In the costal area and inland valleys of Guinea, flash floods occur frequently following heavy rain during the rainy season. Young rice seedlings are particularly vulnerable to submergence stress due to insufficient height and carbohydrate reserves. In an attempt to characterize the physiological responses of young seedlings to flash flood, a wide genetic base including O. sativa, O. glaberrima, and interspecific hybridization progenies (IHP) was used.

Twelve day-old seedlings were submerged for 7 days, and plant height, dry matter accumulation (DMA), and lodging were measured. Upland rice (O. sativa) showed greater shoot elongation, larger reduction in DMA during submergence, and higher lodging, which lead to low flash flooding resistance (FFR). The physiological traits of most O. glaberrima and upland rice (O. sativa) to flash flood were opposite to those of submergence tolerant cultivars, as evidenced from the results of a principal component analysis. The physiological response of Saligbeli was different to other O. glaberrima genotypes in terms of FFR. Saligbeli exhibited enhanced shoot elongation with the increase in DMA during submergence. These features seemed to be a unique way to cope with submergence. Some IHP adapted to lowland cultivation showed traits similar to Saligbeli. The vigorous growth of Saligbeli and IHP underwater should be further investigated for improving FFR in rice.  相似文献

Leptosphaeria maculans causes blackleg disease of canola (Brassica napus L.). Bacteria isolated from soil, canola stubble and plant parts were assayed for suppression of blackleg. In plate assays, the bacteria isolated from canola stubble had the highest agar-diffusible antifungal activity (75%), which was fungitoxic. In plant cotyledon assays, endophytes had the highest disease suppression. Bacteria with the highest disease suppression in cotyledon assays also had significant disease suppression at the three- to four-leaf stage. PCR screening for bacterial biosynthetic genes, commonly thought to be involved in plant disease suppression, revealed 22 bacteria to be positive for pyrrolnitrin. Pseudomonas chlororaphis and P. aurantiaca isolates contained the phenazine biosynthetic gene. Three Bacillus cereus isolates had the zmaR resistance gene. This study generated a novel set of primers specific to the zwittermicin A biosynthetic cluster. The PCR screening has confirmed the presence of genes encoding pyrrolnitrin (55%), phenazine (10%), zwittermicin A biosynthesis (7.5%) and zwittermicin A resistance (7.5%) from the canola phyllosphere and rhizosphere, which seems more widely distributed than genes for 2,4-diacetylphloroglucinol and pyoluteorin.  相似文献

Broomrapes (Orobanche spp.) are parasitic angiosperms, which attach to the roots of the hosts to take water and nutrients from them. No complete control measures are available to date, but breeding for resistance remains as one of the most feasible and environmentally friendly methods. However, the mechanisms governing the interaction between these parasites and the host are not yet well understood. We studied the cellular changes associated with the resistance to Orobanche crenata in faba bean as mechanisms involved or responsible for resistance. Two cultivars of faba bean, resistant and susceptible to O. crenata infection, were used. The evolution of the infection and the changes in the cell and tissue organisation and wall components of the host cells were followed and evaluated in both genotypes. Samples of compatible and incompatible interactions were fixed and sectioned, and specific cytochemical methods for different cell components were applied, results being analysed under light and epifluorescence microscopy. A higher proportion of O. crenata seedlings unable to penetrate the root was found on the resistant genotype. Reinforcement of cell walls by callose deposition hampers parasite penetration through the cortex. Lignification of endodermal cells prevents further penetration of the parasite into the central cylinder.  相似文献

The ability of different bacteria to inhibit Orobanche radical elongation was examined. Seeds of Orobanche aegyptiaca Pers. and O. cernua Loefl. were induced to germinate with the Growth Regulator GR24 in the presence or absence of test bacterium. Radical lengths were estimated microscopically on a scale relative to seed length (0–5 seed lengths) after 5 days of incubation at 25°C. The results indicated that Pseudomonas aeruginosa QUBC1, P. fluorescens QUBC3, Bacillus atrophaeus QUBC16, and B. subtilis QUBC18 significantly inhibited radical elongation (P≤0.01) of both O. aegyptiaca and O. cernua relative to control radicals, whereas Microbacterium hydrocarbonoxydans QUBC11 and Ochrobactrum anthropi QUBC13 showed less inhibitory effects. Other bacterial isolates had no inhibitory effects. Bacterial isolates were identified using the universal method in addition to morphological and biochemical features. The establishment of the inhibitory effect of the most promising isolates, B. atrophaeus QUBC16 and P. aeruginosa QUBC1 on radical elongation of both Orobanche spp. is a step towards utilizing such bacteria as biocontrol agents against O. aegyptiaca, O. cernua, and potentially other Orobanche species.  相似文献

O. crenata on the alga Halimeda tuna were sampled every 2 wk for a year off Tossa de Mar, northeastern Spain. The hydroids were most abundant from November to April when three cohorts, identified by size frequency, were present. From April to October the population consisted of two cohorts, except in June and July when the population density was low and only one cohort, of young hydroids, was identifiable. The paucity of O. crenata in summer was attributed to the high rate of turnover of host thalli, interspecifuc competition with other epiphytes, and a possible shortage of suitable planktonic prey. Recruitment was predominantly asexual, by stolons, and occurred throughout the year. Gonozoids were found only from late October to early December when recruitment by planula larvae also occurred. In winter, when the cohorts lived for the maximum of ≈6 wk, colony sizes quintuplicated and triplicated in successive 15-day periods. Although colonies (ramets) of O. cretana on H. tuna have a short life span reproduction and dispersal to other thalli by stolons means that the genets can survive beyond the life of a host, and perhaps indefinitely.  相似文献

Orobanche foetida is a chlorophyll-lacking holoparasite that subsists on the roots of plants and causes significant damage to the culture of some leguminous plants particularly faba bean. To evaluate the effect of rhizobia as a biological agent and the effect of nitrogen (N) and/or phosphorus (P) to control O. foetida parasitism in faba bean (Vicia faba minor), a trial was conducted in infested and non-infested fields with O. foetida in the Oued Beja Agricultural Experimental Unit, Tunisia. This field trial was performed during two consecutive cropping seasons using the susceptible cv. Badï, two selected rhizobia strains (Bj1 and Mat) and N combined or not with P. Mat strain showed an antagonistic effect which displayed a parasitism index two-fold lower than the control and carried two times less of emerged parasite spikes at crop maturity. Seed-yield losses caused by O. foetida infestation were very high and reached 95% for the control. Despite this high reduction, faba bean inoculated with Mat strain showed an average yield three-fold higher than the control. Broomrape parasitism did not significantly affect the protein/starch ratio of seeds for all treatments. The Mat strain is a potential candidate for developing an integrate method to control O. foetida parasitism on faba bean.  相似文献

在温室盆栽条件下,研究了接种尖孢镰刀菌蚕豆专化型和丛枝菌根(AM)真菌(摩西球囊霉Gm、扭形球囊霉Gt、根内球囊霉Gi及幼套球囊霉Ge)对灭菌连作土壤中蚕豆幼苗生长、枯萎病发生、根际镰刀菌数量和微生物代谢功能多样性的影响.结果表明: 接种AM真菌能显著增加蚕豆幼苗地上部和地下部鲜质量;接种Gm、Gt、Gi和Ge真菌使蚕豆枯萎病病情指数分别显著降低94.0%、60.0%、64.0%和94.0%,使蚕豆根际镰刀菌数量分别显著降低98.6%、74.3%、77.8%和90.4%,以Gm和Ge真菌对蚕豆枯萎病的抑制效应最好.接种Gm、Gt和Ge显著提高了根际微生物对糖类(CH)、氨基酸类(AA)、羧酸类(CA)和酚酸类(PA)碳源的利用,使蚕豆根际微生物的AWCD值分别显著提高34.4%、31.5%和50.8%;而接种Gi对AA、CA、PA类碳源利用和AWCD值均无显著影响,使微生物对CH类碳源的利用显著降低59.3%.主成分和相关分析表明, 接种AM真菌明显改变了蚕豆根际微生物的群落结构.接种Gt真菌提高了蚕豆根际微生物对AA类碳源(β-甲基-D-葡萄糖苷、D-半乳糖酸-γ-内酯、D-甘露醇、N-乙酰基-D-葡萄糖胺、D-纤维二糖)和CA类碳源(D-半乳糖醛酸)的利用,而接种Gi真菌降低了微生物对以上碳源的利用;接种Gm和Ge真菌提高了根际微生物对L-精氨酸和4-羟基苯甲酸的利用.表明糖类和羧酸类是接种Gt真菌后蚕豆根际土壤微生物利用的主要碳源, 氨基酸和酚酸类是接种Gm和Ge后蚕豆根际土壤微生物利用的主要碳源.接种AM真菌显著提高了蚕豆根际微生物的活性,改变了微生物群落结构并抑制病原菌的增殖,是AM真菌减轻了蚕豆枯萎病的发生的重要原因.不同AM真菌减轻了蚕豆枯萎病的发生与其改变微生物的碳源利用密切相关.  相似文献

本文以发芽率、发芽速度指数、发芽指数、根长、茎长和生物量为种子萌发和幼苗生长参数,研究不同生长时期巴茅草叶片和茎秆水浸提液对白菜、生菜、水稻的化感作用。结果表明: 巴茅草叶水浸提液化感作用强于茎秆水浸提液,叶水浸提液处理后受体植物的发芽指数和生物量均显著低于茎水浸提液。枯萎期巴茅草的化感作用强于生长旺盛期。不同浓度巴茅草叶水浸提液对3种作物的化感作用存在明显的量效关系,浸提液浓度越高,巴茅草的化感抑制作用越强。巴茅草叶水浸提液对白菜和生菜各萌发指标100%抑制的浓度分别为0.075和0.10 g·mL-1;而0.10 g·mL-1巴茅草叶水浸提液对水稻发芽率、发芽速度指数、发芽指数的抑制率分别为13.8%、27.2%、19.3%。巴茅草叶水浸提液对白菜和生菜各生长指标100%抑制的浓度分别为0.05和0.10 g·mL-1;而0.10 g·mL-1巴茅草叶水浸提液对水稻根长、茎长、生物量的抑制率分别为64.6%、92.9%、21.8%。结合种子萌发和幼苗生长的综合化感指数,3种供试作物对巴茅草化感作用的敏感程度为白菜>生菜>水稻。  相似文献

通过田间小区试验,研究了小麦与蚕豆间作对蚕豆枯萎病发病率、病情指数、根际镰刀菌数量、蚕豆根系抗氧化酶活性和膜质过氧化的影响.采用Biolog ECO板分析了根际土壤微生物的代谢功能多样性,通过高效液相色谱(HPLC)测定了蚕豆根际土壤中酚酸含量.结果表明: 与单作蚕豆相比,小麦与蚕豆间作有降低蚕豆枯萎病发病率的趋势;发病盛期和发病末期,间作使蚕豆枯萎病的病情指数比单作显著降低15.8%和22.8%,明显提高了蚕豆根际微生物活性(AWCD值),根际微生物的Shannon多样性指数显著提高4.4%和5.3%,丰富度指数显著提高19.4%和37.1%.主成分分析表明,发病盛期和发病末期,间作明显改变了蚕豆根际微生物的群落结构,蚕豆根际镰刀菌数量分别降低53.8%和33.1%;并显著降低了蚕豆根际土壤中对羟基苯甲酸、香草酸、丁香酸、阿魏酸、苯甲酸和肉桂酸的含量.发病盛期和发病末期,间作蚕豆根系的过氧化物酶(POD)和过氧化氢酶(CAT)活性分别显著提高了20.0%、31.3%和38.5%、66.7%,丙二醛(MDA)含量显著降低36.3%和46.3%;发病初期间作对蚕豆根系的POD、CAT酶活性和MDA含量无显著影响.小麦与蚕豆间作显著提高了蚕豆根际微生物的活性、多样性和根系抗氧化酶活性,降低了蚕豆根际土壤中酚酸含量和膜质过氧化程度,减少了土壤镰刀菌的数量,从而提高了蚕豆对枯萎病的抗性,降低了枯萎病的危害程度.  相似文献

Bacillus subtilis strain BSCBE4 and Pseudomonas chlororaphis strain PA23 (=P. aureofaciens) were effective biocontrol agents against Pythium aphanidermatum, the causal agent of damping-off of hot pepper (Capsicum annum L.) in greenhouse vegetable production systems. Application of strains BSCBE4 and PA23 at the rate of 20 g kg-1 of seed significantly increased the growth of hot pepper seedlings. The efficacies of various carriers in sustaining the population of these strains in storage were assessed. Both the antagonists survived up to 180 days of storage in peat and talc-based formulations. The two bacterial strains induced development of plant defence-related enzymes including phenylalanine ammonia lyase, peroxidase, polyphenol oxidase, phenol content, suppressed incidence of damping-off and increased growth of hot pepper seedlings.  相似文献

The L-form of Pseudomonas syringae pv. phaseolicola has been proved to induce resistance to bean halo blight.Various procedures were tested to induce the L-form of Pseudomonas syringae pv. pisi for its potential use as biocontrol agent of pea bacterial blight. Cell-wall deficient cells were induced in a liquid medium with penicillin following a protocol described for P. s. pv. phaseolicola. Cell growth on solid induction medium developed as typical granular and vacuolated structures, and characteristic colonies were observed in the first transfer. However, there was poor growth in subsequent transfers and some reversion to the parental type. To improve the induction procedure, the following new procedures were applied: (1) viability of cells was monitored during induction. The optimum induction time in liquid medium with penicillin was lower for pv. pisi than for pv. phaseolicola. Viability of L-forms in solid induction medium with penicillin was low and decreased in time. (2) the inducer ticarcillin was combined with clavulanic acid, which prevented the reversion to the parental type and (3) a range of concentrations of penicillin and ticarcillin/clavulanic acid was applied by the spiral gradient endpoint method for calculation of minimum inhibitory concentrations (MIC). Based on the results from these tests an induction method for P. s. pv. pisi L-form is proposed and the relevance of L-form is discussed for practice.  相似文献

Based on an assessment of 706 fungal isolates obtained from Canada and Europe, a group of Colletotrichum sp. isolates, tentatively identified as C. truncatum, was moderately efficacious for biocontrol of scentless chamomile (Matricaria perforata). In this study, 19 C. truncatum isolates, 11 from Canada and eight from Europe, were compared for virulence, crop safety, and minimum dew requirement for infection to narrow the selection of candidates. Applied at 1×106 spores mL-1, these isolates expressed variable virulence under controlled environments, with slightly higher variations observed on the Canadian isolates. There was also a slight difference in host specificity among the isolates tested; most isolates caused disease only on chamomile species (M. perforata and M. recutita) but two Canadian isolates also infected lentil, flax, or both. At 20°C, most isolates required more than 20 h dew for maximum infection. This requirement can be an impediment for using this fungus as a biocontrol agent in western Canada where the climate is semi-arid. Treatment of scentless chamomile at the 10-leaf stage with the herbicide metribuzin 48 h prior to fungal inoculation increased weed control to 72%, compared to 40 and 47% by the herbicide and fungus applied alone. However, a similar treatment using the herbicide bentazon did not enhance the weed control significantly as compared to the herbicide alone.  相似文献

A survey of pathogens of the root-parasitic weed Orobanche aegyptiaca Pers. was carried out in Nepal. More than 70% of the fungal strains isolated from infected plants belonged to the genus Fusarium. Other fungi isolated were Acremonium fusidioides, Alternaria alternata, Cladosporium cladosporioides, Epicoccum nigrum, Mortierella alpina, Papulaspora sp., Phoma spp., Sordaria fimicola , Rhizoctonia sp., Trichoderma spp. and Trichothecium roseum.  相似文献

The pathogenic fungus Aspergillus alliaceus has been shown to have potential for the biocontrol of Orobanche spp. (broomrape), a root parasitic plant. The effectiveness of A. alliaceus in reducing Orobanche infection was analysed using pesta granules prepared with different food formulations. The results showed that pesta granules comprising of fungal mycelia/spore mixtures from liquid and solid culture, sclerotia and fungal mycelia reduced Orobanche infection to a greater extent in below ground conditions when applied early and at high doses before crop sowing. In addition, pesta granules eliminated the risk of broomrape contamination within a 0.2–0.3 cm diameter of the granules. The sclerotial pathogenicity of A. alliaceus was compared with those of other fungi reported in other studies. In addition, some morphological and histological studies on the fungal pathogenicity on broomrape plants after infection are presented. The present study reveals the potential of sclerotial A. alliaceus pesta granule applications for long-term broomrape biocontrol under field conditions.  相似文献

A prediction model based on predicting the development time of the egg to the infective stage of the parasite O. ostertagi using temperature is described for bovine ostertagiasis. Two isolates of Ostertagia spp., having different capacities for infectivity and fecundity are considered. Predictions based on the capacities of each of the two isolates are compared with that observed in an animal management system studied in 1979. Infectivity and fecundity are shown to markedly influence the predicted patterns of pasture contamination.  相似文献

Crenate broomrape (Orobanche crenata Forsk.) is a serious long‐standing parasitic weed problem in Algeria, mainly affecting legumes but also vegetable crops. Unresolved questions for parasitic weeds revolve around the extent to which these plants undergo local adaptation, especially with respect to host specialization, which would be expected to be a strong selective factor for obligate parasitic plants. In the present study, the genotyping‐by‐sequencing (GBS) approach was used to analyze genetic diversity and population structure of 10 Northern Algerian O. crenata populations with different geographical origins and host species (faba bean, pea, chickpea, carrot, and tomato). In total, 8004 high‐quality single‐nucleotide polymorphisms (5% missingness) were obtained and used across the study. Genetic diversity and relationships of 95 individuals from 10 populations were studied using model‐based ancestry analysis, principal components analysis, discriminant analysis of principal components, and phylogeny approaches. The genetic differentiation (F ST) between pairs of populations was lower between adjacent populations and higher between geographically separated ones, but no support was found for isolation by distance. Further analyses identified four genetic clusters and revealed evidence of structuring among populations and, although confounded with location, among hosts. In the clearest example, O. crenata growing on pea had a SNP profile that was distinct from other host/location combinations. These results illustrate the importance and potential of GBS to reveal the dynamics of parasitic weed dispersal and population structure.  相似文献

Fusarium oxysporum f.sp. orthoceras, a potential biocontrol agent against the parasitic weed Orobanche cumana Wallr., has proven to be efficacous under greenhouse conditions when formulated as wheat-kaolin granules. To help minimize the loss of viable fungal propagules during the formulation process, the addition of a commercial fertilizer containing the iron chelate of EDDHA to the formulation was investigated. The proportion of surviving propagules was significantly increased after adding the fertilizer. However, growing conidia in fertilizer-amended liquid medium did not reduce losses in viability during formulation. The efficacy of the formulated fungus and the storability at room temperature for the first 3 months was not affected by the iron fertilizer. The protective effect could neither be obtained with the chelator EDDHA alone nor with FeEDTA or FeSO4.  相似文献

Rhizobacteria were isolated from seedlings of seven economically important weeds and characterized for potential phytopathogenicity, effects on seedling growth, and antibiosis to assess the possibility of developing deleterious rhizobacteria as biological control agents. The abundance and composition of rhizobacteria varied among the different weed species. For example, fluorescent pseudomonads represented from 11 to 42% of the total rhizobacterial populations from jimsonweed and lambsquarters, respectively. Other bacteria frequently isolated were nonfluorescent pseudomonads, Erwinia herbicola, Alcaligenes spp., and Flavobacterium spp. Only 18% of all isolates were potentially phytopathogenic, based on an Escherichia coli indicator bioassay. However, the proportion of isolates that inhibited growth in seedling assays ranged from 35 to 65% depending on the weed host. Antibiosis was most prevalent among isolates of fluorescent Pseudomonas spp., the activity of which was due to siderophore production in over 75% of these isolates. Overall, rhizobacterial isolates exhibited a complex array of properties that were inconsistent with accepted definitions for plant growth-promoting and deleterious rhizobacteria. It is suggested that for development of effective biological control agents for weed control, deleterious rhizobacteria must be screened directly on host seedlings and must possess several properties including high colonizing ability, specific phytotoxin production, and resistance or tolerance to antibiotics produced by other rhizosphere microorganisms, and they must either synthesize or utilize other bacterial siderophores.  相似文献

Salinity exerted a distinctly differential effect on fructose-1,6-bisphosphatase (EC. 3.1.3.11) isolated from salt-sensitive and salt-tolerant rice (Oryza sativa) varieties. Cytosolic and chloroplastic isoforms of the enzyme from salt-sensitive rice seedlings exhibited decreased catalytic activity during growth in the presence of salt. Furthermore, chloroplastic fructose 1,6-bisphosphatase purified from salt-sensitive (O. sativa cv. IR26) and from the wild halophytic rice Porteresia coarctata differed in their in vitro salt tolerance property although they exhibited otherwise identical biochemical and immunological properties. This decline in enzyme activity was not correlated with de novo synthesis of the chloroplastic fructose-1,6-bisphosphatase protein in the presence of salt. The inhibitory effect of increasing concentration of NaCl on in vitro enzymatic activity could be prevented by preincubation of the enzyme with a number of osmolytes with an effectiveness in the order polyol>sugars. Further, the intrinsic tryptophan fluorescence of the purified rice enzyme is altered in vitro with increasing NaCl concentration which could be prevented by preincubation with inositol. Purified chloroplastic fructose-1.6-bisphosphatase from P. coarctata however, exhibits no such inhibition of enzyme activity in vitro or alteration in tryptophan fluorescence with increasing NaCl concentration.  相似文献
