The synthesis of bioactive lipids in three species of lower mycelial fungi of the genus Pilaira, the family Pilobolaceae, has been studied. The pigmentation of these fungi was found to be determined by the presence of beta-carotene in amounts of 14.8 (P. moreaui), 115.7 (P. caucasica), and 312.9 (P. anomala) microg/g. The fatty acid profiles of the fungi, which are distinguished by the presence of up to 50% essential fatty acids, is typical of zygomycetes of the order Mucorales. The fungi grown in submerged cultures showed a correlation of the unsaturation indices of their fatty acids and the contents of beta-carotene.  相似文献

Summary The lignin-degrading fungiPhanerochaete chrysosporium, P. sordida, Trametes hirsuta, andCeriporiopsis subvermispora were evaluated for their ability to decrease the concentration of pentachlorophenol (PCP) and to cause dry weight loss in PCP-treated wood. Hardwood and softwood materials from PCP-treated ammunition boxes that were chipped to pass a 3.8-cm screen were used. All four fungi caused significant weight losses and decreases in the PCP concentration. The largest PCP decrease (84% in 4 weeks) was caused byT. hirsuta, and the smallest decrease was caused byC. subvermispora (37% in 4 weeks). After 4 weeks, the fate of spiked14C[PCP] in softwood chips inoculated withT. hirsuta was as follows: 27% was mineralized, 42.5% was non-extractable and bound to the chips, 23.5% was associated with fungal hyphae, and 6% was organic-extractable. Decreases of PCP byP. chrysosporium andP. sordida averaged 59% and 57%, respectively. PCP decreases caused byPhanerochaete spp. were not significantly affected by wood type or sterilization and were primarily due to methylation of PCP that resulted in accumulation of pentachloroanisole. Softwood weight losses caused byT. hirsuta, P. chrysosporium andC. subvermispora were respectively, 24, 6.5, and 17%, after 4 weeks. These weight losses are comparable to reported weight losses by these organisms in non-treated softwood. Nutrient supplementation significantly increased weight loss but not percentage decrease of PCP. The results of this research demonstrate the potential for using lignin-degrading fungi to destroy PCP-treated wood.  相似文献

【背景】植物内生真菌对宿主植物促生长、抗旱和增强抗病能力等方面有着重大的研究和利用价值,尤其对兰科植物的生长起到重要的作用。【目的】通过对掌裂兰根部内生真菌和根际土真菌多样性进行系统分析,掌握掌裂兰根部内生真菌与根际土真菌群落结构,为进一步探究掌裂兰植物与真菌共生规律提供参考。【方法】采用Illumina MiSeq高通量测序技术分析掌裂兰根部内生真菌和根际土真菌多样性。【结果】掌裂兰根部内生真菌隶属于7门89属,优势菌属为瘤菌根菌属(Epulorhiza)(16.93%)、头梗霉属(Cephaliophora)(10.41%)、酵母属(Saccharomyces)(5.73%)、角担菌属(Ceratobasidium)(5.32%)和镰刀菌属(Fusarium)(5.12%),其中Epulorhiza和Ceratobasidium为兰科植物菌根真菌;根际土真菌隶属于11门269属,优势菌属为镰刀菌属(Fusarium)(8.09%)、丛赤壳属(Neonectria)(6.79%)、Plectosphaerella (3.39%)和被孢霉属(Mortierella)(3.01%)。通过...  相似文献

The non-polar lipid content and fatty acid (FA) composition of 11 mushroom species of the family Boletaceae were determined. The non-polar lipid content ranged from 2.0 (Leccinum aurantiacum and Boletus erythropus) to 5.4 % (w/w) d.w. (Suillus grevillei) with an average value of 2.9 %. More than 25 different FAs were found in the mushroom lipids. Unsaturated FAs, mainly linoleic and oleic acids, accounted for about 83 % of the total FAs, while palmitic acid was the main saturated FA. Some FAs are identified for the first time in Boletaceae and in higher Basidiomycetes (cis-11,12-methyleneoctadecanoic acid, 7-cis,10-cis hexadecadienoic) or in fungi (cis-11,12-methyleneoctadecanoic acid). There were significant differences (P < 0.05) in the contents of specific FAs between mushroom species.  相似文献

为了探究桐花树内生真菌在抑菌方面的价值,该文以内生真菌发酵产物的抑菌作用为评价指标筛选活性菌株,采用生物活性跟踪方法结合多种色谱技术分离活性菌株的化学成分,通过波谱与文献数据比对鉴定单体化合物结构,并利用微孔板法测定单体化合物的抑菌活性。结果表明:(1)从桐花树分离得到的16株内生真菌分属2纲7目10科10属,镰刀菌属(Fusarium)为优势菌属。内生真菌GXIMD02029和GXIMD02039的发酵产物对枯草芽孢杆菌、表皮葡萄球菌、耐甲氧西林金黄色葡萄球菌、藤黄微球菌、粘性放线菌和金黄色葡萄球菌有不同程度的抑制作用,GXIMD02038发酵产物对耐甲氧西林金黄色葡萄球菌、藤黄微球菌和金黄色葡萄球菌有抑制作用。(2)7个化合物从内生真菌Phomopsis sp. GXIMD02029中被分离并鉴定为(15R)-acetoxydothiorelone A(1)、cytosporone B(2)、pestalotiopsone H(3)、pestalotiopsone B(4)、4-Hydroxybenzaldehyde(5)、p-Hydroxybenzoic acid(6)、N-(2-phenylethyl)acetamide(7)。(3)化合物1和2有不同程度的抑菌作用,化合物1对枯草芽孢杆菌、表皮葡萄球菌、耐甲氧西林金黄色葡萄球菌的MIC值为16.25 SymbolmA@ g·mL-1,对藤黄微球菌和粘性放线菌的MIC值为7.812 5 SymbolmA@ g·mL-1,对金黄色葡萄球菌的MIC值为31.25 SymbolmA@ g·mL-1。化合物2对藤黄微球菌的MIC值为62.5 SymbolmA@ g·mL-1,对枯草芽孢杆菌、表皮葡萄球菌、耐甲氧西林金黄色葡萄球菌、粘性放线菌的MIC值为125 SymbolmA@ g·mL-1,对金黄色葡萄球菌的MIC值为250 SymbolmA@ g·mL-1。该文筛选了3株活性菌株,首次报道化合物1具有抗菌活性,为桐花树内生真菌在抗菌价值方面提供了依据。  相似文献

The taxonomic composition of marine mycelial fungi was determined in the interstitial habitats of the Vostok Bay littoral (Peter the Great Bay, the Sea of Japan). A total of 39 species of ascomycetes and anamorphic fungi were detected and identified. The predominant species of the intertidal zone were Corollospora maritima, C. lacera, Carbosphaerella leptosphaerioides, Arenariomyces trifurcatus (Ascomycota), Alternaria alternata, Scolecobasidium arenarium, and Zalerion maritimum (anamorphic fungi). This survey is concerned only with filamentous fungi living in the soil layer. The observed diversity of soil fungi largely depends on the method used and the numbers of isolates obtained. Particle-plating usually yields higher numbers of taxa than dilution plating. The Centraalbureau voor Schimmelcultures (CBS) preserves a great diversity of soil fungi. The CBS database contained 2,210 species of soil fungi in 2001, an estimated 70% of the known species available in culture. Thus, the current estimate for described culturable soil fungi is approximately 3,150 species, many of which have a cosmopolitan distribution. Adding the ca. 150 spp. of nonculturable Glomerales results in 3,300 species of currently known soil fungi. Molecular studies in such groups as Fusarium, Trichoderma, Penicillium, and Aspergillus are finding a number of more narrowly distributed cryptic species. Thus the number of species of soil fungi is expected to be considerably higher than the 3300 species currently known.  相似文献

【目的】分离并鉴定具有较好除草活性的植物病原真菌菌株,进一步分离活性化合物,为开发新型生物源除草剂奠定一定的基础。【方法】采用固体培养基接种法分离纯化植物病原真菌,通过形态学特征观察和5.8S rDNA测序鉴定目标菌株;在活性筛选追踪下,对活性组分进行追踪分离及纯化,经波谱分析确定活性单体化合物结构;采用培养皿生物分析法测定活性单体化合物的除草活性及对常见作物的安全性。【结果】茶叶致病菌CY-H具有较好的除草活性,其发酵液对稗草和反枝苋根的抑制率分别为94.6%和77.3%。CY-H被鉴定为间座壳属菌(Diaporthe sp.)。从CY-H菌中分离得到单体化合物CY1被鉴定为cytosporaphenones C。在供试浓度为100μg/mL时,CY1具有较好的抑制反枝苋根的活性,抑制率为57.1%,且其对小麦和油菜的安全性较好,抑制率均在20%左右。【结论】茶叶致病菌CY-H具有开发成微生物源除草剂的潜力。  相似文献

【背景】松树的内生真菌会影响红脂大小蠹及其伴生真菌的生长及扩散,从而影响红脂大小蠹的入侵。【目的】掌握赤峰地区红脂大小蠹寄主树种内生真菌的物种多样性,筛选对其伴生菌有拮抗作用的菌株,为红脂大小蠹的生物防治提供资源。【方法】采用组织分离法、形态学鉴定和internal transcribed spacer (ITS)序列分析相结合的方法对红脂大小蠹寄主树种油松、樟子松和潜在寄主落叶松进行内生真菌多样性研究,并用两点对峙法进行拮抗菌株筛选。【结果】松树内生真菌鉴定为2门6纲10目18科19属,其中落叶松韧皮部分离到的内生真菌数量最多,为39株,隶属于9属12种,真菌检出率为43.33%;油松次之,为30株,隶属于7属8种,真菌检出率为33.33%;樟子松最少,为29株,隶属于10属13种,真菌检出率为32.22%。3个树种的内生真菌相似性较低,无共有的菌种,青霉属(Penicillium)和篮状菌属(Talaromyces)是唯二的共有菌属,且青霉属在3个树种中均为优势菌属。平板对峙结果表明90%以上的树木内生真菌均能够与伴生真菌形成稳定的对峙,抑制率在50%-86%之间,且Phialocephala sp.和Pochonia bulbillosa对伴生菌的抑制率能高达93.7%。【结论】松树韧皮部的内生真菌具有较高的生防潜力,Phialocephala sp.和P. bulbillosa对红脂大小蠹伴生菌有较好的抑制效果,可作为红脂大小蠹的生防资源。  相似文献

The effects of the herbicide tebuthiuron (0.36, 0.6, and 1.01 kg/ha in pellet form) on nontarget organisms, vesicular-arbuscular mycorrhizal fungi, were observed in sagebrush semidesert in central Utah. Only the highest level of tebuthiuron application showed any significant effects on mycorrhizal fungi compared to the untreated control. The introduced annual Bromus tectorum L. had both a reduced percent mycorrhizal root infection and reduced spore density in its rhizosphere with the highest herbicide level. The herbicide did not significantly affect mycorrhizal root infection of Sitanion hystrix, a short-lived perennial grass, at any level of application. As part of a wider study into the role of soil fungi in the ecology of the winter annual grass, Vulpia ciliata ssp. ambigua (Le Gall) Stace & Auquier, we applied the fungicides benomyl and prochloraz to three natural populations of the grass growing in East anglia, United Kingdom. The rhizosphere and rootinfecting fungi associated with the three populations were analysed each month between February and May 1992 when plants set seed. There were marked differences between the fungal floras associated with each of the three populations of V. ciliata, despite the fact that associated plant species and soil nutrient status were broadly similar between sites. This was attributed to wide differences in soil pH between the three populations. Prochloraz did not affect fungal abundance, but benomyl decreased the isolation frequencies of Fusarium oxysporum from roots and the frequencies of Penicillium and Trichoderma spp. isolated from rhizosphere soil, and increased the frequency of isolation of Mucor hiemalis from the rhizosphere of V. ciliata. There were also significant increases in the isolation frequencies of F. oxysporum from roots and M. hiemalis, Trichoderma spp. and Phoma fimeti from the rhizosphere of V. ciliata as plants matured. The significance of these results for the design of ecological field experiments are discussed in light of a previous study which has shown that asymptomatic root-infecting fungi can affect plant fecundity and hence abundance in natural populations of V. ciliata. We propose that differences in microbial communities between sites, controlled in part by soil chemistry, are a major factor determining plant performance under field conditions.  相似文献

Three lignocellulolytic fungi, Trametes versicolor, Pleurotus ostreatus, and Coprinus cinereus, and two cellulolytic fungi Trichoderma reesei andChaetomium elatum were tested for their ability to degrade nitrocellulose. They were provided with different carbon and nitrogen sources in liquid cultures. Nitrocellulose (N content above 12%) was added as nitrogen source (in solution in acetone) alongside amino acids or as sole N source. Either starch or carboxy-methyl cellulose were provided as carbon sources. After 28 days of growth the highest decrease of nitrocellulose was observed with Chaetomium elatum when up to 43% was degraded in a medium containing nitrocellulose as the only nitrogen source. Coprinus cinereus caused a 37% decrease of nitrocellulose when provided with amino acids and starch as co-substrate. In cultures of Trametes versicolor, Pleurotus ostreatus andTrichoderma reesei, only 10%–22% decrease of nitrocellulose was measured in all media. In the presence of nitrocellulose with N content below 12% supplied as 3 mm pellets as the only carbon source, or with nitrocellulose with carboxy-methyl cellulose, the release of nitrite and nitrate from liquid cultures of Chaetomium elatum was measured. Between 6 and 9 days of growth in these media, an increase in both nitrite and nitrate was observed with a loss in weight of nitrocellulose up to 6% achieved after 34 days. The physical nature of the NC pellets may have reduced the rate of degradation in comparison with supplying NC in solution in the cultures.  相似文献

Endophytic fungi in leaves of Lycopersicon esculentum Mill   总被引:1,自引:0,他引:1  
Endophytic fungi were isolated from healthy tomato leaves from field-grown plants. Several surface-sterilization techniques were assayed for removing and killing epiphytes on tomato leaves. Surface-sterilization with an undiluted commercial solution of sodium hypochlorite was employed. To determine the spectrum of endophytes, samples were collected in February 1998 and 1999 from tomato plants (cultivar Tommy) cultivated in the experimental fields of Facultad de Ciencias Agrarias y Forestales, Universidad Nacional de La Plata, Buenos Aires, Argentina. In general, different spectra of species were isolated in 1998 and 1999.Alternaria alternata was the fungus most frequently isolated from tomato leaves in 1999, but it was the second most common species in 1998. In contrast, Colletotrichum gloeosporioides was the fungus most frequently isolated in 1998, but it was not found in 1999. Species of other genera, such as Cladosporium and Penicillium, were isolated in both years.  相似文献

Summary Multigene families are a ubiquitous feature of eukaryotes; however, their presence in Saccharomyces is more limited. The MAL multigene family is comprised of five unlined loci, MAL1, MAL2, MAL3, MAL4 and MAL6, any one of which is sufficient for yeast to metabolize maltose. A cloned MAL6 locus was used as a probe to facilitate the cloning of the other four functional loci as well as two partially active alleles of MAL1. Each locus could be characterized as a cluster of three genes, MALR (regulatory), MALT (maltose transport or permease) and MALS (structural or maltase), encoded by a total of about 7 kb of DNA; however, homologous sequences at each locus extend beyond the coding regions. Our results indicate that there is extensive homology among the MAL loci, especially within their maltase genes. The greatest sequence diversity occurs in their regulatory gene regions. Southern cross analyses of the cloned MAL loci indicate a single duplication of the MAL6R-homologous sequences upstream of the MAL6R gene as well as an extensive duplication of more than 10 kb at the MAL3 locus. The large repeat at the MAL3 locus results in the presence of four copies of MAL3R-homologous sequences and two copies of MAL3T-homologous sequences at that locus. Two naturally occurring inactive alleles of MAL1 show a deletion or divergence of their MALR sequences. The significance of these repeats in the evolution of the MAL multigene family is discussed.  相似文献

Aspergillus niger, Geotrichum candidum and Mucor miehei, when grown on a medium containing beef tallow or poultry fat for 5 days at 28°C, in Roux bottles, shake-flasks or fermenters, synthesized from 0.4 to 3.2 g lipids/I. The degree of fat utilization varied from 15 to 70%. The type of fat in the medium, the species of fungi and its cultivation method all influenced the fatty acid composition of fats remaining in the medium, as well as the intracellular lipids of the fungi.  相似文献

A membrane-bound L-asparaginase (EC 3.5.1.1) of Tetrahymena pyriformis was purified to homogeneity. The purified enzyme is a lipoprotein, since it is inactivated by phospholipase C and its activity is restored by the addition of naturally occuring lipids, such as phosphatidylcholine, triolein and oleyl acetate. The relative effectiveness of a variety of phospholipids, free saturated and unsaturated fatty acids, or neutral lipids, such as esters of fatty, acids and glycerides, with respect to the activation of purified L-asparaginase is compared. Enzyme activity is reconstituted in the presence of lipids and evidence for the formation of an enzyme-phospholipid complex is presented. Contribution of leaf surface fungi to the air spora   总被引:1,自引:0,他引:1  
High concentrations of airborne fungal spores frequently occur from spring through fall in temperate areas of the world. Although it is generally assumed that fungi on leaf surfaces are contributors to the air spora, little data are available comparing the types of fungi found on leaf surfaces with those in the atmosphere. Air sampling was carried out with a Burkard Spore Trap located on the roof of a building on the University of Tulsa campus using standard methods. Leaf samples were aseptically collected from Ulmus americana and Quercus palustris trees on campus, placed in sterile plastic bags, and brought to the lab. For each leaf, 4 cm2 areas of both upper and lower leaf surfaces were swabbed and plated on malt extract agar with streptomycin. Cultures were incubated at room temperature for 5–7 days and then examined microscopically. Results were expressed as colony forming units (CFU)/cm2. Twenty-one fungal taxa were identified from the air samples. The most abundant taxa were Cladosporium, ascospores, basidiospores, and Alternaria; together these four spore types comprised over 90% of the yearly total. Yeasts were the most abundant fungi isolated from both leaf types. Among the mycelial fungi were Phoma species, followed by Cladosporium and Alternaria. Overall twenty genera of filamentous fungi were identified. Yeasts and Phoma are normally splash dispersed and were not identified in the Burkard air samples. However, 10 taxa isolated from leaf surfaces were registered in air samples. Crude estimates of the leaf surface area of each tree suggest that the total fungal load was approximately 5.04×108 CFU for Ulmus and 2.71×108 CFU for Quercus. Of these levels, 19% were from fungi also detected in air samples. The data suggest that some leaf-surface fungi are major contributors to the air spora.  相似文献
