In higher vertebrates, smooth muscle (SM) contains two tissue-specific actin isoforms: α-SMA and γ-SMA, which predominate in vascular and visceral SM, respectively. Whether α-SMA has been extensively studied and recognized for its contractile activity in SM and SM-like cells such as myofibroblasts, myoepithelial and myoid cells, the distribution and role of γ-SMA remained largely unknown. We developed a new specific monoclonal antibody against γ-SMA and confirmed that γ-SMA predominates in the visceral system and is minor in the vascular system, although more expressed in highly compliant veins than in stiff arteries. Contrary to α-SMA, γ-SMA is absent from myofibroblasts in vitro, and in fibrotic diseases in vivo. We raised the hypothesis that, whereas α-SMA is responsible for the “contractile” activity, γ-SMA would be involved in the “compliance” of SM and SM-like cells. Several models support this hypothesis, namely veins vs. arteries and the physiological modifications occurring in the uterus and mammary glands during pregnancy and lactation. Our results suggest that, in addition to enteric smooth muscles, γ-SMA is expressed in all the tissues submitted to an important dilation including veins, gravid uterus, and lactating mammary glands. The hypothesis of two complementary mechanical roles for the two SMA isoforms is sustained by their different intracellular distributions and by functional assays. 相似文献

Translation is a central cellular process and is optimized for speed and fidelity. The speed of translation of a single codon depends on the concentration of aminoacyl-tRNAs. Here, we used microarray-based approaches to analyze the charging levels of tRNAs in Escherichia coli growing at different growth rates. Strikingly, we observed a non-uniform aminoacylation of tRNAs in complex media. In contrast, in minimal medium, the level of aminoacyl-tRNAs is more uniform and rises to approximately 60%. Particularly, the charging level of tRNASer, tRNACys, tRNAThr and tRNAHis is below 50% in complex medium and their aminoacylation levels mirror the degree that amino acids inhibit growth when individually added to minimal medium. Serine is among the most toxic amino acids for bacteria and tRNAsSer exhibit the lowest charging levels, below 10%, at high growth rate although intracellular serine concentration is plentiful. As a result some serine codons are among the most slowly translated codons. A large fraction of the serine is most likely degraded by L-serine-deaminase, which competes with the seryl-tRNA-synthetase that charges the tRNAsSer. These results indicate that the level of aminoacylation in complex media might be a competition between charging for translation and degradation of amino acids that inhibit growth. 相似文献

Proteins and nucleic acids constitute a vast potential reservoir of pattern recognizers that operate on the basis of shape complementarity. It is possible to construct models of computing in which these shape-based interactions contribute directly to recognition of signal patterns at the device (or cell) level. The input-output transform is molded by variation-selection evolution. Such models provide clues as to the organizational features that enable biomolecular matter to acquire nonevolutionary modes of problem solving through the evolutionary process. When resources are patchily distributed in an environment, behavioral ecologists frequently turn to ideal free distribution (IFD) models to predict the spatial distribution of organisms. In these models, predictions about distributions depend upon two key factors: the quality of habitat patches and the nature of competition between consumers. Surprisingly, however, no IFD models have explored the possibility that consumers modulate their competitive efforts in an evolutionarily stable manner. Instead, previous models assume that resource acquisition ability and competition are fixed within species or within phenotypes. We explored the consequences of adaptive modulation of competitive effort by incorporating tug-of-war theory into payoff equations from the two main classes of IFD models (continuous input (CI) and interference). In the models we develop, individuals can increase their share of the resources available in a patch, but do so at the costs of increased resource expenditures and increased negative interactions with conspecifics. We show how such models can provide new hypotheses to explain what are thought to be deviations from IFDs (e.g., the frequent observation of fewer animals than predicted in "good" patches of habitat). We also detail straightforward predictions made uniquely by the models we develop, and we outline experimental tests that will distinguish among alternatives. 相似文献

Cloning by somatic cell nuclear transfer is critically dependent upon early events that occur immediately after nuclear transfer, and possibly additional events that occur in the cleaving embryo. Embryo culture conditions have not been optimized for cloned embryos, and the effects of culture conditions on these early events and the successful initiation of clonal development have not been examined. To evaluate the possible effect of culture conditions on early cloned embryo development, we have compared a number of different culture media, either singly or in sequential combinations, for their ability to support preimplantation development of clones produced using cumulus cell nuclei. We find that glucose is beneficial during the 1-cell stage when CZB medium is employed. We also find that potassium simplex optimized medium (KSOM), which is optimized to support efficient early cleavage divisions in mouse embryos, does not support development during the 1-cell or 2-cell stages in the cloned embryos as well as other media. Glucose-supplemented CZB medium (CZB-G) supports initial development to the 2-cell stage very well, but does not support later cleavage stages as well as Whittten medium or KSOM. Culturing cloned embryos either entirely in Whitten medium or initially in Whittens medium and then changing to KSOM at the late 4-cell/early 8-cell stage produces consistent production of blastocysts at a greater frequency than using CZB-G medium alone. The combination of Whitten medium followed by KSOM resulted in an increased number of cells per blastocyst. Because normal embryos do not require glucose during the early cleavage stages and develop efficiently in all of the media employed, these results reveal unusual culture medium requirements that are indicative of altered physiology and metabolism in the cloned embryos. The relevance of this to understanding the kinetics and mechanisms of nuclear reprogramming and to the eventual improvement of the overall success in cloning is discussed. 相似文献

In the “War of Attrition” model of animal conflict, introduced by Maynard Smith, a reward is obtained by whichever of two opponents displays longer, each individual incurring a cost associated with the length of the contest. This model is generalized to allow more general reward and cost functions, and restrictions on the length of contest permitted. This permits unification of the “War of Attrition” model and the “Graduated Risks” model, and also the extension to models in which contests may end either due to injury, or to retreat.In each case it is demonstrated that either (i) there is no evolutionary stable strategy (ESS) or (ii) there is a unique ESS, which is fully specified. In the case where only a finite number of pure strategies are available, global convergence to the ESS is shown.A variety of interesting conclusions of biological relevance emerge, perhaps the most striking being the occurrence of a dichotomous behavioural pattern in an essentially continuous conflict. 相似文献
