To determine whether prior exposure to Nearctic Ixodes vector ticks protects native reservoir mice from tick-borne infection by Lyme disease spirochetes, we compared their infectivities for white-footed mice and laboratory mice that had been repeatedly infested by noninfected deer ticks. Nymphal ticks readily engorged on tick-exposed laboratory mice, but their feeding success on white-footed mice progressively declined. Tick-borne spirochetes readily infected previously tick-infested mice. Thus, prior infestation by Nearctic ticks does not protect sympatric reservoir mice or Palearctic laboratory mice from infection by sympatric tick-borne spirochetes.  相似文献

Rhipicephalus sanguineus is believed to be the most widespread tick species of the world and its dissemination seems to rely on the diffusion of its main host, the dog. Empirical observations indicate that several bird species in urban areas regularly steal dog food. Such circumstances create a chance for R. sanguineus ticks to climb on birds and carry ticks to another site. In this work we evaluated experimentally the likelihood of birds (chicks) to either feed and/or carry R. sanguineus ticks from an infested site to another and to infest a host (rabbit) in the new location. Chicks were not suitable hosts for R. sanguineus ticks. Not a single adult tick engorged on chicks, yield as well as weight of engorged larvae and nymphs were very low and feeding period of these ticks was very long. However, a few larvae and, chiefly, nymphs were delivered to a new location either mechanically or after attachment and engorging total or partially on chicks. A few of these ticks fed successfully on rabbits. Further evidence on the capacity of birds to introduce R. sanguineus into non-infested dog settings should be provided by systematic examination of birds from urban areas, close to tick infested households.  相似文献

To determine whether replete subadult Ixodes ticks detach more frequently from resting than from active hosts, diverse rodents and lizards were caged in an apparatus designed to record the ticks' sites of detachment relative to the resting site of the host. Replete larval Ixodes ricinus and Ixodes dammini accumulated mainly beneath the resting places of the mice (Apodemus agrarius and Peromyscus leucopus) most frequently parasitized in nature. Although nymphal I. ricinus similarly detached where these mice rested, nymphal I. dammini detached more randomly. When lizards were used as hosts, both subadult stages of I. ricinus tended to detach away from their main resting sites; these ticks detached from squirrels more randomly. Detachment ratios for other rodent hosts, that are abundantly infested by the larvae of these ticks in nature (Apodemus flavicollis and Clethrionomys glareolus), could not be derived because nymphs generally failed to attach. Our observations are consistent with reports that both subadult stages of I. dammini, but not the adult, feed on the same kind of nest-dwelling hosts and that the host range of I. ricinus is less focused. Detachment of mouse-feeding larvae from resting mice promotes subsequent nymphal attachment to conspecific hosts, and the absence of such behavior among nymphs facilitates access of the resulting adults to deer.  相似文献

The vector competence of Ixodes angustus for Borrelia burgdorferi sensu stricto (s.s.) was investigated in the laboratory. The larval progeny of female ticks from Washington State were placed on Swiss-Webster mice that had been inoculated intravenously with 108 spirochetes each of a Californian isolate of B. burgdorferi. Spirochetes were detected in 6 (12%) of 50 nymphs derived from larvae that had fed on these animals. Ten nymphs from the same cohort of experimentally infected ticks were placed on each of 4 naive deer mice (Peromyscus maniculatus). One of the mice seroconverted to B. burgdorferi and spirochetes were isolated from its ear tissues 4 weeks after exposure to ticks. Further vector competence trials were conducted with I. angustus ticks from California. Larvae were fed on deer mice that had been inoculated intradermally with B. burgdorferi along with larvae of I. spinipalpis as a comparison group. There was no significant difference in the prevalence of infection in nymphs of I. angustus (8.2%) versus those of I. spinipalpis (12.1%). We conclude that I. angustus is a competent experimental vector of B. burgdorferi s.s. and its efficiency for acquiring and transstadially passing such spirochetes is similar to that of I. spinipalpis.  相似文献

In order to determine whether the small-scale distribution of immature Ixodes dammini Spielman et al. corresponds closely to the activity patterns of white-tailed deer, Odocoileus virginianus (Zimmerman), these relationships were examined in a site on Long Island, New York, U.S.A. We first determined the extent and temporal pattern of adult ticks feeding on deer by examining twenty-three resident deer tranquilized during September-December 1985. I. dammini adults infested deer throughout this fall period, most abundantly during October and November. With radio-telemetry collars attached to deer we determined the relative frequency that they occupied 0.25 ha quadrats of the study site. During the following summer, we examined white-footed mice, Peromyscus leucopus (Rafinesque), that inhabited these quadrats and removed immature ticks from each. 8975 larval and 163 nymphal I. dammini were removed from 208 mice trapped in forty-three such quadrats. The frequency of deer using these quadrats was positively correlated with both the number of larval and of nymphal ticks per mouse. These results suggest that risk of I. damminiborne zoonotic disease may be decreased by locally reducing deer density in sites that experience intense human activity.  相似文献

From June through August in 1999 and 2000, we conducted an avian serosurvey for eastern equine encephalomyelitis (EEE) virus at Killbuck Marsh Wildlife Area (KMWA), a focus of infection in central Ohio. We also monitored abundance of the suspected enzootic vector, Culiseta melanura Coquillett, in Brown's Lake Bog, an adjacent wetland. Of the 363 birds of 30 species sampled in 1999, three gray catbirds (Dumetella carolinensis) were positive for antibodies to EEE virus, representing 1.2% of the avian samples and 4.2% of the gray catbirds sampled. Given these results and the abundance of gray catbirds at this site, this species became the focus of our sampling efforts in 2000. However, none of the 109 samples collected from 98 catbirds in 2000 was positive for the virus. Culiseta melanura adults were monitored using resting boxes and CDC CO, light traps at both sites in 1999. Culiseta melanura larvae were monitored in 1999 and 2000 at Brown's Lake Bog, the closest known source of this species, approximately 5km from the avian serosurvey site. We suggest that dry conditions reduced the breeding and abundance of Cs. melanura in 2000 and possibly the transmission of EEE virus at KMWA.  相似文献

One hundred ten white-footed mice (Peromyscus leucopus) were captured on horse farms in south-central Maryland, examined for ticks, and tested for specific antibodies to Ehrlichia risticii, the causative agent of Potomac horse fever. Peromyscus leucopus were consistently infested with immature American dog ticks (Dermacentor variabilis), with monthly prevalences as high as 80%. Sera from all 97 P. leucopus tested for antibodies to E. risticii were negative. This indicates that P. leucopus is not a reservoir of E. risticii, and suggests that immature D. variabilis do not acquire E. risticii in feeding upon white-footed mice.  相似文献

Vaccines that target blood-feeding disease vectors, such as mosquitoes and ticks, have the potential to protect against the many diseases caused by vector-borne pathogens. We tested the ability of an anti-tick vaccine derived from a tick cement protein (64TRP) of Rhipicephalus appendiculatus to protect mice against tick-borne encephalitis virus (TBEV) transmitted by infected Ixodes ricinus ticks. The vaccine has a "dual action" in immunized animals: when infested with ticks, the inflammatory and immune responses first disrupt the skin feeding site, resulting in impaired blood feeding, and then specific anti-64TRP antibodies cross-react with midgut antigenic epitopes, causing rupture of the tick midgut and death of engorged ticks. Three parameters were measured: "transmission," number of uninfected nymphal ticks that became infected when cofeeding with an infected adult female tick; "support," number of mice supporting virus transmission from the infected tick to cofeeding uninfected nymphs; and "survival," number of mice that survived infection by tick bite and subsequent challenge by intraperitoneal inoculation of a lethal dose of TBEV. We show that one dose of the 64TRP vaccine protects mice against lethal challenge by infected ticks; control animals developed a fatal viral encephalitis. The protective effect of the 64TRP vaccine was comparable to that of a single dose of a commercial TBEV vaccine, while the transmission-blocking effect of 64TRP was better than that of the antiviral vaccine in reducing the number of animals supporting virus transmission. By contrast, the commercial antitick vaccine (TickGARD) that targets only the tick's midgut showed transmission-blocking activity but was not protective. The 64TRP vaccine demonstrates the potential to control vector-borne disease by interfering with pathogen transmission, apparently by mediating a local cutaneous inflammatory immune response at the tick-feeding site.  相似文献

Tick saliva (or salivary gland extract) potentiates the transmission of Thogoto (THO) virus to uninfected ticks feeding on a non-viraemic guinea-pig. This phenomenon has been named saliva activated transmission (SAT). To investigate the potential of different haematophagous arthropods to mediate SAT, guinea-pigs were infested with uninfected R.appendiculatus Neumann nymphs and inoculated with THO virus and salivary gland extract (SGE) derived from a range of ixodid (metastriate and prostriate) or argasid ticks, or mosquitoes; control guinea-pigs were inoculated with virus alone. Enhancement of THO virus transmission was observed only when SGE was derived from metastriate ticks. Comparison with the vector potential of these various arthropod species revealed that enhancement of THO virus transmission was specific for ticks which were competent vectors of the virus. The data indicate a correlation between vector competence and the ability of haematophagous arthropods to mediate SAT of THO virus.  相似文献

In April 2008, 585 tortoises illegally imported into Italy from North Africa were examined for the presence of ticks. Of these, 221 tortoises (37.8%) were infested with a mean intensity of 3.9 ± 3.1 ticks (range 1–17 ticks). A total of 798 ticks (672 males, 125 females and one nymph) were collected and identified as Hyalomma aegyptium (L.) (Acari: Ixodidae). The overall male : female ratio observed was 5 : 1. The prevalence and mean intensity [±standard deviation (SD)] of ticks were higher among male (67.4%, 4.0 ± 3.2) than female (55.6%, 3.8 ± 3.1) tortoises, although this difference was not significant. By contrast, the prevalence and mean intensity of ticks were significantly higher on tortoises weighing >100 g (61.5%, 4.0 ± 3.2) compared with tortoises weighing <100 g (12.1%, 2.1 ± 1.1). Of the infested tortoises, 89.8% had ticks on their hind limbs, 21.0% on forelimbs, 18.6% on the tail and pre‐anal area, and 4.8% on the head; thus the hind limbs were evidently the preferred attachment site. The present report highlights the need to develop surveillance systems to prevent the introduction and spreading of exotic ticks and tick‐borne pathogens in Italy and other European countries.  相似文献

To determine whether short-tailed shrews (Blarina brevicauda) serve as reservoir hosts for the Lyme disease spirochete (Borrelia burgdorferi) and the agent of human babesiosis (Babesia microti), we examined nymphal ticks that had fed as larvae on shrews collected from 3 enzootic sites in coastal Massachusetts for evidence of infection by either or both of these agents. Xenodiagnosis indicated that 11 of 14 shrews were infected by B. burgdorferi. One of 3 piroplasm-infected shrews also infected ticks with B. microti. In a site where the piroplasm is endemic, 11 of 17 shrews showed patent parasitemias by thin blood smears. Of these, 4 had parasitemias exceeding 40%. Few immature ticks infested shrews, however, suggesting that B. brevicauda, although abundant in some endemic sites and serving as a competent reservoir, would contribute minimally to the population of infected nymphs.  相似文献

A deer was needle-exposed intravenously to Ehrlichia chaffeensis (Rickettsiales: Ehrlichieae) in canine macrophage (DH82) cells and 7 days later was infested with laboratory-reared Amblyomma maculatum (Koch) (Acari:Ixodidae) nymphs for acquisition feeding. After molting, the adult ticks were allowed to feed on a naive deer. The organism was reisolated from the needle-exposed deer by cell culture and E. chaffeensis DNA was detected in the deer's blood by PCR. Similar isolation/recovery techniques were used for the tick-exposed deer and no evidence of infection was found. Although these findings must be considered as preliminary owing to inadequate controls, the data suggest that A. maculatum is probably not a suitable vector for E. chaffeensis.  相似文献

Antisera to BBK32 (a Borrelia burgdorferi fibronectin-binding protein) and BBK50, two Ags synthesized during infection, protect mice from experimental syringe-borne Lyme borreliosis. Therefore, B. burgdorferi bbk32 and bbk50 expression within Ixodes scapularis ticks and the murine host, and the effect of BBK32 and BBK50 antisera on spirochetes throughout the vector-host life cycle were investigated. bbk32 and bbk50 mRNA and protein were first detected within engorged ticks, demonstrating regulated expression within the vector. Then bbk32 expression increased in mice at the cutaneous site of inoculation. During disseminated murine infection, bbk32 and bbk50 were expressed in several murine tissues, and mRNA levels were greatest in the heart and spleen at 30 days. BBK32 antisera protected mice from tick-borne B. burgdorferi infection and spirochete numbers were reduced by 90% within nymphs that engorged on immunized mice. Moreover, 75% of these ticks did not retain spirochetes upon molting, and subsequent B. burgdorferi transmission by adult ticks was impaired. Larval acquisition of B. burgdorferi by I. scapularis was also inhibited by BBK32 antisera. These data demonstrate that bbk32 and bbk50 are expressed during tick engorgement and that BBK32 antisera can interfere with spirochete transmission at various stages of the vector-host life cycle. These studies provide insight into mechanisms of immunity to Lyme borreliosis and other vector-borne diseases.  相似文献

Abstract. A self-medicating applicator for killing ticks on deer and domestic ungulates by passive transfer of acaricides during feeding is described. The applicator consists of a barrel divided into a food bin (above) and a sealed self-contained acaricide reservoir (below) with a vertical, centrally-located ceramic column that extends from the reservoir into the food bin. Acaricide is drawn up the column from the sealed reservoir by absorption. Animals attracted to the food in the device acquire the acaricide during feeding when they contact the uppermost portion of the column.
White-tailed deer (Odocoileus virginianus) readily utilize the applicator in penned and natural conditions. In a field study, examination of hunter-killed deer demonstrated that animals from a treated site were infested with many fewer Ixodes scapularis (3.4 ± 1.1) than those from a control site (10.8 ± 3.0). Chromatographic analysis of hair samples revealed traces of permethrin on three of the four animals examined, suggesting use of applicators by these deer. In a study using penned goats {Capra hircus) infested with Amblyomma americanum ticks, treatment efficacy reached 86.4% within 4 days of exposure to the applicators. Visual observations confirmed that all animals used the applicators. Gas chromatographic analysis of goat hair samples indicated that permethrin was detectable on all of the treated animals exposed to the device.  相似文献

Mist-netted birds were examined for ticks on Jekyll Island, Glynn Co., Georgia (32 bird species) in 1996-1998, and at Fort Morgan, Baldwin Co., Alabama (36 species) in 1998 during fall migration. Sixty-two (14.7%) of 423 birds from Jekyll Island and 22 (13.3%) of 165 birds from Fort Morgan were infested with ticks. The mean number of ticks per infested bird was 2.0 on Jekyll Island and 6.3 at Fort Morgan. Ten species of birds were infested with ticks on Jekyl1 Island where 87% of all ticks were recovered from 3 species: the common yellowthroat (Geothlypis trichas), gray catbird (Dumetella carolinensis), and northern waterthrush (Seiurus noveboracensis). Eight species of birds were infested with ticks at Fort Morgan where 83% of all ticks were recovered from 3 species: the brown thrasher (Toxostoma rufum), swamp sparrow (Melospiza georgiana), and common yellowthroat. Six species of ticks (Amblyomma americanum, Amblyomma maculatum, Haemaphysalis leporispalustris, Ixodes brunneus, Ixodes minor, and Ixodes scapularis) were recovered from the Georgia birds, whereas 3 species (A. maculatum, H. leporispalustris, and Ixodes dentatus) were recovered from the Alabama birds. Attempts to isolate Borrelia burgdorferi sensu stricto, the etiologic agent of Lyme borreliosis, from Ixodes spp. ticks recovered from birds were unsuccessful.  相似文献

Four field experiments were conducted with creole cattle in Guadeloupe to study the attractivity for adult ticks of animals previously infested with males of Amblyomma variegatum producing aggregation attachment pheromones (AAPs). In each experiment, two groups of cattle were used; the first group was infested with adult A. variegatum and the second group was kept free of ticks and pheromones by acaricidal treatment and by washing the hair and skin of previously infested sites. Both groups were then exposed to natural infestation by ticks present in the environment or by ticks bred in the laboratory and released on the pasture. The number of additional ticks infesting animals were compared between the treated and control groups. There were no significant differences between both groups in the numbers of new male ticks infesting the animals during the first days following the beginning of the experiment. Thereafter, comparison of the slopes of infestation indicated that infested cattle were no more attractive for male ticks than uninfested hosts. Uninfested cattle were spontaneously and rapidly infested by male ticks. It is concluded that carbon dioxide or another host component was a major attractant for A. variegatum males. Female ticks attached on predilection sites exclusively in the presence of AAP-producing males, i.e. 5–10 days after the attachment of the first males. Based on these results we postulate that slow-release devices impregnated with pheromones have no additional attractive effect on A. variegatum males under Caribbean conditions. However, they may be useful in inducing the attachment of females, but only in the first days preceding the arrival of pioneer AAP-producing males. © Rapid Science Ltd. 1998  相似文献

Millions of Lyme disease vector ticks are dispersed annually by songbirds across Canada, but often overlooked as the source of infection. For clarity on vector distribution, we sampled 481 ticks (12 species and 3 undetermined ticks) from 211 songbirds (42 species/subspecies) nationwide. Using PCR, 52 (29.5%) of 176 Ixodes ticks tested were positive for the Lyme disease spirochete, Borrelia burgdorferi s.l. Immature blacklegged ticks, Ixodes scapularis , collected from infested songbirds had a B. burgdorferi infection prevalence of 36% (larvae, 48%; nymphs, 31%). Notably, Ixodes affinis is reported in Canada for the first time and, similarly, Ixodes auritulus for the initial time in the Yukon. Firsts for bird-parasitizing ticks include I. scapularis in Quebec and Saskatchewan. We provide the first records of 3 tick species cofeeding on passerines (song sparrow, Swainson's thrush). New host records reveal I. scapularis on the blackpoll warbler and Nashville warbler. We furnish the following first Canadian reports of B. burgdorferi-positive ticks: I. scapularis on chipping sparrow, house wren, indigo bunting; I. auritulus on Bewick's wren; and I. spinipalpis on a Bewick's wren and song sparrow. First records of B. burgdorferi-infected ticks on songbirds include the following: the rabbit-associated tick, Ixodes dentatus, in western Canada; I. scapularis in Quebec, Saskatchewan, northern New Brunswick, northern Ontario; and Ixodes spinipalpis (collected in British Columbia). The presence of B. burgdorferi in Ixodes larvae suggests reservoir competency in 9 passerines (Bewick's wren, common yellowthroat, dark-eyed junco, Oregon junco, red-winged blackbird, song sparrow, Swainson's thrush, swamp sparrow, and white-throated sparrow). We report transstadial transmission (larva to nymph) of B. burgdorferi in I. auritulus. Data suggest a possible 4-tick, i.e., I. angustus, I. auritulus, I. pacificus, and I. spinipalpis, enzootic cycle of B. burgdorferi on Vancouver Island, British Columbia. Our results suggest that songbirds infested with B. burgdorferi-infected ticks have the potential to start new tick populations endemic for Lyme disease. Because songbirds disperse B. burgdorferi-infected ticks outside their anticipated range, health-care providers are advised that people can contract Lyme disease locally without any history of travel.  相似文献

A long-term field study (60 consecutive weeks) was conducted to determine whether a Boophilus annulatus (Say) population could be eradicated through repeated applications of a 1% pour-on formulation of fipronil on cattle held in an infested pasture. Animals treated repeatedly over time (seven applications) were infested with significantly fewer (p < 0.05) female ticks (5 mm in size) than untreated animals in all except one of the 57 weekly tick counts conducted after the first treatment was applied. As the number of treatments increased over time, there was a progressive decrease in the number of instances in which engorging females were observed on the treated cattle. Tick numbers obtained from sentinel cattle placed in pasture with untreated and treated cattle at various intervals throughout the study indicated that a single fipronil treatment had no observable adverse effect on the field tick population. However, after two treatments had been applied, there were always dramatically fewer ticks obtained from sentinel animals placed with treated cattle than were observed on sentinel animals placed with untreated cattle. Furthermore, tick numbers obtained from sentinel cattle placed with treated cattle over time indicated that the tick population in the pasture declined by stages. While a single treatment had no effect on the tick population, the application of two to four fipronil treatments at various intervals dramatically reduced the tick population, even though substantial numbers of females were still present on the sentinel animals. Ultimately, the application of five to seven fipronil treatments resulted in an extremely low tick population in the pasture, although total eradication of the population was not achieved during the study. The mean weight gain of treated animals was significantly higher (p < 0.05) than untreated animals during the study, indicating that the fipronil treatments had a highly beneficial impact on weight gain production.  相似文献

Guinea-pigs infested with male ticks of the speciesRhipicephalus appendiculatus, and rabbits infested withR. evertsi evertsi, acquired immunity to conspecific female ticks. The hosts were first infested with male ticks and thereafter were challenged with males and females of the same species. The mean weight of the engorged females ofR. appendiculatus fed on guinea pigs previously infested with male ticks was 509.0 (±41.4) mg compared with that of females fed on control guinea pigs (651.2±31.8 mg). Similar weight differences were observed forR.e. evertsi females which fed on rabbits previously infested three times with male ticks. The mean weight of the female ticks which fed on these rabbits was 520.1 (±29.8) mg compared with 640.7 (±30.2) mg ofR.e. evertsi females which fed on control hosts. The concentration of gammaglobulins in the sera of rabbits was monitored at various intervals after the first infestation. It was found, for the first time, that infestation of laboratory animals with male ticks conferred immunity, but to a lesser degree than infestation with both sexes. It was also shown that the level of gammaglobulins increased from 3.4±0.28 g l–1 to 7.3±0.24 g l–1 in sera of rabbits hosts as a result of the feeding activity of males, but to a lesser extent than in sera of rabbits on which both sexes had fed (10.8±2.4 g l–1).  相似文献

Ectoparasite and epifaunistic arthropod biodiversity and infestation parameters were compared between 2 sympatric small rodent species, the cotton mouse (Peromyscus gossypinus (Le Conte)) and golden mouse (Ochrotomys nuttalli (Harlan)), in southern Georgia from 1992 to 2003. Because the cotton mouse is known to be a reservoir of more vector-borne zoonotic pathogens than the golden mouse, we hypothesized that it would be parasitized by more ectoparasites that are known to be vectors of these pathogens. Cotton mice (n = 202) were parasitized by 19 species of arthropods, whereas golden mice (n = 46) were parasitized by 12 species. Eleven species of arthropods were recovered from both host species, whereas 7 were recorded only from cotton mice, and 1 species only from golden mice. Infestation prevalences (percent of mice parasitized) were significantly higher for 1 species of arthropod (the tropical rat mite Ornithonyssus bacoti (Hirst)) infesting cotton mice and for 4 species (the flea Peromyscopsylla scotti Fox and the mites Glycyphagus hypudaei Koch, Androlaelaps casalis (Berlese), and Androlaelaps fahrenholzi (Berlese)) infesting golden mice. Mean intensities (mean per infested mouse) were significantly higher for 2 species (the flea Orchopeas leucopus (Baker) and the blacklegged tick Ixodes scapularis Say) infesting cotton mice and for 2 species (G. hypudaei and A. fahrenholzi) infesting golden mice. Ectoparasites that are known to be vectors of zoonotic pathogens were significantly more common on cotton mice than on golden mice. These ectoparasites included the rhopalopsyllid flea Polygenis gwyni (Fox), a vector of the agent of murine typhus; I. scapularis, the principal vector of the agents of Lyme borreliosis, human granulocytic ehrlichiosis, and human babesiosis; and O. bacoti, a laboratory vector of several zoonotic pathogens. However, 2 species of ixodid ticks that can transmit zoonotic pathogens were recovered from both host species. These were the American dog tick Dermacentor variabilis (Say), the principal vector of the agent of Rocky Mountain spotted fever in eastern North America, and Ixodes minor Neumann, an enzootic vector of the agent of Lyme borreliosis. Overall, the cotton mouse was parasitized by significantly more ectoparasites that are known to be vectors of zoonotic pathogens than was the golden mouse. These data support the hypothesis that the cotton mouse has greater epidemiological importance for zoonotic vector-borne pathogen transmission than does the golden mouse.  相似文献
