Megaselia scalaris (Loew) is a cosmopolitan and synanthropic scuttle fly, eclectic in its feeding habits and acts as detritivore, parasite, facultative parasite, and parasitoid. Here we report for the first time M. scalaris infesting laboratory colonies of Triatoma brasiliensis Neiva, the most important Chagas disease vector in semiarid areas of Brazil. M. scalaris larvae were found feeding inside bugs; pupae were found in the esophagus and intestinal regions of T. brasiliensis through dissection. Other relevant information about this finding is also described in this note, including some preventive measures to avoid laboratory colonies infestations.  相似文献

The morphology and life history of a new species of the genus Botryllus belonging to the family Botryllidae are described in detail. This ascidian was collected from the stony shore in the cove near Shimoda Marine Research Center, University of Tsukuba (Shimoda, Shizuoka Prefecture, Japan). The ascidian colony was easily distinguished from colonies of other botryllids because it was very thin and bright pink in color. The arrangement of ovary and testis in this ascidian was the same as that in other species of the genus Botryllus. This ascidian was prolific, with 1-5 embryos on each side of a zooid, and the embryos of this ascidian developed in the peribranchial cavity without any brooding organs as in Botryllus scalaris. We observed the processes and features of the allorecognition reaction in colony specificity and found that allorejection occurred after fusion of the vascular system between two incompatible colonies. This manner of allorejection is also shown in B. scalaris and Botryllus delicatus; however, the reaction speed of allorejection is faster than that of B. delicatus and similar to that of B. scalaris. These results indicate that this ascidian might be closely related to B. scalaris.  相似文献

Sex-lethal (Sxl) and doublesex (dsx) are known to represent parts of the sex-determining cascade in Drosophila melanogaster. We generated cDNA probes of the homologous genes from Megaselia scalaris, a fly species with an epistatic maleness factor as the primary sex determining signal. In Northern blot hybridization of poly(A)+ RNA, the M. scalaris dsx probe detected two bands, one of which had a sex-specific size difference, while the Sxl probe bound to RNAs of equal size in females and males. RT-PCR showed Sxl to be transcribed in gonads of adult females and males but not in somatic tissues. Thus, while dsx appears to have a similar function in M. scalaris and D. melanogaster, Sxl does not. The results suggest that the sex-determining pathway of M. scalaris joins that of D. melanogaster between the Sxl and dsx steps.  相似文献

The ultrastructure of the first and second-instar larvae of Megaselia scalaris (Loew) was studied using scanning electron microscopy (SEM). Significant changes in morphological features were observed in the anterior and posterior spiracles, but only minimal changes in the labium and mouthhooks were seen. The ultrastructure of M. scalaris larvae not only provides chronological transformation of their larval instars, but it can also be used to explain their feeding behavior and mode of respiration. In addition, morphological structures useful for specific identification of first or second-instar larvae collected from human corpses may be used in forensic investigations.  相似文献

Abstract The binding capacity of protein fractions ( M r > 8000) from five lichens, Hypocenomyce scalaris, Hypogymnia physodes, Ramalina fastigiata, Umbilicaria pustulata and Xanthoria parietina was tested towards their freshly isolated phycobionts and towards ten cultures of phycobionts, five of which were from the above-mentioned lichens. The protein fractions from H. scalaris, R. fastigiata, U. pustulata and X. parietina bind intensely to a cultured Pseudotrebouxia from H. physodes and a cultured Trebouxia from X. parietina , but not to the other cultured algae, and not to the freshly isolated algae. A protein fraction between M r 3500 and 8000 from H. physodes shows a faint binding to its own cultured algae. The nature of the binding and its relevance in connection with the mutual recognition of alga and fungus are briefly discussed.  相似文献

The evolutionary transition between oviparity and viviparity in squamate reptiles presumably occurs via a gradual increase in the duration of egg retention, the production of thinner eggshells, and increases in the vascularity of maternal and embryonic tissues. The 'ease' of this transition may differ among taxa. For example, in the genus Sceloporus , the scalaris species group contains both oviparous and viviparous species, and female Sceloporus scalaris can extend egg retention facultatively in response to the absence of a suitable site for oviposition without impairing embryonic development. In contrast, the undulatus species group contains only oviparous species, and, while female Sceloporus virgatus can extend egg retention, doing so retards embryonic development. I tested several hypotheses that would explain the greater ability of 5. scalaris than S. virgatus to extend egg retention. In this study, female S. scalaris retained eggs for 19 d without affecting the mortality of embryos, total developmental time, or dry mass of hatchlings. In contrast, when female S. virgatus retained eggs for 18 d, embryos had very high mortality and eggs took significantly longer to hatch than control (non-retained) eggs, although the dry mass of hatchlings was not affected. The ability of S. scalaris females to retain eggs with little negative effect on embryonic development was associated with relatively large chorioallantois, relatively thin eggshells, and relatively small clutch masses. These observations suggest that phylogenetic differences in the ability to extend egg retention may facilitate or constrain the evolution of viviparity in some lineages.  相似文献

ABSTRACT. Running activity was filmed at high speed in two species of phorid fly, Megaselia scalaris and Puliciphora borinquenensis , which differ in weight by about one order of magnitude. Under all experimental conditions, activity consisted of brief runs regularly interrupted by short halts when the insect was completely immobile. Runs and halts were approximately equal in duration and occurred five to eight times a second. In crowded conditions the flies made longer stops and usually also shorter runs. Maximum instantaneous velocities during running were 0.2 m s-1 in M. scalaris and 0.09 m s-1 in male P. borinquenensis. Maximum relative velocities were 80 and 112 fly-lengths s-1 respectively. In M. scalaris , step frequencies reached at least 57 Hz and this rate can probably be exceeded. Fast running was accompanied by small side-to-side oscillations of the body at the step frequency. Fast turns were normally made at the start of runs and only rarely at other times. Painting the compound eyes or removing the antennae did not disrupt the stop—go pattern of running, although flies so treated were reluctant to run. Possible visual and chemosensory functions for stop—go running are discussed.  相似文献

Twenty-six species of silica-scaled chrysophytes representing the genera Chrysosphaerella (1 species), Mallomonas (19 species) and Synura (6 species) are reported based on transmission electron microscopy from the springtime flora of the slightly acidic bog-lake Baláta-tó, a Nature Reserve, situated in Southwest Hungary. Ten of them ( Mallomonas alata, M. clavus, M. insignis, M. mangofera, M. oviformis, M. papillosa, M. pillula, M. scalaris, M. transsylvanica and Synura multidentata ) are new records for Hungary. Furthermore, two species, M. pillula and M. scalaris are for the first time found in the Carpathian Basin and M. alata f. hualvensis is a new record for the flora of Europe. The well-known sex-determining cascade of Drosophila melanogaster serves as a paradigm for the pathway to sexual development in insects. But the primary sex-determining signal and the subsequent step, Sex-lethal (Sxl), have been shown not to be functionally conserved in non-Drosophila flies. We isolated doublesex (dsx), which is a downstream step in the cascade, from the phorid fly Megaselia scalaris, which is a distant relative of D. melanogaster. Conserved properties, e.g., sex-specific splicing, structure of the female-specific 3' splice site, a splicing enhancer region with binding motifs for the TRA2/RBP1/TRA complex that activates female-specific splicing in Drosophila, and conserved domains for DNA-binding and oligomerization in the putative DSX protein, indicate functional conservation of dsx in M. scalaris. Hence, the dsx step of the sex-determining pathway appears to be conserved among flies and probably in an even wider group of insects, as the analysis of a published cDNA from the silkmoth indicates.  相似文献

Different laboratory cultures of the acarine tick Rhipicephalus (Boophilus) microplus (Canestrini, 1888) (Ixodida: Ixodidae) were infested by small Megaselia scalaris (Loew, 1866) (Diptera: Phoridae) flies. Larvae of this species exhibited opportunistic parasitism predominantly on engorged female ticks, causing severe damage to their cuticle through which the flies were able to reach R. microplus internal organs, on which they fed until developing into pupae in the tick's remains. The flies were kept by continuous propagation on fresh ticks over six generations during which the same parasitoid behaviour was observed. Here we report on an ixodid tick laboratory culture used for rearing M. scalaris.  相似文献

Xenogeneic rejection was observed among colonies of three botryllids, Botryllus scalaris, Botryllus primigenus, and Botrylloides simodensis. Allogeneic recognition occurs in each of these species, but the manner of allogeneic rejection differs among them. We studied xenogeneic rejection reactions among these species under the following conditions: colony contact at natural growing edges, colony contact at artificially cut surfaces, and injection of xenogeneic blood plasma into a vascular vessel. In the first two cases, xenogeneic rejection occurred only in Botryllus primigenus and Botrylloides simodensis. The features of that xenogeneic rejection were similar to those of allogeneic rejection in each of these two botryllids. Injection of xenogeneic blood plasma induced responses similar to those of allogeneic rejection in all three botryllids. It is interesting to note that colonies of Botryllus scalaris never showed any response against injected blood plasma from allogeneic incompatible colonies, unlike the responses seen in colonies of the other two botryllids under the same conditions. On the basis of these results, the relationship between allogeneic and xenogeneic rejection in botryllids is discussed.  相似文献

Environmental contamination with metals such as manganese (Mn) and nickel (Ni) often results in elevated concentrations of these metals in plant tissues. At high concentrations, these metals are known to have detrimental effects on certain insect herbivores. Using laboratory bioassays and artificial diet, we investigated the development and survival of a cosmopolitan insect detritivore, Megaselia scalaris (Diptera: Phoridae), exposed to concentrations of Mn and Ni reaching 2600 mg Mn/kg and 5200 mg Ni/kg dry mass (dm) in artificial diet. Surprisingly, Ni and Mn at the concentrations tested did not harm this fly. Treatment groups from diets with 260–2600 mg Mn/kg dm and 1300–5200 mg Ni/kg dm had significantly shorter larval development times, overall times to adult emergence, and both pupariation and pupal eclosion times compared to a control group. Wing length of females, a correlate of adult fitness, was also greater in metal treatment groups. Other measures including rate of egg hatch, percentage of emerging flies that were female, and wing length of male flies, were not significantly different in metal treatment groups. We conclude that Megaselia scalaris is tolerant of exceptionally high levels of Mn and Ni.  相似文献

We describe TROMB, a new LTR retrotransposon, from the phorid fly Megaselia scalaris. Three full-length copies (4226, 4160 and 4129bp) and a truncated one (319bp) have been isolated. The target site consensus is TATAT, with a 4bp target site duplication TATA. The LTRs are short (142bp) and contain a TATA-box and a polyadenylation signal. The isolated copies are degenerate to different degrees and presumably inactive. The polyprotein coding sequence contains scattered stop codons and deletions/insertions at non-homologous positions. The consensus sequence among the three full-length copies, however, has an uninterrupted open reading frame and, presumably, represents the original sequence of the active element. Southern hybridization experiments showed TROMB to be present at a low copy number in two wild-type strains of M. scalaris and absent in a related species, M. abdita. The order of domains in the polyprotein coding region, the target site specificity for AT-rich sequences, and the protein sequence similarity to blastopia, mdg3 and micropia place TROMB in the gypsy-Ty3 group of LTR retrotransposons.  相似文献

Chloroplasts gather in areas irradiated with weak light to maximize photosynthesis (the accumulation response). They move away from areas irradiated with strong light to minimize damage of the photosynthetic apparatus (the avoidance response). The processes underlying these chloroplast movements can be divided into three parts: photoperception, signal transduction, and chloroplast movement. Photoreceptors for chloroplast movement have been identified recently in various plant species. A blue light receptor phototropin (phot) mediates chloroplast photorelocation movement in the seed plant Arabidopsis thaliana, the fern Adiantum capillus-veneris, the moss Physcomitrella patens and possibly the green alga Mougeotia scalaris. A chimeric photoreceptor between phytochrome and phototropin, neochrome (neo), was found in some advanced ferns and in the green alga M. scalaris. While the mechanism of chloroplast movement is not well understood, it is known that actin filaments play an important role in this process. To understand the molecular mechanisms associated with chloroplast movement, several mutants were isolated in A. thaliana (jac1 and chup1) and the corresponding genes were cloned. In this review, recent progress in photoreceptor research into chloroplast movement in various plant species and the possible factors functioning in signal transduction or the regulation of actin filaments identified in A. thaliana is discussed.  相似文献

We describe some ultrastructure of the third-instar Megaselia scalaris (Diptera: Phoridae) using scanning electron microscopy, with the cephalic segment, anterior spiracle and posterior spiracle being emphasized. Samples were collected from southern, central and eastern regions of Saudi Arabia using Malaise traps and sweep nets. Nine species of Tabanidae were identified, two for the first time from Saudi Arabia, Hybomitra peculiaris (Szilády) and Atylotus pulchellus (Loew).Therefore, the total number of Tabanidae in Saudi Arabia is 31 species. Remarks of the species recorded in this study were given. A key to the genera of Tabanidae occurring in the Arabian Peninsula is also provided. Available literature for Saudi Arabian Tabanidae is summarized and provided. It is concluded that the tabanid fauna of Saudi Arabia is more similar to that of the Palaearctic region than to the Afrotropical region.  相似文献

Apple snails (Ampullariidae: Pomacea ) native to the New World have become agricultural and environmental pests widely in southern and eastern Asia since their introduction in about 1980. Although their impacts have been extensively documented, considerable confusion persists regarding their identities and geographical origins. Efforts to resolve the confusion have suffered from inadequate taxonomic and geographical sampling from both native and introduced ranges. Using phylogenetic and genealogical methods, we analysed 610–655 bp of cytochrome c oxidase subunit I DNA sequences from 783 apple snails from 164 Asian locations and 57 native South American locations. In Asia, we found four species of Pomacea in two clades: (1) Pomacea canaliculata and P. insularum , and (2) P. scalaris and P. diffusa . Parsimony networks and mismatch distributions indicate that the non-native ranges of the two most widespread species, P. canaliculata and P. insularum , probably result from multiple introductions. Molecular analyses are consistent with early accounts; non-native P. canaliculata populations trace back to multiple locations in Argentina and have probably been introduced more than once. In contrast, P. insularum was probably introduced from Brazil and Argentina independently. Multiple introductions may, in part, explain the success and rapid spread of these two species. Unlike P. canaliculata and P. insularum , P. scalaris and P. diffusa were probably introduced through the aquarium trade, derived originally from Argentina and Brazil, respectively. Possible physiological, ecological, and native range differences among these four species highlight the importance of accurate identification in understanding invasion patterns and processes, which is vital in developing and implementing management strategies.  相似文献

The Phoridae is known as 'scuttle flies' because they walk in rapid bursts of movement with short pauses between. In this study, larval locomotive behavior and development was characterized in the phorid, Megaselia scalaris. Comparison was made with the well-characterized fruit fly model, Drosophila melanogaster. Developmentally, the rate of maturation was consistently slower for Megaselia than Drosophila. This disparity was exaggerated at lower temperatures, particularly during larval development. In addition to slower growth, movements in Megaselia were also slower, as evidenced by reduced rates of larval body wall contractions and mouth hook movements. Megaselia larvae also displayed a unique behavior of swallowing air when exposed to a small pool of liquid. This permitted floating upon immersion and, therefore, might prevent drowning in the natural environment. The anatomical and physiological properties of a neuromuscular junction in the phorid larvae were also examined. The innervation of the motor nerve terminals on the ventral abdominal muscle (m6) is innervated by Type Ib and Is axons, similar to Drosophila. As in Drosophila, the Is terminals produce larger excitatory postsynaptic potentials (EPSPs) than the Ib. The amplitudes of the EPSPs in M. scalaris were reduced compared to those of D. melanogaster, but unlike D. melanogaster the EPSPs showed marked facilitation when stimulated with a 20 Hz train. We conclude that there may be differences in synaptic structure of the nerve terminals that could account for the different electrophysiological behaviors.  相似文献
