通过5年10季的小麦/谷子轮作盆栽试验,持续观测了每季0、2.25和22.5 t·hm-2黑炭施用下作物生长和土壤性质变化及氨挥发数量.结果表明:相同NPK肥施用下,与不施黑炭处理相比,黑炭处理能促进作物生长,提高土壤养分供应.22.5 t·hm-2黑炭处理下5年作物累积籽粒和秸秆产量分别增加24.1%和74.1%;相应地上部N、P和K累计吸收量分别增加93.5%、71.2%和46.3%;轮作结束后土壤有效P、K含量及阳离子交换量分别提高262%、274%和58.3%,有机碳提高843%,C/N增至25,容重降低46.6%,土壤pH值则无明显变化.2.25和22.5 t·hm-2黑炭处理的黑炭表观分解率为每年3.5%~5.7%.高量黑炭施用可导致氨挥发数量增加,22.5 t·hm-2黑炭处理下10季氨挥发总量增加102%.  相似文献

Near-isogenic lines (NILs) are ideal materials for precise estimation of quantitative trait loci (QTL) effects and map-based gene isolation. With the completion of the rice genome sequence, QTL isolation based on NILs is becoming a routine. In this study, a trait-performance derived NIL strategy was adopted to develop NILs. Two plants were identified within one inbred line of recombinant inbred lines (RILs, F7 generation), exhibiting a significant difference in panicle size. By marker screening of the whole genome the genetic background of the two plants was estimated to be 98.7% identical. These two plants were selected as parents to produce a near-isogenic F2 (NIL-F2) population, consisting of 125 individuals, in which spikelets per panicle (SPP), grains per panicle (GPP), heading date (HD) and plant height (PH) were recorded. These four traits expressed discontinuous or bimodal distribution in the NIL-F2 population and followed the expected segregation ratios for a single Mendelian factor by progeny tests. A partial dominant QTL for the four traits was mapped to the same interval flanked by RM310 and RM126 on chromosome 8. The QTL region explained 83.0, 80.2, 94.9 and 93.8% of trait variation of SPP, GPP, HD and PH in the progenies, respectively. Progeny tests also confirmed co-segregation of QTL for the four traits, tall plants consistently flowering late and carrying large panicles. 施用生物炭6年后对稻田土壤酶活性及肥力的影响   总被引:4,自引:0,他引:4  
利用田间定位试验,研究0(BC0)、7.5(BC1)、15(BC2)和22.5(BC3)t·hm-2水稻秸秆生物炭及3.75 t·hm-2水稻秸秆(STR)一次性施加6年后对稻田土壤肥力及酶活性的影响.结果表明: 施用生物炭6年后土壤有机碳、有效磷和速效钾含量显著增加,增幅分别为34.6%、12.4%和26.2%,土壤pH值和容重显著降低,但对土壤全氮含量无显著影响.土壤脲酶和酸性磷酸酶的活性显著增加,土壤荧光素二乙酸酯酶(FDA水解酶)和芳基硫酸酯酶的活性受到不同程度的抑制,其中,BC2处理的土壤脲酶活性增加量最大,增幅为36.5%.土壤酸性磷酸酶活性随着生物炭施加量的增加而增加,与土壤速效磷含量呈显著正相关关系;土壤FDA水解酶和脲酶主要与土壤速效钾含量有关;酸性磷酸酶和芳基硫酸酯酶与土壤容重呈显著正相关.施用生物炭6年后土壤脱氢酶和多酚氧化酶活性明显升高,增幅分别为48.8%和27.5%,而过氧化氢酶活性逐渐下降,且显著低于对照BC0.STR处理显著增加了土壤脲酶、FDA水解酶、脱氢酶、酸性磷酸酶和芳基硫酸酯酶的活性,降低了过氧化氢酶和多酚氧化酶的活性,降幅分别为23.4%和15.9%.  相似文献

利用绿豆(Vigna radiata)品种苏绿16-10和潍绿11杂交构建的F2和F3群体发掘调控绿豆产量相关性状的遗传位点。同时对绿豆产量相关性状进行表型鉴定和相关性分析,并利用构建的遗传连锁图谱进行QTL定位。结果表明,单株产量与单株荚数、单荚粒数、百粒重和分枝数均呈正相关。单株产量与单株荚数的相关性最高,这2个性状在F2和F3群体中的相关系数分别为0.950和0.914。在F2群体中,共检测到8个与产量性状相关的QTL位点,其中与单株荚数、单荚粒数和单株产量相关的QTL位点各1个,分别解释11.09%(qNPP3)、17.93%(qNSP3)和14.18%(qYP3)的表型变异;2个与分枝数相关的QTL位点qBMS3和qBMS11,分别解释18.51%和7.06%的表型变异;3个与百粒重相关的QTL位点qHSW3、qHSW7和qHSW10,分别解释5.33%、46.07%和4.24%的表型变异。在F3群体中,qNSP3和qHSW7再次被检测到,表明这2个QTLs有较好的遗传稳定性。同时,开发了1个与百粒重主效QTLqHSW7紧密连锁的InDel标记R7-13.4,并利用自然群体对...  相似文献

The thousand-grain weight and spikelets per panicle directly contribute to rice yield. Heading date and plant height also greatly influence the yield. Dissection of genetic bases of yield-related traits would provide tools for yield improvement. In this study, quantitative trait loci (QTL) mapping for spikelets per panicle, thousand-grain weight, heading date and plant height was performed using recombinant inbred lines derived from a cross between two diverse cultivars, Nanyangzhan and Chuan7. In total, 20 QTLs were identified for four traits. They were located to 11 chromosomes except on chromosome 4. Seven and five QTLs were detected for thousand-grain weight and spikelets per panicle, respectively. Four QTLs were identified for both heading date and plant height. About half the QTLs were commonly detected in both years, 2006 and 2007. Six QTLs are being reported for the first time. Two QTL clusters were identified in regions flanked by RM22065 and RM5720 on chromosome 7 and by RM502 and RM264 on chromosome 8, respectively. The parent, Nanyangzhan with heavy thousand-grain weight, carried alleles with increased effects on all seven thousand-grain weight QTL, which explained why there was no transgressive segregation for thousand-grain weight in the population. In contrast, Chuan7 with more spikelets per panicle carried positive alleles at all five spikelets per panicle QTL except qspp5. Further work on distinction between pleiotropic QTL and linked QTL is needed in two yield-related QTL clusters.  相似文献

A quantitative trait locus (QTL) for grain weight (GW) was detected near SSR marker RM210 on chromosome 8 in backcross populations derived from a cross between the Korean japonica cultivar Hwaseongbyeo and Oryza rufipogon (IRGC 105491). The O. rufipogon allele increased GW in the Hwaseongbyeo background despite the fact that O. rufipogon was the small-seeded parent. Using sister BC3F3 near-isogenic lines (NILs), gw8.1 was validated and mapped to a 6.1 cM region in the interval between RM42 and RM210 (P≤0.0001). Substitution mapping with eight BC3F4 sub-NILs further narrowed the interval containing gw8.1 to about 306.4 kb between markers RM23201.CNR151 and RM30000.CNR99. A yield trial using homozygous BC3F4 sister sub-NILs and the Hwaseongbyeo recurrent parent indicated that the NIL carrying an O. rufipogon chromosome segment across the entire gw8.1 target region out-yielded its sister NIL (containing Hwaseongbyeo chromosome in the RM42–RM210 interval) by 9% (P=0.029). The higher-yielding NIL produced 19.3% more grain than the Hwaseongbyeo recurrent parent (P=0.018). Analysis of a BC3F4 NIL indicated that the variation for GW is associated with variation in grain shape, specifically grain length. The locus, gw8.1 is of particular interest because of its independence from undesirable height and grain quality traits. SSR markers tightly linked to the GW QTL will facilitate cloning of the gene underlying this QTL as well as marker-assisted selection for variation in GW in an applied breeding program.  相似文献

为了鉴定东乡野生稻及其后代群体的耐低氮性,研究低氮和正常氮2种处理下“协青早B//东乡野生稻/协青早B”BC1-F12回交重组自交系株高、抽穗期、穗长、有效穗数、穗实粒数、穗总粒数、着粒密度、结实率、千粒重和单株产量等10个表型性状,利用主成分分析和模糊隶属函数对BILs群体的耐低氮性进行综合评价.结果表明: 株系116、143和157的耐低氮性强,可作为东乡野生稻耐低氮性遗传研究和水稻耐低氮性育种的中间材料.采用逐步回归分析法建立了耐低氮性最优回归方程,筛选到株高、穗总粒数、结实率、千粒重和单株产量等5个性状相对值可作为水稻全生育期耐低氮性的综合评价指标.因此,在水稻耐低氮性遗传改良中,应注重对这5个性状,尤其是穗总粒数和单株产量相对值的选择.

普通野生稻(Oryza Rufipogon)是重要的遗传资源,发掘其优良等位基因将对水稻遗传改良产生重要影响。文章从以珍汕97为轮回亲本,普通野生稻为供体的BC2F1群体中选择一个与珍汕97表型明显不同的单株BC2F1-15,经过连续自交获得回交重组自交系BC2F5群体。均匀分布于12条染色体的126个多态性SSR(Simplesequence repeats)标记基因型分析,发现BC2F1-15单株在30%的标记位点为杂合基因型;利用该群体共检测到4个抽穗期、3个株高、4个每穗颖花数、2个千粒重和1个单株产量QTL。在第7染色体RM481-RM2区间,检测到抽穗期、每穗颖花数和产量QTL,野生稻等位基因表现增效作用;其他3个每穗颖花数QTL位点,野生稻等位基因也均具有增效作用。结果表明野生稻携带有增产相关的等位基因,这些有利等位基因无疑是水稻遗传改良可资利用的新资源。  相似文献

Varietal differences among ten rice cultivars showed that stem diameter is a key factor in lodging resistance (measured in terms of pushing resistance). Two near-isogenic lines (NILs) were selected from a series of chromosome segment substitution lines developed between cultivars Nipponbar and Kasalath, one containing a single stem diameter QTL (sdm8; NIL114), and another with four stem diameter QTLs (sdm1, sdm7, sdm8, sdm12; NIL28). Compared with the Nipponbare control, stem diameters were larger in NIL114 and NIL28 by about 7 and 39%, respectively. Pushing resistance in NIL28 was significantly greater than in Nipponbare, but NIL114 was similar to Nipponbare. The two NILs had greater weight of lower stem and culm wall thickness than Nipponbare. NIL28 had higher plant height, which is a negative effect on lodging resistance, than Nipponbare. The non-structural carbohydrate contents of NIL stems were higher than that of Nipponbare, whereas the silicon contents were lower in the NILs, and cellulose contents were lower only in NIL28. The basal internodes of the two NILs were significantly stiffer than those of Nipponbare. These results suggest that increasing stem diameter in rice breeding programs would improve lodging resistance, although the combination of multiple QTLs would be necessary to produce thicker stems with higher pushing resistance, whereas the higher plant height could also result from the combination of multiple QTLs.  相似文献

稻瘟病是世界范围内影响水稻(Oryza sativa)生产的主要病害。抗稻瘟病基因的发掘和育种利用是控制稻瘟病经济、环保的有效措施。为了揭示云南地方水稻品种子预44广谱持久抗瘟机制, 利用江南香糯和子预44杂交构建的F7重组自交群体, 采用苗期稻瘟病菌自然诱发接种法, 通过调查田间抗瘟性表型数据, 结合基因型数据对子预44中的数量抗瘟性位点进行了分析。结果表明, 在连锁系数(logarithm of odds, LOD)大于2.0的域值上, 共检测出13个QTLs, 分别位于第1、2、6、8、12号染色体上。不同位点表型贡献值差异较大, 范围为5.8%-21.9%, 其中8号染色体上标记RM72-RM404之间的QTLs可解释约61.9%的表型变异, 很可能为一个主效抗瘟QTL位点。多个位点的主效和微效抗性相结合可能是子预44持久稻瘟病抗性的分子基础。  相似文献

Drought is a major abiotic stress limiting rice production and yield stability in rainfed ecosystems. Identifying quantitative trait loci (QTL) for rice yield and yield components under water limited environments will help to develop drought resilient cultivars using marker assisted breeding (MAB) strategy. A total of 232 recombinant inbred lines of IR62266/Norungan were used to map QTLs for plant phenology and production traits under rainfed condition in target population of environments. A total of 79 QTLs for plant phenology and production traits with phenotypic variation ranging from 4.4 to 72.8% were detected under non-stress and drought stress conditions across two locations. Consistent QTLs for phenology and production traits were detected across experiments and water regimes. The QTL region, RM204-RM197-RM217 on chromosome 6 was linked to days to 50% flowering and grain yield per plant under both rainfed and irrigated conditions. The same genomic region, RM585-RM204-RM197 was also linked to harvest index under rainfed condition with positive alleles from Norungan, a local landrace. QTLs for plant production and drought resistance traits co-located near RM585-RM204-RM197-RM217 region on chromosome 6 in several rice genotypes. Thus with further fine mapping, this region may be useful as a candidate QTL for MAB, map-based cloning of genes and functional genomics studies for rainfed rice improvement.  相似文献

Drought is a major abiotic stress factor limiting rice production in rainfed areas. In this study we identified a large-effect quantitative trait locus (QTL) associated with grain yield under stress in five different populations on chromosome 1. The effect of this QTL was further confirmed and characterized in five backcross populations in a total of sixteen stress and non-stress trials during 2006 and 2008. In all the stress trials (eight in total) qDTY1.1 showed strong association with grain yield explaining on average 58% of the genetic variation in the trait. Homozygotes for the tolerant parent allele (Apo) yielded on average 27% more than the susceptible parent allele (IR64) homozygotes. Using an Apo/3*IR64 population, the peak of this QTL (qDTY1.1) was mapped to an interval between RM486 and RM472 at 162.8?cM at a LOD score of 9.26. qDTY1.1 was strongly associated with plant height in all the environments; this was probably due to the presence of the sd1 locus in this genomic region. In a Vandana/3*IR64 population segregating for sd1, a strong relation between plant height and yield under stress was observed. The observed relation between increased height and drought tolerance is likely due to tight linkage between qDTY1.1 and sd1 and not due to pleiotrophy of sd1. Thus there is a possibility of combining reduced plant height and drought tolerance in rice. The large and consistent effect of qDTY1.1 across several genetic backgrounds and environments makes it a potential strong candidate for use in molecular breeding of rice for drought tolerance.  相似文献

以云南元江普通野生稻为供体亲本,在特青的遗传背景下构建了一套BC3高代回交群体。利用117个SSR标记分析383个BC3F2株系的基因型,采用单标记分析法对控制元江普野株高和抽穗期的QTL进行分析。在北京和合肥两个地点试验结果表明,控制株高的QTL分布在第1染色体上,在RM104附近有一个QTL,与sd-1位置相当,其对表现型变异的贡献率在两个地点分别为27%和28%,其加性效应值分别为26.24cm和26.28cm,来自野生稻的等位基因显著提高回交群体的株高;在第1、3、7、8、11染色体共检测到6个控制抽穗期QTL,其中第8染色体RM25附近控制抽穗期的QTL在两个地点的贡献率分别为13%和15%,加性效应值为4.60d和3.65d,来自野生稻的等位基因使回交群体抽穗期延迟。  相似文献

亚铁毒是潜育性水稻土中限制水稻产量的主要因子。利用龙杂8503/IR64的F2和等价的F3群体,在营养液中培养来定位耐亚铁毒的QTLs。通过构建101SSR标记的遗传连锁图谱来确定耐亚铁毒QTLs的位置和特性。借助叶片棕色斑点指数、株高和最大根长3个性状,利用营养液在水稻苗期来评价F2单株、F3群体和亲本龙杂8503、IR64,共检测到叶片棕色斑点指数、株高和最大根长的QTLs20个,分布在水稻的10条染色体上,表明这些性状受多基因控制。控制叶片棕色斑点指数的QTLs分别定位在第1染色体的RM315-RM212、第2染色体的RM6-RM240和第4染色体的RM252-RM451之间。与前人的研究结果比较发现:1)位于第4染色体RM252-RM451之间的控制叶片棕色斑点指数的QTL与水稻功能图谱上控制叶绿素含量减少的QTL的位置一致。另一个位于第1染色体的RM315-RM212之间的控制叶片棕色斑点指数的QTL与水稻功能图谱上位于C178-R2635之间控制叶绿素含量的QTL连锁。2)位于第2染色体RM6-RM240之间的第3个控制叶片棕色斑点指数的QTL与位于RZ58-CD0686的控制钾吸收的QTL连锁。  相似文献

In our previous studies, one putative QTL affecting number of spikelets per panicle (SPP) was identified in the pericentromeric region of rice chromosome 7 using a recombinant inbred population. In order to define the QTL (qSPP7), RI50, a recombinant inbred line with 70% of genetic background same as the female parent of Zhenshan 97, was selected to produce near-isogenic lines for the target region in the present study. In a BC2F2 population consisting of 190 plants, the frequency distribution of SPP was shown to be discontinuous and followed the expected Mendelian ratios (1:2:1 by progeny test) for single locus segregation. qSPP7 was mapped to a 0.4 cM region between SSR marker RM3859 and RFLP marker C39 based on tests of the BC2F2 population and its progeny. Its additive and dominant effects on SPP were 51.1 and 24.9 spikelets, respectively. Of great interest, the QTL region also had effects on grain yield per plant (YD), 1,000 grain weight (GW), tillers per plant (TPP) and seed setting ratio (SR). Significant correlations were observed between SPP and YD (r = 0.66) and between SPP and SR (r = −0.29) in the progeny test. 1082 extremely small panicle plants of a BC3F2 population containing 8,400 individuals were further used to fine map the QTL. It turns out that qSPP7 co-segregated with two markers, RM5436 and RM5499 spanning a physical distance of 912.4 kb. Overall results suggested that recombination suppression occurred in the region and positional cloning strategy is infeasible for qSPP7 isolation. The higher grain yield of Minghui 63 homozygote as compared to the heterozygote suggested that Minghui 63 homozygote at qSPP7 in hybrid rice could further improve its yield. Y. Z. Xing and W. J. 水稻骨干恢复系是指在杂交稻育种中广泛应用的一类恢复系。探明骨干恢复系的遗传基础,发掘其重要农艺性状基因/QTL,对分子标记辅助选择水稻恢复系育种具有重要应用价值。本研究以生产上广泛应用的三系骨干恢复系成恢727和两系骨干恢复系9311为亲本,培育了具有250个系的重组自交系群体。分别在2015年三亚和2016年合肥两个环境下进行了9个重要农艺性状表型和SSR分子标记基因型鉴定,用SAS9.2分析表型数据,用QTL Ici Mapping v4.1进行QTL定位分析。在三亚和合肥两个环境下共检测到39个QTL,三亚检测到16个,分布于第1、2、4、7、8、10、11和12染色体上;合肥检测24个,分布于第1、2、3、7、8、9、10和12染色体上。其中qPH1-1在三亚和合肥两个环境下都能检测到,加性效应分别为-1.75和-2.46。在检测到的39个QTL中,有24个QTL的增效等位基因来自恢复系成恢727,15个QTL的增效等位基因来自9311。共计有26个QTL曾被前人定位,13个属于尚未见文献报道的新QTL。另外,在RM279~RM521、RM336~RM3534、RM25~RM547、RM553~RM160、RM222~RM271区段内检测到5个多效性QTL位点。其中RM25~RM547位点与已经克隆的基因Ghd8位置相近。RM553~RM160位点是一个新的多效性位点,分别控制每穗实粒数、单株产量和结实率,而且效应和表型变异贡献率都较大。其余3个位点在前人的研究中分别有所报道,但其多效性则是在本研究中首次发现。在本研究新发掘到的QTL中,控制穗数的QTL qPN12-1,控制穗长的QTL qPL1-2和qPL10-1,控制总粒数的QTL qSNP2-1和qSNP10-1,控制结实率的QTL qSF3-1,控制千粒重QTL qTGW7-1和控制产量的QTL qGY1-1效应均比较大,解释的表型遗传变异比例也较高。本研究的结果将会为相关性状QTL的精细定位、克隆和育种应用奠定基础。  相似文献

The green rice leafhopper (GRH), Nephotettix cincticeps Uhler, is one of the most serious insect pests affecting cultivated rice (Oryza sativa L.) in temperate regions of East Asia. An accession of the wild rice species, Oryza rufipogon Griff. (W1962), was found to be highly resistant to GRH by an antibiosis test. To understand the genetic basis of the GRH resistance, a BC1F1 population derived from a cross between a susceptible Japonica variety, Taichung 65 (T65), and a highly resistant accession W1962 was analyzed by quantitative trait loci (QTL) mapping. A single major QTL for GRH resistance was detected on rice chromosome 8. A nearly isogenic population containing segments of the targeted QTL region derived from W1962 was then developed through advanced backcrossing with marker-assisted selection. Further molecular mapping using a BC4F2 population revealed that a new resistance gene, designated as Green rice leafhopper resistance 5 (Grh5), was located on the distal region of the long arm of chromosome 8 and tightly linked to the simple sequence repeat markers RM3754 and RM3761. A nearly isogenic line (NIL) carrying Grh5 was subsequently developed in the progeny of the mapping population. The resistance level of Grh5-NIL was compared with those of developed NILs for GRH resistance and was found to have the highest resistance. The DNA markers found to be closely linked to Grh5 would be useful for marker-assisted selection for the improvement of resistance to GRH in rice.  相似文献

N-甲基-N-亚硝基脲(MNU)被用于水稻(Oryza sativa)受精卵的诱变。通过水稻辽盐6号成熟生殖器官的MNU体内同步处理及后代群体筛查, 确立了水稻两性生殖细胞的MNU诱变方法。与辽盐6号受精卵的MNU处理相比, 各组条件下两性生殖细胞的MNU处理明显使M1群体生长发育的指标降低及M1-M2群体中突变性状的发生率升高。两性生殖细胞在含有1.5 mmol?L -1 MNU和10 mmol?L -1 PO4 3-的缓冲液(pH4.8)中处理60分钟, 突变性状发生率是基于受精卵MNU处理的3倍。进一步筛查M3群体, 获得了包含新型植株和籽粒突变体的纯合突变体系列。研究结果表明, 水稻两性生殖细胞的MNU诱变可显著提高广谱诱变效率。该技术的应用可为水稻的未知功能基因鉴定和育种所需的各种突变体规模化开发提供高效的技术支撑。  相似文献
