The movement of kinetin has been investigated in roots of Zeaseedlings, grown in the dark, using kinetin-8-14C. Segments (10 and 11 mm) with and without an apex were used andthe following results obtained: The transport of kinetin is polarized basipetally. Kinetin-8-14C is transported in roots without vascular tissue,but the amount transported is always lower than in roots withvascular tissue. Thin-layer chromatography showed that the molecule of kinetinis metabolized in the root segments, adenine being one of theproducts.  相似文献

Auxin transport was investigated in excised stem segments ofNicotiana tabacum L. by the agar block technique using [1-14C]indol-3yl-acetic acid (IAA). The ability of the stems to transportauxin basipetally increased as secondary development proceeded;by contrast the ability of the pith to transport auxin declinedwith age. By separation of the stem tissues it was shown thatthe great majority of auxin transport took place in cells associatedwith the internal phloem and in cells close to the cambium;in both cases similar velocities of transport were found (c.5.0 mm h–1 at 22°C). The effects of osmotic gradientson auxin transport through the internal phloem were investigated.IAA was found by chromatography to account for practically allthe radioactivity in receiver blocks and other extracts of stemsegments. Pretreatment of 2?0 mm segments of etiolated zucchini (Cucurbitapepo L.) hypocotyl with cycloheximide (CH) or 2-(4-methyl-2,6-dinitroanilino)-N-methylpropionamide(MDMP) eliminated the stimulation by N-1-naphthylphthalamicacid (NPA) of net uptake of [1-14C]indol-3yl-acetic acid ([1-14C]IAA),but had relatively little effect on the net uptake of IAA inthe absence of NPA. The efflux of [1-14C]IAA from preloadedsegments was not substantially affected by inhibitor pretreatmentin the absence of NPA, but CH pretreatment significantly inhibitedthe reduction of efflux caused by NPA. Pretreatment with CHor MDMP did not affect net uptake by segments of the pH probe[2-14C]5,5-dimethyl-oxazolidine-2,4-dione ([2-14C]DMO), or thenet uptake of [14C]-labelled 3-O-methylglucose ([14C]3-0-MeGlu),suggesting that neither inhibitor affected intracellular pHor the general function of proton symporters in the plasma membrane.Both compounds reduced the incorporation of label from [35S]methionineinto trichloroacetic acid (TCA)-insoluble fractions of zucchinitissue, confirming their inhibitory effect on protein synthesis. The steady-state association of [3H]IAA with microsomal vesiclesprepared from zucchini hypocotyl tissue was enhanced by theinclusion of NPA in the uptake medium. The stimulation by NPAof [3H]IAA association with microsomes was substantially reducedwhen the tissue was pretreated with CH. However, CH pretreatmentdid not affect the level of high affinity NPA binding to themembranes indicating that treatments did not result in lossof NPA receptors. It is suggested that the auxin transport site on the effluxcarrier system and the receptor site for NPA may reside on separateproteins linked by a third, rapidly turned-over, transducingprotein. The effects of the morphactin 2-ehloro-9-hydroxyfluorene-9-carboxylicacid methyl ester [CFM] on growth, geotropic curvature and transportand metabolism of indol-3yl-acetic acid [IAA-5-3H] in the coleoptilesof Zea mays and A vena saliva have been investigated. A strongcorrelation has been found to exist between the inhibition ofthe geotropic response and the inhibition of auxin transport.CFM supplied at concentrations sufficient to abolish auxin transporthas been shown to promote the elongation of Zea, but not ofAvena, coleoptile segments. CFM does not change the patternof metabolism of IAA in Zea coleoptile segments. In these segmentsIAA is metabolized when its concentration is high, but the radioactivitytransported basipetally, or laterally in geotropically stimulatedcoleoptiles, is virtually confined to the IAA molecule. Radioactivityexported into the basal receiver blocks is wholly confined toIAA. It is concluded that CFM inhibits the geotropic responsein coleoptiles by suppression of the longitudinal and lateralauxin transport mechanisms. The growth-promoting propertiesof this substance cannot be linked with its effects on eitherauxin metabolism or transport.  相似文献

Reverse-phase high-performance liquid chromatography was usedto analyse [14C]-labelled metabolites of indole-3-acetic acid(IAA) in coleoptile segments of Zeo mays seedlings. After incubationfor 2 h in 10–2 mol m–3 [2-14C]IAA, methanolic extractsof coleoptiles contained between six and ten radioactive compounds,one of which co-chromatographed with IAA. The metabolic productsin coleoptile extracts appeared to be similar to those in rootextracts, with an oxindole-3-acetic-acid-like component as theprincipal metabolite, but the rate of metabolism was slowerin coleoptile than in root segments. Decarboxylation did notappear to play a major role in the metabolism of exogenous IAAduring the short incubation periods. Moreover, external IAAconcentration had little effect on the pattern of metabolism.Coleoptile segments were also supplied with [14C]IAA from agardonor blocks placed at the apical ends, and agar receiver blockswere placed at the basal ends. After incubation for 4 h, theidentity of the single radioactive compound in the receiverblocks was shown to be IAA by both reverse-phase high-performanceliquid chromatography and gas chromatography-mass spectrometrytechniques. The effect of ethylene on the uptake, decarboxylation and basipetaltransport of IAA-1-14C, IAA-2-14C and NAA-1-14C in cotton stemsections (Gossypium hirsutum L., var. Stoneville 213) was studied.A reduction in the capacity of cotton stem sections to transportauxin basipetally appears in sections excised from plants exposedto ethylene for only 3 hr and increases with fumigation time. In addition to reducing transport, increasing ethylene pretreatmentperiods from 3 to 15 hr also progressively reduced the uptakeof 14C and increased the release of 14C as 14CO2 from IAA-1-14C.The effect of ethylene on the decarboxylation of IAA-1-14C wassignificant when expressed as either the cpm of 14C releasedper hr per mg dry weight or the cpm released per hr per mm2in contact with the IAA donor. Comparative experiments usingIAA-1-14C and IAA-2-14C demonstrated that the effect of ethyleneon the decarboxylation of IAA was primarily a cut surface effectwhich apparently contributes to the reduction of IAA uptakeby ethylene. Although ethylene significantly reduced the transport of NAA-1-14C,uptake was significantly increased rather than decreased aswith IAA-1-14C while decarboxylation was unaffected. The movement and metabolism of kinetin-8-14C in the mesocotyland coleoptile of Zea mays have been investigated. The segments (10 mm) of coleoptile with and without an apexwere used. The presence of apex decreases the quantity of countswhich move basipetally in the segments. The quantity transported acropetally in the segments (apex incontact with receiver blocks) was about three times higher thanthat transported basipetally (apex in contact with donor blocks). Using 10 mm coleoptile segments excised I mm below the apex,there was no appreciable difference between acropetal and basipetalmovement. The transport of kinetin in the mesocotyl was studied with andwithout vascular tissue. The quantity transported in segmentswithout vascular tissue was always lower than in the segmentswith vascular tissue. The presence of the node between the coleoptile and mesocotyldoes not disturb the movement of kinetin in the basipetal position(node in contact with donor blocks). Thin layer chromatography of plant extracts after a 48-hourtransport period revealed several radioactive substances whichappeared to be adenine with Rf 0.5 and other components appearedat Rf 0.6.  相似文献

A concentration of 10–5 M tomatine had no effect on leakagefrom, or elongation of, wheat coleoptile segments, but consistentlyreduced IAA-enhanced extension growth by c. 50 per cent. Therewas no evidence of chemical interaction between the alkaloidand the auxin in solution, and IAA action was not affected bypre-treatment for up to 3 h with 10–5 M tomatine. Studieswith [2-14C]IAA revealed that 10–5 M tomatine did notinhibit uptake of auxin into segments. The effect of pre-treatingsegments for up to 3 h with IAA could be virtually nullifiedby 10–5 M tomatine, as could also IAA-induced changesin properties of coleoptile cell walls. Results are discussedin relation to the ability of tomatine to disrupt membrane functionand to current hypotheses implicating membranes in the primaryaction of auxin.  相似文献

When [1-14C]indol-3yl-acetic acid ([1-14C]IAA) was applied to the upper surface of a mature foliage leaf of garden pea (Pisum sativum L. cv. Alderman), 14C effluxed basipetally but not acropetally from 30-mm-long internode segments excised 4 h after the application of [1-14C]IAA. This basipetal efflux was strongly inhibited by the inclusion of 3.10–6 mol· dm3 N-1-naphthylphthalamic acid (NPA) in the efflux buffer. In contrast, when [14C] sucrose was applied to the leaf, the efflux of label from stem segments excised subsequently was neither polar nor sensitive to NPA. The [1-14C]IAA was initially exported from mature leaves in the phloem — transport was rapid and apolar; label was recovered from aphids feeding on the stem; and label was recovered in exudates collected from severed petioles in 20 mM ethylenediaminetetraacetic acid. No 14C was detected in aphids feeding on the stems of plants to which [1-14C]IAA had been applied apically, even though the internode on which they were feeding transported considerable quantities of label. Localised applications of NPA to the stem strongly inhibited the basipetal transport of apically applied [1-14C]IAA, but did not affect transport of [1-14C]IAA in the phloem. These results demonstrate for the first time that IAA exported from leaves in the phloem can be transferred into the extravascular polar auxin transport pathway but that reciprocal transfer probably does not occur. In intact plants, transfer of foliar-applied [1-14C]IAA from the phloem to the polar auxin transport pathway was confined to immature tissues at the shoot apex. Distal applications of indol-3yl-acetic acid (IAA) to debladed cotyledonary petioles of cotton (Gossypium hirsutum L.) seedlings greatly delayed petiole abscission, but similar applications of phenylacetic acid (PAA) slightly accelerated abscission compared with untreated controls. Both compounds prevented abscission for at least 91 h when applied directly to the abscission zone at the base of the petiole. The contrasting effects of distal IAA and PAA on abscission were correlated with their polar transport behaviour-[1-14C]IAA underwent typical polar (basipetal) transport through isolated 30 mm petiole segments, but only a weak diffusive movement of [1-14C]PAA occurred.Removal of the shoot tip substantially delayed abscission of subtending debladed cotyledonary petioles. The promotive effect of the shoot tip on petiole abscission could be replaced in decapitated shoots by applications of either IAA or PAA to the cut surface of the stem. Following the application of [1-14C]IAA or [1-14C]PAA to the cut surface of decapitated shoots, only IAA was transported basipetally through the stem. Proximal applications of either compound stimulated the acropetal transport of [14C]sucrose applied to a subtending intact cotyledonary leaf and caused label to accumulate at the shoot tip. However, PAA was considerably less active than IAA in this response.It is concluded that whilst the inhibition of petiole abscission by distal auxin is mediated by effects of auxin in cells of the abscission zone itself, the promotion of abscission by the shoot tip (or by proximal exogenous auxin) is a remote effect which does not require basipetal auxin transport to the abscission zone. Possible mechanisms to explain this indirect effect of proximal auxin on abscission are discussed.  相似文献

Greater concentrations of auxin at nodes than in internodes,resulting from some nodal barrier to basipetal transport, havelong been postulated as the cause of early differentiation ofinitially isolated xylem and cambium at the nodes. However,this study, using [14C] indole-3-acetic acid (IAA) applied apicallyand gas chromatography-mass spectrometry, found that in stemsofImpatiens sultanithe IAA concentrations (per unit f. wt) atnodes were similar to those in adjacent internodes, though alittle greater at nodes if expressed per unit length of stemand a little less per unit d. wt. By contrast, in decapitatedshoots and in stem explants of dicotyledons, loss of the apicalsource of basipetally flowing auxin can result in auxin drainagewith some auxin retention in the uppermost remaining nodes.When [14C]IAA was applied apically to shoots for 4 h and stemexplants were excised, the explants had no nodal accumulationinitially whereas comparable explants incubated for 20 h revealedsignificant nodal accumulation. If decapitation leads both tonodal auxin accumulation and to adventitious abscission justabove the node, this fits the hypothesis that abscission sitesare positioned where auxin concentration decreases locally inthe apical direction. If segments of Impatiens sultani stem are explanted and incubated,separation layers often form across them and lead to abscission.To test the suggested role of auxin concentration in controllingthe position of abscission sites, explants were labelled byapplying [14C]IAA to the shoot tip 4 h prior to explanting;transport of auxin applied in this way seems to resemble thatof endogenous auxin. During subsequent incubation of explantsfor 20 h, basipetal transport resulted in 14C accumulating justabove the base of the explants (nearly 80 % in the bottom 4mm of 24 mm explants). In internodal explants that had beenwounded at explanting by incising one side so as to sever avascular bundle, and in nodal explants with the leaf removed,the 14C also accumulated just above the wound or node to abouttwice the concentration otherwise expected; this accumulationwas probably due to basipetal transport being impeded by vasculardiscontinuity at the wound or node. Accumulation just abovethe base, or above a wound or node, resulted in gradients of14C concentration (presumably reflecting endogenous auxin concentration)decreasing in the morphologically upward direction at each ofthese three positions where abscission sites tend to occur. Stem segments of non-tumorous Nicotiana glauca and N. langsdorffiiplants and of their tumor-producing amphidiploid F1 hybrid weretreated with 6-furfurylaminopurine (kinetin) prior to transporttests with applied labeled indoleacetic acid (IAA-2-14C). Kinetin-treatmentsincreased the uptake of IAA in non-tumorous shoots; the IAAuptake by N. langsdorffii segments was increased up to 3-fold.The auxin uptake in stem-segments of the tumor-forming hybrid,however, could not be increased significantly by kinetin. Theeven distribution of IAA-14C in segments of normal and tumorproneNicotiana shoots is stimulated by kinetin. The biosynthetic pathway from D-glucose to L-(+)-tartaric acid(TA) in detached leaves of the bean, Phaseolus vulgaris L.,was studied in three cultivars, two of which were known to containTA and one of which lacked TA, with the aid of several putativeradiolabeled intermediates, namely D-[l-14C]glucose, D-[6-14C]glucose,D-[U-14C]glucose, D-[U-14C]gluconate, L-[U-14C]-ascorbic acid,L-[l-l4C]idonate, D-xylo-5-[U-14C]hexulosonate, D-xylo-5-[l-14C]hexulosonate,D-xylo-5-[6-l4C]hexulosonate and L-[U-l4C]threonate. D-[U-14C]Glucoseand D-[U-l4C]gluconate were converted to TA with low isotopicyield but this yield was further reduced when leaf tissues weresupplied with unlabeled D-gluconate or D-xylo-5-hexulosonate.D-xylo-5-[U-14C]Hexulosonate and D-xylo-5-[l-14C]hexulosonatewere good precursors of TA. D-xylo-5-[6-14C]Hexulosonate didnot furnish 14C to TA. Addition of a metabolic product of D-xylo-5-hexulosonate(which was labeled by D-xylo-5-[l-14C]hexulosonate but not byD-xylo-5-[6-14C]hexulosonate) to leaves labeled with D-xylo-5-[l-14C]hexulosonatedoubled the incorporation of 14C into TA. L-[U-14C]Ascorbicacid, L-[l-14C]idonate and L-[U-14C]threonate failed to producelabeled TA. A metabolic scheme to accommodate these observationsis presented. Transport of 14C-photosynthate in decapitated stems of Phaseolusvulgaris explants was dependent on the concentration of indole-3-aceticacid (IAA) applied to the cut surfaces of the stem stumps. Thephysiological age of the stem influenced the nature of the transportresponse to IAA with stems that had ceased elongation exhibitinga more pronounced response with a distinct optimum. Increasednutrient status of the explants had little influence on theshape of the IAA dose-response curve but increased, by two ordersof magnitude, the IAA concentration that elicited the optimalresponse. Applications of the inhibitor of polar auxin transport,1-(2-carboxyphenyl)-3-phenylpropane-1, 3-dione (CPD), affectedIAA-promoted transport of 14C-photosynthates. At sub-optimalIAA concentrations, CPD inhibited transport, whereas at supra-optimalIAA concentrations, 14C-photosynthate transport was marginallystimulated by CPD. Treatment with CPD resulted in a significantreduction in stem levels of [14C]IAA below the site of inhibitorapplication, while above this point, levels of [14C]1AA remainedunaltered. The divergent responses of auxin-promoted transportto CPD treatment are most consistent with a remote action ofIAA on photosynthate transport in the decapitated stems. The effects of applied ethylene on the growth of coleoptilesand mesocotyls of etiolated monocot seedlings (oat and maize)have been compared with those on the epicotyl of a dicot seedling(the etiolated pea). Significant inhibition of elongation by ethylene (10 µll–1for 24 h) was found in intact seedlings of all three species,but lateral expansion growth was observed only in the pea internodeand oat mesocotyl tissue. The sensitivity of the growth of seedlingparts to ethylene is in the decreasing order pea internode,oat coleoptile and oat mesocotyl, with maize exhibiting theleast growth response. Although excised segments of mesocotyland coleoptile or pea internode all exhibit enhanced elongationgrowth in IAA solutions (10–6–2 ? 10–5 moll–1), no consistent effects were found in ethylene. Ethyleneproduction in segments was significantly enhanced by applicationof auxin (IAA, 10–5 mol l–6 or less) in all tissuesexcept those of the eat mesocotyl. Segments of maize show a slow rate of metabolism of applied[2-14C]IAA (30 per cent converted to other metabolites within9 h) and a high capacity for polar auxin transport. Ethylene(10 µl l–1 for 24 h) has little effect on eitherof these processes. The oat has a smaller capacity for polartransport than maize and the rate ef metabolism of auxin isas fast as in the pea (90 per cent metabolized in 6 h). Althoughethylene pretreatment does not change the rate of auxin metabolismin oat, there is a marked reduction in auxin transport. It is proposed that the insensitivity of maize seedlings toethylene is related to the supply and persistence of auxin whichcould protect the seedling against the effects of applied orendogenously produced ethylene. Although the mesocotyl of oatis sensitive to applied ethylene it may be in part protectedagainst ethylene in vivo by the absence of an auxin-enhancedethylene production system. The results are discussed in relationto a model for the auxin and ethylene control of cell growthin the pea.  相似文献

The light-catalysed isomerization of (+)-abscisic acid (ABA)to its trans isomer during isolation from leaves was monitoredby the addition of (±)-[2-14C]ABA to the extraction medium.(+)Trans-abscisic acid (t-ABA) was found to occur naturallyin rose (Rosa arvensis) leaves at 20µg/kg fresh weight;(+)-ABA was present at 594µg/kg. (±)-[2-14D]Trans-abscisicacid was not isomerized enzymically to ABA in tomato shoots. (±)-Abscisic acid was converted by tomato shoots to awater-soluble neutral product, ‘Metabolite B’, whichwas identified as abscisyl-ß-D-glucopyranoside. When(±)-[2-14C]trans-abscisic acid in an equimolar mixturewith (±)-[2-14C}ABA was fed to tomato shoots it was convertedto its glucose ester 10 times faster than was ABA. Trans-abscisyl-ß-D-glucopyrano8ide only was formedfrom (±)-[2-14C]t-ABA in experiments lasting up to 30h. Glucosyl abscisate was formed slowly from ABA and the freeacid fraction contained an excess of the unnatural (–).ABAas did the ABA released from abscisyl-ß-D-glucopyranosideby alkaline hydrolysis. The (+).ABA appeared to be the solesource of the acidic ‘Metabolite C" previously noted. The concentrations of endogenous (+)-, (+)-[2-14C]-, and (–)-[2-14C]ABAremaining as free acid, and also in the hydrolysate of abscisyl-ß-D-glucopyranoside,were measured by the ORD, UV absorption, and scintillation spectrometryof highly purified extracts of ABA from tomato shoots whichhad been supplied with racemic [2-l4C]ABA.  相似文献

The longitudinal and lateral transport of 2, 4-D-[1-14C] hasbeen studied in 6-mm horizontally disposed segments of the hypocotylof Helianthun annuus. The technique involved the asymmetricapplication of the auxin in agar donor blocks to either theupper or lower halves of the cut surface of the segments andthe measurement of 14C accumulating with time in split receiverblocks on the upper and lower halves of the cut surfaces atthe opposite ends. A highly significant effect of gravity inducinga polar migration of 2, 4-D to the lower side has been establishedand represents, under optimal conditions, about 10 per centof the total 2, 4-D in transit. This lateral polar movementshows a tendency to saturate at higher donor concentrations(5 mg/1) and is affected by temperature in precisely the sameway as the basipetal longitudinal transport. The optimal flux(intensity of transport) occurs at about 35 °C or slightlyabove, and above 40 °C both transport systems are seriouslyimpaired. There is some evidence that gravity increases thevelocity of 2, 4-D transport in the lowermost tissues of thehorizontal hypocotyl but does not affect the transport intensity.  相似文献

The terminal (1-year-old) shoot of dormant, 2-year-old Abies balsamea (L.) Mill. seedlings was ringed with 0 or 10 mg Ethrel g-1 lanolin. After 5 weeks of culture under environmental conditions favorable for growth, some of the treated shoots were harvested to measure tracheid number by microscopy and ethylene evolution by gas chromatography-flame ionization detection. The remaining shoots were used to measure basipetal IAA transport in the cambial region by decapitating the shoot apex, applying a pulse of [1-14C]-IAA to the cut surface, and monitoring the subsequent distribution of radioactivity. Ringing with 10 mg Ethrel g-1 lanolin, compared with lanolin alone, stimulated cambial region ethylene evolution about 26-fold at, and 3-fold above and below the ringing site, but promoted tracheid production at the ringing site only. Ethrel ringing also increased the velocity, after 26 h transport, at which the front of the [1-14C]-IAA pulse moved below the ringing site. After 72 h of [1-14C]-IAA transport, when only immobilized radioactivity was present, the amount of radioactivity recovered in shoots ringed with 10 mg Ethrel g-1 lanolin was higher than with lanolin alone at the ringing site but lower below it. No difference was observed above the ringing site. The distribution of radioactivity was the same in shoots ringed with lanolin alone and in unringed shoots. The results support the hypothesis that an abnormally high cambial region concentration of ethylene derived from Ethrel ringing inhibits the capacity of basipetal IAA transport at the ringing site, resulting in a local accumulation of IAA that stimulates tracheid production.  相似文献

Phloem transport in stems of Phaseolus vulgaris was found tobe sensitive to treatment with the auxin transport inhibitor,2,3,5-triidobenzoic acid (TIBA). The response was dependenton the concentration of TIBA applied. A concentration of TIBA(0?5% in lanolin) which did not interfere with normal phloemtransport proved inhibitory to both basipetal transport of IAAand the acropetal component of IAA-promoted metabolite transport.In contrast, both acropetal IAA transport and basipetal IAA-promotedmetabolite transport were unaffected by TIBA treatment. Theinhibitory effect of TIBA on acropetal IAA-promoted transportwas overcome by providing IAA below the point of TIBA application.Both acropetal and basipetal IAA-promoted transport in stemsegments were unaccompanied by any corresponding changes inthe accumulation of [14C]sucrose by the segments.  相似文献
