi associated with common pteridophytes were investigated in Dujiangyan, southwest China. Of the 34 species of ferns from 16 families collected, 31 were colonized by AM fungi. The mean percentage root length colonized was 15%, ranging from 0 to 47%. Nineteen species formed Paris-type and 10 intermediate-type AM. In two ferns, only rare intercellular non-septate hyphae or vesicles were observed in the roots and AM type could not be determined. Of the 40 AM fungal taxa belonging to five genera isolated from rooting-zone soils, 32 belonged to Glomus, five to Acaulospora, one to Archaeospora, one to Entrophospora, and one to Gigaspora. Acaulospora and Glomus were the dominant genera and Glomus versiforme was the most common species. The average AM spore density was 213 per 100 g air-dried soil and the average species richness was 3.7 AM species per soil sample. There was no correlation between spore density and percentage root length colonized by AM fungi.  相似文献

The symbiotic dinoflagellate microalgae of corals (Symbiodinium spp.) contain high concentrations of dimethylsulfoniopropionate (DMSP), a multifunctional metabolite commonly found in many species of marine algae and dinoflagellates. A photoprotective antioxidant function for DMSP and its breakdown products has often been inferred in algae, but its role(s) in the coral–algal symbiosis remains elusive. To examine potential correlations between environmental and physiological parameters and DMSP, total DMSP (DMSPt, from the host coral and zooxanthellae), particulate DMSP (DMSPp, from the zooxanthellae only), coral surface area, and total protein, as well as zooxanthellae density, chlorophyll concentration, cell volume and genotype (i.e., clade) were measured in five coral species from the Diploria-Montastraea-Porites species complex in Bermuda along a depth gradient of 4, 12, 18, and 24 m. DMSPt concentrations were consistently greater than DMSPp concentrations in all species suggesting the possible translocation of DMSP from symbiont to host. D. labyrinthiformis was notably different from the other corals examined, showing DMSPp and DMSPt increases (per coral surface area or tissue biomass) with increasing water depth. However, overall, there were no consistent depth-related patterns in DMSPp and DMSPt concentrations. Further research, investigating dimethylsulfide (DMS), dimethylsulfoxide, and acrylate levels and DMSP-lyase activity in correlation with other biomarker endpoints that have been shown to be depth (i.e., temperature and light) responsive are needed to substantiate the significance of these findings.  相似文献

We examined the morphology and pigment composition of zooxanthellae in corals subjected to normal temperature (27°C) and thermal stress (32°C). We observed several normal and abnormal morphological types of zooxanthellar cells. Normal cells were intact and their chloroplasts were unbroken (healthy); abnormal cells were shrunken and had partially degraded or broken chloroplasts, or they were bleached and without chloroplasts. At 27°C, most healthy zooxanthellar cells were retained in the coral tissue, whereas shrunken zooxanthellae were expelled. Under thermal stress, the abundance of healthy zooxanthellae declined and the proportion of shrunken/abnormal cells increased in coral tissues. The rate of algal cell expulsion was reduced under thermal stress. Within the shrunken cells, we detected the presence of a chl‐like pigment that is not ordinarily found in healthy zooxanthellae. Analysis of the absorption spectrum, absorption maxima, and retention time (by HPLC) indicated that this pigment was 132, 173‐cyclopheophorbide a enol (cPPB‐aE), which is frequently found in marine and lacustrine sediments, and in protozoans that graze on phytoplankton. The production of cPPB‐aE in shrunken zooxanthellae suggests that the chls have been degraded to cPPB‐aE, a compound that is not fluorescent. The lack of a fluorescence function precludes the formation of reactive oxygen species. We therefore consider the formation of cPPB‐aE in shrunken zooxanthellae to be a mechanism for avoiding oxidative stress.  相似文献

We examined zooxanthellae diversity in scleractinian corals from southern Taiwan and the Penghu Archipelago, a tropical coral reef and a subtropical non-reefal community, respectively. Zooxanthellae diversity was investigated in 52 species of scleractinian corals from 26 genera and 13 families, using restriction fragment length polymorphism (RFLP), and phylogenetic analyses of the nuclear small-subunit ribosomal DNA (nssrDNA) and large-subunit ribosomal DNA (nlsrDNA). RFLP and phylogenetic analyses of nuclear-encoded ribosomal RNA genes showed that Symbiodinium clade C was the dominant zooxanthellae in scleractinian corals in the seas around Taiwan; Symbiodinium clade D was also found in some species. Both Symbiodinium clade C and D were found in colonies of seven species of scleractinian corals. Symbiodinium clade D was associated with corals that inhabit either shallow water or the reef edge in deep water, supporting the hypothesis that Symbiodinium clade D is a relatively stress-tolerant zooxanthellae found in marginal habitats.Communicated by Biological Editor H.R. Cryptococcus neoformans, a major pathogen in immunocompromised patients, is a ubiquitous free-living fungus that can be isolated from soils, avian excreta and plant material. To further study potential saprophytic sources of this yeast in the Southern Brazilian State Rio Grande do Sul, we analyzed fecal samples from 59 species of captive birds kept in cages at a local Zoological Garden, belonging to 12 different orders. Thirty-eight environmental isolates of C. neoformans were obtained only from Psittaciformes (Psittacidae, Cacatuidae and Psittacula). Their variety and serotype were determined, and the genetic structure of the isolates was analyzed by use of the simple repetitive microsatellite specific primer M13 and the minisatellite specific primer (GACA)4 as single primers in the PCR. The varieties were confirmed by pulsed-field gel electrophoresis (PFGE). Thirty-three isolates (87%) were from the var. grubii, serotype A, molecular type VNI and five (13%) were Cryptococcus gattii, serotype B, molecular type VGI. All the isolates were mating type α. Isolates were screened for some potential virulence factors. Quantitative urease production by the environmental isolates belonging to the C. gattii was similar to the values usually obtained for clinical ones.  相似文献

A satellite sequence repeat ofAllium cepa was tested by fluorescent in situ hybridization (FISH) for cross-hybridization to chromosomes of 27 species (in 37 accessions) belonging to 14 sections of four subgenera ofAllium. All investigated species of sect.Cepa, with the two subsects.Cepa andPhyllodolon, revealed clear satellite-specific hybridization signals mainly at their chromosome termini. The tested species belonging to other sections/subgenera revealed no hybridization signals. An exception wasA. roylei, assigned to sect.Oreiprason. Its chromosomes also showed strong terminal hybridization signals. This and other features suggest a close relationship ofA. roylei to the species of sect.Cepa in spite of deviating morphological characters. The divergence between the satellite repeats to species to which theA. cepa repeat cross-hybridized was determined and revealed high degrees of similarity. Therefore, we conclude that this satellite sequence had evolved already in progenitor forms of sect.Cepa and remained unusually well conserved during speciation. 脆弱刚毛藻(Cladophora fracta)是一种大型丝状绿藻,生境分布广泛。然而,对于岩溶泉域分布的刚毛藻研究较少,它们的遗传多样性、生物地理亲缘性和生理特性都有待于深入研究。该研究对我国北方地区五个典型岩溶泉域的50个脆弱刚毛藻样本进行了形态学和分子系统学描述。主要研究目标:(1)对我国北方地区五个典型岩溶泉的刚毛藻生境进行描述;(2)根据形态学特征和分子序列对藻体进行鉴定;(3)探究生境对藻体生理特性的影响。结果表明:基于SSU和LSU序列的结果,发现所分析的50株刚毛藻个体为同一种,同时还发现了13个不同的核糖体基因型。基于SSU和LSU的系统发育树,刚毛藻属均未能形成单系分支,分布在三个不同的分支上。13个样本基因型在SSU和LSU树中的位置相似,与Cladophora vagabunda有很高的序列同源性,但是形态特征却差异很大。从显微结构结果来看,五个岩溶泉域采集到的刚毛藻在细胞直径上无显著差异,藻体的形态特征与脆弱刚毛藻相一致。但是,岩溶泉域采集的藻体细胞直径比文献报道中在湖泊和河流中采集的脆弱刚毛藻直径要大。另外,仅在两个地点(XA和ST)采集的标本中发现有假根状分枝。因此,基于形态学和分子序列的结果,将这五个泉域的刚毛藻鉴定为脆弱刚毛藻(Cladophora fracta)。  相似文献

Phenetic analyses of 218 OTUs belonging toVaccinium sectionMyrtillus and scored for 13 characters generated five robust clusters.Vaccinium parvifolium is the most distinct cluster, followed by the “myrtillus-scoparium” complex, thenV. membranaceum, V. caespitosum, and the “ovalifolium-deliciosum” complex. Biosystematic studies suggest that the five clusters comprise seven taxa that possess many of the properties that define biological species. Indeed, the recognition of the seven taxa as species is supported by cytological, phenological, biogeographical and ecological as well as chemical data. 为保护香港特有植物香港鹅耳枥,基于典型样地调查,对香港鹅耳枥所在群落进行了研究。结果表明,在225 m2的样地内,有维管植物37科66属68种。建群种为香港鹅耳枥,个体高度最高3 m,所在群落植株平均高度0.92 m,分层不明显。位于中国香港岛的香港鹅耳枥处于衰退状态,小个体极少。因此,建议采取就地保护和迁地保护措施,以重建种群。  相似文献

Summary. Preparation of homogeneous endoderm cells and culture is a prerequisite to understanding the cellular and molecular mechanism of endosymbiosis in the cnidarian-dinoflagellate association. During the cell isolation from the stony coral Euphyllia glabrescens, various amounts of symbiotic endoderm cells were found to release their symbionts (Symbiodinium spp., or zooxanthellae in generic usage) into the culture. Due to the bulky occupation by zooxanthellae inside the endoderm cell, the symbiotic endoderm cells, or zooxanthellae in hospite, are difficult to be distinguished from released zooxanthellae by microscopic examination. We now report a method for this identification using a fluorescent analogue of sphingomyelin, N-[5-(5,7-dimethyl boron dipyrromethene difluoride)-1-pentanoyl]-D-erythro-sphingosylphosphorylcholine (C5-DMB-SM). Incubation of symbiotic endoderm cells with C5-DMB-SM–defatted bovine serum albumin (DF-BSA) complex results in bright fluorescent membrane staining. Nevertheless, the membrane staining of free-living or released zooxanthellae by this complex is significantly decreased or even diminished. This method has provided a fast and reliable assay to identify symbiotic endoderm cells and will greatly accelerate the progress of endosymbiosis research. Macro and micromorphological properties of intact and mature seeds of 12 taxa (species and varieties) belonging to Nigella L. (Ranunculaceae) was investigated using light and scanning electron microscopy. Material studied covers 11 species of 15 Turkish Nigella. Studied taxa were divided into two types. Type I has ovate to orbicular seeds that includes four species. Type II has triquedrous seeds and includes seven species. Type II was subdivided into two. Type IIa has triquedrous to subpyramidal seeds (five species) and Type IIb has triquedrous to subglobose seeds (two species). Further segregation was performed micromorphologically and an identification key of studied Nigella taxa was given. Studied Nigella taxa have a diverse macro and micromorphological characters that utilize to separate them from each other to assess the systematics of Nigella.  相似文献
