Four healthy Holstein heifers (235 ± 12 kg) fitted with duodenal and pancreatic cannulas were used to investigate infusion of isoleucine (Ile) on the pancreatic exocrine function in a 4 × 4 Latin square design. Three doses of Ile, 10, 20 and 30 g in 2500 ml water, respectively, were infused into the duodenum over a period of 12 h in Experiment (Exp) 1 and over 10 d in Exp 2. Hourly pancreatic juice and jugular blood were taken during the infusion period in Exp 1, and the blood samples were taken in 2-h intervals over the last 2 d in Exp 2. Compared with no Ile infusion, the Ile infusions in both experiments increased the concentration and secretion rate of the protein, activity of ɑ-amylase and trypsin and plasma cholecystokinin. The secretion rate of ɑ-amylase and the activity of trypsin linearly increased with the Ile doses. The pancreatic juice secretion linearly increased with Ile in Exp 2 but not in Exp 1. Isoleucine linearly increased plasma insulin in Exp 1, but not in Exp 2. No effects of Ile on pH of pancreatic juice, the activity of chymotrypsin and lipase and plasma glucose were found. In conclusion, duodenal Ile infusion could increase the pancreatic exocrine function of Holstein heifers, especially ɑ-amylase, and the increment appeared to be dose and time dependent.  相似文献

Abstract

Allelopathic potential of Thymus pulegioides L. chemical polymorphism was investigated under natural and laboratory conditions. A field analysis of 127 natural habitats hosting chemotypes of T. pulegioides with different ratios of phenolics, geraniol, and ɑ-terpinyl acetate was conducted. Effects of chemotypes, and their main compounds on seed germination and radicle growth of Trifolium pratense L. and Poa pratensis L. were conducted under laboratory conditions. Field analysis showed that Poa species were more plentiful in comparison with Trifolium species, independent of the chemotypical composition of T. pulegioides habitats. Laboratory tests with plant-acceptors showed a stronger inhibitory effect of essential oils on the germination and radicle growth of P. pratensis but in some instances germination was stimulated. Dissimilar effects were observed for the same allelochemical through air and water on the same plant-acceptor. Significantly, different effects of essential oils on radicle growth occurred in T. pratense and P. pratensis: with sensitivity to the phenolic chemotype via air and the ɑ-terpinyl acetate chemotype through water. This demonstrates that chemical polymorphism can expand communication opportunities of T. pulegioides with associated plant species. Combining investigations in natural habitats with laboratory experiments can help understand the effect of chemical polymorphism on plant-plant ecological interactions.  相似文献

西双版纳小耳猪群体遗传结构的RFLP分析   总被引:5,自引:3,他引:5  
目的 对西双版纳小耳猪的群体遗传结构进行RFLP分析。方法 采用ECL核酸直接标记和检测系统,以HRP标记ɑ-珠蛋白-3’HVR多基因座小卫星探针,对西双版纳小耳猪的Hinf Ⅰ或HaeⅢ酶切片段进行Southern杂交,以化学发光法进行检测,获得了清晰可辨的、具有高度多态性的DNA指纹图谱。结果 2 kb以上的谱带数在6~15条之间,HinfⅠ酶切产生的图带数低于HaeⅢ。JB亚系内805、807家系和JS亚系内111、121、121、151家系不同个体间的相似系数分别为0.94±0.09、0.85±0.08、0.84±1.7、0.78±1.6、0.83±0.42、0.87±0.07,显著高于各家系间的相似系数,JB亚系内的805家系和JS亚系内的151家系不同个体间相似系数较大,分别为0.94和0.87。结论 西双版纳小耳猪近交程度较高,个体间差异较小,均一性较强。  相似文献

Multidomain proteins are the product of evolutionary selection for diversity of function through concatenation and repurposing of existing modular units of structures. In structures of proteins with multiple domains, components are often globular units stitched together with flexible linkers. Multidomain proteins often fold as multiple distinct order–disorder transitions. However, the relationship between structure and folding is not always straightforward. Tropomyosin binds to actin in muscle and cytoskeletal filaments. The structure is that of a continuous ɑ-helix lacking domain boundaries, but unfolding shows distinct transitions suggesting at least three possible domains do exist. To explore how domains might occur in a continuous structure, we used Lifson-Roig helix-coil models with sequence domains of varying helical nucleation propensities. Of these models, ones with a central folding insulator, separating folding of N- and C-terminal domains, are most consistent with experimental folding studies. The positions of domain boundaries are identified by hydrogen–deuterium exchange mass spectrometry. The presence of structurally cryptic folding domains in tropomyosin could relate to its evolution and explain the uneven distribution of deleterious mutations that lead to various cardiomyopathies.  相似文献

Optimum conditions for glucose syrups production from white sorghum were studied through sequential liquefaction and saccharification processes. In the liquefaction process, a maximum dextrose equivalent (DE) of 10.98 % was achieved using 30 % (w/v) of starch and Termamyl ɑ-amylase from Bacillus licheniformis. Saccharification was performed by free and immobilized amyloglucosidase from Rhizopus mold at 1 % (w/v). DE values of 88.32 % and 79.95 % were obtained from 30 % (w/v) of starch with, respectively, free and immobilized enzyme. The immobilized Amyloglucosidase in calcium alginate beads showed reusable capacity for up to 6 cycles with 46 % of the original activity retained. The kinetic behaviour of immobilized and free enzyme gives Km value of 22.13 and 16.55 mg mL−1 and Vmax of 0.69 and 1.61 mg mL−1 min−1, respectively. The hydrolysis yield using immobilized amyloglucosidase were lower than that of the free one. However, it is relevant to reuse enzyme without losing activity in order to trim down the overall costs of enzymatic bioprocesses as starch transformation into required products in industrial manufacturing. Hydrolysis of sorghum starch using immobilized amyloglucosidase represents a promising alternative towards the development of the glucose syrups production process and its utilization in various industries.  相似文献

该研究对无居民海岛舟山外马廊山岛进行初步踏勘,了解该岛植被特征与植物多样性,探讨海岛植被与大陆植被的差异性,并对海岛植物的主要植物组成、生活型结构、地理成分、群落类型以及多样性和相似性进行了分析。结果表明:外马廊山岛面积小,共有33科51属55种植物,植物种类偏少,留存有普陀狗哇花,滨柃,厚叶石斑木等具有滨海特色植物,具有开发潜力。外马廊山岛热带成分较为明显,与附近地区较为相似。岛上群落优势种或建群种的科为山茶科,大戟科,松科等,与大陆的常绿阔叶林的优势种或建群种的优势科如樟科,金缕梅科,壳斗科,山茶科等不大相同。岛上植物主要以高位芽植物为主(58.18%),但较大陆常绿阔叶林的高位芽植物比例要低。外马廊山岛植物群落结构简单,植物多样性相对较低,物种丰富较低,主要原因在于生态适应性和演替过程的差异,岛上植物群落演替至稳定群落还需要相当一段时间。外来种比例占9.09%,外来种如黑松和红鸡竹能形成群落或在群落中具有较为明显的优势,并可能会造成一定的生态干扰。  相似文献

软儿梨是西北地区特有的一种“冻果”产品,酸甜可口、果香浓郁,是酿造果酒的上乘原料。为明确果酒发酵过程中果香的保留情况,并探究发酵时间的延长对软儿梨果酒品质的影响,以青海省民和县软儿梨冻果为原料,基于带皮渣发酵工艺,先采用顶空固相微萃取(solid?phase microextraction, SPME),再结合气相色谱?质谱联用技术(gas chromatography?mass spectrometry, GC?MS)对原汁(0 d)、前发酵(7、14、28 d)、后发酵(100 d)3个不同时期样品中挥发性风味物质进行了动态跟踪分析,并结合相对气味活度值(relative odor activity value,ROAV)和主成分分析(principal component analysis,PCA)法分别探讨了软儿梨果酒原果香味保留情况和后发酵时间的延长对果酒风味的影响。结果显示:整个发酵过程共检出88种挥发性化合物,其中酯类33种、醇类29种、酸类6种、萜烯类7种以及13种其他类化合物,且风味物质的总含量随发酵的进行呈先上升后略有下降的趋势。ROAV结果表明:软儿梨原汁关键风味物质共8种,分别是丁酸乙酯、己酸乙酯、2?甲基丁酸乙酯、正辛醇、大马士酮、芳樟醇、丁香酚和癸醛;其中果香物质己酸乙酯、正辛醇、芳樟醇、丁香酚和大马士酮在果酒发酵中得到了很好的保留,是决定软儿梨果酒风味的关键物质。主成分分析表明:发酵初期主要的香气贡献物质是具有水果香的丁酸乙酯、3?羟基丁酸乙酯、2?甲基丁酸乙酯;发酵100 d时,乙酸乙酯、α?松油醇、柠檬烯和芳樟醇对香气贡献较大,这些香气化合物共同赋予软儿梨果酒幽香清雅、馥香浓郁的独特风味品质。研究获得了软儿梨果酒关键风味物质及其在不同发酵阶段特征风味物质的变化规律,可为研发高品质软儿梨果酒产品、改进软儿梨果酒发酵工艺提供理论支撑和参考。  相似文献

Summary The aluminium-formaldehyde (ALFA) histofluorescence method was used to study the innervation of the gill of the marine bivalve mollusc Mytilus edulis and the results were contrasted with those obtained with the standard formaldehyde-induced-fluorescence (FIF) method. The ALFA method produced more fluorescing structures than the FIF method, thus revealing fine branches of the branchial nerve running beneath the gill epithelium which previously remained undetected. A molecular mechanical study of two Lys-Ala-Ala tripeptide chains in interaction is presented. The study was carried out to explore the feasibility of dimer formation and explain some features of the crystal structure not completely understood. The role of the counterions present in the crystal structure on the conformation has been included in this study by investigating the effect of removing the charges on the lysyl side chains. The results of the present molecular mechanical study explains well the structural features found by X-ray crystallography, complementing that study.  相似文献

由于在经济生产中的重要意义,猪精液冷冻保存技术成为研究的焦点。早期的研究多集中在对冷冻保护剂和冷冻方法的筛选与优化上,为猪精液冷冻保存进行了深入的探索。但研究表明,无论采用何种冷冻保护剂、何种冷冻方法都损害了精子的受精能力,并伴随有蛋白质的丢失、表达量的改变、功能活性的增减等,这种研究现状迫使该领域的研究向充分揭示冷冻对精子损伤的机理方向转移,希望通过揭示冷冻保存所导致的蛋白质结构和功能改变的实质,充分阐明冷冻损伤的机理,为猪精液冷冻保存提供理论依据,并最终促进猪精液冷冻保存技术的快速发展。  相似文献

The paper presents the capacities of echography in the estimation of the activity of rheumatoid gonitis. A study of knee joints in 126 patients with rheumatoid arthritis has provided the basic ultrasound criteria that are characteristic for the first, second, and third degrees of its progression, which include the pattern of proliferative changes in the synovium, the qualitative and quantitative assessments of intraarticular exudate and its viscosity. The capacities of X-ray study and MRI in the detection of the progression of rheumatoid arthritis are discussed. An algorithm of knee joint radiation study is proposed for patients with rheumatoid arthritis.  相似文献

Since government procurement necessitates comprehensive specifications entirely free from elements of personal equation, the study of dyestuffs for biological stain and therapeutic uses was undertaken in the Medical Department, U. S. Army, early in 1924. Work has been carried on since that time and specifications for the purchase of hematoxylin and eosin Y have been submitted. A number of other dyes have been studied, but the results to date indicate a complex problem which will probably require extensive research for its consummation. Spectrophotometric methods of study are being developed specially in this study and that instrument is being found specially valuable in the study of dyestuffs.  相似文献

The present state of morphological study of variability in the nervous elements under conditions of adaptive change under the influence of afferent effects, age changes during different periods of ontogenesis, under conditions of de- and regeneration of synapses have been considered. This study is a qualitative exploratory analysis of the communities of Turrialba, Siquirres and Guácimo, which border the Parismina River of Costa Rica. The main purpose of this study is to identify the structural weaknesses that are present in the enterprises and the impact of these weaknesses on the capacity for raising the sustainable development competitiveness for the area. The study used semi-structured interviews and focal interviews as instruments of data collection. In addition, the study used the community of Los Santos, located in the Central Valley of Costa Rica, as a reference for comparison. The results indicate weaknesses related to the capacity of organization, financial knowledge, and low technological innovation that demonstrates that the enterprises have a greater direction towards spurious competitiveness.  相似文献

Targeted knock-down is the method of choice to advance the study of sensory and brain functions in the honeybee by using molecular techniques. Here we report the results of a first attempt to interfere with the function of a visual receptor, the long-wavelength-sensitive (L-) photoreceptor. RNA interference to inhibit this receptor led to a reduction of the respective mRNA and protein. The interference effect was limited in time and space, and its induction depended on the time of the day most probably because of natural daily variations in opsin levels. The inhibition did not effectively change the physiological properties of the retina. Possible constraints and implications of this method for the study of the bee's visual system are discussed. Overall this study underpins the usefulness and feasibility of RNA interference as manipulation tool in insect brain research.  相似文献

The developmental study of the skull reveals the following views: The elapid chondrocranium represents a very advanced ophidian type. It more resembles the viperid chondrocranium than the colubrid one. The viperid chondrocranium is only very slightly more advanced than that of the Elapidae. The viperid osteocranium has progressed still further in the development of the poison apparatus than has the elapid osteocranium. The developmental study of the skull advocates the opinion that the family Colubridae has given rise to the family Elapidae, and the Elapidae (or at least the ancestral Elapidae) has given rise to the family Viperidae.  相似文献

The study was done to investigate the content of manmade radionuclides in aquatic organisms of the Yenisei River near the Mining-and-Chemical Combine (MCC) and to estimate the exposure dose rates to organisms from various sources. The results of the investigation and calculations suggest that the main source of radioactive contamination of aquatic organisms is the coolant of the third MCC reactor, which is still being released into the Yenisei. Gamma-spectrometric analysis revealed 23 manmade radionuclides in the biomass of aquatic plants. The aquatic animal Phylolimnogammarus viridis and diatoms also contain manmade radionuclides. Among aquatic organisms, the highest dose rate is received by aquatic plants (up to 39 microGy/day). For most aquatic organisms under study, the dose received from the technogenic irradiation is an order of magnitude higher than the dose received from natural irradiation. The water moss (Fontinalis antipyretica) features the highest capacity to accumulate manmade radionuclides; hence, it accumulates the largest technogenic exposure dose among the study aquatic organisms.  相似文献
