With several pairs of rel+ and rel strains of Escherichia coli, the effects of amino acid starvation on the intracellular concentration of K+ and the rate of uptake of 42K+ were investigated. In the early phase of the experiments, the intracellular concentration of K+ was estimated by the conventional method in which the cell volume per A660 value of the culture was assumed to be constant, being not influenced by the variation of growth condition and strain. Apparently, the K+ concentration of rel+ cells was kept almost constant, while that of rel cells increased about 1.5-fold 2 h after the exposure to amino acid starvation. Unexpectedly, however, the above assumption was found not to be valid in the present study. The cell volume per A660 changed only slightly in CP78 (rel+) cells, while it increased markedly in CP79 (rel) cells after the exposure to amino acid starvation. Reestimation of the K+ concentrations based on the estimated respective values of cell volumes per A660 revealed no significant difference between both strains. After all, the above apparent phenomenon was found to be due to the fact that the increase in cell volume of the rel+ cells was arrested upon amino acid starvation whereas that in the rel cells was not. The 42K+ uptake by the rel+ cells was depressed upon amino acid starvation, whereas that by the rel cells increased. Some regulatory mechanism was suggested to operate in both strains to keep their K+ concentrations constant. When intracellular concentration of a metabolite is to be determined, importance of measurement of cell volume under the respective conditions, without assuming the constancy of the cell volume per A660 of the culture, was pointed out.  相似文献

Serratia marcescens SM-6 when starved for a required amino acid stops synthesizing protein and RNA and accumulates two nucleotides which cochromatograph with ppGpp and pppGpp. These features are characteristic of bacterial strains with stringent RNA control (rel +). Two independent mutants were isolated which resemble relaxed (relA) mutants ofEscherichia coli; they continue to synthesize RNA and accumulate neither ppGpp nor pppGpp when deprived of the required amino acid. The extracellular enzyme activities (nuclease, protease, lipase) of the relaxed mutants are about the same as those of the parental stringent strain when studied under standard growth conditions. Exoenzyme-deficient (nuc; prt) and exoenzyme-hyperproducing (nuc su) mutants were isolated from both stringent and relaxed strains ofS. marcences SM-6 and no change of the cellular ability to form ppGpp and pppGpp could be observed. tRNAs are aminoacylated by the aminoacyl-tRNA synthetases. There are at least 20 natural amino acids, but due to the redundancy of the genetic code, 64 codons on the mRNA. Therefore, there exist tRNA isoacceptors that are aminoacylated with the same amino acid, but differ in their sequence and in the anticodon. tRNA identity elements, which are sequence or structure motifs, assure the amino acid specificity. The Seryl-tRNA synthetase is an enzyme that depends on rather few and simple identity elements in tRNASer. The Seryl-tRNA-synthetase interacts with the tRNASer acceptor stem, which makes this part of the tRNA a valuable structural element for investigating motifs of the protein–RNA complex. We solved the high resolution crystal structures of two tRNASer acceptor stem microhelices and investigated their interaction with the Seryl-tRNA-synthetase by superposition experiments. The results presented here show that the amino acid side chains Ser151 and Ser156 of the synthetase are interacting in a very similar way with the RNA backbone of the microhelix and that the involved water molecules have almost identical positions within the tRNA/synthetase interface.  相似文献

Abstract— The properties of RNA fractions from nuclei of brain cells which were capable of stimulating amino acid incorporation into proteins of an homologous ribosomal system were investigated. RNA was routinely prepared from crude nuclear preparations of rat brain by a method which involved treatment with sodium dodecyl sulphate and phenol at 65°. The capacity of this preparation to stimulate incorporation of radioactivity from a mixture of 15 l -[14C]amino acids was greatly enhanced by preliminary incubation of the ribosomal system from brain for 5–20 min. The response was markedly dependent upon the concentrations of ribosomes and of the pH 5 fraction. The optimal level of Mg2+ for basal incorporation of amino acids into protein was 8 mm ; however, incorporation in the presence of nuclear RNA was greater at higher concentrations of Mg2+. The response to nuclear RNA was also enhanced as the K+ concentration was increased from 25 to 100 mm . The stimulatory effect of nuclear RNA on incorporation of l -[12C]eucine was either unaltered or depressed by addition of a mixture of 19 l -[12C]amino acids each at concentrations, of 10?8, 10?2, or 10?1 mm . Under appropriate conditions of incubation, basal rates of incorporation and rates of incorporation stimulated by nuclear RNA were linear for 30 min. The response was proportional to the concentration of nuclear RNA between 34 and 136 μg. RNA prepared from ribosomes of rat brain essentially failed to stimulate incorporation of amino acids over this range of concentrations. Fractionation of nuclear RNA by centrifugation in sucrose density gradients revealed that 75 per cent of the stimulatory activity was in the fraction which sedimented below 12 S and contained about 25 per cent of the total RNA. Most of the remaining activity was in the 18 S region. Less than 5 per cent of the RNA in the lightest fraction (< 12 S) exhibited amino acid-acceptor activity, The stimulatory action of nuclear RNA on incorporation of amino acids was readily destroyed by mild treatment with pancreatic ribonuclease, whereas amino acid-acceptor activity was relatively resistant to this treatment. The results suggest that the brain may contain low molecular weight RNA with properties of messenger RNA.  相似文献

The in vitro movement of ribosomes over messenger RNA was studied in both the presence and the absence of protein synthesis. For this purpose, labeled polysomes were extracted from rel+ and rel? strains of Escherichia coli grown in the presence of radioactive uracil and incubated in a cell-free system containing tRNA, amino acids, soluble enzymes and a source of energy. The gradual conversion of the labeled polysomes into monosomes and ribosomal subunits was followed by subjecting the reaction mixture to sucrose gradient sedimentation after various incubation times and measuring the radioactivity present in the three relevant ribosomal fractions.It was found that when the conditions of incubation allow protein synthesis to occur, polysomes extracted from rel+ and rel? cells are converted mainly into free monosomes, which can be made to dissociate into subunits by high-sodium or low-magnesium ion concentrations. Under conditions in which protein synthesis cannot occur because a mutant aminoacyl-tRNA synthetase has been rendered inactive, polysome conversion still occurs, though to a reduced extent. When the products of such residual run-off are examined, however, a difference is manifest between polysomes extracted from rel+ and from rel? strains: whereas the polysomes from the rel? strain are still converted into free monosomes even in the absence of protein synthesis, the polysomes from the rel+ strain are now converted mainly into subunits. It can be inferred, therefore, that ribosomes from rel? bacteria, but not those from rel+ bacteria, continue movement over messenger RNA in the absence of protein synthesis.Studies of mixed extracts from rel? and rel+ bacteria have shown that the character of the run-off process does not depend on the source of tRNA and soluble enzymes; the proportions of monosomes and subunits among the run-off products formed in the absence of protein synthesis depend only on the source of the polysomes. It is suggested that the mutation of the rel gene alters the functional architecture of ribosomes.  相似文献

In arelA+ strain ofE. coli starved separately for each of four required amino acids, the intracellular concentration of polysomes decreases as a function of time in all cases: very rapidly in the absence of arginine or leucine, slowly in the absence of threonine or histidine. In a starved isogenicrelA strain, the polysome level is either totally stable or else drops slowly. The decrease in the level, when it occurs, does not significantly affect the polysome size distribution. Models for polysome metabolism in amino acid starved cells are discussed.  相似文献

Populations of transfer ribonucleic acid (tRNA) extracted from control and type 2 adenovirus (Ad2)-infected KB cells were compared. No consistent differences in acceptor activity for 11 amino acids were observed. Comparison of methylated albumin-kieselguhr (MAK) elution profiles of arginyl-tRNA from control and infected cells revealed a minor modification in that the proportion of arginyl-tRNA eluting at high salt concentration was somewhat greater in infected cells. No similar differences were observed in MAK elution profiles of aspartyl-, isoleucyl-, leucyl-, phenylalanyl-, seryl-, tyrosyl-, and valyl-tRNA. Hybridization of 4S RNA from infected cells labeled by incorporation of 3H-uridine with Ad2 deoxyribonucleic acid revealed the presence of a complementary species of RNA in this preparation. Hybridization of 3H-arginyl-tRNA and of 3H-aminoacyl-tRNA labeled by charging with 3H-arginine or a 3H-mixture of amino acids, respectively, failed to detect the presence of virus-specific tRNA in Ad2-infected cells.  相似文献

The Schizosaccharomyces pombe temperature-sensitive mutant snm1 maintains reduced steady-state quantities of the spliceosomal small nuclear RNAs (snRNAs) and the RNA subunit of the tRNA processing enzyme RNase P. We report here the isolation of the pac1 + gene as a multi-copy suppressor of snm1. The pac1 + gene was previously identified as a suppressor of the ran1 mutant and by its ability to cause sterility when overexpressed. The pac1 + gene encodes a double-strand-specific ribonuclease that is similar to RNase III, an RNA processing and turnover enzyme in Escherichia coli. To investigate the essential structural features of the Pac1 RNase, we altered the pac1 + gene by deletion and point mutation and tested the mutant constructs for their ability to complement the snm1 and ran1 mutants and to cause sterility. These experiments identified four essential amino acids in the Pac1 sequence: glycine 178, glutamic acid 251, and valines 346 and 347. These amino acids are conserved in all RNase III-like proteins. The glycine and glutamic acid residues were previously identified as essential for E. coli RNase III activity. The valines are conserved in an element found in a family of double-stranded RNA binding proteins. Our results support the hypothesis that the Pac1 RNase is an RNase III homolog and suggest a role for the Pac1 RNase in snRNA metabolism.  相似文献

Summary The gene braB, encoding the Na-coupled carrier for branched-chain amino acids in Pseudomonas aeruginosa PAO, was cloned on cosmid pMMB34. The cosmid clones carrying the braB gene were identified as those that restored growth at low leucine concentration and Na-dependent leucine transport activity to P. aeruginosa PAO3536 defective in the transport of branched-chain amino acids. Determination of the nucleotide sequence of the DNA fragment shows that the braB gene comprises 1311 bp and encodes a hydrophobic protein of 437 amino acids with a calculated Mr of 45279. The hydropathy profile suggests that there exist in the carrier protein 12 hydrophobic segments long enough to traverse the membrane. The amino acid sequence shows a high degree of homology with thebrnQ product, a branched-chain amino acid carrier of Salmonella typhimurium, while no homology in the nucleotide sequences is found in the braB and brnQ genes.  相似文献

Aminoacyl-tRNA synthetases extracted from healthy and crown gall tumor tissues (induced by Agrobacterium tumefaciens strain B6) from Nicotiana tabacum (strain Wisconsin 38) grown in vitro, showed the same ability to charge Phaseolus vulgaristRNA, for all the 15 amino acids tested. For each amino acid, optimal charging conditions (enzyme concentration, Mg2+/ATP ratios, K+ ion effects) have been determined with Phaseolus vulgaristRNA and were found to be the same whether aminoacyl-tRNA synthetases from healthy or tumor tissues were used. In each case, valyl- and glutamyl-tRNA synthetases were very sensitive to an excess of Mg2+ and K+ ions. Although tRNA's extracted from healthy and tumor tissues gave the same electrophoretic patterns, charging levels obtained with turner tRNAs were generally 45% higher than those obtained with tRNA's from healthy tissues.  相似文献

Summary DNA fragments carrying the recA genes of Rhizobium meliloti and Rhizobium leguminosarum biovar viciae were isolated by complementing a UV-sensitive recA Escherichia coli strain. Sequence analysis revealed that the coding region of the R. meliloti recA gene consists of 1044 by coding for 348 amino acids whereas the coding region of the R. leguminosarum bv. viciae recA gene has 1053 bp specifying 351 amino acids. The R. meliloti and R. leguminosarum bv. viciae recA genes show 84.8% homology at the DNA sequence level and of 90.1% at the amino acid sequence level. recA mutant strains of both Rhizobium species were constructed by inserting a gentamicin resistance cassette into the respective recA gene. The resulting recA mutants exhibited an increased sensitivity to UV irradiation, were impaired in their ability to perform homologous recombination and showed a slightly reduced growth rate when compared with the respective wild-type strains. The Rhizobium recA strains did not have altered symbiotic nitrogen fixation capacity. Summary The ribosomal RNA synthesis in a cell-free system containing the nucleoids and the cytoplasmic fraction prepared from Escherichia coli cells has been investigated. The addition of the 4S fraction from the cytoplasm to the isolated nucleoids induces RNA synthesis by a new chain initiation. In this system a preferential initiation of rRNA chains occurs. The experimental results suggest that the 4S fraction contains at least two activities, one for releasing RNA-polymerases from the nucleoids, and another for the frequent initiation of rRNA chains. No restriction of the rRNA synthesis has been observed in the nucleoids and the 4S fraction from the amino acidstarved rel + cells. The rRNA synthesized in the above system is detected at about 23S and 16S rRNA regions.  相似文献

Summary The allelic state of relA influences the phenotype of Escherichia coli strains carrying the lysA22 mutation: lysA22 relA strains are Lys where lysA22 relA + strains grow (slowly) in the absence of lysine. This physiological effect has been related to an effect of the expression of the relA locus on the regulation of lysine biosynthesis. The fully derepressed levels of some lysine enzymes (aspartokinase III, aspartic semialdehyde dehydrogenase, dihydrodipicolinate reductase) are observed under lysine limitation only in rel + strains. And the induction of DAP-decarboxylase by DAP is much higher in rel + than in rel strains when an amino acid limitation of growth is also realised. These results are in agreement with the hypothesis of Stephens et al. (1975) on a possible role of the stringent regulation as a general signal for amino acid deficiency.  相似文献

Summary Twenty proteins were isolated from the 30S ribosomal subunits of Bacillus subtilis and their amino acid compositions and amino-terminal amino acid sequences were determined. These results were compared with the data of Escherichia coli 30S ribosomal proteins and the structural correspondence of individual ribosomal proteins has been established between B. subtilis and E. coli.Post-translational modifications of amino-terminal amino acids of the ribosomal proteins which have been found in E. coli are almost absent in B. subtilis with the exception of acetylated forms of S9.  相似文献

Summary Growth of Caulobacter crescentus CB15 in minimal glucose-ammonium media is inhibited by nine L-amino acids: cysteine, histidine, isoleucine, leucine, methionine, phenylalanine, serine, threonine, and valine. Addition of 10 mM L-alanine prevents the inhibitory response. Several other amino acids also prevented the inhibitory response but to a lesser extent. Mutants resistant to amino acid inhibition designated Aar-, were isolated and characterized. All mutants exhibited a phenotype of cross-resistance to all amino acids and most analogues, regardless of the selective agent. Representative mutations were linked 80% to cysB by RP4-mediated conjugation, and all mutations were linked 10–25% to cysB by transduction. Although the mechanism of amino acid inhibition is not known, the alanine protection and the Aar- phenotype seem to be due to a reduced rate of uptake of inhibitory amino acids.  相似文献

Summary Mutants in thespoT gene have been isolated as stringent second site revertants of therelC mutation. These show varying degrees of the characteristics associated with thespoT1 gene,viz relative amount and absolute levels of both pppGpp and ppGpp and the decay rate of the latter. The entry of3H-guanosine into GTP and ppGpp pools inspoT + andspoT1 cells either growing exponentially or during amino acid starvation was determined, and the rate of ppGpp synthesis and its decay constant calculated. During exponential growth the ppGpp pool is 2-fold higher, its decay constant 10-fold lower, and its synthesis rate 5-fold lower inspoT - than inspoT + cells; during amino acid starvation the ppGpp pool is 2-fold higher, its decay constant 20-fold lower, and its synthesis rate 10-fold lower inspoT than inspoT + cells. In one of the “intermediate”spoT mutants the rate of entry of3H-guanosine into GTP, ppGpp and pppGpp was measured during amino acid starvation. The data form the basis of a model for the interconversion of the guanosine nucleotides in which the flow is:GDP→GTP→pppGpp→ppGpp→Y. Calculations of the rates of synthesis and conversion of pppGpp and ppGpp under various conditions in variousspoT + andspoT - strains indicate that the ppGpp concentration indirectly controls the rate of pppGpp synthesis. ThespoT1 allele was introduced into various relaxed mutants. It was shown that many phenomena associated with the relaxed response ofrelC and “intermediate”relA mutants were phenotypically suppressed when thespoT1 allele was introduced into these mutants. These double mutants exhibit ppGpp accumulation, rate of RNA accumulation, rate of β-galactosidase synthesis, and heat lability of β-galactosidase synthesized during amino acid starvation similar to the stringent wild-type. It is concluded that the relaxed response is due directly to the lack of ppGpp and that the stringest response is due directly to ppGpp.  相似文献

The time course of 14CO2 dark fixation was studied in leaves of the facultatively halophytic plant species Mesembryanthemum crystallinum cultivated with and without 400 mM NaCl in the nutrient medium. It is generally known from the literature that plants grown under saline conditions incorporate 14C predominately into amino acids. By contrast in leaves of M. crystallinum grown on NaCl and exposed to 14CO2 in the dark, relatively more radioactivity is incorporated in the organic acids (especially malate) than in amino acids. The data obtained are discussed in relation to the NaCl induced Crassulacean acid metabolism in M. crystallinum reported earlier.  相似文献

[目的] 通过克隆苹果蠹蛾气味结合蛋白CpomOBP20基因cDNA序列,分析其序列特征和表达谱,旨在更好地了解OBP基因在苹果蠹蛾生命活动过程中的作用,为该害虫的绿色防控提供理论支撑。[方法] 采用RT-PCR法扩增苹果蠹蛾气味结合蛋白CpomOBP20基因cDNA序列,并使用生物信息学软件对其核苷酸和氨基酸序列进行分析;基于qPCR技术分析CpomOBP20基因在苹果蠹蛾4龄幼虫不同组织(头、血淋巴、表皮、脂肪体、中肠、马氏管和唾液腺)以及雌雄成虫不同末端组织(头、触角、下唇须、喙、足和翅)中的表达情况,利用分子对接研究了CpomOBP20与3种保幼激素的结合能力。[结果] 苹果蠹蛾气味结合蛋白CpomOBP20基因的开放阅读框长459 bp,共编码152个氨基酸,等电点为6.30,蛋白分子质量为16.264 ku,N末端具有20个氨基酸组成的信号肽序列,蛋白质序列中具有6个保守的半胱氨酸残基,属于Classical OBP。序列分析表明,CpomOBP20的氨基酸序列与小菜蛾OBP (XP_011557123.1)的一致性最高,在亲缘关系上更加接近。qPCR结果表明,CpomOBP20基因在苹果蠹蛾4龄幼虫以及雌雄成虫不同组织中均有表达,其中在4龄幼虫的血淋巴中表达量最高,在雌雄成虫表达量最高的分别是翅和足,其次是头部。分子对接结果表明,CpomOBP20与3种保幼激素均具有较好的结合能力,可能参与保幼激素的结合与转运。[结论] 本研究明确了CpomOBP20的核苷酸和氨基酸序列的组成及编码蛋白的理化性质,并推测CpomOBP20的作用可能不仅局限于嗅觉识别,在非嗅觉器官中也可能起着重要的生理作用,为今后更深入地探究气味结合蛋白在苹果蠹蛾生命活动中的作用机理提供数据支撑。  相似文献
