为了解NP配施对平茬后云南松(Pinus yunnanensis)苗木各器官N、P、K化学计量比的影响,分析云南松苗木不同器官(根、茎、叶、萌条)的ω(N)∶ω(P)、ω(N)∶ω(K)、ω(P)∶ω(K)化学计量比的季节变化特征,探讨各器官间N、P、K化学计量比的相关性及其变异来源。采用N、P二因素三水平的3×3回归设计开展不同施肥试验,并对苗木采样测定,研究NP配施对平茬后云南松根、叶、茎及其萌条N、P、K化学计量特征的影响。结果表明:平茬后云南松苗木不同器官的营养元素分配没有统一的规律,展现出丰富的变异。随着施肥季节的变化,ω(N)∶ω(P)在根、茎和萌条中逐渐下降,在叶中先下降后上升,但总体差异不大。单施N肥、P肥和NP配施均对云南松苗木生长的影响产生一定差异,总体来看NP配施更有利于促进苗木的生长,且以处理5(N1P1)表现为极显著(P<0.01)。云南松苗木各器官N、P、K化学计量比主要受N×P交互作用的影响,其次是N,影响最小的是P。除在根和叶中ω(N)∶ω(P)与ω(N)∶ω(K)之间相关性发生改变之外,其余两两间的正负...  相似文献

Complexes of type A4[VO(tart)]2·nH2O, where A = Rb or Cs and tart =d,l-tartrate(4−) (n = 2) or d,d-tartrate(4−) (n = 2 for Rb and n = 3 for Cs), were prepared from an aqueous mixture of V2O5, AOH and H4tart. These complexes were studied by single-crystal X-ray diffraction methods: Rb4[VO(d,l-tart)]2·2H2O, space group P1 with a = 8.156(1),b = 8.246(1),c = 8.719(1)Å, = 66.09(1)°, β = 65.07(1)°, γ = 82.40(1)°,Z = 2, 1917 observed reflections, and final Rw = 0.035; Cs4[VO(d,l-tart)]2·2H2O, space group P21/c with a = 9.350(1),b = 13.728(2),c = 8.479(1)Å, β = 106.77(1)°,Z = 4, 2235 observed reflections, and final Rw = 0.054; Rb4[VO(d,d-tart)]2·2H2O, space group P4122 with a = 8.072(1),c = 32.006(3)Å,Z = 8, 1014 observed reflections and final Rw = 0.038; Cs4[VO(d,d-tart)]2·3H2O, space group P122 with a = 8.184(1),c = 33.680(5)Å,Z = 8, 1310 observed reflections, and final Rw = 0.063. Bulk magnetic susceptibility data (1.5–300 K) for these compounds and A4[VOl,l-tart)]2·nH2O (A = Rb, Cs) were obtained on polycrystalline samples. These data were analyzed in terms of a Van Vleck exchange coupled S = 1/2 model which was modified to include an interdimer exchange parameters Θ. Analysis of the low-temperature (1.5–20 K) susceptibility data gave 2J = +1.30 cm−1 and Θ = −1.86 K for Rb4[VO(d,l-tart)]2·2H2O, 2J = +1.16 cm−1 and Θ = −1.69 K for Cs4[VO(d,l-tart)]2·2H2O, 2J = +1.90 cm−1 and Θ = −0.82 K for Rb4[VO(d,d-tart)]2·2H2O, 2J = +2.04 cm−1 and Θ = −0.80 K for Rb4[VO(l,l-tart)]2·2H2O, 2J = +1.52 cm−1 and Θ = −0.25 K for Cs4[VO(d,d-tart)]2·3H2O, and 2J = +1.64 cm−1 and Θ = −0.31 K for Cs4[VO(l,l-tart)]2·3H2O. These results suggest the magnitudes of intradimer (ferromagnetic and interdimer (antiferromagnetic) exchange interactions are similar in these complexes, as observed for the analogous Na salts.  相似文献

The magnetic susceptibility of singly reduced, camphor-complexed cytochrome P−450 from Pseudomonas putida has been measured at various temperatures. Computer analysis using a spin Hamiltonian gives values of D between 15K and 25K, which agrees with a value of D = 20K found by low-field variable temperature Mössbauer spectroscopy. Krüppel样因子(Krüppel-like factors,KLFs)是锌指蛋白超家族的一个亚家族,参与细胞内的多种生理、病理过程,该家族成员在红细胞分化发育过程中发挥非常重要的作用,但是家族成员间对红系分化的协同调控作用还鲜有报道。本课题组前期研究发现,KIF家族成员KLF1KLF9在已分化的红系细胞中的表达水平显著高于造血干细胞。为进一步探讨二者在红系分化中是否存在协同作用,本研究在K562细胞中分别过表达/敲低表达KLF1KLF9,检测二者表达的相关性,发现KLF1KLF9的基因表达呈现正相关,且二者共表达可以显著促进K562细胞红系分化,特异地增强β-珠蛋白的表达。通过对KLF1KLF9单独和共同过表达、敲低表达的K562细胞转录组数据的分析发现二者可能通过PI3K-Akt和FoxO通路协同调控红系分化,FOS、TF、IL8是协同调控的候选靶基因。本研究结果为后续深入研究KLF1KLF9协同调控红系分化的分子机制奠定了基础。  相似文献

1. The absorption spectrum of chlorophyll b in vivo at 77°K is presented as the difference spectrum between preparations of spinach and chlorophyll b-free Vischeria stellata chloroplasts.

2. A shoulder on this spectrum around 662 nm is due to a component different from chlorophyll b. This component may well be identical with the chlorophyll a form, chlorophyll a (665).

3. The 77°K chlorophyll b absorption spectra in the nonfractionated photosyn-thetic pigment apparatus and in fractions mainly representing Photosystems 1 or 2 are not significantly different.

4. The aerobic irreversible photobleaching of chlorophyll b was studied in the intact pigment complex as well as in fractions mainly consisting of Photosystem 1 or 2. A two-step photobleaching was observed in all cases. The time-course of this bleaching was not significantly different for chlorophyll b in both fractions.

为探索华北落叶松(Larix principis-rupprechtii)人工林土壤有效性钾的钾素形态分布与含量变化规律,定期取样并测定了幼龄林(5a,10a)、中龄林(20a)、近熟林(30a)及成熟林(40a)的土壤有效钾含量。结果表明,速效钾含量在0–20 cm土层高于20–40 cm与40–60 cm土层;在0–20 cm土层中,年内不同生育期不同林龄间速效钾含量差异显著(P0.05);在0–20cm土层,土壤缓效钾和有效钾含量与其它土层含量差异不显著,年内不同生育期间土壤缓效钾和有效钾含量差异未达到显著性水平。随着林龄的增加,土壤速效钾呈先增加后降低的趋势,土壤有效钾与缓效钾呈下降趋势,成熟林(40a)的土壤有效钾与缓效钾含量明显低于其它林龄。研究区域各林龄华北落叶松人工林土壤速效钾与缓效钾在含量分级标准中达到中等及高等含量水平,有效钾含量丰富。  相似文献

In earlier work, immature oocytes of the irradiated population RÖI4 of Drosophila melanpgaster were found to be radioresistant relative to those of the basic population RÖI and to those of the control population Berlin wild (+K). The resistance of RÖI4 relative to RÖI was previously attributed to a hypothetical “factor” rar-3. In the present paper, evidence is presented to show that rar-3 is a single, recessive genetic factor, located on chromosome 3 at a map position of about 49.8. The action of rar-3 is apparently independent of that of rar-1 and rar-2, the factors already present in RÖI.  相似文献

Two new spin-crossover complexes, [Fe(Medpq)(py)2(NCS)2] · py · 0.5H2O (1) and [Fe(Medpq)(py)2(NCSe)2] · py (2) (Medpq = 2-methyldipyrido[3,2-f:2′,3′-h]-quinoxaline, py = pyridine), have been synthesized. The crystal structures were determined at both room temperature (298 K) and low temperature (110 K). Complexes 1 and 2 crystallize in the orthorhombic space group Pbca and monoclinic space group P21/n, respectively. In both complexes, the distorted [FeN6] octahedron is formed by six nitrogen atoms from Medpq, the trans pyridine molecules and the cis NCX groups. The thermal spin transition is accompanied by the shortening of the mean Fe–N distances by 0.194 Å for 2. The mononuclear [Fe(Medpq)(py)2(NCS)2] and [Fe(Medpq)(py)2(NCSe)2] neutral species interact each other via π-stacking, resulting in a one-dimensional extended structure for both 1 and 2. There exist C–HX (X = S, Se) hydrogen bonds for both complexes. Variable-temperature magnetic susceptibility measurements and Mössbauer spectroscopy reveal the occurrence of a gradual spin transition. The transitions are centered at T1/2 = 120 K for 1 and T1/2 = 180 K for 2, respectively.  相似文献

亚高山森林冬季不同厚度雪被斑块下显著的冻融格局差异可能对凋落物分解过程中钾(K)和钠(Na)的动态具有重要影响, 然而已有研究还不足以清晰地认识这一过程。以川西亚高山森林6种代表性树种凋落物为研究对象, 采用凋落物网袋法, 探讨冬季不同厚度雪被斑块下雪被形成期、覆盖期和融化期凋落物分解过程中K和Na元素释放或富集的特征。整个雪被覆盖时期, 6种凋落物分解过程中Na均表现为富集特征, 且以覆盖期最为明显; 而K表现为释放特征, 以雪被融化期释放率最大。相对于其他雪被斑块, 厚型和中型雪被斑块下凋落物K释放率相对较高; 除康定柳(Salix paraplesia)和高山杜鹃(Rhododendron lapponicum)外, 其他物种凋落物在厚型和中型雪被斑块下Na富集率较高。同时, 统计分析结果表明, 物种和雪被显著影响冬季不同关键时期凋落物K和Na元素动态。除红桦(Betula albosinensis)和方枝柏(Sabina saltuaria)凋落物外, 温度因子与凋落物K和Na动态变化呈显著正相关。这些结果表明气候变暖情景下冬季雪被覆盖的减小将抑制亚高山森林冬季凋落物分解过程中K和Na元素的释放, 但是释放程度受凋落物质量和雪被覆盖时期的显著影响。  相似文献

A current goal of malaria vaccine research is the development of vaccines that will cross-protect against multiple strains of malaria. In the present study, the breadth of cross-reactivity induced by a 30K multivalent DNA vaccine has been evaluated in susceptible A/J mice (H-2a) against infection with the Plasmodium chabaudi adami DK strain and a virulent parasite subspecies, Plasmodium chabaudi chabaudi AS. Immunized A/J mice were significantly protected against infection with both P. c. adami DK (31–40% reduction in cumulative parasitemia) and P. c. chabaudi AS parasites, where a 30–39% reduction in cumulative parasitemia as well as enhanced survival was observed. The 30K vaccine-induced specific IFN-γ production by splenocytes in response to native antigens from both P. c. chabaudi AS and P. c. adami DK. Specific antibodies reacting with surface antigens expressed on P. c. adami DS and P. c. chabaudi AS infected red blood cells, and with opsonizing properties, were detected. These results suggest that multivalent vaccines encoding conserved antigens can feasibly induce immune cross-reactivity that span Plasmodium strains and subspecies and can protect hosts of distinct major histocompatibility complex haplotypes.  相似文献

Resonance Raman spectra are reported for native horseradish peroxidase (HRP) and cytochrome c peroxidase (CCP) at 290, 77 and 9 K, using 406.7 nm excitation, in resonance with the Soret electronic transition. The spectra reveal temperature-dependent equilibria involving changes in coordination or spin state. At 290 K and pH 6.5, CCP contains a mixture of 5- and 6-coordinate high-spin FeIII heme while at 9 K the equilibrium is shifted entirely to the 6-coordinate species. The spectra indicate weak binding of H2O to the heme Pe, consistent with the long distance, 2.4 Å, seen in the crystal structure. At 290 K HRP also contains a mixture of high-spin FeIII hemes with the 5-coordinate form predominant. At low temperature, a small 6-coordinate high-spin component remains but the 5-coordinate high-spin spectrum is replaced by another which is characteristic either of 6-coordinate low-spin or 5-coordinate intermediate spin heme. The latter species is definitely indicated by previous EPR studies at low temperature. This behavior implies that, in contrast to CCP, the distal coordination site of HRP is only partially occupied by H2O at any temperature and that lowering the temperature significantly weakens the Fe-proximal imidazole bond. Consistent with this inference, the 77 K spectrum of reduced HRP shows an appreciable fraction of molecules having an Fe-imidazole stretching frequency of 222 cm−1, a value indicating weakened H-bonding of the proximal imidazole.

A field experiment was conducted in the South of Portugal with young (0-3 years old) non-bearing ‘Lane Late’ orange trees. Five nitrogen (N) rates were used in a randomised block design with three replicates. The 180 g N tree−1 over three years led to the greatest canopy width (176 cm) and volume (2,697 dm3). The greatest rate applied (720 g N tree−1 in the three years) led to the largest flower yield. Nitrogen concentration in the flowers significantly increased with fertilizer N, and also with the flowering period up to the 23rd day, declining thereafter. Flower yield was strongly correlated (r = 0.99, p < 0.001) with flower N concentration. Nutrient composition of flowers and of mature leaves from the spring flush was compared. Significant correlations were found for N (r = 0.47, p <0.01), P (r = -0.49, p <0.01), K (r = 0.44, p <0.05) and Ca (r = 0.87, p <0.001), suggesting that flowers can be used as a tool to diagnose the nutritional status of trees. Canonical analysis (with N treatment as dummy-variables) showed strong relationships between canopy width and N, which were greater at the larger rates of fertilizer application, and strong and inverse relationships between K and Mg, also with the greatest N rates.  相似文献

木质素是木材的重要组成成分,主要起机械支持、水分运输及防御病虫害等作用。4CL酶是控制木质素合成途径中的关键酶之一。利用PCR技术从84K杨幼叶cDNA中克隆得到Pag4CL3/4CL5基因,GenBank 登录号分别为MK183033(Pag4CL3)及MK183034(Pag4CL5)。利用生物信息学软件,对这两个基因的功能和特征进行分析。结果发现84K杨中4CL3/4CL5蛋白与毛果杨中4CL3/4CL5蛋白相似度高达97%;此外,均含有SSGTTGLPKGV和GEICIRG两个保守基序;亚细胞定位预测显示Pag4CL3/4CL5蛋白主要定位在内质网中,推测可能是一种膜蛋白;蛋白的亲水性预测Pag4CL3/4CL5均为亲水性蛋白。通过qRT-PCR分析Pag4CL3/4CL5在不同组织部位中的表达差异,结果发现Pag4CL3在叶及茎中表达量较高,在根和顶芽中表达量较低;Pag4CL5在叶及根中表达量较高,在茎和顶芽中表达量较低,两个同源基因表达具有组织部位的差异性。Pag4CL3和Pag4CL5可能在叶中共同行使功能,Pag4CL5主要在根中行使功能,Pag4CL3主要在茎中行使功能。  相似文献

目的:利用CRISPR/Cas9技术对K562细胞系JAK2基因进行编辑,构建JAK2基因敲除的K562细胞系。方法:使用CRISPR在线设计工具,针对JAK2基因设计sgRNA,构建Cas9-sgRNA共表达质粒。使用第二代慢病毒包装系统包装慢病毒并感染K562细胞,提取细胞基因组DNA,Sanger测序和TA克隆检测基因编辑活性。无限稀释法将编辑阳性的细胞接种于96孔板并扩培得到单克隆细胞株,提取基因组DNA,Sanger测序和TA克隆分析敲除JAK2单克隆细胞的基因型。结果:成功构建靶向敲除JAK2基因的lentiCRISPRv2-sgRNA3-1质粒。优化方案得到低细胞毒性高转染效率的感染K562细胞慢病毒量。CRISPR/Cas9系统成功在JAK2基因sgRNA3-1识别位点发挥基因组编辑活性,获得纯合敲除JAK2基因细胞株K562-JAK2~(-/-)(两个等位分别发生移码突变,预期编码没有功能的JAK2蛋白)。结论:CRIAPR/Cas9系统通过慢病毒感染方式获得JAK2基因纯合敲除的K562细胞株,该细胞模型可用于研究在慢性髓系白血病中JAK2基因的作用,为构建K562敲除其他基因细胞系提供实验依据,为探究造血分化机制的研究奠定实验基础。  相似文献
