Summary Formation constants for the calcium(II), magnesium(II) and zinc(II) complexes of the orally effective iron chelator, pyridoxal isonicotinoyl hydrazone (PIH) and three analogues, pyridoxal benzoyl hydrazone (PBH), pyridoxalp-methoxybenzoyl hydrazone (PpMBH) and pyridoxalm-fluorobenzoyl hydrazone (PmFBH) have been determined by potentiometry at 25\dg C andI=0.1 M [KNO3]. The four ligands bind calcium(II) weakly and magnesium(II) only slightly more strongly, as a l: l complex which is formed at pH \s> 8. The chelation of zinc(II) for all the ligands studied was greater than that for calcium(II) and magnesium(II), with complexation generally becoming significant at about pH 5. Thus, chelation of zinc(II) but not calcium(II) or magnesium(II) at physiological pH, 7.4 may be expected. Calculated values of the concentration of uncomplexed metal ion indicate that the selectivity of these ligands towards Fe(III) is comparable to that of the clinically used chelator desferrioxamine.  相似文献

Cations, including calcium, magnesium, potassium, sodium, copper, iron, nickel and zinc, inhibited (up to 40%) extracellular binding and intracellular uptake of cadmium by Lemna polyrhiza in solution culture. Test plants showed a high capacity of extracellular cadmium binding which was competitively inhibited by copper, nickel and zinc; however, calcium, magnesium and potassium caused non-competitive inhibition. Iron and sodium increased K m and decreased V max, thereby causing mixed inhibition of extracellular binding. Intracellular cadmium uptake displayed Michaelis-Menten kinetics. It was competitively inhibited by calcium, magnesium, iron, nickel and zinc. Monovalent cations (sodium and potassium) caused non-competitive and copper caused mixed inhibition of intracellular cadmium uptake. Thus, high levels of cations and metals in the external environment should be expected to lower the cadmium accumulation efficiency of L. polyrhiza.  相似文献

The solubilities of zinc, iron, copper, magnesium, calcium, inorganic phosphate, and citrate in milk decreased when acidic milk preparations were neutralized. In decaseinated bovine milk soluble zinc, iron, and copper were reduced 90%, 60%, and 50%, respectively, as the pH was raised from 4 to 7. Simultaneous precipitation of minerals and citrate was confirmed by analysis of washed precipitate. We propose that the diminished solubilities of zinc, iron, copper, magnesium, and citrate are linked to the precipitation of calcium phosphate through one or more mechanisms of coprecipitation. Such control on mineral solubility may have an impact upon mineral absorption from milk.  相似文献

A study was made of certain cations present in hemolymph and alimentary tract tissues of healthy and diseased European chafer larvae and the spores of Bacillus popilliae collected from diseased hemolymph. The major ions found in the hemolymph, in order of decreasing abundance, were potassium, magnesium, sodium, and calcium. Hemolymph of diseased larvae contained relatively higher concentrations of sodium, potassium, magnesium, iron, and zinc than hemolymph of healthy larvae. Concomitantly, the concentrations of ions were lower in the mid-gut and anterior intestinal tissues of diseased larvae. Only sodium decreased slightly in the diseased tissues of the rectum and rectal sac; other ions remained unchanged or increased. Little or no manganese or copper was detected in the hemolymph or tissues. The major cations of spores of B. popilliae were sodium, calcium, and magnesium. Small amounts of potassium, manganese, copper, iron, and zinc were detected in the spores. Based on calcium and dipicolinic acid determinations of the spores, sufficient calcium was found to allow for the formation of calcium dipicolinate in the expected concentrations.  相似文献

Nonalcoholic fatty liver disease (NAFLD) is the most common chronic liver disease. Its incidence is rising worldwide. However, no specific therapy has been shown to be effective in its treatment. In the present study, the in vitro NAFLD model was established in human SMMC-7721 cells by using oleic acid (OA). Then, content changes of eight cations, including sodium, magnesium, potassium, calcium, iron, copper, zinc, and manganese, were investigated in the experimental model. The results showed that OA induced a decrease in magnesium level, while an increase in iron one. Additionally, the supplementation of magnesium in the cell culture model was studied. It showed that magnesium ameliorated lipid accumulation induced by OA. Our results suggest that magnesium could decrease the risk of NAFLD and be used as a promising candidate for the treatment of NAFLD.  相似文献

The elemental composition of plant tissue culture media was studied in response to (1) different levels of Gelrite and activated carbon (AC) in semisolid media and (2) different levels and types of AC in liquid media. Doubling the level of Gelrite from 2 g/l to 4 g/l reduced available magnesium (20%), calcium (16%), zinc (17%) and manganese (24%) and increased potassium (6%). AC adsorbed copper (90-95%) and zinc (35-51%) from both liquid and semisolid media. Two significantly different ACs gave minor differences in adsorption. No adsorption was indicated for inorganic anions. Nonacid-washed AC released significant levels of magnesium (44% increase), calcium (16% increase) and silica (a 75% increase to 1.8 mg/l). The elemental composition of media may need to be adjusted when increasing the Gelrite level or adding AC.  相似文献

The serum amyloid P component (SAP) has been found to associate in vitro with a variety of polysaccharide and proteinaceous ligands including the yeast cell wall polysaccharide preparation, zymosan, in the presence of calcium at neutral pH. In the present study, we have investigated the role of copper and zinc and other divalent cations and acidic pH on the binding of SAP to zymosan. We report that binding occurs not only in the presence of calcium, but in the presence of copper, zinc, and cadmium as well. No binding occurs in the absence of added metal, or in the presence of barium, cobalt, magnesium, manganese, or nickel. 125I-SAP binding in the presence of metals is inhibited by presaturating the zymosan surface with unlabeled SAP. Whereas calcium-mediated binding decreases by more than 50% as the pH is lowered to 5, copper-mediated binding increases substantially at the more acidic pH values while zinc-mediated binding is essentially unchanged. These data indicate that, in addition to calcium at neutral pH, copper (and zinc) at neutral and particularly acidic pH values mediates SAP binding to polysaccharide ligands. This suggests that SAP may well be considered a copper- as well as a calcium-dependent protein under certain conditions and that this reactivity is favored under those conditions of lowered pH which may result from metabolic processes occurring at local sites of inflammation.  相似文献

The aim of this study was to determine the zinc, iron, copper, calcium, phosphorus, and magnesium levels in blood serum and zinc and copper levels in hair of dogs with canine visceral leishmaniasis. The serum zinc and iron levels were found to be significantly lower in diseased dogs than those of healthy controls. Serum copper levels were significantly higher, whereas no significant differences were observed for calcium, phosphorus, and magnesium. There were no significant differences in the zinc and copper levels in hair. Our results show that the serum zinc, iron, and copper levels are altered in canine leishmaniasis.  相似文献

Ehrlich ascites-tumour cells normally contain a large concentration of Zn-metallothionein. When cells are placed in culture media, containing or pretreated with the metal-ion-chelating resin Chelex-100, they stop growing, remain viable and lose zinc specifically from the metallothionein (MT) pool. The kinetics of loss of zinc are first-order and are very rapid, having a rate constant of greater than or equal to 0.6 h-1. MT protein labelled with 35S is biodegraded with a rate constant of 0.07-0.014 h-1 in control cells, 0.08 h-1 in cells exposed to the zinc-deficient medium and 0.12-0.18 h-1 in cells treated directly with Chelex. Over the 6 h period in which zinc is totally lost from Zn-MT there is relatively little decrease in MT-like protein as measured by cadmium-binding to the 10,000-Mr protein fraction. Other pools of zinc and 35S-labelled protein turn over more slowly. There is no loss of zinc from rat liver Zn-MT that is dialysed against Chelex to model the possible reaction of the resin with Ehrlich-cell Zn-MT. However, Chelex does compete slowly for MT-bound zinc when resin and MT are directly mixed. Analysis of the known and possible pathways of zinc metabolism in cells in relationship to these rate constants shows that biodegradation of MT protein cannot account for the rate of loss of zinc from Zn-MT.  相似文献

Removal of adventitious redox-active metals from buffers by treatment with Chelex resin is a widely used procedure in free radical research. Use of a new batch of Chelex-100 resin in our laboratory coincided with a sudden inability to oxidise low-density lipoprotein with copper. We found that copper-mediated oxidation of ascorbate in water treated with the same batch of Chelex was inhibited when compared with untreated water and water treated with a different batch of the resin. Washing the Chelex removed the inhibitory effect suggesting that material was leaching from the resin. The washing procedure for Chelex-100 described is simple and can be scaled up. Oxidation of ascorbate with low concentrations of copper can be used to test the quality of batches of the resin.  相似文献

Summary Two experiments on cacao seedlings grown in sand culture are described, the first of which was concerned with variations in the levels of nitrogen, phosphorus, potassium, calcium, magnesium and sulfur (the macronutrient experiment) and the second dealing with variations in the levels of iron, copper, zinc, boron, manganese, and molybdenum (the micronutrient experiment).Many of the deficiency symptoms obtained were similar to those reported in the literature and they have not been described again. However, additional information is provided for symptoms of phosphorus, potassium, calcium, iron, manganese, copper, boron, and molybdenum deficiencies.The effects of all treatments on the dry weights of leaves, stems, and roots are presented. The effects of the macronutrient treatments on the levels of nitrogen, phosphorus, potassium, calcium, and magnesium in the leaves of eight month old plants and the effects of micronutrient treatments on the levels of nitrogen, phosphorus, potassium, calcium, magnesium, iron, manganese, copper, zinc, boron, molybdenum, sodium, and aluminium in the leaves of eleven-month-old plants are presented and discussed.  相似文献

The effects of atmospheric pollution on two 10 year old Pinus radiata D. Don forests (Manzanal and Posadero) was studied in the Basque country, analysing the concentration of different nutrients and nutrient input via litterfall. Litterfall production peaked in autumn, and 97.5% of the litterfall consisted of pine needles at Posadero (the less-polluted area), whereas this figure was found to be 88.6% at Manzanal (the polluted area). Posadero had a higher production of litterfall than Manzanal due to the higher tree density. Nutrient concentration showed a similar seasonal variation over the year at both sites. Pine litterfall in the polluted area, Manzanal, had significantly higher concentrations of iron, copper, zinc, sulphur, calcium, sodium and magnesium than in Posadero, while the concentrations of nitrogen, phosphorous and potassium did not differ between sites. Calcium, sodium, magnesium, sulphur, iron and copper input to the forest via litterfall was significantly higher for the polluted area, but the inputs of nitrogen, phosphorous, potassium and zinc did not significantly differ between sites. Both sites showed high nutrient use efficiencies for phosphorous and calcium compared with other coniferous forests. The highest efficiency was for phosphorous, possibly due to the low availability of this ion in the soil. Trees at Posadero were consistently more efficient for calcium than at Manzanal due to the effect of pollution on the latter site. Molar ratios between aluminium and the basic cation nutrients calcium and magnesium in soil were higher at the polluted area, as a result of the increased inputs of anions to the forest soil (Al:Ca = 1.9:1 and Al:Mg = 6.9:1 at Posadero and Al:Ca = 5.7:1 and Al:Mg = 14:1 at Manzanal). The current pollution level of Manzanal is leading to a loss of cations that may cause future changes in the functioning of this forest.  相似文献

The effects of pH and ferrous iron concentration in cultural medium on the bioleaching of copper-zinc concentrate by mesophilic and moderately thermophilic acidophilic microorganisms were studied. It was revealed that the optimum pH for bioleaching in presence of 5 g/L of ferrous iron was 1.4–1.5. It was shown that bioleaching under optimal conditions led to an increase in the copper content in solid phase from 10.1 to 14% and a decrease in the zinc content from 7.4 to 1.4%. The results of the present work demonstrate that acidophilic microorganisms can be used for treatment of complex sulfide concentrates containing copper and zinc.  相似文献

111In-labeled solid meal was prepared by chelation of 111In with Chelex resin bead. The effect of grinding of normal Chelex bead on 111In chelation and retention in solid meal was evaluated in an in vitro system. The Chelex resin beads were ground in a mortar-pestle to form ground Chelex resin beads. Fine particles were removed by resuspension in distilled water and centrifugation (1000 g). One hundred to 150 μCi of 111In chloride was diluted with 0.1 N HC1 and mixed with 1 g of Chelex resin beads. Unbound 111In was removed by centrifugation (1000 g). The 111In-labeled Chelex resin beads were mixed with fresh egg and 111In-labeled solid meal was prepared by heating until solid. The meals were digested with HCl-pepsin (1.2mg/mL of pepsin in 0.1 N HC1) for 4 h in a stirrer-bath (37 °C). Aliquots were collected at intervals for determination of 111In loss from 111In-labeled solid meal.These results suggest that 111In Chelex resin beads were retained in solid meals at a higher level than normal Chelex resin beads and other 111In-tracers.  相似文献

The efficacy of a chelating agent in binding a given metal in a biological system depends on the binding constants of the chelator for the particular metals in the system, the concentration of the metals, and the presence and concentrations of other ligands competing for the metals in question. In this study, we make a comparison of the in vitro binding constants for the chelator, ethylenediaminetetraacetic acid, with the quantitative urinary excretion of the metals measured before and after EDTA infusion in 16 patients. There were significant increases in lead, zinc, cadmium, and calcium, and these increases roughly corresponded to the expected relative increases predicted by the EDTA-metal-binding constants as measured in vitro. There were no significant increases in urinary cobalt, chromium, or copper as a result of EDTA infusion. The actual increase in cobalt could be entirely attributed to the cobalt content of the cyanocobalamin that was added to the infusion. Although copper did increase in the post-EDTA specimens, the increase was not statistically significant. In the case of magnesium, there was a net retention of approximately 85% following chelation. These data demonstrate that EDTA chelation therapy results in significantly increased urinary losses of lead, zinc, cadmium, and calcium following EDTA chelation therapy. There were no significant changes in cobalt, chromium, or copper and a retention of magnesium. These effects are likely to have significant effects on nutrient concentrations and interactions and partially explain the clinical improvements seen in patients undergoing EDTA chelation therapy.  相似文献

The distribution of iron, copper, zinc, and magnesium in hepatic subcellular fractions of male and female rats treated with 2,3,7,8-tetrachloro-dibenzo-p-dioxin (TCDD) was determined. Animals received 40 μg TCDD per kilogram per day for three days by mouth (PO) or the vehicle and were killed seven or nine days posttreatment. Iron, copper, zinc, and magnesium were determined by atomic absorption spectroscopy. The iron content of liver from female animals was twofold higher than male animals. The administration of TCDD increased the iron content of mitochondria in female and male rats and decreased iron content of microsomes of both sexes. Significant increases occurred in the copper content of whole liver, mitochondria, and cytosol of male rats and in whole liver and cytosol of female rats. Decreases in the copper content of the microsomes of male rats were observed following TCDD treatment; however, TCDD produced no changes in the zinc content of hepatic subcellular fractions of either sex. The magnesium content of female TCDD-treated rats increased in whole liver, mitochondria, and cytosol, while the magnesium content of microsomes was not altered. With respect to the subcellular distribution of iron, copper, zinc, and magnesium, TCDD produces differential effects. The altered distribution of some cations may contribute to the broad range of effects of TCDD.  相似文献

BackgroundZinc is an essential trace element. Both its lack and excess are associated with pathological states. The former is more common and can ensue from the excessive treatment with clinically used iron/copper chelators.Aim and methodThe aim of this work was to prepare a reliable, rapid and cheap method for the screening of zinc chelation. Spectrophotometric assessment using a known zinc indicator dithizone was selected.ResultsInitial screening performed by comparison of spectra of dithizone and its complex with zinc suggested 530 and 570 nm as suitable wavelengths for determination of zinc at pH 4.5 while 540 and 590 nm for pH 5.5–7.5. Additional research showed the lower wavelengths to be more suitable. The sensitivity of the method was always bellow 1 μM with good linearity relationship between absorbance and zinc concentration. The method suitability was confirmed by use of two known zinc chelators, ethylenediaminetetraacetic acid (EDTA) and N,N,N′,N′-tetrakis(2-pyridylmethyl)-1,2-ethylenediamine (TPEN).ConclusionThis method represents a sufficiently precise method for zinc chelation screening usable at pathophysiologically relevant pH conditions. Such method can be employed for both screening of novel zinc chelators and for testing affinity of other metal chelators for zinc.  相似文献

Direct batch addition of sterile Chelex ion-exchange resin to Dubecco's modified Eagle's medium supplemented with 10% fetal calf serum with gentle stirring removed a very wide variety of trace metal ions from the medium to varying extents dependent upon Chelex content (between 0.01 and 4% w/v), exposure time (between 5 min and 10 days) and temperature 4, 25 and 37 °C). Prolonged treatment (10 days) with 4% w/v Chelex at 4°C reduced the concentration of zinc, strontium, aluminum, copper, manganese, nickel and chromium from 100 to 2.7, 12.1, 7.7, 22.6, 13.0, 14.7 and 53.3% of their original concentrations, respectively. Re-supplementation of the metal depleted medium with a defined cocktail of metals restored the growth potential of the medium which was then capable of supporting growth over at least three subcultures without a decrease in fibroblast cell yield, demonstrating its suitability in cell culture studies on trace metal ions.  相似文献

通过向设施土壤中添加解磷菌和秸秆,达到活化土壤中作物所需金属元素的含量,促进作物对中微量元素吸收的目的。试验设置对照、秸秆、解磷菌、解磷菌+秸秆4个处理,在不同时间测试土壤及秧苗中钙、镁、铜、铁、锌等的含量,确定解磷菌及秸秆对设施土壤金属元素含量及黄瓜秧苗吸收量的影响。试验结果表明,秸秆、解磷菌、秸秆+解磷菌处理能明显增加黄瓜秧苗吸收钙、镁、铁、铜的能力,对锌的吸收没有影响,其中秸秆+解磷菌处理对黄瓜秧苗吸收钙、镁、铜影响最大,促进作用分别达到6.26%、8.25%和11.57%,3种处理对设施土壤中有效钙、镁、铁的含量没有影响,但是能明显提高土壤中有效铜、锌的含量,盛果后期有效铜含量分别比对照提高12.19%、15.41%、16.49%,盛果后期有效锌含量分别比对照区提高38.8%、33.26%、56.81%。  相似文献
