随着分子生物学技术的发展, 海洋食品物种鉴定方法由原来的蛋白质水平深入到了DNA水平。目前应用于海洋食品物种鉴定的DNA技术主要是FINS(Forensically informative nucleotide sequencing)、PCR-RFLP和物种特异性PCR标记技术等, 能够实现对新鲜、冰冻、腌制或灌装食品物种进行鉴定, 而对混合样本的鉴定及量化分析是尚待解决的一个问题。基因数据库对物种鉴定的影响也越来越大, 是海洋加工食品物种鉴定可利用的另一种重要信息资源。文章综述了DNA技术在海洋食品物种鉴定中的应用研究进展, 并展望DNA技术在海洋食品检测中的发展趋势。  相似文献

The countries of the Arabian Gulf are hot and arid and both water and agricultural resources are strictly limited. The direct use of sea water in pickle processing was investigated. Different vegetables were fermented using brines prepared from filtered sea water, filtered and boiled sea water and distilled water. Average salt concentration of sea water was found to be 4%, which is the required initial salinity for vegetable fermentations. The calcium and magnesium content of sea water was found to improve firmness and colour of the pickled products. The rates of growth of the micro-organisms in the different brines did not vary and the final pH of the brines was 3.8. Average titratable acidities of 1% were obtained. No Pb, Hg of faecal coliforms were detected in the pickled products and an average total coliform count of 2613 ml−1 in the brines was observed. Sensory quality evaluation of the pickled products showed no significant difference between treatments.  相似文献

【背景】泡梨是云南省常见的一种腌渍水果,在云南加工食用已经有一百多年的历史,因其味道酸甜可口、风味独特而深受人们喜爱,而目前对泡梨中微生物种群的系统分析和发酵原理的研究尚未见报道。【目的】研究乳酸菌在云南泡梨中的分布及应用,阐明乳酸菌种类对泡梨发酵中风味物质的影响。【方法】从云南省4个不同地区采集12份泡梨样品,经菌落菌体形态、生理生化特性和16SrRNA基因序列分析进行菌种分离与鉴定。利用分离的乳酸菌为菌种进行泡梨的制备,采用GC-MS技术对人工接种的复合乳酸菌发酵与自然发酵泡梨进行风味物质的分析与感官评价。【结果】分离鉴定出79株植物乳杆菌(Lactobacillus plantarum)、 3株类植物乳杆菌(Lactobacillus paraplantarum)、1株戊糖乳杆菌(Lactobacillus pentosus)、1株干酪乳杆菌(Lactobacillus casei)、2株副干酪乳杆菌(Lactobacillus paracasei)和1株短乳杆菌(Lactobacillus brevis),植物乳杆菌为泡梨发酵中的优势菌。将分离所得乳酸菌用于泡梨制备的结果表明,...  相似文献

Five groups of 20 infested skins with different ectoparasites and different levels of infestation and two groups of negative control skins from sheep and goats were examined for their corresponding defects at the pickled or wet blue stage of processing in tanneries. In addition, an analysis of skin defects was made from randomly selected processed skins at Kombolcha and Dessei tanneries. The prevalence of ‘ekek’ (cockle) at the pickled stage in Damalina ovis and Melophagus ovinus-infested sheep skins were 100 and 95%, respectively. Pickled goat skins affected by sarcoptic mange and Linognathus spp. were 100 and 0% positive for ‘ekek’ (cockle) lesion, respectively. The prevalence in control sheep and goat skins were 15 and 0%, respectively. There was a strong association (p < 0.05) between ‘ekek’ and infestation with M. ovinus and D. ovis in sheep and sarcoptic mange in goats. Follow-up of randomly selected 1000 pickled sheep skins and 1000 wet blue goat skins revealed that 71% of pickled sheep and 42% of wet blue goat skins had ‘ekek’ lesions. As the proportion of ‘ekek’ increased, the quality of graded skins decreased both in sheep and goats. Both on pickled sheep and wet blue goat skins, scratch and scars were found to have a strong association (p < 0.05) with ‘ekek’. The annual economic loses in 2002/2003 due to ‘ekek’ at the two tanneries was estimated to be 1.6 million USD for pickled sheep and 0.6 million USD for wet blue goat skins.  相似文献

The keys to identifying different species normally rely heavily on morphological characteristics. However, when an animal has been killed for food or sport, these markers are often destroyed or intentionally removed from the animal. This presents a problem for government agencies who are involved in determining the species origin of an animal or products derived from it in order to enforce conservation and/or health-related regulations. The problem is compounded if the meat of the animal has been processed in any way. We have developed a procedure called FINS (Forensically Informative Nucleotide Sequencing) that overcomes these problems. FINS has four components. First, methods have been developed that can isolate DNA from a wide range of biological samples including processed foods (e.g., canned, partially cooked, pickled, salted or smoked). Second, a specific segment of DNA is amplified using PCR. Third, the nucleotide sequence of the amplified segment of DNA is determined. Fourth, this nucleotide sequence is subjected to a phylogenetic analysis using a database, and the most closely related species is identified. FINS is a rapid, reliable and reproducible procedure that is based on established techniques. This procedure fills the need for an accurate method of determining the species identity of a specimen when this is not possible by conventional means.  相似文献

Excessive removal of structural material from skin during leather processing results in unattractive crease formation in leather. It is difficult to detect this in pelts at an early processing stage as it only becomes really apparent once the skin is made into leather. There would be great advantages in detecting the problem at the pickled pelt stage (skins treated with sodium sulphide and lime, bated with enzymes, and then preserved in NaCl and sulphuric acid) so that adjustments to the processing could be made to mitigate the effect. A novel bio-sensor for inspection of pickled lamb pelts has been fabricated and developed. The sensor has the planar Interdigital structure. The experimental results show that the sensor has a great potential to predict the quality of leather in a non-invasive and non-destructive way.  相似文献

Fungal infection represents a severe problem that decreases the yield and market value of fruit crops. The use of fungicides is a conventional method to control infections but it is associated with disadvantages, such as hazardous impact on public health, environmental contamination, resistance development among pathogens and high cost of agrochemicals. Biological control is an alternative approach for the treatment of fungal infections. The species of Bacillus, Pseudomonas, Enterobacter, Pantoea, Burkholderia, Lysobacter and Serratia have been successfully used in the control of fungal infections. The mechanisms involved in biocontrol are hyperparasitism or predation, production of antibiotics, lytic enzymes and induction of host resistance. Lactic acid bacteria have been used as biopreservative organisms in food and feed systems. They are a cluster of Gram-positive bacteria and include species of the genera Enterococcus, Lactobacillus, Leuconostoc, Lactococcus and Pediococcus. The ability to produce several antibacterial and antifungal substances confers biopreservation potential to lactic acid bacteria. Many have ‘generally regarded as safe’ status and are considered as safe from both human and environmental points of view. Their isolation is reported from vegetables, aerial plant surfaces, pickled cabbage, grass silage, malted cereals and also from soil. They produce antifungal substances, such as cyclic dipeptides, proteinaceous compounds, organic acids, fatty acids and reuterin. The biocontrol potential of lactic acid bacteria is demonstrated in the prevention of fungal infections of fruits, such as apples and grapes. Thus, living cells or product formulations of antifungal lactic acid bacteria may be prepared and used as an alternative biocontrol technology.  相似文献

分析了腌制蔬菜中的泡菜 ,盐浸渍菜中所含的主要营养成分的种类及其相对含量。结果显示 :泡菜和盐浸渍菜中水分、灰分、粗蛋白、粗脂肪和粗纤维的含量分别为 4 6 1% ,15 95 % ,10 2 8% ,2 19% ,2 3 2 9%和 3 11% ,2 6 49% ,15 19% ,2 42 % ,2 0 0 5 %。其氨基酸总量为 74 5 0 45mg/g ,6 6 4876mg/g ,其中必需氨基酸占 37 46 39%和43 6 76 1%。  相似文献

Aims: To screen from pickled vegetables the potential probiotic lactic acid bacteria (LAB) strains with antagonistic activity against Salmonella invasion in host. Methods and Results: Probiotic properties including acid and bile tolerance as well as inhibition on pathogenic bacteria were used for screening of LAB strains from pickled vegetables. Two strains, i.e Pediococcus pentosaceus MP12 and Lactobacillus plantarum LAP6, were selected and further assayed for their activities against Salmonella invasion in mouse liver and spleen. For these two LAB strains, strain LAP6 was able to adhere to the mouse intestinal epithelium cells. Conclusions: In screening of the probiotic strains able to inhibit the Salmonella invasion in host, factors other than the adherence to host intestinal epithelium may contribute some roles. Significance and Impact of the Study: Probiotic LAB strains with activity against Salmonella invasion in host could be isolated from vegetable origins. These strains may be used for vegetable processing.  相似文献

AIMS: This study was conducted to validate combined heat and acid treatments for inactivating Escherichia coli O157:H7, Listeria monocytogenes and Salmonella typhimurium in an acidified brine containing, or pickled, asparagus model food. METHODS AND RESULTS: A mixture of three strains of E. coli O157:H7, L. monocytogenes and S. typhimurium were inoculated onto pickled asparagus samples. Combinations of various concentrations of acetic acid [0%, 0.25%, 0.5%, 0.75%, 1%, 1.5% and 2% (v/v)] and various temperatures (40 degrees C, 50 degrees C, 60 degrees C and 75 degrees C) were investigated. Following treatment, asparagus samples were stored at room temperature and enumerated at 0, 0.5, 1, 2 and 3 days. Heat and acetic acid treatments were synergistic. The inhibitory effects of these combined treatments on the tested foodborne pathogens were also effective during storage. Loss of green colour in the pickled asparagus significantly increased with increasing concentrations of acetic acid. CONCLUSIONS: Using a combination of mild heat and acetic acid treatments can successfully control E. coli O157:H7, L. monocytogenes and S. typhimurium in pickled asparagus, combinations of heat and acid are synergistic and effective treatments can be selected to reduce adverse effect on colour which occur during product storage. SIGNIFICANCE AND IMPACT OF THE STUDY: Mild heating plus acetic acid treatment are synergistic, so combined treatments can be developed, which would lower the temperature and amount of acetic acid required for minimally processed vegetables while maintaining pathogen control.  相似文献

Two hundred cultures of lactic acid bacteria were isolated from eight yan-taozih (pickled peaches) samples, and four cultures were isolated from the main component of these samples, fresh peaches. Phenotypic and biochemical characteristics identified eight different bacterial groups (A–H) and showed that the majority of the isolates was heterofermentative lactic acid bacteria. The most common bacterial species in yan-taozih was Leuconostoc mesenteroides, although regional diversity was also observed among the lactic acid bacteria strains isolated. The antibacterial activities of the isolates were also determined, and 20 isolates were found to show inhibitory activity against the indicator strain Lactobacillus sakei JCM 1157T. The results of the sensory evaluation suggested that the diversity of lactic acid bacteria has a great effect on the aroma and formation of taste in the food product. Our findings suggest that Leuconostoc mesenteroides is the most abundant lactic acid bacteria in yan-taozih (pickled peaches) and that lactic acid bacteria strains play an important role in affecting the aroma and taste of the food product.  相似文献

以红萝卜为原料,在新配制发酵液时预先添加少量不同浓度梯度的乳酸,通过分析泡萝卜感官评价、各品质指标的动态变化规律以及有机酸和游离氨基酸测定结果,研究在直投式发酵下,经乳酸添加后的泡萝卜发酵过程以及风味品质差异,从而改进我国规模化的工业化泡菜生产工艺。结果表明,乳酸的添加能有效加快泡萝卜颜色、质地变化,缩短盐度、可溶性总糖、总酸含量达到平衡的时间,有机酸、游离氨基酸种类、含量均优于对照组泡萝卜,处理组泡萝卜整体成熟度更高。并最终结合感官评分结果确定出0.45%为最优乳酸添加量,所发酵出的泡萝卜感官品质最佳。  相似文献

榨菜为十字花科植物,是我国特有的经济作物,气候因子是影响榨菜种植分布的重要因素.通过收集榨菜分布的279个坐标点和22个高分辨率环境因子图层,利用MaxEnt模型进行重庆榨菜种植区预测,并基于该模型预测政府间气候变化专门委员会(IPCC)发布的RCP 2.6、RCP 4.5、RCP 6.0和RCP 8.5气候情景下21世纪50和70年代榨菜种植区分布范围.结果表明: MaxEnt模型的预测效果为优秀,其中,最湿月份降水量(贡献率为30.2%).年均温变化范围(17.2%)、最冷月份最低温(9.6%)、等温性(9.1%)、昼夜温差月均值(8.1%)和平均最高温度(7.5%)6个因子为主导因子,累积贡献率高达81.7%,且其各主导因子阈值分别为173~183 mm、27.2~28.3 ℃、1.8~3.8 ℃、22.5~24 ℃、6.2~6.8 ℃和14.8~18.0 ℃.在当前气候条件下,榨菜的适宜种植区比例为4.2%,主要集中在重庆涪陵的东北、西部和东部、长寿的东部和南部、垫江的南部和东南部、丰都的西北部和北部、忠县的东南部区域,以及武隆和南川的少部分区域等,中度适宜种植区面积比例为6.3%.在RCP 2.6、RCP 4.5、RCP 6.0和RCP 8.5气候情景下,预测21世纪50年代榨菜适宜生境的比例下降,分别为2.7%、3.8%、3.1%和3.2%;21世纪70年代比例也下降,分别为3.1%、3.7%、3.5%和2.9%,而中度适宜种植区的比例有所上升.  相似文献

目的为了降低"腌菜"等高盐食品中食盐的含量并防止食品发霉腐烂,探讨生姜、食盐、乳酸协同对空气中落下杂菌和易引起食品发霉腐烂的主要真菌的抗菌作用以及对"腌菜"的协同防腐效果。方法采用把空气中落下的杂菌培养后生长的杂菌总数计数、斜面接种真菌发育抑制和在试管内对黄瓜的防腐实验,检测生姜、食盐、乳酸的协同抗菌防腐作用。结果10%生姜、低浓度的食盐和乳酸对空中杂菌和供试易引起食品发霉腐烂的主要纯培养真菌仅有较弱的抗菌作用,但把三者组合后,对空气中杂菌和供试的大多数易引起食品发霉腐烂的真菌有明显的协同抗菌作用,能发育的空中杂菌总数明显少于各种单独应用的对照,对大多数纯培养真菌的发育阻止天数也明显多于各自单独应用的对照;三者对黄瓜也有协同防腐效果。结论生姜、食盐、乳酸能协同抑制引起食品发霉腐烂微生物的繁殖以及对"腌菜"(黄瓜)有协同防腐作用。  相似文献

Live corals are the key habitat forming organisms on coral reefs, contributing to both biological and physical structure. Understanding the importance of corals for reef fishes is, however, restricted to a few key families of fishes, whereas it is likely that a vast number of fish species will be adversely affected by the loss of live corals. This study used data from published literature together with independent field based surveys to quantify the range of reef fish species that use live coral habitats. A total of 320 species from 39 families use live coral habitats, accounting for approximately 8 % of all reef fishes. Many of the fishes reported to use live corals are from the families Pomacentridae (68 spp.) and Gobiidae (44 spp.) and most (66 %) are either planktivores or omnivores. 126 species of fish associate with corals as juveniles, although many of these fishes have no apparent affiliation with coral as adults, suggesting an ontogenetic shift in coral reliance. Collectively, reef fishes have been reported to use at least 93 species of coral, mainly from the genus Acropora and Porities and associate predominantly with branching growth forms. Some fish associate with a single coral species, whilst others can be found on more than 20 different species of coral indicating there is considerable variation in habitat specialisation among coral associated fish species. The large number of fishes that rely on coral highlights that habitat degradation and coral loss will have significant consequences for biodiversity and productivity of reef fish assemblages.  相似文献

The use of indicator species is popular in ecological monitoring and management. In recent years, new methods to improve the quality and application of indicator data have been proposed and developed. Here we propose the use of detection probability in the selection and application of indicator species. We evaluated environmental and observer factors believed to affect detection of potential species. Observer effects were the most evident factor and may necessitate the greatest consideration in the use of indicator species. Our results call attention to the fact that raw counts are far from accurate and that the use of detection probability can and should be incorporated into sampling protocols, species selection, and the allocation of effort for projects that use indicator species as part of monitoring and management programs.  相似文献

Theoretical predictions suggest that species‐specific signals used in the attraction of mates should evolve to reduce the risk of mismating and hybridization. These predictions lead to the hypothesis that the signals of spatially overlapping (i.e. sympatric or syntopic) species should differ more substantially than those of non‐overlapping species. Earlier studies have tested this prediction for auditory and visual signals. Here we test the hypothesis using olfactory signals, specifically the aggregation pheromones of species from two genera of bark beetles, Dendroctonus and Ips. We found no direct evidence from within these genera regarding the fact that the chemical blends that make up these pheromones differ more substantially in species that overlap in their geographical ranges and/or host‐tree use than in allopatric taxa. However, when comparing between genera, the pheromones of overlapping species appear to be more similar than non‐overlapping species. We hypothesize that the species of host tree utilized by the beetles may have some influence on their pheromone blends. Additionally, within genera, species that overlap in host use tend to be more closely related than species that use different hosts. These results may provide indirect evidence for an effect of species overlap on the evolution of bark beetle pheromones.  相似文献

Land use change alters species' abundance and distributions by affecting habitat availability and quality. The decline of bird populations worldwide is of major concern, and habitat protection and restoration are primary conservation actions. However, conservation decisions largely consider only short-term habitat changes and species’ population dynamics in a given area. Disregarding long term modifications in species’ available habitat, and the role of a given population for a species’ global population may lead to misdirected conservation action. Our goal here was to combine the assessment of conservation responsibility, with that of century-long available habitat dynamics, in order to inform better conservation practice. We compiled available habitat data for 170 bird species in the Carpathian Region from 1860 to 2010 from historic maps and satellite data. We analyzed these species’ range distributions, IUCN extinction risk and population trends, and we identified 29 species of high conservation responsibility, and all of them were forest or and grassland specialists. Furthermore, we found major land use trends including cropland abandonment and increase in forests and grasslands that resulted in increases in potential habitat for the species for which the Carpathians have high conservation responsibility. The loss of row-crop agriculture, on the other hand, reduced habitat for species for which the Carpathians do not have high responsibility, and thus subsidizing agriculture may not be warranted from a conservation perspective. More broadly, many regions worldwide are undergoing rapid land use changes, and we suggest that these should be analyzed relative to a given regions’ conservation responsibility to see if there are opportunities for conservation, i.e., cases similar to the Carpathians, where conservation efforts ‘only’ have to foster current land use trends, and make them permanent, rather than to try to revert the loss of habitat.  相似文献
