The possibility of using an embryo test as a means of determining the reaction of wheat varieties to loose smut ( Ustilago tritici (Pers.) Rostr.) has been investigated. Fifty-seven varieties were inoculated with two physiologic races by the partial vacuum method and the embryos, seedlings, and adult plants were examined for infection. Most of the varieties were fully susceptible to one or both races, and only ten showed real resistance to any one race. Braun R and Molinel proved almost immune to infection. The resistance in other varieties which showed no infection in the field was expressed as embryo susceptibility, i.e. the embryo became infected in much the same way as in field-susceptible varieties.
In the latter varieties infection passes from the embryo into the growing point of the young seedling. In the embryo-susceptible field-resistant varieties, infection does not pass from the scutellum and the growing point is therefore uninfected.
The reaction of most of the varieties tested was straightforward resistance or susceptibility, but in a few varieties a small proportion of the grains reacted differently from the majority. These reactions are discussed.  相似文献

A population of 100 F6-derived recombinant inbred lines was developed from the cross of two diploid (2n = 14) Avena accessions, CI3815 (A. strigosa) and C11994 (A. wiestii). Restriction fragment length polymorphism (RFLP) probes previously mapped in other grass species were used to develop a framework linkage map suitable for comparative genetics. Nine linkage groups were identified among the 181 loci mapped, with an average interlocus distance of 5 cM, and a total genetic map length of 880 cM. A cluster of five tightly linked crown rust resistance genes (Pca) was localized on the map, as were five loci identified by disease resistance gene analogs from maize, sorghum, and wheat. None of the five loci identified by the gene analogs were linked to the Pca locus. The linkage map was compared with previously published diploid and hexaploid linkage maps in an attempt to identify homologous or homoeologous chromosomes between populations. Locus orders and linkage relationships were poorly conserved between the A. strigosa x A. wiestii map and other Avena maps. In spite of mapping complications due to duplications within a basic genome a well as the allopolyploid constitution of many Avena species, such map comparisons within Avena provide further evi dence of substantial chromosomal rearrangement between species within Avena.  相似文献

Fluorescent in situ hybridization (FISH) employing multiple probes was used with mitotic or meiotic chromosome spreads of Avena sativa L. cv. SunII and its monosomic lines to produce physical chromosome maps. The probes used were Avena strigosa pAs120a (which hybridizes exclusively to A-genome chromosomes), Avena murphyi pAm1 (which hybridizes exclusively to C-genome chromosomes), A. strigosa pAs121 (which hybridizes exclusively to A- and D-genome chromosomes), and the wheat rDNA probes pTa71 and pTa794. Simultaneous and sequential FISH employing two-by-two combinations of these probes allowed the unequivocal identification and genome assignation of all chromosomes. Ten pairs were found carrying intergenomic translocations: (i) between the A and C genomes (chromosome pair 5A); (ii) between the C and D genomes (pairs 1C, 2C, 4C, 10C, and 16C); and (iii) between the D and C genomes (pairs 9D, 11D, 13D, and 14D). The existence of a reciprocal intergenomic translocation (10C-14D) is also proposed. Comparing these results with those of other hexaploids, three intergenomic translocations (10C, 9D, and 14D) were found to be unique to A. sativa cv. SunII, supporting the view that 'SunII' is genetically distinct from other hexaploid Avena species and from cultivars of the A. sativa species. FISH mapping using meiotic and mitotic metaphases facilitated the genomic and chromosomal identification of the aneuploid chromosome in each monosomic line. Of the 18 analyzed, only 11 distinct monosomic lines were actually found, corresponding to 5 lines of the A genome, 2 lines of the C genome, and 4 lines of the D genome. 采用常规压片法对砂燕麦、西班牙燕麦和短燕麦3个二倍体燕麦种进行了核型研究。结果表明:砂燕麦染色体核型公式为2n=2x=14=10m+4sm(2SAT),具近中部和中部着丝点染色体,第4对染色体组的短臂上有1对随体,核不对称系数为68.17%;西班牙燕麦染色体核型公式为2n=2x=14=10m+4sm(2SAT),具近中部和中部着丝点染色体,第7对染色体短臂上有1对随体,核不对称系数为59.31%;短燕麦染色体核型公式为2n=2x=14=6m+4sm+4st(2SAT),具近端部、近中部和中部着丝点染色体,第6对染色体组的短臂上有1对随体,核不对称系数为63.91%。虽然3个燕麦种的核型均为2A,但它们的染色体形态有明显不同,比较认为砂燕麦相对进化,短燕麦次之,西班牙燕麦较原始。本研究对燕麦种质资源的核型分析及进化地位研究具有参考价值。  相似文献

Characteristics of accumulation and tolerance of cadmium (Cd) in green manure crops were investigated to identify Cd-accumulating crops and to clarify the mechanisms involved in Cd accumulation and tolerance. Seedlings of eight crop species were treated with Cd (1 mg l(-1) or 5 mg l(-1)) in the growing medium for 4 d. Cd concentration in leaves of Avena strigosa Schreb. cv. New-oat, Crotalaria juncea L. and Tagetes erecta L. cv. African-tall was greater than values used to define Cd-hyperaccumulation (>100 mg Cd kg(-1) DW). However, in leaves of T. erecta, lipid peroxidation level increased significantly, and the activities of superoxide dismutase, ascorbate peroxidase, glutathione reductase, and catalase were depressed by both Cd treatments. By contrast, A. strigosa and C. juncea exhibited high Cd tolerance. Avena strigosa leaves showed higher activities of antioxidative enzymes such as superoxide dismutase and ascorbate peroxidase than those of other species tested. Crotalaria juncea showed higher amounts of total soluble phenolics which, in leaves, were doubled by 5 mg l(-1) Cd treatment. When two Cd-tolerant accumulators (A. strigosa and C. juncea) and the non-accumulator (C. spectabilis) were treated with lower Cd concentrations for 4 weeks, A. strigosa and C. juncea exhibited superior Cd accumulation in the shoots with greater biomass production compared with C. spectabilis. These results indicate that A. strigosa and C. juncea possess the greater potential for Cd accumulation and tolerance than common crops.  相似文献

Genomic fluorescent in situ hybridization was employed in the study of the genome organization and evolution of hexaploid oat (Avena sativa L. cv. Sun II, AACCDD, 2n = 6x = 42). Genomic DNAs from two diploid oat species, Avena strigosa (genomic constitution AsAs, 2n = 14) and Avena pilosa (genomic constitution CpCp, 2n = 14), were used as probes in the study. The DNA from A. strigosa labelled 28 of the 42 (2/3) chromosomes of the hexaploid oat, while 14 of the 42 (1/3) chromosomes were labelled with A. pilosa DNA, indicating a close relationship between the A and D genomes. Results also suggested that at least 18 chromosomes (9 pairs) were involved in intergenomic interchanges between the A and C genomes.  相似文献

Microdissection of metaphase chromosome preparations of diploid oat Avena strigosa (2n = 14) allowed isolation of the three individual chromosomes with distinct morphologies, numbers 2, 3 and 7. Using a PCR approach based on the DNA of microdissected chromosomes, STS derivatives of RFLP markers, genetically mapped in Avena spp. linkage maps, have been physically assigned to these three chromosomes. Based on either two or four RFLP-derived STS markers, the A. strigosa chromosomes 2 and 3 were found to be homoeologous to the oat linkage groups C and E, respectively. With the DNA of chromosome 7, four RFLP-derived STS markers located within the central part of linkage group F and two distal ends of linkage group G were amplified. Accordingly, chromosome 7 corresponds to linkage group F and, most probably, is involved in an A. strigosa-specific chromosomal translocation relative to the diploid species Avena atlantica and Avena hirtula, of which the cross progeny was used for linkage mapping of the tested RFLP clones.  相似文献

A saturated genetic map of diploid oat was constructed based on a recombinant inbred (RI) population developed from a cross between Avena strigosa (Cereal Introduction, C.I. 3815) and A. wiestii (C.I. 1994). This 513-locus map includes 372 AFLP (amplified fragment length polymorphism) and 78 S-SAP (sequence-specific-amplification polymorphism) markers, 6 crown-rust resistance loci, 8 resistance-gene analogs (RGAs), one morphological marker, one RAPD (random amplified polymorphic DNA) marker, and is anchored by 45 grass-genome RFLP (restriction fragment length polymorphism) markers. This new A. strigosa x A. wiestii RI map is colinear with a diploid Avena map from an A. atlantica x A. hirtula F2 population. However, some linkage blocks were rearranged as compared to the RFLP map derived from the progenitor A. strigosa x A. wiestii F2 population. Mapping of Bare-1-like sequences via sequence-specific AFLP indicated that related retrotransposons had considerable heterogeneity and widespread distribution in the diploid Avena genome. Novel amplified fragments detected in the RI population suggested that some of these retrotransposon-like sequences are active in diploid Avena. Three markers closely linked to the Pca crown-rust resistance cluster were identified via AFLP-based bulk-segregant analysis. The derived STS (sequence-tagged-site) marker, Agx4, cosegregates with Pc85, the gene that provides resistance specificity to crown-rust isolate 202 at the end of the cluster. This framework map will be useful in gene cloning, genetic mapping of qualitative genes, and positioning QTL (quantitative trait loci) of agricultural importance.  相似文献

The biosynthesis of cyclic triterpenoids in ten saponin-deficient (sad) mutant varieties of the diploid oat Avena strigosa is reported. Two mutants were found to be deficient in 2,3-oxidosqualene:beta-amyrin cyclase (OSbetaAC) (EC 5.4.99) and thus unable to produce the beta-amyrin necessary for the production of avenacins. The other mutants studied had post beta-amyrin lesions. 2,3-Oxidosqualene:cycloartenol cyclase (OSCC) (EC 5.4.99.8) needed for sterol formation was present in all ten mutants.  相似文献

Allohexaploid wild oat, Avena fatua L. (Poaceae; 2n = 6x = 42), is one of the world's worst weeds, yet unlike some of the other Avena hexaploids, its genomic structure has been relatively little researched. Consequently, in situ hybridisation was carried out on one accession of A. fatua using an 18S-25S ribosomal DNA (rDNA) sequence and genomic DNA from A. strigosa (AA-genome diploid) and A. clauda (CC-genome diploid) as probes. Comparing these results with those for other hexaploids studied previously: (i) confirmed that the genomic composition of A. fatua was similar to the other hexaploid Avena taxa (i.e., AACCDD), (ii) identified major sites of rDNA on three pairs of A/D-genome chromosomes, in common with other Avena hexaploids, and (iii) revealed eight chromosome pairs carrying intergenomic translocations between the A/D- and C-genomes in the accession studied. Based on karyotype structure, the identity of some of these recombinant chromosomes was proposed, and this showed that some of these could be divided into two types, (i) those common to all hexaploid Avena species analysed (3 translocations) and (ii) one translocation in this A. fatua accession not previously observed in reports on other hexaploid Avena species. If this translocation is found to be unique to A. fatua, then this information, combined with more traditional morphological data, will add support to the view that A. fatua is genetically distinct from other hexaploid Avena species and thus should retain its full specific status.  相似文献

2003-2013年在甘肃省农业科学院植物保护研究所兰州温室和甘谷试验站,分别对来自国内35个相关育种单位的冬春小麦品种(系)5001份,其中冬小麦4291份、春小麦710份,进行苗期混合菌、成株期分小种和混合菌抗条锈性接种鉴定,结果表明:全生育期表现免疫近免疫的有兰天31号等479份,高抗的有兰天23号等76份,中抗的有天选49等291份,分别占9.58%、1.52%和5.82%;成株期表现免疫近免疫的有天选50号等840份,高抗的有兰天27号等47份,中抗的有天选52等311份,分别占16.80%、0.94%和6.22%;苗期表现免疫近免疫的有兰天30号等964份,高抗的有天98102等122份,中抗的有00-30等273份,分别占19.28%、2.44%和5.46%.冬小麦有天选49号等914份材料表现全生育期抗病,占18.28%;有97-473等906份成株期表现抗病,占18.12%;有兰天20号等1225份苗期表现抗病,占24.50%.春小麦有定西41号等113份材料全生育期表现抗病,占2.26%;有陇春28号等125份成株期表现抗病,占2.50%;有0109-1等114份苗期对混合菌表现抗病,占2.28%.先后在甘肃天水汪川良种场对相关材料进行成株期抗条锈性评价,结果发现:1154份从小种圃筛选出的抗病材料中,表现抗病的有兰天31号等745份,占64.56%;105份甘肃陇南生产品种中,到2013年表现抗病的仅有兰天28号、中梁31号等30份材料,占28.57%;后备品系中,00-30-2-1、CP04-20、00127-2-3等抗性表现优异;抗源材料中,仅有贵农775、中四、T.Spelta albun、贵协1、贵协3等少数材料表现抗病,重要抗源材料贵农21、贵农22、南农92R、川麦42、Moro从2011年开始在田间表现感病,逐步失去利用价值.其衍生系品种材料如陇鉴9343、天选43号、中梁29号、兰天17号、兰天24号等也在田间逐步感病,条锈病发生流行压力持续增大.  相似文献

Three races of Plasmodiophora brassicae were distinguished by their different capacities to form clubs on different hosts; one failed to club turnip and swede and the others were identified by their effects on different turnip and swede varieties.
In contrast to their effects on clubbing, these races caused equal numbers of root-hair infections with zoosporangia, on all crucifers tested. It is evidently the later stages in the life cycle of the pathogen that cannot develop in clubroot-resistant hosts.
Some partially resistant varieties became clubbed when spore concentrations were increased above the level necessary to cause complete infection of susceptible varieties.
Varieties susceptible to a particular race of P. brassicae do not seem to fall into any taxonomic group within the Cruciferae. Susceptible and resistant varieties may belong to the same species or may occur throughout a number of species and genera.  相似文献

采用苗期分小种和成株期混合优势小种法测定了黄淮麦区227份小麦品种和种质材料对小麦条锈菌的抗病性,结果表明,被测品种在苗期对4个条锈菌生理小种的抗病性有很大的差异,抗条中27、30、31、29号小种的品种分别占被测品种总数的93.00%、72.69%、64.76%和61.67%。成株期用6个菌株混合接种测定选出抗病品种76个,占33.48%,其中免疫的为40个,占17.62%。通过对227份小麦品种资源材料的苗期分小种鉴定及与成株期混合优势小种鉴定结果比较,筛选出了成株期和苗期均抗病的品种48个,占被测品种总数的21.14%;成株期抗病苗期感病的品种30个,占被测品种总数的13.22%。  相似文献

In this treatment, the genera Coincya (synonym: Hutera ) and Rhynchosinapis are regarded as one genus under the name Coincya which has nomenclatural priority. This study is based on data from morphology, population studies, cytology, ecology and reproductive biology reinforced by breeding experiments. Coincya is a west European genus comprising 14 taxa (including six species, five subspecies and three varieties).Eight new combinations are proposed and two species are excluded; R.nivalis is referable to the genus Brassica and R. Leplocarpa is probably referable to Erucastrum.
The chromosome number is either 2 n = 24 or 2 n = 48, which confirmed previous counts; C. Transtagana has not been counted. Breeding experiments were carried out with C. Longirostra, C.rupestris, C.monensis subsp. monensis , diploid and tetraploid races of subsp. recurvata var. recurvata , var. johnslonii , var. setigera , var. granatensis , subsp. hispida and subsp. puberula.
All taxa tested are outbreeding with a genetic self-incompatibility mechanism and are cross-compatible, suggesting that there has been no cytological differentiation between the diploid species in this study. The tetraploids formed semi-sterile triploids when crossed with diploids and although the chromosomes only formed bivalents and trivalents at meiosis it was postulated that the tetraploid was an autotetraploid of polytopic origin.

Cytogenetic examination showed that three diploid oat species containing the As genome are highly similar in karyotype structure and chromosome C-banding patterns. Avena strigosa is more similar to A. wiestii, while A. hirtula is to an extent separated from the two species, differing in the C-banding pattern of chromosome 6. The karyotypes of all three species harbor a small acrocentric chromosome, which is absent from diploid oat species containing other variants of the A genome. The results made it possible to assume genome specificity of the rearrangement resulting in this chromosome.  相似文献

Two repetitive sequences, As32 and As22, of 826 and 742 bp, respectively, were isolated from Avena strigosa (As genome). Databank searches revealed their high homology to different segments of the family of Ty1-copia retrotransposons. Southern hybridization showed them to be present in diploid and polyploid oat species. Polymerase chain reaction with primers designed to amplify the segment between them showed that As32 and As22 sequences are composed of two different Ty1-copia retrotransposons. The segment amplified from the pAs32 insert was 2,264 bp long and contained the entire GAG and AP domains, and more than half of the IN domain. This new element has been designated TAS-1 (transposon, A. strigosa, 1) and appears to contain a long open reading frame that encodes a polypeptide of 625 amino acids. Slot-blot and fluorescence in situ hybridization analyses revealed it to be a component of both A- and D-genome chromosomes. Further, the chromosomes involved in one C-A intergenomic translocation in A. murphyi (AC genomes), one C-D intergenomic translocation in A. byzantina cv. Kanota (ACD genomes), and two C-D intergenomic translocations in A. sativa cv. Extra Klock, were identified. Based on its physical distribution and Southern hybridization pattern, a parental retro-transposon represented by TAS-1 appears to have been active at least twice during the evolution of the genomes in species of Avena.  相似文献

甘蔗品种黑穗病抗性评价体系的建立   总被引:8,自引:0,他引:8  
为了建立甘蔗品种黑穗病抗性评价体系,选用9个引进品种,设计一个包括6个对照品种在内的田间试验。首次采用混合小种进行人工浸渍接种,通过整个新植蔗生长季病害进展曲线下的面积、茎感染率和株感染率3个病情指数,以及病害流行学参数潜伏侵染期和持续发病期的分析,对其扰痛性进行评价。在分析以上参数相关程度的基础上,引进了系统聚类分析法进行进一步评价。结果显示;9个引进甘蔗品种中,ROC26属于感病品种,其余属于抗病或高抗品种;3个病情指数和持续发病期的两两相关均为显著正相关,潜伏侵染期与这些参数的相关为负相关,但未达到显著水平;在类间距离大于1.0的条件下,可将15个品种聚为6类,进一步明确了备品种抗黑穗痛性的相似程度;抗性鉴定标准对照种NC0310、F134、NC0376和Ya71—374的应用,明确了接种源为小种1和小种2,通过一次接种试验,明确了供试品种对2个小种的抗性水平,标准对照种的抗性表现,还说明了本生长季发病条件基本是适宜的。本文建立的甘蔗品种抗黑穗病评价体系是切实可行的。  相似文献

中国板栗(Castanea mollissima Blume)对板栗疫病菌(Cryphonectria parasitica(Murr.)Barr)具有强的抗病力。但近十多年来,我国许多省份均发现有板栗疫病,据调查,广西有19个县市存在栗疫病,有的地方发病还相当严重。为了解释疫病发生的上述情况,并为生产和该病菌的更深入研究提供指导,作者在广西桂林、南宁、柳州、梧州、河池等地收集菌株,对疫病菌致病性的分化进行了研究,现将结果报道如下。 1 材料和方法 1.1 毒力参照菌株 法国已知弱毒株EPF为毒力参照株。 1.2 栗疫病菌的采集、分离和单孢纯化 1.2.1 标本采集:1988年12月~1989年6月,采自广西桂林、南宁、柳州、梧州、河池等地板栗病株。 1.2.2 菌株分离和纯化:将病枝或病树皮,用经灯焰灼烧过的刀片削去表皮,取坏死内皮约5×5mm^2,压于PDA上,28℃,光照培养,产孢后用微块法进行单孢纯化。 1.3  相似文献

Preservation and use of wild oat species germplasm are essential for further improvement of cultivated oats. We analyzed the transferability and utility of cultivated (white) oat Avena sativa (AACCDD genome) microsatellite markers for genetic studies of black oat A. strigosa (A(s)A(s) genome) genotypes. The DNA of each black oat genotype was extracted from young leaves and amplified by PCR using 24 microsatellite primers developed from white oat. The PCR products were separated on 3% agarose gel. Eighteen microsatellite primer pairs amplified consistent products and 15 of these were polymorphic in A. strigosa, demonstrating a high degree of transferability. Microsatellite primer pairs AM3, AM4, AM21, AM23, AM30, and AM35 consistently amplified alleles only in A. sativa, which indicates that they are putative loci for either the C or D genomes of Avena. Using the data generated by the 15 polymorphic primer pairs, it was possible to separate 40 genotypes of the 44 that we studied. The four genotypes that could not be separated are probably replicates. We conclude that A. sativa microsatellites have a high transferability index and are a valuable resource for genetic studies and characterization of A. strigosa genotypes.  相似文献

Sequences analogous to plant resistance genes of the NBS-LRR class were cloned from the genomic DNA of 11 Avena species with different genomes and levels of ploidy. Three pairs of degenerate primers were used, based on conserved DNA sequence motifs belonging to the NBS domain, and 33 sequences were identified. These were subdivided into 7 classes depending on nucleotide sequence identity. Despite the high level of degeneracy, the primers behaved in a highly selective way; the majority of sequences from the different species obtained with every primer combination showed strong identity and were considered homologous. For most species, only one sequence of each class was identified in each genome, suggesting that duplicated sequences are fairly divergent. The strong identity among specific NBS sequences precludes any conclusions being made on the evolution of these species. The genomic organization of the RGA sequences was explored using those of A. strigosa as probes in Southern blots involving digested DNA from 15 Avena species. The hybridization patterns showed wide diversity both among sequences within a species and among species for each sequence. However, the dendrogram generated using the RFLPs showed relationships among species to be in good agreement with those previously established using other molecular markers.  相似文献
