Abstract The ependymal cells of both dorsal and ventral walls lining the lumen of the neurohypophysis of the hagfish, Eptatretus burgeri, absorb peroxidase injected into the third ventricle. Peroxidase diffuses rapidly into the connective tissue separating the neurohypophysis from the adenohypophysis, and also into the connective tissue located between the cell nests of the adenohypophysis.  相似文献

This review briefly summarizes the latest findings on reproductiveendocrinology of Atlantic hagfish (Myxine glutinosa) and implicationsfor fisheries management. In response to a major decline orcollapse of the fisheries (groundfish and anadromous species)industry in the Northeast, species that were once consideredalternative or underutilized have and are being identified thatmay be suitable for commercial harvest, one such example isthe hagfish. Hagfish in recent years have been sought afteras valuable fish not only for their flesh, but also their skin.Currently, there are no regulations governing the harvestingof hagfish along the East Coast. There has been little to noinformation of the life history of hagfish including growthrate, age determination, reproductive biology, life span, andlarval size at hatching. Thus, the level at which a sustainablefisheries for this species can be maintained is unknown. Insome parts of the world, hagfish stocks are being depleted dueto overfishing. In order for fisheries management to manageits hagfish stocks and develop a sustainable commercial hagfishfishery, critical information is needed to assist in determiningthe optimal use of this valuable resource. Key elements of the reproductive system have not been elucidatedin hagfish. However, there is new evidence from recent reproductivestudies that Atlantic hagfish may have a seasonal reproductivecycle. These data include seasonal changes in gonadotropin-releasinghormone (GnRH), gonadal steroids, estradiol and progesterone,corresponding to gonadal reproductive stages along with theputative identity of a functional corpus luteum. This newlyacquired data may provide important information to fisheriesmanagers of the East Coast.  相似文献

The pituitary system of the hagfish remains an enigma. The present study has aimed to detect possible adenohypophysial hormones in the pituitary gland of the brown hagfish, Paramyxine atami, by means of immunohistochemistry in combination with lectin histochemistry. Rabbit antisera raised against ovine luteinizing hormone (LH)β, proopiomelanocortin (POMC)-related peptides, and the growth hormone/prolactin family of tetrapod and fish species were used, and 25 kinds of lectins were tested. Three different types of adenohypophysial cells were revealed in the pituitary of brown hagfish. The first was stained with both anti-ovine LHβ and several D-mannose-binding lectins, such as Lens culinaris agglutinin and Pisum sativum agglutinin. This cell type predominated in the adenohypophysis in adults with developing gonads and thus appeared to be involved in the regulation of gonadal functions. The second was negative for anti-ovine LHβ but was stained with several N-acetylglucosamine-binding lectins, such as wheat germ agglutinin and Lycopersicon esculentum lectin. This cell type exhibited a weak positive reaction with anti-lamprey adrenocorticotropin (ACTH) and thus appeared to be related to POMC-like cells. The second cell type was found in the adenohypophysis regardless of the developmental state of the gonads. The third cell type was negative for both antisera and lectins. Since this cell type was numerous in juveniles and adults without developing gonads, most cells of this type were probably undifferentiated. These findings suggest that GTH and ACTH are major adenohypophysial hormones in the hagfish. Endocrine cells of so-called basal-granulated-open type in the intestinal epithelium of a cyclostome, the Atlantic hagfish (Myxine glutinosa), are characterized ultrastructurally and fluorescence microscopically. These cells regularly extend from the basal lamina to the gut lumen, ending in an apical process with microvilli and a filamentous surface coat. Fasting results in an accumulation of secretion granules in all cytoplasmic portions, except for the terminal web area. A similarity is recorded between the distribution of secretion granules and the finely granular fluorescamine-induced fluorescence, suggesting that the fluorescence is associated to some component(s) of the secretory granules. Granule release may take place at the base after an adequate stimulus (presence of food?) at the luminal portion of the cells. The formaldehyde condensation technique shows that insulin-containing hagfish islet parenchymal cells, but not intestinal endocrine cells, store dopamine after intestinal supply of the amine precursor. Acidification of formaldehyde vapour-fixed intestinal epithelium induces fluorescence in the granules of zymogen cells but not of endocrine cells, indicating a low concentration of tryptophyl-peptide(s) in the secretory granules of hagfish intestinal endocrine cells.  相似文献

The Pacific hagfish, Eptatretus stoutii, is capable of a varietyof immunologic responses including allograft rejection and serumantibody production to soluble and cellular antigens. Interesthas revived in the morphology of hagfish lymphoid tissues. Thesearch for a thymus in young specimens has resulted in the discoveryof a phagocytic and antigen-receptive cell population associatedwith the pharyngeal velar muscles. We suggest that a protothymusor precursor of the thymus of higher vertebrates may be containedwithin this velar muscle complex.  相似文献

Hagfish eyes are markedly basic compared to the eyes of other vertebrates, lacking a pigmented epithelium, a lens and a retinal architecture built of three cell layers: the photoreceptors, interneurons and ganglion cells. Concomitant with hagfish belonging to the earliest-branching vertebrate group (the jawless Agnathans), this lack of derived characters has prompted competing interpretations that hagfish eyes represent either a transitional form in the early evolution of vertebrate vision, or a regression from a previously elaborate organ. Here, we show the hagfish retina is not extensively degenerating during its ontogeny, but instead grows throughout life via a recognizable PAX6+ ciliary marginal zone. The retina has a distinct layer of photoreceptor cells that appear to homogeneously express a single opsin of the RH1 rod opsin class. The epithelium that encompasses these photoreceptors is striking because it lacks the melanin pigment that is universally associated with animal vision; notwithstanding, we suggest this epithelium is a homologue of gnathosome retinal pigment epithelium (RPE) based on its robust expression of RPE65 and its engulfment of photoreceptor outer segments. We infer that the hagfish retina is not entirely rudimentary in its wiring, despite lacking a morphologically distinct layer of interneurons: multiple populations of cells exist in the hagfish inner retina and subsets of these express markers of vertebrate retinal interneurons. Overall, these data clarify Agnathan retinal homologies, reveal characters that now appear to be ubiquitous across the eyes of vertebrates, and refine interpretations of early vertebrate visual system evolution.  相似文献

The localization of a lens forming potency in the iris epithelium was studied by autoradiographic analysis of the distribution of 3H-thymidine labelled cells to be participated in lens regeneration in newts. DNA synthesis started from the dorsal portion of the iris epithelium around 4 days after lentectomy. 5 days after lentectomy, a large number of labelled cells were mostly found in the dorsal sector, showing strong contrast to the ventral and lateral sectors of iris, which contained a few labelled cells. The labelled index (the number of labelled cells/the number of cells in the definite pigmented area of the iris epithelium) of the dorsal sector attained the highest value, 29.7 ± 2.35, on day 7 after lentectomy, and dropped temporarily. This was followed by the second peak on day 12. The dorso-ventral ratio of the labelled index reached to the highest value, 6.87 ± 0.67, on day 5. This ratio decreased rapidly after the completion of a lens rudiment, and it became about 1. In “chase” experiments by diluting the radio-isotope with excess cold thymidine, it was obviously shown that most of the cells labelled with the radio-isotope and distributed in the dorsal marginal iris 5 days after lentectomy participated in the formation of a lens regenerate during the period of chasing. From these results, the following conclusion was drawn. The iris epithelium consists of at least 2 different cell populations; one is capable of transformation into lens cells and is distributed mostly in the dorsal portion of the iris epithelium, while the other has no potency for transformation and is able to grow to compensate a loss of the dorsal marginal cells which transformed into lens cells during the process of lens regeneration.  相似文献

Summary Paraffin sections of brain and pituitary of the hagfish Eptatretus burgeri were immunostained with an antiserum to FMRF-amide. Immunoreactivity was visible in a large number of neurons in the posterior part of the ventromedial hypothalamus and in long neuronal processes extending cranially from the hypothalamus to the olfactory system and caudally to the medulla oblongata. FMRF-amide-like immunoreactivity was also found in cells of the adenohypophysis. These observations suggest that the hagfish possesses a brain FMRF-amide-like transmitter system and pituitary cells containing FMRF-amide-like material.Antisera to ACTH, -MSH and pancreatic polypeptide gave no immunoreaction in hagfish brain or pituitary. Summary The intramembrane organization of the occluding junctions in the gill epithelium of the Atlantic hagfish, Myxine glutinosa, was studied by means of freeze-fracture electron microscopy. Mitochondria-rich cells, characterized by assemblies of rod-shaped particles in the luminal plasma membrane and by an extensive intracellular amplification of the basolateral plasma membrane, are singly distributed between the pavement cells in the gill epithelium of this marine and stenohaline cyclostome. The occluding junctions between mitochondria-rich cells and pavement cells do not differ from those between adjacent pavement cells, concerning the number of superimposed strands (median 6, range 4–9) and their geometrical organization. These observations suggest that, in contrast to marine teleosts, the paracellular pathway plays a minor role in transepithelial ion movements in the hagfish gill epithelium. The findings are in agreement with the absence of hypoosmoregulatory mechanisms in hagfish, as have been evolved in various marine vertebrates. In addition, small communicating junctions are demonstrated between pavement cells; they possibly serve for a coordinated synthesis and secretion of mucus by the pavement cells.Dedicated to Prof. H. Summary Agranular cells are found throughout the adenohypophysis of Myxine glutinosa. They are located at the margin of single hypophysial follicles in the form of a spongy epithelial net. Their relationship to the secretory cells of the adenohypophysis and their morphology are described in adult animals. The distribution and ultrastructure of the agranular cells in the hagfish are in good agreement with the pattern of comparable cells in other vertebrates.This study was supported by the Austrian Fonds zur Förderung der wissenschaftlichen Forschung (grant no. 3204 and 3494). Abstract Intraperitoneal injection of mammalian thyrotropin-releasing hormone (TRH) caused hypertrophy of possible thyrotropic cells in the pituitary gland of the teleost, Chasmichthys dolichognathus. The epithelial cells of the thyroid follicles also appeared hypertrophic after the TRH injection. On the other hand, in the hagfish, Eptatretus burgeri, TRH injection had no effect either on the cytological appearance of the adenohypophysis or on the thyroidal activity in terms of serum thyroxine concentration and epithelial cell height.  相似文献

It has been described the cytology of the following parts of the respiratory system of some South American primates:Callithrix jacchus andCallithrix argentata melanura. The nasal cavities are divided into three parts: a vestibule, covered with a stratified nonkeratinized squamous epithelium; the respiratory portion, consisting of a pseudostratified columnar ciliated epithelium with goblet cells and the olfactory portion which is also covered with a high respiratory epithelium without goblet cells. The trachea is lined with a mucous membrane, whose epithelium is pseudostratified columnar ciliated with scarce goblet cells in the proximal portion unlike to the distal one. In the dorsal portion of the trachea, at the level of the gap between the two ends of incomplete cartilaginous rings, the epithelial lining is of transitional type. The incomplete hyaline cartilaginous rings present centers of calcification. The right and left lungs consist of two and three lobes respectively characteristic for these species, but they are not divided into lobules by connective tissue as in other ones. The bronchi, bronchioles and the respiratory portion, respiratory bronchioles, alveolar ducts, alveolar sacs, and alveoli present the typical respiratory structure with exception of their cartilaginous configuration; the cartilage continues as far as the respiratory bronchioles and alveolar ducts. These last structures are formed by a thin squamous epithelium, in which we observed two types of alveolar lining cells. At the gross anatomical level, hagfish gills show unusual features not seen in any other fish gills. Our study was undertaken to determine if peculiarities also characterize the microscopic anatomy and ultrastructure of hagfish gills. To the contrary, branchial respiratory lamellae of Pacific hagfish were found to resemble the lamellae of lampreys, elasmobranchs, and teleosts, often down to the finest subcellular details. As in other fish, hagfish lamellae are lined by epithelium containing pavement cells with organelles indicative of a secretory function, basal cells showing undifferentiated cell features, and branchial ionocytes. The ionocytes are identical to chloride cells of teleosts in cytostructure, distribution, and abundance. There are pillar and marginal capillaries in hagfish gill lamellae. Pillar cells contain bundles of 5-nm microfilaments, and they associate with collagen columns as in other fish. Hagfish pillar cells do exhibit odd features, however: They cluster (groups of up to nine were seen), and their extracellular collagen columns are rarer than in other fish gills (averaging only two columns per three pillar cells). Other special features of hagfish gills are the following: lipid droplets and smooth endoplasmic reticulum are well developed in all cell types; pavement cells secrete a lipomucous product (stains with periodic acid-Schiff, Alcian blue, and Sudan black B); and goblet cells are absent. The presence of "chloride cells" in hagfish is puzzling, as hagfish body fluids are iso-osmotic to seawater and there is no need to osmoregulate for sodium chloride; the ionocytes contain carbonic anhydrase, suggesting a function in acid/base regulation.  相似文献

We report the discovery of a new agnathan specimen from the Lower Cambrian Chengjiang Lagerstätte of China and thereby provide new evidence on the myomeres (V-shaped), the branchial apparatus (gill filaments and arches), the dorsal fin and the gonads (24-26) of the earliest vertebrates. The new specimen and the co-occurring Myllokunmingia fengjiaoa and Haikouichthys ercaicunensis represent a single species, which is a primitive member of the crown group craniates (vertebrates) and post-dates the origin of the myxinoids (hagfish). The origin of the vertebrate clade is at least as old as Early Cambrian.  相似文献

Summary Neurosecretory axons in the neurohypophysis of the hagfish, Eptatretus burgeri, were statistically classified into six types according to the size of secretory granules. These types are comparable with those in higher vertebrates. The concentration of each axon type is different in three regions: anterior dorsal wall, posterior dorsal wall, and ventral wall. The regional differences of the hagfish neurohypophysis are discussed in relation to the regional differentiation of the tetrapod neurohypophysis into the median eminence and the pars nervosa.We wish to express our appreciation to Professor Hideshi Kobayashi for the guidance of this study. Dorsal-ventral (DV) specification in the early optic vesicle plays a crucial role in the proper development of the eye. To address the questions of how DV specification is determined and how it affects fate determination of the optic vesicle, isolated optic vesicles were cultured either in vitro or in ovo. The dorsal and ventral halves of the optic vesicle were fated to develop into retinal pigment epithelium (RPE) and neural retina, respectively, when they were separated from each other and cultured. In optic vesicles treated with collagenase to remove the surrounding tissues, the neuroepithelium gave rise to cRax expression but not Mitf, suggesting that surrounding tissues are necessary for RPE specification. This was also confirmed in in ovo explant cultures. Combination cultures of collagenase-treated optic vesicles with either the dorsal or ventral part of the head indicated that head-derived factors have an important role in the fate determination of the optic vesicle: in the optic vesicles co-cultured with the dorsal part of the head Mitf expression was induced in the neuroepithelium, while the ventral head portion did not have this effect. The dorsal head also suppressed Pax2 expression in the optic vesicle. These observations indicate that factors from the dorsal head portion have important roles in the establishment of DV polarity within the optic vesicle, which in turn induces the patterning and differentiation of the neural retina and pigment epithelium.  相似文献

In order to reveal possible stimulation of growth of the germinal epithelium of the rat's injured testicle the animals were injected follicle-stimulating and luteinizing hormones and their mixture as well as homogenate of fertilized egg-cells during two weeks. Changes of neurosecretion in large-cell nuclei of the anterior hypothalamus, cellular set in the adenohypophysis and the condition of the germinal epithelium in the gonad under study were investigated. Identical phenomena of reduction (having quantitative distinctions) took place due to effects of exogeneous honadotropins in neurosecretory nuclei and the adenohypophysis. In testicular tubules there occurred pronounced proliferation of the germinal epithelium, but spermatogenesis was absent. Egg-cell homogenate failed to cause changes in neurosecretion and adenohypophysis but resulted in completing the developmental cycle of the germinal epithelium with mature spermatozoa. The effect of gonadotropins upon the system hypothalamus-hypophysis is explained by feed-back mechanism, and the absence of spermatogenesis--by the lack of androgens. The rsults of egg-cell homogenate effects should be associated with local stimulating effects of DNA of the dividing egg-cell.  相似文献

We studied by light microscopy the histological development of the olfactory and vomeronasal organ in tadpoles of the Chinese forest frog, Rana chensinensis, from postembryonic periods to the end of metamorphosis. Unlike Bufo americanus, the olfactory epithelium in larval R. chensinensis is not divided into dorsal and ventral branches in the rostral and mid-nasal regions. The olfactory epithelium in the dorsal portion of the buccal cavity in larval R. chensinensis may correspond to the ventral olfactory epithelium of Bufo, which has been argued to provide a chemosensory function in the tadpoles analogous to the role of taste buds in adults. Bowman's glands were present in the olfactory epithelium of R. chensinensis only after the appearance of the forelimbs during metamorphosis. The appearance of Bowman's glands in the olfactory epithelium at this time suggests that the nose first begins to detect odorants in the air, and this is thus also a metamorphic event. The vomeronasal epithelium appeared a little earlier than the vomeronasal gland in R. chensinensis, unlike in toads (bufonids). This study supports Eisthen's hypothesis that the most recent common ancestor to the tetrapods was aquatic and once had a vomeronasal organ, and that this has been lost in various evolutionary lineages.  相似文献

A new species of hagfish (Myxinidae),Eptatretus eos, is described on the basis of one specimen from the Tasman Sea, west of New Zealand. This five-gilled, deep-sea species differs from previously described hagfish in having the unpaired nostril prolonged into a tube-like snout and in being uniformly pink.  相似文献

Using a monoclonal antibody for the alpha-subunit of the Na+/K(+)-ATPase, DASPEI (a vital mitochondria dye), and confocal laser scanning microscopy, the presence of Na+/K(+)-ATPase in mitochondrion-rich cells of the hagfish gill was confirmed. In addition, the level of Na+/K(+)-ATPase expression in the hagfish gill was compared to that of fishes with different osmoregulatory strategies (little skate, Raja erinacea and mummichog, Fundulus heteroclitus). Immunocytochemistry detected a high density of columnar cells expressing Na+/K(+)-ATPase in the afferent filamental epithelium. Positive cells were also found in the lamellar epithelium but at a much lower density. The distribution of DASPEI staining was similar to that of the Na+/K(+)-ATPase antibody, indicating that the enzyme is expressed in mitochondrion-rich cells. Immunoblot analysis confirmed the specificity of the antibody for the 97 kDa alpha-subunit of the enzyme. The immunoreactive band intensity for the Atlantic hagfish was similar to that of the little skate, but less than half that of the full-strength seawater mummichog. These results are discussed in relation to gill function in early craniates.  相似文献
