The swede midge, Contarinia nasturtii (Kieffer) (Diptera: Cecidomyiidae), is a pest of most cultivated Brassicaceae such as broccoli, canola, cauliflower, cabbage, and Brussels sprouts. The species primarily has a Palaearctic distribution and occurs throughout Europe and southwestern Asia to the Caucasus. Between 1996 and 1999, producers of cruciferous vegetables in Ontario, Canada, reported crop damage that was consistent with damage symptoms characteristic of C. nasturtii feeding and in 2000, field studies confirmed that this damage was caused by C. nasturtii . A bioclimatic model was developed to predict potential range and relative abundance of C. nasturtii in Canada in order to determine the impact of the establishment and spread of C. nasturtii populations. Model output indicated that C. nasturtii could potentially become established in all provinces of Canada, with the risk being greatest in southwestern British Columbia, southern Ontario and Quebec, New Brunswick, Nova Scotia, and Prince Edward Island. Results indicated that C. nasturtii population growth in the Prairie Ecozone of western Canada would be greatest in years with above average precipitation.  相似文献

The midge Contarinia nasturtii Kieffer (Diptera: Cecidomyiidae) was first confirmed in North America in Ontario, Canada, in 2000. The insect is now distributed throughout many counties in the provinces of Ontario and Québec. Nearly 1,200 farms in the northeastern United States that grow cruciferous vegetables are at risk for C. nasturtii infestation if this insect were to spread to that region. Over a period of 3 yr (2002-2004), approximately 3,000 ha of crops on 94 farms in western New York State was scouted for C. nasturtii, but none were found. In 2004, 42 experimental pheromone traps were placed in fields of cruciferous vegetables in eight counties. C. nasturtii males were captured at low levels (1-50 per trap / 8 wk) on four farms in Niagara County, but not at any other site. C. nasturtii larvae were found in plant tissue at one of the four farms. Insect specimens were identified by morphological methods, molecular methods, or both. This is the first confirmation of C. nasturtii in the United States, which we believe was made possible by the combined use of pheromone traps, morphological characters of trapped adults, and molecular methods. The early detection in New York presents an opportunity to implement measures to limit the spread and establishment of C. nasturtii across the state and into other regions of the United States.  相似文献

Studies were conducted on components of resistance to sorghum midge on four resistant (DJ 6514, AF 28, TAM 2566 and IS 15107) and two susceptible cultivars (CSH 1 and Swarna). Data were recorded on the numbers of eggs, larvae, emerged adults and grain damage in panicles of different genotypes infested with 60 midge females/panicle under no-choice conditions. The size of floral parts (glume, lemma, palea, lodicule, stigma, style, ovary and anther), rate of grain development and tannin content of grain were measured. The lengths of glume gl and 82, lemma L1 and L2, palea, lodicule, anther, style and stigma were positively associated with susceptibility to sorghum midge. Rate of grain development (between 3rd and 7th day after anthesis) was negatively associated with susceptibility to sorghum. Tannin content of grain was also negatively correlated with midge susceptibility, although there were distinct exceptions (e.g. DJ 6514 is highly resistant bur has a low tannin content).  相似文献

Synopeas myles (Walker) (Hymenoptera: Platygastridae) is a parasitoid of the swede midge, Contarinia nasturtii (Kieffer) (Diptera: Cecidomyiidae), in Europe. We conducted the first thorough investigation of this parasitoid’s biology. Contrary to the biology reported for all platygastrids to date, exposure of late-instar host larvae to parasitism as opposed to eggs or early larvae yielded more S. myles offspring. Superparasitism was relatively common in the field and among groups of females in the laboratory, but was much less common among single females, providing preliminary evidence for self-discrimination. Observation of immature stages of S. myles inside living hosts revealed that supernumerary larvae in superparasitized hosts were probably eliminated by physical combat soon after hatching. With increasing levels of superparasitism, sex ratios of offspring became more female-biased and their mean development time increased. The probability that offspring would emerge from a host and the size of offspring were unaffected by increasing levels of superparasitism.  相似文献

The midge Contarinia nasturtii (Keiffer), a serious gall-forming insect pest of cruciferous plants in Europe and southwestern Asia, was first reported in the United States in summer 2004. It had not been recorded in North America until its discovery in Ontario, Canada, in 2000. Efficacy of 20 insecticides belonging to 12 different classes was evaluated by using a foliar spray, soil drench, or seed treatment method. The broccoli cultivar 'Packman' was used in all tests at the suitable stage of four to five true leaves. Results indicated that foliar sprays of lambda-cyhalothrin, acephate, acetamiprid, chlorpyrifos, and methomyl reduced C. nasturtii larval populations by 96.7-100%. Except for acetamiprid, the other four insecticides also were effective against adults and provided 100% mortality after 24 h. When applied by drench, acetamiprid, imidacloprid, and thiamethoxam provided 100% control of C. nasturtii larvae, and the duration of efficacy lasted at least 7 wk. When applied as seed treatment, clothianidin and thiamethoxam provided 100% control of larvae and did not significantly affect seed germination. Imidacloprid also provided 100% control but the percentage of germination after treatment was only 62% (96.9% in check). These results indicate that several insecticides may significantly reduce midge populations. The nicotinoid class of insecticides, which has strong systemic activity, is likely to be the first choice. It is necessary to explore and develop other control methods such as cultural control and host resistance to develop an effective integrated pest management system.  相似文献

Visual stimuli influence the orientation behaviour of the sorghum midge, Contarinia sorghicola Coq. (Diptera: Cecidomyiidae). Yellow, red and white colours are attractive to the midge while blue and black are least attractive. Sorghum panicles covered with blue- or black-coloured bags in a headcage showed maximum midge damage, while the reverse was true for panicles covered with yellow, red, and white coloured bags.
Panicles at half-anthesis with viable pollen and receptive stigmata suffered higher damage than those at the pre- and post-anthesis. Physical removal of anthers and stigmata significantly reduced the oviposition by the sorghum midge. Reduced oviposition/adult emergence was also recorded in male sterile sorghum lines (2219A and 296A) or through chemically- (Ethrel) (2-Chloro ethyl-phosphonic acid) induced male sterility in panicles of the sorghum cultivar, Swarna. Chemical stimuli from viable pollen and receptive stigmata and to a limited extent physical stimuli, govern the oviposition behaviour of the sorghum midge.
Sorghum cultivars IS 12573C, S-GIRL-MR1 and IS 2816C showed antixenosis to adult midges. However, these cultivars became susceptible under no-choice conditions in the headcage. DJ 6514 and IS 12666C were attractive to the adult midges, but showed antixenosis to oviposition under natural and no-choice conditions. Genotypes with short florets showed antixenosis for oviposition. Ovary and anther breadth and tannin content of grain showed negative associations with oviposition. Cultivar antixenosis to adult midges and oviposition is an important component of resistance to the sorghum midge.  相似文献

To obtain a set of microsatellite markers for the Queensland fruit fly Bactrocera tryoni , a genomic library was screened with a number of simple repeat oligonucleotide probes. Sequencing recovered 22 repeat loci. The microsatellite sequences were short, with repeat numbers ranging from five to 11. Of these, 16 polymerase chain reaction (PCR) primer sets yielded amplifiable products, which were tested on 53 flies from five widely separated sites. All loci showed polymorphism in the population sample, with the number of alleles ranging from two to 16. Several dinucleotide repeats showed alleles separated by single-base differences and multiple steps, suggesting a mutation process more complex than the stepwise mutation model.  相似文献

The White Stork (Ciconia ciconia) has an extensive European breeding distribution. There have been significant demographic changes since the 1930s, with country-specific extinctions throughout the western distribution since the mid-1940s. Following various reintroduction programs significant concerns have been raised about the geographic origins of introduced storks, and the effect this may have on individual biology, particularly breeding success. Here we report on seven new microsatellite markers developed for C. ciconia that have been successfully combined in a series of multiplex PCR’s with six markers previously described for the Wood Stork.  相似文献

Sixteen polymorphic microsatellite markers were isolated from the European cockchafer, Melolontha melolontha L., based on enriched genomic libraries. Allelic variability was assessed in a collection of 30 individuals obtained from a field in eastern Switzerland. The number of detected alleles ranged from three to 13 with a mean of 6.69 and average observed and expected heterozygosities were 0.52 and 0.66, respectively. The 16 highly polymorphic microsatellite markers represent a powerful molecular tool, which will allow for detailed population genetic analyses on this important pest insect.  相似文献

A simple and rapid method for isolating microsatellite loci in mungbean, Vigna radiata, based on the 5′‐anchored polymerase chain reaction technique revealed 23 microsatellite loci and six cryptically simple sequence repeats. We report on the characterization of seven polymorphic microsatellite loci in V. radiata. The number of alleles per locus ranged from 2 to 5 while the observed heterozygosity ranged from 0 to 0.9048. These markers should prove useful as tools for detecting genetic variation in mungbean varieties for germplasm management and crossbreeding purposes.  相似文献

The European flounder (Platichthys flesus L.) is used in ecotoxicological studies to provide detailed information on the effects of pollution on individual fish. Data on population and evolutionary level effects are, however, limited. Here, the isolation and characterization of 28 novel species specific microsatellite loci are presented. Eight microsatellite markers were developed for the Japanese dormouse (Glirulus japonicus), a natural monument and near‐threatened species in Japan. The markers amplify in individuals from all of the mitochondrial lineages detected in a previous study. Numerous polymorphisms were detected in specimens from a local population in central Honshu (11–21 alleles per locus; n = 31) and from the entire distribution range of the species (19–41 alleles per locus; n = 152). These microsatellites will be useful in conservation genetic studies of G. japonicus.  相似文献

The Korean rockfish (Sebastes schlegeli) is an important commercial fish that is widely used in aquaculture. We isolated and characterized 18 polymorphic microsatellite loci from the Korean rockfish using a (GT)(13)-enriched genomic library. Polymorphism was assessed in 48 individuals from a single population collected from the northern coastal waters of the Yellow Sea. The observed and expected heterozygosities ranged from 0.0244 to 0.7660 (mean 0.4194) and 0.0244 to 0.8758 (mean 0.5002), respectively. Polymorphism at these loci indicated from two to 15 alleles (mean 5.7); 14 of 18 loci conformed to Hardy-Weinberg equilibrium. These markers should be useful for management and conservation studies of this species.  相似文献

Population genomics is a useful tool to support integrated pest management as it can elucidate population dynamics, demography, and histories of invasion. Here, we use a restriction site‐associated DNA sequencing approach combined with whole‐genome amplification (WGA) to assess genomic population structure of a newly described pest of canola, the diminutive canola flower midge, Contarinia brassicola. Clustering analyses recovered little geographic structure across the main canola production region but differentiated several geographically disparate populations at edges of the agricultural zone. Given a lack of alternative hypotheses for this pattern, we suggest these data support alternative hosts for this species and thus our canola‐centric view of this midge as a pest has limited our understanding of its biology. These results speak to the need for increased surveying efforts across multiple habitats and other potential hosts within Brassicaceae to improve both our ecological and evolutionary knowledge of this species and contribute to effective management strategies. We additionally found that use of WGA prior to library preparation was an effective method for increasing DNA quantity of these small insects prior to restriction site‐associated DNA sequencing and had no discernible impact on genotyping consistency for population genetic analysis; WGA is therefore likely to be tractable for other similar studies that seek to randomly sample markers across the genome in small organisms.  相似文献

Given the ecological and commercial importance of the summer flounder (Paralichthys dentatus), there is a surprising paucity of information on the molecular genetics of this species. Some studies published to date are concentrated on the reproduction biology. To address this shortcoming, a microsatellite-enriched genomic DNA library of P. dentatus was generated and screened by sequencing. Twelve dinucleotide microsatellite loci were characterized by genotyping 24 samples. The observed number of alleles ranged from three to thirteen with an average of 8.25, while the effective number of alleles ranged from 2.21 to 8.28 with an average of 5.06. The observed and expected heterozygosities ranged from 0.0833 to 0.9583 and from 0.5594 to 0.8980, respectively. Significant deviations from Hardy–Weinberg expectations were detected at three loci and linkage disequilibrium between two loci was significant after applying Bonferroni correction. In cross-species amplification, three species showed at least two polymorphic loci. The 12 highly polymorphic microsatellite markers represent a powerful molecular tool, which will allow for detailed population genetic analyses on this important marine fish.  相似文献

Primulina tabacum is a rare and endangered perennial herb with highly restricted limestone distribution in southern China. To enrich our scientific conservation for this species, we developed ten microsatellite markers using repetitive DNA enriched libraries. The number of alleles per microsatellite locus varied from two to six. The expected (H E) and observed (H O) heterozygosities varied from 0.4059 to 0.7294 and from 0.1364 to 0.5217, respectively. These markers will be employed in future studies of genetic structure in P. tabacum.  相似文献

We isolated 10 polymorphic microsatellite markers for the skink Mabuya affinis from genomic libraries enriched for (AAGG)n and (AAAG)n repetitive elements. The number of alleles ranged from eight to 13 per locus with the observed heterozygosity ranging from 0.60 to 0.92. These markers will be useful for analysis of questions concerning population genetic structure and models of speciation.  相似文献

In order to study the spatial patterns of genetic diversity of a clonal marine angiosperm, the seagrass Cymodocea nodosa, microsatellite markers were obtained by screening a genomic library enriched for the (CT) dinucleotide motif. Of 38 primer pairs defined, 15 amplified polymorphic microsatellites and are described. These loci identified a number of alleles ranging from two to seven, and showed expected heterozygosity ranging from 0.35 to 0.76, when a group of 40 individuals from Cadiz Bay in Spain was analysed. Additionally, we describe here the multiplexing conditions for 12 of these loci.  相似文献
