Although it is well established that carbon reserves contributeto shoot regrowth of leguminous forage species, little informationis available on nitrogen reserves except in Medicaqo sativaL. and Trifolium subterraneum L. In this study, reserves werelabelled with 15N to demonstrate the mobilization of endogenousnitrogen from roots and stolons to regrowing leaves and newstolons during 24 d of regrowth in white clover (Thfolium repensL.). About 55% and 70%, respectively, of the nitrogen contentsof these organs were mobilized to support the regrowth of leaves.During the first 6 d, nitrogen in regrowing leaves came mainlyfrom N reserves of organs remaining after defoliation. Afterthese first 6 d of regrowth, most of the shoot nitrogen wasderived from exogenous nitrogen taken up while the contributionof nitrogen reserves decreased. After defoliation, the buffer-solubleprotein content of roots and stolons decreased by 32% duringthe first 6 d of regrowth. To identify putative vegetative storageproteins, soluble proteins were separated using SDS-PAGE ortwo-dimensional electrophoresis. One protein of 17.3 kDa instolons and two proteins of 15 kDa in roots seemed to behaveas vegetative storage proteins. These three polypeptides, initiallyfound at high concentrations, decreased in relative abundanceto a large extent during early regrowth and then were accumulatedagain in roots and stolons once normal growth was re-established. Nodul{macron}ted alfalfa plants were grown hydroponically. Inorder to quantify N2 fixation and remobilization of N reservesduring regrowth the plants were pulse-chase-labelled with 15N.Starch and ethanol-soluble sugar contents were analysed to examinechanges associated with those of N compounds. Shoot removalcaused a severe decline in N2 fixation and starch reserves within6 d after cutting. The tap root was the major storage site formetabolizable carbohydrate compounds used for regrowth; initiallyits starch content decreased and after 14 d started to recoverreaching 50% of the initial value on day 24. Recovery of N2fixation followed the same pattern as shoot regrowth. Afteran initial decline during the first 10 d following shoot removal,the N2 fixation, leaf area and shoot dry weight increased sorapidly that their levels on day 24 exceeded initial values.Distribution of 15N within the plant clearly showed that a significantamount of endogenous nitrogen in the roots was used by regrowingshoots. The greatest use of N reserves (about 80% of N incrementin the regrowing shoot) occurred during the first 10 d and thencompensated for the low N2 fixation. The distribution of N derivedeither from fixation or from reserves of source organs (taproots and lateral roots) clearly showed that shoots are thestronger sink for nitrogen during regrowth. In non-defoliatedplants, the tap roots and stems were weak sinks for N from reserves.By contrast, relative distribution within the plant of N assimilatedin nodules was unaffected by defoliation treatment. Plants ofLolium perenneandFestuca rubrawere grown in sand culturereceiving all nutrients as a complete nutrient solution containing1.5 mMNH4NO3, and subjected to one of three defoliation treatments:undefoliated, defoliated on one occasion, or defoliated weekly.15Nlabelling was used to determine the rate of N uptake, allowingthe amount of N remobilized from storage for the growth of thetwo youngest leaves (subsequently referred to as ‘newleaves’) growing over a 14 d period after defoliationto be calculated. The total plant N uptake by both species wasreduced, compared with undefoliated plants, by both a singleand repeated defoliation, although neither caused complete inhibitionof uptake. Regularly defoliatedL. perennehad a greater reductionin root mass, concomitant with a greater increase in N uptakeper g root than did regularly defoliatedF. rubra. In both species,the amount of N derived from uptake recovered in the new leaveswas unaffected by the frequency of defoliation. BothL. perenneandF.rubramobilized nitrogen to the new leaves after a single defoliation,mobilization being sufficient to supply 50 and 41%, respectively,of the total nitrogen requirement. In regularly defoliated plants,no significant nitrogen was mobilized to the new leaves inL.perenne, and only a small amount was mobilized inF. rubra. Plantsachieved greater leaf regrowth when only defoliated once. Weconclude that increasing the frequency of defoliation of bothL.perenneandF. rubrahad little effect on the uptake of nitrogenby roots which was subsequently supplied to new leaves, butdepleted their capacity for nitrogen remobilization, resultingin a reduction in the rate of growth of new leaves. This study was conducted with Lolium temulentum, Festuca pratensis,and the two hybrids L. multiflorum x F. pratensis ‘Elmet’and L. perenne x F. pratensis ‘Prior’. In a comparisonof various durations (7–42 d) of pretreatment at 4 or7 °C the highest yield of microspore-derived callus of L.temulentum was obtained after pretreatment of spikes at 7 °Cfor 28 d, conditions which also proved optimal for panicle pretreatmentwith F. pratensis. For ‘Elmet’, durations of 21–42d were optimal, and for ‘Prior’ the responses tendedto decline with increasing duration. Relationships between nitrate (NO-3) supply, uptake and assimilation,water uptake and the rate of mobilization of seed reserves wereexamined for the five main temperate cereals prior to emergencefrom the substrate. For all species, 21 d after sowing (DAS),residual seed dry weight (d.wt) decreased while shoot plus rootd.wt increased (15–30%) with increased applied NO-3concentrationfrom 0 to 5–20 mM . Nitrogen (N) uptake and assimilationwere as great with addition of 5 mM ammonium (NH+4) or 5 mMNO-3but NH+4did not affect the rate of mobilization of seedreserves. Chloride (Cl-) was similar to NO-3in its effect onmobilization of seed reserves of barley (Hordeum vulgare L.).Increased rate of mobilization of seed reserves with additionalNO-3or Cl-was associated with increases in shoot, root and residualseed anion content, total seedling water and residual seed watercontent (% water) 21 DAS. Addition of NH+4did not affect totalseedling water or residual seed water content. For barley suppliedwith different concentrations of NO-3or mannitol, the rate ofmobilization of seed reserves was positively correlated (r >0.95)with total seedling water and residual seed water content. Therate of mobilization of seed reserves of barley was greaterfor high N content seed than for low N content seed. Seed watercontent was greater for high N seed than for low N seed, 2 DAS.Additional NO-3did not affect total seedling water or residualseed water content until 10–14 DAS. The effects of seedN and NO-3on mobilization of seed reserves were detected 10and 14 DAS, respectively. It is proposed that the increasedrate of mobilization of seed reserves of temperate cereals withadditional NO-3is due to increased water uptake by the seedlingwhile the seed N effect is due to increased water uptake bythe seed directly. Plants of Lolium perenne L. cv. S23 were grown in sand culturesupplied with either ammonium (NH4+) or nitrate (NO3)in an otherwise complete nutrient solution at 12°C or 20°C.Three weeks after germination, plants were clipped weekly tosimulate grazing. After 10 weeks growth all nitrogen (N) wassupplied enriched with 15N to quantify the effects of form ofN supply and temperature on the relative ability of currentroot uptake and remobilization to supply N for laminae regrowth. The form of N supply had no effect on the dry matter partitioning,while at 20°C more dry weight was allocated to laminae regrowthand less to the remaining plant material. The current root uptakeof N, which subsequently appeared in the laminae regrowth, wassimilar for plants supplied with NH4+ or NO3, and bothwere equally reduced at the lower temperature of growth. Remobilizationof N to laminae regrowth was greater for plants receiving NH4+than NO3; remobilization with either form of N supplywas reduced at the lower temperature of growth. Remobilizationwas reduced to a lesser extent at 12°C than current rootuptake. It was concluded that remobilization became relativelymore important in supplying N for regrowth of laminae at lowertemperatures. Nitrogen remobilization from roots and pseudostems during regrowthof Lolium perenne L. was studied in miniswards grown with contrastinglevels of (NH4)2SO4 in solution culture. Growth with a highN supply (5.0 mol m–3) increased theweight of leaf laminae recovered at each of five weekly clippings,and decreased the proportion of photosynthate used for rootgrowth. Clipped plants growing in a steady-state were suppliedwith 15N for 48 h and the recovery of labelled N in laminaemeasured after five weekly cuts. Recovery of labelled N in thelaminae from the second clipping onwards was derived only fromremobilization of N from roots and pseudostem. Miniswards grownwith low N (0.5 mol m–3) relied moreupon remobilization of N for lamina growth than did high N plants.Thus after 14 d 20% of lamina N was labelled in low N plantsbut only 3% was labelled in the high N treatment. Thereafter,N remobilization declined until at the final clipping after35 d, labelled N represented only 4% and 1 % of the lamina Nin the low and high N plants. When plants were not clipped beforethe labelling period, they took up more 15N if grown with highN than cut plants. Thereafter, the remobilization of N followeda similar pattern as in the cut plants. Exponential models were used to calculate the rate of N transferfrom roots and pseudostem to laminae. When grown with low N,the half-life of remobilization was 1.56 weeks. High N miniswardshad an initial rapid remobilization with a half-life of 0.66weeks, and a slower phase with a half-life of 2.98 weeks. The aim of this study was to test the effect of oxygen partialpressure as a possible limiting factor of nitrogen fixationfollowing defoliation. The response of nitrogenase activity(C2H2-reduction) of defoliated and undefoliated white and redclover plants (Trifolium repens L. and Trifolium pratense L.)to either 19 kPa oxygen or 55 kPa oxygen was investigated. Priorto defoliation, white clover plants were grown for five weeksin a growth chamber, and red clover plants for 7 or 11 weeksin a glasshouse. The results included measurements of 16N2-uptake. Increasing oxygen partial pressure from 19 to 55 kPa severelyrestricted nitrogenase activity of undefoliated white cloverplants; however, 2 h after complete defoliation, the same treatmentcaused a significant increase. A fivefold increase in nitrogenaseactivity upon exposure to the elevated oxygen partial pressurewas found at the end of a 24 h period. This beneficial effectdecreased gradually from 1 to 5 d after defoliation. The responseof recently defoliated red clover plants to 55 kPa oxygen partialpressure was similar to that of white clover, independentlyof plant age. The gradual recovery of nitrogenase activity duringthree weeks of regrowth was associated with a simultaneous changein the response to increased oxygen partial pressure, leadingagain to the response of undefoliated plants. These data suggested that lack of oxygen at the site of nitrogenfixation, resulting from a dramatic increase in oxygen-diffusionresistance, is the main factor limiting nitrogenase activityfollowing defoliation. The regrowth pattern in Lolium multiflorum was analysed in relationto carbohydrate levels and developmental characteristics, usingtwo contrasting populations. Liscate (a natural population collectedin Northern Italy) and the cultivar S 22, the former havingmore erect tillers, longer leaves and a lower rate of leaf appearance.The plants were grown in 16 h photoperiods, at 15/10 °C.NAR was lower in S. 22, but because LAR was higher as a resultof the greater SLA, the RGR was similar in Liscate and S. 22Starch levels were also similar in the two populations, butthe level of WSC was higher in Liscate than in S 22. When thetwo populations were defoliated to the same extent, total leafregrowth and rate of leaf elongation were greater in Liscate,but its tillering was more inhibited. Although the rates ofleaf regrowth were eventually similar in both populations, theywere initially lower in S. 22, leading to a higher total regrowthin Liscate. In both populations, WSC levels in the roots andstubble decreased after defoliation, but starch levels remainedunchanged. During regrowth in the dark, starch levels also decreased,but only when most of the WSC in the stubble was depleted. Lolium temulentum seedlings were grown on a nutrient mediumcontaining NH4NO2 at 0, 0·1, 0·5, 1·0 and4·3 mmoll–1 as the sole N source. Relative andabsolute extension rates, maximal leaf size, duration of extensiongrowth, rate of leaf appearance and plastochron index were determinedfrom the parameters of Richards functions fitted to lengthsof laminae measured at intervals after sowing. The final lengthof leaf I was relatively insensitive to N whereas mean relativeextension rate was increased and duration of growth decreasedwith increasing NH4NO2 concentration. Leaves 2 and 3 enlargedprogressively with N at concentrations up to 1·0 mmoll–1but were unresponsive thereafter. There was no significant correlationbetween final length and mean relative extension rate for leaves1 to 3. Leaves 4 to 6 continued to show increasing length beyond1·0 mmoll–1 N and final length was significantlycorrelated with mean relative extension rate. Increasing N increasedthe rate of leaf appearance by decreasing the duration of leafextension and plastochron. These results are discussed in relationto the control of leaf and N turnover. Seed samples of Lolium temulentum L. were harvested at variousstages of maturity and subjected to successive cold- and hot-waterextractions. These extracts were then fractionated by gel-filtrationand the changes in the carbohydrate elution pattern with increasingseed maturity were characterized. Extracts of seeds from earlyharvests contained only free sugars, but the mean chain-lengthincreased with maturity. The hot-water fraction contained nosalient features until 17 days after anthesis but, after thispoint, a completely excluded, high-molecular-weight polysaccharideaccumulated. Infiltration studies using I4C-sucrose and detached seeds showedthat sugar was readily incorporated into polysaccharide, butthat the degree of uptake was directly proportional to the developmentalstage of the seed at the time of detachment. The contribution of nitrogen reserves to regrowth following defoliation was studied in white clover plants (Trifolium repens cv. Huia). This was found to be closely linked to the morphological pattern of development of the aerial parts during the same period. Low temperature (6 degrees C) and short day exposure (8 h photoperiod) were used to induce dwarf development, i.e. to increase branching rate and to enhance new sites of leaf production during a period of regrowth. Treated plants exhibited a large reduction in leaf area and a large increase in leaf pool size for the first 10 d of a subsequent regrowth under standard culture conditions (16 h daylight; 22/18 degrees C day/night). The contribution of nitrogen from storage compounds in organs remaining after defoliation (sources) to regrowing tissues (sinks) was assessed by 15N pulse-chase labelling during regrowth following shoot removal. The mobilization of nitrogen reserves from storage tissues of regrowing clover was closely linked to the pattern of differentiation of the newly developed organs. It appeared that regrowth was supported less by endogenous N for the first 10 d after defoliation in treated plants, compared with control plants grown continuously in standard conditions. It is assumed that dwarf plants exhibit a lower dependence upon the mobilization of soluble proteins previously accumulated in roots and uncut stolons. The relationship between leaf development rate and N-uptake recovery following defoliation is discussed.  相似文献

Single plants of white clover (Trifolium repens) were establishedfrom stolon cuttings rooted in acid-washed silver sand. Allplants were inoculated with Rhizobium trifolii, and receivednutrient solution containing 0·5 mg 15N as either ammoniumor nitrate weekly for 12 weeks (i.e. 6 mg 15N in total). Plantswere then leniently defoliated or left intact, and the labelledN supply was replaced with unlabelled N. Lenient defoliationremoved fully expanded leaves only, leaving immature leaveswhich accounted for 50–55% of the total; growing pointnumbers were not reduced. Nodules, leaves and growing pointswere counted over the following 21 d period, and d. wts, N contents,and 15N enrichments of individual plant organs were determined. Defoliated plants had fewer nodules, but numbers of growingpoints were unaffected by defoliation. The rates of both leafemergence and expansion were accelerated in defoliated plants;in consequence the number of young leaves remained less thanin intact plants until day 21. Total dry matter (DM) and N accumulationwere less in defoliated plants, and a greater proportion oftotal plant DM was invested in roots. About 97 % of plant totalN was derived from fixed atmospheric N, but there was incompletemixing of fixed and mineral N within the plant. Relatively moremineral N was incorporated into roots, whereas there was relativelymore fixed N in nodules. There was isotopic evidence that Nwas remobilized from root and stolon tissue for leaf regrowthafter defoliation; approximately 2 % of plant N turned overdaily in the 7-d period after defoliation, and this contributedabout 50% of the N increment in leaf tissue. Lolium perenne, a main component species in managed grassland, is well adapted to defoliation, fertilization, and regrowth cycles; and hence, to changes in the assimilatory carbon source‐sink ratio. In the Swiss Free Air CO2 Enrichment experiment the source‐sink ratio is (i) increased by elevated partial pressure of CO2 (pCO2), (ii) decreased by enhanced carbon use under high N fertilization, and (iii) gradually increased during regrowth after defoliation. Since sucrose synthesis plays a central role in leaf carbohydrate metabolism in this fructan‐accumulating species, we investigated how sucrose‐phosphate synthase (SPS) responds to the differing assimilatory carbon fluxes and source‐sink ratios in the field. Assimilatory carbon flux, as estimated by leaf gas exchange, strongly depended on pCO2. Surprisingly, the SPS content per leaf area did not increase with pCO2, but increased with N fertilization. During later regrowth, when a dense canopy had formed, the SPS content decreased; in particular, SPS was decreased at high N under elevated pCO2. Further, the higher assimilatory carbon flux through SPS at elevated pCO2 was accompanied by a higher activation state of SPS. The SPS content correlated very strongly with the ratio of free sucrose to free amino acid in leaves, which represents the carbon source‐sink ratio. Hence, SPS content in L. perenne appears to be regulated by the current, strongly nitrogen‐dependent, source‐sink relation.  相似文献

The various pathways of pollen development were investigatedin cultured anthers of Lolium temulentum, Festuca pratensisand the L. multiflorum x F. pratensis hybrid ‘Elmet’.In all three, development from the vegetative cell was the predominantpathway of pollen callus development. However, there were characteristicdifferences in the behaviour of the generative cell. In L. temulentumit remained attached to the pollen wall and degenerated, whereasin F. pratensis it divided. In ‘Elmet’ it detachedfrom the pollen wall and remained undivided. Both polarizedand unpolarized partitioned calluses were observed. Developmentof the fusion product of the vegetative and generative nucleiwere also observed in anthers of L. temulentum. Anomalous grainswere not found to be major source of pollen calluses. Sections of anthers of L. temulentum were used to investigatethe origin of S pollen grains, the small pale-staining grainswhich denote pollen dimorphism. Such grains form out of contactwith the tapetum and are therefore determined before or duringmeiosis (i.e. before harvest of anthers for culture). Sectionswere also used to demonstrate the influence of the durationof pretreatment on the development of the middle layer of theanther wall. Lolium temulentum plants were grown at 20 °C, under an 8-hdaylength, in a controlled-environment chamber, and the kineticsof leaf expansion were observed by measuring the movement ofan optical grid attached to the fourth leaf. The leaf emerged23–24 d after sowing and was fully expanded 9–10d later. Extension rate was maximal between the second and fifthdays after emergence and declined markedly thereafter. Duringthe rapid growth phase the rate of elongation exhibited a distinctdiurnal rhythm, fluctuating between 1.9 to 2.3 mm h–1in the light period, and 1.3 to 1.7 mm h–1 in the dark.A circadian oscillation with a period of about 27 h was observedin leaves elongating in continuous darkness. When plants weretransferred to 5 °C soon after emergence of the fourth leafthere was an immediate reduction in rate of growth to about22 per cent of the rate at 20 °C: the Q10 for the mean elongationrate in the range 20–5 °C was 3.7. When plants weretransferred from 20 to 2 °C at fourth leaf emergence, meanextension rate declined to less than 5 per cent, correspondingto a Q10 in the range 5–2 °C of more than 300. Furthermore,growth at 2 °C was confined almost entirely to the darkphase of the photoperiod cycle. The responsive tissue was shownto be a small area of expanding leafless than 1.5 cm above theshoot apex and the possible mechanisms underlying low temperatureeffects in this region are discussed. An experiment was designed to evaluate the role of N and C reserves on regrowth of Lolium perenne cv. Bravo following defoliation. By using two nitrogen fertilization levels together with three photoperiodic conditions, plants with variable contents of water-soluble carbohydrates (43-216 mg g-1 DW in stubble) and contrasting amounts of nitrogen (7-49 mg g-1 DW) were obtained. Plants were severely defoliated and regrowth was followed for 28 d under the same environmental conditions. The yield of leaf dry matter at the end of the regrowth period was not related to the initial level of carbohydrate reserves. However, levels of fructan in leaf sheaths and in elongating leaf bases strongly influenced the shoot yield during the first 2 d following defoliation. Fructan exohydrolase activity increased 2-3-fold in sheaths and 3.5-5-fold in elongation leaf bases, suggesting that not only fructans from sheaths but also fructans from immature cells may be used as substrates for growth. In contrast, no direct relationship was found between shoot production and nitrogen or soluble protein accumulation in source organs during early regrowth. A significant correlation existed with the initial amount of soluble proteins in sheaths and in elongating leaf bases after only 6 d of regrowth.  相似文献

An experiment was designed to study the role of N and C reserves on regrowth of the shoots following defoliation of forage species. Starch and N accumulation in root and crown tissue of nonnodulated Medicago sativa L. were modified during regrowth by applying different levels of N and different cutting heights. Plants were obtained with similar crown and root dry weights, but having either low starch and high tissue N or high starch and low tissue N. The plants were then submitted to a second defoliation and supplied with optimal N nutrition, and N flow from reserve was quantified using pulse-chase 15N labeling. Maximum yields following the second regrowth were obtained from those plants having a high tissue N, despite their low level of nonstructural carbohydrate. When N in the roots and crown exceeded 5 mg N plant-1 at the beginning of regrowth, about 68% was translocated to regrowing shoots. Highly significant correlations were also found between the amounts of N available in roots and crown at the beginning of regrowth and (a) the amount of N that was mobilized to new tissues, (b) the amount of N taken up during the regrowth period, and (c) the final shoot yield after 24 d of regrowth. No similar correlations were found for plants that varied in their initial starch content of roots and crown. It is suggested that N reserves were used mainly during the first 10 d after defoliation, and that the resulting aerial growth during this period should be sufficient to restore N2 fixation and/or N uptake to levels equal to those prior to defoliation. These data emphasize (a) the importance of root N reserves in initiating and sustaining new shoot growth, and (b) the need for a re-evaluation of the contribution of C reserves to shoot regrowth.  相似文献
