The integration of microalgae-based biofuel and bioproducts production with wastewater treatment has major advantages for both industries. However, major challenges to the implementation of an integrated system include the large-scale production of algae and the harvesting of microalgae in a way that allows for downstream processing to produce biofuels and other bioproducts of value. Although the majority of algal production systems use suspended cultures in either open ponds or closed reactors, the use of attached cultures may offer several advantages. With regard to harvesting methods, better understanding and control of autoflocculation and bioflocculation could improve performance and reduce chemical addition requirements for conventional mechanical methods that include centrifugation, tangential filtration, gravity sedimentation, and dissolved air flotation. There are many approaches currently used by companies and industries using clean water at laboratory, bench, and pilot scale; however, large-scale systems for controlled algae production and/or harvesting for wastewater treatment and subsequent processing for bioproducts are lacking. Further investigation and development of large-scale production and harvesting methods for biofuels and bioproducts are necessary, particularly with less studied but promising approaches such as those involving attached algal biofilm cultures.  相似文献

Terpenes are important compounds in plant trophic interactions. A meta‐analysis of GC‐MS data from a diverse range of apple (Malus × domestica) genotypes revealed that apple fruit produces a range of terpene volatiles, with the predominant terpene being the acyclic branched sesquiterpene (E,E)‐α‐farnesene. Four quantitative trait loci (QTLs) for α‐farnesene production in ripe fruit were identified in a segregating ‘Royal Gala’ (RG) × ‘Granny Smith’ (GS) population with one major QTL on linkage group 10 co‐locating with the MdAFS1 (α‐farnesene synthase‐1) gene. Three of the four QTLs were derived from the GS parent, which was consistent with GC‐MS analysis of headspace and solvent‐extracted terpenes showing that cold‐treated GS apples produced higher levels of (E,E)‐α‐farnesene than RG. Transgenic RG fruit downregulated for MdAFS1 expression produced significantly lower levels of (E,E)‐α‐farnesene. To evaluate the role of (E,E)‐α‐farnesene in fungal pathogenesis, MdAFS1 RNA interference transgenic fruit and RG controls were inoculated with three important apple post‐harvest pathogens [Colletotrichum acutatum, Penicillium expansum and Neofabraea alba (synonym Phlyctema vagabunda)]. From results obtained over four seasons, we demonstrate that reduced (E,E)‐α‐farnesene is associated with decreased disease initiation rates of all three pathogens. In each case, the infection rate was significantly reduced 7 days post‐inoculation, although the size of successful lesions was comparable with infections on control fruit. These results indicate that (E,E)‐α‐farnesene production is likely to be an important factor involved in fungal pathogenesis in apple fruit.  相似文献

Across the energy sector, alternatives to fossil fuels are being developed, in response to the dual drivers of climate change and energy security. For transport, biofuels have the greatest potential to replace fossil fuels in the short‐to medium term. However, the ecological benefits of biofuels and the role that their deployment can play in mitigating climate change are being called into question. Life Cycle Assessment (LCA) is a widely used approach that enables the energy and greenhouse gas (GHG) balance of biofuel production to be calculated. Concerns have nevertheless been raised that published data show widely varying and sometimes contradictory results. This review describes a systematic review of GHG emissions and energy balance data from 44 LCA studies of first‐ and second‐generation biofuels. The information collated was used to identify the dominant sources of GHG emissions and energy requirements in biofuel production and the key sources of variability in published LCA data. Our analysis revealed three distinct sources of variation: (1) ‘real’ variability in parameters e.g. cultivation; (2) ‘methodological’ variability due to the implementation of the LCA method; and (3) ‘uncertainty’ due to parameters rarely included and poorly quantified. There is global interest in developing a sustainability assessment protocol for biofuels. Confidence in the results of such an assessment can only be assured if these areas of uncertainty and variability are addressed. A more defined methodology is necessary in order to allow effective and accurate comparison of results. It is also essential that areas of uncertainty such as impacts on soil carbon stocks and fluxes are included in LCA assessments, and that further research is conducted to enable a robust calculation of impacts under different land‐use change scenarios. Without the inclusion of these parameters, we cannot be certain that biofuels are really delivering GHG savings compared with fossil fuels.  相似文献

As an herbaceous perennial, Miscanthus has attracted extensive attention in bioenergy refinery and ecological remediation due to its high yield and superior environmental adaptability. This review summarizes current research advances of Miscanthus in several aspects including biological properties, biofuels production, and phytoremediation of contaminated soil. Miscanthus has relatively high biomass yield, calorific value, and cellulose content compared with other lignocellulosic bioenergy crops, which make it one of the most promising feedstocks for the production of second‐generation biofuels. Moreover, Miscanthus can endure soil pollutions caused by various heavy metals and survive in a variety of adverse environmental conditions. Therefore, it also has potential applications in ecological remediation of contaminated soil, and reclamation of polluted soil and water resources. Nevertheless, more endeavors are still needed in the genetic improvement and elite cultivar breeding, large‐scale cultivation on marginal land, and efficient conversion to biofuels, when utilizing Miscanthus as a bioenergy crop. Furthermore, more efforts should also be undertaken to translate Miscanthus into a bioenergy crop with the phytoremediation potential.  相似文献

India is the fifth largest primary energy consumer and fourth largest petroleum consumer after USA, China, and Japan. Despite the global economic crisis, India’s economy is expected to grow at 6 to 8?%/year. There is an extreme dependence on petroleum products with considerable risks and environmental issues. Petroleum-derived transport fuels are of limited availability and contribute to global warming, making renewable biofuel as the best alternative. The focus on biogas and biomass-based energy, such as bioethanol and biohydrogen, will enhance cost-effectiveness and provide an opportunity for the rural community. Among all energy sources, microalgae have received, so far, more attention due to their facile adaptability to grow in the photobioreactors or open ponds, high yields, and multiple applications. Microalgae can produce a substantial amount of triacylglycerols as a storage lipid under photooxidative stress or other adverse environmental conditions. In addition to renewable biofuels, they can provide different types of high-value bioproducts added to their advantages, such as higher photosynthetic efficiency, higher biomass production, and faster growth compared to any other energy crops. The viability of first-generation biofuels production is, however, questionable because of the conflict with food supply. In the future, biofuels should ideally create the environmental, economic, and social benefits to the communities and reflect energy efficiency so as to plan a road map for the industry to produce third-generation biofuels.  相似文献

To increase the understanding of poplar and willow perennial woody crops and facilitate their deployment for the production of biofuels, bioproducts, and bioenergy, there is a need for broadscale yield maps. For national analysis of woody and herbaceous crops production potential, biomass feedstock yield maps should be developed using a common framework. This study developed willow and poplar potential yield maps by combining data from a network of willow and poplar field trials and the modeling power of PRISM‐ELM. Yields of the top three willow cultivars across 17 sites ranged from 3.60 to 14.6 Mg ha?1 yr?1 dry weight, while the yields from 17 poplar trials ranged from 7.5 to 15.2 Mg ha?1 yr?1. Relationships between the environmental suitability estimates from the PRISM‐ELM model and results from field trials had an R2 of 0.60 for poplar and 0.81 for willow. The resulting potential yield maps reflected the range of poplar and willow yields that have been reported in the literature. Poplar covered a larger geographic range than willow, which likely reflects the poplar breeding efforts that have occurred for many more decades using genotypes from a broader range of environments than willow. While the field trial data sets used to develop these models represent the most complete information at the time, there is a need to expand and improve the model by monitoring trials over multiple cutting cycles and across a broader range of environmental gradients. Despite some limitations, the results of these models represent a dramatic improvement in projections of potential yield of poplar and willow crops across the United States.  相似文献

ABSTRACT

Microalgae have enormous potential as feedstock for biofuel production compared with other sources, due to their high areal productivity, relatively low environmental impact, and low impact on food security. However, high production costs are the major limitation for commercialization of algal biofuels. Strategies to maximize biomass and lipid production are crucial for improving the economics of using microalgae for biofuels. Selection of suitable algal strains, preferably from indigenous habitats, and further improvement of those ‘platform strains’ using mutagenesis and genetic engineering approaches are desirable. Conventional approaches to improve biomass and lipid productivity of microalgae mainly involve manipulation of nutritional (e.g. nitrogen and phosphorus) and environmental (e.g. temperature, light and salinity) factors. Approaches such as the addition of phytohormones, genetic and metabolic engineering, and co-cultivation of microalgae with yeasts and bacteria are more recent strategies to enhance biomass and lipid productivity of microalgae. Improvement in culture systems and the use of a hybrid system (i.e. a combination of open ponds and photobioreactors) is another strategy to optimize algal biomass and lipid production. In addition, the use of low-cost substrates such as agri-industrial wastewater for the cultivation of microalgae will be a smart strategy to reduce production costs. Such systems not only generate high algal biomass and lipid productivity, but are also useful for bioremediation of wastewater and bioremoval of waste CO2. The aim of this review is to highlight the advances in the use of various strategies to enhance production of algal biomass and lipids for biofuel feedstock.  相似文献

In this study, we analyze the impact of fertilizer‐ and manure‐induced N2O emissions due to energy crop production on the reduction of greenhouse gas (GHG) emissions when conventional transportation fuels are replaced by first‐generation biofuels (also taking account of other GHG emissions during the entire life cycle). We calculate the nitrous oxide (N2O) emissions by applying a statistical model that uses spatial data on climate and soil. For the land use that is assumed to be replaced by energy crop production (the ‘reference land‐use system’), we explore a variety of options, the most important of which are cropland for food production, grassland, and natural vegetation. Calculations are also done in the case that emissions due to energy crop production are fully additional and thus no reference is considered. The results are combined with data on other emissions due to biofuels production that are derived from existing studies, resulting in total GHG emission reduction potentials for major biofuels compared with conventional fuels. The results show that N2O emissions can have an important impact on the overall GHG balance of biofuels, though there are large uncertainties. The most important ones are those in the statistical model and the GHG emissions not related to land use. Ethanol produced from sugar cane and sugar beet are relatively robust GHG savers: these biofuels change the GHG emissions by −103% to −60% (sugar cane) and −58% to −17% (sugar beet), compared with conventional transportation fuels and depending on the reference land‐use system that is considered. The use of diesel from palm fruit also results in a relatively constant and substantial change of the GHG emissions by −75% to −39%. For corn and wheat ethanol, the figures are −38% to 11% and −107% to 53%, respectively. Rapeseed diesel changes the GHG emissions by −81% to 72% and soybean diesel by −111% to 44%. Optimized crop management, which involves the use of state‐of‐the‐art agricultural technologies combined with an optimized fertilization regime and the use of nitrification inhibitors, can reduce N2O emissions substantially and change the GHG emissions by up to −135 percent points (pp) compared with conventional management. However, the uncertainties in the statistical N2O emission model and in the data on non‐land‐use GHG emissions due to biofuels production are large; they can change the GHG emission reduction by between −152 and 87 pp.  相似文献

The seeds of many nondomesticated plant species synthesize oils containing high amounts of a single unusual fatty acid, many of which have potential usage in industry. Despite the identification of enzymes for unusual oxidized fatty acid synthesis, the production of these fatty acids in engineered seeds remains low and is often hampered by their inefficient exclusion from phospholipids. Recent studies have established the feasibility of increasing triacylglycerol content in plant leaves, which provides a novel approach for increasing energy density of biomass crops. Here, we determined whether the fatty acid composition of leaf oil could be engineered to accumulate unusual fatty acids. Eleostearic acid (ESA) is a conjugated fatty acid produced in seeds of the tung tree (Vernicia fordii) and has both industrial and nutritional end‐uses. Arabidopsis thaliana lines with elevated leaf oil were first generated by transforming wild‐type, cgi‐58 or pxa1 mutants (the latter two of which contain mutations disrupting fatty acid breakdown) with the diacylglycerol acyltransferases (DGAT1 or DGAT2) and/or oleosin genes from tung. High‐leaf‐oil plant lines were then transformed with tung FADX, which encodes the fatty acid desaturase/conjugase responsible for ESA synthesis. Analysis of lipids in leaves revealed that ESA was efficiently excluded from phospholipids, and co‐expression of tung FADX and DGAT2 promoted a synergistic increase in leaf oil content and ESA accumulation. Taken together, these results provide a new approach for increasing leaf oil content that is coupled with accumulation of unusual fatty acids. Implications for production of biofuels, bioproducts, and plant–pest interactions are discussed.  相似文献

Thyme plants are known for their production of aromatic oils, whose main component is terpenes. The plants leach terpenes to their surroundings and thereby affect the seed germination and biomass of associated plants, but also potentially themselves. A variation in the dominant terpenes produced by thyme plants is found both within and among species. In Denmark two thyme species (Thymus pulegioides and T. serpyllum) are naturally occurring. The essential oil of T. pulegioides in Denmark is mainly dominated by one monoterpene; ‘carvacrol’. In contrast, the essential oil of T. serpyllum constitutes a mix of two–four different types of terpenes, both mono‐ and sesqui‐terpenes. As the effects of terpenes on plant performance can vary with the type of terpene, and in order to study species‐specific responses, we examined how the dominating T. pulegioides monoterpene ‘carvacrol’ affected germination and growth of both T. pulegioides and T. serpyllum. We compared the performance of seeds and seedlings of both thyme species on soil treated with carvacrol versus control soil. We found no effect of treatment on germination, but we detected a highly significant effect of treatment on seedling biomass. For both thyme species, seedling biomass was significantly higher on terpene soil compared to control soil, suggesting a general adaptation to the presence of terpenes in the soil for both thyme species. Moreover, while no difference in seedling biomass between species on control soil was found, T. pulegiodes seedlings were significantly larger than T. serpyllum when grown on soil treated with its ‘home’ terpene, suggesting an additional species specific response. Dividing the biomass into aboveground and root biomass showed that the increased biomass on terpene‐soil was due to increased aboveground biomass, whereas no difference in root biomass was detected among treatments and species. We discuss whether this response may be caused by an adaptation to a predictable terpene‐mediated alteration in nitrogen‐availability.  相似文献

The genetically tractable microalga Chlamydomonas reinhardtii has many advantages as a model for renewable bioproducts and/or biofuels production. However, one limitation of C. reinhardtii is its relatively low‐lipid content compared with some other algal species. To overcome this limitation, we combined ethane methyl sulfonate mutagenesis with fluorescence‐activated cell sorting (FACS) of cells stained with the lipophilic stain Nile Red to isolate lipid hyperaccumulating mutants of C. reinhardtii. By manipulating the FACS gates, we sorted mutagenized cells with extremely high Nile Red fluorescence signals that were rarely detected in nonmutagenized populations. This strategy successfully isolated several putative lipid hyperaccumulating mutants exhibiting 23% to 58% (dry weight basis) higher fatty acid contents than their progenitor strains. Significantly, for most mutants, nitrogen starvation was not required to attain high‐lipid content nor was there a requirement for a deficiency in starch accumulation. Microscopy of Nile Red stained cells revealed that some mutants exhibit an increase in the number of lipid bodies, which correlated with TLC analysis of triacyglycerol content. Increased lipid content could also arise through increased biomass production. Collectively, our findings highlight the ability to enhance intracellular lipid accumulation in algae using random mutagenesis in conjunction with a robust FACS and lipid yield verification regime. Our lipid hyperaccumulating mutants could serve as a genetic resource for stacking additional desirable traits to further increase lipid production and for identifying genes contributing to lipid hyperaccumulation, without lengthy lipid‐induction periods.  相似文献

Within the bioenergy debate, the ‘food vs. fuel’ controversy quickly replaced enthusiasm for biofuels derived from first‐generation feedstocks. Second‐generation biofuels offer an opportunity to produce fuels from dedicated energy crops, waste materials or coproducts such as cereal straw. Wheat represents one of the most widely grown arable crops around the world, with wheat straw, a potential source of biofuel feedstock. Wheat straw currently has limited economic value; hence, wheat cultivars have been bred for increased grain yield; however, with the development of second‐generation biofuel production, utilization of straw biomass provides the potential for ‘food and fuel’. Reviewing the evidence for the development of dual‐purpose wheat cultivars optimized for food grain and straw biomass production, we present a holistic assessment of a potential ideotype for a dual‐purpose cultivar (DPC). An ideal DPC would be characterized by high grain and straw yields, high straw digestibility (i.e. biofuel yield potential) and good lodging resistance. Considerable variation in these traits exists among current wheat cultivars, facilitating the selection of improved individual traits; however, increasing straw yield and digestibility could potentially have negative trade‐off impacts on grain yield and lodging resistance, reducing the feasibility of a single ideotype. Adoption of alternative management practices could potentially increase straw yield and digestibility, albeit these practices are also associated with potential trade‐offs among cultivar traits. Benefits from using DPCs include reduced logistics costs along the biofuel feedstock supply chain, but practical barriers to differential pricing for straw digestibility traits are likely to reduce the financial incentive to farmers for growing higher ‘biofuel‐quality’ straw cultivars. Further research is required to explore the relationships among the ideotype traits to quantify potential DPC benefits; this will help to determine whether stakeholders along the bioenergy feedstock supply chain will invest in the development of DPCs that provide food and fuel potential.  相似文献

As one of the characteristics of marginal lands, salinization is a major limitation to agricultural production. The present work aimed to investigate the possible factors involved in the tolerance of M. sinensis C4 photosynthesis to salinity stress.
  • Seedlings of two accessions (salt‐tolerant ‘JM0119’ and salt‐sensitive ‘JM0099’) were subjected to 0 mm NaCl (control) or 250 mm NaCl (salt stress treatment) for 2 weeks. The chlorophyll content, parameters of photosynthesis and chlorophyll a fluorescence, activity of C4 enzymes and expression of C4 genes were measured.
  • The results showed that photosynthesis rate, transpiration rate, chlorophyll content, PSII operating efficiency, coefficient of photochemical quenching, activity of phosphoenolpyruvate carboxylase (PEPC) and pyruvate, orthophosphate dikinase (PPDK) and gene expression of PEPC and PPDK under salinity were higher after long‐term salinity exposure in ‘JM0119’ than in ‘JM0099’, while activity of NADP‐malate dehydrogenase (NADP‐MDH) and NADP‐malic enzyme (NADP‐ME), together with expression of NADP‐MDH and NADP‐ME, were much higher in ‘JM0099’ than in ‘JM0119’.
  • In conclusion, the increased photosynthetic capacity under long‐term salt stress in the salt‐tolerant relative to the salt‐sensitive M. sinensis accession was mainly associated with non‐stomatal factors, such as reduced chlorophyll loss, higher PSII operating efficiency, enhanced activity of PEPC and PPDK and relatively lower activity of NADP‐ME.

Determining which factors contribute to the formation and maintenance of genetic divergence to evaluate their relative importance as a cause of biological differentiation is among the major challenges in evolutionary biology. In Spodoptera frugiperda (Smith) (Lepidoptera: Noctuidae) two host strains have been recognized in the 1980s: the corn‐strain prefers maize, sorghum, and cotton, whereas the rice‐strain prefers rice and wild grasses. However, it is not clear to what extent these so‐called ‘strains’, which have also been called ‘host races’ or even ‘sibling species’, are really associated with host plants. Due to the indeterminate evolutionary status, we will use the term ‘host forms’ (sensu Funk). Here, we characterized populations collected from maize, rice, and wild grasses from three countries in South America. Using two mitochondrial cytochrome oxidase I (mtCOI) markers and 10 polymorphisms in the triose phosphate isomerase (Tpi) gene, we found various patterns of host association. Two hundred twenty‐seven nuclear amplified fragment length polymorphisms (AFLPs) markers revealed significant genetic differentiation among populations, which was generally correlated to the host from which the larvae were collected. Using a multivariate discriminant analysis and a Bayesian clustering approach, we found that individuals could be grouped into 2–5 genetically distinct clusters, depending on the method. Together, our results indicate that although host‐associated differentiation is present in this species, it does not account for all observable genetic variation and other factors must be maintaining genetic differentiation between these forms. Therefore, the term ‘host strains’ should be abandoned and ‘host forms’ should be used instead for S. frugiperda.  相似文献

Microalgal neutral lipids [mainly in the form of triacylglycerols (TAGs)], feasible substrates for biofuel, are typically accumulated during the stationary growth phase. To make microalgal biofuels economically competitive with fossil fuels, generating strains that trigger TAG accumulation from the exponential growth phase is a promising biological approach. The regulatory mechanisms to trigger TAG accumulation from the exponential growth phase (TAEP) are important to be uncovered for advancing economic feasibility. Through the inhibition of pyruvate dehydrogenase kinase by sodium dichloroacetate, acetyl‐CoA level increased, resulting in TAEP in microalga Dunaliella tertiolecta. We further reported refilling of acetyl‐CoA pool through branched‐chain amino acid catabolism contributed to an overall sixfold TAEP with marginal compromise (4%) on growth in a TAG‐rich D. tertiolecta mutant from targeted screening. Herein, a three‐step α loop‐integrated metabolic model is introduced to shed lights on the neutral lipid regulatory mechanism. This article provides novel approaches to compress lipid production phase and heightens lipid productivity and photosynthetic carbon capture via enhancing acetyl‐CoA level, which would optimize renewable microalgal biofuel to fulfil the demanding fuel market.  相似文献

Glycolate is produced in autotrophic cells under high temperatures and Ci‐limitation via oxygenation of ribulose‐1,5‐bisphosphate. In unicellular algae, glycolate is lost via excretion or metabolized via the C2 cycle by consuming reductants, ATP and CO2 emission (photorespiration). Therefore, photorespiration is an inhibitory process for biomass production. However, cells can be manipulated in a way that they become glycolate‐producing ‘cell factories’, when the ratio carboxylation/oxygenation is 2. If under these conditions the C2 cycle is blocked, glycolate excretion becomes the only pathway of photosynthetic carbon flow. The study aims to proof the biotechnological applicability of algal‐based glycolate excretion as a new biotechnological platform. It is shown that cells of Chlamydomonas can be cultivated under specific conditions to establish a constant and long‐term stable glycolate excretion during the light phase. The cultures achieved a high efficiency of 82% of assimilated carbon transferred into glycolate biosynthesis without losses of function in cell vitality. Moreover, the glycolate accumulation in the medium is high enough to be directly used for microbial fermentation but does not show toxic effects to the glycolate‐producing cells.  相似文献

Diatoms (Bacillarophyceae) are photosynthetic unicellular microalgae that have risen to ecological prominence in oceans over the past 30 million years. They are of interest as potential feedstocks for sustainable biofuels. Maximizing production of these feedstocks will require genetic modifications and an understanding of algal metabolism. These processes may benefit from genome‐scale models, which predict intracellular fluxes and theoretical yields, as well as the viability of knockout and knock‐in transformants. Here we present a genome‐scale metabolic model of a fully sequenced and transformable diatom: Phaeodactylum tricornutum. The metabolic network was constructed using the P. tricornutum genome, biochemical literature, and online bioinformatic databases. Intracellular fluxes in P. tricornutum were calculated for autotrophic, mixotrophic and heterotrophic growth conditions, as well as knockout conditions that explore the in silico role of glycolytic enzymes in the mitochondrion. The flux distribution for lower glycolysis in the mitochondrion depended on which transporters for TCA cycle metabolites were included in the model. The growth rate predictions were validated against experimental data obtained using chemostats. Two published studies on this organism were used to validate model predictions for cyclic electron flow under autotrophic conditions, and fluxes through the phosphoketolase, glycine and serine synthesis pathways under mixotrophic conditions. Several gaps in annotation were also identified. The model also explored unusual features of diatom metabolism, such as the presence of lower glycolysis pathways in the mitochondrion, as well as differences between P. tricornutum and other photosynthetic organisms.  相似文献
