Chick pineal cells make melatonin rhythmically, even in culture. Light pulses have two effects on these cells: acute suppression of melatonin synthesis and phase shifts (entrainment) of the underlying pacemaker. The two effects use different mechanistic pathways: the first goes through cAMP, and the second goes through the clock. Both converge on serotonin N-acetyltransferase, whose gene has recently been cloned, with cAMP acting on mRNA levels, but primarily on, enzyme activity, and the clock acting primarily on mRNA levels. Aspects of calcium regulation, not yet well-understood, may impinge on both pathways. These cells also have a novel calcium channel and a novel photopigment. 相似文献

AIM: Heart rate variability (HRV) patterns reflect the changing effect of sympathetic and parasympathetic modulation of the autonomic nervous system. While overall and circadian heart rate (HR) and HRV are well characterized by traditional measures, there is currently no method to measure ultradian cycles of HR and HRV. MATERIALS AND METHODS: HR/HRV for each 2-min interval was calculated using normal-to-normal interbeat intervals from overnight polysomnographic ECGs in 113 subjects, aged 58+/-10 years (65 male, 48 female). HR, SDNN2, high-frequency power (HF) and the LF (low-frequency power)/HF ratio were plotted. A curve-fitting algorithm, developed in MatLab, identified cyclic patterns of HR/HRV and extracted parameters to characterize them. Results were compared for older vs. younger patients, males vs. females, with vs. without severe sleep apnea, and for the upper and lower half of sleep efficiency. RESULTS: Ultradian patterns for different HR/HRV indices had variable correspondences with each other and none could be considered surrogates. Differences were seen for all comparison groups, but no one marker was consistently different across comparisons. CONCLUSION: Each HR/HRV parameter has its own rhythm, and the correspondence between these rhythms varies greatly across subjects. Quantification of ultradian patterns of HRV is feasible and could provide new insights into autonomic physiology. 相似文献

Diurnal cycles of glomerular filtration rate (GFR), effective renal plasma flow (ERPF), and of excretion rates of sodium, potassium, magnesium, chloride and phosphate were measured in a 22 year old man with moderately severe heart failure under standardized conditions. Cycles of GFR, ERPF and excretion of potassium, chloride, and phosphate were indistinguishable from those of normals. The phases of the sodium and probably the magnesium excretory cycles were reversed from normal. The significance of some of the observations is discussed. 相似文献

Heart rate and core temperature are elevated by physical activity and reduced during rest and/or sleep. These masking effects may confound interpretation of rhythm waveforms, particularly in situations where the rest-activity rhythm has a different period from that of the core temperature rhythm. Such desynchronization often occurs temporarily as an individual adjusts to a new work shift or to a new time zone following rapid transmeridian travel, making it difficult to assess the impact of such schedule changes on the circadian system. The present experiments were designed to estimate the magnitude of these masking effects, by monitoring the heart rate, rectal temperature, and nondominant wrist activity (2-min samples) of 12 male subjects during 6 days of normal routine outside the lab and during 6 days of strict bedrest. Subjects also kept sleep, dietary, and exercise logs throughout the study. Average (20-min) waveforms were computed for each subject and each rhythm, at home and in bedrest. In addition, data were partitioned according to self-reported sleep and wake times and were analyzed separately for each state. Average waveform comparisons indicated that about 45% of the range of the circadian heart rate rhythm during normal routine was attributable to the masking effects of activity during wake, which also produced a 16% elevation in mean heart rate during wake and an 11% increase in mean heart rate overall. (Analysis of variance indicated that mean heart rate during sleep at home was not significantly different from the mean during sleep in bedrest.) On average, about 14% of the range of the circadian temperature rhythm during normal routine was attributable to the effects of activity masking. However, the change in range of the temperature rhythm, from home to bedrest, was very variable between subjects (-41% to +13%). This variability was not accounted for by age or by reported frequency of exercise at home. Normal activity during wake increased the mean temperature during wake by an average of 0.16 degrees C and the overall mean by about 0.12 degrees C. (Analysis of variance indicated that mean temperature during sleep at home was not significantly different from the mean during sleep in bedrest.) A 10-hr "night" (lights-off from 2200 to 0800 hr) was provided during bedrest, within which subjects could select their own sleep times. ECG alternans is commonly held to be an indicator of electrical instability of the heart, but the development of alternans has not yet been fully understood theoretically. We investigate the onset of alternans and 2:1 rhythms for stimulation at increasing frequencies in the Beeler-Reuter model, a simple ionic model of cardiac tissue. We find hysteresis and bistability at the onset of alternans; well-timed stimuli can switch between the two limit cycles. We determine quantitatively the effect of blocking specific ionic currents. Moreover, we find that calcium buffers generally promote alternans. 相似文献

Early life nutritional adversity is tightly associated with the development of long-term metabolic disorders. Particularly, maternal obesity and high-fat diets cause high risk of obesity in the offspring. Those offspring are also prone to develop hyperinsulinemia, hepatic steatosis and cardiovascular diseases. However, the precise underlying mechanisms leading to these metabolic dysregulation in the offspring remain unclear. On the other hand, disruptions of diurnal circadian rhythms are known to impair metabolic homeostasis in various tissues including the heart and liver. Therefore, we investigated that whether maternal obesity perturbs the circadian expression rhythms of clock, metabolic and inflammatory genes in offspring heart and liver by using RT-qPCR and Western blotting analysis. Offspring from lean and obese dams were examined on postnatal day 17 and 35, when pups were nursed by their mothers or took food independently. On P17, genes examined in the heart either showed anti-phase oscillations (Cpt1b, Pparα, Per2) or had greater oscillation amplitudes (Bmal1, Tnf-α, Il-6). Such phase abnormalities of these genes were improved on P35, while defects in amplitudes still existed. In the liver of 17-day-old pups exposed to maternal obesity, the oscillation amplitudes of most rhythmic genes examined (except Bmal1) were strongly suppressed. On P35, the oscillations of circadian and inflammatory genes became more robust in the liver, while metabolic genes were still kept non-rhythmic. Maternal obesity also had a profound influence in the protein expression levels of examined genes in offspring heart and liver. Our observations indicate that the circadian clock undergoes nutritional programing, which may contribute to the alternations in energy metabolism associated with the development of metabolic disorders in early life and adulthood. 相似文献

We evaluated their circadian rhythms using data from electrocardiographic records and examined the change in circadian period related to normal RR intervals for astronauts who completed a long-term (≥6-month) mission in space. The examinees were seven astronauts, five men and two women, from 2009 to 2010. Their mean?±?SD age was 52.0?±?4.2 years (47–59?yr). Each stayed in space for more than 160 days; their average length of stay was 172.6?±?14.6 days (163–199 days). We conducted a 24-h Holter electrocardiography before launch (Pre), at one month after launch (DF1), at two months after launch (DF2), at two weeks before return (DF3), and at three months after landing (Post), comparing each index of frequency-domain analysis and 24-h biological rhythms of the NN intervals (normal RR intervals). Results show that the mean period of Normal Sinus (NN) intervals was within 24?±?4?h at each examination. Inter-individual variability differed among the stages, being significantly smaller at DF3 (Pre versus DF1 versus DF3 versus Post?=?22.36?±?2.50 versus 25.46?±?4.37 versus 22.46?±?1.75 versus 26.16?±?7.18?h, p?<?0.0001). The HF component increased in 2 of 7 astronauts, whereas it decreased in 3 of 7 astronauts and 1 was remained almost unchanged at DF1. During DF3, about 6 months after their stay in space, the HF component of 5 of 7 astronauts recovered from the decrease after launch, with prominent improvement to over 20% in 3 astronauts. Although autonomic nervous functions and circadian rhythms were disturbed until one month had passed in space, well-scheduled sleep and wake rhythms and meal times served as synchronizers. 相似文献

Using a data logger system, body temperature (dorsal subcutaneous temperature), heart rate, ingestive or ruminating behaviour and posture in adult Shiba goats tied to a stanchion were recorded automatically during 24 hours, to obtain basic information on the biological rhythms. A 600 g of usual ration mixed with hay cube, hay, beet pulp and wheat bran was fed twice a day (morning; 9:00-9:30, evening; 16:00-16:30). Animals kept under an artificial photoperiod (12L-12D, light period; 5:30-17:30) and about 10 degrees C room temperature. 1) Diurnal patterns of the above-mentioned items were recorded, mutual relationships relationships between these items were revealed. 2) The heart rate was higher after morning feeding, or during a light period and decreased gradually from midnight to early morning. Twice feeding greatly increased the heart rate. 3) The body temperature was lower in the early morning and increased gradually after morning feeding and showed the highest level during 1 to 1.5 hours after evening feeding. After that it decreased gradually till the early morning. 4) The numbers of jaw movement (bites/min) were a 70-90 bites in the ingestive behaviour and a 80-90 bites in the ruminating behaviour at dark period. 5) The total heart rate was a 110000 to 120000 beats/day, the total biting time was a 9.5 hours/day, and the mean standing time was a 9.3 to 11.7 hours/day. The standing time during light period (12 hours) was a 7.3 to 9.9 hours and that during dark period (12 hours) was a 1.8 to 2 hours. 相似文献

Activity and the electrocardiogram and respiratory potentials of 17 adult River lampreys, Lampetra fluviatilis , acclimated to 7°± 1°C and a light/dark cycle of 11/13 hours, were recorded throughout the spawning run (November to April). Activity was far greater in the dark in all but the sexually mature animals, reflecting behaviour in the field. A circadian rhythm was also observed in the heart rate during periods of inactivity, and this was correlated with an increase in ventilatory frequency or amplitude. A similar rhythm was not present in the sexually mature animals. Moreover, heart and ventilatory frequencies tended to increase during the spawning run. A whole number ratio between heart rate and ventilatory frequency was seen in only 11 of 425 recordings. 相似文献

The neurobiological substratum of circadian rhythmicity encompasses three levels of integration: firstly, generation of time signals by circadian pacemakers; secondly, entrainment of pacemakers by environmental influences; thirdly, coupling of circadian pacemakers among themselves and with target systems responsible for the expression of overt rhythms. From recent contributions, the notion that circadian organization results from the interaction of independent oscillators and pathways has been strengthened. In addition, recent evidence supports the existence of circadian rhythmicity in single isolated neurons. New information was produced on the gene control of circadian rhythm generation in Drosophila, as well as interesting advances in the understanding of neuronal mechanisms involved in the generation, entrainment and coupling of circadian rhythms in various species. 相似文献

We demonstrated in previous works that the circadian rhythms of blood pressure (BP) and atrial natriuretic peptide (ANP) are antiphasic in normal subjects and in essential hypertension. The aim of the present study was to assess the circadian rhythms of BP and ANP in 20 patients with stable congestive heart failure (CHF), divided into two groups of 10 according to their New York Heart Association functional class. A matched control group of 10 normal volunteers was also studied. Noninvasive BP monitoring at 15-min intervals was performed for 24 h. Peripheral blood samples were also obtained at 4-h intervals starting from 08:00 h. The mean (+/- SEM) circadian mesors of ANP plasma levels were 13.4 +/- 1.7 pmol/L in the control group, 28.6 +/- 2.4 pmol/L in the group of 10 patients in class II, and 81.5 +/- 12 pmol/L in the group of 10 patients in class III-IV. In normal subjects, plasma ANP concentration was highest at 04:00 h (21.5 +/- 2.7 pmol/L) and lowest at 16:00 h (8.8 +/- 2.4 pmol/L; p less than 0.01). Both groups of patients with CHF showed no significant circadian change in the plasma levels of ANP and also a significantly blunted circadian rhythm of BP. Cosinor analysis confirmed the loss of the circadian rhythms of ANP and BP in CHF patients. ABSTRACT: The amount and timing of sleep and sleep architecture (sleep stages) are determined by several factors, important among which are the environment, circadian rhythms and time awake. Separating the roles played by these factors requires specific protocols, including the constant routine and altered sleep-wake schedules. Results from such protocols have led to the discovery of the factors that determine the amounts and distribution of slow wave and rapid eye movement sleep as well as to the development of models to determine the amount and timing of sleep. One successful model postulates two processes. The first is process S, which is due to sleep pressure (and increases with time awake) and is attributed to a 'sleep homeostat'. Process S reverses during slow wave sleep (when it is called process S'). The second is process C, which shows a daily rhythm that is parallel to the rhythm of core temperature. Processes S and C combine approximately additively to determine the times of sleep onset and waking. The model has proved useful in describing normal sleep in adults. Current work aims to identify the detailed nature of processes S and C. The model can also be applied to circumstances when the sleep-wake cycle is different from the norm in some way. These circumstances include: those who are poor sleepers or short sleepers; the role an individual's chronotype (a measure of how the timing of the individual's preferred sleep-wake cycle compares with the average for a population); and changes in the sleep-wake cycle with age, particularly in adolescence and aging, since individuals tend to prefer to go to sleep later during adolescence and earlier in old age. In all circumstances, the evidence that sleep times and architecture are altered and the possible causes of these changes (including altered S, S' and C processes) are examined. 相似文献
