Production of bikaverin has been examined in 130Fusarium isolates belonging to 21 species. The highest yield of bikaverin was produced on autoclaved rice — up to 2.5g/kg of dry culture. Bikaverin was produced by the following species:F verticillioides, F sacchari varsubglutinans, F proliferatum, F anthophilum, F oxysporum, F dlamini, F nygamai, F napiforme, andF solani. SpeciesF coeruleum, F poae, F sporotrichioides, F tricinctum, F chlamydosporum, F culmorum, F graminearum, F cerealis (F crookwellense), F avenaceum, F acuminatum, andF equiseti did not produce bikaverin. The production of bikaverin determines the colour of the mentionedFusarium species cultures on agar media and on rice. The pigment has indicator properties and changes colour from red in acidic solution to violet-blue in alkaline. The role it plays in fungus metabolism is not elucidated. 相似文献

Five new cyclohexadepsipeptides termed as enniatins R – V ( 1 – 5 ) and seven known cyclohexadepsipeptides ( 6 – 12 ) were isolated from the solid culture of Fusarium proliferatum, a fungus isolated from the cadaver of an unidentified insect collected in Tibet. Their structures were elucidated by NMR and MS spectroscopic analysis. The X‐ray single‐crystal structure of 6 was reported for the first time. Enniatins R and S represented the first enniatins incorporating with an unusual 2,3‐dihydroxy‐isovaleric acid (Div) residue. The cytotoxicity and autophagy‐inducing activities of 1 – 12 were evaluated in vitro. Beauvenniatin F ( 11 ) exhibited strong cytotoxicity against K562/A (adriamycin‐resistant K562) with IC50 value of 3.78 μm , and also autophagy‐inducing activity at the concentration of 20 μm in GFP‐LC3 stable HeLa cells. 相似文献

The presence of chitinase activity without inducers in the enzymic precipitates from the culture fluid of 25-day-old autolyzed cultures of 17 Fusarium species has been studied. In all cases endochitinase and -N-acetylglucosaminidase activities were found. The chitinase activity as a joint action of these two enzymes with production of N-acetylglucosamine was also determined. A correlation among endochitinase, -N-acetylglucosaminidase, and chitinase was always found. Fusarium oxysporum f.sp. lini, F. subglutinans, and F. moniliforme were the best producers of chitinase activity. Fusarium species could be a good source of chitinases for production by fungi. 相似文献

Summary The lipid constituents of Fusarium aquaeductuum Lagerh., F. aquaeductuum Lagerh. var. medium and F. sambucinum Fuck. have been studied. In the mycelia of these three fungi -carotene, lycopene and rhodoxanthin were present. Ergosterol was also present and the fatty acid pattern showed a slight qualitative difference between the three fungi.Use of the presence of carotenoids in fungi as a taxonomic tool is discussed and it is concluded that it cannot yet be considered reliable. 相似文献

Contamination of small-grain cereals with the fungal species Fusarium graminearum, F. culmorum, F. poae, F. sporotrichioides and F. equiseti is an important source of trichothecenes, Zearalenone and other mycotoxins which cause serious diseases in human and animals. Additionally, these species contribute to Fusarium Head Blight, a disease which produces important losses in cereal yield. Early detection and control of these Fusarium species is crucial to prevent toxins entering the food chain and a useful tool in disease management practices. We describe the development of specific PCR assays to F. graminearum, F. culmorum, F. poae, F. sporotrichioides and F. equiseti using DNA from pure fungal cultures as well as from naturally infected wheat seeds, using in this case a rapid and easy protocol for DNA isolation. The specific primers were designed on the basis of IGS sequences (Intergenic Spacer of rDNA), a multicopy region in the genome that permits to enhance the sensitivity of the assay in comparison with PCR assays based on single-copy sequences. 相似文献

Fumonisins are polyketide-derived mycotoxins, produced by several Fusarium species, and its biosynthetic pathway is controlled by the FUM cluster--a group of genes exhibiting a common expression pattern during fumonisin biosynthesis. The most common are the B analogues with fumonisin B(1) (FB(1)) being the most prevalent. At least a part of the inter- and intraspecific variation in FBs synthesis level can be explained by the sequence differences inside FUM cluster. The aim of our study was to evaluate the toxin production and sequence variability in FUM genes and intergenic regions among thirty isolates of seven species reported as potential fumonisins producers: Fusarium anthophilum, Fusarium fujikuroi, Fusarium nygamai, Fusarium oxysporum, Fusarium proliferatum, Fusarium subglutinans and Fusarium verticillioides, particularly with respect to FBs synthesis. Fumonisins were produced in high amounts (over 1mg g(-1)) by one isolate of F. subglutinans, three of F. verticillioides and all F. proliferatum isolates except one, regardless of the host organism. The remaining isolates produced low amounts of FBs and two F. verticillioides isolates didn't produce it at all. The lowest variation in amount of toxin produced was found among F. proliferatum isolates. Based on the translation elongation factor 1α (tef-1α) sequence of F. fujikuroi, a species-specific marker was developed. The intergenic region presents similar opportunity for F. nygamai identification. The phylogenetic reconstruction based on FUM1 gene generally reflects the scenario presented by tef-1α sequences. Although the sequence similarities for intergenic regions were lower than in coding regions, there are clearly conserved patterns enabling separation of different subsets of species, including the non-producer species. 相似文献

Production of extracellular cellulase by an isolate of a Fusarium sp. has been studied in shake cultures, and sequential appearance of cellulase components (- glucosidase on the first day, endo--glucanase on second day and exo--glucanase on the third day of growth on insoluble cellulose) was observed. Maximum production of all these components was achieved on the fifth day. The Fusarium produced significantly higher -glucosidase within a short period of time as compared with Trichoderma reesei. The influence of different nitrogen and carbon sources on cellulase has been investigated. Crude cellulolytic enzyme was used for hydrolysis of common agricultural wastes both with and without sodium hydroxide pretreatment. Analysis of hydrolysates indicated glucose as the major constituent (about 83% of total reducing sugar). 相似文献

Interspecies interactions between Fusarium avenaceum, Fusarium culmorum, Fusarium graminearum, Fusarium poae, and Fusarium tricinctum were studied during early growth stages of isolates on model surfaces. Additionally, germination and germ tube growth of the pathogens were studied on attached and detached wheat leaves at 10 °C and 22 °C. Two-species interactions between Fusarium isolates during germination and germ tube growth were assessed after 8 hours of incubation. All species except F. tricinctum germinated and grew faster at higher than lower temperature. All species were able to germinate with more than one germ tube per conidium cell; and germination and germ tube growth were faster on leaves than on glass surface. Interactions among Fusarium species during germination and germ tube growth were predominantly competitive with macroconidia-producing species being more competitive. It is concluded that the type of conidia as well as environmental factors influence the competitiveness of Fusarium species during early stages of growth. 相似文献

Fumonisins B1 (FB1) and B2 (FB2), two structurally related mycotoxins with cancer-promoting activity, were recently isolated from corn cultures of Fusarium moniliforme MRC 826. These toxins have been reported to be produced also by isolates of F. proliferatum. Contamination of foods and feeds by F. moniliforme has been associated with human esophageal cancer risk, and FB1 has been shown to be the causative agent of the neurotoxic disease leukoencephalomalacia in horses. Because of the toxicological importance of the fumonisins, the potential to produce FB1 and FB2 was determined in a study of 40 toxic Fusarium isolates representing 27 taxa in 9 of the 12 sections of Fusarium, as well as two recently described species not yet classified into sections. With the exception of one isolate of F. nygamai, fumonisin production was restricted to isolates of F. moniliforme and F. proliferatum, in the section Liseola. The F. nygamai isolate produced 605 micrograms of FB1 g-1 and 530 micrograms of FB2 g-1, and the identity of the toxins was confirmed by capillary gas chromatography-mass spectrometry. This is the first report of the production of the fumonisins by F. nygamai. 相似文献

Cellulose-acetate electrophoresis (CAE) was used to investigate isozyme polymorphisms among different isolates of Fusarium cerealis, F. culmorum, F. graminearum and F. pseudograminearum. After initial testing of 18 enzymes in three buffer systems for activity and resolution of bands, 12 proved to be appropriate for analysis of the full sample set. Comparing the different electrophoretic types (ETs), adenylate kinase (AK), NADP dependent glutamate dehydrogenase (NADP GDH), peptidase B (PEP B), peptidase D (PEP D) and phosphoglucomutase (PGM) proved to be diagnostic for at least one species examined. However, only PEP D was useful alone as a marker to distinguish the four taxa studied providing a rapid and simple CAE based diagnostic protocol. 相似文献

Isolates of Fusarium were obtained from samples of barley grain at harvest in 1971, 1973 and 1974. The ability of these isolates to produce the myco-toxin zearalenone was examined, the grain also being tested for its presence in 1974. The most common isolate was F. culmorum, 60% of which in 1974 produced zearalenone. Other species isolated which produced this myco-toxin were F. monilijorme, F. graminearum, F. avenaceum, F. nivale and F. sambucinum var coeruleum, the last three species have not previously been recorded as producing zearalenone. 相似文献

An improved TLC method ofFusaria metabolites detection and quantitation has been elaborated. A total 92 isolates of Discolor sectionFusaria from cereals and potato have been examined from the point of view of cultures morphology and ability to produce characteristic mycotoxins. Low nutrient media (CLA, SNA) were found as suitable for production of uniform and typical macroconidia in studied cultures. All 26 isolates ofF. sambucinum Fuckel (=F.sulphureum, Schlecht) formed diacetoxyscirpenol in amount 20-1000 mg/kg and all 17.F. crookwellense Burgess N. & T. produced zearalenone (16-602 mg/kg).F. graminearum Schwabe produced: zearalenone 14/14 isolates, deoxynivalenol 11/14 isolates, both up to 77 mg/kg. Out of 26F. culmorum cultures originating from Poland 22 produced zearalenone up to 675 mg/kg, 17/26 3 acetyldeoxynivalenol up to 280 mg/kg and 16/26 deoxynivalenol up to 220 mg/kg. genus by the following characteristics: whitish colony color, long zigzag-like chains of 0–3(-5)-septate clavate conidia,
