In situ light measurements were used to obtain information oninherent and apparent optical properties. The average verticalattenuation coefficient Kd(ave) varied from 1.1 to 4.6 In unitsm–1 During three periods the variation in Kd(ave) correlatedwith changes in chlorophyll a concentration and specific attenuationcoefficients Ks, of 0.013, 0.014 and 0.022 m2 mg Chl a–1were calculated. Chlorophyll-specific diffuse absorption coefficients(A,) for these periods were 0.012. 0.013 and 0.017 m2 mg Chla–1 and only varied significantly from estimates of Ksin the period when scattering was intense. Absorption coefficientsa(zmid) and scattering coefficients b(zmid) calculated for themid-point of the euphotic zone ranged between 0.45 and 2.9 mand 3.5–52.0 m respectively. Chlorophyll-specific absorptioncoefficients Ka, of 0.005, 0.006 and 0.007 m2 mg Chl a–1and scattering coefficients Kb of 0.05. 0.09 and 0.191 m2 mgChl a–1 were measured during the three periods. The highKb value occurred when gas-vacuolate cyanobactena were dominant.Algal photosynthesis and light absorption were related throughthe maximum quantum yield m which varied between 0.019 and 0.11mol C Einstein–1 while average quantum yields a, variedbetween 0.006 and 0.024 with a mean of 0.013 mol C Einstein–1A comparison of changes in the mean irradiance of the mixedzone and chlorophyll concentration indicated that growth waslight limited below 0.04–0.05 Einsteins absorbed mg Chla–1 day–1.  相似文献

The influence of various treatments and temperature regimeson total chlorophylls and on the chlorophyll a:b ratio of westernwheatgrass and blue grama plants was investigated at differenttime intervals during the 120-day growth period. Western wheatgrass,a C3 species, accumulated greater amounts of chlorophyll thandid blue grama plants, a C4 species. Maximum concentrations(mg gd wt–1) of chlorophylls in western wheatgrass andin blue grama were recorded at the lower (13/7°C) and higher(30/18°C) temperature regimes. Nitrogen fertilizer alonedecreased the chlorophyll content in both species. The chlorophylla:b ratio in blue grama ranged from an average of 2·00under irrigated plus fertilized conditions to 3·00 undercontrol and fertilized conditions. On the other hand, the chlorophylla:b ratio in western wheatgrass remained constant at 3·00throughout the growing season under various treatments and temperatureregimes.  相似文献

Populations of carrot (Daucus carota) were raised over a widerange of densities (79–5763 plants m-2) to examine thedynamics of competition in terms of yield–density relationshipsand size variability, and to investigate the effects of nutrientsupply on competition. While the relationship between shootyield and density was asymptotic, the relationship between rootand total yield and density tended to be parabolic. For a giventime and density series the relationship between yield per unitarea and density could best be described by the model: y=wmD(1+aD)b wherey is the yield per unit area,D is density,wm, a andb arefitted parameters. The calanoid copepod, Eudiaplomus graciloides, was reared fromegg to adult on uni-algal diets (0.1. 0.5 and 2.5 mg dry wt1–1) using the green alga, Chlamydomonas reinhardtii,as food, or on a mixed diet consisting of Lake Esrom water filteredthrough a plankton net with pore size 45 µm and supplementedwith C. reinhardtii (2.5 mg dry wt 1–1). On the mixeddiet at 21.0°C growth in body dry wt (W, µg dry wt)was exponential, and the growth constants were 0.21 day–1in the early to mid juvenile stage (N1 - C4) and 0.11 day–1in the late juvenile to early adult stage (C4-A). At 14.5°Cthe corresponding growth rate constants were 0.10 and 0.08 day–1.Similar growth rates were found at uni-algal concentrationsof 0.5 and 2.5 mg dry wt I–1, and it was argued that thethreshold concentration for growth in Eudiaptomus was closeto 0.1 mg dry wt I–1. The clearance (C, ml h–1)of copepodites was measured on the uni-algal diets. The constantsof the regression (C = aWb) were: a = 0.125, b = 0.858 (2000C. reinhardtii ml–1), a = 0.068, b = 0.849 (10 000), a= 0.028, b = 0.875 (50 000). Ingestion rates were calculatedfrom the clearances and the average algal concentrations. Atthe three food levels the average daily rations were 30, 67and 125% of body dry wt. The respiration rate (R, nl O2 h–1)was measured in individuals reared on the mixed diet. The constantsof the regression (R = aWb) were: a = 4.82, b = 1.07 (nauplii,14.5°C), a = 4.17, b = 0.904 (copepodites and adults, 14.5°C),a = 6.87, b = 0.757 (copepodites and adults, 21.0°C). Nosignificant difference in the respiration rate of copepoditesreared on uni-algal diets and the mixed diet could be demonstrated.Energy budgets were calculated. The assimilation efficiencyand the gross growth efficiency of copepodites decreased markedlywith increasing food concentration, the net growth efficiencyvaried from an average of 0.44 at the lowest algal concentrationto 0.60 on the mixed diet. Monocultures of Vulpia fasciculata were grown over a wide rangeof densities to investigate the influence of crowding and nutrientsupply on growth and self-thinning. For a given time and densityseries the relationship between mean yield per plant (w) andthe density of survivors (N) could be described by the equation w= wm (1+aN)–b. where wm is the yield of an isolated plant, a is the area requiredto achieve a yield of wm and b describes the effectiveness withwhich resources are taken up from the area. All three parametersincreased with time. Adding nutrients changed not only the rate at which the effectsof crowding occurred but also the intensity of crowding since wm = C(ab)D. where C and D are constants. The addition of nutrients resultedin an increase in the value of C. Such an increase means thata larger weight can be supported by a given area because theresources within that area are greater. During the early phases of growth, populations of V. fasciculataconformed to the –3/2 power law, w = cN–3/2, butonly at very high densities with a plentiful supply of nutrients.However, once the maximum standing crop had been reached thetrajectory of the thinning line switched to a slope of justless than –1 when weight was ploted against density onlogarithmic scales. The intercept of the –3/2 thinningline was considerably higher (log c = 5.74) than those for mosttrees and forbs but was similar to those of a number of othergrasses. The causes of interspecific differences in the µ-l relationshipare examined in the context of a mechanistic model which relatesµ to irradiance in terms of six factors:, kc photosyntheticquotient (PQ), Chl a:C, respiration and excretion. The effectof cell size on the light saturated growth rate is also considered.It is shown that photosynthetic efficiency and PQ exhibit remarkablylittle interspecific variability, and average 0.024 ±0.005 µg C(µg Chl a)–1 h–1 (µEm–2 s–1)–1 and 1.5 ± 0.2 mol 02 molC–1 (when NO3 is the nitrogen source) respectively.Two useful relationships were derived: (i) between growth efficiency,g and Chl a:C at µ. = 0; (ii) between the compensationintensity, Ic and the Chl a-specific maintenance respirationrate. Both relationships were independent of temperature anddaylength. Species best adapted to growth at low light werefound to exhibit high Chl a:C ratios and low maintenance respirationrates. As a group, diatoms were consistently the best adaptedfor growth at low irradiance. Chiorophytes, haptophytes, chrysophytesand cryptophytes were intermediate in their performance at lowirradiance. Dinoflagellates exhibited extreme diversity, withspecies spanning the spectrum from very good performance atlow irradiance to very poor. A new µmax-cell carbon relationshipis given based on growth rates normalized to 15°C. Evidenceis presented to show that noise in this relationship can besignificantly reduced by using only carbon-specific growth ratesand using only data for species grown at the same daylength.  相似文献

In situ growth and development of Neocalanus flemingeri/plumchrusstage C1–C4 copepodites were estimated by both the artificial-cohortand the single-stage incubation methods in March, April andMay of 2001–2005 at 5–6°C. Results from thesetwo methods were comparable and consistent. In the field, C1–C4stage durations ranged from 7 to >100 days, dependent ontemperature and chlorophyll a (Chl a) concentration. Averagestage durations were 12.4–14.1 days, yielding an averageof 56 days to reach C5, but under optimal conditions stage durationswere closer to 10 days, shortening the time to reach C5 (fromC1) to 46 days. Generally, growth rates decreased with increasingstage, ranging from 0.28 day–1 to close to zero but weretypically between 0.20 and 0.05 day–1, averaging 0.110± 0.006 day–1 (mean ± SE) for single-stageand 0.107 ± 0.005 day–1 (mean ± SE) forartificial-cohort methods. Growth was well described by equationsof Michaelis–Menten form, with maximum growth rates (Gmax)of 0.17–0.18 day–1 and half saturation Chl a concentrations(Kchl) of 0.45–0.46 mg m–3 for combined C1–3,while Gmax dropped to 0.08–0.09 day–1 but Kchl remainedat 0.38–0.93 mg m–3 for C4. In this study, in situgrowth of N. flemingeri/plumchrus was frequently food limitedto some degree, particularly during March. A comparison withglobal models of copepod growth rates suggests that these modelsstill require considerable refinement. We suggest that the artificial-cohortmethod is the most practical approach to generating the multispeciesdata required to address these deficiencies.  相似文献

Pea plants were grown under intermittent illumination (ImL)conditions. The low dosage of light given to ImL plastids limitedthe rate of chlorophyll (Chl) a and Chl b biosynthesis and,therefore, it retarded the rate of photosynthetic unit formationand thylakoid membrane development. Depending on the developmentalstage of the photosynthetic unit, ImL plastids had variableChl a/Chl b ratios (2.7 <Chl a/Chlb<20) and showed distinctintermediates in the assembly of the chlorophyll a–b light-harvestingcomplex (LHC) of photosystem-II (PSII). The results are consistentwith a step-wise increment in the PSII antenna size involvingthree distinct forms of the PSII unit: (i) a PSII-core formwith about 37 Chl a molecules; (ii) a PSILß form containingthe PSII-core and the LHC-II-inner antenna with a total of about130 Chl (a + b) molecules, and (iii) the mature PSIIa form containingPSIIß and the LHC-II-peripheral antenna with a totalof 210–300 Chl (a + b) molecules. The thylakoid membranecontained polypeptide subunits b, c and d (the Lhcb1, 2 and3 gene products, respectively) when only the LHC-II-inner waspresent. Polypeptide subunit a, (the apoprotein of the chlorophyll-proteinknown as CP29), along with increased amounts of b and c appearedlater in the development of thylakoids, concomitant with theassembly of the LHC-II-peripheral. The results suggest thatpolypeptide subunit d has priority of assembly over subunita. It is implied that, of all LHC-II constituent proteins, subunitd is most proximal to the PSII-core complex and that it servesas a linker in the transfer of excitation energy from the bulkLHC-II (subunits b and c) to the PSII-core. The work also addressesthe origin of low-molecular-weight proteins (Mr = 19, 17.5 and13.4 kDa) which co-isolate with intact developing plastids andwhose abundance decreases during plastid development. A model of carbon isotope discrimination by phytoplankton wasdeveloped which took into account the occurrence of a carbon-concentratingmechanism (CCM). A simple equation was obtained for the modelinvolving CO2 active transport. In the case of HCO3 activetransport, another equation was developed based on a seriesof approximations. The former equation was used to analyse reportedand newly obtained data from culture experiments and field observationsin both freshwater and marine environments. In most cases, alinear relationship between a combined parameter, (1–f)Ci,which was made up of the relative contribution of active CO2uptake to total carbon uptake (f) and the intracellular CO2concentration (Ci), and CO2 concentration in bulk solution (Ce)was obtained as (1–f)Ci = ace–b, with a high correlationcmfficient (r2>0.9). The slope a is suggested as a measureof the ratio of diffusive to total (diffusive+active) CO2 transport,while bla represents CO2 demand.  相似文献

The lacustrine blue-green alga (= Cyanobacterium) Oscillatoriarubescens D.C. was cultured in chemostats with temperature andlight intensity as the only limiting factors. The experimentswere carried out under combinations of three temperatures 10,20 and 30°C and three irradiances 6, 18 and 30 µEm–2 s–1. Dilution rate, photosynthesis rate anda large number of abiotic and cellular factors were regularlymeasured to determine steady-state periods and the level ofthe associated variables. Various mathematical models were fittedwith series of data by a non-linear regression method derivedfrom Marquardt's method. Four models for calculation of specificgrowth rate and photosynthesis rate are presented. The lastone computes growth rate from calculated photosynthesis: µ= µmax pmaxI/[KpKi + I(Kp + Pmax)], with Pmax =100 + 257Tand K1 = 300/chl a. Values of adjusted parameters are discussed.One conclusion confirms that values of maximal growth ratesand half-saturation constants frequently considered in the literatureas absolute species characteristics should always be consideredas relative to associated prevalent growth conditions.  相似文献

Long-term effects of atmospheric carbon dioxide concentration(ambient or 700 µmol mol–1) and air temperature(simulation of field conditions or + 4 C) on leaf photosyntheticrate were examined in Lolium perenne L. cv. Vigor, exposed tonatural illumination during winter. Photosynthetic capacitywas compared over a range of air temperatures and photon fluxdensities of photosynthetically active radiation which wererepresentative of winter climate (5–15 C and 0–500µmol m–2s–1), with CO2 level during measurementsimilar to that during the experimental period. Long-term exposureto increased air temperature reduced leaf CO2 fixation capacityby 23% (averaged over all measurement conditions), resultingfrom a decline in lightsaturated uptake rate, but not in incident-lightquantum efficiency. CO2- stimulation was largely absent in plantsgrown in ambient temperature, but pronounced in plants grownunder +4 C, where it compensated for two-thirds of the 23%drop. This enhancing effect of elevated CO2 level on leaf CO2uptake rate observed in the warmer treatment, was strongly dependenton measurement temperature, increasing from 5% at 5 C, to upto 32% at 15 C. Measurements of chlorophyll fluorescence anddry matter corresponded with the observed changes in assimilationcapacity, which could not be attributed to a deteriorated nitrogenstatus of the leaves as there was a similar N content on anarea basis. Diurnal series of fluorescence and photosynthesis assays wereconducted in high altitude (3803 m), tropical (16°), LakeTiticaca (Peru/Bolivia). Near-surface diurnal thermoclines formedon typical days of high photon flux density (PFD, {small tilde}2000 µE m–2 s–1). In the depth range of diurnalstratification profiles of in vivo fluorescence, both without(Fa and with (Fb DCMU, exhibited a mean decrease of 64% frommorning to mid-day, but little change (mean increase of 1.5%)through the afternoon. Three times during the day surface, mid-depth(3–5 m) and deep (15–20 m) phytoplankton sampleswere incubated with H14CO3 under short (<2 h) exposuresto a range of in situ PFDs. Comparison of phytoplankton in differentsamples (ANOVA) showed identical photosynthetic response insunrise (isothermal) samples but a significant drop in surfaceand mid-depth photosynthesis at all PFDs during times of diurnalstratification. Similarly, both low-light () and light-saturated(P2 max photosynthetic parameters were lower in mid-day surfacesamples compared to deep samples. In addition, previously photoinhibitedsamples had a higher threshold intensity for photoinhibition,IT. These results, together with diurnal time series of fluorescencefrom in situ incubations, demonstrate that recovery from extendedepisodes of photoinhibition during diurnal stratification isslower than suggested by previous observations in vitro. Photosynthesisby near-surface phytoplankton is different in light increasingup to IT than light decreasing from IT. A method is described for the rapid separation of carbonic anhydrase(CA) isozymes by cellulose acetate membrane electrophoresisin which CA activity is detected using the pH-indicating dye,bromcresol purple. This method can detect bovine erythrocyteCA in a 0.3 mm3 sample applied at a concentration of 100 ngcm–3 (total of 30 pg applied) while at higher concentrationsthree isozymes were observed. It was found, using a potentiometrictechnique, that intact cells of Anabaena flos-aquae (Cyanophyceae)and Chlorella ellipsoidea had no detectable activity while C.saccharophila and Chlamydomonas reinhardtii (Chlorophyceae)had external CA activity. CA activity of the extracts suggestedthe presence of internal CA in all species. After electrophoresisit was found that C. saccharophila and C. reinhardtii had twoisozymes while A. flos-aquae and C. ellipsoidea had only a singledetectable band. Spinach had up to five detectable isozymesthat were difficult to resolve. Incubation of spinach extractwith the CA inhibitor ClO4 (500 mol m–3) inhibitedCA activity by 90% using the potentiometric technique, but afterelectrophoresis had no detectable effect. This technique isuseful in identifying isozymes that are substantially differentin electrical charge and in monitoring CA isozyme activity duringenzyme purification. Planktonic primary production in the German Wadden Sea   总被引:8,自引:0,他引:8  
By combining weekly data of irradiance, attenuation and chlorophylla concentrations with photosynthesis (P) versus light intensity(E) curve characteristics, the annual cycle of planktonic primaryproduction in the estuarine part of the Northfrisian WaddenSea was computed for a 2 year period. Daily water column particulategross production ranged from 5 to 2200 mg C m–2 day–1and showed a seasonal pattern similar to chlorophyll a. Budgetcalculation yielded annual gross particulate primary productionsof 124 and 176 g C m–2 year–1 in 1995 and 1996,respectively. Annual amounts of phytoplankton respiration, calculatedaccording to a two-compartment model of Langdon [in Li,W.K.W.and Maestrini,S.Y. (eds), Measurement of Primary Productionfrom the Molecular to the Global Scale. International Councilfor the Exploration of the Sea, Copenhagen, 1993, pp. 20–36],and dissolved production in 1996, were both in the range of24–39 g C m–2 year–1. Annual total net productionwas thus very similar to particulate gross production (127 and177 g C m–2 year–1 in 1995 and 1996, respectively).Phytoplankton growth was low or even negative in winter. Inspring and summer, production/biomass (Pr/B) ratios varied from0.2 up to 1.7. Phytoplankton growth during the growth seasonalways surpassed average flushing time in the area, thus underliningthe potential of local phytoplankton bloom development in thispart of the Wadden Sea. The chlorophyll-specific maximum photosyntheticrate (PBmax) ranged from 0.8 to 9.9 mg C mg–1 Chl h–1and was strongly correlated with water temperature (r2 = 0.67).By contrast, there was no clear seasonal cycle in B, which rangedfrom 0.007 to 0.039 mg C mg–1 Chl h–1 (µmolphotons m–2 s–1)–1. Its variability was muchless than PBmax and independent of temperature. The magnitudeand part of the variability of PBmax and B are presumably causedby changes in species composition, as evidenced from the rangeof these parameters found among 10 predominant diatom speciesisolated from the Wadden Sea. The ratio of average light conditionsin the water column (Eav) to the light saturation parameterEk indicates that primary production in the Wadden Sea regionunder study is predominantly controlled by light limitationand that nutrient limitation was likely to occur for a few hoursper day only during 5 (dissolved inorganic nitrogen) to 10 (PO4,Si) weeks in the 2 year period investigated.  相似文献

When disks excised from leaves fumigated with SO2 at 2 ppm for2 hr were illuminated, chlorophyll a and carotenoids were brokendown, while they were not destroyed in darkness. The destructionof these pigments was suppressed under nitrogen. Chlorophylla destruction was inhibited by l,2-dihydroxybenzene-3,5-disulfonate(tiron), hydro-quinone and ascorbate, but not by l,4-diazabicyclo-[2,2,2]-octane(DABCO), methio-nine, histidine, benzoate and formate. Chlorophylla destruction was inhibited by phenazine methosulfate but stimulatedby methyl viologen. Addition of superoxide dismutase (SOD) tothe homogenate of SO2-fumigated leaves inhibited the chlorophylla destruction. The activity of endogenous SOD was reduced to40% by 2-hr fumigation before the loss of chlorophyll was observed.These results suggest that chlorophyll a destruction by SO2was due to superoxide radicals (O2). Moreover, malondialdehyde (MDA), a product of lipid peroxidation,was formed in SO2-fumigated leaves. MDA formation was inhibitedby tiron, hydroquinone and DABCO but not by benzoate and formate.MDA formation was increased by D2O. A rapid induction of sulfate transport was observed in the greenalga Chlorella ellipsoidea during sulfur-limited growth. Bothaffinity and Vmax increased about five-fold within 6 h of transferringcells from Bold's basal medium with 350 µM MgSO4 to sulfur-deficientBold's medium. High affinity sulfate transport was induced within15 min and reached maximum rate within 3 h of transferring cellsto sulfur-deficient condition, indicating that a new, high-affinity-sulfatetransport system is induced by sulfur starvation in C. ellipsoidea.Eadie-Hofstee plots of initial rates of sulfate uptake indicatedthat the K of sulfur-starved cells was about 17 µM. Bothsulfur-starved and unstarved cells grown in air had a Vmax of1.5 times higher than that of high-CO2 grown cells. Sulfatetransport was completely inhibited by 30 µM CCCP or 800µMKCN both in the light and the dark but transport in the lightwas not inhibited by 20 µM DCMU. Treatment with 50 µMor 500 µM vanadate caused 50% inhibition of uptake. Therate of sulfate uptake in the dark was twice that in the lightand was stimulated by low pH. These results suggest that thesulfate transport system in C. ellipsoidea is operated by protonsymport across the plasmamembrane which is partially mediatedby P-type ATPase and that these systems depend exclusively onenergy derived from oxidative phosphorylation in the mitochondria. Effects of diffusion and upwelling on the formation of red tides   总被引:4,自引:0,他引:4  
In this paper, records on the timing and location of specificred tides monitored once or twice a week in Mikawa Bay, Japan,are related to horizontal and vertical mixing rates determinedfrom a numerical model. Horizontal (Kh) and vertical (Kz) diffusioncoefficients, and upwelling velocities, were estimated usinga box model analysis. In the wind-mixed period and in the upperlayer during the stratified period, Kh was estimated to be ofthe order of 102 m2 s–1. During the stratified period,Kz was estimated to be of the order of 10–5 m2 s–1.The upwelling velocity was calculated to be in the range 0.35–5.1m day–1 with an average of 1.5 m day–1. Comparisonbetween the literature values of the specific growth rate (µ)of the red tide-forming diatoms and calculated Kh values duringthe red tides show that diatoms which have a low µ cannotform red tides in a strongly diffusive environment, while specieshaving a high µ can form red tides even in a strong diffusiveenvironment. On the other hand, no clear relationship was foundbetween µ of the flagellate group and Kh, although theflagellate group formed red tides even in severe diffusive conditions.From the comparison between the literature values of sinkingrate and swimming speed and the physical parameters associatedwith vertical processes, it was concluded that flagellates willform red tides, even in severe diffusive conditions, by usingtheir swimming ability, while diatoms form red tides by theirhigh growth rates with the aid of vertical diffusion and theupwelling movement of water.  相似文献

Controlled environment experiments were performed to determinethe effects of temperature and water potential on germination,radicle elongation and emergence of mungbean (Vigna radiata(L.) Wilczek cv. IPB-M79-17-79). The effects of a range of constant temperatures (15–45°C) and water potentials (0 to –2.2 MPa) on germinationand radicle elongation rates were studied using an osmoticumtechnique, in which seeds were held against a semi-permeablemembrane sac containing a polyethylene glycol solution. Linearrelationships were established between median germination time(Gt50) and water potential at different temperatures, and betweenreciprocal Gt50 (germination rate) and temperature at differentwater potentials. Germination occurred at potentials as lowas –2.2 MPa at favourable temperatures (30–40 °C),but was fastest at 40 °C when water was not limiting, withan estimated base temperature (Tb) of about 10 °C. Subsequentradicle elongation, however, was restricted to a slightly narrowertemperature range and was fastest at 35 °C. The conceptof thermal time was used to develop an equation to model thecombined effects of water potential and temperature on germination.Predictions made using this model were compared with the actualgermination obtained in a related series of experiments in columnsof soil. Some differences observed suggested the additionalimportance of the seed/soil/water contact zone in influencingseed germination in soil. Seedling emergence appeared to reflectfurther the radicle elongation results by occurring within anarrower range of temperatures and water potentials than germination.Emergence had an estimated Tb of 12.6 °C and was fastestat 35 °C. A soil matric potential of not less than about–0.5 MPa at sowing was required to obtain 50% or moreseedling emergence. Key words: Germination, temperature, water potential  相似文献
