Leaf orientations and light environments were recorded for 40 juvenile Pseudopanax crassifolius trees growing in New Zealand in a partially shaded, secondary forest environment. Efficiencies of interception of diffuse and direct light by the observed leaf arrangments were calculated relative to those of three hypothetical leaf arrangements. Canopy gaps above the study plants were unevenly distributed with respect to azimuth and elevation above the horizon. Our results indicate that photosynthetically active radiation (PAR) received from the sides is more important than that received from directly above. In 33 of the plants leaf orientation was found to be significantly clustered towards one azimuth. The mean azimuth and the mean angle of declination were different for each plant. Leaves were steeply declined, and oriented towards the largest canopy gap at each site. Steep leaf angles reduced interception of direct and diffuse PAR when compared to interception by plant with a hypothetical horizontal leaf arrangement. When compared to a hypothetical arrangement with steep leaf declination and a uniform azimuth distribution, the observed leaf arrangement increased the efficiency of interception of diffuse PAR, but had a variable effect on the interception of direct PAR. Results indicate that the developing leaves of juvenile P. crassifolius orient towards the strongest sources of diffuse light, regardless of their value as a source of direct light. By maximising diffuse light interception while reducing direct light interception, leaf orientation may be a partial determinant of the types of habitats exploited by this species. This study emphasises the importance of considering diffuse light interception for plants growing in partially shaded environments.  相似文献

Inactivation of a host-specific toxin, RS-toxin, induced by Rhizoctonia solani, the cause of rice sheath blight disease was investigated. A putative -glucosidase identified based on enzyme assay and Western blot analysis was purified from coconut (Cocos nucifera; the only known non-host of R. solani) leaves and tested for its efficacy in degrading RS-toxin. SDS–PAGE analysis showed the appearance of a 97 kDa protein, which appeared in proteins extracted from coconut leaf bits during 48 and 96 h after RS-toxin-treatment and the protein eventually disappeared. A comparison of the u.v. spectra read at 150–300 nm revealed conspicuous disturbances in the absorbance at 24 h of incubation of RS-toxin with the coconut leaf protein extracts as compared to that at 12 h, indicating the possible degradation of RS-toxin by coconut leaf -glucosidase during incubation. Incubation of rice leaf sheath bits with coconut leaf protein extracts significantly reduced electrolyte leakage due to RS-toxin 30 min after the toxin treatment. Simultaneously, there was a significant reduction in sheath blight symptoms when the incubation of rice leaf sheaths with the coconut leaf protein extracts was extended up to 96 or 120 h. This appears to be the first report of purification and characterization of a putative plant -glucosidase.  相似文献

Nitrogen fertilization strategies were widely adopted to enhance grain production and improve nitrogen utilization in rice all over the world. For fertilization timing strategy, ear fertilization was usually employed in recent years. For fertilization amount strategy, nitrogen fertilization would continually increase to meet the demands of increasing people for food. However, under heavy ear fertilization as well as great nitrogen amount (NA), physiological N-use efficiency (PE, defined as grain production per unit nitrogen uptake by plants) decreased. Under three NA and two ratios of fertilization given during ear development period to total NA (ear fertilization distribution ratio, EFDR), net photosynthetic rate (Pn), Pn to nitrogen content per unit area (photosynthetic N-use efficiency, Pn/N), nitrogen accumulation in plant tissues and PE of three rice (Oryza sativaL.) genotypes, Jinyou 253, Liangyoupeijiu and Baguixiang were screened in the first and second seasons in 2002 so as to understand the fluctuation patterns of Pn/N and nitrogen distribution in leaf blades under great NA & EFDR and relationship with PE in rice. Results showed that under greater NA & EFDR, Pn in flag leaves at heading and plant nitrogen accumulation at maturity always increased and PE & Pn/N always decreased in spite of increased grain production. Rice distributed more nitrogen in leaf blade under greater NA and EFDR. PE indicated significantly (P<0.05) positive relationship with Pn/N and negative relationship with nitrogen distribution ratio in leaf blades at heading and maturity, and no association with Pn in two growing seasons. Results suggested that low PE in rice under great NA and heavy ear fertilization is associated to more nitrogen distribution in leaf blades and decreases in photosynthetic efficiency.  相似文献

Prereproductive adults of the grasshopper, Melanoplus sanguinipes (F.) (Orthoptera, Acrididae), demonstrated orientation and movement towards both visual and olfactory stimulus sources in a still-air chamber. Visual stimuli (wheat and lima bean foliage, vertical black or yellow-green stripes, and a yellow-green broad leaf pattern) were approached more frequently than the control white background surface. Olfactory stimuli (chopped wheat foliage and a four-component, synthetic, grass odor blend of volatiles) elicited an even greater positive response than the visual stimuli. Changing the proportions of the four volatiles in the blend significantly reduced positive orientation responses to the stimulus source. Visual cues of wheat foliage and olfactory cues of either chopped wheat odor or the grass odor blend gave greater responses when combined than when presented separately.In flowing air or wind, nearly all insects demonstrated a rapid positive response to odors of chopped wheat and the grass odor blend, significantly greater than the response to the same stimuli in still air. However, positive responses to visual cues were not significantly greater in wind than in still air. When combined with the olfactory stimuli in flowing air, visual cues did not increase the incidence of response. Grasshoppers responding to grass odors in wind moved more rapidly and directly toward the source, and stopped less often and for shorter durations than insects responding to odor in still air or to visual cues.We conclude from these studies that M. sanguinipes adults show orientation behavior to both visual and olfactory stimuli from food plant sources, although leaf odors elicit a stronger positive response particularly when carried by wind.  相似文献

Isoprene synthase (ISPS) catalyzes the formation of isoprene, an important volatile terpenoid with strong effects on global atmospheric chemistry and protective physiological functions in plant leaves. Many terpene synthase genes including isoprene synthase, a member of the TPS-b cluster of this numerous gene family, were already functionally analysed but much less is known about regulation of their promoters. To study regulation of the PcISPS gene in detail we developed transgenic Grey poplar (Populus × canescens) and Arabidopsis thaliana plants in which the PcISPS promoter is fused to enhanced green fluorescent protein (E-GFP) and β-glucuronidase (GUS) reporter genes. We analysed these reporters during plant development, for organ specificity and in plants subjected to different light and temperature regimes. We observed low promoter activity in non-isoprene emitting tissue like roots where ISPS gene is transcribed but no active enzyme is detectable. In leaves we demonstrate that light and temperature directly modulate ISPS promoter activity. Moreover, with confocal laser scanning microscopy we show a cell specific gradient of ISPS promoter activity within the leaf parenchyma depending on light direction. Our results indicate that ISPS promoter activity, which correlates with basal isoprene emission capacity, is not uniformly distributed within leaf tissue and that it can adapt rapidly towards internal as well as external environmental stimuli.  相似文献

Summary The effects on plant growth and stomatal physiology of alterations in light quantity and quality during development were investigated in the C3 monocot, Commelina communis. Reduction in light intensity resulted in decreased branching and stem elongation, with effects more severe under neutral shade (R:FR1.0) than under leaf shade (R:FR0.4) conditions. Shade treatments had no effect on the leaf area or stomatal density of newly expanded leaves. Gas exchange measurements on leaves that had expanded under the different treatments indicated that a reduction in light intensity decreased the magnitude and slowed the kinetics of stomatal responses to pulses of blue light, particularly in plants from the neutral shade treatment. These results indicate that the specific stomatal response to blue light is plastic, and is modulated by the light environment prevailing during leaf development.  相似文献

A thermotolerant fungal strainAspergillus terreus produced high activities of cellulolytic enzymes when grown in shake flasks for 8 days at 40°C or 14 days at 28°C in medium containing 2.5% (w/v) cellulose powder and 1% (w/v) wheat bran. There was little difference between the final activities of endo-(1,4)--glucanase (ca. 14.4 U/ml); filter paper activity (ca. 1.3 U/ml) and -glucosidase (ca. 10 U/ml). Endoglucanase had maximum activity at 60°C and pH 3.8; the other two enzymes were optimal at 60°C and pH 4.8. The maximum hydrolysis of different cellulosic substrates (about 50%) was obtained within 48 h when 1.1 U/ml of filter paper cellulase activity were employed to saccharify 100 mg alkali-treated cotton, filter paper, bagasse, and rice straw at 50°C and pH 4.8. The major end-product, glucose, was produced from all substrates, with traces of cellobiose and other larger oligosaccharides being present in rice straw hydrolysates.  相似文献

A synthetic brassinosteroid (BR), 2,3,22ß,23ß-tetrahydroxy-24ß-methyl-B-homo7-oxa-5-cholestan-6-one, an isomer of the growth promoter brassinolide,when applied to seedlings of dwarf rice Oryza sativa var. Tan-ginbozuand Waito-C, induced a significant bending of the second leaflamina at 100 ng/plant and higher dosages. Promotion of thesecond leaf sheath elongation, the characteristic response ofdwarf rice varieties to gibberellins, was significantly butmodestly enhanced by BR at a dosage of 10,000 ng/plant, fiveorders of magnitude higher than the minimal dosage responseto GA3. Gibberellin A3 had no significant effect on the bendingof the second leaf lamina, nor did any synergism exist betweenBR and GA3 in leaf lamina bending or leaf sheath elongation.Neither ethylene nor (2-chloroethyl)phosphonic acid (ethephon)caused the bending of the second leaf lamina, and neither synergizedthe BR effect. However, IAA and -naphthaleneacetic acid causedsignificant bending at 5,000 ng/plant, and both auxins significantlysynergized the effect of BR on the bending, IAA being effectiveat 500 ng/ plant in this regard. The antiauxins, 2,3,5-triiodobenzoicacid (TIBA) and -(p-chlorophenoxy)isobutyric acid (PCIB) completelynullified both the BR-induced bending and the BR$IAA-synergizedbending. Triacontanol, a 30-carbon primary alcohol, applied in nutrient culture solutions to rice (Oryza sativa L.) seedlings at 2.3×10-8 M (10 g/l), caused an increase in dry weight and leaf area of the whole plants. The response could be observed as early as 3 h of treatment. It was observed at relatively high and low light intensities as well as in the dark where control plants lost but triacontanol-treated plants gained in dry weight. The dry weight gain in the dark was, however, eliminated by removing CO2 from the atmosphere. Triacontanol-treated plants also increased their content of Kjeldahl-N and contained 30% more total N per plant than controls after 6 h in the dark.  相似文献

To analyse the control of rice phytochrome A (phyA) overexpression (wild type or variously mutated) on gene regulation, transgenic tobacco lines overexpressing various rice phyA constructs were crossed with transgenic tobacco lines containing mustard Lhcb1 or Chs1 promoters fused to the uidA reporter gene (-glucuronidase). It was demonstrated that the temporal pattern of competence to respond to phytochrome was not altered by rice phyA overexpression. Also, overexpression of rice phyA did not change the spatial pattern of gene expression. The responsiveness to red and far-red light, on the other hand, depended on the type of overexpressed rice phyA in a structure-function relation: the serine-to-alanine mutant mediated an enhanced response both under continuous red and far-red light, whereas the N-terminal deletion mutant showed a dominant negative effect under continuous far-red light and even after red light pulses. However, the effectiveness of rice phyA overexpression depended on the promoter construct and the developmental stage of the seedlings. The Lhcb1 promoter also conferred -glucuronidase activity in etiolated seedlings. Introduction

The interactions between plants and insect herbivores are complex and multifaceted. Rice and its specialist insect pest the brown planthopper (BPH), Nilaparvata lugens Stål (Hemiptera: Delphacidae) constitute an ideal system for studying plant–insect interactions.

Objectives

Combined metabolomics analyses of rice plant and BPH were conducted to understand the mechanism of host rice plant defense and BPH insect response.

Methods

Metabolite dynamics in rice leaf sheath and BPH honeydew was investigated using the gas chromatography–mass spectrometry (GC–MS) method. The GC–MS data were analyzed by principal component analysis and partial least squares-discriminant analysis.

Results

Twenty-six metabolites were detected in the leaf sheath extracts. Rice leaf sheath metabolomics analysis results show that BPH feeding induces distinct changes in the metabolite profiles of YHY15 and TN1 plants. These results suggest that BPH infestation enhance fatty acid oxidation, the glyoxylate cycle, gluconeogenesis and the GABA shunt in TN1 plants, and glycolysis and the shikimate pathway in YHY15. We propose that the BPH15 gene mediates a resistance reaction that increases the synthesis of secondary metabolites through the shikimate pathway. Thirty-three metabolites were identified in BPH honeydew. Honeydew metabolomics analysis results show that when BPH insects were fed on resistant YHY15 plants, most of the amino acids in honeydew were significantly decreased compared to those of BPH fed on TN1 plants. Based on metabolomics results, we propose that BPH feeding on resistant YHY15 plants would enhance amino acid absorption. At the same time, urea was significantly increased in BPH fed on YHY15.

Conclusion

Metabolomics study is valuable in understanding the complex and multifaceted interaction between plants and insect herbivores and provide essential clue for development of novel control BPH strategies.

The object of this study was to test the alternative hypotheses of magnetoreception by photopigments and magnetoreception based on magnetite. Migratory European Robins, Erithacus rubecula, were tested under light of different wavelengths; after these tests, they were subjected to a brief, strong magnetic pulse designed to alter the magnetization of single domain magnetite. In control tests under white light, the birds preferred the normal, seasonally appropriate migratory direction. Under 571 nm green light, they continued to be well oriented in the migratory direction, whereas under 633 nm red light, their behaviour was not different from random. The magnetic pulse had a significant effect on migratory orientation, but the response varied between individuals: some showed a persistent directional shift, while others exhibited a change in scatter; one bird was seemingly unaffected.These findings indicate a light-dependent process and, at the same time, suggest an involvement of magnetizable material in migratory orientation. They are in agreement with the model of a light-dependent compass and a magnetite-based map, even if some questions concerning the effect of the pulse remain open.  相似文献

Overexpression of phytochrome A results in an increased inhibition of hypocotyl elongation under red and far-red light. We used this approach to assay for the function of N-terminal mutations of rice (Oryza sativa L.) phytochrome A. Transgenic tobacco seedlings that express the wild-type rice phytochrome A (RW), a rice phytochrome A lacking the first 80 amino acids (NTD) or a rice phytochrome A with a conversion of the first 10 serines into alanine residues (S/A) were compared with untransformed wild-type tobacco (Nicotiana tabacum L. cv. Xanthi) seedlings. Experiments under different fluence rates showed that RW and, even more strongly, S/A increased the response under both red and far-red light, whereas NTD decreased the response under far-red light but hardly altered the response under red light. These results indicate that NTD not only lacks residues essential for an increased response under red light but also distorts the wild-type response under far-red light. Wild-type rice phytochrome A and, even more so, S/A mediate an enhanced phytochrome A as well as phytochrome B function, whereas NTD interferes with the function of endogenous tobacco phytochrome A as well as that of rice phytochrome A when co-expressed in a single host. Experiments with seedlings of different ages and various times of irradiation under far-red light demonstrated that the effect of NTD is dependent on the stage of development. Background and Aims

At present most process-based models and the majority of three-dimensional models include simplifications of plant architecture that can compromise the accuracy of light interception simulations and, accordingly, canopy photosynthesis. The aim of this paper is to analyse canopy heterogeneity of an explicitly described tomato canopy in relation to temporal dynamics of horizontal and vertical light distribution and photosynthesis under direct- and diffuse-light conditions.

Methods

Detailed measurements of canopy architecture, light interception and leaf photosynthesis were carried out on a tomato crop. These data were used for the development and calibration of a functional–structural tomato model. The model consisted of an architectural static virtual plant coupled with a nested radiosity model for light calculations and a leaf photosynthesis module. Different scenarios of horizontal and vertical distribution of light interception, incident light and photosynthesis were investigated under diffuse and direct light conditions.

Key Results

Simulated light interception showed a good correspondence to the measured values. Explicitly described leaf angles resulted in higher light interception in the middle of the plant canopy compared with fixed and ellipsoidal leaf-angle distribution models, although the total light interception remained the same. The fraction of light intercepted at a north–south orientation of rows differed from east–west orientation by 10 % on winter and 23 % on summer days. The horizontal distribution of photosynthesis differed significantly between the top, middle and lower canopy layer. Taking into account the vertical variation of leaf photosynthetic parameters in the canopy, led to approx. 8 % increase on simulated canopy photosynthesis.

Conclusions

Leaf angles of heterogeneous canopies should be explicitly described as they have a big impact both on light distribution and photosynthesis. Especially, the vertical variation of photosynthesis in canopy is such that the experimental approach of photosynthesis measurements for model parameterization should be revised.  相似文献

1.  Wildtype Oregon-R Drosophila melanogaster were trained in the ambient magnetic field to a horizontal gradient of 365 nm light emanating from one of the 4 cardinal compass directions and were subsequently tested in a visually-symmetrical, radial 8-arm maze in which the magnetic field alignment could be varied. When tested under 365 nm light, flies exhibited consistent magnetic compass orientation in the direction from which light had emanated in training.
2.  When the data were analyzed by sex, males exhibited a strong and consistent magnetic compass response while females were randomly oriented with respect to the magnetic field.
3.  When tested under 500 nm light of the same quantal flux, females were again randomly oriented with respect to the magnetic field, while males exhibited a 90° clockwise shift in magnetic compass orientation relative to the trained direction.
4.  This wavelength-dependent shift in the direction of magnetic compass orientation suggests that Drosophila may utilize a light-dependent magnetic compass similar to that demonstrated previously in an amphibian. However, the data do not exclude the alternative hypothesis that a change in the wavelength of light has a non-specific effect on the flies' behavior, i.e., causing the flies to exhibit a different form of magnetic orientation behavior.

Summary The carbon supply of peppermint plants was manipulated by growing clonal propagules under three carbon dioxide regimes (350, 500 and 650 l l-1). Feeding by fourth instar caterpillars of Spodoptera eridania increased with elevated CO2 hostplant regime, as well as with low leaf nitrogen content and by a high proportion of leaf volatile terpenoids. Leaf weight increased significantly with the increased carbon supply, but the amount of nitrogen per leaf did not change. The amount of volatile leaf mono-and sesquiterpenes increased proportionately with total leaf dry weight and hence was not influenced by CO2 supply. These results are consistent with ecological hypotheses which assume that allocation to defense is closely regulated and not sensitive to carbon supply per se.  相似文献

Preference studies of the white-backed planthopper, Sogatella furcifera (Horvath) (Delphacidae: Homoptera) (WBPH) for different growth stages of rice, Oryza sativa L., indicated that WBPH selected its preferred variety for feeding by alighting on the plant and feeding briefly, tasting the plant. Differential preference for shelter, food or oviposition by WBPH were observed for different rice varieties and for different stages of a variety. With the advancement of plant age, rice varieties became more unsuitable for feeding and a process progressing more rapidly in resistant varieties. Most of the resistant varieties viz. Pundia, Landi Sarakanti and Ptb-33, unsuitable for shelter and feeding were usually not preferred for oviposition. However, the tolerant variety Mahia bankoi, though suitable for shelter and feeding was also not preferred for oviposition. Therefore, when screening varieties by only measuring feeding preference sources having tolerance mechanisms of resistance may be overlooked.  相似文献

Pure phloem sap was collected from insects feeding on rice (Oryza sativa L.) leaves by a laser technique similar to the aphid stylet technique. Rapid circulation of nitrogen in the sieve tubes was demonstrated directly using 15N as a tracer. Application to the roots of the metabolic inhibitors of amino acids, aminooxyacetate and methioninesulfoximine, changed the amino acid composition in the sieve tubes. Feeding methionine to leaf tips resulted in its bulk transfer into the sieve tubes. In vitro experiments confirmed the existence of protein kinases in the pure rice phloem sap. The phosphorylation status of the sieve tube sap proteins was affected by the light regime. The possibility that changes in chemical composition or protein modification such as phosphorylation in the sieve tubes might affect plant growth are discussed.Analysis of pure phloem sap collected from rice plants by insect laser technique has shown dynamic changes in the chemical composition and the quality of proteins in the sap.  相似文献
