Protein refolding using a simple dilution method in a microchannel often led to the formation of protein aggregates, which bound to the microchannel wall, resulting in low refolding yields. To inhibit aggregation and improve refolding yields, an artificial chaperone-assisted (ACA) refolding, which employed detergents and β-cyclodextrin was used. Model proteins, hen egg white lysozyme and yeast α-glucosidase, were successfully refolded in a microchannel. The microscopic observation showed that the ACA method suppressed protein aggregation and facilitated the refolding of lysozyme, whereas significant aggregation was observed when a simple dilution method was employed. The ACA method increased the lysozyme refolding yield by 40% over the simple dilution approach. Similarly, for α-glucosidase, the refolding yield using the ACA method (ca. 50%) was approximately three times compared with the simple dilution method. The ACA refolding method is a suitable approach to use in the refolding of proteins using a microfluidic system.  相似文献

Refolding of GuHCl-denatured α-amylase was investigated using the artificial chaperone-assisted method. Three different cationic detergents (CTAB, TTAB and DTAB) and two nonionic detergents (Tween 80 and Triton X-100) were evaluated as the capturing reagents along with α- and β-CD as the stripping agents. The refolding yields, at a final protein concentration of 0.15 mg/ml, were 82, 71 and 66% in the presence of β-CD and CTAB, TTAB or DTAB, respectively. To improve the refolding yield and to suppress the extent of aggregation, the initial rate of the stripping step was slowed down by maintaining the refolding environment at 4°C for about 3 min followed by raising the temperature to 25°C. Under this thermal procedure, the refolding yield and the extent of aggregation were changed from 82 and 25% at 25°C to 94 and 7% at 4°C, respectively. These findings may assist the activity recovery of recombinant proteins at relatively high concentrations.  相似文献

Periplasmic metal binding protein characterized by high histidine content was cloned from moderate halophile, Chromohalobacter salexigens. The protein, termed histidine-rich metal binding protein (HP), was expressed in and purified from E. coli as a native form. HP bound to Ni- and Cu-loaded chelate columns with high affinity, and Co- and Zn-columns with moderate affinity. Although the secondary structure was not grossly altered by the addition of 0.2–2.0 M NaCl, the thermal transition pattern was considerably shifted to higher temperature with increasing salt concentration: melting temperature was raised by ~20 °C at 2.0 M NaCl over the melting temperature at 0.2 M NaCl. HP showed reversible refolding from thermal melting in 0.2–1.15 M NaCl, while it formed irreversible aggregates upon thermal melting at 2 M NaCl. Addition of 0.01–0.1 mM NiSO4 stabilized HP against thermal melting with high reversibility, while addition above 0.5 mM resulted in irreversible melting due to aggregation.  相似文献

Aggregation of photosynthetic light-harvesting complexes strongly influences their spectroscopic properties. Fluorescence yield and excited state lifetimes of the main light-harvesting complex (LHC II) of higher plants strongly depend on its aggregation state. Detergents are commonly used to solubilize membrane proteins and/or to circumvent their aggregation in aqueous environments. Nonlinear polarization spectroscopy in the frequency domain (NLPF) was performed with LHC II over a wide concentration range of the mild detergent n-dodecyl β-d-maltoside (β-DM). Additionally, conventional absorption-, fluorescence- and circular dichroism-spectra were measured. The results indicate that: (i) conventional spectroscopic techniques are not well suited to investigate aggregation effects. NLPF provides a novel approach to overcome this problem: NLPF spectra display dramatic alterations upon even minor β-DM concentration changes. (ii) Commonly used detergent concentrations (around or slightly above the critical micellar concentration) apparently do not lead to complete trimerization of LHC II. A long-wavelength species in the NLPF spectra (peaking at about 685 nm), indicative of residual aggregation, persists up to DM-concentrations of 0.06%. (iii) High-resolution NLPF spectra indicate the existence of a species with a considerably shortened excited state lifetime. (iv) No indication of denaturation was found even at the highest β-DM concentrations used. (v) A specific change in interaction between certain chlorophyll(s) b and a xanthophyll molecule, probably neoxanthin, was detected upon aggregation as well as at higher β-DM concentrations. The results are discussed with respect to the still elusive mechanism of nonradiative dissipation of excess excitation energy in the antenna system.  相似文献

The denatured monomers of an integral membrane protein OmpF porin were refolded and reassembled into its sodium dodecyl sulfate-resistant trimer in mixtures of n-octyl β-d-glucopyranoside and lipids. Effective reassembly was observed with a yield of 60–70% when the denatured monomers (0.1 mg/mL) were solubilized at 25 °C for 24 h in a refolding medium (pH 6.9) containing 7 mg/mL n-octyl β-d-glucopyranoside, 1 mg/mL sodium dodecyl sulfate and 2–2.5 mg/mL soybean asolectin. The reassembled species was characterized in the presence of sodium dodecyl sulfate by physicochemical methods. Low-angle laser light scattering measurements revealed that the molecular weight of the reassembled species is 115,000 ± 3,500 which corresponds to that of the trimer of this protein. Circular dichroism spectra suggested that the reassembled species is composed of the same β-structure as the native one. Synchrotron radiation small-angle X-ray scattering measurements confirmed that the reassembled species is a trimer that has the same compactness as the native one.  相似文献

l-Arginine was used to suppress the aggregation of recombinant mink and porcine growth hormones in the refolding process from E. coli inclusion bodies by solubilization–dilution protocol at high protein concentration and pH 8.0. The influence of l-arginine concentration on the renaturation yield of both proteins was investigated. l-Arginine effectively suppressed the precipitation of growth hormones during dilution, but did not inhibit soluble oligomers formation. The results of mink and porcine growth hormones purification from 4 g of biomass are presented.  相似文献

Production of natural pertactin for pharmaceutical use is limited by its low abundance. In this study, recombinant pertactin was highly expressed in the form of inclusion bodies in E. coli. However, up to 75% of the soluble turned out as aggregates when refolding by pulse-fed batch dilution. The conceivable route for aggregate formation was proposed as that the C-terminus of partially folded intermediate with a strong hydrophobic core would intertwine with that region of newly added denatured protein, resulting in aggregation between proteins with different folding states. The key factor for prevention of aggregate formation was to improve the synchronization of refolding. For this purpose, flash-batch dilution was conducted at a scale of 5 L and achieved a monomeric refolding yield of above 70%. Aggregates formed were efficiently removed along with impurities by one-step chromatography of Ni-resin. The purity of monomeric pertactin was >98%. An overall yield was 320 mg per liter fermentation liquor with a total recovery of about 59%. The purified protein was characterized by MALDI-TOF, circular dichroism, fluorescence, HPLC and DSC, and showed similar physiochemical properties compared to its natural counterpart. Animal study showed similar immunological responses and antibodies elicited demonstrated a comparable reactivity.  相似文献

Aggregation is a serious obstacle for recovery of biologically active heterologous proteins from inclusion bodies (IBs) produced by recombinant bacteria. E. coli transformed with a vector containing the cDNA for Bothropstoxin-1 (BthTx-1) expressed the recombinant product as IBs. In order to obtain the native toxin, insoluble and aggregated protein was refolded using high hydrostatic pressure (HHP). IBs were dissolved and refolded (2 kbar, 16 h), and the effects of protein concentration, as well as changes in ratio and concentration of oxido-shuffling reagents, guanidine hydrochloride (GdnHCl), and pH in the refolding buffer, were assayed. A 32% yield (7.6 mg per liter of bacterial culture) in refolding of the native BthTx-1 was obtained using optimal conditions of the refolding buffer (Tris–HCl buffer, pH 7.5, containing 3 mM of a 2:3 ratio of GSH/GSSG, and 1 M GdnHCl). Scanning electron microscopy (SEM) showed that that disaggregation of part of IBs particles occurred upon compression and that the morphology of the remaining IBs, spherical particles, was not substantially altered. Dose-dependent cytotoxic activity of high-pressure refolded BthTx-1 was shown in C2C12 muscle cells.  相似文献

Biosynthesis of the (1,3)-β-d-glucan (curdlan) in Agrobacterium sp., is believed to proceed by the repetitive addition of glucosyl residues from UDP-glucose by a membrane-embedded curdlan synthase (CrdS) [UDP-glucose: (1,3)-β-d-glucan 3-β-d-glucosyltransferase; EC 2.4.1.34]. The catalytic module of CrdS (cm-CrdS) was expressed in good yield from a cDNA encoding cm-CrdS cloned into the pET-32a(+) vector, containing a coding region for thioredoxin, and from the Champion™ pET SUMO system that possesses a coding region of a small ubiquitin-related modifier (SUMO) partner protein. The two DNA fusions, designated pET-32a_cm-CrdS and SUMO_cm-CrdS were expressed as chimeric proteins. High yields of inclusion bodies were produced in E. coli and these could be refolded to form soluble proteins, using a range of buffers and non-detergent sulfobetaines. A purification protocol was developed, which afforded a one-step on-column refolding and simultaneous purification of the recombinant 6xHis-tagged SUMO_cm-CrdS protein. The latter protein was digested by a specific protease to yield intact cm-CrdS in high yields. The refolded SUMO_cm-CrdS protein did not exhibit curdlan synthase activity, but showed a circular dischroism spectrum, which had an α/β-type-like conformation. Amino acid sequences of tryptic fragments of the SUMO_cm-CrdS fusion and free cm-CrdS proteins, determined by MALDI/TOF confirmed that the full-length proteins were synthesized by E. coli, and that no alterations in amino acid sequences occurred. A three-dimensional model of cm-CrdS predicted the juxtaposition of highly conserved aspartates D156, D208, D210 and D304, and the QRTRW motif, which are likely to play roles in donor and acceptor substrate binding and catalysis.  相似文献

L-arginine (Arg) is a widely used additive for suppressing protein aggregation during refolding. Systematic screening of Arg analogs provides superior additives that enhance the refolding yield more effectively than Arg. The refolding yield of hen egg lysozyme in the presence of 500 mM L-argininamide (ArgAd) increases 1.7-fold higher than Arg. Thermal unfolding experiments indicate that ArgAd has a greater denaturing effect than Arg. The refolding yield positively relates to the net charge of Arg analogs. Moreover ArgAd was also effective for the refolding of bovine carbonic anhydrase. High potency to increase the refolding yield of ArgAd compared to Arg results from high positive net charge and the denaturing property.  相似文献

This study tested the hypothesis that l-arginine (Arg) may stimulate cell proliferation and prevent lipopolysaccharide (LPS)-induced death of intestinal cells. Intestinal porcine epithelial cells (IPEC-1) were cultured for 4 days in Arg-free Dulbecco’s modified Eagle’s-F12 Ham medium (DMEM-F12) containing 10, 100 or 350 μM Arg and 0 or 20 ng/ml LPS. Cell numbers, protein concentrations, protein synthesis and degradation, as well as mammalian target of rapamycin (mTOR) and Toll-like receptor 4 (TLR4) signaling pathways were determined. Without LPS, IPEC-1 cells exhibited time- and Arg-dependent growth curves. LPS treatment increased cell death and reduced protein concentrations in IPEC-1 cells. Addition of 100 and 350 μM Arg to culture medium dose-dependently attenuated LPS-induced cell death and reduction of protein concentrations, in comparison with the basal medium containing 10 μM Arg. Furthermore, supplementation of 100 and 350 μM Arg increased protein synthesis and reduced protein degradation in both control and LPS-treated IPEC-1 cells. Consistent with the data on cell growth and protein turnover, addition of 100 or 350 μM Arg to culture medium increased relative protein levels for phosphorylated mTOR and phosphorylated ribosomal protein S6 kinase-1, while reducing the relative levels of TLR4 and phosphorylated levels of nuclear factor-κB in LPS-treated IPEC-1 cells. These results demonstrate a protective effect of Arg against LPS-induced enterocyte damage through mechanisms involving mTOR and TLR4 signaling pathways, as well as intracellular protein turnover.  相似文献

In this study, we have investigated the relationship between chaperonin/co-chaperonin binding, ATP hydrolysis, and protein refolding in heterologous chaperonin systems from bacteria, chloroplast, and mitochondria. We characterized two types of chloroplast cpn60 oligomers, ch-cpn60 composed of α and β subunits (α7β7 ch-cpn60) and one composed of all β subunits (β14 ch-cpn60). In terms of ATPase activity, the rate of ATP hydrolysis increased with protein concentration up to 60 μM, reflecting a concentration at which the oligomers are stable. At high concentrations of cpn60, all cpn10 homologs inhibited ATPase activity of α7β7 ch-cpn60. In contrast, ATPase of β14 ch-cpn60 was inhibited only by mitochondrial cpn10, supporting previous reports showing that β14 is functional only with mitochondrial cpn10 and not with other cpn10 homologs. Surprisingly, direct binding assays showed that both ch-cpn60 oligomer types bind to bacterial, mitochondrial, and chloroplast cpn10 homologs with an equal apparent affinity. Moreover, mitochondrial cpn60 binds chloroplast cpn20 with which it is not able to refold denatured proteins. Protein refolding experiments showed that in such instances, the bound protein is released in a conformation that is not able to refold. The presence of glycerol, or subsequent addition of mitochondrial cpn10, allows us to recover enzymatic activity of the substrate protein. Thus, in our systems, the formation of co-chaperonin/chaperonin complexes does not necessarily lead to protein folding. By using heterologous oligomer systems, we are able to separate the functions of binding and refolding in order to better understand the chaperonin mechanism.  相似文献

Developing novel methanol-feeding strategies for the improved production of heterologous proteins in high cell-density fed-batch cultures of Pichia pastoris has been of great interest during recent years. In this study, a recombinant P. pastoris strain (GS115/His+ Mut+) producing β-galactosidase (β-Gal) was used to investigate conventional feeding strategies and to develop a new strategy to increase the recombinant protein production during fedbatch cultures on methanol. Three types of conventional methanol-feeding strategies, including μ-stat, dissolved oxygen-stat (DO-stat) and constant methanol concentration were investigated and compared with respect to alcohol oxidase (AOX), formate dehydrogenase (FDH) and β-gal activities, and cell dry weight (CDW), methanol, and formaldehyde concentration variations during the production phase. Methanol feeding with μ-stat 0.025/h exhibited the highest β-gal activity. Supplementing ammonium and magnesium in μ-stat 0.025/h did not affect the cell growth or methanol or formaldehyde concentrations throughout the fermentation but did improved the maximum β-gal activity from 148.2 to 158.1 kU/mL. A new three-step methanol-feeding strategy was developed based on the results obtained from conventional feeding strategies, which started with μ-stat 0.025/h for 5 h, then μ-stat 0.030/h, and finally, was switched to DO-stat when maintaining the DO above 20% air saturation became difficult. Implementation of this new feeding strategy resulted in a CDW of 107.2 ± 0.7 g/L, AOX specific activity of 0.1890 ± 0.0030 U/mg CDW, and β-gal activity of 173.5 ± 2.1 kU/mL after 29 h of fermentation, which shows a 5.6, 29.1, and 15.7% increase in CDW, AOX, and β-gal activity, respectively, compared to that of μ-stat at 0.025/h.  相似文献

This study was undertaken to investigate how the length of the extraction period influences the (1 → 3)-β-d-glucan (β-glucan) yield and also to examine the background concentration of β-glucan as airborne β-glucan in outdoor environments in different seasons and as concentrations in airborne and floor dust in offices. To ensure compatibility between results obtained in different laboratories, it is important to use optimal and standardised methods to extract and quantify β-glucan. In this study, an extraction period of 60 min gave the highest β-glucan yield. The median concentration of β-glucan in 44 floor dust samples was 597 μg g−1 dust. The median concentration of airborne β-glucan in offices was 5.1 ng m−3 in the summer and 2.3 ng m−3 in the winter, and the outdoor median concentration in towns was 6.8 ng m−3. The outdoor airborne concentration of β-glucan was significantly lower in January, November and December than during the rest of year. In July, the median airborne concentration of β-glucan was 14 times higher than in January. Furthermore, the airborne concentration of β-glucan was significantly higher in July than in March, April, May, September and October. In the summertime, we found that the indoor airborne concentration of β-glucan was lower than outdoor concentrations. This is in accordance with measurements of concentrations of airborne pollen and culturable fungal spores showing higher outdoor than indoor concentrations during the summer months.  相似文献

BALB/c-derived tumor cells were transfected with recombinantEscherichia coli β-galactosidase (β-gal) genes which were inserted into IgM heavy chain gene derivatives, leading to expression of the resulting fusion protein in different cellular compartments. A β-gal-specific, major histocompatibility complex (MHC) class I-restricted CD8+CD4 cytotoxic T lymphocyte (CTL) line of BALB/c origin raised against one transfectant expressing cytoplasmic β-gal also lysed transfectants expressing β-gal as membrane-inserted fusion protein, as well as transfectants secreting β-gal. Our data show that MHC class I-restricted CTL can recognize fragments of nonviral cellular proteins, be they expressed as intracellular, membrane-inserted, or secreted products. The findings confirm and extend a hypothesis on the nature of minor histocompatibility (H) antigens formulated earlier.  相似文献

Neotermes koshunensis is a lower termite that secretes endogenous β-glucosidase in the salivary glands. This β-glucosidase (G1NkBG) was successfully expressed in Aspergillus oryzae. G1NkBG was purified to homogeneity from the culture supernatant through ammonium sulfate precipitation and anion exchange, hydrophobic, and gel filtration chromatographies with a 48-fold increase in purity. The molecular mass of the native enzyme appeared as a single band at 60 kDa after gel filtration analysis, indicating that G1NkBG is a monomeric protein. Maximum activity was observed at 50 °C with an optimum pH at 5.0. G1NkBG retained 80% of its maximum activity at temperatures up to 45 °C and lost its activity at temperatures above 55 °C. The enzyme was stable from pH 5.0 to 9.0. G1NkBG was most active towards laminaribiose and p-nitrophenyl-β-d-fucopyranoside. Cellobiose, as well as cello-oligosaccharides, was also well hydrolyzed. The enzyme activity was slightly stimulated by Mn2+ and glycerol. The K m and V max values were 0.77 mM and 16 U/mg, respectively, against p-nitrophenyl-β-d-glucopyranoside. An unusual finding was that G1NkBG was stimulated by 1.3-fold when glucose was present in the reaction mixture at a concentration of 200 mM. These characteristics, particularly the stimulation of enzyme activity by glucose, make G1NkBG of great interest for biotechnological applications, especially for bioethanol production.  相似文献

Although GroE chaperonins and osmolytes had been used separately as protein folding aids, combining these two methods provides a considerable advantage for folding proteins that cannot fold with either osmolytes or chaperonins alone. This technique rapidly identifies superior folding solution conditions for a broad array of proteins that are difficult or impossible to fold by other methods. While testing the broad applicability of this technique, we have discovered that osmolytes greatly simplify the chaperonin reaction by eliminating the requirement for the co-chaperonin GroES which is normally involved in encapsulating folding proteins within the GroEL–GroES cavity. Therefore, combinations of soluble or immobilized GroEL, osmolytes and ATP or even ADP are sufficient to refold the test proteins. The first step in the chaperonin/osmolyte process is to form a stable long-lived chaperonin–substrate protein complex in the absence of nucleotide. In the second step, different osmolyte solutions are added along with nucleotides, thus forming a ‘folding array’ to identify superior folding conditions. The stable chaperonin–substrate protein complex can be concentrated or immobilized prior to osmolyte addition. This procedure prevents-off pathway aggregation during folding/refolding reactions and more importantly allows one to refold proteins at concentrations (~mg/ml) that are substantially higher than the critical aggregation concentration for given protein. This technique can be used for successful refolding of proteins from purified inclusion bodies. Recently, other investigators have used our chaperonin/osmolyte method to demonstrate that a mutant protein that misfolds in human disease can be rescued by GroEL/osmolyte system. Soluble or immobilized GroEL can be easily removed from the released folded protein using simple separation techniques. The method allows for isolation of folded monomeric or oligomeric proteins in quantities sufficient for X-ray crystallography or NMR structural determinations.  相似文献

Sucrose presence and concentration modulated in different ways and to different extents the activity of six plant glycoside hydrolases (PGHs) extracted from apple callus cultures, both in the water soluble fraction (WS-F) and in the NaCl-released fraction (NaCl-F). β-d-Glucosidase activity increased because of sucrose starvation and the addition of sucrose decreased both WS-F and NaCl-F β-d-glucosidase from calli grown in a Murashige and Skoog’s basal medium with (MSH) or without (MS0) plant growth regulators (PGRs). WS-F and NaCl-F α-l-arabinofuranosidase, NaCl-F β-d-galactosidase and NaCl-F β-d-xylosidase activity reached a maximum when 0.045 M sucrose was added to the MS0 medium with an ensuing decline at higher sucrose concentrations. α-d-Galactosidase and α-d-xylosidase activity reached a maximum when 0.045 M sucrose was supplied and did not decline significantly in 0.09 M sucrose-supplied calli. When the effects of PGR presence or absence were analysed, NaCl-F β-d-glucosidase, α-d-galactosidase, β-d-galactosidase, α-d-xylosidase and β-d-xylosidase activities were found to be higher in MS0 than in MSH. To assess whether sugar effects were sucrose-specific, other sugars (glucose, fructose, galactose, maltose, lactose, raffinose, sorbitol and mannitol) were tested, with or without PGR supplementation. In general, sugar alcohols (mannitol, sorbitol) and some monosaccharides (fructose and glucose in particular) were better inducers of NaCl-F α-l-arabinofuranosidase, β-d-galactosidase and β-d-xylosidase activity than disaccharides (sucrose, maltose, and lactose) or the trisaccharide raffinose. This trend was not widespread to all PGHs assessed since sucrose-supplemented calli displayed higher NaCl-F α-d-galactosidase than those supplemented with glucose, galactose, sorbitol or mannitol. These results show that sugars supplied to callus tissue cultures as a carbon source can also modulate PGH activity. Modulation is different for each PGH, sugar-specific and, at least in the case of sucrose, concentration-dependent. Results also suggest the existence of regulatory interactions between PGRs and sugars as part of an intricate sensing and signalling network. Combination of PGR, sugar type and concentration should be taken into account to maximize each PGH activity for further enzyme studies.  相似文献

Since red pine (Pinus densiflora Sieb. et Zucc.) often forms sparse forest floors where herbaceous plants do not grow well, allelopathy of red pine was investigated. A growth inhibitory substance was isolated from an aqueous methanol extract of red pine needles and determined by spectral data as abscisic acid-β-d-glucopyranosyl ester (ABA-GE). This substance inhibited root and shoot growth of cress and E. crus-galli seedlings at concentrations greater than 0.1 μM. The concentrations required for 50% growth inhibition on roots and shoots of cress were 0.23 and 0.61 μM, respectively, and those of E. crus-galli were 1.1 and 2.8 μM, respectively. The activity of ABA-β-d-glucosidase, which liberates free ABA from ABA-GE, in cress and E. crus-galli seedlings was 13–29 nmol mg−1 protein min−1. Endogenous concentration of ABA-GE in the pine needles was 4.1–21.5 μmol kg−1 and the concentration in soil water of the pine forest was 2.5 μM. The effectiveness of ABA-GE on growth inhibition and the occurrence of ABA-GE in pine needles and soil water suggest ABA-GE may play an important role in the allelopathy of red pine resulting in the formation of sparse forest floors.  相似文献

Human muscle creatine kinase (CK) is an enzyme that plays an important physiological role in the energy metabolism of humans. It also serves as a typical model for studying refolding of proteins. A study of the refolding and reactivation process of guanidine chloride-denatured human muscle CK is described in the present article. The results show that the refolding process can be divided into fast and slow folding phases and that an aggregation process competes with the proper refolding process at high enzyme concentration and high temperature. An intermediate in the early stage of refolding was captured by specific protein molecules: the molecular chaperonin GroEL and alpha(s)-casein. This intermediate was found to be a monomer, which resembles the "molten globule" state in the CK folding pathway. To our knowledge, this is the first monomeric intermediate captured during refolding of CK. We propose that aggregation is caused by interaction between such monomeric intermediates. Binding of GroEL with this intermediate prevents formation of aggregates by decreasing the concentration of free monomeric intermediates, whereas binding of alpha(s)-casein with this intermediate induces more aggregation.  相似文献
