Chlorpyrifos is a widely applied insecticide that permeates on most waterways and affects aquatic organisms. The growth performances, hematological and histological impacts on Nile tilapia, Oreochromis niloticus following a 60 day of exposure to varying concentrations of chlorpyrifos 20 EC (T1 08 µgL?1, T2 16 µgL?1, and T3 32 µgL?1) were compared to a control Tc 0 µgL?1. The 96-hour LC50 of chlorpyrifos 20 EC was calculated as 46.80 μgL?1. The water quality parameters were recorded regularly. The value of dissolved O2 and NH3 stayed rather steady, although temperature varied considerably. It was revealed that as chlorpyrifos levels go up, the percentage of weight gain (WG %), specific growth rates (SGR), and survival rate decreases. The control group Tc had the highest percentages of SGR weight (1.16 ± 0.58) and the T3 group had the lowest percentages of SGR weight (0.25 ± 0.77). The hematological assessment showed significant differences of hemoglobin concentration, white blood cell counts and red blood cell numbers between chlorpyrifos treatment and control group (P < 0.05). Histological alterations in the liver, gills, and muscle tissues reported to be worse for T3 as compared to others. There were no statistical differences in GSI, HSI values between control and treatment groups. The chlorpyrifos 20 EC was shown to be highly toxic to O. niloticus at sub-lethal dosages.  相似文献

Investigating the determinants of the reproductive biology of fishes is an essential component of fisheries research. Tilapia breeding patterns were investigated to determine the impact of non‐native Oreochromis niloticus on the native congeneric Oreochromis macrochir in the upper Kabompo River in the Northwest of Zambia using the gonadosomatic index and the sex ratios. Oreochromis niloticus was the most abundant fish caught (221, 63.5%) than Omacrochir (127, 36.5%). Results showed that the overall gonadosomatic index means of Omacrochir in both sections were similar. Oreochromis macrochir bred in December and February–March, with no reproduction in June. However, Oniloticus in the invaded section indicated all year reproduction through reduced spawning in May–June, with increased spawning activity in February–March. The sex ratio (females: males) was 1:1.3 and 1:1.7 for Oniloticus and Omacrochir, respectively, and both significantly deviated from the sex ratio of 1:1 (ꭓ2 = 8.42 and 9.37, p < .05). Our study has revealed that Oniloticus was able to spawn across all sampled months with a 23% higher breeding population than Omacrochir, which might explain the suppression in the abundance of native Omacrochir. Due to the superior breeding patterns of Oniloticus, fisheries, wildlife, and aquaculture practitioners need to make contingency plans to alleviate its impacts further downstream of the Kabompo River.  相似文献

This work aims to evaluate the antibacterial activity of biological zinc nanoparticles (BIO-ZnONPs) against pathogenic fish bacteria and assess the effect of BIO-ZnONPs on the performance, behavior, and immune response in Nile tilapia (Oreochromis niloticus) as compared to chemical zinc nanoparticles (CH- ZnONPs). Aspergillus niger TS16 fabricated the BIO-ZnONPs were spherical shape with the average size of 45 nm and net charge of ?27.23 mV. Generally, the results indicate that BIO-ZnONPs were more effective than CH- ZnONPs in enhancing the performance properties of Nile tilapia. Five experimental groups of Nile tilapia (initial body weight of 20.2 g) were treated with two concentrations of 0.5 and 1 mg L?1 from biological and chemical ZnONPs, while the fifth group was served as a control. After ten weeks of treated water with ZnONPs, the performance, feed efficiency parameters, feeding, and swimming behaviors significantly improved in BIO-ZnONPs treated groups (P < 0.05). The liver function, LYZ activity, and NBT values were significantly enhanced in the 0.5 mg L?1 BIO-ZnONPS group compared to CH- ZnONPs group and control (P < 0.05). Furthermore, the lowest cortisol and the highest testosterone and growth hormone levels were recorded in 1 mg L?1 BIO-ZnONPs group. Regarding the antibacterial effects, BIO-ZnONPs displayed the lower total bacterial loads in water and fish tissues (intestine, gills, skin, and muscle) and the maximum antibacterial properties against pathogenic bacteria (Listeria monocytogenes, Bacillus cereus, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, and Aeromonas hydrophila). Our study exemplifies novel findings of BIO-ZnONPs in the promotion of fish health and production and its antibacterial properties in Nile tilapia.  相似文献

This study was carried out to determine the median lethal concentrations (LC50) of Zinc nanoparticles (ZnNPs) on Oreochromis niloticus and Tilapia zillii. The biochemical and molecular potential effects of ZnNPs (500 and 2000 μg L−1) on the antioxidant system in the brain tissue of O. niloticus and T. zillii were investigated. Four hundred fish were used for acute and sub-acute studies. ZnNP LC50 concentrations were investigated in O. niloticus and T. zillii. The effect of 500 and 2000 μg L−1 ZnNPs on brain antioxidants of O. niloticus and T. zillii was investigated. The result indicated that 69 h LC50 was 5.5 ± 0.6 and 5.6 ± 0.4 for O. nilotica and T. zillii, respectively. Fish exposed to 500 μg L−1 ZnNPs showed a significant increase in reduced glutathione (GSH), total glutathione (tGSH) levels, superoxide dismutase (SOD), catalase (CAT), glutathione reductase (GR), glutathione peroxidase (GPx) and glutathione-S-transferase (GST) activity and gene expression. On the contrary, malondialdehyde (MDA) levels significantly decreased. Meanwhile, fish exposed to 2000 μg L−1 ZnNPs showed a significant decrease of GSH, tGSH levels, SOD, CAT, GR, GPx and GST activity and gene expression. On the contrary, MDA levels significantly increased. It was concluded that, the 96 h LC50 of ZnNPs was 5.5 ± 0.6 and 5.6 ± 0.4 for O. nilotica and T. zillii, respectively. ZnNPs in exposure concentrations of 2000 μg/L induced a deleterious effect on the brain antioxidant system of O. nilotica and T. zillii. In contrast, ZnNPs in exposure concentrations of 500 μg L−1 produced an inductive effect on the brain antioxidant system of O. nilotica and T. zillii.  相似文献

We here propose a novel Raman spectroscopy method that permits the noninvasive measurement of blood glucose concentration. To reduce the effects of the strong background signals produced by surrounding tissue and to obtain the fingerprint Raman lines formed by blood analytes, a laser was focused on the blood in vessels in the skin. The Raman spectra were collected transcutaneously. Characteristic peaks of glucose (1125 cm-1) and hemoglobin (1549 cm-1) were observed. Hemoglobin concentration served as an internal standard, and the ratio of the peaks that appeared at 1125 cm-1 and 1549 cm-1 peaks was used to calculate the concentration of blood glucose. We studied three mouse subjects whose blood glucose levels became elevated over a period of 2 hours using a glucose test assay. During the test, 25 Raman spectra were collected transcutaneously and glucose reference values were provided by a blood glucose meter. Results clearly showed the relationship between Raman intensity and concentration. The release curves were approximately linear with a correlation coefficient of 0.91. This noninvasive methodology may be useful for the study of blood glucose in vivo.  相似文献

In this study we investigated the acute toxicity of Zinc Nano Particles (ZnO NPs) and bulk ZnO on tilapia fish (Oreochromis mossambicus). Oreochromis mossambicus was exposed to the different concentration of ZnO NPs, ZnO and mixed solution of both ZnO NPs and ZnO (20 ppb, 20 ppb, 20 ppb) respectively for 96 h. A very high impact was recorded in hematological parameters which shows significant increased (p < 0.05) in count of white WBCs and platelets in all the experimental groups compared tocontrol group. The count of RBCs, Hb, hematocrit and MCHC were significantly decreased. The remarkable changes which were recorded during this study were histopathological lesions in the gills of exposed fish including, disorganization of gill lamella, cartilaginous core disruption, lifting of epithelium, loss of secondary gill lamellae, blood congestion, fusion of secondary gills lamellae, shortening of secondary gills lamellae, atrophy and curling. Disassembly were seen in plasma membrane of liver along with blood congestion, pyknosis, necrosis, hyperplasia and formation of vacuoles. Intestinal alterations which were observed include shortening of villi, necrosis, detachment and fusion of villi and extreme goblet cells formation. It is concluded from the present study that high level of ZnO NPs, ZnO and mixed solution has a strong tendency to alter hematological parameters, histological architecture, therefore, the indiscriminate use of ZnO NPs and ZnO can subsidize in reducing the population of Oreochromis mossambicus in natural water bodies.  相似文献

The occurrence of Nα-acetylhistidine (NAH) in skeletal muscle of 91 species of freshwater fish and 9 species of other ectothermic vertebrates was investigated, with consideration of phylogenetic relationships. Of the 91 freshwater fish species examined, 13 species (7 cichlids, 5 anabantids, and 1 catfish) contained considerable amounts (> 1 µmol/g) of NAH in their skeletal muscles. The highest level (10.37 µmol/g) of NAH was found in the tissue of Betta splendens (Siamese fighting fish). Moreover, the NAH contents in the tissues of Trichogaster trichopterus (three spot gourami), Kryptopterus bicirrhis (glass catfish), Oreochromis niloticus (Nile tilapia), Mikrogeophagus ramirezi (ram cichlid) and Parachromis managuensis (Guapote tigre) were 3.17–6.16 µmol/g. The skeletal muscle of amphibians (5 species) and reptiles (4 species) had a low level (< 0.25 µmol/g) of NAH. The present findings clearly demonstrate NAH as the fifth imidazole-related compound, in addition to histidine, carnosine, anserine and ophidine (balenine), recognized as a major non-protein nitrogenous constituent in the skeletal muscle of vertebrate animals.  相似文献

Microplastic particles have increasingly been detected in aquatic biota, from zooplankton to fish, raising concern for potential effects on aquatic organisms. In addition, they may potentially influence the toxicity of other contaminants in the marine environment. The aim of this study was to clarify whether polyethylene microspheres (1–5 μm) modulate short-term toxicity of the polycyclic aromatic hydrocarbon pyrene to juveniles (0+ group) of the common goby (Pomatoschistus microps). Fish were exposed for 96 h to pyrene (20 and 200 μg L−1) in the absence and presence of microplastics (0, 18.4 and 184 μg L−1). Mortality, bile pyrene metabolites, and biomarkers involved in neurotransmission, aerobic energy production, biotransformation and oxidative stress were quantified. Microplastics delayed pyrene-induced fish mortality and increased the concentration of bile pyrene metabolites. Microplastics, alone or in combination with pyrene, significantly reduced acetylcholinesterase (AChE) activity, an effect also observed for pyrene alone. The mixture also decreased isocitrate dehydrogenase (IDH) activity. No significant effects were found for glutathione S-transferase activity or lipid peroxidation. Overall, results show that: (i) microplastics modulate either the bioavailability or biotransformation of pyrene; (ii) simultaneous exposure to microplastics and pyrene decrease the energy available through the aerobic pathway of energy production; and (iii) microplastics inhibit AChE activity. The mechanism for AChE inhibition appeared to be different for pyrene and microplastics, since simultaneous exposure to both did not increase significantly the inhibitory effect. The observed neurotoxic effects of microplastics per se and the effects on IDH activity of the two stressors combined are of concern because they may increase mortality in natural fish populations. More studies need to be carried out on possible combined effects of microplastics and polycyclic aromatic hydrocarbons on fish, particularly juveniles.  相似文献

Cichlid fishes have played an important role in evolutionary biology and aquaculture industry. Nile tilapia (Oreochromis niloticus), blue tilapia (Oreochromis aureus) and Mozambique tilapia (Oreochromis mossambicus), the useful models in studying evolutionary biology within Cichlid fishes, are also mainly cultured species in aquaculture with great economic importance. In this paper, the complete nucleotide sequence of the mitochondrial genome for O. niloticus and O. aureus were determined and phylogenetic analyses from mitochondrial protein-coding genes were conducted to explore their phylogenetic relationship within Cichlids. The mitogenome is 16,625 bp for O. niloticus and 16,628 bp for O. aureus, containing the same gene order and an identical number of genes or regions with the other Cichlid fishes, including 13 protein-coding genes, two rRNA genes, 22 tRNA genes and one putative control region. Phylogenetic analyses using three different computational algorithms (maximum parsimony, maximum likelihood and Bayesian method) show O. niloticus and O. mossambicus are closely related, and O. aureus has remotely phylogenetic relationship from above two fishes.  相似文献

To measure oxygen saturation (SO2) of flowing erythrocytes in blood vessels of living animals, our “in vivo cryotechnique” (IVCT) was combined with confocal Raman microscopy at low temperature (−150 °C), referred to as cryomicroscopy. We evaluated two resonance Raman (RR) shifts around 1355 and 1378 cm−1, reflecting de-oxygenated and oxygenated hemoglobin molecular structures, respectively. Judging from the calibration analyses of quickly frozen human whole blood for the control experiment in vitro, the two RR shifts were well retained at the low temperature, and their calculated ratios mostly reflected the relative SO2 measured with a blood-gas analyzer. In blood vessels of living mouse organs prepared with the IVCT, their RR spectral peaks were also detected at the same RR shifts obtained in human blood. In the blood vessels of living mouse small intestines, some arterioles and venules were clearly distinguishable by monitoring different peak patterns of their RR shifts. The different ratios of the RR shift-areas were calculated even in the arterial vessels. In blood vessels of mouse livers, the Raman spectra showed a lower peak shift of 1378 cm−1 compared to that of 1355 cm−1, indicating an SO2 decrease in hepatic blood circulation. Thus, the new cryopreparation technique will enable us to directly analyze the in vivo SO2 in various tissues of a whole animal body prepared with the IVCT, reflecting their living states.  相似文献

Based on central composite rotatable experimental design and response surface method, the interacting effects of temperature (20 °C–34 °C) and dietary protein level (25%–50%) on hematological parameters including red blood cell (RBC), white blood cell (WBC) and hemoglobin (Hb) of juvenile Oreochromis niloticus were studied under laboratory conditions. The experiment lasted for 7 weeks. After the feeding trial, fish were challenged with Streptococcus iniae and mortality was recorded for within 8 days. Results showed that the linear and quadratic effects of temperature on RBC, WBC and Hb were highly significant (P < 0.01). When the dietary protein level was 25%–50%, the RBC, WBC and Hb were increased firstly and then decreased, but the linear and quadratic effects of protein level were insignificant (P > 0.05). The interacting effects of temperature and protein level on RBC and Hb were significant (P < 0.05). The regression equations of RBC, WBC and Hb toward the two factors of interest were established, with the coefficients of determination being 0.870, 0.836 and 0.881, respectively (P < 0.01). These equations could be used for prediction in practice. After the challenge, the mortalities for the combinations of 22.1 °C/28.7% and 20.0 °C/37.5% were significantly higher than 27.0 °C/37.5% (P < 0.05). The optimal temperature/dietary protein level combination was obtained at 27.9 °C/38.1% at which the lowest mortality (13.76%) was attained. This value was close to the optimal temperature/dietary protein level combination (29.4 °C/41.9%) for the greatest levels of RBC (2.560 × 106 μL?1), WBC (270.648 × 103 μL?1) and Hb (92.851 g L?1). The results of this study indicated that preferred temperature/dietary protein level combination might strengthen the non-specific immunity and reduce susceptibility to S. iniae.  相似文献

This study investigated the in vitro antioxidant, proapoptotic and anti-proliferative activity of phycocyanin extracted from Ulva lactuca (Chlorophyta) algae extract loaded on albumin nanoparticle (ULANP). The characterization of ULANP profile was done by using FTIR and its cytotoxicity was investigated by using MTT assay against HepG2 and MCF7 cell lines. The proapoptotic markers caspase 8 & 9 were measured. Analysis of ULANP by FTIR showed the characteristic band (2100 cm−1 ~3700 cm−1) that is indicated primarily by COO, CO and conjugated double bond. These bonds showed the spectral band at peaks of 2985 cm−1 and 2860 cm−1, 2986 cm−1 respectively. The antioxidant potential and radical scavenging property of ULANP was also appreciable as compared to the vitamin C and gallic acid. The antiproliferative assay carried out by WST-1 suggests that ULANP was effective against both HepG2 (93.17%) and MCF7 (91.3%). Caspase-8 and −9 were significantly elevated (p < 0.001) in both the cell lines of breast and liver cancer. It was concluded that ULANP induced anti-proliferative and pro-apoptotic activities on liver and breast cancer. It is promising as a novel antitumor activity for further investigation the mechanistic pathways mediated this action.  相似文献

Objective

The purpose of this study was to comparatively analyze the signature Raman spectra of genomic DNA, nuclei, and tissue of normal gastric mucosa and gastric cancer and to investigate the biochemical transformation of molecules associated with gastric mucosa malignancy.

Method

Genomic DNA, nuclei, and tissue from normal gastric mucosa and gastric cancer were analyzed by Raman spectroscopy.

Results

1) The Raman spectrum of gastric cancer genomic DNA showed that two peaks appeared, one at approximately 1090 cm-1 with a higher intensity than the peak at 1050 cm-1 in the spectrum. Characteristic peaks appeared at 950 cm-1, 1010 cm-1, and 1100-1600 cm-1. 2) Using a hematoxylin and eosin (H&E)-stained section, the intensity of the characteristic peak of nucleic acids at 1085 cm-1 was increased and shifted to 1088 cm-1 in cancer cells. The relative intensity of the characteristic peaks of nucleoproteins at 755 cm-1 and 1607 cm-1 was significantly increased in cancer cells compared with normal cells. 3) Compared with normal tissues, the peak representing PO2- symmetric stretching vibration shifted from 1088 cm-1 to 1083 cm-1 in cancer tissue, and the characteristic peak for collagen at 938 cm-1 shifted to 944 cm-1. In addition, an extra characteristic peak indicating C = C stretching vibration appeared at 1379 cm-1 in the lipid spectrum in cancer tissue.

Conclusions

The position, intensity, and shape of peaks in the Raman spectra of DNA, nuclei, and tissue from gastric cancer were significantly different compared with those of normal cells. These results indicate that the DNA phosphate backbone becomes unstable in cancer cells and might be broken; the relative content of histones is increased and stable; the relative collagen content is reduced, facilitating cancer cell metastasis; and the relative content of unsaturated fatty acids is increased, increasing the mobility of the plasma membrane of cancer cells.  相似文献

Exposures to particulate matter with a diameter of 2.5 μm or less (PM2.5) may influence the risk of birth defects and make you allergic, which causes serious harm to human health. Bamboo charcoal can adsorb harmful substances,that was of benefitto people’s health. In order to figure out the optimal adsorbtion condition and the intrinsic change of bamboo charcoal, five chemicals were adsorbed by bamboo charcoal and were analyzed by FT-IR. The optimal blast time was 80 min of Na2SO3, 100 min of Na2S2O8, 20 min of Na2SO4, 120 min of Fe2(SO4)3 and 60 min or 100 min of S. FT-IR spectra showed that bamboo charcoal had five characteristic peaks of SS stretch, H2O stretch, OH stretch, CO stretch or CC stretch, and NO2 stretch at 3850 cm−1, 3740 cm−1, 3430 cm−1, 1630 cm−1 and 1530 cm−1, respectively. For Na2SO3, the peaks at 3850 cm−1, 3740 cm−1, 3430 cm−1, 1630 cm−1 and 1530 cm−1 achieved the maximum at 20 min. For Na2S2O8, the peaks at 3850 cm−1, 3740 cm−1, 3430 cm−1 and 1530 cm−1 achieved the maximum at 40 min. For Na2SO4, the peaks at 3850 cm−1, 3740 cm−1 and 1530 cm−1 achieved the maximum at 40 min. For Fe2(SO4)3, the peaks at 3850 cm−1, 3740 cm−1, 1630 cm−1 and 1530 cm−1 achieved the maximum at 120 min. For S, the peaks at 3850 cm−1 and 3740 cm−1 achieved the maximum at 40 min, the peaks at 1630 cm−1 and 1530 cm−1 achieved the maximum at 40 min. It proved that bamboo charcoal could remove sulfur powder from air to restrain sulfur allergies.  相似文献

The concentrations of Fe, Mn, Ni, Pb and V in water, sediment and the gill, liver and muscle tissues of Synodontis resupinatus, Heterotis niloticus and Clarias gariepinus, all commercially important fish species of the lower Niger River, were investigated in 2015. Water, sediment and fish samples were collected for six months and heavy metals were determined using an Atomic Absorption Spectrometer. Fe ranked highest in water and sediment, with concentrations of 2.74 mg l?1 and 61.60 mg kg?1, respectively. Metals followed the magnitude of Fe > Mn > Ni > V > Pb in the water and Fe > Mn > V > Ni > Pb in the sediments. Metal concentrations were higher in the tissues of S. resupinatus compared with H. niloticus and C. gariepinus. Fe was also highest in the gills, liver and muscle of the three fish species. Its highest concentration of 132.97 mg kg?1 dry weight was recorded in the gills of S. resupinatus. Bioconcentration factors of metals ranged from 8.79 for Mn in H. niloticus muscle to 67.99 for Ni in S. resupinatus gills. The fish species studied pose no health risk for all metals studied, because the target hazard quotient was less than 1 and the estimated daily intakes of the metals were below the reference doses.  相似文献

Botryococcus braunii, B race is a unique green microalga that produces large amounts of liquid hydrocarbons known as botryococcenes that can be used as a fuel for internal combustion engines. The simplest botryococcene (C30) is metabolized by methylation to give intermediates of C31, C32, C33, and C34, with C34 being the predominant botryococcene in some strains. In the present work we have used Raman spectroscopy to characterize the structure of botryococcenes in an attempt to identify and localize botryococcenes within B. braunii cells. The spectral region from 1600–1700 cm−1 showed ν(C=C) stretching bands specific for botryococcenes. Distinct botryococcene Raman bands at 1640 and 1647 cm−1 were assigned to the stretching of the C=C bond in the botryococcene branch and the exomethylene C=C bonds produced by the methylations, respectively. A Raman band at 1670 cm−1 was assigned to the backbone C=C bond stretching. Density function theory calculations were used to determine the Raman spectra of all botryococcenes to compare computed theoretical values with those observed. The analysis showed that the ν(C=C) stretching bands at 1647 and 1670 cm−1 are actually composed of several closely spaced bands arising from the six individual C=C bonds in the molecule. We also used confocal Raman microspectroscopy to map the presence and location of methylated botryococcenes within a colony of B. braunii cells based on the methylation-specific 1647 cm−1 botryococcene Raman shift.  相似文献

The acute toxicity of household detergent (Ariel) on blood parameters and histology of Oreochromis niloticus was investigated using static bioassay for 96 h. Linear alkylbenzene sulfonate (LAS) is an anionic surfactant widely used in detergents and cleaners, both in industrial and household applications. LAS contaminating aquatic ecosystems as a potential toxic pollutant, was investigated in the present study for acute toxicity. The fish samples were divided into six groups, including 20 fish in each group. Normal feed was given to control group without detergents treatment. Hematological parameters (RBC count, Hb, Ht and platelets) were significantly declined, while WBC count showed a highly significant increase. Compared with the control group, significant elevation of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) was recorded in fish treated with different concentrations of detergent. Catalase (CAT), Superoxide dismutase (SOD) activities and Reduced Glutathione (GSH) concentration showed a highly significant reduction. Total proteins showed significant decrease, while total lipids, cholesterol and triglycerides significantly increased. The mean lethal concentration (LC50) for 96 h of Ariel was at concentration 10 mg/L. Relative percentage of detergent residues in fish muscles was increased with higher detergent concentrations. In conclusion, exposure to detergents resulted in great alterations in the histological structure of liver and gills.  相似文献

For diet reconstruction studies using stable isotopes, accurate estimates of trophic shift (Δδtrophic) are necessary to get reliable results. Several factors have been identified which affect the trophic shift. The goal of the present experiment was to test whether measurements of the activities of enzymes could improve the accuracy of estimation of trophic shift in fish. Forty-eight Nile tilapia (Oreochromis niloticus) were fed under controlled conditions with two diets differing in their protein content (21 and 41%) each at four different levels (4, 8, 12 and 16 g kg? 0.8 d? 1). At the end of the feeding experiment, proximate composition, whole body δ13C and δ15N as well as the activities of enzymes involved in anabolism and catabolism were measured. Step-wise regression specified contributing variables for Δδ15N (malic enzyme, aspartate aminotransferase and protein content) and Δδ13Clipid-free material (aspartate aminotransferase and protein content). Explained variation by using the significant main effects was about 70% for Δδ15N and Δδ13Clipid-free material, respectively.
