Changes in the abscisic acid (ABA) levels in embryo axes of seeds, belonging to the orthodox (Norway maple — Acer platanoides L.) and recalcitrant (sycamore — Acer pseudoplatanus L.) categories, were investigated throughout maturation using an ELISA (enzyme-linked immunosorbent assay) test. Concentration
of ABA in embryo axes substantially differed depending on species and sampling date. ABA was always higher in Norway maple
except at the end of seed maturation when ABA content was similar in both species. During maturation ABA decreased in both
species but the decline was more marked in Norway maple than in sycamore (11 vs. 3 fold). These species also differed in the pattern of ABA changes, which in sycamore embryo axes was very regular, while
in Norway maple a sharp decrease was recorded after acquisition by the seeds of tolerance to desiccation. Dehydration of embryo
axes of Norway maple caused a further significant decrease of ABA level. In contrast, in dehydrated sycamore embryo axes ABA
content did not decrease, but slightly increased. The role of ABA in desiccation tolerance and dormancy of Norway maple and

The process of tracheary element formation by cultured isolated mesophyll cells of Zinnia elegans has been investigated by the use of scanning and transmission electron microscopy. Freshly isolated cells are typified by a thin primary wall, starch-filled chloroplasts and a nucleus with a small nucleolus and peripherally located chromatin. During culture the nucleolus expands and becomes vacuolate, and the chromatin in the nucleus is dispersed. Wall thickening is accompanied by a localisation of microtubules over the thickenings and a slight increase in dictyosome activity. Wall hydrolysis appears to proceed by a previously undescribed mechanism, and gives rise to a granularparticulate appearance in the hydrolysed wall. Tri-iodobenzoic acid inhibits the process of differentiation and induces a polar pattern of growth in the cultured cells.Abbreviation TIBA

The activity of a -(1-4)-xylan synthetase, a membrane-bound enzymic system, was measured in particulate enzymic preparations (1,000 g and 1,000–100,000 g pellets) obtained from homogenates of cambial cells, differentiating xylem cells and differentiated xylem cells isolated from actively growing trees of sycamore (Acer pseudoplatamus) and poplar (Populus robusta). The specific activity (nmol of xylan formed min–1 mg–1 of protein) as well as the activity calculated on a per cell basis (nmol of xylan formed min–1 cell–1) of this enzymic system, markedly increased as cells differentiate from the vascular cambium to xylem. This increase is closely correlated with the enhanced deposition of xylan occurring during the formation of secondary thickening. The possible control of xylan synthesis during the biogenesis of plant cell wall is discussed. 相似文献

After a pre-treatment with red light, hair formation at the growing tip of the siphonaceous green alga Acetabularia mediterranea Lamour. (= A. acetabulum (L.) Silva) can be induced by a pulse of blue light. Red light is needed again after the inductive blue-light pulse if the new whorl of hairs is to develop within the next 24 h. In order to investigate the role of this red light, the duration of the red irradiation was varied and combined with periods of darkness. The response of hair-whorl formation was dependent on the total amount of red light, regardless of whether the red irradiation followed the blue pulse immediately or was separated from it by a period of darkness. Furthermore, periods of exposure to the photosynthesis inhibitor 3-(3,4-dichlorophenyl)-1-1dimethylurea had a similar effect to darkness. Both observations indicate that this red irradiation acts as a light source for photosynthesis. Whether or not the red light had an additional effect via phytochrome was tested in another type of experiment. The dependence of hair-whorl formation on red-light irradiance in the presence of simultaneous far-red irradiation was determined for the pre-irradiation period as well as for the irradiation period after the blue pulse. In both experiments, far-red light caused a small promotion of hair-whorl formation when low irradiances of red light were used. However, these differences were attributable to a low level of photosynthetic activity (which in fact was measurable) caused by red light reflected in the growth chamber. Furthermore, lowering the proportion of active phytochrome by far-red light would be expected to suppress hair-whorl formation. The influence of far-red light was also tested in a strain of Acetabularia mediterranea that developed hair whorls in about 20% of cells even when kept in complete darkness after the blue-light pulse. Far-red irradiation had no effect. These results strongly indicate that phytochrome is not involved in hair-whorl formation. Facilitation is known to be an important process structuring natural plant communities. However, much less is known about
its role in facilitating the invasion of ecosystems by non-native plant species. In this study we evaluated the effects of
invasive (Acer platanoides) and native (Pseudotsuga menziesii) forest types on the performance of A. platanoides seedlings, and related these effects to structural and functional properties associated with the two forest types, in a native
P. menziesii forest that is being invaded by A. platanoides. Acer platanoidesseedlings had higher densities, recruitment, and survival, and experienced less photoinhibition and water stress when beneath
conspecific canopies than in the adjacent P. menziesii forest. Soil moisture and canopy cover were greater in the invaded patch than the native forest. There was no difference
in soil fertility or understory light levels between locations. These demographic (i.e. seedling survival), physiological,
and environmental differences appeared to be due to the effects of A. platanoides and P. menziesii trees. Thus, Acer trees appear to produce a more mesic environment by modifying the structure and phenology of the forest canopy and by altering
the timing of transpirational water loss relative to P. menziesii. Environmental modification by invaders that lead to positive effects on conspecifics may help us to understand the dramatic

The photosynthetic activity of intact sugar maple leaves has been assessed in the presence of exogenous sulphite, bisulphite and sulphate under varying light conditions using photo-acoustic and modulated fluorescence methods. In the light, bisulphite was found to be more toxic than the other two, sulphate being the least toxic of all. Interestingly, the vitality index, Rrd, measured as the ratio of the modulated fluorescence decrease (Fd) to the steady-state fluorescence (Fs), which indicates the efficiency of the whole photosynthetic activity, was more affected than the total photosynthetic energy storage (PES) of PSII and PSI during linear and cyclic electron transport, and Fv/Fm (PSII activity). The severity of the damage appeared to be a function of light intensity. Bisulphite treatment in darkness resulted in a dramatic decrease in Rrd, a moderate decrease in PES and a marginal decrease in Fv/Fm. As for sulphite, the effect was negligible with a tendency for enhanced activity. It is inferred that the Calvin cycle is a good candidate for the primary site of bisulphite and sulphite action. Recovery of activity, especially at the Rrd level, obtained in the presence of ascorbate, glutathione and L-cysteine, indicated a contribution of O2 free radicals to the observed inhibition of the photosynthetic activity in the light. 相似文献

Germinability of isolated embryos from developing fruits of Acer platanoides was high at the earliest developmental stage assessed (90 dpa), but fell subsequently and at seed maturity was very low. These observations showed an inverse correlation with changes in endogenous free abscisic acid (ABA) levels in the embryo, which were low during early ontogeny, but reached maximum levels late in development (150–160 dpa). These observations suggest the possibility that dormancy may be induced during development as a result of ABA accumulation in the embryo, an argument strengthened by the obvious inhibitory effect of added ABA on the germinability of isolated embryos. The cotyledons appear to exert an inhibitory influence on embryo germinability that may result from their free ABA content although the embryonic axis itself possesses an innate dormancy that may reflect its own free ABA content. The increased germinability of isolated embryos resulting form added kinetin serves only to emphasise the complexity of the system and the dangers of simplistic interpretation.The correlation between germinability and ABA content is not complete, however, since much of the reduction in germinability had occurred before any appreciable increase in free ABA levels in the embryo was observed. Indeed the failure of the intact seed to respond to endogenous changes in embryonal ABA levels suggests that even though free ABA in the embryo may influence embryo germinability, it has little effect in the intact seed, where the presence of an intact testa may be a more important factor.The absence of a desiccation phase in the embryo during the late stages of development suggests that the large increases in endogenous free ABA did not cause dormancy by inhibiting water uptake, nor did they result from water stress in the embryonal tissues. 相似文献

Despite the widespread recognition that urban areas are frequently dominated by exotic and invasive plants, the consequences
of these changes in community structure have not been explicitly considered as an explanation for the pattern of advanced
leaf phenology, or early greenup, reported in many urban areas. As such, we evaluated two hypotheses that could account for
advanced greenup in forests along an urban to rural gradient: advanced phenology within individual species or differences
in woody plant community. We monitored the spring leafing phenology of Aesculus glabra (Ohio buckeye), Lonicera maackii (Amur honeysuckle), and Acer negundo (box elder) in 11 forests spanning an urban to rural gradient in central Ohio, USA. From February to April 2006, we monitored
these species, recorded woody plant composition, and documented daily minimum and maximum temperatures at each site. We found
a weak but general trend of advanced phenology within species in more urban landscapes. Monthly average minimum temperatures
were higher with increasing urbanization while monthly average maximum temperatures were similar across the urban to rural
gradient. We also found evidence for shifts in woody plant communities along the urbanization gradient, mainly driven by the
abundance of L. maackii, an invasive exotic species, in the more urban forests. Because L. maackii leafs out weeks earlier than native woody species and is very abundant in urban forests, we suggest that the invasion of
forests by this species can generate earlier greenup of urban forests. 相似文献

Scenedesmus obliquus, strain 633, which synthesizes ketocarotenoids and sporopollenin, also forms pink-red-colored cell walls. Both the cell walls left over after autospore liberation and those from homogenates of disrupted green cells have similar carotenoid pigmentation. Canthaxanthin, astaxanthin, an unidentified ketocarotenoid, and lutein were found as integral cell wall components. A band of cells closest to the cambium in the xylem of tobacco (Nicotiana tabacum L. cv. Samsun) stems oxidized 2,2-azinobis-(3-ethylbenzo-thiazoline-6-sulphonate) (ABTS), o-dianisidine and syringaldazine in the absence of exogenously added hydrogen peroxide. The oxidation was not prevented by catalase which suggests that the oxidation is not dependent on the production and utilisation of endogenous hydrogen peroxide by cell-wall peroxidases. Cell walls, isolated from tobacco xylem, also oxidized these substrates in the absence of added hydrogen peroxide. The cell walls consumed molecular oxygen whilst oxidizing a range of compounds including coniferyl alcohol. The substrate preference and sensitivity to inhibitors suggest the presence of laccasetype polyphenol oxidases (p-diphenol:O2 oxidoreductase EC 1.14.18.1) which are covalently bound to the wall. The oxidation of coniferyl alcohol by the xylem cell walls was confirmed by assays based on the disappearance of coniferyl alcohol and was not affected by the presence of 500 units·mi-1 catalase or Superoxide dismutase. Prolonged incubation of cell walls with coniferyl alcohol led to the production of a yellow-orange water-insoluble material that precipitated with the cell walls. Although a proportion of this material was soluble in methanol, the majority was tightly associated with the cell walls. These coloured cell walls had elevated lignin contents when assayed by the acetyl-bromide method. Fourier transforminfrared spectroscopic analysis of the coloured cell walls indicated that the increased lignin content is due to the deposition of guaiacyl-type lignin. Digestion of the xylem cell walls with Driselase, a mixture of fungal glycases, produced a wall residue that had a dramatically reduced ability to oxidize ABTS in the absence of added H2O2. However, oxidase activity could not be detected in the Driselase-solubilized extract, although small amounts of oxidase activity could be recovered from the Driselaseresistant wall residue by extraction in 3 M CaCl2.Abbreviations ABTS

Diurnal changes of xylem pressure in the lianaTetrastigma voinierianum have been measured under greenhouse conditions by means of the recently developed xylem pressure probe. During the early morning hours, tensions in the vessels developed more or less rapidly with time, depending on light intensity. On sunny days, absolute negative pressures down to about -0.4 MPa (atmospheric = 0.1 MPa) were recorded around noon in petiolar or stem xylem vessels, whereas on rainy or cloudy days the xylem pressure remained in the positive sub-atmospheric or slightly negative pressure range. Towards the evening the tension in the vessels always decreased, i.e. the xylem pressure shifted to about atmospheric, or even above-atmospheric, values during the night. Simultaneous xylem pressure recordings at heights of 1 and 5 m frequently yielded either no gradient in tension at all, or far less than expected from the Cohesion Theory. Occasionally, tension gradients were even opposite to those predicted by this theory. Stem-toleaves pressure gradients in accord with the Cohesion Theory were recorded only when tension had been developed during sunny days in the upper branches of the liana, because increases in tension were not immediately propagated to the xylem of the leaves at ground level, as would be expected from a strictly coupled hydraulic system. Parallel recordings of the xylem tension using the pressure chamber yielded rather variable values ranging from 0.1 to 1 MPa; diurnal pressure changes could not be detected at all. The data are discussed on the basis of the equation for the chemical activity of water. They strongly suggest that the xylem tension induced by transpiration is not the sole force for water ascent. Other forces, such as osmotic pressure or convectional and interfacial forces, which to a remarkable extent have already been postulated for decades, seem to be equally important.Abbreviation R.H.

Summary Cell spreading of isolated melanophores in medium containing fibronectin was observed in the wild type and two mutants of the medaka, Oryzias latipes. Isolated and cultured melanophores of the wild type and the mm mutant were different in appearance from those within scales but dendritic in shape and with fully dispersed pigment granules. Isolated melanophores of the cm mutant were stellate with dispersed pigment granules, whereas in scales the pigment granules are condensed. In the presence of fibronectin, spreading of cultured melanophores of wild type and cm mutant was observed. Spreading of melanophores from the mm mutant was observed only among dendritic melanophores, but not among condensed melanophores. The increase of spreading was inhibited by antibody against fibronectin. To test the involvement of cytoskeletal elements, colchicine, vinblastine or cytochalasin B were added to the culture medium; spreading did not increase, even in the presence of fibronectin. Summary A method for determining the mass flow rate of xylem water in thin stems under natural field conditions is presented. Diurnal courses of xylem water flow and stomatal conductance of the vines Entadopsis polystachya, Cyclanthera multifoliolata, and Serjania brachycarpa were examined in a tropical deciduous forest on the west coast of Mexico. E. polystachya (leaf area 23.6 m2) had a maximum water flow rate of 6.50 kg h-1 or 1.44 kg cm-2
stem basal area h-1; daily water use was 2.00 kg m-2
leaf area day-1. S. brachycarpa (leaf area 4.5 m2) and C. multifoliolata (leaf area 3.6 m2) had a maximum water flow rate of 0.72 and 0.19 kg h-1 or 0.63 and 0.92 kg cm-2
stem basal area h-1. Daily water use was 1.26 and 0.39 kg m-2
leaf area day-1, respectively. The daily courses of xylem water flow were strongly influenced by the orientation of the leaf area to irradiance and its intensity. While leaves of E. polystachya had a constant high stomatal conductance during the day, S. brachycarpa had a maximum stomatal opening in the morning followed by continuous closure during the rest of the day. In contrast to the woody species, the herbaceous C. multifoliolata exhibited a strong midday depression of stomatal conductance and wilting of its leaves. The leaf biomass accounted for 8% (Entadopsis), 16% (Serjania), and 23% (Cyclanthera) of above-ground biomass. The relation of sapwood area to leaf area supplied (Huber value) was 0.19 (Entadopsis), 0.18 (Serjania), and 0.06 (Cyclanthera) cm2 m-2 相似文献

We monitored the distribution of death of secondary xylem cells in a conifer, Abies sachalinensis. The cell death of tracheids, which are tracheary elements, occurred successively and was related to the distance from cambium. Thus, it resembled programmed cell death. By contrast, the death of long-lived ray parenchyma cells had the following features: (1) ray parenchyma cells remained alive for several years or more; (2) in many cases, no successive cell death occurred even within a given radial cell line of a ray; and (3) the timing of cell death differed among upper and lower radial cell lines and other lines of cells within a ray. These results indicate that the death of long-lived ray parenchyma cells involves a different process from the death of tracheids. The initiation of secondary wall formation and the lignification of ray parenchyma cells in the current year's annual ring were delayed in the upper and lower radial cell lines of a ray. In addition, the density of distribution and orientation of cortical microtubules in such cells were different from those in cells in other radial lines. Ray parenchyma cells in the previous year's annual ring within the upper and lower radial cell lines of a ray contained many starch grains. Our results indicate that positional information is an important factor in the control of the pattern of differentiation and, thus, of the functions of ray parenchyma cells that are derived from the same cambial ray cells. 相似文献

Death and decay of trees of Mango (Mangifera indica L.) var. Kesar due to fungal infection was studied histologically. Fungal infection in the trees was observed due to various reasons like mechanical injuries in the stem, pruning of the branches, through the inflorescences, attack of Ambrosia beetle and termites. In the initial stage, fungal spores get settled on the flowers due to presence of nectar, followed by their germination and entry of the hypha into peduncle, which gradually spreads into younger branches. The inflorescences were first attacked by Fusarium moniliformis followed by other fungi like Alternaria, Chetomium sp., Aspregillus ellipticus, Aspregillus niger, etc. Fungal mycelia gradually invade the xylem tissues from the top of the branches and spread basipetally ultimately causing death of the infected branches. During monsoon, the crevices on the surface of bark of the healthy plants supported the growth of fungi like Pleurotus, Auricularia, Xyleria, Daldinia sp., and Polyporous sp. The removal of bark from such infected trees revealed minute holes on the surface of the woody cylinder made by Ambrosia beetles. During wet season fungal mycelia makes an easy entry into the xylem through the wounded portion of the stem or pruned branches. Initial entry of the hyphae into xylem was seen through the ray cells. Then the hyphae enter into the lumen of axial elements lining the ray cells through pits and intracellular spaces. The vessel elements located in the xylem (transition zone) between healthy and infected portion were filled with tyloses while axial and ray parenchyma showed heavy accumulation of tannin contents. On the other hand, the infected xylem was also found devoid of reserve metabolites while in normal trees, axial and ray parenchyma showed heavy accumulation of starch grains. 相似文献

Balance trials were performed to investigate the effects of experimental Eimeria bovis coccidiosis on the metabolism of water, sodium and potassium in calves. Non-infected pair-fed controls and controls fed according to plan were included in the study to allow differentiation between the effects due to infection and due to changes in feed intake. Primary infection with 5 × 104 (group A) or 1 × 105 (group B) oocysts caused mild diarrhoea in three out of four group A calves and mild to severe haemorrhagic diarrhoea in all five group B calves. Losses of sodium and potassium via faeces tended to increase in the infected calves during patency and apparent digestibility (AD) of these minerals was comparably low. In the urine of the infected calves the Na/K-ratio decreased due to a reduced urinary excretion of sodium. The retention (RT) of sodium was particularly high in the calves that had received the higher oocyst dose. Potassium RT did not underlie significant changes during the course of coccidiosis. In the infected calves the plasma level of sodium was reduced transiently while the level of potassium remained fairly stable. Infections with the higher oocyst dose caused a distinct reduction of fluid excretion via urine which compensated for the increased faecal water losses during severe diarrhoea. Reinfection of the group A calves with 1 × 105 oocysts did not cause any significant metabolic impairment. The results of this study indicate that although acute sublethal bovine coccidiosis alters electrolyte and water metabolism the overall balance of electrolytes and water is largely maintained by physiologic adaptation. 相似文献

Monthly uptake rates and the annual deposition of gaseous SO2 via the stomata of six Norway spruce canopies (Picea abies (L.) Karst.) in Germany (Königstein im Taunus, Witzenhausen, Grebenau, Frankenberg, Spessart, Fürth im Odenwald) were calculated (i) from statistical response functions of stomatal aperture depending on meteorological data, and (ii) from the synchronously measured SO2 immission at these stands. The stomatal response functions had been derived on the basis of thorough stomatal water conductance measurements in the field. Calculations of the SO2 conductance of spruce twigs and SO2 uptake rates via stomata need continuously measured complete data sets of the (i) light intensity, (ii) air temperature, (iii) air humidity and (iv) SO2 concentration in spruce forests from all the year. These data were recorded half hourly in different German spruce forests. The apparent needle water vapour pressure difference and transpiration rates were calculated from meteorological data. Additional use of canopy through flow data in dry years allowed the estimation of the mean stomatal conductance for H2O and SO2 of whole spruce canopies. The annual SO2 uptake of a mean unit needle surface in spruce forests was 32% of the SO2 uptake rate of exposed needles at the top of spruce crowns. There is significant SO2 uptake all the year. The mean SO2 dose at all sites and years received through the stomata was (0.25±0.07) mol SO2 m-2 (total needle surface) (nPa Pa-1)-1 (annual mean of SO2 immission; 1 nPa (SO2) Pa-1 (air) = 1 ppb) day-1 (vegetation period per year). Comparison of calculated SO2 uptake rates into needles with measured SO4
2- accumulation rates in needles from the mentioned sites and additionally from Würzburg, Schneeberg (Fichtelgebirge) and from three sites in the eastern Erzgebirge (Höckendorf, Kahleberg, Oberbärenburg) revealed that oxidative SO2 detoxification (SO4
2- formation) dominates only at sites with high SO2 immission and short vegetation periods. Under these conditions 70 to 90% of the annual stomatal SO2 uptake is detoxified via SO4
2- accumulation in needles. Cations are needed for neutralization of accumulating SO4
2- which are inavailable to support growth. Thus, SO2 induces a dominant and competitive additional nutrient cation demand, cation deficiency symptoms and enhanced needle loss (spruce decline symptoms) mainly at sites, where the ratio R=(SO2 immission): (length of the vegetation period) is higher than R=0.07 nPa Pa-1 day-1. Correlation analysis of the relative needle loss versus the SO2-dependent SO4
2- formation rate revealed a significant increase of needle loss at the 98% level (Student). At sites with small SO2 immission and long vegetation periods (R<0.07 nPa Pa-1 day-1) reductive SO2 detoxification via growth (and/or phloem export of SO4
2-) is not kinetically overburdened. Under these conditions only 30% of the annual SO2 uptake is detoxified via SO4
2- formation and spruce decline is small or absent. On the basis of the critical value R0.07 nPa Pa-1 day-1 recommended SO2 immission limits can be deduced on a mere ecophysiological basis. These deduced values are close to the proposed SO2 immission limits of the IUFRO, WHO and the UNECE. 相似文献

凤丹(Paeonia suffruticosa Andr.)为芍药科多年生植物,是一种重要的传统中药资源。在其生长发育的周期中与土壤微生物尤其是根际土壤微生物有密切的关系。通过构建16S rRNA基因克隆文库及文库的限制性片段长度多态性分析(ARDRA),对中国药用植物凤丹5大主要分布区域的根际土壤细菌群落多样性进行了研究。采用限制性内切酶HinfI和Csp6I对克隆文库中随机挑选的1000个白色克隆子进行了酶切分型,根据酶切图谱的不同,将其分为324个OTUs,并对38个优势OTUs进行了测序和系统发育分析。16S rRNA基因序列分析结果表明,凤丹根际土壤细菌种群主要包括:变形菌门(包括alpha、beta、gamma、detla亚门)、酸杆菌门、放线菌门、拟杆菌门及厚壁菌门等11类细菌,此外还包含了3个未归类的细菌。变形菌门和酸杆菌门为文库中的主要菌群,分别占克隆总数的47.34%和14.36%,其中Pseudomonas sp.、Burkholderia sp.和Arthrobacter sp.为优势菌属。研究结果表明,我国药用植物凤丹5大主要分布区域的根际土壤细菌种群不仅具有丰富的多样性,还存在丰富的潜在新菌种。 相似文献
