Ethylene and supraoptimal levels of 2,4-dichlorophenoxyacetic acid inhibit the growth of the apical hook region of etiolated Pisum sativum (var. Alaska) seedlings by stopping almost all cell divisions. Cells are prevented from entering prophase. The hormones also retard cell division in intact root tips and completely stop the process in lateral buds. The latter inhibition is reversed partially by benzyl adenine. In root tips and the stem plumular and subhook regions, ethylene inhibits DNA synthesis. The magnitude of this inhibition is correlated with the degree of repression of cell division in meristematic tissue, suggesting that the effect on cell division results from a lack of DNA synthesis. Ethylene inhibits cell division within a few hours with a dose-response curve similar to that for most other actions of the gas. Experiments with seedlings grown under hypobaric conditions suggest that the gas naturally controls plumular expansion and cell division in the apical region.  相似文献

A combined autoradiographic and enzyme cytochemical study ofroot apices from Pisum sativum has permitted the demonstrationof a high naphthol AS-D esterase activity in the central groupof meristematic cells immediately outside the quiescent centre.This high naphthol AS-D esterase activity indicates an expressionof the programming of these cells to form elements of the stele.A consideration is given to the need for a quantal mitosis andof its possible occurrence in the quiescent centre. It is concludedthat the programming and mitotic activity may be two physiologicalevents which whilst occurring in the same cell, are not necessarilylinked events. Phosfon-S, a substance which inhibits stem elongation, alters nucleic acid metabolism in Pisum sativum Alaska. Methylated albumin kieselguhr (MAK) columns were used to fractionate 32P-labeled nucleic acids. Phosfon-S treatment of the plants resulted in a decrease in soluble RNA and an increase in ribosomal RNA. Specific activities of the various nucleic acid fractions were lower as a result of treatment. The nucleic acids from treated tissues were more resistant to RNase degradation, and endogenous RNase activity was lower in treated tissues. When RNase treated nucleic acids were fractionated on MAK columns, the DNA-RNA fractions from treated plants had a higher specific activity than that of the control, which was not true before nuclease treatment. Spectrophotometric examination of this fraction revealed a difference in absorption spectra, possibly indicating a Phosfon-S nucleic acid complex. It is suggested that these alterations in nucleic acid metabolism could in turn alter a wide variety of metabolic processes, resulting in retarded growth.  相似文献

Incorporation of adenine-8-C14 into chromosomal nucleic acids of Drosophila salivary glands has been observed with the autoradiographic technique. RNA-C14 and DNA-C14 were detected as the fractions extractable by ribonuclease digestion and resistant to ribonuclease, respectively. Extractions with desoxyribonuclease and acids were also used to identify the nucleic acids. Time-course curves were determined from grain counts. RNA-C14 concentration reached a maximum in 2 hours, and decreased after the 4th hour. DNA-C14 concentration reached its maximum within 8 hours, and showed no decreases during a 48-hour experiment. In the latter part of the period of observation, morphological differentiation of the gland occurred, the decrease in RNA-C14 became very rapid, and a large increase in DNA-C14 was observed. Marked decrease in RNA-C14 and increase in DNA-C14 were detectable in a few hours when isotope was administered shortly before visible differentiation of the gland. Measurements of nuclear size indicated no significant decreases in RNA-C14 amount prior to the period of differentiation. During this later period, a large decrease in RNA-C14 amount occurs suddenly, and the same amount of C14 is added simultaneously to the DNA fraction, as expected if RNA-C14 is utilized in the synthesis of DNA.  相似文献

Root tips (1.0 cm) were excised, transferred to culture medium,sampled at intervals, embedded in paraffin and sectioned. Themitotic index in each primary meristem in the 2.0 mm tips wasreduced immediately after excision. After 8–12 h mitosisresumed. The position of mitotic figures in the protoderm, groundmeristem, procambium, and root cap was determined. The greatestnumber of mitotic figures occurred in the procambium, and uponexcision the smallest percent reduction also occurred in thisregion. The ground meristem showed the greatest reduction inmitotic figures. After excision, the relative contribution tothe total number of mitotic figures increased in the procambiumtissue and decreased in the ground meristem. Cell division inall primary meristems of the root tip is reduced after tip excision,but the ground meristem (cortical region) is the primary targettissue.  相似文献

Tanimoto, E. 1985. Axial distribution of glycosidases in relationto cellular growth and ageing in Pisum sativum root.—J.exp. Bot. 36: 1267–1274. Eight glycosidase activities were measured in relation to cellulargrowth and ageing along the axis of pea roots. The highest activities of ß-D-fucosidase and ß-D-glucosidasewere in the apical 1.0 mm segment containing the root cap andmeristem. Activities of -L-arabinosidase, -D-mannosidase, ß-D-galactosidase, -D-glucosidase and ß-D-xylosidase werehighest in the segment between 1 and 2 mm from the tip and containingyoung elongating cells with high growth potential. The activityof -D-galactosidase was high between 1 and 2 mm from the roottip, decreasing to a minimum 4-5 mm from the tip and increasingagain up to the base of root. This distribution of enzyme activitiesrelative to the root tip remained unchanged during 28 h whilethe root length doubled. The planes of cell division and growth were examined in thecourse of a single plastochron in the shoot apical meristemby observing the orientations of mitotic spindles. In the I1region of the apical dome, cell divisions were at first anticlinalbut 30 h before a leaf primordium emerged at this site 20 percent of the cell divisions became periclinal. These periclinaldivisions were found only in the corpus. Periclinal divisionsin the tunica were coincident with the appearance of the primordiumas a bulge. The change in the direction of growth in I1 at thesite of the incipient leaf primordium occurred without any changein the rate of growth in this region of the meristem.  相似文献

A method for measuring rates of ribonucleic acid (RNA) and deoxyribonucleic acid (DNA) syntheses using a single radioactive precursor has been devised and tested using bacterial cultures and natural assemblages of marine and freshwater microorganisms. The procedure is based upon the uptake and incorporation of exogenous [3H]adenine into cellular adenosine triphosphate and deoxyadenosine triphosphate pools which serve as the immediate precursors for the adenine incorporated into RNA and DNA, respectively. It is proposed that the DNA/RNA rate ratio is correlated with the specific growth rate of microorganisms and can be used as an index for estimating and comparing the productivities of microbial assemblages in nature. This technique can also be used to detect discontinuous growth and cell division processes which frequently occur in surface plankton populations. The DNA/RNA rate ratios measured in a variety of aquatic ecosystems ranged from 3.3 to 31.8% without significant correlation to total microbial biomass.  相似文献

The effects of abscisic acid (ABA) on individual plants of Lemna minor L. were studied. The effects on growth and metabolism of the roots were the most noticeable and the most desirable to measure. Two mg/1 of ABA inhibited the root growth rate by 60% and this was accompanied by a 60% deceleration in the rate of uridine incorporation. The uptake of uridine and leucine and the incorporation of leucine were not affected by ABA. The latent period of root growth inhibition was 1 hour, whereas the inhibition of ribonucleic acid (RNA) synthesis occurred 2 to 4 hours after application. The growth inhibition caused an accumulation of starch in the peripheral, differentiated cell layer of the cortex. Apparently, the growth inhibition by ABA was not entirely due to an inhibition of RNA synthesis, and other plausible mechanisms of growth inhibition are discussed.  相似文献

0.5-30mg.L^-1的氯化铅溶液对豌豆根尖生长无影响,但微核总数及微核率明显增高,并诱发产生染色体断片,粘连,滞后,染色体桥等异常分裂。诱导的比率与氯化铅溶液浓度和作用时间呈正相关。用40mg.L氯化铅处理的豌豆根尖生长发育受抑,微核率下降,据此认为废水排放标准中应增加遗传毒理学指标。  相似文献

Illumination or gibberellic acid treatment of etiolated barley leaf segments stimulates unrolling and results in an increased level of RNA. In contrast, segments treated with abscisic acid do not unroll and have a lower content of RNA. Gibberellic acid treatment enhanced the capacity of segments to incorporate radioactivity from 32P-orthophosphate into all the RNA components detected by gel electrophoresis; abscisic acid greatly restricted the incorporation of precursors into all the RNA fractions. In conjunction with a changed capacity for RNA synthesis it was observed that abscisic acid-treated segments had a lowered soluble DNA-dependent RNA polymerase level in comparison to gibberellic acid-treated or illuminated segments. However, the influence of growth regulators on RNA polymerase content of the segments was associated with general effects on protein level rather than a specific effect on the synthesis of polymerase enzyme.  相似文献
