Seeds were harvested separately from the sequentially formed umbels to obtain information about the origin of variation in seed density in celery. The study involved seeds from the lowest order (primary) to the highest order (quarternary) umbels from the cvs Mars, Monarch, Selfira and Tall Utah. With increasing umbel order the percentage germination decreased and the mean germination time increased. Seed density increased with increasing umbel order, but was more highly correlated with the length of the seed ripening period which differed for each umbel order due to simultaneous harvesting. The decrease in seed density during seed development was correlated with an increase in air volume in the seeds. It is concluded that the variation in seed density in celery seed lots originated from differences in seed maturity.  相似文献

The morphology of flower initiation and early development in glasshouse celery (Apium graveolens L. var. dulce (Miller) Pers.) cv. Celebrity was studied by means of apical dissections and cryo-scanning electron microscopy. Easily recognisable morphological features were used to define seven stages in the early development of the inflorescence. A highly significant linear regression was established between the logarithm of the apical diameter (measured diametrically across the apical dome between the two most recently initiated leaf or inflorescence primordia) and these discrete floral stages. There was no strong evidence that either the origin or the slope of the regression varied with different combinations of temperature (viz. 10°C or 14°C) and daylength (viz. natural, short or long) which were conducive for the initiation and development of an inflorescence. It is suggested that both apical diameter and floral stage may be used as parameters for assessing the influence of environmental factors such as temperature and daylength on the floral development of glasshouse celery.  相似文献

Seeds of 4 annual cultivars of celery ( Apium graveolens L. No. 15, 16, 17 and 143) were able to germinate in the dark, under high temperature, in relatively high concentrations of NaCl solutions and in the presence of relatively high concentrations of the inhibitors of the biosynthesis of gibberellin, paclobutrazol and ancymidol. Seeds of two biennial cultivars [Florida and Earlibell (EB)] showed very low germinability, or failed to germinate under the same conditions. These results are discussed in terms of endogenous gibberellin levels, which may be higher in the seeds of annual cultivars as compared to biennial ones.  相似文献

Seed dormancy of a highly-dormant cultivar of celery (Apium graveolens L.) was broken by combinations of plant-derived smoke extract or N6-benzyladenine (BA) and gibberellins A4/7 (GA4/7) in the dark at temperatures between 18 and 26°C. A less dormant cultivar which responded to GA4/7 alone showed no additional response to smoke extract or BA. Neither smoke extract nor BA affected either cultivar in the dark in the absence of GA4/7. The partial dormancy-breaking effect of short exposures to red-light was also enhanced by smoke extracts in this highly-dormant cultivar. We report the development of nine microsatellite markers in globe artichoke (Cynara cardunculus L. var. scolymus). Four markers were obtained from sequences available in GenBank and five were isolated using a biotin/streptavidin capture technique for (CA)n and (CT)n motifs directly from artichoke genomic DNA. Polymorphism was explored in 15 artichoke accessions that represent the genetic variation within cultivated varietal types. Inter‐specific amplification was tested using cultivated cardoon (C. cardunculus L. var. altilis DC.) and wild cardoon (C. cardunculus L. var. sylvestris Lam.). Primers and conditions for polymerase chain reaction (PCR) amplification of detected loci are described.  相似文献

Plants of annual celery ( Apium graveolens L.) were treated with paclobutrazol during anthesis. Seeds collected from the treated plants showed a marked reduction in germination in light and failed to germinate in the dark. Application of GA4/7 to the imbibition solution reversed the inhibitory effect of paclobutrazol while gibberellic acid (GA3) was ineffective. Benzyladenine (BA) interaction with GA4/7 was light and concentration-dependent. At relatively low concentrations in the dark there was a synergistic effect, but at higher concentrations, especially in the light, BA, antagonized the GA4/7 effect. Seedlings emerging from the seeds from paclobutrazol-treated plants were only slightly shortened. It is suggested that paclobutrazol applied to the mother plants inhibited the biosynthesis of endogenous GAs, which normally enable the germination of annual seeds under unfavorable conditions. Exogenously applied GA4/7 fulfills the function of the absent endogenous GAs.  相似文献

The temperature-dependent, primary dormancy of cv. Florida 683 celery seeds in darkness was partially broken by a 30 min light exposure on the third day of incubation at 20–22°C, resulting in c 50 percent germination after 20 days. This light stimulation was negated by including different inhibitors of gibberellin biosynthesis in the incubation medium. Subsequent addition of a solution of the gibberellins A4 and A7 or of the gibberellin-active compound (1-3-chlorophthalimido)-cyclohexane carboxamide (AC94,377) overcame the inhibitory effects on germination of these GA-biosynthesis inhibitors. A Robertsonian translocation found in a cultivar of celery is described which is unusual because half the small product of the exchange survives as a telocentric chromosome. The change has no drastic effect on the overall fitness of the translocation homozygote. The consequences of such a change for karyotype evolution are shown to be, karyotype asymmetry, the possible genesis of B-chromosomes and the possibility of a chromosome change becoming established even though it has no adaptive value.  相似文献

Celery (Apium graveolens Linn, Family: Apiaceae) is a common edible herb used as a spice in the traditional medicine of several nations since time immemorial. The whole plant is extensively used in cooking as soups and salads. A. graveolens has various pharmacological properties such as anticancer, anti-obesity, anti-hepatotoxic, and antihypertensive agents. Hence, it is of interest to document the in vitro cytotoxic, antioxidant, and antimicrobial activity of A. graveolens. The plants were collected in the local market, shade dried, and different parts of the plants were extracted with 70% ethanol using a cold maceration process. Antioxidant tests were performed based on the various radical scavenging methods. Antimicrobial activity and MIC were completed using the respective cup-plate and two-fold serial dilution method. In vitro cytotoxic studies were achieved by the MTT; Sulphorhodamine B assayed total cell protein content. DLA and ESC cells determined the short-term toxicity. The leaf extract exhibited significant antioxidant properties against NO, DPPH, ABTS, LPO, and HPO methods. Thus, potential inhibition against Gram-positive, Gram-negative, and fungal strains within the MIC ranges of 250-500 µg/ml was observed. All the extracts of the plant presented in the study revealed greater cytotoxicity effects against five respective cancer cell lines, L6, Vero, BRL 3A, A-549, L929, and L-929 with the ranging of 443-168.5 µg/ml. Thus, we show that A. graveolens possess a potential cytotoxic, antioxidant, and antimicrobial activity.  相似文献

The germination of seeds of celery (Apium graveolens L.) becomes progressively thermoinhibited on incubation in the dark at high temperatures, the inhibitory temperature being dependent on the cultivar used. In two high-dormancy cultivars of celery, the production of germination inhibitors in seeds incubated in the dark at 26°C gradually increased over a 7-day period. Inhibitor production was measured by incubating seeds of the low-dormancy cultivar Florida 683 in homogenates of the thermoinhibited seeds of the high-dormancy cultivars and recording germination either in the light or with the gibberellins A4 and A7 (GA4/7) in the dark. Most Florida 683 seeds which failed to germinate in the homogenates after 15 days were induced to germinate by addition of N6-benzyladenine (BA). The presence of BA in addition to GA4/7 throughout incubation in the dark completely overcame the inhibitory effects of homogenates. This indicates that thermoinhibition of celery seeds is associated with the accumulation of a germination inhibitor which interacts with cytokinins. The 2 major photosynthetic products and translocated carbohydrates in celery ( Apium graveolens L.) are sucrose and the sugar alcohol, mannitol. Sucrose is produced and utilized in leaves of all ages. Mannitol, however, is synthesized primarily in mature leaves, utilized in young leaves and stored in all leaves. Here we show that mannitol export was lower from young, expanding leaves than from older leaves. After a 10 min pulse of 14CO2 and a 2 h chase in the light or dark there was more radioactivity in sucrose than in mannitol in petiole tissues from leaves of all ages. However, after a chase of 15 h in the dark or 6 h in the light followed by 9 h in the dark, mannitol was the predominant [14C]-labeled carbohydrate remaining in all leaf and petiole tissues. Thus, newly synthesized sucrose was apparently exported at a faster rate than mannitol and more mannitol was partitioned into vacuolar storage pools than was sucrose. It also appears that in the light both sucrose and mannitol were exported, but in the dark, once sucrose pools were depleted, mannitol remained as the predominant substance translocated. Both mannitol and sucrose were unloaded into petiole storage parenchyma tissue, but sucrose was hydrolyzed prior to storage.  相似文献

Celery (Apium graveolens L.) plants cv. Jason overwintered in a polythene tunnel flowered earlier and grew taller than similar plants given a 10-week cold-treatment at 5°C prior to transplanting in the same tunnel in mid-February. However, there was no significant difference in the yield of seeds obtained from both treatments, plants grown at a density of 4m-2 yielded less seeds than those at 2m-2, though the yield per unit area was slightly higher from the high density treatment. Treatment with 100 mgl-1 GA3 applied twice just prior to flowering and during anthesis increased flower stalk, flower pedicel and stamen length but delayed flower opening and seed ripening and decreased seed set and seed yield. Treatment with a mixture of 1000 mgl-1 GA4 and GA7 plus 1000 mgl-1 ethephon on three occasions during seed ripening decreased seed yield and reduced seed germination though those seeds capable of germinating were less dormant than seeds from untreated plants. The size distribution of seeds was unaffected by any treatment other than the preseeding spray with GA3 which reduced the percentage of medium-size seeds.  相似文献

为研究野生水芹的营养价值,以湖北野生水芹和旱芹为材料,对新鲜叶片和叶柄的主要营养成分进行了比较分析。结果表明:两种材料叶片和叶柄的淀粉、粗纤维和维生素B1含量的差异不明显,游离氨基酸和维生素B2含量比旱芹低,但野生水芹含有丰富的蛋白质和钙,其叶柄中可溶性糖和磷的含量分别达29.70 mg/g和0.77mg/g,分别比旱芹高57.23%和37.50%;其叶片维生素C、铁含量分别达0.19 mg/g和0.35 mg/g,比旱芹分别高137.50%和191.67%,整株黄酮类物质含量是旱芹含量的2倍。这说明野生水芹具有较高的食用价值,是一种适合作为涝渍地区保护性开发的野生蔬菜资源。  相似文献

The N-substituted phthalimide AC 94377 (1-(3-chlorophthalimido)-cyclohexanecarboxamide) was equally effective as a mixture of the gibberellins A4 and A7 (GA4/7) in breaking dormancy and stimulating germination of celery seeds when either was used in combination with ethephon or daminozide as a seed soak. Whereas seedlings emerging from GA4/7-treated seeds became etiolated in comparison with those from untreated seeds, those from AC 94377-treated seeds showed normal development. Preharvest sprays of gibberellic acid (GA3) increased the height of mature plants in comparison with untreated controls by about 16 per cent whereas AC 94377 was ineffective. The yield from GA3-treated plots was about 10 per cent greater than that from AC 94377-treated plots.  相似文献

Underdeveloped (small) embryos embedded in abundant endosperm tissue, and thus having morphological dormancy (MD) or morphophysiological dormancy (MPD), are considered to be the ancestral state in seed dormancy evolution. This trait is retained in the Apiaceae family, which provides excellent model systems for investigating the underpinning mechanisms. We investigated Apium graveolens (celery) MD by combined innovative imaging and embryo growth assays with the quantification of hormone metabolism, as well as the analysis of hormone and cell-wall related gene expression. The integrated experimental results demonstrated that embryo growth occurred inside imbibed celery fruits in association with endosperm degradation, and that a critical embryo size was required for radicle emergence. The regulation of these processes depends on gene expression leading to gibberellin and indole-3-acetic acid (IAA) production by the embryo and on crosstalk between the fruit compartments. ABA degradation associated with distinct spatiotemporal patterns in ABA sensitivity control embryo growth, endosperm breakdown and radicle emergence. This complex interaction between gibberellins, IAA and ABA metabolism, and changes in the tissue-specific sensitivities to these hormones is distinct from non-MD seeds. We conclude that the embryo growth to reach the critical size and the associated endosperm breakdown inside MD fruits constitute a unique germination programme.  相似文献

The emu (Dromaius novaehollandiae), a member of ratite family, is native to Australia and has been introduced to other countries worldwide. In this work, 10 polymorphic microsatellite loci were isolated and characterized for emu from public sequences. Polymorphism was surveyed in 22 individuals from two different populations kept in captivity. Between two and 11 alleles were found per locus, and the observed heterozygosity ranged from 0.05 to 0.85, in accordance with expectations. These markers will be useful as tools for detecting levels of genetic variation, reconstructing pedigrees (for quantitative genetic analysis) and identifying markers associated to fitness traits in emu populations.  相似文献

Somaclonal variation derived from tissue culture is a potential source of variation that can be used in crop improvement programmes. The characteristics of this variation are first shown in the regenerant generation and their heritability is then confirmed by examination of the progeny. There would be savings of time, space and labour if this variation could be detected in vitro using easily assessed visual cues. The aim of this study was to relate variation in the source of explant and the morphology of the newly initiated callus to the characteristics of the regenerant plant, of which the most important was resistance to leaf spot disease caused by Septoria apiicola. Associations were investigated by isolating four stem explants from each of 564 surface sterile seedlings, var. Celebrity, on a callus initiation medium (MS medium, 30 g litre‘1sucrose, 0.5 mg litre’12,4-D, 0.6 mg litre‘1kinetin) and assessing the morphology and colour of the callus. After this initial culture (8 wk), each callus was transferred to a regeneration medium (MS medium, 30 g litre“1sucrose). Plantlets were regenerated from many of the callus cultures and these were transferred to the glasshouse. When all of the surviving regenerant plants (276) were mature, leaf shape, amount and composition of the essential oils and resistance to late blight were assessed. Statistical analysis revealed that the character of the newly initiated callus (width, height, colour, organogenesis) showed poor correlation with all aspects of the regenerated plant measured. However, it was shown that increased variation resulted from different seedlings more than from plants derived from within seedlings or within callus.  相似文献

Non-embryogenic cells (NEC) and embryogenc cells (EC) were separated from cell clusters derived from the hypocotyl segments of celery seedlings, which had been suspension-cultured in MS medium supplemented with 105 M 2,4-D. The EC formed globular embryos in medium without 2,4-D. The globular embryo developed through heart-shaped, torpedo to cotyledonary embryos within 10 days. The EC and developing embryos were fractionated into symplastic [MeOH, hot water (HW), starch (S)] and apoplastic [pectin, hemicellulose, TFA (trifluoroacetic acid)-soluble and cellulose] fractions. The EC contained lower levels of sugar in the MeOH fraction and higher levels of starch than NEC. In the apoplastic fractions, there were no differences of total sugar amounts between NEC and EC. Cellulose contents were about 10% of the wall polysaccharides. During somatic embryogenesis, total sugar contents of the MeOH and HW fractions increased till the heart-shaped embryo stage, and then decreased during the torpedo and cotyledonary embryo stages. The sugar contents of the starch, pectin, TFA-soluble, and cellulose fractions did not change during the stages mentioned above. However, the hemicellulose substances remarkably increased during embryogenesis, and then decreased as the development proceeded. The neutral sugar components of the hemicellulosic fractions were analyzed. Arabinose increased markedly in EC to the globular embryo stage, but decreased as the development proceeded. Galactose increased only at the torpedo and cotyledonary embryo stages. Xylose was present at lower levels in all stages of embryogenesis than in the differentiated hypocotyl cell walls. These results suggest that there was a high turnover of arabinogalactan polysaccharides during embryogenesis, and that xylan accumulated in the cell walls of differentiated cells  相似文献

Celery seeds were less thermoinhibited when dried back after a seed soak treatment with the gibberellins A4 and A7 (GA4/7) plus ethephon (G+E) or an osmotic priming treatment in the light with polyethylene glycol (PEG). At temperatures between 18 and 25° in the dark, 50 percent of the PEG-treated seeds germinated after 3 days whereas G+E-treated seeds required 7 days and untreated seeds did not germinate at all.Irrespective of treatment, dry control and dried-back, treated seeds contained very little detectable cytokinin activity. However, when such seeds were imbibed for 18 h in the dark and then analysed immediately with the soybean callus bioassay, less cytokinin activity was detected in both G+E and PEG seeds than in the untreated seeds. In particular, cytokinins with the HPLC properties of zeatin and its riboside were decreased in G+E seeds and virtually absent from PEG seeds. Conversely, extracts from PEG and to a lesser extent G+E seeds contained activity which chromatographically resembled cytokinin glucosides whereas this was absent from untreated seeds.  相似文献
