Selfish genetic elements called transposons can insert themselves at new locations in host genomes to modify gene structure and alter gene expression. Expansion of transposons can occur when novel transposition events are transmitted to subsequent generations after germline hopping. Therefore, organisms seem likely to have evolved defense mechanisms to silence transposons in the germline. Recently, small RNAs interacting with Piwi proteins (piwi-interacting RNAs: piRNAs) have been demonstrated to be involved in genomic defense mechanism against transposons. Here, we show that piRNA-like small RNAs are present abundantly in the Bombyx ovary. We cloned 38,493 kinds of Bombyx small RNA from the ovary and performed functional characterization. Bombyx small RNAs showed a unimodal length distribution with a peak at 28nt and a strong bias for U at the 5' end. We found that 12,869 kinds of Bombyx small RNAs were associated with transposons or repetitive sequences. We classified them as repeat-associated small interfering RNAs (rasiRNAs), a subclass of piRNAs. Notably, antisense rasiRNAs have a strong bias toward U at 5' ends; in contrast, sense rasiRNAs have a strong bias toward A at nucleotide position 10, indicating that the piRNA amplification loop proposed in Drosophila is evolutionarily conserved in Bombyx. These results suggest that Bombyx small RNAs regulate transposon activity.  相似文献

[目的]Fox家族蛋白是调控动物发育的关键转录因子.本文分析了鳞翅目昆虫家蚕Bombyxmori的BmFoxG亚家族蛋白在精巢发育中的作用,为基于生殖系统的家蚕性状优化或害虫不育技术的开发提供理论依据.[方法]利用PCR克隆家蚕BmFoxG-1 和 BmFoxG-2基因;通过生物信息学工具对BmFoxG蛋白的结构及理化性质进行分析;采用qPCR技术检测BmFoxG基因在家蚕不同发育阶段的精巢中的表达变化;在家蚕Bm12细胞株中过表达BmFoxG-1,并通过qRT-PCR分析BmFoxG-1调控的靶基因.[结果]本文克隆获得了 BmFoxG-1(933 bp)和 BmFoxG-2(702 bp)两个基因.BmFoxG-1 和 BmFoxG-2蛋白均包含保守的Forkhead结构域,但BmFoxG-1蛋白在C端多出约60个氨基酸.BmFoxG-2基因在家蚕不同发育时期的精巢中的表达量均较低;BmFoxG-1 的表达均显着高于BmFoxG-2,且随着发育时期而变化,暗示BmFoxG-1参与精巢的发育.生殖细胞发育基因BmVasa、BmCyclinA等多个细胞周期基因以及精子鞭毛发育相关基因BMSK0009828在不同发育阶段的精巢中的表达趋势与BmFoxG-1类似,且BmFoxG-1在家蚕细胞中的过表达可以显著上调BmVasa、BmCyclinA和BMSK0009828等精巢发育相关基因的表达.[结论]我们推测BmFoxG-1蛋白可能通过调节精巢细胞周期或生殖细胞的功能来调节家蚕精巢的发育.  相似文献

【目的】非编码RNA(non-coding RNA,ncRNA)在家蚕Bombyx mori发育过程中具有重要调控作用。本研究旨在探索ncRNA参与家蚕神经系统发育的分子机理。【方法】采用实时荧光定量PCR技术对22个中等长度的ncRNA及3个ncRNA的邻近编码基因在家蚕幼虫神经系统中的表达水平进行检测。【结果】8个ncRNA(包括1个C/D box snoRNA,4个H/ACA box snoRNA和3个不能归类的ncRNA)在家蚕5龄幼虫神经组织和非神经组织中均有表达,且在胸腹神经中的表达量明显高于头部神经中的表达量。其中,snoRNA Bm-51,Bm-18和Bm-86在胸腹神经中的表达量分别是头部神经中的23,5和4.7倍。进一步研究发现,这3个内含子起源的ncRNA与其宿主基因在家蚕神经系统中的表达趋势一致,宿主基因在胸腹神经中的表达量也明显高于头部神经中的表达量。【结论】本研究筛选到的在家蚕不同神经部位存在差异表达的ncRNA,特别是在胸腹神经中高表达的ncRNA可能协同其邻近编码基因,参与家蚕神经发育或神经活动过程。该结果为研究ncRNA参与家蚕神经系统发育提供了分子依据,为从非编码RNA角度探索鳞翅目害虫防治提供了新的思路。  相似文献

1. Ovarian tissue from Bombyx mori L. larvae about to pupate was cultured in Trager's (1935) salt solution and 10 per cent hemolymph, with indifferent results. Improvement of cultures was sought by modifying the culture medium. 2. To reduce the activity of the tyrosinase, hemolymph for culture medium was heated for 5 minutes at 60°C., and the coagulated protein removed. 3. A physiological solution was formulated containing cations and amino acids as they occur normally in silkworm hemolymph. In both hanging-drop and small tube cultures use of this medium brought about increased cell number, improved cell appearance, more rapid mitoses, and longer life of cultures. 4. To the solution formulated from analyses, tryptophan, cystine, cysteine, malate, fumarate, succinate, and α-ketoglutarate were added after testing individually, resulting in improved growth in cultures. 5. Use of a silkworm egg extract prepared 4 to 5 days after acid treatment produced an increase in cell number. 6. In small roller tube cultures, when the new medium was changed twice a week, the cells spread over the walls of the tube in 4 or 5 days (Figs. 8 and 9), rapid mitoses were observed after 2 weeks, and transparent active cells were present at 3 weeks. In the silkworm, Bombyx mori, two storage proteins named SP-1 and SP-2 were shown to decline in concentration in the haemolymph and increase in the fat body during the larval-pupal transformation, when protein granules are formed in the fat body at the same time as the degeneration of mitochondria and endoplasmic reticulum. At the larval-pupal ecdysis, in females the two proteins account for 60% of total fat body protein (80% of the soluble protein), while males have very little SP-1 and SP-2 comprises only 20% of the total fat body protein. The concentration of protein granules in the fat body cytoplasm is much greater in females than in males, and the granules in females have partially crystalline inner zones. This is different from males where granules with non-crystalline structure are most numerous.The properties of these proteins purified from pupal fat body are similar to those of Cecropia storage proteins and calliphorin, all of which have molecular weights of around 500,000 and are composed of subunits of mol. wt about 85,000. SP-1 differs from SP-2 by having an exceptionally high content of methionine, but much less glutamate, phenylalanine and tyrosine. SP-1 resembles another female-specific protein, vitellogenin and SP-2 resembles calliphorin in amino acid composition.From these results, it is concluded that SP-1 and SP-2 have storage roles and are deposited in protein granules.  相似文献

Small non-protein coding RNAs (ncRNAs) play important roles in development, stress response and other cellular processes. Silkworm is an important model for studies on insect genetics and control of Lepidopterous pests. We have previously identified 189 novel intermediate-size ncRNAs in silkworm Bombyx mori, including 40 ncRNAs that showed altered expression in different developmental stages. Here we characterized the functions of these 40 ncRNAs by measuring their expressions in six tissues of the fifth instar larvae using Northern blot and real-time polymerase chain reaction assays. We identified nine ncRNAs (four small nucleolar RNAs and five unclassified ncRNAs) that were enriched in silk gland, including four ncRNAs that showed silk gland-specific expression. We further showed that three of nine silk gland-enriched ncRNAs were predominantly expressed in the anterior silk gland, whereas another three ncRNAs were highly accumulated in the posterior silk gland, suggesting that they may play different roles in fibroin synthesis. Furthermore, an unclassified ncRNA, Bm- 152, exhibited converse expression pattem with its antisense host gene gartenzwerg in diverse tissues, and might regulate the expression of gartenzwerg through RNA-protein complex. In addition, two silk gland-enriched ncRNAs Bm-102 and Bm-159 can be found in histone modification complex, which indicated that they might play roles through epigenetic modifications. Taken together, we provided the first expression and preliminary functional analysis of silk gland-enriched ncRNAs, which will help understand the molecular mechanism of silk gland-development and fibroin synthesis.  相似文献

Since insects are unable to biosynthesize sterols de novo, sterols must be obtained from dietary sources. Although it has been reported that beta-sitosterol is crucial for larval growth in the silkworm, Bombyx mori, little has been investigated concerning the dietary selection of sterols by Bombyx larvae. Here, we demonstrate that Bombyx larvae have the following sterol preference: beta-sitosterol > ergosterol > cholesterol = stigmasterol. Interestingly, Bombyx larvae preferred ergosterol, an inhibitory sterol on larval growth, indicating that sterol selection following first contact of the diet with the mouth part might be different from the sterol recognition mechanism present in sterol metabolism.  相似文献

目的:通过观察家蚕Bombyx mori吞噬细胞的微细结构,来确定拟绛色细胞是否也具有吞噬功能。方法:用荧光小球微量注射家蚕pnd pS品系的幼虫,经荧光染色剂丫啶橙和碘化丙啶染色循环血细胞后,在荧光显微镜下观察并扫描拍摄。结果:观察发现除颗粒细胞和浆血细胞外,一些原血球细胞(血干细胞)和拟绛色细胞(多酚氧化酶)也能吞噬荧光小球。在拟绛色细胞里还发现许多和颗粒细胞一样的能被丫啶橙染色的颗粒。尽管在小球细胞中没有发现被吞噬的荧光小球,但该类血球有比较多的能被丫啶橙染色的大颗粒,这表明它们可能是已经被吞噬的凋亡小体。结论:除颗粒细胞和浆血细胞外,一些原血球细胞和拟绛色细胞也能吞噬荧光小球。说明拟绛色细胞也具有吞噬功能。  相似文献

Fenoxycarb treatment before and after pupal ecdysis of Bombyx mori disturbed adult eclosion and the animals were unable to escape from the pupal exuviae. This effect of fenoxycarb was dose and time dependent with the highest sensitivity around the pupal ecdysis. The sensitivity rapidly diminished within 20 hours of pupal ecdysis. Twenty-hydroxyecdysone (20E) produced similar effects. Fenoxycarb injection at the pupal ecdysis induced higher ecdysteroid production by the prothoracic glands and higher PTTH-secretory activity in the brain-corpora cardiaca-corpora allata complexes. As a result, the fenoxycarb treated pupae contained higher ecdysteroid titres in the haemolymph. Both the fenoxycarb and the 20E treatments resulted in the lack of development of the rectum in pharate adults. This was the main cause of high ecdysteroid titres in the pharate adult stage. This effect was mimicked by either removal of the rectum early in the pharate adult stage or a surgical extirpation of the hindgut at the time of pupal ecdysis. These results suggest that the disturbance of adult eclosion by fenoxycarb is due in part to the inability of the formation of the rectum in the pharate adult stage.  相似文献

Topical application of fenoxycarb (1 μg per animal) at 129 or 132 h of the fifth instar larvae of the silkworm, Bombyx mori, did not induce morphological abnormalities in the pupal stage, but these animals became dauer (permanent) pupae. This condition of B. mori and the endocrine events leading to permanent pupae are discussed in this work. Application of fenoxycarb at 132 h of the fifth instar elicited a high ecdysteroid titre in the pharate pupal stage and a steadily high ecdysteroid titre in the pupal stage. The fenoxycarb-induced permanent pupae had non-degenerating prothoracic glands that secreted low amounts of ecdysteroid and did not respond to recombinant prothoracicotropic hormone (rPTTH) late in the pupal stage. The Bombyx PTTH titre in the haemolymph, determined by a time-resolved fluoroimmunoassay, was lower than that of controls at the time of pupal ecdysis, but higher than controls later in the pupal stage in fenoxycarb-treated animals. After application of fenoxycarb, its haemolymph level, measured by ELISA, reached a peak at pupal ecdysis, then remained low. These results suggest that the fenoxycarb-mediated induction of permanent pupae is only partially a brain-centred phenomenon. It also involves alterations in the hormonal interplay that govern both the initiation of pupal-adult differentiation and changes in the steroidogenic pathway of the prothoracic glands of B. mori.  相似文献

Sterols in silkworm larvae were analyzed. Cholesterol was predominantly detected in all tissues examined. Dietary phytosterols and desmosterol, a putative biosynthetic intermediate from phytosterols to cholesterol, were also detected, indicating that imperfect intestinal conversion from phytosterols to cholesterol influences the sterol composition in larval tissues.  相似文献

The Tudor-sn protein, which contains four staphylococcal nuclease domains and a Tudor domain, is a ubiquitous protein found in almost all organisms. It has been reported that Tudor-sn in mammals participates in various cellular pathways involved in gene regulation, cell growth, and development. In insects, we have previously identified a Tudor-sn ortholog in the silkworm, Bombyx mori, and detected its interactions between with Argonaute proteins. The role of Tudor-sn in silkworm, however, still remains largely unknown. In this study, we demonstrated that silkworm Tudor-sn is a stress granule (SG) protein, and determined its interactions with other SG proteins using Bimolecular Fluorescence Complementation assay and Insect Two-Hybrid method. Depletions of Argonaute proteins and SG-marker protein Tia1 by RNAi impaired the involvement of Tudor-sn in the SG formation. Protein domain deletion analysis of Tudor-sn demonstrated that SN2 is the key domain required for the aggregation of Tudor-sn in SGs.  相似文献

It is generally accepted that the prothoracicotropic hormone (PTTH) is the stimulator of ecdysteroidogenesis by prothoracic glands in larval insects. In the present study, we investigated activation of ecdysteroidogenesis by bovine insulin in prothoracic glands of the silkworm, Bombyx mori. The results showed that the insulin stimulated ecdysteroidogenesis during a long-term incubation period and in a dose-dependent manner. In addition, insulin also stimulated both DNA synthesis and viability of prothoracic glands. Insulin-stimulated ecdysteroidogenesis was blocked by either LY294002 or wortmannin, indicating involvement of the phosphatidylinositol 3-kinase (PI3K) signaling pathway. Activation of ecdysteroidogenesis by insulin appeared to be developmentally regulated. Moreover, in vitro activation of ecdysteroidogenesis of prothoracic glands by insulin was also verified by in vivo experiments: injection of insulin into day 6 last instar larvae greatly increased both hemolymph ecdysteroid levels and ecdysteroidogenesis 24 h after the injection, indicating its possible in vivo function. Phosphorylation of Akt and the insulin receptor was stimulated by insulin, and stimulation of Akt phosphorylation appeared to be PI3K-dependent and developmentally regulated. Insulin did not stimulate extracellular signal-regulated kinase (ERK) signaling of the prothoracic glands. These results suggest that in silkworm prothoracic glands, in addition to the PTTH and an autocrine factor, ecdysteroidogenesis is also stimulated by insulin during development.  相似文献

NHL家族蛋白具有调控细胞增殖与分化的功能,在哺育动物中被广泛研究。本文克隆得到家蚕NHL蛋白家族成员BmBrat基因,通过RACE技术获得该基因cDNA全长序列为3 614 bp,其ORF为2 580 bp,编码859个氨基酸,预测其蛋白分子量为94.3 kDa,等电点为6.65。利用RT-PCR技术检测其在五龄3 d家蚕各组织表达情况,结果表明其在幼虫各组织均有表达,包括丝腺、中肠、脂肪体、马氏管等,且卵巢和头部表达量最高;胚胎时期表达谱分析显示其在胚胎发育第4天和第5天有高量表达。经原核表达、蛋白纯化及免疫小鼠后获得家蚕BmBrat多克隆抗体,且Western blotting及免疫荧光检测显示该抗体可以特异检测家蚕BmBrat蛋白;免疫荧光结果表明BmBrat蛋白定位于家蚕血细胞胞质中,为进一步研究BmBrat基因的生物学功能奠定了基础。  相似文献
