To confirm the effect of the 7 21 Robertsonian translocation on fertility in Japanese Black Cattle, cytogenetic studies were performed on embryos collected from the following 3 mating groups: normal bull cross normal cow, translocation carrier bull cross normal cow, and normal bull cross translocation carrier cow. All the analyzable embryos showed normal chromosome complements when the parents had a normal karyotype. In the group sired by the 7 21 translocation heterozygous bulls, a total of 56 embryos had metaphases suitable for chromosome analyses. Out of these embryos, 28 had normal chromosome complements and 25 were embryos with a balanced karyotype. However, 3 (5.4%) were monosomic and trisomic embryos, presumably resulting from the fertilization of normal ova by aneuploid spermatozoa. Unbalanced embryos were also observed in the chromosome analyses of embryos derived from the 7 21 translocation heterozygous cows. These results suggest that the 7 21 translocation in the heterozygous state may be associated with a slight reduction in reproductive efficiency.  相似文献

The effect of the 1;29 Robertsonian translocation on fertility was studied using embryos resulting from matings of nine carrier cows and two carrier bulls. Embryos were collected from the following three mating groups utilizing superovulation: normal bull cross normal cow, normal bull cross translocation carrier cow, and translocation carrier bull cross normal cow. The proportion of ova which were fertilized did not vary among the groups, indicating that fertilization rates were not affected by the translocation. The translocation cows did yield fewer embryos on average than did cows with normal karyotypes, which may suggest ovulation rates are reduced (at least after superovulation attempts) in cattle carrying the 1;29 translocation. Twenty of 39 embryos successfully karyotyped had abnormal chromosome complements. All four of the theoretically predicted karyotypes and two additional abnormal combinations were found. Eight of 39 (20.5%) embryos karyotyped had unbalanced karyotypes which would have resulted in embryonic loss. The proportion of embryos with unbalanced karyotypes, was slightly higher when the cow (36%) carried the translocation than when the bull (19%) did. Results of this study indicate that fertility is impaired due to the presence of this translocation. The major loss in reproductive potential appears to be due to embryonic loss rather than fertilization failure.  相似文献

A new Robertsonian translocation, rob(15;25), was discovered in a Portuguese Barrosa cow. The animal (2n = 59,XX) was found by G- and R-banding to be a heterozygous carrier of a centric fusion translocation involving chromosomes 15 and 25. C-banding revealed the dicentric nature of this new centric fusion. Comparison of this new translocation with the well-known Robertsonian translocation rob(1;29), which is often found in the same breed, confirmed that two different chromosomes (25 and 29) were involved in the short arms of these two Robertsonian translocations.  相似文献

Chromosomes were harvested from two fibroblast lines derived from a phenotypically normal cow and her albino daughter, both known to be heterozygous for an X/autosome translocation. QFH-banding after early BrdU incorporation identified the translocated autosomal material as chromosome 23 bands 13-25, and revealed that the centromeric portion of the translocated chromosome 23 had been retained. Fluorescent in situ hybridization of a BoLA Class I cDNA probe to the normal chromosome 23 and to the translocated autosomal material confirmed the identity of the translocation, and allowed a more precise sublocalization of the MHC in cattle than that previously reported.  相似文献

Here, we demonstrate that pancreatic microsomal membranes from pigs, sheep, or cattle destined for human consumption can be used as a valuable and ethically correct alternative to dog microsomes for cell-free protein translocation. By adding adequate ribonuclease (RNase) inhibitors to the membrane fraction, successful in vitro co-translational translocation of wild-type and chimeric pre-prolactin into the lumen of rough microsomes was obtained. In addition, the human type I integral membrane proteins CD4 and VCAM-1 were efficiently glycosylated in RNase-treated microsomes. Thus, RNase-neutralized pancreatic membrane fractions from pig, cow, or sheep are a cheap, easily accessible, and fulfilling alternative to canine microsomes.  相似文献

Superovulation and embryo collection procedures were used to study the effect of the 14;20 Robertsonian translocation on fertility and embryo viability. Karyotypes were successfully completed on cells from 77 of the 279 embryos prepared for such analysis. Embryos from 4 cows heterozygous for the translocation were studied. Two bulls with the same condition were studied by using their semen in artificial insemination of cows with normal karyotypes. The proportions of fertilized ova and transferable embryos were not different between cows with the 14;20 translocation and those with normal karyotypes, indicating that fertilization rates were not affected by the translocation. Twenty-two percent of the embryos which were karyotyped had an unbalanced karyotype and would theoretically not have survived to term. All of the theoretically predicted chromosome complements from such a translocation were observed as were three 58,XX,t karyotypes and a 58,XX karyotype. There was no difference in the percentage of embryos with abnormal karyotypes whether the cow or bull was the carrier. Results therefore indicate that fertility is rather severely impaired in carriers of the 14;20 translocation, as was observed with the 1;29 translocation, with most loss due to embryo mortality rather than a lowered conception rate.  相似文献

An experiment was conducted to determine the effects of prostaglandin E(2) (PGE(2)) on ovarian progesterone secretion during the estrous cycle in the cow. Intraluminal uterine catheters were implanted in three beef cows (2 treated, 1 control), and 1.3 mg of PGE(2) were infused into the uterus every 4 hours from days 9 through 21 post-estrus. Blood samples were collected from the jugular vein at 2-hour intervals from days 9 to 21 and twice daily from day 22 to 28 post-estrus. Progesterone was measured by applying a specific, direct plasma radioimmunoassay in all samples without extraction. Intrauterine infusion with PGE(2) resulted in maintenance of luteal-phase progesterone secretion until day 21 post-estrus, 4 days after luteal regression occurred in the vehicle-treated cow. In this study, we demonstrated that PGE(2) can prolong the presence of luteal phase plasma progesterone concentrations by possibly stimulating in vivo steroidogenesis by the corpus luteum during the estrous cycle in the cow.  相似文献

Mosaic evolution of ruminant stomach lysozyme genes   总被引:1,自引:0,他引:1  
The genomes of ruminant artiodactyls, such as cow and sheep, have approximately 10 lysozyme genes, 4 of which are expressed in the stomach. Most of the duplications of the lysozyme genes occurred 40-50 million years ago, before the divergence of cow and sheep. Despite this, the coding regions of stomach lysozyme genes within a species (e.g., cow, sheep, or deer) are more similar to each other than to lysozyme genes in other ruminants. This observation suggests that the coding regions of the stomach lysozyme genes have evolved in a concerted fashion. Our previous characterization of 3 cow stomach lysozyme genes suggested that it was only the coding exons that had participated in concerted evolution. To determine whether the introns and flanking regions of ruminant stomach lysozyme genes are evolving in a concerted or a divergent fashion, we have isolated and characterized 2 sheep stomach lysozyme genes. Comparison of the sequences of the sheep and cow stomach lysozyme genes clearly shows that the introns and flanking regions have evolved, like the 3' untranslated region of the mRNAs, in a divergent manner. Thus, if the four coding exons are evolving by concerted evolution, then a mosaic pattern of concerted and divergent evolution is occurring in these genes. The independent concerted evolution of coding exons of the ruminant stomach lysozyme gene may have assisted in the accelerated adaptive evolution of the lysozyme to new function in the early ruminant.  相似文献

A study was conducted to investigate the effect of the 7/21 Robertsonian translocation on fertilization and subsequent development of bovine oocytes matured in vitro. Semen from Japanese Black bulls, 2 with a normal karyotype (Bulls A and B) and 2 that were heterozygous for the 7/21 translocation (Bulls C and D), was used in this study. In vitro matured bovine oocytes were inseminated with frozen-thawed sperm capacitated with heparin. After insemination, oocytes were cultured at 38.5 degrees C on a monolayer of cumulus cells in TCM-199 supplemented with 5% superovulated cow serum and 0.5 mM sodium pyruvate in an atmosphere of 2% CO2 in air. Cleavage rate was evaluated at 54 h after insemination, and development of embryos to the blastocyst stage was observed 7 to 10 d post insemination. There was no difference in the fertilization rate among the 4 bulls. Although the cleavage rate of oocytes inseminated with semen from Bull C (heterozygote) was lower (P < 0.05) than that obtained with semen from Bull B (normal), the blastocyst formation rate did not differ among the 4 bulls. These results indicate that the 7/21 Robertsonian translocation had no effect on the fertilization and blastocyst formation rates of bovine in vitro-matured oocytes.  相似文献

Abstract. Statistically significant differences were observed in the population density of the horn fly, Haematobia irritans irritans L., on Angus cows having significantly different frame sizes. Angus cows, averaging <112.5 cm in height at the hip, had significantly lower numbers of horn flies than Angus cows that measured 112.5–117.5 cm, 117.5–120 cm, 120–126 cm and >126 cm in height at the hip. The Angus I cows(126 cm). The estimated heritability (h2) of horn fly resistance was 0.43 ± 0.07 and 0.95 ± 0.31 for 1989 and 1990, respectively. Horn fly counts on the Angus I herd (<112.5 cm in height) was 118.1 (probable breeding value, PBV = -20.69) to 165 horn flies per cow (PBV = 26.9 flies per cow in 1989) and from 75.9 (PBV = -29.1) to 134.5 (PBV = 29.5) flies per cow in 1990. Angus I bulls had PBV = -23.7 to 13.4 and from-26.5 to 14.75 in 1989 and 1990, respectively. The Angus II cows had horn fly counts that ranged from 159.6 (PBV of-23.5) to 208.1 (PBV of 25) per cow in 1989 and from 232.3 (PBV of-56.2) to 378.7 (PBV of 90) per cow in 1990. Angus II bulls had PBVs that ranged from-17.1 to 18.9 in 1989 and from -28.1 to 48.8 in 1990. The Angus I cows had significantly (P < 0.0001) lower numbers of hom flies (mean of 63.8 horn flies per m2) than the small, medium or large Angus II cows (mean of 129.4, 149.6 and 145.5 hom flies per m2, respectively). The data indicated that some specific factor(s) associated with cow size contribute(s) to innate resistance of cattle to the horn fly.  相似文献

以小麦品种石麦15(SM15)为对象,以牛粪为有机肥,设置不施氮肥、单施尿素、单施牛粪和有机无机肥配施4种施肥方式,研究不同施肥处理对冬小麦群体光合速率(CAP)、旗叶净光合速率(Pn)、叶面积指数(LAI)、叶绿素荧光参数及产量的影响.结果表明: 无机肥主要作用于生育前期,小麦的CAP、Pn及LAI最高,其次为配施处理,单施有机肥处理最低.花后10 d开始,与单施尿素处理相比,有机肥和有机无机肥配施处理小麦衰老进程较缓慢,在灌浆中后期保持较高的抗氧化酶活性及较长的绿叶面积持续期,进而维持较高的光合物质生产能力.有机无机肥配施处理延缓效果更为明显,籽粒产量最高.可见,有机无机肥配施可以延缓灌浆中后期叶片衰老,维持合理的冠层结构,使小麦具有较强的光合性能,进而获得较高的籽粒产量.  相似文献

威岭黑麦(Weiling rye)是一个高抗白粉病(Erysiphe gramininis f.sp.tritici)的中国矮杆栽培黑麦。以Weiling rye作为白粉病抗源,高感白粉病小麦栽培品种My8443为母本,从Weiling rye与小麦My8443远缘杂交的BC_2F_6后代中鉴定出一个新的小麦-黑麦易位系No.147,以实现威岭黑麦白粉病抗性向普通栽培小麦的转移。No.147及其亲本的抗白粉病特性通过苗期和成株期优势生理小种混合接种和室内单生理小种接种鉴定,改良的染色体C-分带和基因组原位杂交技术(GISH。Ge- nomic in situ hybridization)被用于鉴定小麦和黑麦的染色质,酸性聚丙烯酰胺凝胶电泳(APAGE)被用于鉴定黑麦醇溶蛋白1RS特异条带,11个黑麦种属特异性标记SCM(Secale cereale marker)引物被用于扩增分析黑麦特异性简单重复序列(SSR)。研究结果证实No.147是一个新的高抗白粉病的1BL/1RS小麦-黑麦染色体易位系,并对其产生的细胞学机制进行了分析。论文对中国栽培黑麦抗性基因资源的利用和该易位系在小麦遗传育种改良中的利用价值进行了讨论。  相似文献

用改良的Giemsa C-带技术、DNA原位杂交和酸性聚丙烯酰胺凝胶电泳(A-PAGE)对来源于小麦品种绵阳11与不同黑麦自交系远缘杂交获得的高代株系(BC1F7)的染色体结构和醇溶蛋白进行了研究。结果发现,在鉴定的200个株系中,有45个株系经C-带和A-PAGE检测均一致地发现它们含有一对1RS /1BL易位染色体,而一个株系843-1-1,C-带鉴定、原位杂交结果均证明它含有一对1RS/1BL易位染色体,但A-PAGE醇溶蛋白图谱却不具有黑麦1RS染色体臂的黑麦碱特征带,而表达出既不同于黑麦碱又不同于亲本绵阳11的醇溶蛋白带型。这一结果表明,利用不同的黑麦亲本资源,可以获得黑麦碱基因Sec-1表达缺失的新的1RS/1BL易位系。这种新的1RS/1BL易位系缺失了影响小麦品质的黑麦碱蛋白,因此是进一步研究1RS/1BL 易位对小麦品质影响的珍贵材料。研究指出,在利用外源基因的植物育种中,外源种供体材料的遗传多样性是值得重视的基因资源。  相似文献

应用Biolog方法研究了温室盆栽番茄条件下,施用不同种类及不同腐熟水平的有机物料对土壤微生物群落的影响,施用有机物料60d后取土分析土壤微生物群落多样性及土壤微生物对Biolog微平板中胺、氨基酸、糖、羧酸、聚合物和其它类碳源的利用情况,结果表明,施用有机物料可提高土壤微生物群落多样性,施用新鲜酒糟的多样性指数略高于施用腐熟10d酒糟,牛粪不同腐熟水平对多样性影响显著,且对多样性具有正向或负向的影响;对照和施用酒糟的土壤微生物对聚合物的利用率高于施用牛粪处理,施用新鲜物料处理的土壤微生物对聚合物的利用率高于施用腐熟物料处理。  相似文献

The presence of intermediate filament proteins in vascular tissue cells has been examined by immunofluorescence microscopy on frozen sections of the aortic wall of diverse vertebrates (rat, cow, human and chicken) and by gel electrophoresis of cytoskeletal proteins from whole aortic tissue or from stripped tunica media of cow and man. Most cells of the aortic wall in these species contain vimentin filaments, including smoooth muscle cells of the tunica media. In addition, we have observed aortic cells that are positively stained by antibodies to desmin. The presence of desmin in aortic tissue has also been demonstrated by gel electrophoresis for rat, cow and chicken. In aortic tissue some smooth muscle cells contain both types of intermediate filament proteins, vimentin and desmin. Bovine aorta contains, besides cells in which vimentin and desmin seem to co-exist, distinct bundles of smooth muscle cells, located in outer regions of the tunica media, which contain only desmin. The results suggest that (i) intermediate-sized filaments of both kinds, desmin and vimentin, can occur in vascular smooth muscle in situ and (ii) smooth muscle cells of the vascular system are heterogeneous and can be distinguished by their intermediate filament proteins. The finding of different vascular smooth muscle cells is discussed in relation to development and differentiation of the vascular system.  相似文献

Greenhouse and field trial experiments were performed to evaluate the use of Chromolaena odorata with various soil amendments for phytoextraction of Pb contaminated soil Pb mine soils contain low amount of nutrients, so the additions of organic (cow manure) and inorganic (Osmocote and NH4NO3 and KCl) fertilizers with EDTA were used to enhance plant growth and Pb accumulation. Greenhouse study showed that cow manure decreased available Pb concentrations and resulted in the highest Pb concentration in roots (4660 mg kg(-1)) and shoots (389.2 mg kg(-1)). EDTA increased Pb accumulation in shoots (17-fold) and roots (11-fold) in plants grown in soil with Osmocote with Pb uptake up to 203.5 mg plant(-1). Application of all fertilizers had no significant effects on relative growth rates of C. odorata. Field trial study showed that C. odorata grown in soil with 99545 mg kg(-1) total Pb accumulated up to 3730.2 and 6698.2 mg kg(-1) in shoots and roots, respectively, with the highest phytoextraction coefficient (1.25) and translocation factor (1.18). These results indicated that C. odorata could be used for phytoextraction of Pb contaminated soil. In addition, more effective Pb accumulation could be enhanced by Osmocote fertilizer. However, the use of EDTA in the field should be concerned with their leaching problems.  相似文献

The effects of infection by Mycobacterium avium paratuberculosis (Map) on dairy cows are poorly documented and quite controversial. This retrospective study aimed at quantifying the variation in non-return to service of Holstein dairy cows according to their Map-infection status. Three different statuses were defined based on both individual and herd tests results: ELISA positive cow, all tests negative cow in a negative herd and all tests negative cow in a positive herd. Whatever the age at Map testing, the status was attributed to a cow from its first lactation onwards. Non-return to service was determined at 200 days after first and second services. The study was performed from 1999 to 2007 on 185,950 AI from 48,914 cows in early stage of the infection in 1069 herds by logistic regression controlling for known factors influencing non-return rate. Non-return rate was higher for infected cows compared to negative cows from negative herds (RR of 1.10 or +3.9 points of % of non-return rate). The effect was significant for parities 1 and 2 (RR of 1.11 and 1.12, respectively) but not for higher ones. This effect was lower when comparing positive cows to negative cows in the same herds but relative risks were still above 1. The hypothesis that the effect of Map on non-return depends upon the stage of infection is formulated.  相似文献

Due to hostile condition of red mud (RM), its utilization for vegetation is restricted. Therefore, RM with biowastes as soil amendment may offer suitable combination to support plant growth with reduced risk of metal toxicity. To evaluate the effects of RM on soil properties, plant growth performance, and metal accumulation in lemongrass, a study was conducted using different RM concentrations (0, 5, 10, and 15% w/w) in soil amended with biowastes [cow dung manure (CD) or sewage-sludge (SS)]. Application of RM in soil with biowastes improved organic matter and nutrient contents and caused reduction in phytoavailable metal contents. Total plant biomass was increased under all treatments, maximally at 5% RM in soil with SS (91.4%) and CD (51.7%) compared to that in control (no RM and biowastes). Lemongrass acted as a potential metal-tolerant plant as its metal tolerance index is >100%. Based on translocation and bioconcentration factors, lemongrass acted as a potential phytostabilizer of Fe, Mn, and Cu in roots and was found efficient in translocation of Al, Zn, Cd, Pb, Cr, As, and Ni from roots to shoot. The study suggests that 5% RM with biowastes preferably SS may be used to enhance phytoremediation potential of lemongrass.  相似文献

Thus far the life cycle of Trypanosoma (Megatrypanum) theileri has not been studied. We collected tabanids during the mass hatching, when only few tabanids are infected with trypanosomes. Tabanids were caught immediately after attacking a bait cow to serve as controls or after they had been allowed to engorge on the Trypanosoma (M.) theileri-infected cow. Tabanids were kept in the laboratory and used to study the developmental cycle of T. (M.) theileri in the tabanid gut. From day 1 to day 10 the presumably unfed controls and the engorged tabanids were dissected and cytological smears made from the mid- and hindgut. In total 2.6% (1/38) of the controls and 39% (23/59) of the engorged tabanids were positive for trypanosomes in the 1991 season. From day 1 to day 4 after engorgement trypanosomes were found in the midgut. Epimastigotes with a length of 29 μm on day 1 after infection multiplied by inequal division to form smaller epimastigotes of 26 μm on day 3. On day 4 morphologically indistinguishable trypanosomes of 21 μm total length were found in both mid- and hindgut. From day 5 to day 10 trypanosomes were found only in the hindgut in which the transformation to metacyclics was demonstrated, i.e. epimastigotes transformed to amastigote stages of 5 μm in total length.  相似文献

In Denmark earthworms play an important role in the disappearance of cow pats on pastures. Field investigations were designed to study the influence of earthworms on the transmission of infective Cooperia oncophora larvae from experimental cow pats to grass. Results from the present experiment showed that cow pats, protected from attack by earthworms, disappeared at a much lower rate than unprotected cow pats. Development in faeces and transmission to grass of C. oncophora larvae were disturbed by the disintegration of cow pats. The rapid disintegration of unprotected cow pats resulted in an approximately 50% reduction of infective C. oncophora larvae on grass in the vicinity of these cow pats, as compared with larval contamination of grass around protected cow pats. How earthworms may cause a reduction in the transmission of infective C. oncophora larvae to grass by creating adverse conditions for the survival of parasite larvae in the cow pats, by eating and killing the parasite larvae, and by depositing parasite larvae in the soil is discussed.  相似文献
