The present investigation was targeted towards a highly desirable everbearing trait in strawberry (Fragaria × ananassa Duch.) via marker assisted selection while seeing its worldwide commercial applicability through the extended harvest season. The crosses were made between everbearing and june-bearing cultivars to raise the F1 individuals. Morphological characters (plant, floral, and fruit) were assessed that showed significant differences among the strawberry cultivars. Molecular characterization was carried out between everbearing and non-everbearing cultivars using RAPD and SSR markers. For phenotyping, a chi-square test was performed and revealed that out of all four cross combinations, the best fitted cross found to be in Mendelian segregation ratio (1:1) was ‘Confectura’ × ‘Torrey’ with χ2-value 1.58. Further, the identified polymorphic markers were assessed across the F1 individuals of cross ‘Confectura’ × ‘Torrey’ for its genotyping. It could be revealed that the targeted everbearing trait is governed by a dominant gene(s) in the subjected strawberry genotypes. Strawberry (Fragaria × ananassa Duch), a fruit of economic and nutritional importance, is also a model species for fleshy fruits and genomics in Rosaceae. Strawberry fruit quality at different harvest stages is a function of the fruit's metabolite content, which results from physiological changes during fruit growth and ripening. In order to investigate strawberry fruit development, untargeted (GC-MS) and targeted (HPLC) metabolic profiling analyses were conducted. Principal component analysis (PCA) and partial least squares discriminant analysis (PLS-DA) were employed to explore the non-polar and polar metabolite profiles from fruit samples at seven developmental stages. Different cluster patterns and a broad range of metabolites that exerted influence on cluster formation of metabolite profiles were observed. Significant changes in metabolite levels were found in both fruits turning red and fruits over-ripening in comparison with red-ripening fruits. The levels of free amino acids decreased gradually before the red-ripening stage, but increased significantly in the over-ripening stage. Metabolite correlation and network analysis revealed the interdependencies of individual metabolites and metabolic pathways. Activities of several metabolic pathways, including ester biosynthesis, the tricarboxylic acid cycle, the shikimate pathway, and amino acid metabolism, shifted during fruit growth and ripening. These results not only confirmed published metabolic data but also revealed new insights into strawberry fruit composition and metabolite changes, thus demonstrating the value of metabolomics as a functional genomics tool in characterizing the mechanism of fruit quality formation, a key developmental stage in most economically important fruit crops.  相似文献

Effect of GA3 on postharvest ripening in strawberry fruit was evaluated through different biochemical parameters. Strawberry slices at different ripening stages were incubated with GA3. A significant decrease on respiratory activity depending on GA3 concentration was obtained. Also GA3 was applied to whole and deachened fruit at white and green ripening stages. Our results show that GA3 has an inhibitory effect on strawberry fruit ripening, evidenced by a decrease in the respiratory activity and a delay in anthocyanin synthesis and chlorophylls degradation.  相似文献

Studies were conducted to determine the effect of vermicompost on growth, physiological disorders, fruit yield and quality of 'Chandler' strawberry. For this, 4 levels of vermicompost (2.5, 5.0, 7.5 and 10.0tha(-1)) were supplemented with inorganic fertilizers to balance fertilizer requirement of strawberry under semi-arid region of northern India. The vermicompost was incorporated into top 10cm layer of soil, which was supplemented on the basis of chemical analysis, with amount of inorganic N, P, K fertilizer calculated to equalize the recommended dose of nutrients. Vermicompost application increased plant spread (10.7%), leaf area (23.1%) and dry matter (20.7%), and increased total fruit yield (32.7%). Substitution of vermicompost drastically reduced the incidence of physiological disorders like albinism (16.1-4.5%); fruit malformation (11.5-4.0%) and occurrence of grey mould (10.4-2.1%) in strawberry indicating that vermicompost had significant role in reducing nutrient-related disorders and disease like Botrytis rot, and thereby increasing the marketable fruit yield up to 58.6% with better quality parameters. Fruit harvested from plant receiving vermicompost were firmer, have higher TSS, ascorbic acid content and lower acidity, and have attractive colour. All these parameters appeared to be dose dependent and best results were achieved @ 7.5tha(-1), however, beyond this dose of vermicompost, there was not significant influence on these parameters.  相似文献

During banana ripening there is a massive conversion into sugars, mainly sucrose, which can account for more than 10% of the fresh weight of the fruit. An ethylene burst is the trigger of the banana ripening process but there is evidence that other compounds can act as modulators of some biochemical pathways. As previously demonstrated, gibberellic acid (GA3) can impair the onset of starch degradation and affect some degradative enzymes, but effects on the sucrose biosynthetic apparatus have not yet been elucidated. Here, the activity and amount of sucrose synthase (SuSy; E.C. 2.4.1.13) and sucrose–phosphate synthase (SPS; E.C. 2.4.1.14), respiration rates, ethylene production, and carbohydrate levels, were evaluated in GA3-infiltrated and non-infiltrated banana slices. The exogenous supply of gibberellin did not alter the respiration or the ethylene profile but delayed sucrose accumulation by at least 2 days. While SuSy activity was similar in control and treated slices, SPS increase and sucrose accumulation was related in treated slices. Western blotting with specific antiserum showed no apparent effects of GA3 on the amount of SuSy protein, but impaired the increase in SPS protein during ripening. The overall results indicate that although GA3 did not block carbohydrate mobilisation in a irreversibly way, it clearly affected the triggering of starch breakdown and sucrose synthesis. Also, the delayed sucrose accumulation in GA3-infiltrated slices could be explained by the disturbance of SPS activity. In conclusion, gibberellins can play an important role during banana ripening and our results also reinforce the idea of multiple regulatory components in the ripening pathway, as evidenced by the GA3 effects.  相似文献

In the short-day plant, strawberry (Fragaria ananassa Duch.), polyamines (putrescine, spermidine and spermine), conjugated spermidine (water-insoluble compounds) and bound amines (putrescine, spermidine, phenylethylamine, 3-hydroxy, 4-methoxyphenylethylamine) accumulated in the shoot tips during floral induction and before floral emergence. Different associations of free amines and conjugated amines were observed during floral induction, as compared with the reproductive phase. During the whole period of floral development, phenylethylamine (an aromatic amine) was the predominant amine, representing 80 to 90% of the total free amine pool. Phenylethylamine conjugates (water-insoluble compounds) were the predominant amides observed prior to fertilization. These substances decreased drastically after fertilization. In vegetative shoot tips from plants grown continously under long days, free polyamines (putrescine, spermidine) and bound polyamines (putrescine, spermidine) were low and no change was observed. Free amines (spermine and phenylethylamine), bound aromatic amines (phenylethylamine, 3-hydroxy, 4-methoxyphenylethylamine), conjugated spermidine and phenylethylamine did not appear. Male-sterile flowers were distinguished by their lack of conjugated spermidine and phenylethyalamine and by a decrease in free phenylethylamine. In normal and sterile strawberry plants -DL-difluoromethylornithine (DFMO), a specific irreversible inhibitor of ornithine decarboxylase (ODC), caused inhibition of flowering and free and polyamine conjugates. When putrescine was added, polyamine titers and flowering were restored. A similar treatment with -DL-difluoromethylarginine (DFMA), a specific, irreversible inhibitor of arginine decarboxylase (ADC), did not affect flowering and polyamine titers. These results suggest that ornithine decarboxylase (ODC) and polyamines are involved in regulating floral initiation in strawberry. Wild strawberry (Fragaria vesca L.) is an attactive model system for studying ripening in non-climacteric fruit, because of its small diploid genome, its short reproductive cycle, and its capacity for transformation. We have isolated eight ripening-induced cDNAs from this species after differential screening of a cDNA library. The predicted polypeptides of seven of the clones exhibit similarity to database protein sequences, including acyl carrier protein, caffeoyl- CoA 3-O-methyltransferase, sesquiterpene cyclase, major latex protein, cystathionine -synthase, dehydrin and an auxin- induced gene. A ninth cDNA clone that was constitutively expressed is predicted to encode a metallothionein-like protein. None of these proteins appear to be directly related to events generally associated with ripening such as cell wall metabolism or the accumulation of sugars and pigments, rather, their putative functions are indicative of the wide range of processes upregulated during fruit ripening.  相似文献

BACKGROUND AND AIMS: A number of strawberry varieties were surveyed for their total ellagic acid concentration, and attempts were made to determine if ellagic acid and ascorbic acid concentrations of two strawberry cultivars could be increased by polythene reflective mulches. METHODS: After adjusting crop yields and cultivation using polythene mulches with two different PAR reflective capacities, field- and polytunnel-grown strawberries were analysed for ellagic acid and ascorbic acid concentrations by HPLC. Comparative measurements of yield and fruit quality were determined along with plant developmental changes. KEY RESULTS: Ellagic acid concentration varied widely with strawberry cultivar (60-341 microg g(-1) frozen weight), as did the ratio of conjugated ellagic acid : free ellagic acid. Also, there was significant year-to-year variation in total ellagic acid concentration with some cultivars. Mulches with different reflective capacities impacted on strawberry production; highly reflective mulches significantly increased growth and yield, the latter due to increases in fruit size and number. CONCLUSIONS: Highly reflective mulches significantly increased total concentrations of ellagic acid and ascorbic acid relative to control in fruit of different cultivars. The potential of agronomic practices to enhance the concentration and amounts of these important dietary bioactive compounds is discussed.  相似文献

Studies were conduced to test the effects of various cytokinins on somatic embryogenesis from chickpea (Cicer arietinum L.) immature cotyledons. Zeatin (13.7 μmol) added, to B5 basal medium, supplemented with 1.5 % sucrose and 0.2 μmol indole-3-acetic acid, was the most effective cytokinin. Lobular structures obtained from cotyledons cultures were transferred to B5 basal medium supplemented with gibberellic acid and indole-3-butyric acid at different concentrations. The most effective treatment was B5 medium containing 14.4 μmol gibberellic acid plus 1.0 μmol indole-3-butyric acid in which 42.8 % of lobular structures cultured formed normal somatic embryos. High conversion of embryos into plantlets (61.0–65.2 % embryos regenerated plants) was observed when germinated embryos were placed on plant development medium.  相似文献

Changes in gene expression during foliar senescence and fruit ripening in tomato (Lycopersicon esculentum Mill.) were examined using in-vitro translation of isolated RNA and hybridization against cDNA clones.During the period of chlorophyll loss in leaves, changes occurred in mRNA in-vitro translation products, with some being reduced in prevalence, whilst others increased. Some of the translation products which changed in abundance had similar molecular weights to those known to increase during tomato fruit ripening. By testing RNA from senescing leaves against a tomato fruit ripening-related cDNA library, seven cDNA clones were identified for mRNAs whose prevalence increased during both ripening and leaf senescence. Using dot hybridization, the pattern of expression of the mRNAs corresponding to the seven clones was examined. Maximal expression of the majority of the mRNAs coincided with the time of greatest ethylene production, in both leaves and fruit. Treatment of mature green leaves or unripe fruit with the ethylene antagonist silver thiosulphate prevented the onset of senescence or ripening, and the expression of five of the seven ripening- and senescence-related genes.The results indicate that senescence and ripening in tomato involve the expression of related genes, and that ethylene may be an important factor in controlling their expression.Abbreviations cDNA copy-DNA - MW molecular weight - PAGE polyacrylamide gel electrophoresis - SDS sodium dodecyl sulphate  相似文献

A sand culture experiment assessed whether gibberellic acid(GA3) could alleviate the adverse effects of salt stress on thegrowth, ion accumulation and photosynthetic capacity of two spring wheatcultivars, Barani-83 (salt sensitive) and SARC-I (salt tolerant).Three-week-oldplants of both cultivars were exposed to 0, 100 and 200 molm–3 NaCl in Hoagland's nutrient solution. Threeweeks after the initiation of salt treatments, half of the plants of eachcultivar were sprayed overall with 100 mg L–1GA3 solution. Plants were harvested 3 weeks after theapplication of GA3. Fresh and dry weights of shoots and roots, plantheight and leaf area were decreased with increasing supply of salt, butgibberellic acid treatment caused a significant ameliorative effect on both thecultivars with respect to these growth attributes. However, GA3caused no significant change in grain yields but increased grain size in boththe cultivars. Saline growth medium caused a marked increase in theconcentrations of Na+ and Cl in shoots androots of both the lines. However, with the application of GA3accumulation of Na+ and Cl was enhanced inboth shoots and roots of both wheat lines, but more ions accumulated in saltsensitive Barani-83 than in salt tolerant SARC-1. Net CO2assimilation rate (A) of both wheat lines decreased consistently withincreasingsupply of NaCl, but application of GA3 alleviated the effect of saltstress on this variable in both the cultivars. However, the ameliorative effectof the hormone was more pronounced in Barani-83 than in SARC-1. Althoughwater-use efficiency (A/E=CO2assimilation/transpiration) and intrinsic water use efficiency(A/gs=CO2 assimilation/stomatalconductance) decreased significantly with increasing salt concentration of thegrowth medium in both the cultivars, GA3 was more effective inenhancing both the water-use attributes in Barani-83 than in SARC-1. Overall,GA3 treatment stimulated the vegetative growth of both cultivars ofwheat under salt stress, but it caused a slight reduction in grain yield.GA3 treatment enhanced the accumulation of Na+ andCl in both shoots and roots of wheat plants under saltstress.It also caused a significant increase in photosynthetic capacity in both linesat the vegetative stage under both saline and non-saline media.  相似文献

During germination, the transmembrane electric potential (PD) of cortical cells of the embryonal axis of radish seeds (Raphanus sativus L.) rises from-120 mV initially to a maximum of-150 mV after 5 h incubation, then falls again to stable values of around-120 mV. Treatments inhibiting germination block the transitory PD increase. Administration of uncoupling agents or low temperatures, during the process of germination, produces a marked fall of the PD transitory increase. Abscisic Acid has a parallel inhibitory effect on PD and germination, while fusicoccin produces a rise in both; administration of abscisic acid with fusicoccin inhibits germination, while the PD remains at the high levels given by fusicoccin. Abscisic acid (ABA) was identified by combined gas liquid chromatography-mass spectrometry in sieve-tube exudate collected from the cut stylar ends of white lupin fruit. Water stress caused an increase in ABA levels in leaf, seed and pod tissues and phloem exudate. When compared with levels in extracts of these tissues, the concentration of ABA in sieve-tube sap was very high. Amino acid loss from the roots of 25-day-old, sterile and non-sterile sand-grown forage rape plants, was determined over periods of up to 3.5 hours. Amino acid accumulation in the root-zone of sterile plants was concentration-dependent giving a convex accumulation profile. Amino acid levels in the root zone of non-sterile plants rapidly attained steady state values. Microbial assimilation of amino acids within the root zone appeared to lower amino acid concentrations, resulting in an underestimation of rates of amino acid loss from roots. The concentrations of most amino acids were higher after selected amino acids were supplied to the root zone. The response to exogenous acids was dependent on the concentration and composition of the acids added. Addition of a mixture containing ASN, GLN and GABA, each at 0.25 mM resulted in a greater increase in individual and total acid levels compared with a mixture containing ALA, SER, GLY and THR at the same concentration. Apparently, amino acids supplied exogenously competed with acids lost from the plant, by providing an alternative nutrient source for root zone micro-organisms. Addition of glucose and citric acid had a similar effect to addition of ALA, SER, GLY and THR, but were less effective than ASN, GLN and GABA at all concentrations tested. The nitrogen-rich amino acids ASN and GLN, and the -amino acid, GABA, appeared to compete more effectively with plant-derived acids than did ALA, SER, GLY and THR, the most abundent constituents of the plant-derived acids, which had the highest calculated rates of microbial consumption. Therefore, although bacterial consumption showed a dependence on amino acid concentration, a degree of selectivity for nitrogen-rich acids and gaba was also apparent.  相似文献
