Evaluation of enzyme activities in combination with taxonomic analyses may help define the mechanisms involved in microbial decomposition of orgaic amendments and biological control of soilborne pathogens. In this study, powdered pine bark was added to nematode-infested soil at rates of 0, 5, 10, 15, 20, 25, 30, 35, 40, 45, and 50 g kg–1. Total fungal populations did not differ among treatments immediately after application of pine bark. After 7 days, fungal populations were positively correlated with increasing levels of pine bark. This increase was sustained through 14 and 21 days.Penicillium chrysogenum andPaecilomves variotii were the predominant fungal species isolated from soil amended with pine bark. Total bacterial populations did not change with addition of pine bark at 0, 7, and 14 days after treatment. At 21 and 63 days, total bacterial populations declined in soil receiving the highest rates of pine bark. Addition of pine bark powder to soil caused a shift in predominant bacterial genera fromBacillus spp. in nonamended soil, toPseudomonas spp. in amended soil. Soil enzyme activities were positively correlated with pine bark rate at all sampling times. Trehalase activity was positively correlated with total fungal populations and with predominant fungal species, but was not related to bacterial populations. The number of non-parasitic (non-stylet bearing) nematodes andMeloidogyne arenaria in soil and roots were not correlated with pine bark rate. Concentrations of ions were measured in soil solutions from beech (Nothofagus) forests in remote areas of New Zealand and in solutions from beech (Fagus sylvatica) and Norway spruce (Picea abies) forests in North-East Bavaria, West Germany, to compare the chemistry of soil solutions which are unaffected by acid deposition (New Zealand) with those that are affected (West Germany). In New Zealand, soil solution SO4 2– concentrations ranged between <2 and 58 mol L–1, and NO3 concentrations ranged between <1 and 3 mol L–1. In West Germany, SO4 2– concentrations ranged between 80 and 700 mol L–1, and NO3 concentrations at three of six sites ranged between 39 and 3750 mol L–1, but was not detected at the remaining three sites. At all sites in New Zealand, and at sites where the soil base status was moderately high in West Germany, pH levels increased, and total Al (Alt) and inorganic monomeric Al (Ali) levels decreased rapidly with increasing soil depth. In contrast, at sites on soils of low base status in West Germany, pH levels increased only slightly, and Al levels did not decline with increasing soil depth.Under a high-elevation Norway spruce stand showing severe Mg deficiency and dieback symptoms in West Germany, soil solution Mg2+ levels ranged between 20 and 60 mol L, and were only half those under a healthy stand. Alt and Ali levels were substantially higher the healthy stand than under the unhealthy stand, indicating that Al toxicity was not the main cause of spruce decline.  相似文献

Flax anther culture: effect of genotype,cold treatment and media   总被引:2,自引:0,他引:2  
We report on screening of wide range of flax cultivars for androgenic response and on testing of induction conditions for flax (Linum usitatissimum L.) anther culture and plant regeneration. Anthers were cultured on four different media: Mo, N6, MS and N&N supplemented with various combinations of growth regulators. The induction of callus formation from cultured anthers was the highest on N6 (with cultivar PR FGL 77 – 12 %) and N&N media (with cultivar Carolin – 2.8 %), preferentially after cold pretreatment (7days at 8 °C). Shoots were formed on calli derived from the microspores inside the cultured anthers on media N&N and N6 supplemented with 1mgl–1 zeatin or 1mgl–1BAP + 1mgl–1NAA, respectively and elongated on MS medium supplemented with 2mgl–1 zeatin. The highest number of shoots (120) was observed with cultivar Red Wing. Shoots were rooted on MS medium supplemented with 2mgl–1IAA. Our experiments resulted in total in 62 % anther response and 155 plants regenerated and transferred into soil.  相似文献

The distribution of labile Cd and Zn in two contrasting soils was investigated using isotopic exchange techniques and chemical extraction procedures. A sewage sludge amended soil from Great Billings (Northampton, UK) and an unamended soil of the Countesswells Association obtained locally (Aberdeen, UK) were used. 114Cd and 67Zn isotopes were added to a water suspension of each soil and the labile metal pool (E-value) determined from the isotope dilution. Samples were obtained at 13 time points from 1h to 50 days. For the sewage sludge amended soil, 29 g Cd g–1 (86% of total) and 806 g Zn g–1 (65% of total) were labile and for the Countesswells soil the value was 8.6 g Zn g–1 (13% of total); limits of detection prevented a Cd E-value from being measured in this soil. The size of the labile metal pool was also measured by growing plants for 90 days and determining the isotopic content of the plant tissue (L-value). Thlaspi caerulescensJ. & C. Presl (alpine penny cress), a hyperaccumulator of Zn and Cd, Taraxacum officinale Weber (dandelion) and Hordeum vulgare L. (spring barley) were used. L-values were similar across species and lower than the E-values. On average the L-values were 23±0.8 g Cd g–1 and 725±14 g Zn g–1 for the Great Billings soil and 0.29±0.16 g Cd g–1 and 7.3±0.3 g Zn g–1 for the Countesswells soil. The extractable metal content of the soils was also quantified by extraction using 0.1 M NaNO3, 0.01 M CaCl2, 0.5 M NaOH, 0.43 M CH3COOH and 0.05 M EDTA at pH 7.0. Between 1.3 and 68% of the total Cd and between 1 and 50% of the total Zn in the Great Billings soil was extracted by these chemicals. For the Countesswells soil, between 6 and 83% of the total Cd and between 0.1 and 7% of the total Zn was extracted. 0.05 M EDTA and 0.43 M CH3COOH yielded the greatest concentrations for both soils but these were less than the isotopic estimates. On the whole, E-values were numerically closer to the L-values than the chemical extraction values. The use of isotopic exchange provides an alternative estimate of the labile metal pool within soils compared to existing chemical extraction procedures. No evidence was obtained that T. caerulescens is able to access metal within the soil not freely available to the other plants species. This has implications for long term remediation strategies using hyperaccumulating plant species, which are unlikely to have any impact on non-labile Cd and Zn in contaminated soil.  相似文献

Methane in sulfate-rich and sulfate-poor wetland sediments   总被引:2,自引:1,他引:1  
Wetlands of northern Belize provide a unique opportunity to study methane production and emissions from marshes dominated by identical species (Typha domingensis, Cladium jamaicense) and genus (Eleocharis spp.), but differing substantially in the amount of sulfates present in the sediments. Some marshes occur on limestone marls rich in gypsum (CaSO4) while others are underlain by alluvial sands poor in sulfates. Concentrations of methane and sulfates in the sediment interstitial water are one or more orders of magnitude different for these two geological substrata averaging 139.2 and 14.9 M of CH4, and 0.08 and 11.53 mM of SO4 –2 2 on alluvial sands and limestone respectively. The amount of methane found in the internal atmosphere of plants from alluvial sands is significantly higher (6.3 M) than in plants from limestone (0.19 M). The average methane emissions measured in wetlands located on alluvial sands were 25.2 mg m–2 h–1 while emissions from marshes on limestone were only 2.4 mg m–2 h–1. These values extrapolated for the entire year and the respective wetland areas resulted in the estimate of total CH4 emissions from northern Belize of 0.066 Tg per year.  相似文献

The preparation of a GM1-ganglioside (GM1) [14C]-labelled in the sialic acid residue is reported. This can be obtained by re-N-acetylation in the presence of [1-14C]-acetic anhydride, of a GM1 derivative de-N-acetylated specifically on the sialic acid residue by alkaline hydrolysis of GM1 with tetramethylammonium hydroxide. The radiolabelled GM1 is utilized to investigate the binding properties and the mode of interaction of GM1 with cultured fibroblasts. Three different forms of association (one serum-removable, one trypsin-removable and one trypsin-stable) have been recognized to occur in a way that depended on cell culture conditions (presence or absence of fetal calf serum), ganglioside concentration (from, 5×10–9 M to 10–4 M) and incubation time (up to 24 h). Summary The frequency of intra- and interchromosomal recombination was determined in RAD18 and rad18 deletion and rad18-3 mutant strains. It was found that spontaneous interchromosomal recombination at trp5, his1, ade2, and MAT was elevated 10- to 70-fold in the rad18-3 and rad18 mutants as compared to the RAD + strains. On the other hand the frequencies of spontaneous intrachromosomal recombination for the his33, his35 and the his4C , his4A duplications and for heterothallic mating type switching were only marginally elevated in the rad18 deletion mutant, and recombination between ribosomal DNA repeats was only 2-fold elevated in the rad18-3 mutant. These differences may be due to a haploid versus diploid specific difference. However interchromosomal recombination was elevated 40-fold and intrachromosomal recombination was only marginally (1.5-fold) elevated in a diploid homozygous for rad18, arguing against a haploid versus diploid specific difference. Possible explanations for the difference in the elevated levels of intra- versus interchromosomal spontaneous recombination are discussed.  相似文献

A chlorine-containing non-protein amino acid which was recently discovered from the fruit bodies ofAmanita gymnopus (2S)-2-amino-5-chloro-4-hydroxy-5-hexenoic acid, was isolated and crystallized for the first time from the fruit bodies of an unknown member ofAmanita belonging to the sectionRoanokenses, subsectionSolitariae. Senescence-induced changes in the xanthophyll cycle activity and chlorophyll (Chl) fluorescence parameters were compared in detached barley (Hordeum vulgare L.) leaf segments kept for 6 d in darkness or under continuous white light (90 mol m–2 s–1). Before detachment of the leaf segments, the plants were grown at periodic regime [12 h light (90 mol m–2 s–1)/12 h dark]. The de-epoxidation state of the xanthophyll cycle pigments (DEPS) in the leaf samples was determined immediately (the actual DEPS), after 1 h of dark-adaptation (the residual DEPS), and during 14 min of a high-irradiance (HI) exposure (500 mol m–2 s–1) (HI-induced DEPS). In the light-senescing segments, senescence was delayed pronouncedly compared to dark-senescing ones as the Chl content, the photosystem 2 photochemistry, and electron transport processes were highly maintained. Further, the actual DEPS increased, probably due to the increased mean photon dose. The HI-induced increase in the DEPS was stimulated in the light-senescing segments, whereas it was slowed down in the dark-senescing ones. However, after the 14 min HI-exposure of the dark-senescing segments the HI-induced DEPS was not markedly lower than in the mature leaves, which indicated the maintenance of the xanthophyll cycle operation.  相似文献

A tissue culture procedure was developed for the establishment and propagation of a colchi-autotetraploid of Rauvolfia serpentina for possible commercial exploitation. Multiplication of autotetraploid shoots was obtained either through axillary bud elongation on Murashige and Skoog [1] medium (MS) containing 2.65 M (0.5 mgl–1) -naphthaleneacetic acid and 0.33 M (0.05 mgl–1) kinetin, or via multiple shoot formation on MS medium supplemented with 4.44 M (1.0 mgl–1) 6-benzylaminopurine and 0.53 M (0.1 mgl–1) -naphthaleneacetic acid. Rooting could be induced by transferring the shoots to MS medium containing 7.95 M (1.5 mgl–1) -naphthaleneacetic acid alone. The plantlets, thus formed, were tetraploid in nature by cytological observations of the root tips. They exhibited 80–90% success in establishment under glass house and field conditions.  相似文献

The tetrasaccharides GalNAcß1-4[NeuAc2-3]Galß1-4Glc and GalNAcß1-4[NeuAc2-3]Galß1-4GlcNAc were synthesised by enzymic transfer of GalNAc from UDP-GalNAc to 3-sialyllactose (NeuAc2-3Galß1-4Glc) and 3-sialyl-N-acetyllactosamine (NeuAc2-3Galß1-4GlcNAc). The structures of the products were established by methylation and1H-500 MHz NMR spectroscopy. In Sda serological tests the product formed with 3-sialyl-N-acetyllactosamine was highly active whereas that formed with 3-sialyllactose had only weak activity.  相似文献

Globally, land-use change is occurring rapidly, and impacts on biogeochemical cycling may be influenced by previous land uses. We examined differences in soil C and N cycling during long-term laboratory incubations for the following land-use sequence: indigenous forest (soil age = 1800 yr); 70-year-old pasture planted after forest clearance; 22-year-old pine (Pinus radiata) planted into pasture. No N fertilizer had been applied but the pasture contained N-fixing legumes. The sites were adjacent and received 3–6 kg ha–1 yr–1volcanic N in rain; NO3 -N leaching losses to streamwater were 5–21 kg ha–1 yr–1, and followed the order forest < pasture = pine. Soil C concentration in 0–10 cm mineral soil followed the order: pasture > pine = forest, and total N: pasture > pine > forest. Nitrogen mineralization followed the order: pasture > pine > forest for mineral soil, and was weakly related to C mineralization. Based on radiocarbon data, the indigenous forest 0–10 cm soil contained more pre-bomb C than the other soils, partly as a result of microbial processing of recent C in the surface litter layer. Heterotrophic activity appeared to be somewhat N limited in the indigenous forest soil, and gross nitrification was delayed. In contrast, the pasture soil was rich in labile N arising from N fixation by clover, and net nitrification occurred readily. Gross N cycling rates in the pine mineral soil (per unit N) were similar to those under pasture, reflecting the legacy of N inputs by the previous pasture. Change in land use from indigenous forest to pasture and pine resulted in increased gross nitrification, net nitrification and thence leaching of NO3 -N.  相似文献

2,3-Sialylation of the lactosamine type N-glycans with trans-sialidase from Trypanosoma cruzi is reported. Trans-sialidase (160 kDa, pI 5.35–5.65) and its catalytic fragment (70 kDa, pI 6.0–6.3) were isolated from T. ruzi cells and immobilized on ConA-Sepharose. The resulting preparation retained its activity for several months and was repeatedly used for obtaining mono-, di-, tri-, and tetrasialylated 7-amino-4-metylcoumarin-labeled oligosaccharides with various numbers of antennas and for 2,3-sialylation of glycans within glycoproteins and neoglycoconjugates.  相似文献

Summary Addition of marl (CaCO3) and/or manure to an acid soil in pots caused lime-induced chlorosis inPinus radiata seedlings, especially after excessive irrigation for 15 days. Chlorotic symptoms and their intensity were found to be related more to soil moisture and to the HCO3 concentrations, than to percentage of free CaCO3, in the soil mixtures.Comparative chemical analysis showed lower total Fe and Mn concentrations and higher concentrations of cations and organic anions in the needles of seedlings with chlorotic symptoms than in the needles of healthy ones.  相似文献

Phytotoxicity and inhibitory effects of the fusarial toxins fumonisin B1 (FB1) [m.p. 103–105 °C], fusaric acid [m.p. 106–107 °C], butenolide (4-acetamido-4-hydroxy-2-butenoic acid lactone) [116–117 °C], 9, 10-dihydroxyfusaric acid [m.p. 150–155 ° C], and moniliformin on chlorophyll synthesis in the aquatic macrophyte Lemna minor (duckweed) were examined. FB1 proved to be most active, reducing the growth of L. minor fronds and their ability to synthesize chlorophyll by 53% and 59%, respectively, at 0.7 g/ml. The growth rate of L. minor was reduced 59% by 6.7 g/ml fusaric acid, 62% by 66.7 g/ml butenolide, and 22% by 66.7 g/ml 9,10-dihydroxyfusaric acid. Moniliformin was the least phytotoxic to L. minor, with only a 16% suppression of growth rate and a 54% reduction in chlorophyll at 66.7 g/ml.The mention of firm names or trade products does not imply that they are endorsed or recommended by the US Department of Agriculture over other firms or similar products not mentioned.  相似文献

Summary A procedure for the regeneration of fertile transgenic white mustard (Sinapis alba L.) is presented. The protocol is based on infection of stem explants of 7–9 day old plants with an Agrobacterium tumefaciens strain harboring a disarmed binary vector with chimeric genes encoding neomycin phosphotransferase and -glucuronidase. Shoots are regenerated from callus-forming explants within 3–4 weeks. Under selection, 10% of the explants with transgenic embryonic callus develop into fertile transgenic plants. Rooting shoots transferred to soil yield seeds within 14–16 weeks following transformation. Integration and expression of the T-DNA encoded marker genes was confirmed by histochemical glucuronidase assays and Southern-DNA hybridization using primary transformants and S1-progeny. Seedlings ofAlnus incana (nodulated and non-nodulated) andBetula papyrifera were fertilized with varying amounts (0, 10, 25, 50, 100, 250 and 500 g N g–1 soil) of labelled ammonium-N or nitrate-N ( 5.2 A% excess15N as ammonium sulphate or potassium nitrate). After 4 months in the greenhouse,15N excess in the plants were determined and an isotope dilution equation was applied to determine the percent of biomass N fixed by theA. incana/Frankia system. When ammonium was used as the sole N source and birch as the non-fixing reference, N-fixation accounted for 95%, 87% and 60% of the plant nitrogen yields with 10, 25 and 50 g N g–1 rates, additions respectively. At the 100 g N g–1 fertilization and above N-fixation accounted for less than 10% of the N yield. Similar results were obtained when non-nodulatedA. incana was used as non-fixing reference. With nitrate as the sole N source, N-fixation accounted for 98%, 97%, 97%, 86%, 56% and 12% of N yield with 10, 25, 50, 100, 250 and 500 g N g–1 additions respectively. These values were similar for both types of reference plants. The direct isotope dilution method was compared to that of the total nitrogen difference method. There was good agreement between the two methods up to 50 g N g–1 for ammonium and up to 100 g N g–1 for nitrate. The difference method produced negative values at high concentrations of nitrogen fertilization. Again similar results were obtained by the two reference plants. The results indicate that birch can be used as a non-fixing control in isotope dilution studies but that care must be exercised in selecting the type and quantity of labelled nitrogen fertilizer.  相似文献

A field experiment was carried out at two sites off Yucatan State, Southeast Mexico, in order to determine the feasibility of culturing the red seaweedEucheuma isiforme; this was done during May–September 1989. At both sites (Uaymitun and Dzilam) the 25 days harvest and 14 algae per line plant density growth rates (2.21% day–1 and 1.21% day–1, respectively) were significantly higher (p<0.05) than those obtained with other combinations of the two factors tested (50, 75, 100 and 125 days harvest and 9 and 14 algae per line plant density). The mean carrageenan content of the cultured algae was 35.8% and 31.4% at Uaymitun and Dzilam, respectively.  相似文献

Strawberry (Fragaria ananassaDuch. cv. Fengxiang) plantlets were cultured under two in vitroenvironments for rooting, and then acclimatized under two ex vitroirradiance conditions. At the end of rooting stage plant height, fresh weight and specific leaf area of T1-plants grown under high sucrose concentration (3 sucrose), low photosynthetic photon flux density (30 mol m–2 s–1) and normal CO2 concentration (350–400 l l–1) were significantly higher than those of T2-plantlets grown under low sucrose concentration (0.5), high photosynthetic photon flux density (90 mol m–2 s–1) and elevated CO2 concentration (700–800 l l–1). But T2-plantlets had higher net photosynthetic rate (Pn), effective photochemical quantum yield of PSII (PSII), effective photosynthetic electron transport rate (ETR), photochemical quenching (qP) and ratio of chlorophyll fluorescence yield decrease (Rfd). After transfer, higher irradiance obviously promoted the growth of plantlets and was beneficial for the development of photosynthetic functions during acclimatization. T2-plantlets had higher fresh weight, leaf area, PSII and ETR under higher ex vitroirradiance condition.  相似文献

The determination of thelacZ mutant frequency in gt10lacZ phage vectors isolated from the transgenic mouse strain 40.6 (MutaMouse), requires the screening of large numbers of phages on -galactosidase activity. Existing methods rely on distinguishing a few white plaques on X-gal containing plates amongst a multide of blue ones which is both time-consuming and expensive. The new screening method described here employs the galactose sensitiveEscherichia coli C lacZ recA galE strain into which a multicopy plasmid has been introduced, which results in over-expression of thegalK andgalT genes. In the presence of phenyl--d-galactopyranoside, a substrate for -galactosidase, this leads to the suppression of lacZ + phage propagation without affecting the ability of lacZ phages to form plaques. With this method it is possible to screen 1.5×106 phages on a single 9-cm Petri dish. Furthermore, the need for blue/white screening has been eliminated.  相似文献
