膏桐饼作为生物柴油产业中产生的最大宗副产物,其高蛋白、低纤维含量的特性有望使其开发成为一种优良的蛋白饲料,但毒性限制了其直接作为饲料利用.发酵是多种饼粕脱毒的常用方法.分离、纯化膏桐根际微生物并分别发酵膏桐饼,用甲醇提取发酵膏桐饼的毒性成分并溶于水中,通过鲤鱼存活时间来评价不同菌株发酵膏桐饼的毒性,筛选获得了一株能够在膏桐饼上快速生长并能有效脱除膏桐饼毒性的短帚霉.脱毒小桐子饼组的小鲤鱼存活时间与对照组相比延长了1.22倍.  相似文献

对云南省两个不同地区膏桐(Jatropha curcas)种子形态特征,粗脂肪含量与环境因子关系开展研究,结果表明:湿热环境的景洪较干热环境的元谋膏桐种子成熟散布期提前,形态特征也优于元谋膏桐种子(P〈0.01);土壤有机质是影响膏桐种子粗脂肪积累最重要的因子,对膏桐种子粗脂肪积累起显著正效应;干旱胁迫也是影响膏桐种子粗脂肪积累的一个重要的限制因子;景洪膏桐种子粗脂肪含量、含水量较元谋膏桐种子更高,但粗脂肪含量与含水量比率较低(P〈0.01);具有较高粗脂肪含量的膏桐种子与其含水量的比率可能是植物对干旱胁迫适应的一种机制,其对物种的适应有着重要的生态学意义。  相似文献

膏桐为一种重要的油料作物,其种子可以提炼生物柴油,生物柴油在我国将成长为有一定规模的产业。为了增加膏桐结籽率,提高其产量,给膏桐规模化栽培方面提供传粉生物学的科学依据,于2006年对云南元江坝区半栽培膏桐(Jatropha curcas L.)居群的访花昆虫及主要传粉昆虫种类进行调查。膏桐开花示样醒目,具花香与蜜腺,花形态没有特化,这些特征使得它提供的酬物适合于不同的昆虫采食,其访花昆虫种类较丰富,共有35种访花昆虫,分属5目20科。综合访花昆虫的传粉数量,及传粉质量包括访花行为、访花频率与日活动规律等确定主要传粉昆虫,确定大头金蝇Chrysomya(Compsomyia)megacephala Fabricius、中华蜜蜂Apiscerana Fabricius、迁粉蝶(淡色型)Catopsilia pomona f.Crocale为该居群的主要传粉昆虫。3种传粉昆虫具有不同的访花习性和日活动规律。雌雄迁粉蝶日活动规律相似。访花者的组成受气象因素、开花示样及生境条件的影响。最后,探讨膏桐与访花昆虫的关系,以及主要传粉者的组成随时空变异而改变的规律。  相似文献

玉米赤霉烯酮(zearalenone,ZEN)是霉变谷物中常见的霉菌毒素之一,主要出现在霉变的玉米、小麦等谷物中,给畜禽和人类带来一定程度的健康危害,如生殖毒性、免疫毒性、肝毒性和肾毒性等。目前,解决玉米赤霉烯酮污染问题的方法包括物理、化学和生物3个途径。虽然传统的物理和化学脱毒方法已经运用在许多的饲料生产中,但同时也存在着二次污染的风险。生物降解法是一种利用微生物吸附和降解玉米赤霉烯酮的脱毒方法,具有安全环保、高效、特异性强和脱毒率高的特性,且不影响谷物的营养价值,已成为玉米赤霉烯酮降解研究的热点。本文主要介绍了近年来降解玉米赤霉烯酮的微生物种类,并将其归纳分类,从微生物的脱毒能力、脱毒方法和脱毒产物进行了叙述,综述了微生物脱毒的优点及前景,以期为微生物降解玉米赤霉烯酮的理论研究及实际应用提供新的视角。  相似文献

从土壤中分离纯化真菌并鉴定为烟曲霉L-3。以菌株L-3作为固定化载体,将地衣芽孢杆菌固定在真菌上组成固定化体系。研究了混菌菌丝球,菌丝饼,发酵混合液,粗漆酶液对刚果红染料废水的降解情况并对染料废水进行毒性试验。结果表明,菌丝球对染料废水的降解效果最显著,降解率高达99.96%,菌丝饼仅用20 s降解率为91%,发酵混合液与粗漆酶液的处理效果并不显著。该体系对染料废水的去毒率较明显,尤其是菌丝球的去毒率可达到78%。可见,固定化体系对染料废水不但有较高的降解能力,也有较高的去毒率。  相似文献

研究了黑曲霉与酿酒酵母混合固态发酵对棉籽饼粕的脱毒效果及提高棉籽饼粕营养价值的作用。结果表明,混合发酵优化后的条件为:发酵温度30℃,接种量15%,初始水分含量0.96g水/g干基,初始pH值为6,发酵时间60 h;发酵后棉籽饼粕中游离棉酚脱毒率为95.51%,真蛋白质含量从32.99%提高到 42.40%,必需氨基酸中赖氨酸、蛋氨酸和苏氨酸分别较发酵前相比增加了31.03%、25.00%和44.23%,氮溶解指数(NSI)增加了1.49倍,从而改善了棉粕饼粕的品质。  相似文献

高效降解棉酚菌株的分离鉴定及诱变选育   总被引:2,自引:1,他引:1  
作为一种蛋白资源, 棉籽粕因其含有毒素——游离棉酚限制了其在饲料工业中的应用。为获得能高效降解棉籽粕中棉酚的菌株, 以醋酸棉酚为唯一碳源培养基从16份样品中筛选出一株高效降解棉酚菌株(Y-2), 经生理生化及18S rDNA鉴定为Pichia guilliermondii, 此菌种为非致病酵母, 且首次报道用于棉酚的降解, 在工业生产中具有潜在的应用前景。通过紫外诱变获得一株棉酚降解率更高的突变株YUV-51。通过对突变株YUV-51发酵温度、时间及接种量的初步优化, 获得其固态发酵优化条件: 30 °C培养48 h, 接种量0.025 g湿菌体/g棉籽粕, 初始水分含量50%。为避免游离棉酚在前处理中大量降解棉籽粕, 不进行湿热处理, 经接种发酵后脱毒率可达到58%。这使微生物脱毒在实际生产中应用成为可能。  相似文献

本实验对前期研究得到的具有高产红色素能力的产紫青霉突变菌株Penicillium purpurogenum Li-3-9的发酵条件进行优化的同时作出了对红色素的安全性评价。在单因素试验的基础上,采用Box-Behnken试验设计优化了蔗糖浓度、酵母膏浓度和装液量对红色素色价的影响;并且研究了P. purpurogenum Li-3-9产生的红色素对斑马鱼胚胎的毒性效应。结果表明,最佳发酵条件为:蔗糖40g/L、酵母膏4g/L、装液量为45mL、接种量4%、培养温度32℃、初始pH值6.9、培养时间168h、摇床转速150r/min,红色素色价最高达到了11.4U/mL。通过计算斑马鱼胚胎致死率、心率、畸形率及孵化率可知,该色素无致死毒性,并且对斑马鱼胚胎发育具有一定的促进作用。  相似文献

Gibel carp ( Carassius auratus gibelio Bloch) is a natural gynogenetic fish which requires sperm of the same or related species to activate egg development. The eggs of one gibel carp were divided into two batches. One batch was 'fertilized' with sperm from gibel carp (strain DD), and the other 'fertilized' with sperm from red common carp ( Cyprinus carpio red variety) (strain DR). The juveniles were transferred to the laboratory 36 days post-hatch. Triplicate groups of each strain were fed a formulated diet at either 3% or satiation ration for 8 weeks. At both the restricted and satiation rations, specific growth rate was significantly higher in strain DR than in strain DD. At the 3% ration, there was no significant difference in feeding rate or feed conversion efficiency between the two strains. At the satiation ration, strain DR had a significantly lower feeding rate but higher feed conversion efficiency than strain DD. At the satiation ration, strain DR had a significantly lower intake protein, but higher recovered protein than strain DD. There was no significant difference in faecal protein loss between the two strains. At the 3% ration, strain had no significant effects on intake protein, faecal protein or recovered protein. Neither faecal energy loss nor recovered energy was affected by strain or ration. At both the 3% and satiation ration, final body contents of dry matter and lipid were significantly lower in strain DR than strain DD, while there was no significant difference in protein and energy content between the two strains at either ration level. The results suggested that gibel carp 'fertilized' with sperm of common carp grew faster than those 'fertilized' with sperm of gibel carp through increased feed conversion efficiency and protein retention.  相似文献

为探究饲料中高剂量的有机硒和无机硒对异育银鲫(Carassius auratus gibelio var. CAS V)的生长性能、硒蓄积和血浆生化指标等方面的影响,以硒代蛋氨酸(有机硒)和亚硒酸钠(无机硒)作为不同类型的硒源,初始体重为(62.95±0.23) g异育银鲫为研究对象,进行了为期90d的养殖实验。实验结果表明,饲料中添加0、10和20 mg/kg的有机硒和无机硒对异育银鲫的存活和饲料干物质的表观消化率无显著影响;有机硒处理组硒的表观消化率随饲料有机硒的添加量增加而显著升高(P<0.05);饲料中添加无机硒对硒消化率无显影响(P>0.05)。在饲料中添加有机硒可以提高异育银鲫的生长(P<0.05),在20 mg/kg处理组中达到最高(P<0.05);而饲料中添加10 mg/kg无机硒处理组未显著改变异育银鲫的特定生长率(P>0.05),但高浓度的无机硒则显著降低了其特定生长率(P<0.05)。饲料中添加有机硒显著降低了异育银鲫的肝体比,添加10 mg/kg无机硒显著降低异育银鲫的肝体比(P<0.05);饲料中添加有机硒和无机硒对异...  相似文献

鲤饲料转化率性状的QTL 定位及遗传效应分析   总被引:1,自引:0,他引:1  
数量性状(QTL)定位是实现分子标记辅助育种、基因选择和定位、培育新品种及加快性状遗传研究进展的重要手段。饲料转化率是鲤鱼的重要经济性状和遗传改良的主要目标, 而通过QTL 定位获得与饲料转化率性状紧密连锁的分子标记以及相关基因是遗传育种的重要工具。研究利用SNP、SSR、EST-SSR 等分子标记构建鲤鱼(Cyprinus carpio L.)遗传连锁图谱并对重要经济性状进行QTL 定位。选用174 个SSR 标记、41 个EST-SSR 标记、345 个SNP 标记对德国镜鲤F2 代群体68 个个体进行基因型检测, 用JoinMap4.0 软件包构建鲤鱼遗传连锁图谱。再用MapQTL5.0 的区间作图法(Interval mapping, IM)和多QTL 区间定位法(MQMMapping, MQM)对饲料转化率性状进行QTL 区间检测, 通过置换实验(1000 次重复)确定连锁群显著性水平阈值。结果显示, 在对饲料转化率性状的多QTL 区间定位中, 共检测到15 个QTLs 区间, 分布在9 个连锁群上, 解释表型变异范围为17.70%—52.20%, 解释表型变异最大的QTLs 区间在第48 连锁群上, 为52.20%。HLJE314-SNP0919(LG25)区间标记覆盖的图距最小, 为0.164 cM; 最大的是HLJ1439-HLJ1438(LG39)区间,覆盖图距为24.922 cM。其中区间HLJ1439-HLJ1438、HLJ922 -SNP0711 解释表型变异均超过50.00%, 可能是影响饲料转化率性状的主效QTLs 区间。与饲料转化率相关的15 个QTLs 的加性效应方向并不一致, 有3个区间具有负向加性效应, 平均为?0.027; 12 个正向加性效应, 平均值为0.06。研究检测出的与鲤鱼饲料转化率性状相关的QTL 位点可为鲤鱼分子标记辅助育种和更进一步的QTL 精细定位打下基础。    相似文献

The influence of traditional Chinese medicine (TCM) formulation from Astragalus Root (Radix astragalin seu Hedysari) and Chinese Angelica Root (R. Angelicae Sinensis) at a ratio of 5:1 (w/w) on non-specific immunity of Jian carp, Cyprinus carpio var. Jian was investigated. The number of NBT-positive cells in the blood and lysozyme and complement activities in the serum of Carp fed with commercial feed supplemented with 1.0% (diet 1) and 1.5% (diet 2) TCM at 10 day of post-feeding were not different from those of the control group fed with feed unsupplemented TCM 10 days post-feeding (P>0.05), but at 20 and 30 days they increased significantly (P<0.05). The values of diet 1 group and diet 2 group at 20 day and at 30 day were not significantly different (P>0.05) from each other. In addition, the TCM formula increased body weight of experimental fish by about 16.84% (diet 1) and 19% (diet 2) above that of the control group. Therefore, these data suggest that the TCM formula could elevate the function of non-specific immunity of Jian carp. The optimal dosage added to commercial carp feed was 1.0% (w/w) and the oral administration time as a course of treatment was 20 days.  相似文献

A bacterial strain with putative immunomodulatory properties was isolated from Xi'an hot springs in China. Comparison of 16S rRNA gene revealed a 97% similarity between the tested strain (designated XA-1) and Anoxybacillus kamchatkensis. Two compounds isolated from the secondary metabolites of XA-1 were identified by spectral data (infrared, nuclear magnetic resonance and mass spectrometry) as: (1) cyclo (Gly-L-Pro) and (2) cyclo (L-Ala-4-hydroxyl-L-Pro). Two cyclic dipeptides showed stimulatory properties towards a range of parameters when a dose of 20mg kg(-1) body weight was intraperitoneally injected in naive common carp, Cyprinus carpio. Innate immune parameters (serum SOD, lysozyme and bactericidal activity, and phagocytic activity by peripheral blood leucocytes) along with the expression of two immune-related genes (IL-1β and iNOS) in blood were examined after 7, 14, 21, and 28 days of injection. In the absence of infection, immunomodulators should ideally not affect normal physiology and immunity of the host; possible negative outcomes of activated immune responses in the naive state are discussed. Protection by two bacterial dipeptides was assessed in an intraperitoneal injection challenge trial with live Aeromonas hydrophila. Both compounds reduced mortality, with the highest survival rate observed in the group that received compound 2 (80%) followed by the group that received compound 1 (65%) while control group scored the worse (15%). Elucidation of the involved protective mechanisms in carp requires future studies.  相似文献

本文通过细菌的分离培养,首次从临床症状表现为溃疡、腐烂的患病江豚表皮分离到一株杀鲑气单胞菌XJ-JT株。通过细菌理化性质鉴定、遗传进化分析、药敏试验、致病性试验对其生物学特性进行分析,结果表明:菌株XJ-JT为革兰氏阴性短杆菌,两端钝圆,无芽孢;细菌分离鉴定的结果显示分离菌株为杀鲑气单胞菌;系统进化分析揭示其基因序列与银鲫源性杀鲑气单胞菌分离株高度同源;20种抗生素的药敏试验结果表明分离菌株对阿米卡星、克拉霉素、克林霉素等8种药物敏感,对头孢噻肟、头孢曲松、卡那霉素中介,对阿莫西林、氨苄西林、四环素等9种药物耐药;人工感染试验,结果显示分离菌对鲫鱼有较强的致病性。  相似文献

对从患病锦鲤(Cyprinus carpio L.)肝组织中分离的1株纯培养菌(HC050630C-1)进行了形态特征、主要理化特性、对健康鲤鱼的致病作用、药物敏感性等方面的检验;同时测定了该株菌的16S rRNA基因序列,构建了系统发育树。结果表明,被检菌为吉氏库特菌(Kurthia gibsonll),对健康鲤鱼未显示明显的致病作用;所测菌株的16S rRNA基因序列长度1459bp,在GenBank中登录号为EF611423;该菌株16SrRNA基因序列与GenBank数据库中吉氏库特菌的16S rRNA基因序列同源性在99%;药敏试验结果显示,对供试的氨苄青霉素等27种药物呈现高度敏感或敏感,对头孢吡肟等6种呈低度敏感,对苯唑青霉素等4种耐药。  相似文献

本实验室2009年从广东省患典型出血病的养殖草鱼中分离到一株具强致病性的水生呼肠孤病毒GCRV-GD108株,该毒株具有11个节段双链RNA。全基因组序列分析显示与草鱼呼肠孤病毒GCRV及水生呼肠孤病毒属其它已知种存在较大的分子差异。本研究进一步检测了广东、福建、湖南等地草鱼出血病流行毒株的分子特性。根据已克隆到的GCRV-GD108株11个节段序列分别设计合成特异引物,从各地收集患出血病草鱼,提取组织总RNA,RT-PCR检测。结果表明各检测样品均可扩增到特异性条带,而GCRV标准株则无特异条带;同时根据GCRV标准株序列合成的特异引物进行扩增,GCRV标准株有特异性条带,而各检测样品则均无带。测序结果显示各样品间相应片段序列的同源性很高(95.2%~99.4%),与GCRV-GD108的相应序列也具高同源性(95.0%~99.8%),说明检测样品与GCRV-GD108株具有相似的分子特性,均与GCRV及水生呼肠孤病毒属的其它种存在较大差异。本研究结果提示我国养殖草鱼出血病病毒存在着不同的分子类型,GCRV-GD108株在南方具有一定的代表性,在病害防控上尤其是疫苗研制与使用上应予关注。此外,从上述引物中筛选出适合于双重PCR的引物对,建立了双重PCR检测方法,可在一次PCR反应中鉴别所感染的病毒属于GCRV或GCRV-GD108株。  相似文献

We used customized fish tanks as model fish ponds to observe grazing, swimming, and conspecific social behavior of common carp (Cyprinus carpio) under variable food-resource conditions to assess alterations in feeding niche. Different food and feeding situations were created by using only pond water or pond water plus pond bottom sediment or pond water plus pond bottom sediment and artificial feeding. All tanks were fertilized twice, prior to stocking and 2 weeks later after starting the experiment to stimulate natural food production. Common carp preferred artificial feed over benthic macroinvertebrates, followed by zooplankton. Common carp did not prefer any group of phytoplankton in any treatment. Common carp was mainly benthic in habitat choice, feeding on benthic macroinvertebrates when only plankton and benthic macroinvertebrates were available in the system. In the absence of benthic macroinvertebrates, their feeding niche shifted from near the bottom of the tanks to the water column where they spent 85% of the total time and fed principally on zooplankton. Common carp readily switched to artificial feed when available, which led to better growth. Common carp preferred to graze individually. Behavioral observations of common carp in tanks yielded new information that assists our understanding of their ecological niche. This knowledge could be potentially used to further the development of common carp aquaculture.  相似文献
