It was previously shown that a one-dimensional Ising model could successfully simulate the equilibrium binding of myosin S1 to regulated actin filaments (T. L. Hill, E. Eisenberg and L. Greene, Proc. Natl. Acad. Sci. U.S.A. 77:3186-3190, 1980). However, the time course of myosin S1 binding to regulated actin was thought to be incompatible with this model, and a three-state model was subsequently developed (D. F. McKillop and M. A. Geeves, Biophys. J. 65:693-701, 1993). A quantitative analysis of the predicted time course of myosin S1 binding to regulated actin, however, was never done for either model. Here we present the procedure for the theoretical evaluation of the time course of myosin S1 binding for both models and then show that 1) the Hill model can predict the "lag" in the binding of myosin S1 to regulated actin that is observed in the absence of Ca++ when S1 is in excess of actin, and 2) both models generate very similar families of binding curves when [S1]/[actin] is varied. This result shows that, just based on the equilibrium and pre-steady-state kinetic binding data alone, it is not possible to differentiate between the two models. Thus, the model of Hill et al. cannot be ruled out on the basis of existing pre-steady-state and equilibrium binding data. Physical mechanisms underlying the generation of the lag in the Hill model are discussed.  相似文献

Methionine is an essential amino acid for both prokaryotic and eukaryotic organisms; however, little is known concerning its utilization in African trypanosomes, protozoa of the Trypanosoma brucei group. This study explored the Michaelis-Menten kinetic constants for transport and pool formation as well as metabolic utilization of methionine by two divergent strains of African trypanosomes, Trypanosoma brucei brucei (a veterinary pathogen), highly sensitive to trypanocidal agents, and Trypanosoma brucei rhodesiense (a human pathogenic isolate), highly refractory to trypanocidal arsenicals. The Michaelis-Menten constants derived by Hanes-Woolf analysis for transport of methionine for T. b. brucei and T. b. rhodesiense, respectively, were as follows: K(M) values, 1. 15 and 1.75 mM; V(max) values, 3.97 x 10(-5) and 4.86 x 10(-5) mol/L/min. Very similar values were obtained by Lineweaver-Burk analysis (K(M), 0.25 and 1.0 mM; V(max), 1 x 10(-5) and 2.0 x 10(-5) mol/L/min, T. b. brucei and T. b. rhodesiense, respectively). Cooperativity analyses by Hill (log-log) plot gave Hill coefficients (n) of 6 and 2 for T. b. brucei and T. b. rhodesiense, respectively. Cytosolic accumulation of methionine after 10-min incubation with 25 mM exogenous methionine was 1.8-fold greater in T. b. rhodesiense than T. b. brucei (2.1 vs 1.1 mM, respectively). In African trypanosomes as in their mammalian host, S-adenosylmethionine (AdoMet) is the major product of methionine metabolism. Accumulation of AdoMet was measured by HPLC analysis of cytosolic extracts incubated in the presence of increasing cytosolic methionine. In trypanosomes incubated for 10 min with saturating methionine, both organisms accumulated similar amounts of AdoMet (approximately 23 microM), but the level of trans-sulfuration products (cystathionine and cysteine) in T. b. rhodesiense was double that of T. b. brucei. Methionine incorporation during protein synthesis in T. b. brucei was 2.5 times that of T. b. rhodesiense. These results further confirm our belief that the major pathways of methionine utilization, for polyamine synthesis, protein transmethylation and the trans-sulfuration pathway, are excellent targets for chemotherapeutic intervention against African trypanosomes.  相似文献

Cooperative binding of myosin S-1.ADP to regulated F-actin was previously reported and has been interpreted by a two-state model in which an important source of cooperativity is nearest neighbor interactions between the 7-actin.tropomyosin (TM).troponin units (functional units) (Hill, T.L., Eisenberg, E., and Greene, L. (1980) Proc. Natl. Acad. Sci. U.S.A. 77, 3186-3190). It has been postulated that the head-to-tail overlap between adjacent TM molecules is the structural basis of the nearest neighbor interactions. We tested the hypothesis by examining S-1.ADP binding to reconstituted regulated F-actin containing either intact TM or nonpolymerizable TM from which the COOH-terminal 11 residues were removed. In the absence of Ca2+, substitution of nonpolymerizable TM for TM reduced significantly the slope of the steeply rising phase of the sigmoidal S-1.ADP binding curve. Nevertheless, considerable residual cooperativity remained. Analysis of the data using the two-state model of Hill et al. suggests that removal of TM overlap abolishes nearest neighbor interactions, while the concerted change of the state of 7 actins in a functional unit can account for the residual cooperativity.  相似文献

We describe a mutant of Escherichia coli citrate synthase, CS R319L, in which the arginine residue at position 319 of the sequence has been replaced by leucine. In this mutant, saturation by the substrate acetyl-CoA is changed from sigmoid (Hill parameter = 1.75 +/- 0.2) to hyperbolic (Hill parameter = 1.0 +/- 0.1) and dependence on the activator KCl is greatly reduced. Further mutations at this site and at position 343 (which model building predicts is close enough to allow a side-chain interaction with position 319) are also described. In the wild-type enzyme, the model suggests the possibility of a salt-bridge interaction between Arg-319 (located on the P helix in the small domain) and Glu-343 (in the Q helix in the same domain), but mutation of Glu-343 to Ala (CS E343A) produced a much smaller difference in the kinetic properties than the ARg-319 to Leu mutation did. Small changes in kinetic properties were also obtained with an Arg-319----Glu (CS R319E) mutation. In CS R319L, oxaloacetate, the first substrate to bind, induces an ultraviolet difference spectrum which is obtained with wild-type enzyme only in the presence of KCl. To account for these observations we postulate that wild-type E. coli citrate synthase exists in two conformational states, T and R, which are equilibrium; T state binds NADH, the allosteric inhibitor, while R state binds substrates and can be converted to another substrate-binding state, R', by KCl.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

盐生植物黄花补血草广泛分布于我国西北地区、东北西部以及华北北部,对改良盐碱土壤具有重要的生态作用。以黄花补血草(Limonium aureum(L.)Hill)为材料,研究分析了不同浓度NaCl胁迫对其种子萌发和幼苗生长产生的抑制效应及作用机制。结果表明:低浓度NaCl(25 mmol/L和50 mmol/L)处理不影响黄花补血草种子萌发和幼苗生长,25 mmol/L NaCl甚至促进了根生长,而高浓度NaCl(100 mmol/L和150 mmol/L)处理明显抑制种子萌发及幼苗生长。利用荧光探针的检测结果表明,NaCl处理的幼苗根中过氧化氢(H2O2)和一氧化氮(NO)含量明显高于对照水平。碘化丙啶(PI)染色结合激光共聚焦显微镜观察及检测相对电导率结果显示,高浓度NaCl处理抑制了幼苗根尖伸长区细胞的伸长生长,增加了细胞膜的通透性,对根细胞造成了明显的伤害。此外,高浓度NaCl处理诱导叶片丙二醛(MDA)含量显著升高。以上结果说明,黄花补血草对低浓度的盐具有一定的耐盐性,但高浓度盐降低了种子的萌发率,使幼苗根中H2O2产生增加,抑制根尖伸长区细胞的伸长生长,对根、叶造成明显氧化损伤,从而抑制黄花补血草幼苗的生长。  相似文献

Effects of plant growth regulators on Hill activity during induced senescence of leaves of three submerged aquatic plants Vallisneria spiralis L., Hydrilla verticillata (L.f.) Royle and Potamogeton pectinatus L., and a terrestrial plant Spinacia oleracea L. were studied. Hill activity was reduced by 39.3, 42.7, 45.2 and 245.1 μmol DCIP (2,6-dichloroindophenol) (mg chl)?1h?1 in Vallisneria, Hydrilla, Potamogeton and Spinacia, respectively. During induced senescence of isolated mature leaves, Hill activity declined with increasing incubation time in all species. Kinetin (0.23 mM) treatment reduced the loss of Hill activity; while both 0.69 mM ehthrel and 0.075 mM ABA treatments decreased it in each species. The effect of kinetin was greatest in Spinacia, followed by Potamogeton, Hydrilla and Vallisneria, while the effect of either ethrel or ABA or both was greatest in Potamogeton, followed by Spinacia, Vallisneria and Hydrilla. Kinetin pre-treatment for an optimal period (12 h) followed by treatment with either ethrel or ABA partially removed the inhibitory effect of the latter on Hill activity. Pre-treatment of tissues with either ethrel or ABA solution, restricted to 12 h, followed by kinetin treatment markedly reduced the promotive effect of kinetin on the Hill activity of these species.  相似文献

The composition of free fatty acids (FFA) in relation to Hill reaction activity and photoperoxidation of lipids was studied in chloroplasts isolated from fresh, cold and dark-stored as well as illuminated leaves of Lycopersicon esculentum Mill., Phaseolus vulgaris L. and Cucumis sativus L. Following the cold and dark-storage of leaves the loss of Hill reaction activity is accompanied by approximately a 5-fold increase in the amount of FFA and by an increase in the percentage of unsaturated FFA, particularly that of linolenic acid. Illumination of the cold- and dark-stored leaves restores both Hill reaction activity and the content and composition of chloroplast FFA. Following the second and third cycles of cold storage and illumination of leaves the percentage of unsaturated fatty acids in chloroplasts increases while that of saturated ones decreases despite of the significant restoration of Hill reaction activity. Since the illumination of cold-stored leaves results in peroxidation of inhibitory fatty acids it seems likely that this phenomenon could, at least partially, be responsible for the restoration of Hill reaction activity. Inhibition of Hill reaction activity by exogenous linolenic acid in chloroplasts of fresh, cold-stored as well as cold-stored and illuminated leaves could be reversed following the incubation of chloroplast suspension with BSA, however only to a value measured in the absence of unsaturated fatty acid. The model of myosin regulation by a continuous tropomyosin chain is generalized to a chain of tropomyosin-troponin units. Myosin binding to regulated actin is cooperative and initially inhibited by the chain as before. In the absence of calcium, myosin is further inhibited by the binding of troponin-I to actin, which through the whole of troponin pins the tropomyosin chain in a blocking position; myosin and TnI compete for actin and induce oppositely-directed chain kinks. The model predicts equilibrium binding curves for myosin-S1 and TnI as a function of their first-order affinities K(S1) and L(TI). Myosin is detached by the actin binding of TnI, but TnI is more efficiently detached by myosin when the kink size (typically nine to ten actin sites) spans the seven-site spacing between adjacent TnI molecules. An allosteric mechanism is used for coupling the detachment of TnI to calcium binding by TnC. With thermally activated TnI kinks (kink energy B approximately k(B)T), TnI also binds cooperatively to actin, producing cooperative detachment of myosin and biphasic myosin-calcium Hill plots, with Hill coefficients of 2 at high calcium and 4-6 at low calcium as observed in striated muscle. 用不同浓度的NaCl处理盐生植物黄花补血草和大叶补血草愈伤组织,研究其中H2O2、MDA及渗透性调节物质等生理指标含量的变化,从细胞水平分析比较2种补血草愈伤组织对盐环境的适应机制.结果表明:随NaCl胁迫浓度的升高,2种补血草愈伤组织的H2O2和MDA含量逐渐显著增加;愈伤组织中脯氨酸含量增加幅度在75和150mmol/LNaCl下明显表现为黄花补血草大于大叶补血草,而其中的可溶性糖含量增加程度于相同NaCl浓度下明显呈现出黄花补血草小于大叶补血草的趋势;可溶性蛋白含量在黄花补血草中随NaCl浓度升高而逐渐增加,但在大叶补血草中却表现为低浓度(75mmol/L)比对照升高,而高浓度(150和300mmol/L)比对照明显减少的趋势.研究发现,2种补血草愈伤组织中脯氨酸、可溶性糖和可溶性蛋白这些有机渗透性调节物质对盐胁迫的反应特性存在差异,并与其耐盐性有关.  相似文献

Titrations of mitochondrial apo-aspartate aminotransferase with pyridoxal 5'-phosphate in the presence of AMP, contrary to what has been observed in the case of the cytosolic isoenzyme [(1983) FEBS Lett. 153, 98-102], show sigmoidal isotherms, with Hill coefficients ranging from nH = 1.4, in the absence of AMP, to nH = 1.8, in the presence of 5.9 mM AMP. The experimental data were successfully fitted by the Monod-Wyman- Changeaux model. The best fit, in the absence of AMP, was obtained with L = 30, KR = 4.72 X 10(-7) M and KT = 1.18 X 10(-5) M. Binding curves in the presence of AMP fit the model by keeping KR as a constant. This implies that AMP could bind to the apoenzyme only in the T state. In contrast, binding curves in the presence of phosphate ion (Pi) showed a less pronounced cooperativity, the Hill coefficient dropping to nH = 1.0 in the presence of 0.1 mM Pi. The above results suggest a regulatory role of AMP and Pi in the reconstitution of aspartate aminotransferase.  相似文献

In order to establish differences in the chilling sensitivity of domestic and wild Lycopersicon species, galactolipase (EC 3.1.1.26) activity, free fatty acid (FFA) level and Hill reaction activity were measured in chloroplasts isolated from control and cold treated leaves of L. esculentum Mill., cv. Norton, L. hirsutum Humb. and Bonpl., L. peruvianum var. glandulosum Mill. Galactolipase activity was higher in chloroplasts from Lycopersicon species with high chilling sensitivity than in chloroplasts of more chilling-resistant ones. A similar relationship was observed for FFA level in chloroplasts from both cold-stored and control leaves. Decrease in Hill reaction activity due to cold stress was greater in chloroplasts of more chilling-sensitive species. The changes are accompanied by a decline of photochemical activity. Considering the changes in the three parameters noted above, an increasing order of chilling tolerance was established: L. esculentum < L. hirsutum (700 m) < L. hirsutum (3100 m) < L. peruvianum (3400 m). It is suggested that measurements of galactolipase activity and FFA may be useful in an evaluation of differences in resistance to chilling injury of closely related species.  相似文献

为了研究黄花补血草挥发油的化学成分,本文采用了水蒸气蒸馏法从黄花补血草中提取挥发油,并用GC-MS法采用最佳分析条件对化学成分进行鉴定,用峰面积归一化法测定各化合物在挥发油中的相对百分含量;通过研究,鉴定出70种化合物,其峰面积相对含量约占挥发油总量82.12%。黄花补血草挥发油的主要组分为2-硝基乙醇(59.63%)、正二十四烷(3.71%)、二苯胺(2.31%)、6,10,14-三甲基-2-十五烷酮(1.79%)、正二十一烷(1.57%)、丙二醇(1.40%)等。  相似文献

A method was described to determine root growth respiration and root maintenance respiration rate of plants, grown in culture solutions of high and low oxygen concentrations, during linear growth. Root growth respiration of aerobically grown plants was three to four times lower in the flood-intolerant Senecio jacobaea L. than in the flood-tolerant species. Senecio aquaticus Hill. Root growth respiration of Senecio aquaticus Hill decreased by a factor two to three upon transplantation to a culture solution of low oxygen tension. The difference in root growth respiration between aerobically and anaerobically grown Senecio aquaticus Hill was ascribed to the presence of a highly active non-phosphorylating oxidase.  相似文献

Excised lateral buds of taro [Colocasia esculenta var. esculenta (L.) A.F. Hill] developed into plantlets and formed callus if cultured on media containing taro extract. α-Naphthaleneacetic acid enhanced the process but only if taro extract was also present. An improved procedure is described for the characterization of solute self-association by sedimentation equilibrium. Whereas previous statistical-mechanical approaches to allowance for the effects of thermodynamic nonideality have entailed tedious iteration because of their specification of activity coefficients in terms of the equilibrium concentrations of all species, such reliance upon knowledge of the solution composition is avoided by the adaptation of an alternative statistical-mechanical formulation [T. L. Hill and Y. D. Chen (1973) Biopolymers, Vol. 12, pp. 1285–1312] in which thermodynamic nonideality is expressed in terms of total solute concentration. The development of an analysis in terms of a relationship with total solute concentration as the experimental variable allows this attribute of the Adams-Fujita approach to be retained without sacrifice of statistical-mechanical rigor. Its use is illustrated by application to Rayleigh interferometric records of sedimentation equilibrium distributions reflecting α-chymotrypsin dimerization and lysozyme self-association. © 1996 John Wiley & Sons, Inc.  相似文献

In isolated broken chloroplasts photosynthetic electron transport requires the presence of CO2 and/or bicarbonate. This bicarbonate effect on electron flow was measured in a medium containing 100 m M sodium formate. In this medium a dark incubation time with bicarbonate is required for the reactivation of the Hill reaction. We have measured the kinetics of the reactivation of electron flow by varying the dark incubation of CO2-depIeted pea ( Pisum sativum L., cv. Rondo) chloroplasts with bicarbonate. The half-time of this reactivation appears to be 25 s when 2 m M bicarbonate is added.
The dinitrophenol herbicide, i -dinoseb, is shown to be a competitive inhibitor of the bicarbonate dependent Hill reaction with an inhibitor constant (Ki) of 31 n M . In the presence of 100 n M i-dinoseb or 100 n M DCMU the half-time of the reactivation by 2 m M bicarbonate appears to increase to about 58 s. We provide an explanation for these phenomena by analyzing the bicarbonate-thylakoid interaction on the basis of a simple reaction scheme. The binding of bicarbonate to the thylakoids appears to be a second order reaction with pseudo-first order kinetics. According to our analysis, any inhibitor, which is competitive with respect to the bicarbonate stimulation of the Hill reaction, should increase the half-time of the reactivation of the Hill reaction.  相似文献

The effect of troponin T treatment on the Ca(2+)-activated tension of single glycerinated rabbit skeletal muscle fibers was examined. The tension of the fiber was completely desensitized to Ca2+ by incubation in a solution containing an excessive amount of troponin T and reached a level of about 70% of the maximum tension of the control fiber. SDS/PAGE showed that most of troponins C and I was removed from the fiber by troponin T treatment. During the course of troponin T treatment, the cooperativity of Ca2+ activation (Hill coefficient) was decreased while pCa at half-maximal Ca(2+)-sensitive tension (pK) increased. Using the 26-K fragment of troponin T, the study indicated that the removal of troponins C and I was due to the replacement of the troponin C.I.T complex in the myofibrils of the fiber with the added troponin T. The troponin-T-treated fiber was again sensitized to Ca2+ by the addition of troponin C.I. The removal of troponin C by treatment with trans-1,2-cyclohexanediamine-N,N,N',N'-tetraacetic acid did not change the minimum tension of the fiber, from which troponin C.I was partially removed by troponin T treatment, but it decreased the height of maximum tension with a concomitant decrease in the Hill coefficient as well as a decrease in pK. The above findings suggested that pK is determined by the balance between two opposite actions through troponins C and I, while the extent of cooperativity of Ca2+ activation seemed to be related mainly to the content of troponin C.  相似文献
