Ilumination did not affect the pathway of shoot regeneration at the cut edges of epicotyl explants of Troyer citrange (Moreira-Dias et al. 2000, 2001), but signigficantly affected the number of developed shoots and the response to exogenous cytokinins. Shoot regeneration at the apical end occurred through a direct organogenic pathway without callus formation. For explants incubated in the light, this regeneration did not require cytokinin addendum, but the number of shoots formed was significantly increased by benzyl adenine, but not by zeatin or kinetin. Incubation in the dark almost suppressed shoot formation at the apical end. The addition of benzyl adenine or kinetin, but not of zeatin, restored shoot formation in the dark to the value obtained in the light. At the basal end of the explants shoot regeneration occurred through an indirect organogenic pathway after the formation of a primary callus. In explants incubated in the light, callus formation and shoot growth was supported by a low (0.5–1 mg l−1) benzyl adenine concentration and by zeatin. Kinetin did not support callus growth. Shoot formation was higher in the presence of benzyl adenine (0.5–1 mg l−1) than of zeatin, but was inhibited by a high (5 mg l−1) benzyl adenine concentration. Incubation in the dark increased callus growth and shoot formation at the basal cut as compared to explants incubated in the light. The three cytokinins tested supported callus growth and shoot formation in the dark, zeatin being the most effective and kinetin the least. In terms of number of shoots developed, the optimum cytokinin addendum depended on the pathway of organogenesis and the conditions of incubation. The maximum number of shoots developed at the apical end was obtained when the incubation was performed in the light in the presence of benzyl adenine. At the basal end, the optimal conditions were incubation in the dark in the presence of zeatin. It was not always possible to define an optimal cytokinin concentration as the curve concentration/response varied from experiment to experiment, which seemed unrelated to the endogenous cytokinin concentration in the explants.  相似文献

In an attempt to optimize somatic embryo formation in Oncidium ‘Gower Ramsey’, the effects of five auxins (2,4-D, IAA, IBA, NAA and picloram) and five cytokinins (2iP, BA, kinetin, TDZ and zeatin), used alone, was tested in vitro using root-derived callus. In general, kinetin (0.5 and 2 mg l−1) and zeatin (0.5 mg l−1) were found to be more effective than other auxin and cytokinin treatments to induce somatic embryogenesis from root-derived callus.  相似文献

Rapid Effects of Abscisic Acid on Ion Uptake in Sunflower Roots   总被引:1,自引:0,他引:1  
Short-term effects of ABA, ABA + kinetin and kinetin on ion (86Rb-potassium and phosphate) and water uptake in sunflower plants (Helianthus annuus var. californicus) were examined with a continuous-recording technique. Ion uptake in the roots and transport to the shoots were also investigated by conventional tracer uptake experiments and by sap bleeding experiments with excised roots. After addition of 5 × 10?6-4 × 10?5M ABA to the root medium there was an immediate decrease (30–70%) in the rate of ion uptake which lasted 30–70 min. The rate of water uptake was not significantly affected as measured with this method. Ion transport to the shoots and to the bleeding sap of excised roots was decreased by ABA. ABA-induced inhibition of ion uptake was abolished by the presence of kinetin, and uptake was slightly stimulated by 2 × 10?5M kinetin alone. We suggest that concentration gradients of ABA or rapid changes in the ABA-kinetin balance in the roots affect ion uptake and transport.  相似文献

The effect of N-(2-chloro-4-pyridyl)-N-phenylurea (4PU-30) on the growth and content of endogenous cytokinins of adenine type in tobacco (Nicotiana tabacum L.) callus was investigated. Biomass accumulation in calli grown on Murashige and Skoog (MS) medium with 4PU-30 was higher than that on MS medium with kinetin. The obvious presence of isopentenyladenine type cytokinins and traces of zeatin type cytokinins supposes modification in the endogenous cytokinin metabolism of the tobacco callus grown on 4PU-30.  相似文献

Summary Yellowish compact callus, induced from cowpea hypocotyls on Murashige and Skoog(MS) medium (1962) containing 0.2 mg/l(0.93 μM) kinetin and 0.4 mg/l (1.81 μM) 2,4-dichlorophenoxyacetic acid (2,4-D), was subcultured on MS medium containing cytokinin alone, auxin alone, or auxins plus cytokinins in order to determine the effect of cytokinins on root organogenesis in callus cultures. The callus actively proliferated on the same medium but did not show any organogenic activity macroscopically as well as microscopically. On medium with N6-benzyladenine (BA) and 1-naphthaleneacetic acid (NAA), the yellowish compact callus first changed to pale green compact callus and then many green spots appeared on its surface under light culture. But the yellowsih compact callus remained yellowish and white spots appeared on its surface in dark culture. These spots gradually became white nodular structures. Adventitious root formation from the nodular structures occurred not only on the same medium, but also on medium with either auxin or cytokinin but not both. Yellowish compact callus on medium with auxin alone was transformed to yellowish friable callus, which did not develop adventitious roots. The yellowish friable callus could gain rhizogenic activity only after morphological modification to pale green compact callus on medium with auxin plus cytokinin. The modified callus did not form adventitious roots on medium with auxins but only with cytokinins. Therefore, it is suggested that cytokinins have stimulating effects on root formation from callus that previously did not show rhizogenic activity on medium with auxins alone. In addition, the rhizogenic potential of cowpea callus was discriminated from that of leaf explants, which formed adventitious roots directly on medium with auxin alone.  相似文献

Kinetin has been shown to act synergistically with indole-3-acetic acid (IAA) or calcium ion (Ca2+) to stimulate ethylene production. Several commercially available cytokinins (kinetin, kinetin-riboside, benzyladenine, benzyladenine-riboside, isopentenyladenine, isopentenyladenine-riboside, and zeatin) as well as noncytokinin bases (adenine and xanthine) were administered to mung bean (Phaseolus aureus Roxb.) hypocotyls to study their effects, alone or in combination with IAA or Ca2+, on ethylene production. In the presence of IAA or Ca2+, all cytokinins tested synergistically stimulated ethylene production and were as effective or nearly as effective as kinetin. Noncytokinin bases (adenine and xanthine) were, however, inactive in this system.  相似文献

Influence of roots on the vegetative or floral development of cotyledonary buds of Scrofularia arguta Sol.: A possible cytokinin role. This study shows that the presence of “nonabsorbing roots” insures a vegetative development of cotyledonary buds cultured in vitro whereas buds growing without roots produce flowers early. In the same way, roots suppress floral expression of axillary meristems of the same cotyledonary buds and induce these buds to vegetative functioning. Various trophic modifications in the culture medium are ineffective on non-rooted buds as also are gibberellin As and adenine. On the contrary, several cytokinins (kinetin, benzyladenine and zeatin) exert the same influence as roots. These results suggest that roots regulate meristematic functioning through cytokinins.  相似文献

The combined application of 10–6 M adenine and 10–6M mevalonic acid to soya bean callus accelerated its growth.Two biologically active compounds that co-chromatographed withzeatin and isopentenyl adenine were extracted from this callus.Studies with labelled adenine and mevalonic acid indicated thatthe cytokinin-dependent soya bean callus incorporated only avery small amount of the radioactive precursors into the biologically-activecompounds, making it extremely difficult to determine whetherthese compounds were synthesized de novo or whether they aroseas by-products of tRNA turnover. In this study gas chromatographic-mass spectrometric techniques have been used to identify and quantify the metabolic incorporation of [15N5]adenine into zeatin and its metabolites by 3-week-old Datura innoxia Mill, crown gall tissue. In a parallel study the levels of endogenous cytokinins were also determined by the stable isotope dilution technique using deuterium (2H)-labeled internal standards. Incorporation levels of the [15N5]adenine after 8 hours of incubation, expressed as a percentage of the endogenous cytokinins, were as follows: zeatin (1.0%), zeatin riboside (1.5%), and zeatin riboside 5′-phosphate (10.2%). These results are consistent with those observed in complementary experiments using [U-14C]adenine, and support the proposal that the cytokinin biosynthesis occurs primarily at the nucleotide level. The effect of tissue age on cytokinin biosynthesis, determined by [U-14C]adenine incorporation into cytokinins by tissues at varying growth stages, indicated a steady increase with time reaching maximal synthesis at five weeks following subculture after which the level of 14C incorporation into cytokinins declined.  相似文献

In vitro culture is currently used to produce plant material for ex situ conservation of endangered species. In this study, an efficient protocol for shoot regeneration from leaves and roots was developed for Centaurea ultreiae, a critically endangered species. Organogenesis from leaf and root explants was promoted by incubating these explants on half-strength Murashige and Skoog (MS) medium in the presence of one of four different cytokinins [6-benzyladenine (BA), zeatin, kinetin or N6-(2-isopentenyl) adenine (2iP)], each provided at five different levels. Shoot organogenesis was induced in both explants. The best response, 90% of leaf explants producing a mean of 2.48 shoots per explants and 94.3% of root explants producing a mean of 5.60 viable shoots per explants, was observed when explants were incubated on a medium containing 0.55 μM BA. Histological studies revealed connectivity between vascular tissues of regenerated shoots and cambial cells of leaf explants. Moreover, adventitious shoots were derived from pericycle cells of root explants and parenchymatic cells of callus tissues.  相似文献

The crown-gall tissue of Vinca rosea converts labelled adenine into cytokinins. The principal initial products appear to be ribosylzeatin phosphates; zeatin and ribosylzeatin are also produced in appreciable quantities. The efficiency of conversion of adenine into cytokinins suggests a pathway of synthesis independent of turnover of tRNA. Isopentenyl adenine or its derivatives do not appear to be intermediates in the conversion of adenine to zeatin compounds. Summary Stem segments from apical shoot tips of Polygala myrtifolia were used as primary explants to establish in vitro cultures. Axillary shoots produced on non-contaminated explants were excised and recultured in the same medium to increase the stock of shoot cultures. Equal molar concentrations of five cytokinins [2-isopentenyladenine, kinetin, zeatin, N 6-benzyladenine (BA), and adenine] were tested for ability to induce axillary shoot development from double-node stem segments. The highest rate of axillary shoot proliferation was induced on Murashige and Skoog agar medium supplemented with 1.8 μM BA. Seven indole-3-acetic acid (IAA) concentrations (0, 2.9, 5.7, 8.6, 11.4, 14.3, 17.1 μM) were tested to determine the optimum conditions for in vitro rooting of microshoots. Up to 72% of the microshoots rooted with 14.3 μM IAA. Other auxins tested, α-naphthaleneacetic acid and indole-3-butyric acid, were less effective than IAA in inducing adventitious root formation. All rooted plantlets having more than three roots were successfully established in soil.  相似文献

The cytokinins kinetin and zeatin increased stomatal opening at 15°C in the dark by up to 50% in detached epidermis of the CAM plant Kalanchoe daigremontiana. The effect of different concentrations and activities of cytokinins on the morphogenesis of regenerated Rhododendron forrestii Balf. f. ex Diels. shoots taken from nodal segments were tested. We evaluated zeatin, zeatin riboside, izopentyladenine, izopentyladenine riboside, kinetin, kinetin riboside, benzylaminopurine, benzylaminopurine riboside. The experimental results were evaluated by mathematical methods and regression analysis describing the effect of isoprenic and aromatic type of cytokinins. On the basis of this modelling, maximum axillary shoot production was attained with medium supplemented with 2.0 mg·l−1 izopentyladenine riboside, 2.0 mg·l−1 benzylaminopurine and 20 g·l−1 sucrose. Minimal axillary shoots were produced with kinetin and kinetin riboside.  相似文献

When care was taken to minimise the effects of phosphatase activity during extraction ofVinca rosea crown-gall tumour tissue, a large proportion of extractable cytolinin activity was present in the nucleotide fraction. Analysis using ion-exchange chromatography followed by enzymic or chemical degradation and subsequent identification of the biologically active material indicated that this activity was due to zeatin riboside 5′-monophosphate. This was also the major radiolabelled cytokinin formed when this tissue was supplied with [14C]adenine. The incorporation of radioactivity from [14C]adenosine into free cytokinins was also shown, but no incorporation of radioactivity was found when [3H]mevalonic acid lactone was supplied to this tissue under the same conditions. In parallel experiments using normal stem callus tissue ofV. rosea, no incorporation of [14C]adenine into free cytokinins was observed. The significance of these results is discussed in relation to a possible transfer-RNA-independent pathway of cytokinin biosynthesis, operating primarily at the mononucleotide level.  相似文献

Factors influencing adventitious bud and root development, callus induction and embryogenesis were investigated in stem and leaf cultures of Petunia inflata R. E. Fries and Petunia hybrida cv. Cascade and cv. Rose du ciel grown on a synthetic nutrient medium. Indoleacetic acid caused limited callus development and root formation whereas naphthaleneacetic acid Induced abundant roots. 2,4-Dichlorophenoxyacetic acid promoted callus growth and differentiation of embryos which eventually developed into plantlets. Cytokinins such as benzyladenine, zeatin and kinetin induced bud development. A combination of auxins and cytokinins caused an interaction which was manifested in altered morphogenetic response. Thus 2,4-dichlorophenoxyacetic acid in conjunction with benzyladenine caused suppression of bud development and retarded differentiation of embryos. Likewise, when benzyladenine was used with indoleacetic acid root development was totally inhibited and abundant buds were produced.  相似文献

The naturally occurring cytokinins, zeatin, zeatin riboside and dihydrozeatin did not promote the germination of celery (Apium graveolens L.) seeds and 6-Δ2-isopentenyladenine (2iPA) and its riboside were only moderately active. Of the synthetic cytokinins, kinetin, kinetin riboside, and the disubstituted urea, N-phenyl-N′-pyridyl urea (NC5392) were moderately active, and 6-benzyl-aminopurine (BA) and its derivatives BA riboside and 6-benzyl-amino-9(tetrahydropyran-2yl)purine (SD8339) were the most active cytokinins tested. 6-(o-hydroxybenzyl)aminopurine (hyd-BA) and its naturally occurring riboside inhibited germination under normally inductive conditions. All the cytokinins examined were more active in promoting germination of lettuce (Lactuca sativa L.) than celery seeds. BA, BA riboside and SD8339 were again the most active cytokinins. In contrast to the results with celery, zeatin and zeatin riboside were highly active. The other cytokinins also showed high activity with the exception of dihydrozeatin, hyd-BA and hyd-BA riboside which were less active. Cytokinin ribosides were less active than the corresponding free bases during the early period of the lettuce seed incubation but total germination after 90 h was similar.  相似文献

Apparently free-base cytokinins can interact with cupric ions in a specific manner. Oxidation of NADH by a horseradish peroxidase system was strongly promoted by such cytokinins provided cupric ions were present. Oxidation was promoted by 5 micromolar kinetin, zeatin, 6-benzylaminopurine (BA), or 6-(Δ2-isopentenylamino)purine (2iP) but not by adenine, 6-methylaminopurine or 6,6-dimethylaminopurine. The 6-methylaminopurine promoted oxidation at 500 micromolar but adenine and 6,6-dimethylaminopurine did not. Activity of the free-base purines correlated well with their activity in cell-division assays. However, addition of methoxymethyl-, cyclohexyl-, or tetrahydropyranyl- at N-9 of BA or of ribosyl- at N-9 of BA, 2iP, kinetin, or zeatin eliminated activity in the peroxidase system. In a nonenzymic system containing cupric ions, all of the bases, including adenine, inhibited the Cu2+ -stimulated oxidation of ascorbic acid. As in the peroxidase system, the N-9 derivatives were inactive. The cytokinin promotion of NADH oxidation by peroxidase may result from an interaction of the hormones with copper, with peroxidase conferring a specificity similar to the cytokinin specificity observed in growth and development.  相似文献

Kinetin has a stimulating effect in the Avena straight-growth test. The action of different concentrations of kinetin, 2.5 × 10?7, 2.5 × 10?6 and 2.5 × 10?5M, in combination with different concentrations of IAA was studied in this test. It was shown that the effect of low IAA concentrations, 0.25 × 10?7 and 1 × 10?7M, was strongly enhanced by the addition of all the kinetin concentrations investigated. The effect of the highest IAA concentrations, 25 × 10?7 and 100 × 10?7M, on the other hand, was inhibited relatively strongly by the highest employed concentration of kinetin. The results are explained as due to a kinetin-produced increase of auxin in the coleoptile segment, which in combination with low IAA concentrations can lead to a growth stimulation and with high IAA concentrations to a growth inhibition. Since kinetin in purification and chromatography of auxin can partly follow IAA, thereby affecting the quantitative yield, it is emphasized that, prior to the test, auxin extracts containing cytokinins should be freed from the latter by, for example, gel filtration or paper electrophoresis.  相似文献

Summary A novel protocol has been developed for inducing somatic embryogenesis from leaf cultures of Decalepis hamiltonii. Callus was obtained from leaf sections in Murashige and Skoog (MS) medium supplemented with α-naphthaleneacetic acid (NAA)+N6-benzyladenine (BA) or 2,4-dichlorophenoxyacetic acid (2,4-D)+BA. Nodular embryogenic callus developed from the cut end of explants on media containing 2,4-D and BA, whereas compact callus developed on media containing NAA and BA. Upon subsequent transfer of explants with primary callus onto MS media containing zeatin and/or gibberellic acid (GA3) and BA, treatment with zeatin (13.68μM) and BA (10.65 μM) resulted in the induction of the highest number of somatic embryos directly from nodular tissue. The maturation of embryos took place along with the induction on the same medium. Embryogenic calluses with somatic embryos were subcultured onto MS basal medium supplemented with 4.56μM zeatin+10.65 μM BA. After 4wk, more extensive differentiation of somatic embryos was observed. The mature embryos developed into complete plantlets on growth regulator-free MS medium. A distinct feature of this study is the induction of somatic embryogenesis from leaf explants of Decalepis hamiltonii, which has not been reported previously. By using this protocol, complete plantlets could be regenerated through indirect somatic embryogenesis or organogenesis from leaf explants in 12–16 wk.  相似文献
