We tested hypotheses concerning the specificity of interactions between insect vectors and mollicute plant pathogens in a 22-month study of leafhoppers collected at three agricultural field sites in Mexico. The common species collected, Dalbulus maidis, D. elimatus, D. gelbus, and D. guevari were equally likely to test positive for corn stunt spiroplasma (CSS) in ELISA, and to transmit maize bushy stunt phytoplasma (MBSP) to test maize seedlings. We documented intraspecific variation in the ability of D. maidis to transmit confirmed CSS infections. Dalbulus guevari and D. gelbus were less successful in transmitting CSS than D. maidis from the same population. Our results suggest this vector-plant pathogen interaction is not specific to a single Dalbulus-mollicute combination, and that both the range of potential vectors in agricultural fields, and intraspecific variation across populations of these vectors, should be the focus of future work.  相似文献

The importance of color stimuli in host-finding by Dalbulus maidis (DeLong & Wolcott), D. gelbus DeLong, and D. quinquenotatus DeLong & Nault (Homoptera: Cicadellidae) was determined by comparing pre- and post-contact responses of leafhoppers to maize seedlings (Zea mays L.) with those to vertical models varying in hue or value (total amount of reflected light). For each species, more contacts were made with maize by jumping/flying than by walking, and both sexes remained on maize for similar times after contact. When presented simultaneously with seven models (vertical wooden rods) painted in different colors, each species made more contacts with yellow. Yellow also received more contacts when it was present in a group of four colors. There were no differences in contacts made with the yellow model by jumping/flying or by walking for D. maidis and D. gelbus, but D. quinquenotatus made more contacts by jumping/flying than by walking. If yellow was not present in the group, the color reflecting maximally between 500–580 nm received more contacts compared to the other three colors. Responses to a series of neutrals indicated contacts were elicited primarily because of hue, and not changes in value. Yellow also elicited tenure as long or significantly longer than other colors after contact with a model. For each species, there were no differences in tenure between males and females on models. The importance of color stimuli in host-finding by Dalbulus leafhoppers is suggested to be influenced by differences in flight behavior and habitat stability of field hosts.
Résumé Nous avons examiné l'importance de la couleur dans la découverte de l'hôte, en comparant au laboratoire les réactions avant et après contacts avec des plants de maïs,-à 4 feuilles-, et avec des leurres peints présentant des différences de nuance ou de quantité de lumière réfléchie, chez 3 espèces de Dalbulus: D. maidis,-spécialiste du maïs-, D. gelbus,-qui consomme le maïs et Tripsacum, et D. quinquenotatus,-spécialiste de Tripsacum. Les espèces entrent en contact avec le maïs plus en voltigeant que par la marche, mâles et femelles séjournant plusieurs heures après être entrés en contact. En utilisant des leurres végétaux,-des morceaux de bois verticaux peints de différentes couleurs-, une forte réponse au jaune, due à la teinte plus qu'à l'intensité, a été mise en évidence. Il n'y a pas de différence dans les contacts après voltige ou après marche avec des leurres jaunes pour D. maidis et D. gelbus, tandis que chez D. quinquenotatus, ils sont plus fréquents après voltige. Ces 3 espèces de Dalbulus continuent à être influencées par la couleur après contact, puisque chaque espèce reste plus longtemps sur le jaune que sur les autres couleurs. L'importance de la stimulation par la couleur dans la découverte des hôtes par Dalbulus est semble-t-il due au comportement de vol et à la stabilité de l'habitat dans la nature.

Divergence of abundant genomic elements among the Zea and Tripsacum genera was examined cytologically and a tool kit established for subsequent studies. The LTR regions from the CRM, Huck, Grande, Prem1, Prem2/Ji, Opie, Cinful-1, and Tekay retroelement families were used as FISH probes on mitotic chromosome spreads from a "trispecies" hybrid containing chromosomes from each of three species: Zea mays (2n = 20), Z. diploperennis (2n = 20), and Tripsacum dactyloides (2n = 36). Except for Tekay, which painted both Zea and Tripsacum chromosomes with nearly equal intensity, the retroelement probes hybridized strongly to the Zea chromosomes, allowing them to be distinguished from those of Tripsacum. Huck and Grande hybridized more intensely to maize than to Z. diploperennis chromosomes. Tripsacum genomic clones containing retroelement sequences were isolated that specifically paint Tripsacum chromosomes. The retroelement paints proved effective for distinguishing different genomes in interspecific hybrids and visualizing alien chromatin from T. dactyloides introgressed into maize lines. Other FISH probes (180-bp knob, TR-1, 5S, NOR, Cent4, CentC, rp1, rp3, and alpha-ZeinA) could be simultaneously visualized with the retroelement probes, emphasizing the value of the retroelement probes for cytogenetic studies of Zea and Tripsacum.  相似文献

Plant biomass has attracted interest as a feedstock for biofuels production, but much of this work has been focused on relatively few plant species. In this study, three relatively-unstudied species of warm-season perennial grasses, grown at multiple locations in the eastern and central US and harvested over a three year period, were examined for fermentability via in vitro ruminal gas production and dry matter digestibility assays, and near-infrared reflectance calibrations were developed for these fermentation parameters. Big bluestem (Andropogon gerardii Vitman) displayed greater fermentability than did sand bluestem (Andropogon hallii Hack) or eastern gamagrass [Tripsacum dactyloides (L.) L.], but displayed lower biomass yields. The bluestems also displayed lower N contents and less variation in fermentability over different growth environments (geographic locations and harvest years), suggesting a more consistent biomass quality than for eastern gamagrass. Thus, in addition to their use as forage for ruminant animals, bluestems may be of particular interest as feedstocks for bioconversion to ethanol and other products via direct microbial fermentation (consolidated bioprocessing) schemes, and thus merit additional efforts to enhance biomass yield potential.  相似文献

为分析玉米着丝粒卫星DNA(CentC)和着丝粒反转录转座子(CRM)在玉米种的亚种及近缘种中的保守性,采用双色荧光原位杂交检测了这两种重复序列在墨西哥玉米、二倍体多年生类玉米、多年生类玉米、摩擦禾、薏苡、高粱中的存在和分布。CentC、CRM探针在墨西哥玉米、二倍体多年生类玉米和多年生类玉米的所有染色体的着丝粒区产生了强或较强的杂交信号, 而且不同染色体的杂交信号的强度存在差异, 表明两种玉米着丝粒重复序列在不同着丝粒中的数量不同; 此外, 部分着丝粒中的CentC和CRM信号的强度存在差异, 不完全重叠。CentC、CRM探针仅在摩擦禾的多数染色体的着丝粒区产生了弱的杂交信号。在薏苡和高粱中仅测检到主要分布在着丝粒区的较强或强的CRM信号。这些结果表明, CentC在玉米种的亚种间及玉蜀黍属的物种间高度保守, 在与玉蜀黍属亲缘关系最近的摩擦禾属物种中也具有较高的保守性; CRM在与玉蜀黍属亲缘关系较近和较远的禾本科种属中都具有保守性。  相似文献

Although the corn leafhopper Dalbulus maidis (DeLong and Wolcott) is the most important vector of maize pathogens in Latin America, little is known about how and where it overwinters (passes the dry season), particularly in Mexico. The objectives of this study were (1) to monitor the abundance of D. maidis adults throughout the dry season in maize and maize-free habitats and (2) to determine where and how D. maidis adults, exposed or nonexposed to the maize pathogen Spiroplasma kunkelii Whitcomb, overwinter in a maize-free habitat. Work for the first objective was done during the two consecutive dry seasons of 1999-2000 and 2000-2001; the second objective was done during the dry seasons of 2003-2004 and 2005-2006. During the dry winter seasons, D. maidis was prevalent as long as maize was present in irrigated areas. The leafhopper was found in 52 of the 58 irrigated maize fields sampled in Mexico at the end of the dry seasons of 1999-2000 and 2000-2001. However, leafhopper adults were not found in nonirrigated maize-free habitats at high elevation during the dry winter season (February, March, and April), although leafhopper adults were prevalent on perennial wild grasses in January after maize harvest. Additional experiments revealed, however, that corn leafhopper adults, although few in number, survived the entire dry season in these nonirrigated maize-free fields. Also, no detectable difference in survival existed between leafhoppers exposed and those not exposed to S. kunkelli during the two dry seasons in the maize-free habitat.  相似文献

Combined with a system for identifying each of the chromosomes in a genome, visualizing the location of individual genetic loci by fluorescence in situ hybridization (FISH) would aid in assembling physical and genetic maps. Previously, large genomic clones have been successfully used as FISH probes onto somatic chromosomes but this approach is complicated in species with abundant repetitive elements. In this study, repeat-free portions of sequences that were anchored to particular chromosomes including genes, gene clusters, large cDNAs, and portions of BACs obtained from public databases were used to label the corresponding physical location using FISH. A collection of probes that includes at least one marker on each chromosome in the maize complement was assembled, allowing a small-target karyotyping system to be developed. This set provides the foundation onto which additional loci could be added to strengthen further the ability to perform chromosomal identification in maize and its relatives. The probes were demonstrated to produce signals in several wild relatives of maize, including Zea luxurians, Z. diploperennis, and Tripsacum dactyloides.  相似文献

In Zea mays L., we studied the molecular evolution of Shrunken2 (Sh2), a gene that encodes the large subunits of a major enzyme in endosperm starch biosynthesis, ADP-glucose pyrophosphorylase. We compared 4669 bp of the Sh2 coding region on 50 accessions of maize and teosinte. Very few nucleotide polymorphisms were found when compared with other genes in Z. mays, revealing an effect of purifying selection in the whole species that predates domestication. Additionally, the comparison of Sh2 sequences in all Z. mays subspecies and outgroups Z. diploperennis and Tripsacum dactyloides suggests the occurrence of an ancient selective sweep in the Sh2 3' region. The amount and nature of nucleotide diversity are similar in both maize and teosinte, confirming previous results that suggested that Sh2 has not been involved in maize domestication. The very low level of nucleotide diversity as well as the highly conserved protein sequence suggest that natural selection retained effective Sh2 allele(s) long before agriculture started, making human selection inefficient on this gene.  相似文献

Eastern gamagrass, (Tripsacum dactyloides L.) is a perennial, warm-season grass that is being developed as a forage plant. Shoots were derived from callus initiated from immature embryos and immature inflorescences of diploid (2n=2x=36) gynomonoecious eastern gamagrass. These shoots were induced to microtiller in the presence of 3 mg/l benzyladenine. Amiprophosmethyl (10, 15, or 20 μm) was applied to 27 microtillers for 3–5 days to induce chromosome doubling. All 14 surviving plants were tetraploid, (2n=4x=72), as determined by flow cytometry or chromosome counts. These plants were morphologically normal and produced seed. Test crosses were made with a known diploid. Flow cytometry and chromosome counts showed that the progeny were triploid, proving that the induced tetraploids reproduce sexually. A relationship has been established between the expression of apomixis in natural polyploids of Tripsacum dactyloides and fertility as measured by percent seed set. Thus, fertility may be reliably used as a defining phenotype for apomixis when scoring the progeny from diploid (2n = 2x = 36) x tetraploid (2n = 4x = 72) crosses in Tripsacum. By exploiting the relationship between apomixis and fertility, as defined by seed set, analyses were performed on a set of related second-generation triploid populations segregating for apomixis. These populations were derived from sexual (diploid) x apomictic (tetraploid) crosses. Six out of 25 genome-dispersed restriction fragment length polymorphism (RFLP) markers co-segregate with fertility. Five of these markers were previously reported and include: php20855, tda48, tda53, umc62, and umc83, and are linked to Tripsacum genetic linkage groups F, I, H, L, and A, respectively. Significantly, we report here the syntenic relationships of the maize chromosome intervals to Tripsacum that segregate for numerous meiosis-specific and fertility-associated genes. Utilizing RFLP locus comparative mapping based on conservation of chromosome (genic) regions between related species, it may be concluded that the genes controlling fertility have been preserved in both Tripsacum and maize. A sixth marker, umc166, has also been shown to co-segregate with fertility and is conserved in both grass species. Specifically, umc166 is linked to Tripsacum linkage group D and, by syntenic comparison, to the short arm of maize chromosome 5. Encoded within this marked interval is the gene Ameiotic1 (Am1) whose function is required for the initiation of meiosis in both micro- and megaspore mother cells and whose absence of expression in the female is, in all likelihood, a prerequisite for the expression of apomixis.  相似文献

Some angiosperms reproduce by apomixis, a natural way of cloning through seeds. Apomictic plants bypass both meiosis and egg cell fertilization, producing progeny that are genetic replicas of the mother plant. In this report, we analyze reproductive development in Tripsacum dactyloides, an apomictic relative of maize, and in experimental apomictic hybrids between maize and Tripsacum. We show that apomictic reproduction is characterized by an alteration of developmental timing of both sporogenesis and early embryo development. The absence of female meiosis in apomictic Tripsacum results from an early termination of female meiosis. Similarly, parthenogenetic development of a maternal embryo in apomicts results from precocious induction of early embryogenesis events. We also show that male meiosis in apomicts is characterized by comparable asynchronous expression of developmental stages. Apomixis thus results in an array of possible phenotypes, including wild-type sexual development. Overall, our observations suggest that apomixis in Tripsacum is a heterochronic phenotype; i.e., it relies on a deregulation of the timing of reproductive events, rather than on the alteration of a specific component of the reproductive pathway.  相似文献

We have examined the structure of nuclear genes coding for ribosomal RNAs in maize and its wild relatives, the teosintes and Tripsacum. Digestion of the rDNA (genes coding for 18S, 5.8S and 26S RNAs) with 15 restriction endonucleases (with six base pair recognition sites) yields essentially a single map for the approximately 10,000 repeat units within an individual plant or species. Both length and site variation were detected among species and were concentrated in the intergenic spacer region of the rDNA repeat unit. This result is in agreement with patterns of rDNA change observed among wheat and its relatives (Triticeae), and among vertebrate species. Digestion of these nuclear DNAs with BamHI and subsequent hybridization with a 5S RNA gene-specific probe allowed determination of the size of the 5S gene repeat unit in maize, teosintes, and Tripsacum. Groupings in the genus Zea were characterized by distinct repeat unit types five Tripsacum species examined shared a 260 base pair major repeat unit type. Additionally, several other restriction endonuclease cleavage patterns differentiated among the 5S DNAs within the genus Zea. The rDNA and 5S DNA restriction site variation among the species can be interpreted phylogenetically and agrees with biochemical, karyotypic, and morphological evidence that places maize closest to the Mexican teosintes. For both gene arrays, contributions from each parental genome can be detected by restriction enzyme analysis of progeny from crosses between maize and two distantly related teosintes, Zea luxurians or Zea diploperennis, but certain teosinte arrays were underrepresented in some of the hybrids.  相似文献

Greenhouse and field studies were conducted to evaluate the feasibility of phytoremediation for clean-up of highly contaminated sediments from Indiana Harbor. In the greenhouse study, plant species evaluated were willow (Salix exigua), poplar (Populus spp.), eastern gamagrass (Tripsacum dactyloides), arrowhead (Sagitaria latifolia), switchgrass (Panicum virgatum), and sedge (Carex stricta). Sediments with sedge, switchgrass, and gamagrass had significantly less residual total petroleum hydrocarbons (TPH) after one year of growth (approximately 70% reduction) than sediments containing willow, poplar, or no plants (approximately 20% reduction). Although not all polycyclic aromatic hydrocarbons (PAH) had concentration differences due to the presence of plants, residual pyrene concentrations in the unvegetated pots were significantly higher than in pots containing sedge, switchgrass, arrowhead, and gamagrass. As evaluated by TPH dissipation in the upper section of the pots, the sedge, switchgrass, and gamagrass treatments had higher TPH degradation than the unvegetated, willow and poplar treatments. These trends were similar for soil at the bottom of the pots, with the exception that in the switchgrass treatment, degradation was not significantly different than in the unvegetated soil. Two target contaminants, pyrene and benzo[b]fluoranthene, showed differences in degradation between planted and unvegetated treatments. In the field study, phytoremediation plant species were eastern gamagrass (T. dactyloides), switchgrass (P. virgatum), and sedge (C. stricta). In addition, rhizosphere characteristics of arrowhead (S. latifolia) and sedge were assessed. Arrowhead- and sedge-impacted soils were found to contain significantly more PAH-degrading bacteria than unvegetated soils. However, over the 12-month field study, no significant differences in contamination were found between the planted and unplanted soils for TPH and PAH concentrations. TPH concentrations near the canal were greater than concentrations further from the canal, indicating that the canal may have served as a continuous source of contamination during the study.  相似文献

 DNA fingerprinting verified hybrid plants obtained by crossing Eastern gamagrass, Tripsacum dactyloides L., and perennial teosinte, Zea diploperennis Iltis, Doebley & R. Guzmán. Pistillate inflorescences on these hybrids exhibit characteristics intermediate to the key morphological traits that differentiate domesticated maize from its wild relatives: (1) a pair of female spikelets in each cupule; (2) exposed kernels not completely covered by the cupule and outer glumes; (3) a rigid, non-shattering rachis; (4) a polystichous ear. RFLP analysis was employed to investigate the possibility that traits of domesticated maize were derived from hybridization between perennial teosinte and Tripsacum. Southern blots of restriction digested genomic DNA of parent plants, F1, and F2 progeny from two different crosses were probed with RFLP markers specifically associated with changes in pistillate inflorescence architecture that signal maize domestication. Pairwise analysis of restriction patterns showed traits considered missing links in the origin of maize correlate with alleles derived from Tripsacum, and the same alleles are stably inherited in second generation progeny from crosses between Tripsacum and perennial teosinte. We studied the corn stunt spiroplasma (CSS), Spiroplasma kunkelii (Mycoplasmatales: Spiroplasmataceae) and its vector the corn leafhopper Dalbulus maidis (Hemiptera: Cicadellidae) under field conditions in Mexico. We surveyed for the presence of CSS in D. maidis by using PCR on samples of adults collected during the 2000–01 and 2003–04 winter (dry) seasons from irrigated low‐elevation sites and un‐irrigated high‐elevation sites. Also, we determined the body size and number of mature eggs of D. maidis females collected during the dry season in 2004 and in females collected on maize seedlings in the first months (June and July) of the wet (summer) season in 2005. Our PCR results showed that CSS was present in leafhopper adults collected during the 2000–01 and 2003–04 dry seasons in irrigated low‐elevation sites. However, in un‐irrigated high‐elevation sites, CSS was present in corn leafhopper adults caught before, but not during, the dry seasons. In these un‐irrigated high‐elevation sites, adult leafhoppers without CSS were recovered during the first 2 months (November and December) of the dry season from the foliage of wild perennial grasses. Females collected on wild perennial grasses in December 2004 showed similar head width and wing length to females caught on maize seedlings in June 2005. However, females collected on maize seedlings in July 2005 had the widest heads, longest wings and highest number of mature eggs. The largest body size of these females that arrived in July 2005 indicates that they are immigrants and support the Roff’s hypothesis that insect migrants should be larger than nonmigrants.  相似文献

The susceptibility of 22 plant species to Meloidogyne marylandi and M. incognita was examined in three greenhouse experiments. Inoculum of M. marylandi was eggs from cultures maintained on Zoysia matrella "Cavalier" or Cynodon dactylon x C. trasvaalensis "Tifdwarf". Inoculum of M. incognita was eggs from cultures maintained on Solanum lycopersicum 'Rutgers'. In each host test the inoculum density was 2,000 nematode eggs/pot. None of the three dicot species tested (Gossypium hirsutum, Arachis hypogaea, and S. lycopersicum) were hosts for M. marylandi but, as expected, M. incognita had high levels of reproduction on G. hirsutum and S. lycopersicum. Meloidogyne marylandi reproduced on all of the 19 grass species (Poaceae) tested but reproduction varied greatly (P = 0.05) among these hosts. The following grasses were identified for the first time as hosts for M. marylandi: Buchloe dactyloides (buffalograss), Echinochloa colona (jungle rice), Eragostis curvula (weeping lovegrass), Paspalum dilatatum (dallisgrass), P. notatum (bahiagrass), Sorghastrum, nutans (indiangrass), Tripsacum dactyloides (eastern gamagrass), and Zoysia matrella (zoysiagrass). No reproduction of M. incognita was observed on B. dactyloides, Cyndon dactylon (common bermudagrass), E. curvula, P. vaginatum (seashore paspalum), S. nutans, T. dactyloides, Z. matrella or Z. japonica. Reproduction of M. incognita was less than reproduction of M. marylandi on the other grass species, except for the Zea mays inbred line B73 on which M. incognita had greater reproduction than did M. marylandi (P = 0.05) and Stenotaphrum secundatum (St. Augustinegrass) on which M. incognita and M. marylandi had similar levels of reproduction.  相似文献

Pseudomonas bacteria capable of inhibiting maize fungal pathogens were isolated from caryopses of three of 11 populations of eastern gamagrass ( Tripsacum dactyloides ) collected from eastern and central United States. The bacteria inhibited fungal pathogens in pure cultures and a zone of fungal inhibition 1–2 cm wide occurred around surface sterilized maize kernels that were treated with the bacteria and placed in culture dishes containing 2% maize meal agar. In cold-soil germination tests, there were no significant ( P < 0.05) differences in germination of scarified maize kernels that were treated with gum arabic (sticking agent) and one of the bacterial strains (98%) or the commercial fungicide, Captan (96%). Both of these treatments resulted in significantly higher germination than scarification (50%) or scarification and gum arabic alone (58%). Non-scarified controls had 85% germination. The results indicate that Pseudomonas may be effective in preventing invasion of damaged seeds by pathogens capable of inhibiting germination and destroying seed embryos.  相似文献

Ribosomal internal transcribed spacer (ITS) sequences were used to evaluate the phylogenetics of Zea and Tripsacum. Maximum likelihood and polymorphism parsimony were used for phylogenetic reconstructions. Zea ITS nucleotide diversity was high compared to other plant species, but approximately equivalent to other maize loci. Coalescence of ITS alleles was rapid relative to other nuclear loci; however, there was still much diversity within populations. Zea and Tripsacum form a clade clearly differentiated from all other Poaceae. Four Zea ITS pseudogenes were identified by phylogenetic position and nucleotide composition. The phylogenetic position of Z. mays ssp. huehuetenangensis was clearly established as basal to the other Z. mays. The ITS phylogeny disfavored a Z. luxurians and Z. diploperennis clade, which conflicted with some previous studies. The introgression of Z. mays alleles into Z. perennis and Z. diploperennis was also established. The ITS data indicated a near contemporary divergence of domesticated maize and its two closest wild relatives.   相似文献

The effect of corn stunt spiroplasma (CSS) on survival and fecundity of three Dalbulus leafhopper species was determined. CSS significantly reduced the survival, as measured by the time to 50% (t50) and 25% (t25) survival, and by the scale parameter (b) of the Weibull model, for D. elimatus and D. gelbus. Fecundity of these two species, as measured by the net and gross reproductive rates, was also reduced by CSS. CSS did not significantly affect the corn leafhopper, D. maidis. In a separate experiment, maize bushy stunt mycoplasma (MBSM) reduced the survival and fecundity of D. maidis at temperatures from 20 to 29 °C. The effect of MBSM on D. maidis survival was less severe than CSS on D. elimatus and D. gelbus; t25, but not t50, was reduced by MBSM. Survival times and the cohort generation time generally declined with increasing temperature. Fecundity, however, generally increased with increasing temperature.  相似文献
