碳量子点(Carbon quantum dots,CQDs)是一种新型碳纳米材料,具有独特的性质,逐渐得到广泛关注。由于CQDs主要是通过"自下而上"的方法制备,其表面具有更加丰富多样的基团,因此更容易衍生化和功能化,进而更容易得到具有特殊性质和功能的CQDs。同时,CQDs在水溶性、可修饰性、耐光漂白等方面具有明显的优势,并且还兼具低生物毒性和良好的生物相容性,使其在生物成像、生物传感和生物分子/药物传递等方面具有潜在的应用价值。随着CQDs研究的增多,各种不同功能化的CQDs得到发展,并被越来越多地用于抗菌方面。根据CQDs目前在医学领域的发展,本文对CQDs在抗菌方面的最新研究进展作一综述。  相似文献

The extraction of lactoperoxidase (EC 1.11.1.7) from whey was studied using single step reverse micelles‐assisted extraction and compared with reverse micellar extraction. The reverse micelles‐assisted extraction resulted in extraction of contaminating proteins and recovery of lactoperoxidase in the aqueous phase leading to its purification. Reverse micellar extraction at the optimized condition after forward and backward steps resulted in activity recovery of lactoperoxidase and purification factor of the order of 86.60% and 3.25‐fold, respectively. Whereas reverse micelles‐assisted extraction resulted in higher activity recovery of lactoperoxidase (127.35%) and purification factor (3.39‐fold). The sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS‐PAGE) profiles also evidenced that higher purification was obtained in reverse micelles‐assisted extraction as compared of reverse micellar extracted lactoperoxidase. © 2010 American Institute of Chemical Engineers Biotechnol. The liquid-liquid extraction of alpha-lactalbumin based on reverse micellar organic solvents was investigated. Forward extraction of the protein in the reverse micellar organic phase from aqueous feed solutions was strongly dependent on the initial pH of the feed solution and the complete forward extraction of 0.03 mM alpha-lactalbumin was successfully achieved at pH 6.0. The forward extraction percentage steeply decreased with increasing KCl concentration, while in the NaCl system the forward extraction was independent on the salt concentration below 1 M. From the circular dichroic measurement, higher order structure of the recovered alpha-lactalbumin through the extraction process was well preserved.  相似文献

Reversed micellar extraction was applied for the fractionation of IgG fragments. With isooctane solution containing 50 mM AOT, Fab, Fe and IgG were extracted to the micellar phase. Each protein had an optimum pH range in the extraction. Fab and Fc were separated from the mixture at pH 7.0, with Fab being extracted to the micellar phase and Fe remaining in the aqueous phase. Extracted Fab fragments were recovered by bringing them into contact with 6M guanidine/HCl followed by dilution with PBS. Fab and Fc fragments were separated and recovered by reversed micellar extraction from IgG lysate digested with papain. Since the procedure is simple and rapid compared with column chromatography, mass preparation of the fragments can be expected by this method.  相似文献

用反胶束技术分离纯化蛋白质,具有高选择性、易于大规模操作等优点,具有良好的工业应用前景。但是离子型表面活性剂形成的反胶束体系萃取蛋白质容易引起蛋白质的变性,这是由于离子型表面活性剂的强电荷作用所导致的。对用AOT/异辛烷反胶束体系从胰酶粗提物中萃取胰蛋白酶进行了研究,通过在反胶束相加入乙醇,解决了反胶束萃取蛋白质时蛋白质变性失活的问题。并且由于乙醇的加入大大减少了分相的时间,简化了实验步骤,优化了实验方法,使此技术在工业上的大规模应用成为可能。通过优化各种实验条件,胰蛋白酶的前萃取率达到90%,反萃取率接近100%。最终得率为88%。纯化后的比活提高了5倍多,从300U/mg左右提高到了1800U/mg。  相似文献

Reverse micelles are formed in apolar solvents by spontaneous aggregation of surfactants. Surfactant sodium bis (2-ethylhexyl) sulfosuccinate (AOT) is most often used for the reverse micellar extraction of enzymes. However, the inactivation of enzyme due to strong interaction with AOT molecules is a severe problem. To overcome this problem, the AOT/water/isooctane reverse micellar system was modified by adding short chain polyethylene glycol 400 (PEG 400). The modified AOT reverse micellar system was used to extract Mucor javanicus lipase from the aqueous phase to the reverse micellar phase. The extraction efficiency (E) increased with the increase in PEG 400 addition and the maximum E in PEG 400 modified system was twofold higher than that in the PEG 400-free system. Upon addition of PEG 400, the water activity (a(w)) of aqueous phase decreased, whereas a(w) of reverse micellar phase increased. The circular dichroism spectroscopy analysis revealed that PEG 400 changes the secondary and tertiary structure of lipase. The maximum specific activity of lipase extracted in PEG 400-modified reverse micellar system was threefold higher than that in the PEG-free system.  相似文献

The purification of industrial alpha-amylase by liquid-liquid extraction with Aliquat 336 reversed micellar solution as the extractant was studied. Seven kinds of Aliquat 336 reversed micellar solution, formed by using seven kinds of straight chain alkyl alcohols as cosolvent, have been utilized to extract industrial a-amylase. It was found that these seven kinds of reversed micellar solution can all achieve a high protein transfer efficiency in the forward extraction process. After a full forward and backward extraction cycle, however, only the reversed micelles with n-butanol as the cosolvent was found to be able to maintain the activity of alpha-amylase in the stripping solution. By using the reversed micelles of Aliquat 336/isooctane/1% (v/v) n-butanol to perform a full extraction cycle, it was found that 85% of the total activity of alpha-amylase in the industrial a-amylase could be recovered at the end of an extraction cycle and the specific activity of alpha-amylase could be concentrated about 1.5-fold; meanwhile, most of the neutral protease in the industrial a-amylase could be removed. The separation factor of alpha-amylase to neutral protease at the end of an extraction cycle can reach about 10. (c) 1995 John Wiley & Sons, Inc.  相似文献

研究了CTAB-正辛醇-正庚烷交束溶液萃取牛血红蛋白(pHb)时、pH值、表面活性剂浓度、助表面活性剂浓度、离子种类和离子强度、溶剂比以及蛋白质浓度等因素对萃取效果的影响,并以蛋白质分子与表面活性剂分子间的相互作用以及反胶束大空间阻碍作用上进行了解释。研究表明,水相PH值在10.5 ̄12.5之间,KC1浓度为0.1mol/l,反胶束溶液中表面活性剂浓度为0.02mol/l,正辛醇与正庚烷之比为0.  相似文献

Reverse micellar extraction is a promising technique in large-scale bioseparation. However, low recovery and high salt concentration in back extraction limit its application. In CTAB/n-octane/n-hexanol reverse micellar system, the enzyme, pancreatic kallikrein could be effectively enwrapped into reverse micelles in forward extraction, but was difficult to be released during back extraction. In this study, dilute chaotropes (urea and GuHCl) were introduced to enhance the release of enzyme instead of high salts in back extraction. Kallikrein enwrapped in reverse micelles was released effectively in the presence of dilute urea and GuHCl during back extraction. Nearly 100% activity recovery of kallikrein from commercial product was obtained by adding 0.60 M urea, and for kallikrein from crude material, the recovery increased greatly by adding 0.80 M urea and 0.08 M GuHCl in the stripping solution. The mechanism of chaotrope for enhancing the release of enzyme from micelles was explored and dynamic light scatter analysis showed that the chaotrope would influence the sizes of micelles during reverse micellar extraction.  相似文献

Sodium di(2-ethylhexyl) sulfosuccinate, referred to as Aerosol-OT or AOT, was used to remove lysozyme from an aqueous phase via reverse micellar extraction and precipitation method. For both methods, when the surfactant was in excess, a complete removal of lysozyme from the aqueous phase was obtained at the values of pH below the pI of lysozyme. However, for the reverse micellar method, a solubilization limit of lysozyme in the organic phase was observed, and a white precipitate was formed at the aqueous-organic interface. This observation suggested using AOT directly as a precipitating ligand. The lysozyme precipitated with AOT was fully recovered, with its original enzymatic activity, using acetone as a recovery solvent. A mechanism is suggested to explain the solubilization of lysozyme in an AOT reverse micellar system. It is shown that a direct precipitation method can be used with advantage instead of using the reverse micellar extraction method to recover lysozyme from an aqueous phase.  相似文献
