The primary aim of this long‐term angler‐led tagging programme was to gain information about seasonal changes in distribution of the starry smooth‐hound shark Mustelus asterias, along the Dutch coast for management and conservation purposes. Between 2011 and 2014, M. asterias comprised 92·6% (n = 2418) of the total elasmobranch catch (n = 2612) by the licenced group of taggers within the Dutch Delta of which 2244 M. asterias were fin‐tagged with plastic rototags. Sex and total length (LT) composition inside the eastern tidal basin (Oosterschelde) were significantly different, i.e. more females and larger individuals, than outside indicating a pupping ground, which was confirmed by the capture of 30 newborn pups (≤32 cm). The distribution pattern of reported recaptured M. asterias (return‐rate 3·6%, n = 80) showed a circannual migration between summering in the southern North Sea and wintering in the English Channel and the Bay of Biscay, and suggests that M. asterias is philopatric. The Dutch angling season for M. asterias runs from approximately mid‐May to mid‐October when the water temperature is above 13° C. Recaptures of eight mature females, but no males in the Bay of Biscay, indicate partial spatial segregation by sex, where mature females migrate over larger distances than immature females and males. These observations, with the absence of recaptures in other known summering areas (i.e. the Irish Sea and Bristol Channel), suggest that the southern North Sea is used by a separate population. Implications for management and recommendations to improve and expand the study approach are discussed.  相似文献

At-sea observer programmes can provide spatio-temporal data on the sizes and quantities of fish being either discarded or retained, including for species and size categories that may not be sampled effectively during scientific surveys. Such data were analysed for English and Welsh fisheries operating on the continental shelf of the North Sea ecoregion (ICES Divisions 4a–c and 7d) and Celtic Seas ecoregion (ICES Divisions 6a, 7a–c, e–k) for the period (2002–2016). These data were collated for four main gear types: beamtrawl, nephrops and otter trawls, and nets (including gillnets, tangle nets and trammel nets). Beamtrawlers caught proportionally more smaller individuals than otter trawlers, whilst nets were the most size-selective gear for larger sharks. Size-related discarding was observed, with smaller spurdog Squalus acanthias and starry smooth-hound Mustelus asterias (< 60 cm total length, LT) usually being discarded. Such size-based selection was not evident for lesser-spotted dogfish Scyliorhinus canicula, which was generally landed as bait. Temporal changes in the discard–retention patterns were evident for S. acanthias and porbeagle Lamna nasus, with increased regulatory discarding as management measures became restrictive. A corresponding increase in the proportion of the non-quota M. asterias retained was also observed over the study period. Knowledge of the size-based discard–retention patterns by métier can also help inform on where future studies on discard survival could usefully be undertaken.  相似文献

Multiple paternity was investigated in the starry smooth‐hound shark, Mustelus asterias Cloquet, 1821. Analysis of 12 pregnant females and their embryos, at four microsatellite loci, showed that at least 58% carried multiply‐sired litters. Paternal skew was observed, with one male siring most of the embryos within a litter, although no patterns of association were detected between the identity of the father and the size or the uterine position of the embryos. Sperm storage was observed in the oviducal glands of all twelve females. The Northeast Atlantic and Mediterranean skates (Rajidae) showed remarkable species diversity but with high morphological and ecological conservatism. Since skates are particularly vulnerable to the bottom trawl fishery, species-specific demographic surveys as well as studies defining life history and evolutionary traits are important in prioritising conservation programs. However, the identification of juveniles and adults of some species may be difficult using referenced guidelines and identification keys. Therefore, we attempt to develop markers for species identification through the parallel analysis of a 16S rDNA gene sequence and of several morphological characters on 135 individuals collected by trawl surveys in the Adriatic Sea and putatively assigned to six taxa. Species-specific haplotypes were defined for Raja miraletus, Raja montagui, Dipturus oxyrinchus, since a solid accordance between species boundaries and well-differentiated haplotypes was observed. Comparative analysis of 16S rDNA sequences allowed the identification of three juvenile specimens of Leucoraja circularis, a species that rarely occurs in the Adriatic Sea. On the contrary, morphological traits and haplotype distribution were largely discordant in Raja asterias and R. clavata. While all putative R. clavata individuals showed a unique haplotype (H-CLA), only 8 of 30 putative R. asterias individuals possessed a second weakly divergent haplotype (H-AST). The remaining 22 R. asterias carried the H-CLA. The multivariate analyses of morphometric and meristic characters in putative R. clavata and R. asterias revealed the clustering of individuals regardless of haplotypes. However, a bimodal distribution of R. asterias and R. clavata samples would suggest that two separated taxa might exist, both sharing the two 16S rDNA haplotypes. The haplotype distribution appeared to be significantly correlated only to the standardised disc length/total length (DL/TL) variation. Three alternative explanations may support this scenario: (i) an incomplete lineage sorting process in two morphologically yet distinct taxa; (ii) a recent hybridisation between the two taxa; (iii) the two taxa are morphologically plastic species and all considered morphological characters may be misleading in discriminating between them at all maturity stages, except for the DL/TL. However, further analyses on larger data sets and using molecular key markers (i.e. nuclear genes) will be needed to definitely resolve the status of these taxa. Molecular relationships among rajid taxa are largely consistent with systematics based on internal and external anatomical features. This multidisciplinary study contributed to defining the pattern of species diversity and abundance of rajids in the Adriatic Sea.  相似文献

In this study, elasmobranch length–weight relationships were estimated for five species caught between June 1999 and May 2003 from the Cilician Basin shelf waters (Northeastern Mediterranean): Raja clavata, Raja radula, Raja asterias, Dasyatis pastinaca, and Gymnura altavela. There were no significant differences between males and females, and only Raja clavata females showed isometric growth.  相似文献

Nine polymorphic microsatellite DNA markers from the sorghum plant bug, Stenotus rubrovittatus, were isolated and characterized. These markers were used to analyse 22 individuals from a single field population. The number of alleles at these nine loci ranged from two to 28 (mean = 11.4) and heterozygosity ranged from 0.27 to 0.86 (mean = 0.58). Stenotus rubrovittatus has shown rapid population growth in the decades since the first report in the 1980s of serious damage to a rice crop. These microsatellite markers will be of value for studying both the population genetics and population dynamics of S. rubrovittatus.  相似文献

We isolated and characterized 60 novel microsatellite markers from the closely related oldfield mouse (Peromyscus polionotus) and deer mouse (Peromyscus maniculatus) for studies of conservation, ecological, quantitative and population genetics. We assessed all 60 markers in a wild population of Peromyscus polionotus rhoadsi (N = 20) from central Florida and found an average of nine alleles per marker and an observed heterozygosity (HO) of 0.66 (range = 0.00–1.00). These polymorphic markers contribute to the growing number of genomic resources for Peromyscus, an emerging model system for ecological and evolutionary research.  相似文献

Cacopsylla melanoneura is one of the main vectors of ‘Candidatus Phytoplasma mali’. We developed and described 10 microsatellite markers. Forty‐five psyllids from one population were analysed. The number of alleles ranged from six to 24 and the observed heterozygosity from 0.42 to 0.84. These markers will be useful tools for population genetic studies on C. melanoneura.  相似文献

Ten novel polymorphic microsatellite loci were developed for the freshwater sleeper Micropercops swinhonis, a relatively abundant and broadly distributed lake‐dwelling fish in Asia, using next generation sequencing. These markers were tested in a population of 48 individuals, and characteristics suggest that they may be useful for population genetic studies. The average number of alleles per marker was 6.6 (range: 2–26), the average expected heterozygosity was 0.55 (range: 0.081–0.955), and the average polymorphic information content value per marker was 0.507 (range: 0.077–0.942). No marker showed significant deviation from Hardy‐Weinberg Equilibrium following Bonferroni correction of significance values, and no markers showed evidence of null alleles, large allele dropout, or scoring errors from stutter bands. Two markers had successful amplification in Odontobutis potamophila, and one marker had successful amplification in Rhinogobius giurinus. These novel markers may be useful for further research in population genetic studies involving M. swinhonis.  相似文献

We developed 10 polymorphic microsatellite markers for the federally endangered razorback sucker, Xyrauchen texanus. PCR optimization and cross-species amplification experiments indicated that these markers will be useful for analysis of fine-scale population structure in razorback and two other sucker species; the white sucker, Catostomus commersonii and the Rio Grande sucker, C. plebeius. Alleles at locus Xte4 did not overlap when compared between razorback and bluehead (C. discobolus) suckers and permits detection of F1 hybrids. These microsatellite markers appear widely applicable for identifying genetic consequences of population decline, hatchery rearing and release, and hybridization in razorback and other castostomine suckers.  相似文献

The Roanoke logperch (Percina rex Jordan and Evermann), an endangered fish, occurs in only six watersheds in the Roanoke and Chowan river drainages of Virginia, USA. The species’ population genetic structure is poorly known. We developed 16 microsatellite markers that were reliably scorable and polymorphic P. rex. Markers were also screened in seven other darter species of the genus Percina. Most markers exhibited successful amplification and polymorphism in several species. These markers may therefore prove useful for population genetic studies in other darters, a diverse but highly imperiled group.  相似文献

We developed microsatellite markers for Maculinea nausithous and Maculinea alcon, two of five species of endangered large blue butterflies found in Europe. Two separate microsatellite libraries were constructed. Eleven markers were developed for M. nausithous and one for M. alcon. The primers were tested on both species as well as on the three other European Maculinea species. The number of alleles per locus ranged from two to 14. These markers will be useful tools for population genetic studies of Maculinea species.  相似文献

We identified 11 polymorphic microsatellite markers for Alouatta caraya. Three markers were isolated from an enriched genomic library of A. caraya (AC14, AC17, AC45), five were previously described in Homo sapiens (TGMS1, TGMS2, D5S117, D8S165, D17S804), and three were identified for Lagothix lagotricha (1110, 1118, 157). Forty‐eight individuals from one Argentinean population were genotyped, yielding heterozygosity values between 0.146 and 0.792. These markers provide an exclusion power of 0.922 when neither parent is known (0.992 when one parent is known) and are suitable for parentage analysis, population genetics and phylogeographical studies of A. caraya, the southernmost primate in the New World.  相似文献

We report here development and characterization of 48 novel microsatellite markers for Ropalidia marginata, a tropical, primitively eusocial polistine wasp from peninsular India. Thirty‐two microsatellites showed polymorphism in a wild population of R. marginata (N = 38) collected from Bangalore, India. These markers will facilitate answering some interesting questions in ecology and evolutionary biology of this wasp, such as population structure, serial polygyny, intra‐colony genetic relatedness and the pattern of queen succession.  相似文献

Coffee leaf rust due to Hemileia vastatrix is one of the most serious diseases in Arabica coffee (Coffea arabica). A resistance gene (SH3) has been transferred from C. liberica into C. arabica. The present work aimed at developing sequence-characterized genetic markers for leaf rust resistance. Linkage between markers and leaf rust resistance was tested by analysing two segregating populations, one F2 population of 101 individuals and one backcross (BC2) population of 43 individuals, derived from a cross between a susceptible and a SH3-introgressed resistant genotype. A total of ten sequence-characterized genetic markers closely associated with the SH3 leaf rust resistance gene were generated. These included simple sequence repeats (SSR) markers, sequence-characterised amplified regions (SCAR) markers resulting from the conversion of amplified fragment length polymorphism (AFLP) markers previously identified and SCAR markers derived from end-sequences of bacterial artificial chromosome (BAC) clones. Those BAC clones were identified by screening of C. arabica genomic BAC library using a cloned AFLP-marker as probe. The markers we developed are easy and inexpensive to run, requiring one PCR step followed by gel separation. While three markers were linked in repulsion with the SH3 gene, seven markers were clustered in coupling around the SH3 gene. Notably, two markers appeared to co-segregate perfectly with the SH3 gene in the two plant populations analyzed. These markers are suitable for marker-assisted selection for leaf rust resistance and to facilitate pyramiding of the SH3 gene with other leaf rust resistance genes.  相似文献

 White rust, caused by Albugo candida (Pers.) Kuntze, is an economically important disease of Brassica juncea (L.) Czern. and Coss mustard, particularly in India. The most efficient and cost-effective way of protecting mustard plants from white rust disease is through genetic resistance. The objective of this study was to identify RAPD markers for white rust resistance in an F1-derived doubled-haploid (DH) population originating from a cross between white rust-susceptible and white rust-resistant breeding lines of B. juncea from the canola-quality B. juncea breeding project of the Agriculture and Agri-Food Canada-Saskatoon Research Centre. The DH population was used to screen for RAPD markers associated with white rust resistance/susceptibility using bulked segregant analysis. Two markers, WR2 and WR3, linked to white rust resistance, flanked the resistance locus Ac2 1 and were highly effective in identifying the presence or absence of the resistance gene in the DH population. These two markers were shown to be specific to the Russian source of white rust resistance utilized in this project. Details of six highly polymorphic dinucleotide and one highly polymorphic tetranucleotide microsatellite markers are provided for Pagellus erythrinus. These markers are highly polymorphic, with an expected heterozygosity ranging from 0.713 to 0.959 and allele numbers ranging from seven to 36. Eleven polymorphic microsatellite markers were developed to delineate population structure of Ammodytes personatus. These markers had between 8 and 27 alleles. Expected heterozygosity ranged from 0.818 to 0.965, whereas the observed heterozygosity ranged from 0.441 to 0.886. Five of the eleven markers conformed to Hardy–Weinberg equilibrium. We believe the markers will be useful for genetic diversity study of A. personatus.  相似文献

Thirteen polymorphic microsatellite markers have been developed for the little greenbul, Andropadus virens. The number of alleles range from two to 15 per locus with the observed heterozygosity ranging from 0.15 to 0.92. Eight sets of primers also amplify in closely related species, Andropadus laterostris and Andropadus tephrolaemus. These markers will be useful for analysis of questions concerning population genetic structure and models of speciation.  相似文献
