The epidermis of the mudskipper Periophthalmus magnuspinnatus consisted of three layers: the outermost layer, middle layer and stratum germinativum. Extensive vascular capillary networks were present near the superficial layer of epidermis and outermost layer. The diffusion distance between the vascular capillaries and the surface of epidermis was c . 1.5 ± 0.9μm. The middle layer consisted of small or voluminous cells swollen by epidermal cells. Due to the swollen cells, the thickness of the epidermis increased and the epidermis appeared web-like. The swollen cells contained tonofilaments, lucent contents and desmosomes. Fine blood capillaries were also discernible in this layer. Well-developed lymphatic spaces containing lymphocytes existed in the stratum germinativum. Numerous blood capillaries were present under the basement membrane. The dermis consisted of a stratum laxum and stratum compactum, and there was a definite area with acid mucopolysaccharides and a small scale in the stratum laxum. The skin had an epidermal pigment cell, dendritic melanophores (-cytes) containing melanin granules within their cytoplasm, and two kinds of dermal pigment cells, melanophores and colourless pigments containing reflecting platelets.  相似文献

Mucus glycoprotein (RGP) was purified and characterized from the skin mucus of rainbow trout, Salmo gairdneri. RGP was found to contain 30.1% NeuAc, 26.0% GalNAc, 5.0% Gal, and 26.0% amino acids. The protein moiety of RGP is very rich in Thr (32.4 mol%). Neither NeuGc nor KDN (2-keto-3-deoxy-d-glycero-d-galacto-nononic acid) was found in RGP. Alkaline borohydride treatment of RGP yielded a major disaccharide alditol, NeuAcα2→6GalNAc-ol and more than 4 minor oligosaccharide alditols including NeuAc→(GalNAcα1→)GalNAc-ol. It was evident that an average RGP molecule has approximately 500 NeuAc-containing oligosaccharide chains, which are attached to the Thr and Ser residues of the protein moiety and spaced at an average of 3 amino acids apart.  相似文献

We describe the adhesive nature of the pectoral and pelvic fins in the catfish Pseudocheneis sulcatus, as examined by scanning electron microscopy. The outer rays of these fins are modified into structures that bear prominent transverse ridges and grooves. The outer epidermal cells of the ridges are thrown into elongated spines. Mucous pores (openings of mucous glands) are frequently present (100m apart) in the epidermis of the ridges and show entangled mucus droplets. In the pectoral fins, they are present towards the contour areas of the outer rays, but they are absent in the pelvic fins. In the latter, mucous pores are present near the base of the ridges (distal to the inner rays). Spines as well as mucous pores are absent in the cells that line the groove between two adjacent ridges. We suggest that in this species adhesion is effected by suction pressure generated by the musculature attached to the grooves and ridges, and that mucus and the spines aid in this process.  相似文献

Skin is a highly heterogeneous tissue. Intra-dermal structures include hair follicles, arrector pili muscles, epidermal specializations (such as Merkel cell clusters), sebaceous glands, nerves and nerve endings, and capillaries. The spatial arrangement of these structures is tightly controlled on a microscopic scale - as seen, for example, in the orderly arrangement of cell types within a single hair follicle - and on a macroscopic scale - as seen by the nearly identical orientations of thousands of hair follicles within a local region of skin. Visualizing these structures without physically sectioning the skin is possible because of the 2-dimensional geometry of this organ. In this protocol, we show that mouse skin can be dissected, fixed, permeabilized, stained, and clarified as an intact two dimensional object, a flat mount. The protocol allows for easy visualization of skin structures in their entirety through the full thickness of large areas of skin by optical sectioning and reconstruction. Images of these structures can also be integrated with information about position and orientation relative to the body axes.  相似文献

A thick, elaborate glycocalyx was associated with α and β chloride cells of rainbow trout gills, a less elaborate one with surface epithelial cells and a scanty and inconspicuous one with mucous cells. Linkage studies on seven erythrocyte antigen loci, the major histocompatibility complex (MHC) and the haemoglobin beta gene cluster (HBB) were performed in 48 Norwegian goat families. Close linkage was excluded between the erythrocyte antigen B-system and the MHC, between the erythrocyte antigen N7 locus and HBB, and between the MHC and the HBB.  相似文献

The phenotype and frequency of cells in normal human peripheral blood spontaneously secreting IL-2, IL-4, IL-6, IL-10, IFN and TNF-α ex vivo was determined using ELIspot assays. CD4+T cells were the dominant source of IL-2 and IL-4 while multiple cell types (primarily CD8+lymphocytes) produced IFN. Fewer than 0.05% of mononuclear cells were spontaneously secreting these T cell derived factors. By comparison, IL-6, IL-10 and TNF-α were produced by 0.7–20% of PBMC. The primary sources of the latter cytokines were CD14+macrophages/monocytes. A significant positive correlation was found in the frequency of cells secreting IL-6, IL-10 and TNF-α ex vivo, suggesting that the release of such factors was coordinately regulated. No such correlation was found among IL-2, IL-4 and IFN secreting cells, indicating that the production of predominantly T cell derived cytokines was regulated independently.  相似文献

Abstract In some living osteichthyans (e.g. the armoured catfishes) the postcranial dermal skeleton exhibits tooth-like structures (odontodes) similar to those present in the dermal skeleton of the ancient craniates. We have undertaken this work to compare odontode with tooth development, structure, attachment to a bony support and replacement. We studied the odontodes fixed on the scutes (i.e. postcranial dermal plates) in a growth series of Corydoras aeneus using light, scanning and transmission electron microscopy. Odontodes are constituted of a pulp cavity surrounded by a cone of dentine itself capped with hypermineralized substance. The pulp cavity is devoid of nerves and blood vessels and there are no odontoblastic processes in the dentine. The dentine cone is firmly attached to a circular bony protuberance of the scute surface, the pedicel or attachment bone, by means of a ligament. An odontode anlage develops as a small invagination of a dermal papilla projecting into the epidermis, the basal cell layer of which constitutes a dental epithelium. First, dentine is deposited, next the hypermineralized substance, then the ligament and attachment bone. Odontodes develop in two positions with regard to the scute surface: a primary position when new odontodes form at the posterior border of the enlarging scute; a secondary position when new odontodes replace old odontodes that have been shed during thickening of the scute. In this case, the ligament and part of the base of the dentine cone are resorbed but not the pedicel of attachment bone, which is covered by deposition of scute matrix after the odontode has been shed. Within the scute matrix, the embedded pedicels of successive generations of odontodes are preserved, forming piles in the scutes of adult specimens.  相似文献

The subterranean fish Phreatobius sanguijuela, originally described from Bolivia, was captured in different wells near São Francisco do Guaporé, Rondônia State, Brazil. Thirty wells were investigated in April and July 2012, and September 2013. These surveys resulted in the capture of 58 individuals from eight wells and comprised three to 14 individuals per well. The capture of the individuals allowed a detailed evaluation of their colours in life, behaviour in the field and in captivity, aspects of their biology, confirmation of the species identification and provided new diagnostic characteristics to distinguish between P. sanguijuela and Phreatobius dracunculus. Cannibalism, territorialism, agonistic interactions and phototaxis behaviour were not observed. Phreatobius sanguijuela exhibited cryptobiotic habits and two behaviours under stressful conditions. The analysis of stomach contents reveals that this species apparently feeds on invertebrates, almost exclusively on earthworms. The sex ratio was 1:1. The absence of opercular movement during the resting period associated with intense blood irrigation of the skin indicates a possible cutaneous respiration as an alternative form of gas exchange. Local people often mistake P. sanguijuela for helminths and have the habit of releasing non‐native fishes into the wells or to use chemicals to eliminate them. The consequence of this habit for the conservation of the species requires further evaluation.  相似文献

目的:探讨红细胞分布宽度(RDW)、中性粒细胞与淋巴细胞比值(NLR)与晚期非小细胞肺癌(NSCLC)患者临床病理特征及预后的关系。方法:选择2017年5月至2019年5月我院收治的106例确诊为晚期NSCLC患者(NSCLC组)和门诊接诊的102例体检正常者(对照组)作为研究对象。检测RDW、NLR,分析RDW、NLR与NSCLC患者临床病理特征以及预后的关系。结果:NSCLC组RDW、NLR高于对照组(P<0.05),年龄≥60岁、淋巴结转移NSCLC患者RDW高于年龄<60岁、无淋巴结转移NSCLC患者(P<0.05),TNM分期为Ⅳ期、淋巴结转移NSCLC患者NLR高于TNM分期为Ⅲ期、无淋巴结转移患者(P<0.05)。Kaplan-Meier生存曲线分析结果显示高RDW组、高NLR组NSCLC患者生存率低于低RDW组、低NLR组(P<0.05)。单因素COX回归分析显示分化程度、TNM分期、淋巴结转移、RDW、NLR与NSCLC预后有关(P<0.05),多因素COX回归分析显示淋巴结转移、RDW、NLR与NSCLC患者预后相关(P<0.05)。结论:晚期NSCLC患者RDW、NLR较高,RDW与年龄、淋巴结转移有关,NLR与TNM分期和淋巴结转移有关,高水平RDW、NLR预示着NSCLC预后不良,可作为预后评估的辅助指标。  相似文献

Abstract. The internal parenchyma of the leaf and rhizome in 36 species of Sansevieria is made of dead cells and living cells arranged in a regular pattern. Intercellular spaces are lacking. The walls of dead cells consist of an inner amorphous layer positive to the fluorescence test for callose, a middle suberin-like layer and an outer fibrillar layer. In about half of the species examined, the inner layer forms distinctive thickenings. Detached leaves of Sansevieria lose water very slowly, and are able to recover it quickly. The pattern of leaf dehydration appears to be related to leaf morphology, whereas no relation is evident between the pattern of leaf rehydration and leaf morphology. Neither leaf dehydration nor leaf rehydration pattern is affected by the presence of wall thickenings in the dead parenchyma cells. The fresh weight per unit volume of both turgid and droughted leaves is nearly 1, denoting that the dead cells are filled with water and do not undergo substantial cavitation during drought. The data indicate that the dead parenchyma cells of Sansevieria are a specialized water-storing system.  相似文献

Water extracts of the bark of Uncaria tomentosa, a vine indigenous to South America, has been used for generations as an "immuno modulator". To understand the basis of this immuno modulatory effect we fed mice in their drinking water with C-Med 100, which is a commercially available water extract from Uncaria tomentosa. We found a dose-dependent increase in spleen cell numbers in the supplemented mice, but the proportions of B cells, T cells, NK cells, granulocytes, and memory lymphocytes were normal. However, there were no detectable changes of the lymphoid architecture of the spleen even after long-term treatment. Further, when C-Med 100 treatment was interrupted the cellularity returned to normal level within four weeks. The increased number of lymphocytes was most likely not due to increased production because C-Med 100 did not have any significant effect on precursor cells nor on the accumulation of recent thymic emigrants in the spleen. We conclude that accumulation is most likely due to prolonged cell survival, because adoptive transfer experiments demonstrated that C-Med 100 treatment significantly prolonged lymphocyte survival in peripheral lymphoid organs, without increasing their proliferation rate. Since the accumulation was reversible and without detectable pathological effects, these results suggest the use of C-Med 100 as a potential agent for clinically accelerating the recovery of patients from leukopenia.  相似文献

The DNA encoding soluble B lymphocyte stimulator (134–285 amino acids, sBLyS) mutant with residues 217–224 replaced by two glycines (named msBLyS) was constructed. The sequence encoding a foreign immunodominant T-helper epitope from ovalbumin (OVA) was then coupled to the 5′-end of msBLyS cDNA. After being sequenced, the recombinant DNA was ligated into the prokaryotic expression vector pQE-80L. The recombinant protein was produced in E. coli DH5α after induction with IPTG with the yield of more than 40% of total bacterial protein. The recombinant protein was purified with Ni-NTA chromatography and Sepharcryl S200 chromatography to a purity of more than 98%. The BALB/c mice, immunized with the recombinant protein, produced anti-BLyS antibodies at a high level, which indicated that the recombinant BLyS mutant modified with T-helper epitope elicited polyclonal antibodies with cross-reactivity with BLyS in vivo. This recombinant protein may therefore be used as immune inhibitor of BLyS for treating BLyS -associated autoimmune diseases.  相似文献

The population density and summer daytime habitat characteristics of the endangered torrent catfish, Liobagrus reini, in the Urano River, a tributary of the Chikuma River in Nagano, central Japan, were investigated. A total of 105 individuals were captured in 2001–2003, and the estimated population density 100m–2 in the whole study area ranged from 0.20 to 2.96. Liobagrus reini were captured from under cobbles and boulders, interstices in bedrock, and in clumps of roots of terrestrial vegetation along banks. Compared with the distribution of physical factors in the study area, the fish selected shallow habitats with the riverbed covered by cobbles and boulders. Boulders and larger cobbles were chosen by larger individuals as a habitat. Habitat selection was not correlated with current velocity, and some individuals were sampled near (<1m) the riverbank with no current. For the conservation of L. reini, the preservation of natural riverbanks and areas with boulders and large cobbles in the riverbed is concluded to be important.  相似文献

It was determined by means of light and electron microscopy that the granular blood cells of Pacific oysters could absorb basic dyes from dye-coated plastic beads injected into the mantles of oysters; in doing so, the granular blood cells became altered morphologically and behaviorally. In addition, it was observed that oysters could absorb the basic dye, toluidine blue O, in low concentration from sea water. It was shown that the absorption of toluidine blue O caused a shift in the blood cell levels of oysters, and it was suggested that this shift was occasioned by the absorption of toluidine blue O by granulocytes.  相似文献

Abstract Adults of the European lancelet were collected at Banyuls-sur-Mer (Mediterranean France) in mid-spring, shortly before the onset of the breeding season. The dorsal and ventral fin rays were studied by light microscopic histochemistry and by transmission electron microscopy (TEM). Each fin ray is a mass of extracellular material that accumulates beneath the mesothelium of a fin box coelom. The fin ray material is rich in lipids, proteins, and neutral mucopolysaccharides. TEM reveals no lipid droplets in this material, which consists entirely of a packed mass of 15–20 nm granules of medium electron density. It is likely that these granules consist of glycoproteins or glycolipoproteins. Our results are consistent with the proposal of Azariah (1965, Journal of the Marine Biological Association of India 7: 459–661) that lancelet fin rays are nutritional reserves supporting gametogenesis during the breeding season.  相似文献

应用扫描和透射电镜技术对金鱼(Carassius auratus)孵出1 d3、d9、d龄仔鱼的体表进行了观察。仔鱼的上皮细胞呈扁平、多边形,彼此之间由增厚的边缘嵴状突紧密连接,细胞向内凹陷的表面有指纹状嵴突。孵出3 d的仔鱼体表两侧各有一列间隔有序呈丘状突起的味蕾,外被单层上皮细胞,味孔处有一根粗圆的感觉毛。粘液细胞出现在仔鱼头部、腹部、体侧的表皮上,并开口于多个相邻上皮细胞间的连接处。粘液细胞胞质比例大,内含丰富、平行排列的粗面内质网,旁边有大量大小不一、内含物液化状、近圆形的粘原颗粒。9 d后仔鱼体表完全被粘液覆盖,初步建立了以粘液细胞为基础的防病和抗病机制,以机械和化学感觉为主的Ⅰ型味蕾觅食方式。  相似文献
