We examined the linkages between plant functional group richness and microbial community structure and functions in a full-scale vertical flow constructed wetland (VFCW), where five plant functional group richness levels (including zero (control), one, two, three and four functional groups) were applied. Most diagnostic phospholipid fatty acid (PLFA) abundances and enzyme activities were higher in planted treatments than in the control (p < 0.05). Among the diagnostic PLFAs determined, only the fungal PLFA (18:2ω6,9) abundance was related to plant functional group richness level (p < 0.05). For the enzyme activities determined, dehydrogenase, invertase, urease and acid phosphatase activities responded positively to plant functional group richness (p < 0.05). Redundancy analysis (RDA) identified that the Gram-positive and Gram-negative bacteria, actinomycetes and fungi abundances affected enzyme activities. Principal component analyses (PCAs) revealed that the enzyme profiles had greater resolving power in distinguishing plant functional group richness than the PLFA profiles. We conclude that plant functional group richness was closely linked to microbial community structure and functions in the full-scale VFCW we studied.

The effects of the heavy metals Cd and Pb on the activity of the enzyme ferric chelate reductase (FC-R, E.C. 1.6.99.13) have been studied in excised sugar beet root tips. The activity of this enzyme is markedly increased by iron deficiency. Metals were used as chloride salts or chelated with EDTA, and chemical speciation was carried out to predict the metal chemical species in equilibrium both in the ferric reductase assay and in the nutrient solutions. Three different heavy metal treatments were used. First, effects of Cd and Pb on the functioning of the FC-R were assessed in Fe-deficient plants, by including metals in the enzyme assay medium only. Results indicate that 50 μM CdCl2 or Cd-EDTA did not affect FC-R activities even when assay time was as long as 2 h, whereas Pb slightly decreased enzyme activity only at concentrations of 2 mM. Second, short-time Cd and Pb pre-treatments (30–60 min) were imposed on intact Fe-deficient plants before carrying out the assay of FC-R activity. These short-term treatments induced significant decreases in the FC-R activities previously induced by Fe deficiency. With Cd, effects were more pronounced at higher concentrations, and they were stronger when Cd was in the free ion form than when present in the form of Cd-EDTA chelate. Third, prolonged Cd and Pb treatments were imposed on plants grown on 45 μM Fe-EDTA to assess the long-term effects of heavy metals on the induction of the FC-R enzyme. These long-term heavy metal treatments caused a significant increase in the root FC-R activities, indicating that Cd and Pb induce a deficiency in Fe in sugar beet that in turn elicits FC-R activity. The increases, however, are not as large as those found in total absence of Fe.

Abstract

This study was performed heavy metals (As, Cd, Cr, and Pb) in water, soil and frogs around an electronic-waste dump site. The bioaccumulation factors (BAFs) of heavy metals in three frog species and potential human health risks were assessed. Heavy metals were analyzed using inductively coupled plasma-optical emission spectrometry. The Cd and Pb concentrations in water samples and As and Pb concentrations in soil samples from within the e-waste dump site exceeded the standards. The heavy metal concentrations in the muscles of three frog species were as follows: Cr?>?Pb?>?As?>?Cd, and there were no significant differences among frog species except in the case of Pb (p?<?0.05). Only the Cr concentrations exceeded the food quality standards. The relative order of the BAFs for heavy metals in frogs as a result of uptake from the water and soil was Cr?>?As?>?Pb?>?Cd and Cr?>?As?>?Cd?>?Pb, respectively, which indicated that the uptake from water was greater than that from the soil. The assessment of the health risk index and carcinogenic risk (CR) indicated potential human health effects from As, Cr, and Pb via the consumption of frogs.

Although potentially harmful effects of heavy metals are well known, limited numbers of studies exist regarding their relationship with autism. The aim of this study was to investigate urine levels of some heavy metals such as of chromium (Cr), cadmium (Cd), and lead (Pb) in children with autism and healthy subjects. Urine levels of Cr, Cd, and Pb were measured by atomic absorption spectrometry in 30 children with autism and compared with 20 healthy controls. Urine Cd and Pb levels were found as significantly decreased in children with autism compared to healthy subjects (p?<?0.05). On the other hand, urine Cr levels were significantly higher in children with autism than healthy subjects (p?<?0.05). This study suggested that autism may be associated with significant decrease in excretion rate of Cd and Pb and a significant increase excretion rate in the levels of Cr in the urine. These results have indicated that further studies are warranted for investigation of possible roles of heavy metals in autism.

Four gramineous energy plants, Miscanthus sacchariflorus, M. floridulus, Phragmites australis, and Arundo donax were grown on copper tailings in the field for four years. Their phytoremediation potential was examined in terms of their effects on the fractions of heavy metals and soil enzyme activities. Results showed that plantation of these four gramineous plants has improved the proportion of organic material (OM)-binding fraction of heavy metals in copper tailings as a whole, and reduced the proportion of exchangeable and residual fractions. In particular, M. sacchariflorus growth improved significantly the proportion of the OM-binding fractions of Cu (1.73 times), Cd (1.71 times), Zn (1.18 times), and Pb (3.14 times) (P < 0.05) and reduced markedly the residual fractions of Cu (64.45%), Cd (82.38%), Zn (61.43%), and Pb (73.41%) (P < 0.05). Except for A. donax, the growth of other three energy plants improved the activity of phosphatase, urease and dehydrogenase in copper tailings to some extent. In particular, the activity of soil phosphatase and urease in planted tailings differed significantly from that of control (P < 0.05). The effect of M. sacchariflorus growth on soil enzyme was the highest, followed by P. australis, M. floridulus, and A. donax. The content of each heavy metal fraction in soil was correlated with soil enzyme activities, especially the content of OM-binding fraction, which correlated significantly with the activities of phosphatase, urease and dehydrogenase in soil. According to the effects of four gramineous plants growth on activity of soil enzymes and fractions of heavy metals, M. sacchariflorus had the optimal effects for phytoremediation. Therefore, M. sacchariflorus was a candidate plant with great potential for the revegetation of heavy metal tailings.

A thymidine incorporation technique was used to determine the tolerance of a soil bacterial community to Cu, Cd, Zn, Ni, and Pb. An agricultural soil was artificially contaminated in our laboratory with individual metals at three different concentrations, and the results were compared with the results obtained by using the plate count technique. Thymidine incorporation was found to be a simple and rapid method for measuring tolerance. Data obtained by this technique were very reproducible. A linear relationship was found between changes in community tolerance levels obtained by the thymidine incorporation and plate count techniques (r = 0.732, P < 0.001). An increase in tolerance to the metal added to soil was observed for the bacterial community obtained from each polluted soil compared with the community obtained from unpolluted soil. The only exception was when Pb was added; no indication of Pb tolerance was found. An increase in the tolerance to metals other than the metal originally added to soil was also observed, indicating that there was multiple heavy metal tolerance at the community level. Thus, Cu pollution, in addition to increasing tolerance to Cu, also induced tolerance to Zn, Cd, and Ni. Zn and Cd pollution increased community tolerance to all five metals. Ni amendment increased tolerance to Ni the most but also increased community tolerance to Zn and, to lesser degrees, increased community tolerance to Pb and Cd. In soils polluted with Pb increased tolerance to other metals was found in the following order: Ni > Cd > Zn > Cu. We found significant positive relationships between changes in Cd, Zn, and Pb tolerance and, to a lesser degree, between changes in Pb and Ni tolerance when all metals and amendment levels were compared. The magnitude of the increase in heavy metal tolerance was found to be linearly related to the logarithm of the metal concentration added to the soil. Threshold tolerance concentrations were estimated from these linear relationships, and changes in tolerance could be detected at levels of soil contamination similar to those reported previously to result in changes in the phospholipid fatty acid pattern (Å. Frostegård, A. Tunlid, and E. Bååth, Appl. Environ. Microbiol. 59: 3605-3617, 1993).

Concentrations and spatial distribution of Zn, Cu, Cd, and Pb along two landscapes including a wastewater-irrigated area and a control area were determined to assess the impact of long-term wastewater irrigation and landscape properties on heavy metal contamination. Some disturbed and undisturbed soil samples were taken from soil trenches and soil cores, located on three main landscape positions (upper slope, midslope, and lower slope) in northwestern Iran. The investigation showed that the mean concentration of the heavy metals followed the order Zn > Cu > Pb > Cd in the wastewater-irrigated soil and Pb > Zn > Cu > Cd in the control soils. On average, compared to similar positions in the control region, the wastewater-irrigated regions contained 3.0 (midslope) to 4.9 (lower slope), 2.7 (midslope) to 4.6 (lower slope), 3.3 (upper slope) to 4.1 (lower slope), and 1.7 (upper slope) to 2.6 (lower slope) times higher amounts of Zn, Cu, Cd, and Pb, respectively. Significant positive relationships (P < 0.05) were recorded between the heavy metals concentration with <0.002 mm particle-size fraction and organic matter content, the fractions linked to runoff and soil erosion. It is believed that the two soil fractions play a crucial role in the distribution of the metals along the wastewater-irrigated landscape. Despite the significant increase of heavy metals (P < 0.05) in the wastewater-irrigated soils compared with control soils, the concentration of all evaluated metals was below the maximum accepted limits (Zn < 300 mg/kg, Cu < 100 mg/kg, Cd < 5 mg/kg, and Pb < 100 mg/kg), and grouped as “not-enriched” to “moderately-enriched” categories regarding the topsoil enrichment index. Overall, the lower slope was shown to be more contaminated with the heavy metals compared to the other positions.

Twenty five water samples were collected along the Taizihe River, the concentration and health risks of Zn, Cu, Pb, Cr and Cd were detected and evaluated, and the pollution sources was analyzed through principal components analyses. The results indicated that the order of average concentration of heavy metals was follows: Pb > Cr > Cu > Zn and Cd. Among that, the concentrations of Zn, Cu and Cr were at the permissible levels, but Pb and Cd exceeded grade V standard at some sites. The concentrations of Zn and Cu in the wet season were significant higher than that in the dry season (p < 0.05), but the average concentrations of Pb, Cr and Cd were not significantly different in the two seasons (p > 0.05). The annual average risks of human health caused by Cd and Cr were 10−3/a and 10−4/a, respectively, which were higher than the recommended maximum acceptable risk level. The human health risk values of Zn, Pb and Cu were all concentrated at 10−8/a or 10−9/a levels, which did not exceed the recommended standard. On the whole, Cd and Cr were the main health risk pollutants of Taizihe River. Pollution sources of Pb was different from other heavy metals in wet and dry season, Cd and Cr were similar in the wet and dry season. The mainly pollution source of heavy metals was industry, especially mining, metal smelting and electroplating industry.

Abstract

Using biodegradable chelators to assist in phytoextraction may be an effective approach to enhance the heavy-metal remediation efficiencies of plants. A pot experiment was conducted to investigate the effects of ethylenediamine disuccinic acid (EDDS), citric acid (CA), and oxalic acid (OA) on the growth of the arsenic (As) hyperaccumulator Pteris vittata L., its arsenic (As), cadmium (Cd), and lead (Pb) uptake and accumulation, and soil microbial responses in multi-metal(loid)-contaminated soil. The addition of 2.5-mmol kg?1 OA (OA-2.5) produced 26.7 and 14.9% more rhizoid and shoot biomass, respectively compared with the control, while EDDS and CA treatments significantly inhibited plant growth. The As accumulation in plants after the OA-2.5 treatment increased by 44.2% and the Cd and Pb accumulation in plants after a 1-mmol kg?1 EDDS treatment increased by 24.5 and 19.6%, respectively. Soil urease enzyme activities in OA-2.5 treatment were significantly greater than those in the control and other chelator treatments (p?<?0.05). A PCR–denatured gradient gel electrophoresis analysis revealed that with the addition of EDDS, CA and OA enhanced soil microbial diversity. It was concluded that the addition of OA-2.5 was suitable for facilitating phytoremediation of soil As and did not have negative effects on the microbial community.

Microbial community composition and activity were characterized in soil contaminated with lead (Pb), chromium (Cr), and hydrocarbons. Contaminant levels were very heterogeneous and ranged from 50 to 16,700 mg of total petroleum hydrocarbons (TPH) kg of soil−1, 3 to 3,300 mg of total Cr kg of soil−1, and 1 to 17,100 mg of Pb kg of soil−1. Microbial community compositions were estimated from the patterns of phospholipid fatty acids (PLFA); these were considerably different among the 14 soil samples. Statistical analyses suggested that the variation in PLFA was more correlated with soil hydrocarbons than with the levels of Cr and Pb. The metal sensitivity of the microbial community was determined by extracting bacteria from soil and measuring [3H]leucine incorporation as a function of metal concentration. Six soil samples collected in the spring of 1999 had IC50 values (the heavy metal concentrations giving 50% reduction of microbial activity) of approximately 2.5 mM for CrO42− and 0.01 mM for Pb2+. Much higher levels of Pb were required to inhibit [14C]glucose mineralization directly in soils. In microcosm experiments with these samples, microbial biomass and the ratio of microbial biomass to soil organic C were not correlated with the concentrations of hydrocarbons and heavy metals. However, microbial C respiration in samples with a higher level of hydrocarbons differed from the other soils no matter whether complex organic C (alfalfa) was added or not. The ratios of microbial C respiration to microbial biomass differed significantly among the soil samples (P < 0.05) and were relatively high in soils contaminated with hydrocarbons or heavy metals. Our results suggest that the soil microbial community was predominantly affected by hydrocarbons.

The present study was conducted to assess the suitability of sewage sludge amendment (SSA) in soil for Beta vulgaris var. saccharifera (sugar beet) by evaluating the heavy metal accumulation and physiological responses of plants grown at a 10%, 25%, and 50% sewage sludge amendment rate. The sewage sludge amendment was modified by the physicochemical properties of soil, thus increasing the availability of heavy metals in the soil and consequently increasing accumulation in plant parts. Cd, Pb, Ni, and Cu concentrations in roots were significantly higher in plants grown at 25% as compared to 50% SSA; however, Cr and Zn concentration was higher at 50% than 25% SSA. The concentrations of heavy metal showed a trend of Zn > Ni > Cu > Cr > Pb > Cd in roots and Zn > Cu > Ni > Cr > Pb > Cd in leaves. The only instance in which the chlorophyll content did not increase after the sewage sludge treatments was 50%. There were approximately 1.12-fold differences between the control and 50% sewage sludge application for chlorophyll content. The sewage sludge amendment led to a significant increase in Pb, Cr, Cd, Cu, Zn, and Ni concentrations of the soil. The heavy metal accumulation in the soil after the treatments did not exceed the limits for the land application of sewage sludge recommended by the US Environmental Protection Agency (US EPA). The increased concentration of heavy metals in the soil due to the sewage sludge amendment led to increases in heavy metal uptake and the leaf and root concentrations of Ni, Zn, Cd, Cu, Cr, Pb, and Zn in plants as compared to those grown on unamended soil. More accumulation occurred in roots and leaves than in shoots for most of the heavy metals. The concentrations of Cd, Cr, and Pb were more than the permissible limits of national standards in the edible portion of sugar beet grown on different sewage sludge amendment ratios. The study concludes that the sewage sludge amendment in the soil for growing sugar beet may not be a good option due to risk of contamination of Cr, Pb, and Cd.

The present study was designed to assess the effects of three different metals (cadmium, lead, and zinc) at the same concentrations on Hibiscus rosa sinensis during metal uptake. The effects of different metals at the same concentrations were assessed on biomass, root-shoot length, and biochemical parameters like chlorophyll and antioxidant enzymes like SOD and CAT to establish the tolerance potential and toxic effects on plants in different metals. The accumulation of metals by plants was found to be in the following order: Zn > mixed metals > Cd > Pb, where Zn was accumulated approximately 79.6% in plant tissues. Plants removed from Cd showed more enzyme activities than the other two metals. DNA stability was investigated by a Random Amplified Polymorphic DNA (RAPD) technique, which demonstrated that the samples in Cd and mixed metal showed similar trends, whereas samples in Zn and Pb showed similar band intensity to the control. Results suggested that Cd and/or heavy metal stress influences antioxidant status and also induces DNA changes during remediation. Therefore, these studies could be a useful biomarker assay for better treatment for metals’ remediation from soil by means of phytoremediation.

High-throughput 16S rRNA and 18S rRNA sequencing were performed to study the changes of soil microbial diversity and community structure under different heavy metal pollution levels in Chengxian lead–zinc mining area, Gansu Province. In this study, we characterized the main physicochemical properties, multiple heavy metal pollution, and microbial community structure of the soil in the tailings. The results show that the soil near the tailings pond was alkaline, barren and the heavy metals were seriously polluted. The microbial diversity and richness of S1 and S2 sites were significantly lower than that of CK2 site (P < 0·05), indicating that the heavy metal pollution could change the physicochemical properties and microbial community structure in soil. Among 97 identified core operating taxa of fungal communities, Ascomycota, Teguta and Basidiomycota were dominant at the phylum level, while among 1523 identified core operating taxa of bacterial communities, Actinomycota was dominant at the phylum level. In addition, the redundancy analysis and Spearman correlation analysis showed that the physicochemical properties and the heavy metal concentration had significant effects on the composition and distribution of soil microbial community. The basic characteristics of soil physicochemical properties, multiple heavy metal pollution and microbial community structure in the tailings were revealed, hoping to provide a basis for ecological rehabilitation of tailings by revealing the variance rule of microbial community diversity in the future.

The present study was carried out in natural stands of Typha domingensis in Lake Burullus, Egypt, to investigate (1) nutrient dynamics and heavy metals accumulation in its organs, (2) the phytoextractive potential of its organs and (3) the amount of nutrients and heavy metals released back into the water after decomposition of the dead tissues. Nitrogen concentrations were higher in the shoot than in the root and rhizome, while P, Ca, Cu, Fe, Zn and ash concentrations were higher in the root than in the rhizome and shoot. Significant differences in the concentrations of Mg, Cd, Cu and ash were assessed during the growing season of T. domingensis. The content of most nutrients and heavy metals in the shoot increased rapidly during the early growing season in February, reached maximal values in July and then decreased again. The nutrient and heavy metal contents in the below-ground portion of the plant showed an opposite trend compared to the shoot; they decreased sharply during the spring, when they were translocated, supporting the heterotrophic phase of shoot growth. However, they increased slightly from July to September and then decreased again. The transfer factors of all nutrients and heavy metals from the sediment to the below-ground organs were greater than unity. The higher translocation ratio of N in T. domingensis shoots makes it suitable for N phytoextraction from water and sediment, while the lower translocation ratios for Cd, Cu, Fe, Pb and Zn make it suitable for metal ion phytostabilisation. The dead shoot biomass of the stands at the end of 2010 amounted to 1950 g DM m−2, when the seasonal decomposition process began. With a decay rate of 0.0049 day−1, 1624 g DM m−2 is decomposed in the lake in a year. This is equivalent to releasing the following nutrient and heavy metals into the surrounding water (in g m−2): 23.4 N, 0.8 P, 19.2 Ca, 1.8 Mg, 5.6 Na, 32.8 K, 0.01 Cd, 0.01 Cu, 0.84 Fe, 0.12 Pb and 0.03 Zn.

Anthropogenic metal pollutants bioaccumulated in benthic animals by means of feeding and osmotic diffusion. These metals may affect the physiology of the benthos. In this study, we exposed Capitella sp. I to three metals (Cd, Pb, and Ni), each in eight different concentrations, to determine the effects of metals on the animals. Growth rate, ingestion rate, and percent survival were estimated in three separated experiments. The growth and feeding of the worms were sensitive to even the lowest concentrations of each metal added to the sediments. The lowest observable adverse effect levels for Cd, Ni, and Pb were 0.03, 1.59, and 0.41 μmol g 1 sediment, respectively. Growth rates in the elevated metal contaminant treatments decreased drastically at slightly contaminated levels, lessened detrimental effects at moderately contaminated levels, and showed incompensable intoxication at heavily contaminated levels. The trends in ingestion rates were similar to those of growth rates. No significant difference in survivorship was found among the different contaminant levels for any of the three heavy metals. Capitella sp. I was most sensitive to Cd, followed by Ni and Pb, which had similar effects. The rapid physiological responses of Capitella sp. I allowed the animals to survive metal exposure. Sediment productivity remained unchanged at different contamination levels of Ni and Pb, but was drastically reduced at 4.75 μmol g 1 Cd in the sediment. This further demonstrated Capitella sp. I can adjust their ingestion rates to maintain constant sediment productivities in moderate pollution conditions; however, when threshold concentration was exceeded, homeostasis collapsed.

To assess the extent and potential hazards of heavy metal pollution at Shanghai Laogang Landfill, the largest landfill in China, surface soil samples were collected near the landfill and concentrations of Cu, Zn, Cd, Pb, and Cr were determined. The results revealed that the concentrations of heavy metals, except Pb, were higher in the surface soil near the landfill than in the background soil. Principal component analysis and hierarchical cluster analysis suggested that the enrichment of Cu in soil was probably related to agricultural activities and Cd and Pb to landfill leachates, whereas Zn and Cr concentrations were probably controlled by soil matrix characteristics. The pollution indices (PIs) of the metals were: Cd > Cu > Cr > Zn > Pb. Among the five measured metals, Cd showed the largest toxic response and might cause higher ecological hazards than other metals. The integrated potential eco-risk index (RI) of the five metals ranged from 26.0 to 104.9, suggesting a low-level eco-risk potential. This study indicated the accumulations of Cu, Zn, Cd, Pb, and Cr did not reach high pollution levels, and therefore posed a low eco-risk potential in surface soil near the landfill.
