Androctonus crassicauda is one of the Southeastern Anatolian scorpions of Turkey with ethno-medical and toxicological importance. Two toxic peptides (Acra1 and Acra2) were isolated and characterized from the venom of this scorpion. In this communication, the isolation of an additional toxin (Acra3) by chromatographic separations (HPLC and TSK-gel sulfopropyl) and its chemical and functional characterization is reported. Acra3 is a 7620Da molecular weight peptide, with 66 amino acid residues crosslinked by four disulfide bridges. The gene coding for this peptide was cloned and sequenced. Acra3 is anticipated to undergo post-translational modifications at the C-terminal region, having an amidated serine as last residue. Injection of Acra3 induces severe neurotoxic events in mice, such as: excitability and convulsions, leading to the death of the animals within a few minutes after injection. Electrophysiological assays conducted with pure Acra3, using cells that specifically expressed sodium channels (Nav1.1-Nav1.6) showed no clear effect. The exact molecular target of Acra3 remained undiscovered, similar to three other scorpion peptides that clustered very closely in the phylogenetic tree included here. Fluorescent derivatives of scorpion toxin V from Leiurus quinquestriatus quinquestriatus have been prepared so that the topographical, dynamic, and cellular properties of the neurotoxin receptor site on the voltage-dependent sodium channel could be studied. Four different modification strategies have been pursued in which acylated, amidinylated, thio-amidinylated, and reductively alkylated scorpion toxins were prepared. Acylation induces a loss of net positive charge on the toxin and these derivatives are purified by preparative isoelectric focusing and ion-exchange chromatography. Amidinylation and reductive alkylation preserve the protonation state of the toxin and maintain the native tertiary structure of the toxin. Because the native toxin does not contain cysteine, we have introduced new sulfhydryls through modification with the cyclic imidoester 2-iminothiolane which also preserves the net charge on the toxin. Novel purification methods with small amounts of toxin by immunoprecipitation using antibodies directed against the chromophores or through covalent thiol-disulfide exchange chromatography have been utilized. The biological activities, equilibrium binding, and spectroscopic properties indicate that these derivatives retain high affinity for the sodium channel and are as active or only 2-3 times less active than L. quinquestriatus V toxin itself. The spectroscopic properties of these fluorescent derivatives cover the absorption range from 290 to 470 nm, and fluorescence emissions range from 360 to 550 nm where suitable filters and spectral overlap with previously synthesized fluorescent tetrodotoxin can be found. The fluorescent properties in particular show excellent environmental sensitivity and are suitable for probing the molecular dynamics of the toxin receptor and for topographic mapping of the sodium channel by fluorescence resonance energy transfer measurements.  相似文献

The venom of the African scorpion Leiurus quinquestriatus was subjected to high-performance ion-exchange chromatography. Among a large number (greater than 25) of small proteins and other substances, a protein component of approx. 6500 Da was purified. The effect of this toxin was tested on single myelinated nerve fibres of the frog Rana esculenta. Toxin concentrations less than 10 nM produced clear effects. Activation rather than inactivation of the voltage-dependent sodium channel was strongly affected. Thus, this toxin from an African scorpion acts like the beta-toxins present in the venom of North American scorpions.  相似文献

Scorpion (Leiurus quinquestriatus) venom (ScV) stimulated accumulations of cyclic AMP and turnover of phosphatidylinositol in guinea pig cortical synaptoneurosomes. The concentrations of ScV that were necessary to increase cyclic [3H]AMP accumulation were lower than those required to stimulate formation of [3H]inositol phosphates from phosphatidylinositol. In the presence of 10 microM 2-chloroadenosine, ScV induced a dose-dependent synergistic accumulation of cyclic AMP with an EC50 value that was comparable to the EC50 required for stimulation of phosphatidylinositol turnover. Tetrodotoxin partially inhibited cyclic AMP accumulations elicited by ScV indicating that at least part of such responses are due to activation of voltage-dependent sodium channel. Tetrodotoxin virtually completely blocked formation of inositol phosphate stimulated by ScV. High concentrations of Mg2+ (30 mM) did not block responses to ScV indicating that release of neurotransmitters was not involved. Membrane potential changes could not be detected at concentrations of ScV that triggered the biochemical responses. Stimulation of phosphatidylinositol turnover by ScV appears to depend on an increase in influx of Na+ in synaptoneurosomes, presumably due to slowing of the inactivation of voltage-dependent sodium channels by alpha-scorpion toxin, a component of ScV. At least in part, the stimulation of cyclic AMP accumulation by ScV correlates with increases in phosphatidylinositol turnover.  相似文献

An inhibitor of apamin binding has been purified to homogeneity in three chromatographic steps from the venom of the scorpion, Leiurus quinquestriatus hebraeus. The inhibitor, which we have named leiurotoxin I, represents less than 0.02% of the venom protein. It is a 3.4-kDa peptide with little structural homology to apamin although it has some homology to other scorpion toxins such as charybdotoxin, noxiustoxin, and neurotoxin P2. Leiurotoxin I completely inhibits 125I-apamin binding to rat brain synaptosomal membranes (Ki = 75 pM). Thus, it is 10-20-fold less potent than apamin. Leiurotoxin I is not a strictly competitive inhibitor of this binding reaction. Like apamin, leiurotoxin I blocks the epinephrine-induced relaxation of guinea pig teniae coli (ED50 = 6.5 nM), while having no effect on the rate or force of contraction in guinea pig atria or rabbit portal vein preparations. Thus, leiurotoxin I of scorpion venom and apamin of honeybee venom demonstrate similar activities in a variety of tissues, yet are structurally unrelated peptides. These two peptides should be useful in elucidating the role of the small conductance, Ca2+-activated K+ channels in different tissues.  相似文献

Hydatidosis is a zoonotic disease that commonly occurs in several places around the world, especially in the Middle East, due to infection by the larval stage of Echinococcus granulosus. This disease impacts an immense effect on the economic and public health of both humans and animals. Despite their effectiveness, the unacceptable side effects and progressive resistance to scolicidal agents may limit their use. According to their biopharmaceutical activity and benefits, numerous studies have reported that scorpion venom and its derivatives represent important resources for therapeutic applications. Therefore, this study was designed to investigate the in vitro scolicidal consequences of the crude venom of Androctonus crassicauda on E. granulosus. For this purpose, protoscolices from infected organs of camel containing hydatid cysts were collected, separated, and washed. The scolicidal impacts of three different concentrations of the crude venom (20, 50, and 100 µg/mL) were tested at different times of exposure (30, 60, 120, and 240 min). Particularly, eosin exclusion test was used to examine the viability of the protoscolices. The study results showed that the crude venom at 100 μg/mL destroys all protoscolices after 240 min incubation. Also, the scolicidal activity of venom increased significantly according to the time of exposure. In conclusion, the crude venom of A. crassicauda demonstrated high scolicidal activity in vitro against protoscolices of hydatid cysts in low concentration and short exposure time. However, the efficacy of scorpion venom remains to be evaluated in vivo for the treatment of hydatidosis in both humans and domesticated animals.  相似文献

The hemocyanins of the scorpions Leiurus quinquestriatus and Androctonus australis, the tarantula Eurypelma californicum (all 24-mers), and the lycosid spider Cupiennius salei (dodecamer) were dissociated into subunits, the subunits isolated and studied by two-dimensional immunoelectrophoresis for interspecific cross-reactivities. Androctonus hemocyanin yielded a pattern of 8 subunit types in agreement with data from Lamy et al. (1979, Arch. Biochem. Biophys. 193, 140-149). Leiurus hemocyanin is also composed of 8 immunologically distinct subunits which could be assigned to the pattern of Androctonus in a subunit-to-subunit correlation. The subunit designations 1 to 6 of Lamy et al. could be adopted for both scorpion hemocyanins; however, in the present communication, Lamy's subunits 3A/3B are designated as 3'/3", because we could not unequivocally decide if 3' = 3A and 3" = 3B or vice versa. The 7 subunit types a to g of Eurypelma hemocyanin could be correlated with the scorpion hemocyanin subunits as follows: a = 3', b = 5B, c = 3C, d = 5A, e = 6, f = 2, g = 4. Additional cross-reactivities were detected between e/4, and f/5A, respectively. No subunit of Eurypelma hemocyanin is homologous to scorpion 3", which could not be precipitated by anti-Eurypelma antiserum. Antiserum against Cupiennius hemocyanin precipitated subunit f of Eurypelma and subunits 2 and 5A of scorpion hemocyanin. The published models of quaternary structure and a possible subunit phylogeny of arachnidan hemocyanins are discussed in view of the present results.  相似文献

Lumpy skin disease caused by a capripoxvirus was observed in a captive-bred female Arabian oryx (Oryx leucoryx) at the National Wildlife Research Center, Taif, Saudi Arabia. Clinical signs included severe general depression with fever, anorexia, greater than 1,000 nodular cutaneous lesions and gradual recovery over 2 mo. The virus was found by electron microscopy and paired sera showed an increasing virus neutralization antibody titer against capripoxvirus. A serologic survey of the herd of 90 oryx showed a low prevalence (2%) of this infection. This report describes the first case of lumpy skin disease in an Arabian oryx.  相似文献

Two novel antimicrobial and cytotoxic triterpenoids, isopseudolarifuroic acids A (1) and B (2), were isolated from the bark of Pseudolarix kaempferi. The structural elucidation of two novel compounds was carried out mainly by spectroscopic methods, and also by computer modeling. Compounds 1 and 2 exhibited significant cytotoxic activities against several tumor cell lines. Compound 1 also showed most potent antimicrobial activities against both Gram-positive and Gram-negative bacteria.  相似文献

Reproductive data on captive Arabian oryx (Oryx leucoryx) were collected from June 1986 through April 1992 at the National Wildlife Research Center (Taif, Saudi Arabia). Oryx females are polyestrous. The estrous cycle averaged 22 days and mean gestation length was 260 ± 5.5 days (S.D). Sex ratio at birth was unbiased and mean weight was 6.5 ± 0.7 kg (S.D.), with no difference between sexes. Under captive breeding conditions, births occurred throughout the year. Females gave birth to a single calf at any time during the day and produced 1.03 young per year. Abortion rate was 3.6%. Mortality rate of young was 6.1% before weaning at 3 months of age. The interbirth interval averaged 295 ± 42 days (S.D.), with 53% lasting between 270 and 279 days. The circadian rhythms of locomotor activity of the scorpion Leiurus quinqueslriatus were examined under different light-dark cycles and in free-running conditions. The circadian rhythm is bimodal in LD 12:12 with alternating cycles of temperature (35°-25°C) with high intensity (1300 lux) or in LD 12: 12 with constant temperature 35° C with 300 lux. In LD 12:12 (1300 lux), in long or in short light spans with constant temperature, the bimodal pattern is slightly changed with the appearance of a third minor peak of activity. In free-running conditions, the bimodal rhythm of locomotor activity persists in DD with T about 24 hr, but in LL the rhythm becomes unimodal with T about 24 hr. Cosinor and power spectrum analysis showed the presence of more than one periodic component. It seems that there is a correlation between the range of light regimens, temperature, light intensity and the coincidence of these components. These components are independently entrained by the environmental light cycle. The mechanism of entrainment of components is discussed.  相似文献

The non disulphide-bridged peptides (NDBPs) of scorpion venoms are attracting increased interest due to their structural heterogeneity and broad spectrum of biological activities. Here, two novel peptides, named AcrAP1 and AcrAP2, have been identified in the lyophilised venom of the Arabian scorpion, Androctonus crassicauda, through “shotgun” molecular cloning of their biosynthetic precursor-encoding cDNAs. The respective mature peptides, predicted from these cloned cDNAs, were subsequently isolated from the same venom sample using reverse phase HPLC and their identities were confirmed by use of mass spectrometric techniques. Both were found to belong to a family of highly-conserved scorpion venom antimicrobial peptides - a finding confirmed through the biological investigation of synthetic replicates. Analogues of both peptides designed for enhanced cationicity, displayed enhanced potency and spectra of antimicrobial activity but, unlike the native peptides, these also displayed potent growth modulation effects on a range of human cancer cell lines. Thus natural peptide templates from venom peptidomes can provide the basis for rational analogue design to improve both biological potency and spectrum of action. The diversity of such templates from such natural sources undoubtedly provides the pharmaceutical industry with unique lead compounds for drug discovery.  相似文献

Two new species of Heliotropium L. (Boraginaceae, subfam. Heliotropioideae), growing in sandy loam soils in Jizan, south-west Saudi Arabia, Heliotropium chaudharyanum Al-Turki, Omer & A. Ghafoor and Heliotropium jizanense Al-Turki, Omer & A. Ghafoor, are described and illustrated. The relationships of the new species within the small-flowered Heliotropium europaeum L. species group are discussed. Notes on distribution and habitat are provided, together with a map.  相似文献

The organic solvent extract of Kumazasa leaves (Sasa albo-marginata) showed antimicrobial activity against bacteria, fungi and yeast. Kumazasa at a concentration of 0.2-1.0% showed stronger antimicrobial activity than potassium sorbate or sodium benzoate at the same concentration. Both acidic and phenolic fractions of the extract showed strong antimicrobial activity. Thirty acidic and phenolic compounds were identified by GC and GC-MS analysis. Acetic, propionic, benzoic, phenylacetic, salicylic, 3-hydroxybenzoic and o-anisic acids, and guaiacol, phenol, 4-ethylphenol, xylenol and 4-vinylphenol were the main components. It was estimated that these components play an important role in the formation of the antimicrobial activity of Kumazasa extract.  相似文献

Fifteen fecal samples from peacocks (Pavo cristatus) in Saudi Arabia contained oocysts of Eimeria riyadhae n. sp. in two peacocks and oocysts of E. arabica n. sp. in one peacock. Sporulated oocysts of Eimeria riyadhae are ellipsoidal, 27-30.5 x 20.5-25 (28.8 +/- 1.3 x 22.4 +/- 1.6) micron, with a two-layered wall and bilobed polar body, but without a micropyle or residuum. The sporocysts are ovoid, 11-14.5 x 6.5-8 (13.2 +/- 1.2 x 7.2 +/- 0.6) micron with a thick, knob-like Stieda body and a residuum. Sporulated oocysts of Eimeria arabica are spheroidal, 17.5-21.5 x 17.5-21.5 (19.2 +/- 1.6 x 19.2 +/- 1.6) micron, with a two-layered wall and two refractile polar bodies, but without a micropyle or residuum. The sporocyts are elongate ovoid, 9.5-12 x 4-6.5 (11.2 +/- 0.9 x 5.5 +/- 0.88), with a small crescent-shaped Stieda body. The host bird belongs to the order Galliformis.  相似文献
