This paper reports on the effect of different methods of collagen cross-linking on its dielectric properties. In order to obtain collagen-hyaluronic acid (HA) scaffolds, collagen was first dehydrated by a combination of thermal and vacuum drying (DHT) and then treated with the chemical reagent carbodiimide (EDC/NHS) for final cross-linking. The measurements of the relative permittivity varepsilon' and the dielectric loss varepsilon' for all materials were carried over the frequency range of 10 Hz-100 kHz and at temperatures from 22 to 260 degrees C. The results for these samples reveal distinct relaxation processes at low temperatures, below 140 degrees C and at higher temperatures as broad peak around 230 degrees C. The first and second relaxation are associated with changes in the secondary structure of collagen accompanied by the release of water and with the denaturation of dry collagen, respectively. The influence of cross-linking on the permittivity of collagen is significant over the entire temperature range.  相似文献

With an aim to reveal the mechanism of protein-water interaction in a predominantly two phase model protein system this study investigates the frequency and temperature dependence of dielectric constant epsilon' and loss factor epsilon' in cow horn keratin in the frequency range 30 Hz to 3 MHz and temperature range 30-200 degrees C at two levels of hydration. These two levels of hydration were achieved by exposing the sample to air at 50% relative humidity (RH) at ambient temperature and by evacuating the sample for 72 h at 105 degrees C. A low frequency dispersion (LFD) and an intermediate frequency alpha-dispersion were the two main dielectric responses observed in the air-dried sample. The LFD and the high frequency arm of the alpha-dispersion followed the same fractional power law of frequency. Within the framework of percolation cluster model these dispersions, respectively have been attributed to percolation of protons between and within the clusters of hydrogen-bonded water molecules bound to polar or ionizable protein components. The alpha-dispersion peak, which results from intra-cluster charge percolation conformed to Cole-Cole modified Debye equation. Temperature dependence of the dielectric constant in the air-dried sample exhibited peaks at 120 and 155 degrees C which have been identified as temperatures of onset of release of water bound to polar protein components in the amorphous and crystalline regions, respectively. An overall rise in the permittivity was observed above 175 degrees C, which has been identified as the onset of chain melting in the crystalline region of the protein.  相似文献

A model was developed and evaluated as a tool for predicting the formation of soluble products from staged thermochemical treatment of lignocellulosic materials under acidic conditions typical of autohydrolysis. The model was used to predict the general trend of hemi-cellulose and cellulose hydrolysis between pH 2 and 4 and temperatures of 170-230 degrees C, and results were compared with experimental data. When the model was evaluated for this range of temperatures and pH values, results indicated: (1) a relatively low temperature (175 degrees C) during the first stage allows hydrolysis of the hemi-cellulose polysaccharides without significant mono-saccharide decomposition, (2) subsequent stages at higher temperatures (equal or greater than 200 degrees C) are needed for significant cellulose hydrolysis, but glucose decomposition will also occur, and, (3) a pH in the range of 2-2.5 will enhance polysaccharide hydrolysis while limiting monosaccharide decomposition. The model's predictions, indicating that the formation of biodegradable products could be optimized using Pretreatments at pH 2-2.5 for the pH range evaluated, were confirmed in experiments with white fir as a representative lig nocellulose.  相似文献

Two heteroduplexes d(C1A2C3T4C5G6C7A8C9A10C11)-d (G12T13G14T15G16G17A18G19T20G21) containing a bulged guanine either unmodified or modified with the carcinogen N-2-acetylaminofluorene (AAF) have been studied by nuclear magnetic resonance (NMR) as models of slipped mutagenic intermediates (SMI). Conformational equilibria are observed in both the unmodified and the AAF-modified heteroduplexes. The major conformation of the unmodified duplex is one where the extra guanine is stacked in the helix and the major conformation of the AAF-modified heteroduplex is one where the AAF is external to the helix. Unusual sugar proton chemical shifts of C5- and G6-AAF indicate that the AAF ring is pointing out in the 5' direction. A strong increase in the modified heteroduplex melting temperature (+15 degrees C) is observed. Moreover, in contrast to the unmodified heteroduplex, which shows extensive melting in the vicinity of the bulged guanine, the base pairs around the bulge in the AAF-modified heteroduplex remain paired at temperatures up to 30 degrees C. This exceptional stability of the site around the bulged modified guanine is suggested to be responsible for the high rate of -1 mutation induced by AAF at repetitive sequences.  相似文献

Our recent IR study demonstrated that hydrogen-bond structure in cellulose Ibeta drastically changes around 220 degrees C (Watanabe et al. Biomacromolecules 2006, 7, 3164). In the present study, temperature-dependent IR spectra of cellulose Ialpha from 30 to 260 degrees C were analyzed by use of perturbation-correlation moving-window two-dimensional correlation spectroscopy. It was observed that as in the case of cellulose Ibeta abrupt changes in the hydrogen-bond structure occur around 220 degrees C in cellulose Ialpha. It was also revealed that although weakly hydrogen-bonded OH groups in Ibeta are stable below 230 degrees C thermal oxidation of those in Ialpha is accelerated around 220 degrees C. In this way, the present study has clarified a difference between the thermal behavior of Ialpha and that of Ibeta at the functional group level. Our result suggests that the drastic change in the hydrogen-bond structure around 220 degrees C makes cellulose Ialpha much more unstable than Ibeta.  相似文献

1. A fragment of human subcomponent C1q was prepared by limited proteolysis with pepsin at 37 degrees C for 20 h, and at pH4.4, followed by gel filtration on Sephadex G-200. This fragment was shown to contain all the collagen-like features known to be present in the intact molecule [Reid (1976) Biochem. J. 155, 5-17]. 2. Circular-dichroism studies showed the presence of positive bands at 230 and 223 nm in the intact subcomponent C1q and pepsin fragment respectively, compared with a positive band at 220 nm obtained for lathyritic rat skin collagen. These bands were abolished by collagenase treatment, which suggested that there may some collagen-like triple-helical structure in subcomponent C1q and that this structure resides in the pepsin-resistant portion of the molecule. However, the 230 and 223 nm bands had a substantially lower magnitude than that obtained for the unaggregated single fibres of totally triple-helical collagen. 3. Thermal-transition temperatures obtained for subcomponent C1q, the pepsin fragment and the reduced and alkylated pepsin fragment were 48 degrees, 48 degrees and 39 degrees C respectively, compared with a value of 38 degrees C obtained for lathyritic rat skin collagen. 4. Only the unreduced pepsin fragment regained significant amounts (up to 60%) of collagen-like structure, after heat denaturation and cooling, as estimated by circular-dichroism measurements. 5. Electron-microscopy studies of subcomponent C1q and the collagen-like pepsin-resistant fragment of subcomponent C1q showed that the six peripheral globular regions of the molecule were fragmented by pepsin leaving the six collagen-like connecting strands and fibril-like central portion intact.  相似文献

Effect of microwave radiation with the frequency of 1000 +/- 10 MHz and specific absorption rate of 220-580 mV/g on the ferricyanide reduction by human red blood cells in the presence of methylene blue (carrier of oxidation-reduction equivalents through the membrane) was studied at different temperatures in the region of 23-34 degrees C. The temperature dependence of the ferricyanide reduction rate in Arrhenius plots shows two sharp "anomalous" sites with apparently negative activation energy at 26-27 and 29-30 degrees C. Broadness and expression of the "anomalous" sites increased with an increase of the blood storage time. The increase of the ferricyanide reduction rate under microwave irradiation was observed only in the temperature regions corresponding to the "anomalous" sites of the temperature dependence.  相似文献

The alpine cockroach Celatoblatta quinquemaculata is common at altitudes of around 1500 m on the Rock and Pillar range of Central Otago, New Zealand where it experiences freezing conditions in the winter. The cockroach is freeze tolerant, but only to c. -9 degrees C. The cause of death at temperatures below this is unknown but likely to be due to osmotic damage to cells (shrinkage). This study compared the effect of different ice nucleation temperatures (-2 and -4 degrees C) on the viability of three types of cockroach tissue (midgut, Malpighian tubules and fat body cells) and cooling to three different temperatures (-5, -8, -12 degrees C). Two types of observations were made (i) cryomicroscope observations of ice formation and cell shrinkage (ii) cell integrity (viability) using vital stains. Cell viability decreased with lower treatment temperatures but ice nucleation temperature had no significant effect. Cryomicroscope observations showed that ice spread through tissue faster at -4 than -2 degrees C and that intracellular freezing only occurred when nucleated at -4 degrees C. From temperature records during cooling, it was observed that when freezing occurred, latent heat immediately increased the insect's body temperature close to its melting point (c. -0.3 degrees C). This "rebound" temperature was independent of nucleation temperature. Some tissues were more vulnerable to damage than others. As the gut is thought to be the site of freezing, it is significant that this tissue was the most robust. The ecological importance of the effect of nucleation temperature on survival of whole animals under field conditions is discussed.  相似文献

A family of injectable, biodegradable, and thermosensitive copolymers based on N-isopropylacrylamide, acrylic acid, N-acryloxysuccinimide, and a macromer polylactide-hydroxyethyl methacrylate were synthesized by free radical polymerization. Copolymers were injectable at or below room temperature and formed robust hydrogels at 37 degrees C. The effects of monomer ratio, polylactide length, and AAc content on the chemical and physical properties of the hydrogel were investigated. Copolymers exhibited lower critical solution temperatures (LCSTs) from 18 to 26 degrees C. After complete hydrolysis, hydrogels were soluble in phosphate buffered saline at 37 degrees C with LCSTs above 40.8 degrees C. Incorporation of type I collagen at varying mass fractions by covalent reaction with the copolymer backbone slightly increased LCSTs. Water content was 32-80% without collagen and increased to 230% with collagen at 37 degrees C. Hydrogels were highly flexible and relatively strong at 37 degrees C, with tensile strengths from 0.3 to 1.1 MPa and elongations at break from 344 to 1841% depending on NIPAAm/HEMAPLA ratio, AAc content, and polylactide length. Increasing the collagen content decreased both elongation at break and tensile strength. Hydrogel weight loss at 37 degrees C was 85-96% over 21 days and varied with polylactide content. Cell adhesion on the hydrogels was 30% of that for tissue culture polystyrene but increased to statistically approximate this control surface after collagen incorporation. Non-catalytic hydrothermal decomposition of sericin and fibroin from silk waste into useful protein and amino acids was examined in a closed batch reactor at various temperatures, reaction times, and silk to water ratios to examine their effects on protein and amino acid yields. For the decomposition of sericin, the highest protein yield was found to be 0.466 mg protein/mg raw silk, obtained after 10 min hydrothermal reaction of silk waste at 1:100 silk to water ratio at 120 degrees C. The highest amino acid yield was found to be 0.203 mg amino acids/mg raw silk, obtained after 60 min of hydrothermal reaction of silk waste at 1:20 silk to water ratio at 160 degrees C. For the hydrothermal decomposition of fibroin, the highest protein yield was 0.455 mg protein/mg silk fibroin (1:100, 220 degrees C, 10 min) and that of amino acids was 0.755 mg amino acids/mg silk fibroin (1:50, 220 degrees C, 60 min). The rate of silk fibroin decomposition could be described by surface reaction kinetics. The soluble reaction products were freeze-dried to obtain sericin and fibroin particles, whose conformation and crystal structure of the particles were shown to differ from the original silk materials, particularly in the case of fibroin, in which the change from beta-sheet conformation to alpha-helix/random coil was observed.  相似文献

The effects of temperature on the assembly of collagen fibrils were examined in a system in which collagen monomers are generated de novo and in a physiological buffer by specific enzymic cleavage of type I pC-collagen, an intermediate in the normal processing of type I procollagen to type I collagen. Increasing the temperature of the reaction in the range of 29-35 degrees C decreased the turbidity lag and increased the rate of propagation as assayed by turbidity. The effect of temperature on the turbidity propagation rate gave a linear Arrhenius plot with a negative slope. The predicted value of the activation energy of propagation was 113 kJ/mol. However, the effects of temperature on the rate of assembly above 37 degrees C were opposite to the effects seen at temperatures below 37 degrees C. In the range of 37-41 degrees C, the turbidity propagation rate decreased markedly with temperature. Also, the turbidity lag increased. Therefore, much longer times were required for monomers to reach equilibrium with fibrils. A large fraction of the collagen monomers remaining in solution at temperatures above 37 degrees C was sensitive to rapid digestion by trypsin and alpha-chymotrypsin. Cooling the solutions to 25 degrees C made the monomers resistant to protease digestion. The results are consistent with the conclusion that, although formation of collagen fibrils is a classical example of an entropy-driven process of self-assembly, the rate of assembly between 37 and 41 degrees C is limited by reversible micro-unfolding of the monomer.  相似文献

Hydrolysis of the straw material Paja Brava, a sturdy grass characteristic for the high plains of Bolivia, was studied in order to find suitable conditions for hydrolysis of the hemicellulose and cellulose parts. Dried Paja Brava material was pre-steamed, impregnated with dilute sulfuric acid (0.5% or 1.0% by wt), and subsequently hydrolyzed in a reactor at temperatures between 170 and 230 degrees C for a reaction time between 3 and 10 min. The highest yield of xylose (indicating efficient hydrolysis of hemicellulose) were found at a temperature of 190 degrees C, and a reaction time of 5-10 min, whereas considerably higher temperatures (230 degrees C) were needed for hydrolysis of cellulose. Fermentability of hemicellulose hydrolyzates was tested using the xylose-fermenting yeast species Pichia stipitis, Candida shehatae and Pachysolen tannophilus. The fermentability of hydrolyzates decreased strongly for hydrolyzates produced at temperatures higher than 200 degrees C.  相似文献

Increased decomposition rates in boreal peatlands with global warming might increase the release of atmospheric greenhouse gases, thereby producing a positive feedback to global warming. How temperature influences microbial decomposers is unclear. We measured in vitro rates of decomposition of senesced sedge leaves and rhizomes (Carex aquatilis), from a fen, and peat moss (Sphagnum fuscum), from a bog, at 14 and 20 degrees C by the three most frequently isolated fungi and bacteria from these materials. Decomposition rates of the bog litter decreased (5- to 17-fold) with elevated temperatures, and decomposition of the sedge litters was either enhanced (2- to 30-fold) or remained unaffected by elevated temperatures. The increased temperature regime always favoured fungal over bacterial decomposition rates (2- to 3-fold). Different physiological characteristics of these microbes suggest that fungi using polyphenolic polymers as a carbon source cause greater mass losses of these litters. Litter quality exerted a stronger influence on decomposition at elevated temperatures, as litter rich in nutrients decomposed more quickly than litter poorer in nutrients at higher temperatures (8.0%-25.7% for the sedge litters vs. 0.2% for the bryophyte litter). We conclude that not all peatlands may provide a positive feedback to global warming. Cautious extrapolation of our data to the ecosystem level suggests that decomposition rates in fens may increase and those in bogs may decrease under a global warming scenario.  相似文献

Dynamic viscoelastic properties of collagen solutions with concentrations of 0.5-1.5% (w/w) were characterized by means of oscillatory rheometry at temperatures ranging from 20 to 32.5 degrees C. All collagen solutions showed a shear-thinning flow behavior. The complex viscosity exhibited an exponential increase and the loss tangent decreased with the increase of collagen concentration (C(COL)) when the C(COL)> or =0.75%. Both storage modulus (G') and loss modulus (G') increased with the increase of frequency and concentration, but decreased with the increase of temperature and behaved without regularity at 32.5 degrees C. The relaxation times decreased with the increase of temperature for 1.0% collagen solution. According to a three-zone model, dynamic modulus of collagen solutions showed terminal-zone and plateau-zone behavior when C(COL) was no more than 1.25% or the stated temperature was no more than 30 degrees C. The concentrated solution (1.5%) behaved being entirely in plateau zone. An application of the time-temperature superposition (TTS) allowed the construction of master curve and an Arrhenius-type TTS principle was used to yield the activation energy of 161.4 kJ mol(-1).  相似文献

Temperature is an important factor regulating microbial activity and shaping the soil microbial community. Little is known, however, on how temperature affects the most important groups of the soil microorganisms, the bacteria and the fungi, in situ. We have therefore measured the instantaneous total activity (respiration rate), bacterial activity (growth rate as thymidine incorporation rate) and fungal activity (growth rate as acetate-in-ergosterol incorporation rate) in soil at different temperatures (0-45 degrees C). Two soils were compared: one was an agricultural soil low in organic matter and with high pH, and the other was a forest humus soil with high organic matter content and low pH. Fungal and bacterial growth rates had optimum temperatures around 25-30 degrees C, while at higher temperatures lower values were found. This decrease was more drastic for fungi than for bacteria, resulting in an increase in the ratio of bacterial to fungal growth rate at higher temperatures. A tendency towards the opposite effect was observed at low temperatures, indicating that fungi were more adapted to low-temperature conditions than bacteria. The temperature dependence of all three activities was well modelled by the square root (Ratkowsky) model below the optimum temperature for fungal and bacterial growth. The respiration rate increased over almost the whole temperature range, showing the highest value at around 45 degrees C. Thus, at temperatures above 30 degrees C there was an uncoupling between the instantaneous respiration rate and bacterial and fungal activity. At these high temperatures, the respiration rate closely followed the Arrhenius temperature relationship.  相似文献

Native collagen polypeptides exist in a unique triple helical conformation resistant to most proteinases. In this study, the stability of type I collagen triple helix, employing a mixture of trypsin and alpha-chymotrypsin as a proteolytic probe, was examined. The degradation of type I [3H]collagen was monitored as 3H-labeled peptides soluble in trichloroacetic acid (TCA) or by sodium dodecyl sulfate (SDS)-polyacrylamide slab gel electrophoresis. In one set of experiments, collagen substrates were preincubated at various temperatures for up to 8 h, followed by a 15-min proteolytic treatment at the same temperature. At 43 degrees C, most of the collagen was degraded, while the fraction of the substrate degraded at 40, 38, and 35 degrees C was 53, 41 and 19%, respectively. This fraction was independent of the preincubation time which varied from 10 to 480 min. Thus, at any given temperature, a constant fraction of the collagen substrate was susceptible to proteolysis. Measurement of the midpoint temperature (Tm) of the helix to coil transformation for type I collagen, at neutral pH employing an increasing temperature gradient and brief proteolysis at the individual temperatures, indicated a value of 38.8 degrees C. However, determination of the Tm by employing proteolytic digestions at a constant temperature (30 degrees C) using conditions under which the nonhelical peptides are readily digested to TCA-soluble peptides while native collagen resists such proteolysis, indicated a value of 42.7 degrees C. In further studies, collagen was subjected to continuous proteolysis for up to 24 h. A large fraction of collagen was digested at 30 or 34 degrees C, temperatures well below the Tm of the helix to coil transformation. SDS-polyacrylamide gel electrophoresis of the degradation products obtained at these temperatures revealed multiple cleavage fragments. Finally, temperature double-jump experiments indicated that the destabilization of the triple helix is reversible provided that the Tm of the substrate is not exceeded. The results provide evidence for reversible and local relaxation of the collagen triple helix.  相似文献

Complex dielectric constants of human erythrocyte suspensions over a frequency range from 45 MHz to 26.5 GHz and a temperature range from 5 to 40 degrees C have been determined with the open-ended coaxial probe technique using an automated vector network analyzer (HP 8510). The spectra show two separate major dispersions (beta and gamma) and a much smaller dispersion between them. The two major dispersions are analyzed with a dispersion equation containing two Cole-Cole functions by means of a complex nonlinear least squares technique. The parameters of the equation at different temperatures have been determined. The low frequency behavior of the spectra suggests that the dielectric constant of the cell membrane increases when the temperature is above 35 degrees C. The real part of the dielectric constant at approximately 3.4 GHz remains almost constant when the temperature changes. The dispersion shifts with temperature in the manner of a thermally activated process, and the thermal activation enthalpies for the beta- and gamma-dispersions are 9.87 +/- 0.42 kcal/mol and 4.80 +/- 0.06 kcal/mol, respectively.  相似文献

The effect of temperature on the translocation frequency of the Tn3 element was investigated. The temperature optimum for translocation of Tn3 was in the range from 26 to 30 degrees C. At temperatures above 30 degrees C, the translocation frequency decreased rapidly and linearly; at 36 degrees C it was only 5% of the frequency observed at 30 degrees C. The duration and reversibility of the temperature effect were utilized to demonstrate a requirement for protein synthesis in the translocation process.  相似文献

1. Temperatures of different body surface regions and deep body temperature (Tb) of unrestrained adult Mongolia gerbils exposed to ambient temperatures (Ta) of -10-35 degrees C were measured using infrared (i.r.) thermography and a thermocouple. 2. A strong positive linear relationship between the surface temperature and Ta was found. For Ta range -4-35 degrees C, the slope was lowest for the areas around the eyes and dorsal head, and steepest for the body extremities. At -10 degrees C, surface temperatures of the areas around the eyes and dorsal head were significantly lower then predicted. 3. Tb was lowest at Ta of 25 and 30 degrees C, increased at all temperatures above and up to Ta of -4 degrees C below this range, and began decline at -10 degrees C. 4. The thermoneutral zone (TNZ) is probably between 28 and 32 degrees C, and the absolute lower critical temperature (Tabsl) is probably -4 degrees C. 5. The Mongolian gerbil shows little control of surface temperature and in contrast to larger mammals it has not developed any special thermoregulatory surface areas to regulate heat exchange with its environment. At temperatures below -4 degrees C, this species is unable to maintain the surface temperature of body extremities above the freezing point. 6. It is suggested that the Mongolian gerbil uses mainly behavioral and ecological adaptive strategies to attenuate the stressful effects of its habitat.  相似文献

To examine the thermal stability of the helical structure of type IV collagen within basement membranes in situ, we have employed indirect immunofluorescence histochemistry performed at progressively higher temperatures using a conformation-dependent antibody, IV-IA8. We previously observed by competition enzyme-linked immunosorbent assay that, in neutral solution, the helical epitope to which this antibody binds undergoes thermal denaturation over the range of 37-40 degrees C. In the present study, we have reacted unfixed cryostat tissue sections with this antibody at successively higher temperatures. We have operationally defined denaturation as the point at which type IV- specific fluorescence is no longer detectable. Under these conditions, the in situ denaturation temperature of this epitope in most basement membranes is 50-55 degrees C. In capillaries and some other small blood vessels the fluorescent signal is still clearly detectable at 60 degrees C, the highest temperature at which we can confidently use this technique. We conclude that the stability of the helical structure of type IV collagen within a basement membrane is considerably greater than it is in solution, and that conformation-dependent monoclonal antibodies can be useful probes for investigations of molecular structure in situ.  相似文献
