This study reports the results of both a small-scale and a large-scale field treatment to assess control of the haematophagous fly Hippobosca maculata Leach a serious pest of equines in a stud in India using a deltamethrin based formulation, Butox. In the small-scale field trial application of 2 litres of deltamethrin at 0.001-0.003% concentration gave 90-100% control over 30 days. At 0.004% and 0.005% concentrations complete control was recorded for 45 and 90 days respectively. Mass application of 2 litres of 0.005% deltamethrin to equines and bovines controlled infestations of H. maculata for 1 year.  相似文献

The efficacy of 10% (w/w) permethrin impregnated eartags for the control of the haematophagous fly pests Stomoxys calcitrans L., Haematopota dissimilis Meigen and Hippobosca maculata Leach on equines in India was determined. The tags were found to be effective for 1-2 months against S. calcitrans and H. dissimilis but completely ineffective against H. maculata. No tags were lost during the study. Tags can be used as part of an integrated control programme. This is the first reported use of eartags for control of haematophagous flies on horses.  相似文献

The effectiveness of two pyrethroids, fenvalerate and deltamethrin, against the fly Haematobia irritans exigua de Meijere (Diptera: Muscidae) on buffalo was considered in a field trial. Fenvalerate provided 100% control for 1, 2 and 4 weeks at concentrations of 0.03, 0.04 and 0.05%, respectively. Concentrations of 0.01 and 0.02% were less effective. One hundred percent control of this fly was obtained with deltamethrin for 2, 3 and 6 weeks at concentrations of 0.003, 0.004 and 0.005%, respectively. Deltamethrin concentrations of 0.001% and 0.002% achieved fly control for only 1-2 weeks, respectively.  相似文献

The efficacy of tags impregnated with pheromone and acaricide for control of Amblyomma variegatum on cattle in Guadeloupe was determined for a 13-week trial. Comparisons were made between untreated cattle and cattle with tags containing either pheromones alone (o-nitrophenol, methyl salicylate, 2,6-dichlorophenol and phenylacetaldehyde), pheromones plus acaricide (cyfluthrin or deltamethrin), or acaricide alone. Tags were fastened to cattle both on collars on the neck and with adhesive to tail hairs, with over 98 and 90% of tags retained, respectively, during the trial. By the end of the trial, tick infestations increased on untreated cattle (311.7%) and cattle with pheromone tags (154.8%) but decreased on cattle with pheromone/cyfluthrin (–45.0%), cyfluthrin (–42.8%), pheromone/deltamethrin (–68.7%) and deltamethrin tags (–87.6%). Cattle with pheromone tags had greater proportions of ticks on the hind regions (81%) compared to untreated cattle (62.5%) and on the front regions (18.2%) compared to untreated cattle (8.2%) indicating that ticks aggregated in response to pheromones. Analysis of hair samples by gas chromatography indicated that, within 2 days of placement, cyfluthrin spread rapidly over the body from the tags. Both cyfluthrin and deltamethrin were present in detectable levels on all eight body regions examined throughout the trial. Analysis of the tags indicated that they all still contained pheromone components after 13 weeks under field conditions.  相似文献

Deltamethrin‐based campaigns to control Triatoma infestans (Klug) (Hemiptera: Reduviidae) have decreased in success as a result of the development of insecticide resistance. We compared the in vitro effects of the pyrethroid deltamethrin and two doses of the organophosphate fenitrothion, presented on different materials, on T. infestans from La Esperanza, Argentina. Laboratory tests demonstrated a decrease in susceptibility to deltamethrin in the field population [LD50: 30.32 nanograms per insect (ng/i)] compared with the reference population (LD50: 0.13 ng/i), giving a high resistance ratio of 233.42. By contrast, similar susceptibility to fenitrothion was assessed in both the field and reference populations (LD50: 21.65 ng/i and 21.38 ng/i, respectively). The effectiveness of the formulated insecticides varied according to the surfaces to which they were applied. The application of fenitrothion formulations to glass or brick resulted in mortality of 90–100%. The application of fenitrothion formulations to wood or mud caused mortality in the range of 6.7–56.7%. Resistant insects presented low mortality when exposed to the deltamethrin formulation and high mortality when exposed to fenitrothion formulations. Moreover, the insecticides demonstrated residual activity only when applied to glass. The present work demonstrates that fenitrothion is an alternative to pyrethroids for the management of deltamethrin‐resistant insects in La Esperanza. However, this effectiveness is not sustained over time.  相似文献

评价使用氟氯氰菊酯、溴氰菊酯、高效氯氟氰菊酯喷雾和喇叭口点施处理对草地贪夜蛾的活性与防效,为草地贪夜蛾综合防控提供技术支持。采用喷雾法测定了3种农药原药对草地贪夜蛾3龄幼虫毒力;采用喷雾和喇叭口点施2种方式田间施用5.7%氟氯氰菊酯乳油、25 g/L溴氰菊酯乳油、5.0%高效氯氟氰菊酯乳油,药后第1天、第3天、第7天调查挂牌标记玉米上草地贪夜蛾活虫数,计算防治效果。3种拟除虫菊酯农药对草地贪夜蛾3龄幼虫LC 50值大小顺序依次为氟氯氰菊酯(29.80 mg/L)<高效氯氟氰菊酯(42.39 mg/L)<溴氰菊酯(49.88 mg/L);3种拟除虫菊酯类农药在54.00 g a.i./ha剂量下均能明显降低草地贪夜蛾田间虫口数量,同等剂量防效氟氯氰菊酯乳油>高效氯氟氰菊酯乳油>溴氰菊酯微乳剂。由于草地贪夜蛾低龄幼虫和高龄幼虫取食部位有差异,喷雾法防治低龄幼虫的效果好,喇叭口点施防治高龄幼虫的效果好。拟除虫菊酯类农药对草地贪夜蛾具有较好的防治效果,喇叭口点施方式节省药剂、提高对高龄幼虫的防效,在防治草地贪夜蛾等害虫方面具有广阔的应用前景。  相似文献

Abstract:   The toxicity of four insecticides used to control the Colorado potato beetle, Leptinotarsa decemlineata (Say) (Coleoptera: Chrysomelidae), imidacloprid (Admire®), cryolite (Kryocide®), cyromazine (Trigard®), and Bacillus thuringiensis var. tenebrionis (Novodor®), to one of its natural enemies, the 12-spotted lady beetle, Coleomegilla maculata lengi Timberlake (Coleoptera: Coccinellidae) was determined in the laboratory. Toxicity assays against C. maculata adults and larvae consisted of (1) topical applications and (2) exposures to treated foliage and prey, using concentrations up to 10 times the manufacturer's recommendations. Over a 6-day period, cyromazine (insect growth regulator) and B. t . var. tenebrionis (microbial insecticide) had no lethal effects on first and third instars C. maculata . For both larval and adult stages, cryolite (inorganic insecticide) caused very low predator mortality when topically applied and moderate mortality when ingested through contaminated eggs of Colorado potato beetles. Imidacloprid (systemic organic insecticide) was highly toxic to adult and larval C. maculata . Its estimated LD50 at 6 days following treatment, corresponded to 0.02–0.09 times the recommended field concentration, depending on the developmental stage and mode of contamination. These results indicate that integrated pest management programmes for Colorado potato beetles using imidacloprid or, to a lesser degree, cryolite, would be detrimental to C. maculata . Cyromazine and B. t . var. tenebrionis seem to present a better compatibility with the protection of C. maculata populations.  相似文献

A recent study reported neutralizing antibodies to West Nile virus (WNV) in horses from four ranches of southern Pantanal. To extend that study, a serosurvey for WNV and 11 Brazilian flaviviruses was conducted with 760 equines, 238 sheep and 61 caimans from 17 local cattle ranches. Among the tested equines, 32 were collected from a ranch where a neurologic disorder outbreak had been recently reported. The sera were initially screened by using a blocking ELISA and then titrated by 90% plaque-reduction neutralization test (PRNT90) for 12 flaviviruses. Employing the criterion of 4-fold greater titer, 78 (10.3%) equines were seropositive for Ilheus virus, 59 (7.8%) for Saint Louis encephalitis virus, 24 (3.2%) for WNV, two (0.3%) for Cacipacore virus and one (0.1%) for Rocio virus. No serological evidence was found linking the neurological disease that affected local equines to WNV. All caimans and sheep were negative by blocking ELISA for flaviviruses. There were no seropositive equines for Bussuquara, Iguape, Yellow fever and all four Dengue virus serotypes. The detection of WNV-seropositive equines in ten ranches and ILHV and SLEV-seropositive equines in fourteen ranches of two different sub-regions of Pantanal is strong evidence of widespread circulation of these flaviviruses in the region.  相似文献

Six insecticides and their eight combinations were tested for their efficacy against brinjal fruit and shoot borer, Leucinodes orbonalis. Endosulfan + deltamethrin (0.07%, 0.0025%) and endosulfan + fenvalerate (0.07% + 0.005%) were highly effective against fruit borer that recorded only 13.3% damage as compared to 69.8% in control. The other promising treatments which significantly reduced the fruit damage over the control were in the order: carbaryl + fenvalerate = dichlorvos + fenvalerate (14.9%) > malathion + fenvalerate (16.4%) > fenvalerate + deltamethrin (16.6%) > dichlorvos = carbaryl + deltamethrin = malathion = dichlorvos + deltamethrin = malathion + deltamethrin (18.3%) > endosulfan (20.0%) > carbaryl (21.6%) with mean percentage of damage 14.9, 16.4, 18.3, 20.0, 21.6 and 69.8%, respectively. Carbaryl was least effective, but its combinations with pyrethroids were proved superior over carbaryl alone. Cost - benefit ratio ranged from a minimum of 1: 5.10 (carbaryl) to a maximum of 1: 20.44 (fenvalerate). Dichlorvos + fenvalerate combination gave the highest yield of 263.45 q/ha, whereas carbaryl was least effective giving 225.7 q/ha. with a net gain of Rupees 42,443.00 (US$ 886.00) and 28,141.00 (US$ 587.49), respectively. The other treatments were intermediate between the two insecticide regimes. However, all the treatments were superior over the control which produced 113.58 q/ha with a net gain of Rupees 340.00 only.  相似文献

The spatial and temporal effects of the pyrethroid insecticide, deltamethrin, on populations of cereal aphids and their primary parasitoids and hyperparasitoids were investigated in a large scale field experiment in winter wheat. Four hectare plots were treated at the recommended field concentration or a reduced concentration representing one twentieth of field rate. A control plot was left unsprayed. Invertebrate populations were sampled at sites within a grid layout covering the whole plots, enabling the use of geostatistical analysis. Hymenopteran populations were monitored using transparent sticky traps and suction sampling. Aphid populations were recorded by visual counts. Monitoring continued for 36 days after treatment. The full rate deltamethrin treatment resulted in initial reductions of aphid populations by 78%. Primary parasitoid and hyperparasitoid populations were reduced in suction samples by 90% and 47% respectively, when corrected for control fluctuations. The reduced deltamethrin concentration caused reductions of aphid, primary parasitoid and hyperparasitoid populations of 40, 60 and 54% respectively. Aphid population recovery over the full rate plot occurred slowly and in a patchy manner following treatment. No significant reinvasion gradients were detected. Aphid population density recovered more rapidly in the reduced rate treatment; with initial evidence for increased densities at the plot centre. Significant patterns of reinvasion were initially detected for both groups of Hymenoptera in the full rate treatment, suggesting that reinvasion of the sprayed area was taking place from untreated surrounding reservoirs. It was concluded that experiments that examine pesticide impacts within small plots will lead to underestimates of effects on dispersive groups including parasitic Hymenoptera.  相似文献

The toxicity of the pyrethroid insecticide deltamethrin to a cereal aphid and a coccinellid beetle predator was assessed. Deltamethrin gave effective aphid control in winter wheat at dose-rates of 6.25, 3.13 and 1.56g a.i./ha. The direct exposure of adultCoccinella septempunctata L. (Coleoptera: Coccinellidae) to spray drops was estimated at a range of positions in a cereal crop canopy from volumetric analysis of fluorescent tracer deposits. These measurements were used to calculate exposure to deltamethrin at the three experimental dose-rates. Observations of coccinellid beetle distribution through a cereal crop canopy permitted a realistic range of direct contact doses tobe calculated and the toxic effects of these levels of exposure to be predicted from laboratory dose-response data. Estimated beetle mortalities from direct exposure were 19, 8 and 3% at the three experimental dose-rates.In situbioassays with adultC. septempunctata which exposed beetles continuously to deltamethrin residues on flag leaves, resulted in 100, 94 and 39% mortality respectively at these dose-rates during the 10 days after spray application. Additionalin situ bioassays exposed beetles to deltamethrin residues on flag leaves for 24 h and then transferred surviving beetles to the soil under the cereal crop canopy for a further 9 days. This resulted in 89, 69 and 29% beetle mortality respectively at the three dose-rates. Mortality predictions combining both direct contact and residual exposure were made for the three dose-rates.to determine the maximum impact of summer sprays of deltamethrin on adult coccinellid populations in cereals. These worst case predictions suggested that a reduction in dose-rate by as much as three quarters of the recommended application rate in UK cereals may be necessary to preserve approximately 60% of adultC. septempunctata in the crop over the 10 days after a deltamethrin spray application. The methodology described may be appropriate for estimating selective dose-rates for key enemies in a range of crops.  相似文献

Controlled drop (CDA) and conventional applications of difenzoquat to pot-grown Avena fatua were compared. With the recommended surfactant (0.5% v/v Agral), very low volume CDA was less effective than conventional spray application. However, addition of extra Agral or various blends of paraffinic oil with Agral and the surfactants Burtemul A2 or Burtemul P2 improved the effects of the CDA treatments. When difenzoquat was absent the additives were inactive against A. fatua. They had little direct effect on wheat and barley and did not substantially increase the phytotoxicity of difenzoquat to these crops. Oil/surfactant mixtures were less viscous than high concentrations of Agral, and so easier to spray. In a pot experiment, smaller (150 μm diameter) drops of difenzoquat solution were more active against A. fatua than larger (200 μm-300 μm) drops. Reduction of the spray volume within the range 40 litres/ha to 5 litres/ha also reduced phytotoxicity. An oil/surfactant additive improved the activity of all difenzoquat CDA treatments. There was slightly more effect at the lowest spray volume but interactions between additive and application treatments were not statistically significant. When simulated rain treatments were applied 2 h or 5 h after spraying, difenzoquat controlled drop application was much less phytotoxic than a conventional 150 litres/ha treatment. However, addition of an oil/surfactant mixture markedly improved the performance of CDA. When rain was withheld for 24 h the additive had relatively less effect. In the field an oil/surfactant mixture improved control of A. fatua by difenzoquat with both conventional and controlled drop treatments. The additive did not increase injury to the wheat crop. The oil/surfactant mixtures markedly improved the spreading and wetting properties of sprays. This reduced localised contact injury, which, it is suggested, improved uptake and translocation of difenzoquat.  相似文献

Three field populations of Cx. pipiens (L.) mosquitoes were collected from three different localities in Riyadh city. They were tested for developing resistance against commonly used insecticides to control mosquitoes in Riyadh. Two populations from Wadi Namar (WN1 and WN2) were highly resistant to deltamethrin (187.1- and 161.4-folds respectively). The field population from AL-Wadi district (AL-W) showed low resistance to lambda-cyhalothrin (3.8-folds) and moderate resistance to beta-cyfluthrin and bifenthrin (14- and 38.4-folds respectively). No resistance to fenitrothion was observed in WN1 population. Fenitrothion concentrations required to inhibit 50% of Acetylcholinesterase (AChE) activity in both WN1 population and the laboratory susceptible strain (S-LAB) were 0.073 and 0.078 ppm respectively. Piperonyl butoxide suppressed resistance to pyrethroid insecticides (>90%) in field populations indicating that oxidases and/or esterases play an important role in the reduction of pyrethroids toxicity. These results should be considered in the current mosquitoes control programs in Riyadh.  相似文献

BackgroundAedes albopictus is one of the most invasive species in the world as well as the important vector for mosquito-borne diseases such as dengue fever, chikungunya fever and zika virus disease. Chemical control of mosquitoes is an effective method to control mosquito-borne diseases, however, the wide and improper application of insecticides for vector control has led to serious resistance problems. At present, there have been many reports on the resistance to pyrethroid insecticides in vector mosquitoes including deltamethrin to Aedes albopictus. However, the fitness cost and vector competence of deltamethrin resistant Aedes albopictus remain unknown. To understand the impact of insecticide resistant mosquito is of great significance for the prevention and control mosquitoes and mosquito-borne diseases.Methodology/Principal findingsA laboratory resistant strain (Lab-R) of Aedes albopictus was established by deltamethrin insecticide selecting from the laboratory susceptible strain (Lab-S). The life table between the two strains were comparatively analyzed. The average development time of Lab-R and Lab-S in larvae was 9.7 days and 8.2 days (P < 0.005), and in pupae was 2.0 days and 1.8 days respectively (P > 0.05), indicating that deltamethrin resistance prolongs the larval development time of resistant mosquitoes. The average survival time of resistant adults was significantly shorter than that of susceptible adults, while the body weight of resistant female adults was significantly higher than that of the susceptible females. We also compared the vector competence for dengue virus type-2 (DENV-2) between the two strains via RT-qPCR. Considering the results of infection rate (IR) and virus load, there was no difference between the two strains during the early period of infection (4, 7, 10 day post infection (dpi)). However, in the later period of infection (14 dpi), IR and virus load in heads, salivary glands and ovaries of the resistant mosquitoes were significantly lower than those of the susceptible strain (IR of heads, salivary glands and ovaries: P < 0.05; virus load in heads and salivary glands: P < 0.05; virus load in ovaries: P < 0.001). And then, fourteen days after the DENV-2-infectious blood meal, females of the susceptible and resistant strains were allow to bite 5-day-old suckling mice. Both stains of mosquito can transmit DENV-2 to mice, but the onset of viremia was later in the mice biting by resistant group as well as lower virus copies in serum and brains, suggesting that the horizontal transmission of the resistant strain is lower than the susceptible strain. Meanwhile, we also detected IR of egg pools of the two strains on 14 dpi and found that the resistant strain were less capable of vertical transmission than susceptible mosquitoes. In addition, the average survival time of the resistant females infected with DENV-2 was 16 days, which was the shortest among the four groups of female mosquitoes, suggesting that deltamethrin resistance would shorten the life span of female Aedes albopictus infected with DENV-2.Conclusions/SignificanceAs Aedes albopictus developing high resistance to deltamethrin, the resistance prolonged the growth and development of larvae, shorten the life span of adults, as well as reduced the vector competence of resistant Aedes albopictus for DENV-2. It can be concluded that the resistance to deltamethrin in Aedes albopictus is a double-edged sword, which not only endow the mosquito survive under the pressure of insecticide, but also increase the fitness cost and decrease its vector competence. However, Aedes albopictus resistant to deltamethrin can still complete the external incubation period and transmit dengue virus, which remains a potential vector for dengue virus transmission and becomes a threat to public health. Therefore, we should pay high attention for the problem of insecticide resistance so that to better prevent and control mosquito-borne diseases.  相似文献

Alternative methods of pest control can and should be encouraged, especially those that consider the reality of smallholder family farmers. Here, we evaluated the potential of Agave americana (agave) extracts for the control of the aphid Brevicoryne brassicae in cabbage (Brassica oleracea L. var acephala) in the field and laboratory. The field experiments consisted of the evaluation of the proportion of dead aphids on cabbage plants after application of agave extracts. In the field, agave mixed with cow milk caused mortality above 80% and was the most effective extract. Agave mixed with water and agave mixed with ethanol elicited mortality above 60%. In the laboratory, we evaluated the mortality of aphids after the application of different concentrations of aqueous agave extracts; the commercial insecticide deltamethrin was included as positive control. Evaluation took place at 3, 6, 12, 24, 48 and 72 hr after applying the treatment. As expected, deltamethrin was the most effective treatment. However, agave extract at concentrations of 0.750 and 0.500 g/mL caused >70% mortality 3 hr after application. We conclude that A. americana extracts decreased aphid populations and is a promising alternative to the commercial insecticide against aphids in cabbage.  相似文献

Studies were carried out in Zimbabwe of the responses of tsetse to cattle treated with deltamethrin applied to the parts of the body where most tsetse were shown to land. Large proportions of Glossina pallidipes Austen (Diptera: Glossinidae) landed on the belly ( approximately 25%) and legs ( approximately 70%), particularly the front legs ( approximately 50%). Substantial proportions of Glossina morsitans morsitans Westwood landed on the legs ( approximately 50%) and belly (25%), with the remainder landing on the torso, particularly the flanks ( approximately 15%). Studies were made of the knockdown rate of wild, female G. pallidipes exposed to cattle treated with a 1% pour-on or 0.005% suspension concentrate of deltamethrin applied to the (a) whole body, (b) belly and legs, (c) legs, (d) front legs, (e) middle and lower front legs, or (f) lower front legs. The restricted treatments used 20%, 10%, 5%, 2% or 1% of the active ingredient applied in the whole-body treatments. There was a marked seasonal effect on the performance of all treatments. With the whole-body treatment, the persistence period (knockdown > 50%) ranged from approximately 10 days during the hot, wet season (mean daily temperature > 30 degrees C) to approximately 20 days during the cool, dry season (< 22 degrees C). Restricting the application of insecticide reduced the seasonal persistence periods to approximately 10-15 days if only the legs and belly were treated, approximately 5-15 days if only the legs were treated and < 5 days for the more restricted treatments. The restricted application did not affect the landing distribution of tsetse or the duration of landing bouts (mean = 30 s). The results suggest that more cost-effective control of tsetse could be achieved by applying insecticide to the belly and legs of cattle at 2-week intervals, rather than using the current practice of treating the whole body of each animal at monthly intervals. This would cut the cost of insecticide by 40%, improve efficacy by 27% and reduce the threats to non-target organisms and the enzootic stability of tick-borne diseases.  相似文献

在害虫治理中, 在消费或贮藏粮食加工产品的建筑设施或场所进行害虫防治需要将杀虫剂施用在各种基质表面上。为了测定不同基质表面上杀虫剂的药效, 将四纹豆象Callosobruchus maculatus (F.)成虫接触田间推荐剂量的阿维菌素、 溴氰菊酯和毒死蜱。结果表明: 施用在玻璃、 瓷砖、 塑料和纸盘表面上, 阿维菌素对四纹豆象成虫的致死率分别为63.33%, 22.41%, 12.9%和11.9%, 而溴氰菊酯在这4种基质表面上对四纹豆象成虫的致死率分别为 55%, 44.2%, 41.3%和 37.4%。在所有基质表面上接触毒死蜱, 四纹豆象成虫的死亡率均为100%。对数据进行的Probit 分析表明, 毒死蜱制剂在玻璃、 瓷砖、 塑料和纸盘上对四纹豆象成虫 的LC50 值分别为 8.66, 13.6, 29.16和 56.5 μg/mL, 阿维菌素制剂的相应数值分别为119.4, 446.2, 774.2 和 836.4 μg/mL, 溴氰菊酯制剂的相应数值分别为 1 008, 1 131, 1 210和 1 336 μg/mL。据此推断, 毒死蜱对四纹豆象的毒性最强, 且在玻璃、 瓷砖、 塑料和纸盘表面上的毒性依次降低。  相似文献

Hypocryphalus mangiferae Stebbing is one of the most destructive insect pests of mango trees and is found to be associated with the transmission of causal organisms of mango sudden death disease. The present study was carried out to evaluate the toxicity of deltamethrin, bifenthrin, chlorpyrifos, emamectin benzoate, imidacloprid and spinosad in laboratory and field trials. Bioassay results showed that the toxicity of chlorpyrifos was significantly higher than deltamethrin but similar to bifenthrin. Deltamethrin and bifenthrin toxicity, however, increased significantly (P < 0.01) from day 1 to day 3. Spinosad was the least toxic compound while emamectin was the most toxic among new chemical insecticides tested, but its toxicity increased significantly from day 1 to day 5. Comparison of the efficacies of the insecticides using lethal times to produce 50% mortality (LT50) and 90% mortality (LT90) showed that the relative potencies of chlorpyrifos, emamectin, imidacloprid and spinosad were greater than bifenthrin and deltamethrin. The results of field trials showed the highest number of beetles emerged from the control twigs while significantly fewer beetles emerged from the twigs treated with bifenthrin (P < 0.05), which accounted for 12% for bifenthrin compared to that of the control. Deltamethrin and other pyrethroids have been extensively used in Argentina since 1980, for the chemical control of Triatoma infestans Klug (Hemiptera: Reduviidae). Recently, resistance to deltamethrin was detected in field populations by the survival of bugs exposed by topical application to the diagnostic dose estimated on the CIPEIN susceptible strain. Results of the current study showed low resistant ratios (RRs) to deltamethrin for the resistant populations (RR ranged from 2.0 for San Luis colony to 7.9 for Salta colony). Biochemical studies were made on the most resistant colony (Salta) and the susceptible strain (CIPEIN), in order to establish the importance of degradative mechanisms as a cause of the detected resistance. Esterase activity was measured on 3 days old first instars through phenylthioacetate and a-naphtyl acetate activities. The results showed a significant difference in no cholinesterase esterase activity from susceptible (7.6 +/- 0,7 micro M S./i.min.) and Salta resistant colony (9.5 +/- 0.8 microM S./i.min.). Cytochrome p450 mono-oxygenase (p450) activity was measured on individual insects through ethoxycoumarine deethylase (ECOD) activity using a fluorescence microplate reader. The dependence of ECOD activity on age and body region of the nymphs, and pH and time of incubation were studied in order to optimize the measurement. As a result, comparative studies were performed on abdomens of 2 days old first instars at pH 7.2 and 4 h incubation time. ECOD activity of first nymphs was significantly lower in the susceptible colony (61.3 +/- 9.08 pg ECOD/ insect) than in the resistant one (108.1+/- 5.7 pg ECOD/ insect). These results suggest that degradative esterases (no-cholinesterase) and mono-oxygenases cytochrome p450, play an important role in the resistance to deltamethrin in Salta colony from Argentina.  相似文献

The effect of Decis (deltamethrin as active ingredient) on callus tissue of Solanum tuberosum L. cv. Désirée was studied. Decis is an agrochemical currently used on field grown potato plants to control the Colorado beetle, a potato pest. Deltamethrin added to the culture medium interferes with the behaviour of callus tissue. After 5 h of culture, the level of total proteins was higher in treated tissue than in the control, and SDS-PAGE showed that deltamethrin promoted the increase of some soluble proteins. After 24 h and 14 days of culture, the level of total proteins became similar in both treated and control material. This similarity between control and treated tissues, after 14 days of culture, occurred as a result of the treatment with deltamethrin which caused a decrease of soluble proteins, but an increase in insoluble proteins whereas the opposite was observed for control callus. SDS-PAGE of both soluble and insoluble proteins showed that only quantitative dissimilarities occurred. This longer treatment also increased the chlorophyll content. Ultrastructural study of the cells revealed that tissue callus cultured for 14 days in the presence of deltamethrin had plastids containing a more developed membranous system, with a higher number of grana and with more compartments than in control cells. Deltamethrin also promoted the abundance of vesicles associated with dictyosomes. This response of the callus tissue to Decis added to the callus medium parallels the behaviour of potato plants treated with this agrochemical under field conditions (Fidalgo, Santos & Salema, 1993).  相似文献
