The isolation and characterization of six polymorphic loci from a Testudo hermanni genomic library is reported. Enrichment was performed for AC but four of the characterized microsatellites present also an additional motif. Variability was tested on populations of the two recognized subspecies, Testudo hermanni hermanni and Testudo hermanni boettgeri. For one locus, a size range specific for the subspecies T. h. hermanni was observed. These are the first primers identified directly in the genome of T. hermanni  相似文献

We report here development and characterization of 48 novel microsatellite markers for Ropalidia marginata, a tropical, primitively eusocial polistine wasp from peninsular India. Thirty‐two microsatellites showed polymorphism in a wild population of R. marginata (N = 38) collected from Bangalore, India. These markers will facilitate answering some interesting questions in ecology and evolutionary biology of this wasp, such as population structure, serial polygyny, intra‐colony genetic relatedness and the pattern of queen succession.  相似文献

Eight tetranucleotide microsatellite loci were characterized for white hake, Urophycis tenuis, a commercially exploited demersal gadoid fish distributed in the Northwest Atlantic. The eight loci are polymorphic with number of alleles and observed heterozygosities ranging from seven to 40 and from 0.544 to 0.978, respectively, in samples of between 39 and 47 individuals collected from throughout the species range. We report eight (CA)10?35 unlinked microsatellite loci from the Neotropical freshwater catfish, Pimelodella chagresi (Siluriformes: Pimelodidae). These loci were characterized with 23 individuals collected in Panama. Number of alleles per locus varied from 7 to 23 (mean = 12.9) and observed heterozygosity ranged from 0.522 to 0.909 (mean = 0.732). These loci will be used to investigate the existence of cryptic species within the P. chagresi clade, and to study fine‐scale population structure.  相似文献

I isolated the first set of polymorphic microsatellite markers from the house finch, Carpodacus mexicanus, a well‐studied North American bird species, as part of an effort to compare levels of genetic diversity in introduced and native populations. Here, I describe eight independently assorting microsatellite loci screened for polymorphism using 40 house finches. Polymorphism levels ranged from six to 14 alleles (mean = 10.6), making these markers a powerful tool for paternity and population level analyses of this widely distributed North American species.  相似文献

The longheaded eagle ray, Aetobatus flagellum feeds mainly on bivalves, and there have been some indications that the current reduction in the bivalve population in southern Japan may be due to an increase in the number of longheaded eagle rays. Consequently, the 'predator control programme' for reducing the longheaded eagle ray population was established in 2001 in Japan. For studying the population genetics of the longheaded eagle ray, we isolated eight polymorphic microsatellite loci (two to six alleles per locus; expected heterozygosity, 0.172-0.700) from this species.  相似文献

The desert tortoise, Gopherus agassizii, is a threatened species native to the North American desert southwest and is recognized as having distinct Mojave and Sonoran populations. We identified six polymorphic microsatellite loci in the desert tortoise. All six loci were polymorphic in Sonoran samples. Five of the loci were variable in Mojave samples with varying degrees of amplification success. Two of the loci exhibited low allelic variation (2–3 alleles) while four were highly variable (8–27 alleles).  相似文献

Eight polymorphic microsatellite loci were isolated from the rainforest canopy tree, Syzygium sayeri, in order to study parentage and subsequently pollen dispersal among individuals in wild populations. Screening of one natural population (n = 64) mapped in a 500 × 500 m area at Cape Tribulation, north Queensland, Australia, yielded two to 11 alleles per locus with observed levels of heterozygosity ranging from 0.07 to 0.70. One locus was significantly out of Hardy–Weinberg equilibrium after Bonferroni correction for multiple tests. These loci should provide a useful tool in further understanding the dispersal patterns of this species.  相似文献

Primers were developed for 10 microsatellite loci for two species of Oceanodroma storm-petrels. Variability was tested in 27 O. castro and 22 O. monteiroi from the Azores, and 24 O. leucorhoa from Norway. At least six loci amplified reliably and were polymorphic in each species. The number of alleles per locus averaged 4.6, and observed heterozygosities averaged 0.41. Most primers also yielded polymerase chain reaction products in O. tethys, O. hornbyi and Pterodroma phaeopygia. These loci are being used to assay population genetic structure in storm-petrels.  相似文献

Six polymorphic microsatellite loci containing dinucleotide repeats were developed for the freshwater turtle Mauremys rivulata. The number of alleles ranged from five to 18. The observed and expected heterozygosities ranged from 0.19 to 0.79 and 0.46 to 0.90, respectively. Primer sequences for 16 microsatellite loci were developed from Schizolobium parahyba, a tropical tree species. Twelve loci were found to be polymorphic after screening diversity in individuals from Belize. A total of 39 alleles were found at nine loci. The isolation of molecular markers in Asparagus acutifolius, a wild edible plant species, is important to characterize local ecotypes that could be cultivated and preserved. We isolated and characterized polymorphic microsatellite loci from A. acutifolius by constructing and screening an enriched DNA library. Primer pairs were designed for 12 loci. Seven primer pairs worked well during amplification reactions and were tested on a wild population from Pontecagnano (SA), Italy. These loci showed a high level of genetic variability, with the numbers of alleles identified ranging from two to five and observed heterozygosity ranging from 0.20 to 0.73.  相似文献

We provide a taxonomic review of the extinct testudinid Testudo catalaunica, based on published and unpublished material from several Miocene (late Aragonian and early Vallesian) sites of the Vallès‐Penedès Basin (north‐east Iberian Peninsula). We show that Testudo catalaunica irregularis is a junior subjective synonym of T. catalaunica, and further provide an emended diagnosis of the latter based on newly reported material. Contrary to some recent suggestions, this emended diagnosis discounts an alternative attribution of T. catalaunica to Paleotestudo. The latter is merely recognized as a subgenus of Testudo, based on a cladistic analysis that assessed the phylogenetic position of all extant and most extinct species of Testudo currently recognized as valid (including T. catalaunica). Our phylogenetic analysis (which recovers the molecular phylogeny of extant Testudo s.l.) supports a taxonomic scheme in which the three extant subgenera of Testudo are represented in the fossil record. Testudo s.s. is retrieved as the sister taxon of Testudo (Agrionemys) + [Testudo (Paleotestudo) + Testudo (Chersine)]. The extinct Testudo (Paleotestudo) is therefore the sister taxon of the Testudo (Chersine) clade. The latter subgenus reveals as the most diverse clade of Testudo s.l. in the fossil record, with T. catalaunica Testudo steinheimensis constituting a subclade distinct from that including Testudo hermanni.  相似文献

Ligula intestinalis is a species with a complex biology. Several strains, distinct in the host specificity and biogeography, are supposed to exist; however, their morphological or molecular identification remains unresolved. We describe the isolation of 16 polymorphic microsatellite markers. The number of alleles ranged from four to 14 in a population of 21 L. intestinalis individuals. Loci showed amplification success across samples of worldwide distribution. A cross‐amplification of 14 markers in Digramma interrupta (Cestoda: Pseudophyllidea) was observed.  相似文献

Weigela coraeensis var. coraeensis is a deciduous shrub species distributed in Japan on the mainland, Honshu, whereas its variety W. coraeensis var. fragrans is endemic to the Izu Islands located south of Honshu. We isolated eight polymorphic microsatellite loci from the species and characterized these loci for 20 individuals from a population in Honshu. The number of alleles per locus ranged from 7 to 15 and the observed and expected heterozygosities ranged from 0.60 to 0.90 and from 0.65 to 0.90, respectively. These eight polymorphic microsatellites will be useful for examining intraspecific genetic differentiation in W. coraeensis.  相似文献

The Mediterranean yellowtail Seriola dumerilii (Risso, 1810) is a semi‐pelagic fish economically important to fisheries and aquaculture. We developed nine microsatellite primer pairs for this species. Microsatellites were isolated from a partial genomic library enriched for an AC motif. The developed loci proved to be polymorphic showing three to 11 alleles per locus. They can be employed for population studies as well as for parentage assessment. Cross‐species amplification was successfully tested in Seriola fasciata and Seriola rivoliana.  相似文献

We have developed a set of eight polymorphic nuclear microsatellite markers for the Mediterranean shrub Pistacia lentiscus by means of an enriched library method. Characterization for the eight loci was carried out on 42 individuals from two populations sampled in southern Spain. The overall number of alleles detected was 59, ranging from three to 13 per locus. Expected heterozygosity per locus and population ranged from 0.139 to 0.895. Two loci albeit only in one population (Seville) departed significantly from Hardy-Weinberg equilibrium expectations and no linkage disequilibrium between pairs of loci was detected. These markers will be used in studies of gene flow across a fragmented landscape.  相似文献

Here I report on the isolation of eight microsatellites from the winged-kelp, Alaria nana, using an enrichment protocol. Using 55 individuals from Tatoosh, WA and complete data at all loci, loci displayed 6-11 alleles with observed and expected heterozygosities of 0.396-0.754, and 0.769-0.872 respectively. Using an isolated, randomly breeding subset, six of the eight loci were determined to be in Hardy-Weinberg equilibrium. These markers will be useful in assessing population structure in the intertidal and inferring patterns of dispersal.  相似文献

We developed 11 new microsatellite markers for the European tree frog (Hyla arborea), and tested patterns of polymorphism in 54 adults (27 males and 27 females) from two ponds close to Lausanne (Western Switzerland). One marker was sex linked and two pairs displayed linkage disequilibrium. Comparisons of allele numbers with heterozygosity values support a stepwise-mutation model at neutral equilibrium, with mutation rates spanning nearly two orders of magnitude. These markers will prove useful for population genetic studies and fine-scale investigations requiring genetic assignment techniques.  相似文献
